    • Aims: The aim of this study was to determine the incidence of neonatal morbidity in extremely preterm infants and to identify associated risk factors. Methods: Population based study of infants born before 27 gestational weeks and admitted for neonatal intensive care in Sweden during 2004-2007. Results: Of 638 admitted infants, 141 died. Among these, life support was withdrawn in 55 infants because of anticipation of poor long-term outcome. Of 497 surviving infants, 10% developed severe intraventricular haemorrhage (IVH), 5.7% cystic periventricular leucomalacia (cPVL), 41% septicaemia and 5.8% necrotizing enterocolitis (NEC); 61% had patent ductus arteriosus (PDA) and 34% developed retinopathy of prematurity (ROP) stage >= 3. Eighty-five per cent needed mechanical ventilation and 25% developed severe bronchopulmonary dysplasia (BPD). Forty-seven per cent survived to one year of age without any severe IVH, cPVL, severe ROP, severe BPD or NEC. Tocolysis increased and prolonged mechanical ventilation decreased the chances of survival without these morbidities. Maternal smoking and higher gestational duration were associated with lower risk of severe ROP, whereas PDA and poor growth increased this risk. Conclusion: Half of the infants surviving extremely preterm birth suffered from severe neonatal morbidities. Studies on how to reduce these morbidities and on the long-term health of survivors are warranted.

    • Objective: Intrapartum antibiotic prophylaxis (IAP) reduces the incidence of neonatal early onset group B streptococcal infections. The present study investigated if an automated PCR-assay, used bedside by the labor ward personnel was manageable and could decrease the use of IAP in a setting with a risk-based IAP strategy. Methods: The study comprises two phases. Phase 1 was a multicenter, randomized, controlled trial. Women with selected risk-factors were allocated either to PCR-IAP (prophylaxis given if positive or indeterminate) or IAP. A vaginal/rectal swab and superficial swabs from the neonate for conventional culture were also obtained. Phase 2 was non-randomized, assessing an improved version of the assay. Results: Phase 1 included 112 women in the PCR-IAP group and 117 in the IAP group. Excluding indeterminate results, the assay showed a sensitivity of 89% and a specificity of 90%. In 44 % of the PCR assays the result was indeterminate. The use of IAP was lower in the PCR group (53 versus 92%). Phase 2 included 94 women. The proportion of indeterminate results was reduced (15%). The GBS colonization rate was 31%. Conclusion: The PCR assay, in the hands of labor ward personnel, can be useful for selection of women to which IAP should be offered.

  • Angiomotin regulates endothelial cell migration during embryonic angiogenesis

    • The development of the embryonic vascular system into a highly ordered network requires precise control over the migration and branching of endothelial cells (ECs). We have previously identified angiomotin (Amot) as a receptor for the angiogenesis inhibitor angiostatin. Furthermore, DNA vaccination targeting Amot inhibits angiogenesis and tumor growth. However, little is known regarding the role of Amot in physiological angiogenesis. We therefore investigated the role of Amot in embryonic neovascularization during zebrafish and mouse embryogenesis. Here we report that knockdown of Amot in zebrafish reduced the number of filopodia of endothelial tip cells and severely impaired the migration of intersegmental vessels. We further show that 75% of Amot knockout mice die between embryonic day 11 (E11) and E11.5 and exhibit severe vascular insufficiency in the intersomitic region as well as dilated vessels in the brain. Furthermore, using ECs differentiated from embryonic stem (ES) cells, we demonstrate that Amot-deficient cells have intact response to vascular endothelial growth factor (VEGF) in regard to differentiation and proliferation. However, the chemotactic response to VEGF was abolished in Amot-deficient cells. We provide evidence that Amot is important for endothelial polarization during migration and that Amot controls Rac1 activity in endothelial and epithelial cells. Our data demonstrate a critical role for Amot during vascular patterning and endothelial polarization.

    • This article analyses how UN Security Council resolution 1325 and the Women, Peace, Security (WPS) agenda have been adopted in the Middle East and North Africa. The region faces huge security challenges triggered by recurrent armed conflicts, terrorism and wars both between and within states. Four thematic areas have been chosen: (1) terrorism; (2) peace negotiations; (3) state-building; and (4) democratization. The article uses empirical vignettes from Jordan, Syria, Yemen, Iraq and Tunisia to assess the normative framework of the WPS agenda and its National Action Plans. The article concludes by highlighting some of the core problems of implementation, such as the persistence of patriarchal societal values, institutionalized hypermasculinity in and among armed groups, and the number of unresolved conflicts in the region. It also underlines the problem of instrumentalizing the WPS agenda and the strategic essentialization of women in peacekeeping, peace-building and post-conflict reconstruction.

    • Resilience and gender have become new buzzwords for expressing renewal in peacebuilding. This article unpacks the gender-resilience nexus in theory and analyses global trends and variation in peacebuilding policy and practice. It advances an analytical framework based on three central pillars of peacebuilding: process, outcome, and expertise. A comprehensive analysis of 49 international peacebuilding handbooks, produced by leading international organisations for policymakers and practitioners in the field, is conducted. The results show how the integration of the gender-resilience nexus signals new ways of understanding conflict dynamics and peacebuilding. Yet, gender peace expertise is ‘thin’ in regards to policies and practices of resilient conflict transformation. By way of conclusion, we suggest three directions to be taken in research to advance and refine the gender-resilience nexus. First, the politics and contestation of peacebuilding need to be problematised and explored further. Second, the understanding of resilience in peacebuilding needs to shift emphasis from conflict management to conflict transformation. Third, the positionality of peacebuilding actors and local contexts need to be probed further.

