    • The first generation of ELT instruments includes an optical-infrared high resolution spectrograph, indicated as ELT-HIRES and recently christened ANDES (ArmazoNes high Dispersion Echelle Spectrograph). ANDES consists of three fibre-fed spectrographs (UBV, RIZ, YJH) providing a spectral resolution of similar to 100,000 with a minimum simultaneous wavelength coverage of 0.4-1.8 mu m with the goal of extending it to 0.35-2.4 mu m with the addition of a K band spectrograph. It operates both in seeing- and diffraction-limited conditions and the fibre-feeding allows several, interchangeable observing modes including a single conjugated adaptive optics module and a small diffraction-limited integral field unit in the NIR. Its modularity will ensure that ANDES can be placed entirely on the ELT Nasmyth platform, if enough mass and volume is available, or partly in the Coude room. ANDES has a wide range of groundbreaking science cases spanning nearly all areas of research in astrophysics and even fundamental physics. Among the top science cases there are the detection of biosignatures from exoplanet atmospheres, finding the fingerprints of the first generation of stars, tests on the stability of Nature's fundamental couplings, and the direct detection of the cosmic acceleration. The ANDES project is carried forward by a large international consortium, composed of 35 Institutes from 13 countries, forming a team of more than 200 scientists and engineers which represent the majority of the scientific and technical expertise in the field among ESO member states.

  • Three planets transiting the evolved star EPIC 249893012: a hot 8.8-Mearth super-Earth and two warm 14.7 and 10.2-Mearth sub-Neptunes

    • We report the discovery of a new planetary system with three transiting planets, one super-Earth and two sub-Neptunes, that orbit EPIC 249893012, a G8 IV-V evolved star (M⋆ = 1.05 ± 0.05 M☉, R⋆ = 1.71 ± 0.04 R☉, Teff = 5430 ± 85 K). The star is just leaving the main sequence. We combined K2 photometry with IRCS adaptive-optics imaging and HARPS, HARPS-N, and CARMENES high-precision radial velocity measurements to confirm the planetary system, determine the stellar parameters, and measure radii, masses, and densities of the three planets. With an orbital period of 3.5949-0.0007+0.0007days, a mass of 8.75-1.08+1.09 M⊕, and a radius of 1.95-0.08+0.09 R⊕, the inner planet b is compatible with nickel-iron core and a silicate mantle (ρb = 6.39-1.04+1.19 g cm-3). Planets c and d with orbital periods of 15.624-0.001+0.001 and 35.747-0.005+0.005 days, respectively, have masses and radii of 14.67-1.89+1,84 M⊕ and 3.67-0.14+0.17 R⊕ and 10.18-2.42+2.46 M⊕ and 3.94-0.12+0.13 R⊕, respectively, yielding a mean density of 1.62-0.29+0.30 and 0.91-0.23+0.25 g cm-3, respectively. The radius of planet b lies in the transition region between rocky and gaseous planets, but its density is consistent with a rocky composition. Its semimajor axis and the corresponding photoevaporation levels to which the planet has been exposed might explain its measured density today. In contrast, the densities and semimajor axes of planets c and d suggest a very thick atmosphere. The singularity of this system, which orbits a slightly evolved star that is just leaving the main sequence, makes it a good candidate for a deeper study from a dynamical point of view.

  • Validation of a Novel Multivariate Method of Defining HIV-Associated Cognitive Impairment

    • BackgroundThe optimum method of defining cognitive impairment in virally suppressed people living with HIV is unknown. We evaluated the relationships between cognitive impairment, including using a novel multivariate method (NMM), patient– reported outcome measures (PROMs), and neuroimaging markers of brain structure across 3 cohorts.MethodsDifferences in the prevalence of cognitive impairment, PROMs, and neuroimaging data from the COBRA, CHARTER, and POPPY cohorts (total n = 908) were determined between HIV-positive participants with and without cognitive impairment defined using the HIV-associated neurocognitive disorders (HAND), global deficit score (GDS), and NMM criteria.ResultsThe prevalence of cognitive impairment varied by up to 27% between methods used to define impairment (eg, 48% for HAND vs 21% for NMM in the CHARTER study). Associations between objective cognitive impairment and subjective cognitive complaints generally were weak. Physical and mental health summary scores (SF-36) were lowest for NMM-defined impairment (P < .05).There were no differences in brain volumes or cortical thickness between participants with and without cognitive impairment defined using the HAND and GDS measures. In contrast, those identified with cognitive impairment by the NMM had reduced mean cortical thickness in both hemispheres (P < .05), as well as smaller brain volumes (P < .01). The associations with measures of white matter microstructure and brain-predicted age generally were weaker.ConclusionDifferent methods of defining cognitive impairment identify different people with varying symptomatology and measures of brain injury. Overall, NMM-defined impairment was associated with most neuroimaging abnormalities and poorer self-reported health status. This may be due to the statistical advantage of using a multivariate approach.

