  • Switched enantiopreference of Humicola lipase for 2-phenoxyalkanoic acid ester homologs can be rationalized by different substrate binding modes
  • 1999
  • Ingår i: Tetrahedron. - 0957-4166 .- 1362-511X. ; 10:21, s. 4191-4202
  • Tidskriftsartikel (refereegranskat)abstract
    • Humicola lanuginosa lipase was used for enantioselective hydrolyses of a series of homologous 2-phenoxyalkanoic acid ethyl esters. The enantioselectivity (E-value) of the enzyme changed from an (R)-enantiomer preference for the smallest substrate, 2-phenoxypropanoic acid ester, to an (S)-enantiomer preference for the homologous esters with longer acyl moieties. The E-values span the range from E=13 (R) to E=56 (S). A molecular modeling study identified two different substrate-binding modes for each enantiomer. We found that the enantiomers favored different modes. This discovery provided a model that offered a rational explanation for the observed switch in enantioselectivity. • Lipases from Rhizomucor miehei and Humicola lanuginosa : Modification of the lid covering the active site alters enantioselectivity
  • 1993
  • Ingår i: Journal of Protein Chemistry. - 0277-8033. ; 12:6, s. 749-757
  • Tidskriftsartikel (refereegranskat)abstract
    • The homologous lipases from Rhizomucor miehei and Humicola lanuginosa showed approximately the same enantioselectivity when 2-methyldecanoic acid esters were used as substrates. Both lipases preferentially hydrolyzed the S- enantiomer of 1-heptyl 2-methyldecanoate (R. miehei: E(S) = 8.5; H. lanuginosa: E(S) = 10.5), but the R-enantiomer of phenyl 2-methyldecanoate (E(R) = 2.9). Chemical arginine specific modification of the R. miehei lipase with 1,2-cyclohexanedione resulted in a decreased enantioselectivity (E(R) = 2.0), only when the phenyl ester was used as a substrate. In contrast, treatment with phenylglyoxal showed a decreased enantioselectivity (E(S) = 2.5) only when the heptyl ester was used as a substrate. The presence of guanidine, an arginine side chain analog, decreased the enantioselectivity with the heptyl ester (E(S) = 1.9) and increased the enantioselectivity with the aromatic ester (E(R) = 4.4) as substrates. The mutation, Glu 87 Ala, in the lid of the H. lanuginosa lipase, which might decrease the electrostatic stabilization of the open-lid conformation of the lipase, resulted in 47% activity compared to the native lipase, in a tributyrin assay. The Glu 87 Ala mutant showed an increased enantioselectivity with the heptyl ester (E(S) = 17.4) and a decreased enantioselectivity with the phenyl ester (E(R) = 2.5) as substrates, compared to native lipase. The enantioselectivities of both lipases in the esterification of 2-methyldecanoic acid with 1-heptanol were unaffected by the lid modifications.

  • 2-Methylalkanoic acids resolved by esterification catalysed by lipase from Candida rugosa : Alcohol chain length and enantioselectivity
  • 1993
  • Ingår i: Tetrahedron Asymmetry. - 0957-4166. ; 4:8, s. 1869-1878
  • Tidskriftsartikel (refereegranskat)abstract
    • Enantiomerically pure (R)-2-methyldecanoic acid and (S)-2-methyl-1-decanol were prepared in a multi gram scale by esterification reactions catalysed by lipase from Candida rugosa. The enantiomeric ratios (E-values) were determined as a function of the chain length of the alcohol used as the complementary substrate in cyclohexane. In the resolution of 2-methyldecanoic acid the highest value (E = 37 ± 5) was obtained, when either 2-hexanol, 1-heptanol or 1-octanol were used. In contrast, when resolving 2-methyloctanoic acid, the E-values increased continually with increasing chain length of the alcohol used. 1-Hexadecanol gave the highest value: E > 100. The E-values were determined from the enantiomeric excess (ee) of the product at a conversion below 0.4. After two consecutive esterification reactions enantiomerically pure (R)-2-methyldecanoic acid, >99.8% ee, and after subsequent reduction of the ester produced, (S)-2-methyl-1-decanol, 96.7% ee, were obtained.