  • Report of the Benchmark Workshop on Baltic Cod Stocks (WKBALTCOD)

    • The ICES Benchmark Workshop on Baltic Cod Stocks (WKBALTCOD), chaired by External Chair Jean-Jacques Maguire, Canada and ICES Chair Marie Storr-Paulsen, Denmark, and attended by two invited external experts Verena Trenkel, France and Meaghan Bryan, USA met in Rostock, Germany, 2–6 March 2015 with 39 participants and six countries represented. The objective of WKBALTCOD was to evaluate the appropriateness of data and methods to determine stock status and investigate meth-ods appropriate to use in the single-stock assessment for the cod stock in SD 22–24 and cod in SD 25–32 in the Baltic. Participants in the workshop were a large group with diverse backgrounds representing the industry, fisheries, NGOs, managers and scientists.The single-stock analytic assessment of the eastern Baltic stock was not accepted by the assessment working group (WGBFAS) in 2014 due to severe problems with the input data. The advice for the eastern Baltic cod was, therefore, based on the ICES approach for data-limited stocks. As an outcome ICES decided to establish a bench-mark for both cod stocks and to scope an integrated assessment for the Baltic cod stocks. The first meeting (WKSIBCA) was therefore meant to introduce the interces-sional work conducted since the assessment working group in April 2014, and to reach some conclusions on how to proceed both in the short term (Benchmark in March 2015) and longer term (2–3 years) and was seen as a data compilation work-shop, there is produced a separate report from this workshop. The WKBALTCOD was the 2nd meeting in the benchmark process and was intended to come up with a final stock assessment method, stock annex and input data for both stocks. As it was not possible to reach conclusive decision on the final model to be used for the east Baltic cod stock during the benchmark meeting and as more work on the preferable models was needed, it was decided by the ACOM leadership to prolong the bench-mark process until the assessment working group meeting in April 2015. This deci-sion has led to a relatively long process partly mixed with the assessment working group WGBFAS.It became clear during the benchmark process that although large effort has been put into explaining the underlying processes leading to the changes in the Baltic ecosys-tem, there is still some lack of understanding of the present situation in the eastern Baltic cod stock. Therefore, it was not possible to reach firm conclusions on the final model to be used and therefore not possible to set reference points. It was decided to continue to explore the most promising models and to continue to improve the input data until the assessment working group started in April.The main challenges still to be solved for the Eastern Baltic cod stock is the quantifi-cation of increased natural mortality and decrease in growth. Through several presentations during the workshop (both WKSIBCA and WKBALTCOD) it became clear that natural mortality very likely has increased in later years, due to decreased condition and increased parasite infection. A decrease in growth also seems plausible duo to a decrease in condition and/or selectivity-induced mortality of the largest in-dividuals. However, as none of these parameters are easily estimated, especially with the severe ageing problems, different model assumptions made the output very shaky.For the western Baltic cod, stock identification issues were examined in area SD 24, the intermediate area: based on otolith characteristics and genetics. Due to the results showing a large proportion of east cod in this area, it was decided to split the catch2 | ICES WKBALTCOD REPORT 2015and survey from SD 24 into either the western or eastern Baltic cod stock. It was pos-sible to derive proportions of eastern and western cod in SD 24 back to the mid-1990s.For the western Baltic cod stock a modelled survey indices was included in the as-sessment covering the western part of SD 24 and Area 22+23 and based on a smoothed ALK.Both cod stocks have in the past used commercial tuning fleet to have a better cov-ered of older age groups. It was decided to abound this time-series duo quality issues such as a limited coverage and problems with technical creeping.WKBALTCOD was not able to explore and define reference points for the Western Baltic cod stock during the meeting due to time constraints, but these were calculated and decided by correspondence after the meeting. The recent protocols on estimation procedures developed by WKMSYREF3 for stocks with a full analytical assessment and for data-limited stocks served as objective guidelines to obtain reference point estimates.

  • Research and development challenges for Swedish biofuel actors – three illustrative examples : Improvement potential discussed in the context of Well-to-Tank analyses

The EU has, for example, set a target for the use of renewable fuels in the transportation sector stating that all EU member states should use 10% renewable fuels for transport by 2020. Fulfilling this ambition will lead to an enormous market for biofuels during the coming decade. To avoid increasing production of biofuels based on agriculture crops that require considerable use of arable area, focus is now to move towards more advanced second generation (2G) biofuels that can be produced from biomass feedstocks associated with a more efficient land use. Climate benefits and greenhouse gas (GHG) balances are aspects often discussed in conjunction with sustainability and biofuels. The total GHG emissions associated with production and usage of biofuels depend on the entire fuel production chain, mainly the agriculture or forestry feedstock systems and the manufacturing process. To compare different biofuel production pathways it is essential to conduct an environmental assessment using the well-to-tank (WTT) analysis methodology. In Sweden the conditions for biomass production are favourable and we have promising second generation biofuels technologies that are currently in the demonstration phase. In this study we have chosen to focus on cellulose based ethanol, methane from gasification of solid wood as well as DME from gasification of black liquor, with the purpose of identifying research and development potentials that may result in improvements in the WTT emission values. The main objective of this study is thus to identify research and development challenges for Swedish biofuel actors based on literature studies as well as discussions with the the researchers themselves. We have also discussed improvement potentials for the agriculture and forestry part of the WTT chain. The aim of this study is to, in the context of WTT analyses, (i) increase knowledge about the complexity of biofuel production, (ii) identify and discuss improvement potentials, regarding energy efficiency and GHG emissions, for three biofuel production cases, as well as (iii) identify and discuss improvement potentials regarding biomass supply, including agriculture/forestry. The scope of the study is limited to discussing the technologies, system aspects and climate impacts associated with the production stage. Aspects such as the influence on biodiversity and other environmental and social parameters fall beyond the scope of this study. We find that improvement potentials for emissions reductions within the agriculture/forestry part of the WTT chain include changing the use of diesel to low-CO2-emitting fuels, changing to more fuel-efficient tractors, more efficient cultivation and manufacture of fertilizers (commercial nitrogen fertilizer can be produced in plants which have nitrous oxide gas cleaning) as well as improved fertilization strategies (more precise nitrogen application during the cropping season). Furthermore, the cultivation of annual feedstock crops could be avoided on land rich in carbon, such as peat soils and new agriculture systems could be introduced that lower the demand for ploughing and harrowing. Other options for improving the WTT emission values includes introducing new types of crops, such as wheat with higher content of starch or willow with a higher content of cellulose. From the case study on lignocellulosic ethanol we find that 2G ethanol, with co-production of biogas, electricity, heat and/or wood pellet, has a promising role to play in the development of sustainable biofuel production systems. Depending on available raw materials, heat sinks, demand for biogas as vehicle fuel and existing 1G ethanol plants suitable for integration, 2G ethanol production systems may be designed differently to optimize the economic conditions and maximize profitability. However, the complexity connected to the development of the most optimal production systems require improved knowledge and involvement of several actors from different competence areas, such as chemical and biochemical engineering, process design and integration and energy and environmental systems analysis, which may be a potential barrier.