  • Into the storm : Diving into the winds of the ultra-hot Jupiter WASP-76 b with HARPS and ESPRESSO

    • Context. Despite swift progress in the characterisation of exoplanet atmospheres in composition and structure, the study of atmospheric dynamics has not progressed at the same speed. While theoretical models have been developed to describe the lower layers of the atmosphere, and independently, the exosphere, little is known about the intermediate layers up to the thermosphere. Aims. We aim to provide a clearer picture of atmospheric dynamics for the class of ultra-hot Jupiters, which are highly irradiated gas giants, based on the example of WASP-76 b. Methods. We jointly analysed two datasets that were obtained with the HARPS and ESPRESSO spectrographs to interpret the resolved planetary sodium doublet. We then applied the MERC code, which retrieves wind patterns, speeds, and temperature profiles on the line shape of the sodium doublet. An updated version of MERC, with added planetary rotation, also provides the possibility of modelling the latitude dependence of the wind patterns. Results. We retrieve the highest Bayesian evidence for an isothermal atmosphere, interpreted as a mean temperature of 3389 ± 227 K, a uniform day- to nightside wind of 5.5-2.0+1.4 km s-1 in the lower atmosphere with a vertical wind in the upper atmosphere of 22.7-4.1+4.9 km s-1, switching atmospheric wind patterns at 10-3 bar above the reference surface pressure (10 bar). Conclusions. Our results for WASP-76 b are compatible with previous studies of the lower atmospheric dynamics of WASP-76 b and other ultra-hot Jupiters. They highlight the need for vertical winds in the intermediate atmosphere above the layers probed by global circulation model studies to explain the line broadening of the sodium doublet in this planet. This work demonstrates the capability of exploiting the resolved spectral line shapes to observationally constrain possible wind patterns in exoplanet atmospheres. This is an invaluable input to more sophisticated 3D atmospheric models in the future.

    • Ultra-hot Jupiters are tidally locked with their host stars, dividing their atmospheres into a hot dayside and a colder nightside. As the planet moves through transit, different regions of the atmosphere rotate into view, revealing different chemical regimes. Highresolution spectrographs can observe asymmetries and velocity shifts and offer the possibility for time-resolved spectroscopy. The ultra-hot Jupiter WASP-189 b has recently been found to possess a rich transmission spectrum with evidence for atmospheric dynamics and chemical inhomogeneity. In this study, we search for other atoms and molecules in the planet’s transmission spectrum and investigate asymmetric signals. We analysed and combined eight transits of the ultra-hot Jupiter WASP-189 b collected with the HARPS, HARPS-N, ESPRESSO, and MAROON-X high-resolution spectrographs. Using the cross-correlation technique, we searched for neutral and ionised atoms as well as oxides, and we compared the obtained signals to model predictions. We report significant detections for H, Na, Mg, Ca, Ca+, Ti, Ti+, TiO, V, Cr, Mn, Fe, Fe+, Ni, Sr, Sr+, and Ba+. Of these, Sr, Sr+, and Ba+ are detected for the first time in the transmission spectrum of WASP-189 b. In addition, we robustly confirm the detection of titanium oxide based on observations with HARPS and HARPS-N using the follow-up observations performed with MAROON-X and ESPRESSO. By fitting the orbital traces of the detected species by means of time-resolved spectroscopy using a Bayesian framework, we inferred posterior distributions for orbital parameters as well as line shapes. Our results indicate that different species must originate from different regions of the atmosphere to be able to explain the observed time dependence of the signals. Throughout the course of the transit, most signal strengths are expected to increase due to the larger atmospheric scale height at the hotter trailing terminator. For some species, however, we instead observed that the signals weaken, either due to the ionisation of atoms and their ions or the dissociation of molecules on the dayside.