Three important results from the lignocellulosic ethanol study are: (i) the production systems could be far more complex and intelligently designed than previous studies show, (ii) the potential improvements consist of a large number of combinations of process integration options wich partly depends on specific local conditions, (iii) the environmental performance of individual systems may vary significantly due to systems design and local conditons.From the case study on gasification of solid biomass for the production of biomethane we find that one of the main advantages of this technology is its high efficiency in respect to converting biomass into fuels for transport. For future research we see a need for improvements within the gas up-grading section, including gas cleaning and gas conditioning, to obtain a more efficient process. A major challenge is to remove the tar before the methanation reaction. Three important results from the biomethane study are: (i) it is important not to crack the methane already produced in the syngas, which indicates a need for improved catalysts for selective tar cracking, (ii) there is a need for new gas separation techniques to facilitate the use of air oxidation agent instead of oxygen in the gasifier, and (iii) there is a need for testing the integrated process under realistic conditions, both at atmospheric and pressurized conditions. From the case study on black liquor gasification for the production of DME we find that the process has many advantages compared to other biofuel production options, such as the fact that black liquor is already partially processed and exists in a pumpable, liquid form, and that the process is pressurised and tightly integrated with the pulp mill, which enhances fuel production efficiency. However, to achieve commercial status, some challenges still remain, such as demonstrating that materials and plant equipment meet the high availability required when scaling up to industrial size in the pulp mill, and also proving that the plant can operate according to calculated heat and material balances. Three important results from the DME study are: (i) that modern chemical pulp mills, having a potential surplus of energy, could become important suppliers of renewable fuels for transport, (ii) there is a need to demonstrate that renewable DME/methanol will be proven to function in large scale, and (iii) there is still potential for technology improvements and enhanced energy integration. Although quantitative improvement potentials are given in the three biofuel production cases, it is not obvious how these potentials would affect WTT values, since the biofuel production processes are complex and changing one parameter impacts other parameters. The improvement potentials are therefore discussed qualitatively. From the entire study we have come to agree on the following common conclusions: (i) research and development in Sweden within the three studied 2G biofuel production technologies is extensive, (ii) in general, the processes, within the three cases, work well at pilot and demonstration scale and are now in a phase to be proven in large scale, (iii) there is still room for improvement although some processes have been known for decades, (iv) the biofuel production processes are complex and site specific and process improvements need to be seen and judged from a broad systems perspective (both within the production plant as well as in the entire well-to-tank perspective), and (v) the three studied biofuel production systems are complementary technologies. Futher, the process of conducting this study is worth mentioning as a result itself, i.e. that many different actors within the field have proven their ability and willingness to contribute to a common report, and that the cooperation climate was very positive and bodes well for possible future collaboration within the framework of the f3 center. Finally, judging from the political ambitions it is clear that the demand for renewable fuels will significantly increase during the coming decade. This will most likely result in opportunities for a range of biofuel options. The studied biofuel options all represent 2G biofuels and they can all be part of the solution to meet the increased renewable fuel demand.

Furthermore, the cultivation of annual feedstock crops could be avoided on land rich in carbon, such as peat soils and new agriculture systems could be introduced that lower the demand for ploughing and harrowing. Other options for improving the WTT emission values includes introducing new types of crops, such as wheat with higher content of starch or willow with a higher content of cellulose.From the case study on lignocellulosic ethanol we find that 2G ethanol, with co-production of biogas, electricity, heat and/or wood pellet, has a promising role to play in the development of sustainable biofuel production systems. Depending on available raw materials, heat sinks, demand for biogas as vehicle fuel and existing 1G ethanol plants suitable for integration, 2G ethanol production systems may be designed differently to optimize the economic conditions and maximize profitability. However, the complexity connected to the development of the most optimal production systems require improved knowledge and involvement of several actors from different competence areas, such as chemical and biochemical engineering, process design and integration and energy and environmental systems analysis, which may be a potential barrier.Three important results from the lignocellulosic ethanol study are: (i) the production systems could be far more complex and intelligently designed than previous studies show, (ii) the potential improvements consist of a large number of combinations of process integration options wich partly depends on specific local conditions, (iii) the environmental performance of individual systems may vary significantly due to systems design and local conditons.From the case study on gasification of solid biomass for the production of biomethane we find that one of the main advantages of this technology is its high efficiency in respect to converting biomass into fuels for transport. For future research we see a need for improvements within the gas up-grading section, including gas cleaning and gas conditioning, to obtain a more efficient process. A major challenge is to remove the tar before the methanation reaction.Three important results from the biomethane study are: (i) it is important not to crack the methane already produced in the syngas, which indicates a need for improved catalysts for selective tar cracking, (ii) there is a need for new gas separation techniques to facilitate the use of air oxidation agent instead of oxygen in the gasifier, and (iii) there is a need for testing the integrated process under realistic conditions, both at atmospheric and pressurized conditions.From the case study on black liquor gasification for the production of DME we find that the process has many advantages compared to other biofuel production options, such as the fact that black liquor is already partially processed and exists in a pumpable, liquid form, and that the process is pressurised and tightly integrated with the pulp mill, which enhances fuel production efficiency. However, to achieve commercial status, some challenges still remain, such as demonstrating that materials and plant equipment meet the high availability required when scaling up to industrial size in the pulp mill, and also proving that the plant can operate according to calculated heat and material balances. Three important results from the DME study are: (i) that modern chemical pulp mills, having a potential surplus of energy, could become important suppliers of renewable fuels for transport, (ii) there is a need to demonstrate that renewable DME/methanol will be proven to function in large scale, and (iii) there is still potential for technology improvements and enhanced energy integration.Although quantitative improvement potentials are given in the three biofuel production cases, it is not obvious how these potentials would affect WTT values, since the biofuel production processes are complex and changing one parameter impacts other parameters. The improvement potentials are therefore discussed qualitatively. From the entire study we have come to agree on the following common conclusions: (i) research and development in Sweden within the three studied 2G biofuel production technologies is extensive, (ii) in general, the processes, within the three cases, work well at pilot and demonstration scale and are now in a phase to be proven in large scale, (iii) there is still room for improvement although some processes have been known for decades, (iv) the biofuel production processes are complex and site specific and process improvements need to be seen and judged from a broad systems perspective (both within the production plant as well as in the entire well-to-tank perspective), and (v) the three studied biofuel production systems are complementary technologies. Futher, the process of conducting this study is worth mentioning as a result itself, i.e. that many different actors within the field have proven their ability and willingness to contribute to a common report, and that the cooperation climate was very positive and bodes well for possible future collaboration within the framework of the f3 center.Finally, judging from the political ambitions it is clear that the demand for renewable fuels will significantly increase during the coming decade. This will most likely result in opportunities for a range of biofuel options. The studied biofuel options all represent 2G biofuels and they can all be part of the solution to meet the increased renewable fuel demand.