    • Context. WASP-121 b is a hot Jupiter that was recently found to possess rich emission (day side) and transmission (limb) spectra, suggestive of the presence of a multitude of chemical species in the atmosphere. Aims. We survey the transmission spectrum of WASP-121 b for line-absorption by metals and molecules at high spectral resolution and elaborate on existing interpretations of the optical transmission spectrum observed with the Hubble Space Telescope (HST). Methods. We applied the cross-correlation technique and direct differential spectroscopy to search for sodium and other neutral and ionised atoms, TiO, VO, and SH in high-resolution transit spectra obtained with the HARPS spectrograph. We injected models assuming chemical and hydrostatic equilibrium with a varying temperature and composition to enable model comparison, and employed two bootstrap methods to test the robustness of our detections. Results. We detect neutral Mg, Na, Ca, Cr, Fe, Ni, and V, which we predict exists in equilibrium with a significant quantity of VO, supporting earlier observations by HST/WFC3. Non-detections of Ti and TiO support the hypothesis that Ti is depleted via a cold-trap mechanism, as has been proposed in the literature. Atomic line depths are under-predicted by hydrostatic models by a factor of 1.5 to 8, confirming recent findings that the atmosphere is extended. We predict the existence of significant concentrations of gas-phase TiO2, VO2, and TiS, which could be important absorbers at optical and near-IR wavelengths in hot Jupiter atmospheres. However, accurate line-list data are not currently available for them. We find no evidence for absorption by SH and find that inflated atomic lines can plausibly explain the slope of the transmission spectrum observed in the near-ultraviolet with HST. The Na I D lines are significantly broadened (FWHM ∼50 to 70 km s-1) and show a difference in their respective depths of ~15 scale heights, which is not expected from isothermal hydrostatic theory. If this asymmetry is of astrophysical origin, it may indicate that Na I forms an optically thin envelope, reminiscent of the Na I cloud surrounding Jupiter, or that it is hydrodynamically outflowing.

  • A strong H-opacity signal in the near-infrared emission spectrum of the ultra-hot Jupiter KELT-9b

    • We present the analysis of a spectroscopic secondary eclipse of the hottest transiting exoplanet detected to date, KELT-9b, obtained with the Wide Field Camera 3 aboard the Hubble Space Telescope. We complement these data with literature information on stellar pulsations and Spitzer/Infrared Array Camera and Transiting Exoplanet Survey Satellite eclipse depths of this target to obtain a broadband thermal emission spectrum. Our extracted spectrum exhibits a clear turnoff at 1.4 μm. This points to H- bound-free opacities shaping the spectrum. To interpret the spectrum, we perform grid retrievals of self-consistent 1D equilibrium chemistry forward models, varying the composition and energy budget. The model with solar metallicity and C/O ratio provides a poor fit because the H- signal is stronger than expected, requiring an excess of electrons. This pushes our retrievals toward high atmospheric metallicities ([M/H] = 1.98-0.21+0.19) and a C/O ratio that is subsolar by 2.4σ. We question the viability of forming such a high-metallicity planet, and therefore provide other scenarios to increase the electron density in this atmosphere. We also look at an alternative model in which we quench TiO and VO. This fit results in an atmosphere with a slightly subsolar metallicity and subsolar C/O ratio ([M/H] = -0.22-0.13+0.17, log (C/O) = -0.34-0.34+0.19). However, the required TiO abundances are disputed by recent high-resolution measurements of the same planet.

    • When observing transmission spectra produced by the atmospheres of ultra-hot Jupiters (UHJs), large telescopes are typically the instrument of choice given the very weak signal of the planet’s atmopshere. The aim of the present study is to demonstrate that, for favourable targets, smaller telescopes are fully capable of conducting high-resolution cross-correlation spectroscopy. We apply the cross-correlation technique to data from the 2.1 m telescope at the Wendelstein Observatory, using its high-resolution spectrograph FOCES, in order to demonstrate its efficacy in resolving the atmosphere of the UHJ KELT-9 b. Using three nights of observations with the FOCES spectrograph and one with the HARPS-N spectrograph, we conduct a performance comparison between FOCES and HARPS-N. This comparison considers both single-transit and combined observations over the three nights. We then consider the potential of 2 m class telescopes by generalising our results to create a transit emulator capable of evaluating the potential of telescopes of this size. With FOCES, we detected seven species in the atmosphere of KELT-9 b: Ti II, Fe I, Fe II, Na I, Mg I, Na II, Cr II, and Sc II. Although HARPS-N surpasses FOCES in performance thanks to the mirror of the TNG, our results reveal that smaller telescope classes are capable of resolving the atmospheres of UHJs given sufficient observing time. This broadens the potential scope of such studies, demonstrating that smaller telescopes can be used to investigate phenomena such as temporal variations in atmospheric signals and the atmospheric loss characteristics of these close-in planets.