    • Human hepatocytes display substantial functional inter-individual variation regarding drug metabolizing functions. In order to investigate if this diversity is mirrored in hepatocytes derived from different human pluripotent stem cell (hPSC) lines, we evaluated 25 hPSC lines originating from 24 different donors for hepatic differentiation and functionality. Homogenous hepatocyte cultures could be derived from all hPSC lines using one standardized differentiation procedure. To the best of our knowledge this is the first report of a standardized hepatic differentiation procedure that is generally applicable across a large panel of hPSC lines without any adaptations to individual lines. Importantly, with regard to functional aspects, such as Cytochrome P450 activities, we observed that hepatocytes derived from different hPSC lines displayed inter-individual variation characteristic for primary hepatocytes obtained from different donors, while these activities were highly reproducible between repeated experiments using the same line. Taken together, these data demonstrate the emerging possibility to compile panels of hPSC-derived hepatocytes of particular phenotypes/genotypes relevant for drug metabolism and toxicity studies. Moreover, these findings are of significance for applications within the regenerative medicine field, since our stringent differentiation procedure allows the derivation of homogenous hepatocyte cultures from multiple donors which is a prerequisite for the realization of future personalized stem cell based therapies.

    • Background: Amyotrophic lateral sclerosis is a progressive neurodegenerative disorder leading to muscle weakness and respiratory failure. Arimoclomol, a heat-shock protein-70 (HSP70) co-inducer, is neuroprotective in animal models of amyotrophic lateral sclerosis, with multiple mechanisms of action, including clearance of protein aggregates, a pathological hallmark of sporadic and familial amyotrophic lateral sclerosis. We aimed to evaluate the safety and efficacy of arimoclomol in patients with amyotrophic lateral sclerosis.Methods: ORARIALS-01 was a multinational, randomised, double-blind, placebo-controlled, parallel-group trial done at 29 centres in 12 countries in Europe and North America. Patients were eligible if they were aged 18 years or older and met El Escorial criteria for clinically possible, probable, probable laboratory-supported, definite, or familial amyotrophic lateral sclerosis; had an ALS Functional Rating Scale-Revised score of 35 or more; and had slow vital capacity at 70% or more of the value predicted on the basis of the participant's age, height, and sex. Patients were randomly assigned (2:1) in blocks of 6, stratified by use of a stable dose of riluzole or no riluzole use, to receive oral arimoclomol citrate 1200 mg/day (400 mg three times per day) or placebo. The Randomisation sequence was computer generated centrally. Investigators, study personnel, and study participants were masked to treatment allocation. The primary outcome was the Combined Assessment of Function and Survival (CAFS) rank score over 76 weeks of treatment. The primary outcome and safety were analysed in the modified intention-to-treat population. This trial is registered with ClinicalTrials.gov, NCT03491462, and is completed.Findings: Between July 31, 2018, and July 17, 2019, 287 patients were screened, 245 of whom were enrolled in the trial and randomly assigned. The modified intention-to-treat population comprised 239 patients (160 in the arimoclomol group and 79 in the placebo group): 151 (63%) were male and 88 (37%) were female; mean age was 57·6 years (SD 10·9). CAFS score over 76 weeks did not differ between groups (mean 0·51 [SD 0·29] in the arimoclomol group vs 0·49 [0·28] in the placebo group; p=0·62). Cliff's delta comparing the two groups was 0·039 (95% CI –0·116 to 0·194). Proportions of participants who died were similar between the treatment groups: 29 (18%) of 160 patients in the arimoclomol group and 18 (23%) of 79 patients in the placebo group. Most deaths were due to disease progression. The most common adverse events were gastrointestinal. Adverse events were more often deemed treatment-related in the arimoclomol group (104 [65%]) than in the placebo group (41 [52%]) and more often led to treatment discontinuation in the arimoclomol group (26 [16%]) than in the placebo group (four [5%]).Interpretation: Arimoclomol did not improve efficacy outcomes compared with placebo. Although available biomarker data are insufficient to preclude future strategies that target the HSP response, safety data suggest that a higher dose of arimoclomol would not have been tolerated.Funding: Orphazyme.

  • Does employer involvement in primary health care enhance return to work for patients with stress-related mental disorders? a cluster randomized controlled trial

    • BackgroundStress-related disorders have become a major challenge for society and are associated with rising levels of sick leave. The provision of support to facilitate the return to work (RTW) for this patient group is of great importance. The aim of the present study was to evaluate whether a new systematic procedure with collaboration between general practitioners (GPs), rehabilitation coordinators (RCs) and employers could reduce sick leave days for this patient group.MethodEmployed patients with stress-related diagnoses seeking care at primary health care centres (PHCCs) were included in either the intervention group (n = 54), following the systematic intervention procedure, or the control group (n = 58), receiving treatment as usual (TAU). The intervention included a) a training day for participant GPs and RCs, b) a standardised procedure for GPs and RCs to follow after training, c) the opportunity to receive clinical advice from specialist physicians in the research group. Outcome measures for RTW were sick leave days.ResultsThe median number of registered gross sick leave days was lower for the control group at six, 12 and 24 months after inclusion, but the difference was not statistically significant. The control group had significantly fewer net sick leave days at three months (p = 0.03) at six months (p = 0.00) and at 12-months follow-up (p = 0.01). At 24 months, this difference was no longer significant.ConclusionsThe PRIMA intervention, which applied a standardized procedure for employer involvement in the rehabilitation process for patients with stress-related disorders, actually increased time to RTW compared to TAU. However, at 24 months, the benefit of TAU could no longer be confirmed. The study was registered on 16/01/2017 (ClinicalTrials.gov, NCT03022760).