  • An atlas of resolved spectral features in the transmission spectrum of WASP-189 b with MAROON-X

    • Exoplanets in the ultra-hot Jupiter regime provide an excellent laboratory for testing the impact of stellar irradiation on the dynamics and chemical composition of gas giant atmospheres. In this study, we observed two transits of the ultra-hot Jupiter WASP-189 b with MAROON-X/Gemini-North to probe its high-altitude atmospheric layers, using strong absorption lines. We derived posterior probability distributions for the planetary and stellar parameters by calculating the stellar spectrum behind the planet at every orbital phase during the transit. This was used to correct the Rossiter- McLaughlin imprint on the transmission spectra. Using differential transmission spectroscopy, we detect strong absorption lines of Ca+, Ba+, Na, Hα, Mg, Fe, and Fe+, providing an unprecedented and detailed view of the atmospheric chemical composition. Ca+ absorption is particularly well suited for analysis through time-resolved narrow-band spectroscopy, owing to its transition lines formed in high-altitude layers. The spectral absorption lines show no significant blueshifts that would indicate high-altitude day-to-night winds, and further analysis is needed to investigate the implications for atmospheric dynamics. These high signal-to-noise observations provide a benchmark data set for testing high-resolution retrievals and the assumptions of atmospheric models. We also simulate observations of WASP-189 b with ANDES/ELT, and show that ANDES will be highly sensitive to the individual absorption lines of a myriad of elements and molecules, including TiO and CO.

    • Context. The population of strongly irradiated Jupiter-sized planets has no equivalent in the Solar System. It is characterised by strongly bloated atmospheres and large atmospheric scale heights. Recent space-based observations of SO2 photochemistry have demonstrated the knowledge that can be gained about Earth's uniqueness from detailed atmospheric studies of these unusual planets. Aims. Here we explore the atmosphere of WASP-172 b, a planet similar in terms of temperature and bloating to the recently studied HD 149026 b. We characterise the atmospheric composition and subsequently the atmospheric dynamics of this prime target. Methods. We observed a particular transit of WASP-172 b in front of its host star with ESO's ESPRESSO spectrograph and analysed the spectra obtained before, during, and after transit. Results. We detect the absorption of starlight by WASP-172 b's atmosphere by sodium (5.6 ) and hydrogen (19.5 ) and obtained a tentative detection of iron (4.1 ). We detect strong yet varying blueshifts, relative to the planetary rest frame, of all of these absorption features. This allows for a preliminary study of the atmospheric dynamics of WASP-172 b. Conclusions. With only one transit, we were able to detect a wide variety of species that clearly track different atmospheric layers with possible jets. WASP-172 b is a prime follow-up target for a more in-depth characterisation with both ground- and space-based observatories. If the detection of Fe is confirmed, this may suggest that radius inflation is an important determinant for the detectability of Fe in hot Jupiters, as several non-detections of Fe have been published for planets that are hotter but less inflated than WASP-172 b.

    • Planet formation processes or evolution mechanisms are surmised to be at the origin of the hot Neptune desert. Studying exoplanets currently living within or at the edge of this desert could allow disentangling the respective roles of formation and evolution. We present the High Accuracy Radial velocity Planet Searcher (HARPS) transmission spectrum of the bloated super-Neptune WASP-166b, located at the outer rim of the Neptune desert. Neutral sodium is detected at the 3.4σ level (0.455†±â€ 0.135%), with a tentative indication of line broadening, which could be caused by winds blowing sodium farther into space, a possible manifestation of the bloated character of these highly irradiated worlds. We put this detection into context with previous work claiming a non-detection of sodium in the same observations and show that the high noise in the trace of the discarded stellar sodium lines was responsible for the non-detection. We highlight the impact of this low signal-to-noise ratio remnant on detections for exoplanets similar to WASP-166b.