    • This article pays interest to the intersection between policy and practice when implementing entrepreneurship in the educational system. Taking a point of departure in Mahieu’s (2006) call for knowledge of the interplay between different policy levels and Backström-Widjeskog’s (2010) conclusion about tensions occurring when teachers are introduced to the concept, the intention is to develop knowledge about conflicts and tensions at the intersection between policy and practice. From analysing policy documents and narratives from entrepreneurship education implementation projects during a time when entrepreneurship education started to be promoted in Sweden three figures of thought are found (economic/humanistic, biological/social, and individual/collective) which are proposed to be involved in creating tensions and conflicts in the intersection between policy and practice. Theoretically, these figures of thought can be seen as a contribution to understanding processes in which the concept of entrepreneurship education has deliberately been moved, by way of policy, to the educational practice. Reflecting on these thought figures may enhance teachers’ translation processes when starting to work with entrepreneurship education in practice.

    • The ICES/HELCOM Working Group on Integrated Assessments of the Baltic Sea(WGIAB) was established in 2007 as a forum for developing and combining ecosystembasedmanagement efforts for the Baltic Sea. The group intends to serve as a scientificcounterpart and support for the ICES Baltic Fisheries Assessment Working Group(WGBFAS) as well as for efforts and projects related to Integrated Ecosystem Assessments(IEA) within ICES and HELCOM. The group works in cooperation with similargroups within the ACOM/SCICOM Steering Group on Integrated Ecosystem Assessments(SSGIEA).The 2015 WGIAB meeting was held in Cádiz, Spain, from 9–13 March, back-to-backwith the meeting of its counterpart in the Working Group on Ecosystem Assessmentof Western European Shelf Seas (WGEAWESS). The meetings had joint sessions as wellas WG specific work, and some participants effectively participated in both meetings.The WGIAB meeting was attended by 27 participants from nine countries. The meetingwas chaired by Christian Möllmann, Germany, Laura Uusitalo, Finland and Lena Bergström,Sweden.This was the last year of the ongoing three-year Terms of Reference (ToR) for WGIAB.The main working activities in 2015 were to i) conduct studies on Baltic Sea ecosystemfunctioning with the goal to publish case studies from different parts of the Baltic Seain peer-reviewed journals, ii) work on the demonstration exercise to develop ecosystem-based assessment and advice for Baltic fish stocks focusing on cod (DEMO) withmultiple approaches, iii) plan further how to integrate the social and economic aspectsmore tightly in the WGIAB work, and iv) discuss the future focus and format of theWGIAB work.The Baltic ecosystem functioning activity focused on identifying and exploring keytrends and linkages in the Baltic Sea foodweb. This was pursued by presentation andfurther discussion of ongoing intersessional work on foodweb modelling and integratedanalyses, and by exercises to develop conceptual models Baltic Sea foodwebsand the links to ecosystem function. Long-term monitoring datasets on the abiotic andbiotic parts of the Baltic Sea Proper ecosystem were updated for use in the continuedwork to develop environmental indicators for fisheries and marine management.The focus of the DEMO 3 (DEMOnstration exercise for Integrated Ecosystem Assessmentand Advice of Baltic Sea cod) was on finding a way to use the results from theDEMO1 and DEMO2 workshops in short and midterm projections/scenarios of Balticcod dynamics based on different types of modelling, as well as designing methodologyand modelling data for practical implementation of Integrated Advice for Baltic cod.The WGIAB was positively inclined towards including social and economic aspectsinto the integrated assessment. Openings to this path were provided by presentationon ongoing project work, and discussing their linkages to ecological aspects. It wasseen as crucial that experts on social and economic analysis should be included andtake an active part in the future work of the group.The group concluded that its upcoming work should focus more closely on functionaldiversity, which was identified as a recurring issue in the Baltic Sea. This approach wasalso identified as a useful connection point between scientific and management aspectsin order for the group to continue serving as a forum for developing ecosystem-basedmanagement efforts in the Baltic Sea. A focus on functional diversity was also seen as2 | ICES WGIAB REPORT 2015a potentially feasible way of bringing together management aspects for different sectors,by linking to ecosystem services concepts.The group proposed Saskia Otto, Germany and Martin Lindegren, Denmark as newincoming Chairs, together with Lena Bergström, Sweden and Laura Uusitalo, Finland.Having four Chairs is justified due to the wide scope of the group's work, as well asthe increased work load due to the planned new foci.

    • In the setting of prior myocardial infarction, the oral antiplatelet ticagrelor added to aspirin reduced the risk of recurrent ischemic events, especially, in those with diabetes mellitus. Patients with stable coronary disease and diabetes are also at elevated risk and might benefit from dual antiplatelet therapy. The Effect of Ticagrelor on Health Outcomes in diabEtes Mellitus patients Intervention Study (THEMIS, NCT01991795) is a Phase 3b randomized, double-blinded, placebo-controlled trial of ticagrelor vs placebo, on top of low dose aspirin. Patients >= 50 years with type 2 diabetes receiving anti-diabetic medications for at least 6 months with stable coronary artery disease as determined by a history of previous percutaneous coronary intervention, bypass grafting, or angiographic stenosis of >= 50% of at least one coronary artery were enrolled. Patients with known prior myocardial infarction (MI) or stroke were excluded. The primary efficacy endpoint is a composite of cardiovascular death, myocardial infarction, or stroke. The primary safety endpoint is Thrombolysis in Myocardial Infarction major bleeding. A total of 19 220 patients worldwide have been randomized and at least 1385 adjudicated primary efficacy endpoint events are expected to be available for analysis, with an expected average follow-up of 40 months (maximum 58 months). Most of the exposure is on a 60 mg twice daily dose, as the dose was lowered from 90 mg twice daily partway into the study. The results may revise the boundaries of efficacy for dual antiplatelet therapy and whether it has a role outside acute coronary syndromes, prior myocardial infarction, or percutaneous coronary intervention.

    • Background: Propofol is known to interact with the γ-aminobutyric acidA (GABAA) receptor, however, activating the receptor alone is not sufficient for producing anaesthesia. Propofol tyresine phosphorylates the GABAA receptor and reorganises the actin cytoskeleton, eausing ring structures and rnembrane ruffles. Propofol, but not GABA, the endogenous tigand for the GABAA receptor, tyresine phosphorylates actin, both in the membrane and cytoskeletal fractions of the neuron.Aim: How does propofol cause the actin reorganisation and is this a specific effect of propofol? Is the small membrane associated G-protein rho involved in the signal cascade towards the actin reorganisation?Methods: Westem blotting (WB) was used to visualize tyresine phosphorylated immunoprecipitated proteins and changes in actin between the different cellularcompartments after inhibition with rho (C3 exotoxin) and rho kinase (ROK) (HA-1077) inhibitors. Fluoreseenee mireoscopy after rhodamine-phalloidin labelling of actin was used to calculate the number of actin ring structures caused by propofol or GABA, in same experiments combined with pre-incubation with C3 exotoxin, HA- 1077 or the tyrosine kinase inhibitor Herbimycin A. Propofol-stimulated cells were studied with confocal microscopy.Results: Propofol eaused an increased tyresine phosphorylation, that was reduced by C3 exotoxin, of a 160 kDa protein after two minutes stimulation. The 160 kDa protein is still unidentified. The actin ring structures caused by propofol was shown with confocal microscopy to go almost through the entire cell. The amount of rings were reduced by C3 exatoxin as well as HA-1077. Furthermore, w hen a tyrosine kinase bioeker was used no ring structures were formed. However, GABA did not produce any ring structures. When the actin content of the cellular campartments were analysed, C3 exatoxin treated cells showed an increased amount of actin in the cytoskeletal fraction, simultaneausly with a decrease in both the membrane and the cytosol fractions. The ROK bioeker on ly eaused a reduction of actin in the cytosol/membrane fractions, but no increase was observed in the cytoskeleton.Conclusion: Propofol, but not GABA, eauses actin ring structures in neurons. Propofol uses the rho and rho kinase pathway to reorganize the actin cytoskeleton into ring structures, which is also dependent on a tyresine klnase. Propofol also eauses an unidentified rho dependent 160 kDa protein to be tyresine phosphorylated. The activation eaused by propofol of rho and rho kinase causes actin to be moved from the cytoskeleton to the cell membrane and cytosol. This reorganisation of actin might influence the GABAA receptor by keeping it open, thus allowing the cell to be hyperpolarized for longer time, and consequently maintain anaesthesia.

  • Speleothems from Warm Climates : Holocene Records from the Caribbean and Mediterranean Regions

    • This thesis contributes to increased knowledge on Holocene climate and environmental variability from two complex and sparsely studied areas. Using a speleothem from Gasparee Cave, Trinidad, as a paleoclimate archive, the local expression of the 8.2 ka (thousand years before 1950) climate event and associated patterns of the inter-tropical convergence zone (ITCZ) and rainfall is provided. Subsequent speleothem studies using multi-proxy analysis of stalagmites from Kapsia Cave and Alepotrypa Cave, Greece, provide records of climate, vegetation and human induced changes in the cave environment during parts of the Holocene.The speleothems from the well-studied Neolithic habitation site, Alepotrypa Cave, have produced a climate and habitation record which covers the period of 6.3-1.0 ka. The cave was inhabited between 8.0-5.2 ka and was closed by a tectonic event, which has preserved the settlement. The stable oxygen record shows the first well-dated and robust expression of the 4.2 ka dry event in the Peloponnese, places the timing of the 3.2 ka dry event within an ongoing dry period, and shows a final dry event at 1.6 ka. The North Atlantic as well as more regional drivers, such as the North Sea Caspian Pattern Index is proposed to, in a complex interplay, govern many of the climate trends and events observed.Trace element variation after the site is abandoned indicate what is interpreted as two volcanic eruptions, the Minoan eruption of Thera (Santorini) around 3.6 ka and the 2.7 ka eruption of Somma (Vesuvius). Variations in trace elements during the habitation period show clear human influence, indicating an association with specific cave activities. One of the most interesting prospects for continued work on Alepotrypa Cave is this successful marriage of speleothem studies and archeology. A framework of dates which constrain some behavior of people living in the cave is only the beginning, and there is great potential to continue finding new clues in the speleothem data.

    • The eastern Baltic (EB) cod (Gadus morhua) stock was depleted and overexploited for decades until the mid-2000s, when fishing mortality rapidly declined and biomass started to increase, as shown by stock assessments. These positive developments were partly assigned to effective management measures, and the EB cod was considered one of the most successful stock recoveries in recent times. In contrast to this optimistic view, the analytical stock assessment failed in 2014, leaving the present stock status unclear. Deteriorated quality of some basic input data for stock assessment in combination with changes in environmental and ecological conditions has led to an unusual situation for cod in the Baltic Sea, which poses new challenges for stock assessment and management advice. A number of adverse developments such as low nutritional condition and disappearance of larger individuals indicate that the stock is in distress. In this study, we (i) summarize the knowledge of recent changes in cod biology and ecosystem conditions, (ii) describe the subsequent challenges for stock assessment, and (iii) highlight the key questions where answers are urgently needed to understand the present stock status and provide scientifically solid support for cod management in the Baltic Sea.

    • Besides traditional timber production, other forest functions, such as biodiversity and recreation, have gained increasing importance during the last few decades. Demands on forests have become more diversified, thus making forest management and planning more complex. To meet these challenges, there is a growing interest in a more diversified silviculture, for which a number of different management options are available. However, it remains unclear how the various management options affect economic, ecological, and social aspects of sustainable forest management. Hence, in this study, we assess the consequences of various management options on different aspects of sustainable forest management through scenario analysis using a forestry decision support system. We evaluate 10 different forest management scenarios for two contrasting municipalities in Sweden, based on expert participation by way of a web-based multi-criteria decision analysis framework. We asked experts in economic, ecological, and social forest values, as well as those in reindeer husbandry, to weigh a number of indicators in their field of expertise against each other, and to create value functions for each indicator. We then determined scenario ranking for different sets of weights for economic, ecological and social forest values. Our results indicate that current management practices are favorable for economic aspects (wood production), while a number of scenarios would be better suited to fulfill the Swedish co-equal forest policy goal of production and consideration of environmental issues, such as scenarios with longer rotation periods, a larger share of set-asides and a higher share of continuous cover forestry. These measures would be beneficial not only for ecological values, but also for social values and for reindeer husbandry. Furthermore, we found that expert participation through the web-tool was a promising alternative to physical meetings that require more commitment in terms of time and resources.