    • Chromatin changes within the context of DNA repair remain largely obscure. Here we show that DNA damage induces monoubiquitylation of histone H2A in the vicinity of DNA lesions. Ultraviolet (UV)-induced monoubiquitylation of H2A is dependent on functional nucleotide excision repair and occurs after incision of the damaged strand. The ubiquitin ligase Ring2 is required for the DNA damage-induced H2A ubiquitylation. UV-induced ubiquitylation of H2A is dependent on the DNA damage signaling kinase ATR (ATM- and Rad3-related) but not the related kinase ATM (ataxia telangiectasia-mutated). Although the response coincides with phosphorylation of variant histone H2AX, H2AX was not required for H2A ubiquitylation. Together our data show that monoubiquitylation of H2A forms part of the cellular response to UV damage and suggest a role of this modification in DNA repair-induced chromatin remodeling.

    • Context. The atmospheric and surface characterization of rocky planets is a key goal of exoplanet science. Unfortunately, the measurements required for this are generally out of reach of present-day instrumentation. However, the planet Mercury in our own solar system exhibits a large exosphere composed of atomic species that have been ejected from the planetary surface by the process of sputtering. Since the hottest rocky exoplanets known so far are more than an order of magnitude closer to their parent star than Mercury is to the Sun, the sputtering process and the resulting exospheres could be orders of magnitude larger and potentially detectable using transmission spectroscopy, indirectly probing their surface compositions. Aims. The aim of this work is to search for an absorption signal from exospheric sodium (Na) and singly ionized calcium (Ca+) in the optical transmission spectrum of the hot rocky super-Earth 55 Cancri e. Although the current best-fitting models to the planet mass and radius require a possible atmospheric component, uncertainties in the radius exist, making it possible that 55 Cancri e could be a hot rocky planet without an atmosphere. Methods. High resolution (R similar to 110 000) time-series spectra of five transits of 55 Cancri e, obtained with three different telescopes (UVES/VLT, HARPS/ESO 3.6 m and HARPS-N/TNG) were analysed. Targeting the sodium D lines and the calcium H and K lines, the potential planet exospheric signal was filtered out from the much stronger stellar and telluric signals, making use of the change of the radial component of the orbital velocity of the planet over the transit from -57 to +57 km s(-1). Results. Combining all five transit data sets, we detect a signal potentially associated with sodium in the planet exosphere at a statistical significance level of 3 sigma. Combining the four HARPS transits that cover the calcium H and K lines, we also find a potential signal from ionized calcium (4.1 sigma). Interestingly, this latter signal originates from just one of the transit measurements with a 4.9 sigma detection at this epoch.     • High-resolution precision spectroscopy provides a multitude of robust techniques for probing exoplanetary atmospheres. We present multiple VLT/ESPRESSO transit observations of the hot-Jupiter exoplanet WASP-19b with previously published but disputed atmospheric features from lowresolution studies. Through spectral synthesis and modelling of the Rossiter-McLaughlin (RM) effect we calculate stellar, orbital and physical parameters for the system. From narrow-band spectroscopy we do not detect any of HI, FeI, MgI, CaI, NaI, and KI neutral species, placing upper limits on their line contrasts. Through cross-correlation analyses with atmospheric models, we do not detect Fe I and place a 3σ upper limit of log (XFe/X⊙) ≈ -1.83 ± 0.11 on its mass fraction, from injection and retrieval. We show the inability to detect the presence of H2O for known abundances, owing to lack of strong absorption bands, as well as relatively low S/N ratio. We detect a barely significant peak (3.02±0.15 σ) in the cross-correlation map for TiO, consistent with the sub-solar abundance previously reported. This is merely a hint for the presence of TiO and does not constitute a confirmation. However, we do confirm the presence of previously observed enhanced scattering towards blue wavelengths, through chromatic RM measurements, pointing to a hazy atmosphere. We finally present a reanalysis of low-resolution transmission spectra of this exoplanet, concluding that unocculted starspots alone cannot explain previously detected features. Our reanalysis of the FORS2 spectra of WASP-19b finds a ~100× sub-solar TiO abundance, precisely constrained to log XTiO ≈ -7.52 ± 0.38, consistent with the TiO hint from ESPRESSO. We present plausible paths to reconciliation with other seemingly contradicting results.