    • The present day distribution of Miombo savanna-woodland in Mozambique has been attributed to an expansion due to the clearing of original coastal forests through agriculture and use of fire. Here, we test this hypothesis using palaeoecological data from Lake Nhauhache, situated in the Vilankulo region. Our analysis shows that Brachystegia, one of the main constituents of the Miombo, has varied over time, and its variability seems to be driven by hydrological changes related to climatic variability rather than by land-use changes. The analyses show that Brachystegia was most common during ad 200-700 when a marshy forest/shrub community was dominant. After ad 700, this community changes to a dominance of Syzygium and Fagara linked to gradually rising water levels. Brachystegia remains in low abundance and fluctuating over time. From ad 1000, a general decline in trees/shrubs in favour of grasses concurs with an increase in grass pollen (possibly cereal) and charcoal, most probably as a result of farming activities. The decline in tree taxa was probably exacerbated by periodic droughts after c. ad 1200 as indicated by the diatom assemblage. In the period ad 1700 to late 1800, arboreal pollen is well represented, and this is concurrent with the diatom record suggesting high lake levels.

    • The long-term responses of vegetation to climate variability are of relevance for predicting present and future vegetation change, and have implications for the management of savanna and riparian ecosystems. This paper explores the links between regional rainfall, hydrology and vegetation dynamics in the savannas and riverine forests of the lower Limpopo Valley, southern Africa, from 800 AD to the present, reviewing palaeoecological data (fossil pollen, spores, diatoms and lithology) from several hydrological systems in Kruger National Park (KNP), South Africa and Limpopo National Park (PNL), Mozambique. The PNL-KNP records show that riverine arboreal taxa expanded during wetter periods, including 800-1400 AD and after 1800 AD. Between 1400 and 1800 AD, grasses, savanna taxa and generalist taxa were favored over riparian taxa, a change that is linked with the onset of dry spells in the region (corresponding to the so-called Little Ice Age). The most extreme drought events around 1700 AD resulted in a marked decline of riparian forest taxa near Lake Mapimbi, KNP. In contrast, many water-scarce sequences away from the riverine environment, such as Radio Pan, Mafayeni Pan, Malahlapanga Pan and Lake Makwadzi show stable grassland vegetation throughout the last 1200 years. The results demonstrate the resilience of the grassland-savanna ecosystems to projected climate change with warmer and overall drier climate. The riverine forests are predicted to be more vulnerable especially as more extreme weather events are projected.

    • The eastern Mediterranean, with its long archaeological and historical records, provides a unique opportunity to study human responses to climate variability. We review paleoclimate data and reconstructions from the region with a focus on the last 6000 years. We aim to provide an up-to-date source of information on climate variability and to outline present limitations and future opportunities. The review work is threefold: (1) literature review, (2) spatial and temporal analysis of proxy records, and (3) statistical estimation of uncertainties in present paleoclimate reconstructions (temperature, C). On a regional scale the review reveals a wetter situation from 6000 to 5400 yrs BP (note: all ages in this paper are in calibrated years before present (i.e. before 1950), abbreviated yrs BP, unless otherwise stated). This is followed by a less wet period leading up to one of fully-developed aridity from c. 4600 yrs BP. There is a need for more high-resolution paleoclimate records, in order to (i) better understand regional patterns and trends versus local climate variability and to (ii) fill the gap of data from some regions, such as the Near East, Greece and Egypt. Further, we evaluate the regional occurrence of a proposed widespread climate event at 4200 yrs BP. This proposed climate anomaly has been used to explain profound changes in human societies at different locations in the region around this time. We suggest that although aridity was widespread around 4200 yrs BP in the eastern Mediterranean region, there is not enough evidence to support the notion of a climate event with rapidly drying conditions in this region.

  • Climate in the eastern Mediterranean during the Holocene and beyond – A Peloponnesian perspective

    • This thesis contributes increased knowledge about climate variability during the late Quaternary in the eastern Mediterranean. Results from a paleoclimate review reveal that regional wetter conditions from 6000 to 5400 years BP were replaced by a less wet period from 5400 to 4600 years BP and to fully arid conditions around 4600 years BP. The data available, however, show that there is not enough evidence to support the notion of a widespread climate event with rapidly drying conditions in the region around 4200 years ago. The review further highlights the lack of paleoclimate data from the archaeologically rich Peloponnese Peninsula. This gap is addressed in this thesis by the provision of new paleoclimate records from the Peloponnese. One stalagmite from Kapsia Cave and two stalagmites from Glyfada Cave were dated and analyzed for stable oxygen (δ18O) and carbon (δ13C) isotopes. The Glyfada record covers a period from ~78 ka to ~37 ka and shows that the climate in this region responded rapidly to changes in temperatures over Greenland. During Greenland stadial (interstadial) conditions colder (warmer) and drier (wetter) conditions are reflected by depleted (enriched) δ13C-values in the speleothems. The Kapsia record covers a period from ~2900 to ~1100 years BP. A comparison between the modern stalagmite top isotopes and meteorological data shows that a main control on stalagmite δ18O is wet season precipitation amount. The δ18O record from Kapsia indicates cyclical humidity changes of close to 500 years, with rapid shifts toward wetter conditions followed by slowly developing aridity. Superimposed on this signal is a centennial signal of precipitation variability. A second speleothem from Kapsia with multiple horizons of fine sediments from past flood events intercalated with the calcite is used to develop a new, quick and non-destructive method for tracing flood events in speleothems by analyzing a thick section with an XRF core scanner.