    • Cross-correlation spectroscopy is an invaluable tool in the study of exoplanets. However, aliasing between spectral lines makes it vulnerable to systematic biases. This work strives to constrain the aliases of the cross-correlation function to provide increased confidence in the detections of elements in the atmospheres of ultra-hot Jupiters (UHJs) observed with high-resolution spectrographs. We use a combination of archival transit observations of the UHJ KELT-9 b obtained with the HARPS-N and CARMENES spectrographs and show that it is possible to leverage each instrument's strengths to produce robust detections at a substantially reduced signal-to-noise. Aliases that become present at low signal-to-noise regimes are constrained through a linear regression model. We confirm previous detections of H I, Na I, Mg I, Ca II, Sc II, Ti II, Cr II, Fe I, and Fe II, and detect eight new species, Ca I, Cr I, Ni I, Sr II, and Tb II, at the 5δ level, and Ti I, V I, and Ba II above the 3δ level. Ionised terbium (Tb II) has never before been seen in an exoplanet atmosphere. We further conclude that a 5δ threshold may not provide a reliable measure of confidence when used to claim detections, unless the systematics in the cross-correlation function caused by aliases are taken into account.

  • 3D non-LTE modeling of the stellar center-To-limb variation for transmission spectroscopy studies

    • Context. Transmission spectroscopy is one of the most powerful techniques used to characterize transiting exoplanets, since it allows for the abundance of the atomic and molecular species in the planetary atmosphere to be measured. However, stellar lines may bias the determination of such abundances if their center-To-limb variations (CLVs) are not properly accounted for. Aims. This paper aims to show that three-dimensional (3D) radiation hydrodynamic models and the assumption of non-local ther-modynamic equilibrium (non-LTE) line formation are required for an accurate modeling of the stellar CLV of the Na I D1 and K I resonance lines on transmission spectra. Methods. We modeled the CLV of the Na I D1 and K I resonance lines in the Sun with 3D non-LTE radiative transfer. The synthetic spectra were compared to solar observations with high spatial and spectral resolution, including new data collected with the CRISP instrument at the Swedish 1-m Solar Telescope between μ = 0.1 and μ = 1.0. Results. Our 3D non-LTE modeling of the Na I D1 resonance line at 5896 A and the K I 7699 A resonance line in the Sun is in good agreement with the observed CLV in the solar spectrum. Moreover, the simulated CLV curve for a Jupiter-Sun system inferred with a 3D non-LTE analysis shows significant differences from the one obtained from a 1D atmosphere. The latter does indeed tend to overestimate the amplitude of the transmission curve by a factor that is on the same order of magnitude as a planetary absorption depth (i.e., up to 0.2%). Conclusions. This work highlights the fact that to correctly characterize exoplanetary atmospheres, 3D non-LTE synthetic spectra ought to be used to estimate the stellar CLV effect in transmission spectra of solar-like planet hosts. Moreover, since different spectral lines show different CLV curves for the same geometry of the planet-star system, it is fundamental to model the CLV individually for each line of interest. The work will be extended to other lines and FGK-Type stars, allowing for synthetic high-resolution spectra to mitigate the stellar contamination of low-resolution planetary spectra, for example, those drawn from JWST.

    • Context. Determining the chemical properties of the atmosphere of young forming gas giants might shed light on the location their formation occurred and the mechanisms involved. Aims. Our aim was to detect molecules in the atmosphere of the young forming companion PDS70 b by searching for atmospheric absorption features typical of substellar objects. Methods. We obtained medium-resolution (R ≈ 5075) spectra of the PDS70 planetary system with the SINFONI integral field spectrograph at the Very Large Telescope. We applied molecular mapping, based on cross-correlation with synthetic spectra, to identify signatures of molecular species in the atmosphere of the planet. Results. Although the planet emission is clearly detected when resampling the data to lower resolution, no molecular species could be identified with the cross-correlation technique. We estimated upper limits on the abundances of H2O, CO, and CH4 (log(Xmol) < -4.0, - 4.1, and - 4.9, respectively) assuming a clear atmosphere, and we explored the impact of clouds, which increase the upper limits by a factor of up to 0.7 dex. Assuming that the observations directly probe the planet's atmosphere, we found a lack of molecular species compared to other directly imaged companions or field objects. Under the assumption that the planet atmosphere presents similar characteristics to other directly imaged planets, we conclude that a dusty environment surrounds the planet, effectively obscuring any feature generated in its atmosphere. We quantify the extinction necessary to impede the detection (AV ≈ 16-17 mag), pointing to the possibility of higher optical thickness than previously estimated from other studies. Finally, the non-detection of molecular species conflicts with atmospheric models previously proposed to describe the forming planet. Conclusions. To reveal how giant planets form a comprehensive approach that includes constraints from multiple techniques needs to be undertaken. Molecular mapping emerges as an alternative to more classical techniques like SED fitting. Specifically tuned atmospheric models are likely required to faithfully describe the atmospheres of forming protoplanets, and higher spectral resolution data may reveal molecular absorption lines despite the dusty environment enshrouding PDS70 b.

  • Replication Fork Stability in Mammalian Cells

    • Maintaining replication fork integrity is vital to preserve genomic stability and avoid cancer. Physical DNA damage and altered nucleotide or protein pools represent replication obstacles, generating replicative stress. Numerous cellular responses have evolved to ensure faithful DNA replication despite such challenges. Understanding those responses is essential to understand and prevent or treat replication-associated diseases, such as cancer. Re-priming is a mechanism to allow resumption of DNA synthesis past a fork-stalling lesion. This was recently suggested in yeast and explains the formation of gaps during DNA replication on damaged DNA. Using a combination of assays, we indicate the existence of re-priming also in human cells following UV irradiation. The gap left behind a re-primed fork must be stabilised to avoid replication-associated collapse. Our results show that the checkpoint signalling protein CHK1 is dispensable for stabilisation of replication forks after UV irradiation, despite its role in replication fork progression on UV-damaged DNA. It is not known what proteins are necessary for collapse of an unsealed gap or a stalled fork. We exclude one, previously suggested, endonuclease from this mechanism in UV-irradiated human fibroblasts. We also show that focus formation of repair protein RAD51 is not necessarily associated with cellular sensitivity to agents inducing replicative stress, in rad51d CHO mutant cells. Multiple factors are required for replication fork stability, also under unperturbed conditions. We identify the histone methyltransferase SET8 as an important player in the maintenance of replication fork stability. SET8 is required for replication fork progression, and depletion of SET8 led to the formation of replication-associated DNA damage. In summary, our results increase the knowledge about mechanisms and signalling at replication forks in unperturbed cells and after induction of replicative stress.

    • The atmospheres of ultra-hot Jupiters are highly interesting and unique chemical laboratories. Due to the very high atmospheric temperatures, their chemical composition is dominated by atoms and ions instead of molecules, and the formation of aerosols on their day-sides is unlikely. Thus, for these planets detailed chemical characterisations via the direct detection of elements through high-resolution day-side and transit spectroscopy are possible. This in principle allows the element abundances of these objects to be directly inferred, which may provide crucial constraints on their formation process and evolution history. In the recent past, several chemical species, mostly in the form of atoms and ions, have already been detected using high-resolution spectroscopy in combination with the cross-correlation technique. As part of the Mantis network, we provide a grid of standard templates in this study, designed to be used together with the cross-correlation method. This allows for the straightforward detection of chemical species in the atmospheres of hot extrasolar planets. In total, we calculate high-resolution templates for more than 140 different species across several atmospheric temperatures. In addition to the high-resolution templates, we also provide line masks that just include the position of line peaks and their absorption depths relative to the spectral continuum. A separate version of these line masks also takes potential blending effects with lines of other species into account.

    • Isotope abundance ratios have an important role in astronomy and planetary sciences, providing insights into the origin and evolution of the Solar System, interstellar chemistry and stellar nucleosynthesis1,2. In contrast to deuterium/hydrogen ratios, carbon isotope ratios are found to be roughly constant (around 89) in the Solar System1,3, but do vary on galactic scales with a 12C/13C isotopologue ratio of around 68 in the current local interstellar medium4–6. In molecular clouds and protoplanetary disks, 12CO/13CO ratios can be altered by ice and gas partitioning7, low-temperature isotopic ion-exchange reactions8 and isotope-selective photodissociation9. Here we report observations of 13CO in the atmosphere of the young, accreting super-Jupiter TYC 8998-760-1 b, at a statistical significance of more than six sigma. Marginalizing over the planet’s atmospheric temperature structure, chemical composition and spectral calibration uncertainties suggests a 12CO/13CO ratio of 31−10+17(90% confidence), a substantial enrichment in 13C with respect to the terrestrial standard and the local interstellar value. As the current location of TYC 8998-760-1 b at greater than or equal to 160 astronomical units is far beyond the CO snowline, we postulate that it accreted a substantial fraction of its carbon from ices enriched in 13C through fractionation.