    • This paper offers new high-resolution oxygen and carbon isotope data from Stalagmite S1 from Mavri Trypa Cave, SW Peloponnese. Our data provide the climate background to the destruction of the nearby Mycenaean Palace of Nestor at Pylos at the transition from Late Helladic (LH) IIIB to LH IIIC, similar to 3150-3130 years before present (before AD 1950, hereafter yrs BP) and the subsequent period. S1 is dated by 24 U-Th dates with an averaged precision of +/- 26 yrs (2s), providing one of the most robust paleoclimate records from the eastern Mediterranean for the end of the Late Bronze Age (LBA). The delta O-18 record shows generally wetter conditions at the time when the Palace of Nestor at Pylos was destroyed, but a brief period of drier conditions around 3200 yrs BP may have disrupted the Mycenaean agricultural system that at the time was likely operating close to its limit. Gradually developing aridity after 3150 yrs BP, i.e. subsequent to the destruction, probably reduced crop yields and helped to erode the basis for the reinstitution of a central authority and the Palace itself.

    • We present a speleothem based stable isotope (δ13C, δ18O) record from southern Greece covering a period from 79±5.8 ka to 37±3 ka, i.e. the end of Marine Isotope Stage (MIS) 5a and large parts of MIS 4 and MIS 3. The record from Glyfada Cave provides a U-Th dated proxy paleoclimate record from Greece, covering this time period, and shows that the climate over the Peloponnese rapidly responded to interstadial and stadial conditions over Greenland. During stadial (interstadial) conditions colder (warmer) and drier (wetter) conditions are reflected by depleted (enriched) δ13C-values in the speleothems from Glyfada Cave. Depositional hiatuses in Glyfada Cave correspond to periods of severe cold conditions in the northern Hemisphere and reduced precipitation over the Peloponnese most likely due to a southward displacement of Mediterranean cyclone tracks due to expanding northern ice sheets and increased snow cover over the European continent. By comparing our independently dated record with previously published pollen studies from Greece a time lag between the records from Ioannina and Megali Limni and Glyfada is evident. The match in time between the Glyfada speleothem record and the Tenaghi Philippon pollen record, when tuned to NGRIP, is rather precise.

    • We present stable isotope data (delta O-18, delta C-13) from a detrital rich stalagmite from Kapsia Cave, the Peloponnese, Greece. The cave is rich in archeological remains and there are reasons to believe that flooding of the cave has directly affected humans using the cave. Using a combination of U-Th and C-14 dating to constrain a site-specific correction factor for (Th-232/U-238) detrital molar ratio, a linear age model was constructed. The age model shows that the stalagmite grew during the period from ca. 950 BC to ca. AD 830. The stable oxygen record from Kapsia indicates cyclical changes of close to 500 yr in precipitation amount, with rapid shifts towards wetter conditions followed by slowly developing aridity. Superimposed on this signal, wetter conditions are inferred around 850, 700, 500 and 400-100 BC, and around AD 160-300 and AD 770; and driest conditions are inferred to have occurred around 450 BC, AD 100-150 and AD 650. Detrital horizons in the stalagmite indicate that three major floods took place in the cave at 500 BC, 70 BC and AD 450. The stable carbon isotope record reflects changes in biological activity being a result of both climate and human activities. (c) 2014 University of Washington.

    • The work was divided into two different series of light gas oil (LLGO) and heavy gas oil (HGO). The rapeseed oil was blended with the gas oils in the following concentrations: 10%, 20% and 30%. The pressure was maintained constant. The flow rate, temperature were varied. It is possible to blend rape seed oil into HGO and LLGO in the production of low sulfur diesel fuel. The conversion of the rapeseed oil to hydrocarbon was higher when blended into LLGO compared to HGO. Low reaction temperatures and high flow rates give less amount of conversion of the rape seed oil.

    • Histone H1 is an important constituent of chromatin which undergoes major structural rearrangements during mitosis. However, the role of H1, multiple H1 subtypes and H1 phosphorylation is still unclear. In normal human fibroblasts, phosphorylated H1 was found located in nuclei during prophase and in both cytoplasm and condensed chromosomes during metaphase, anaphase and telophase as detected by immunocytochemistry. Moreover, we detected remarkable differences in the distribution of the histone H1 subtypes H1.2, H1.3 and H1.5 during mitosis. H1.2 was found in chromatin during prophase, and almost solely in the cytoplasm of metaphase and early anaphase cells. In late anaphase it appeared in both chromatin and cytoplasm, and again in chromatin during telophase. H1.5 distribution pattern resembled that of H1.2, but some H1.5 remained situated in chromatin during metaphase and early anaphase. H1.3 was detected in chromatin in all cell cycle phases. We propose therefore, that H1 subtype translocation during mitosis is controlled by phosphorylation, in combination with H1 subtype inherent affinity. We conclude that H1 subtypes, or their phosphorylated variants, may be signalling molecules in mitosis or that they leave chromatin in a regulated way to give access for chromatin condensing factors or transcriptional regulators during mitosis.

    • Tumour-loaded dendritic cells (DCs) from patients with chronic lymphocytic leukaemia (CLL) matured using an α-type 1-polarized DC cocktail (IL-1β/TNF-α/IFN-α/IFN-γ/poly-I:C;αDC1) were recently shown to induce more functional CD8+ T cells against autologous tumour cells in vitro than DCs matured with the ‘standard’ cocktail (IL-1β/TNF-α/IL-6/PGE2;PGE2DCs). However, the ability of vaccine DCs to induce a type 1-polarized immune response in vivo probably relies on additional features, including their ability to induce a CXCR3-dependent recruitment of NK cells into vaccine-draining lymph nodes. Moreover, their guiding of rare tumour-specific CD8+ T cells to sites of DC–CD4+ T cell interactions by secretion of CCL3 and CCL4 is needed. We therefore analysed the chemokine profile and the lymphocyte-attracting ability in vitro of monocyte-derived PGE2DCs and αDC1s from patients with CLL. αDC1s produced much higher levels of CXCR3 ligands (CXCL9/CXCL10/CXCL11) than PGE2DCs. Functional studies further demonstrated that αDC1s were superior recruiters of both NK and NKT cells. Moreover, αDC1s produced higher levels of CCL3/CCL4 upon CD40 ligation. These findings suggest that functional αDC1s, derived from patients with CLL, produce a desirable NK-, NKT- and CD8+ T cell-attracting chemokine profile which may favour a guided and Th1-deviated priming of CD8+ T cells, supporting the idea that αDC1-based vaccines have a higher immunotherapeutic potential than PGE2DCs.