  • Reversed enantiopreference of Candida rugosa lipase supports different modes of binding enantiomers of a chiral acyl donor
  • 1998
  • Ingår i: Journal of Molecular Catalysis - B Enzymatic. - 1381-1177 .- 1873-3158. ; 5:1-4, s. 283-287
  • Tidskriftsartikel (refereegranskat)abstract
    • Molecular modelling identifies two different productive modes of binding the enantiomers of a 2-methyldecanoic acid ester to the active site of Candida rugosa lipase (CRL). The fast reacting S-enantiomer occupies the previously identified acyl-binding tunnel of the enzyme, whereas the R- enantiomer leaves the tunnel empty. The modelling suggested that if both enantiomers were forced to bind to the active site leaving the tunnel empty, the enzyme would reverse its enantiopreference to become R-enantioselective. To test this hypothesis, we designed a structural analogue to 2- methyldecanoic acid, 2-methyl-6-(2-thienyl)hexanoic acid, which was expected to be too bulky to fit its acyl moiety into the acyl-binding tunnel. The CRL- catalysed hydrolysis of the ethyl ester of this substrate resulted in the preferential conversion of the R-enantiomer as predicted by molecular modelling. This represents the first kinetic evidence supporting the existence of two different modes of binding the enantiomers of a 2- methyldecanoic acid ester to the active site of CRL. We have shown that a rational 3D based approach in combination with substrate engineering can be used to predict and control the stereochemical outcome of a lipase catalysed reaction.

  • Monoclonal antibodies selective for α-synuclein oligomers/protofibrils recognize brain pathology in Lewy body disorders and α-synuclein transgenic mice with the disease-causing A30P mutation
  • 2013
  • Ingår i: Journal of Neurochemistry. - : Wiley-Blackwell. - 0022-3042 .- 1471-4159. ; 126:1, s. 131-144
  • Tidskriftsartikel (refereegranskat)abstract
    • Inclusions of intraneuronal alpha-synuclein (-synuclein) can be detected in brains of patients with Parkinson's disease and dementia with Lewy bodies. The aggregation of -synuclein is a central feature of the disease pathogenesis. Among the different -synuclein species, large oligomers/protofibrils have particular neurotoxic properties and should therefore be suitable as both therapeutic and diagnostic targets. Two monoclonal antibodies, mAb38F and mAb38E2, with high affinity and strong selectivity for large -synuclein oligomers were generated. These antibodies, which do not bind amyloid-beta or tau, recognize Lewy body pathology in brains from patients with Parkinson's disease and dementia with Lewy bodies and detect pathology earlier in -synuclein transgenic mice than linear epitope antibodies. An oligomer-selective sandwich ELISA, based on mAb38F, was set up to analyze brain extracts of the transgenic mice. The overall levels of -synuclein oligomers/protofibrils were found to increase with age in these mice, although the levels displayed a large interindividual variation. Upon subcellular fractionation, higher levels of -synuclein oligomers/protofibrils could be detected in the endoplasmic reticulum around the age when behavioral disturbances develop. In summary, our novel oligomer-selective -synuclein antibodies recognize relevant pathology and should be important tools to further explore the pathogenic mechanisms in Lewy body disorders. Moreover, they could be potential candidates both for immunotherapy and as reagents in an assay to assess a potential disease biomarker.

  • 2008

  • 2020
  • Ingår i: International Journal of Qualitative Studies on Health and Well-being. - Abingdon : Taylor & Francis. - 1748-2623 .- 1748-2631. ; 15:sup1
  • Tidskriftsartikel (refereegranskat)abstract
    • Purpose: This is a study on a sport-based intervention, with a focus on physical activity, social relations, and learning, to promote health and well-being in the workplace lived space. Lived space is situated and associated with social and cultural conventions which affect the quality of the perceived space at work. The aim of the paper is to elucidate the participant’s experiences of the intervention and how health and well-being were affected. Methods: The intervention was conducted with employees from the warehouse of a company within the retail sector. The design consisted of one initial workshop as a baseline, a sport-based intervention, three group interviews, and a final workshop. A hermeneutic phenomenological analysis focused on experiences of the intervention and the meaning of the workplace as the lived space. Results: Three themes emerged in the analysis; Expressing positive individual effects, Expressing improved work environment and The meaning of the workplace as lived space. The themes are discussed in relation to three basic health foci: physical activity, social relations and learning. Conclusion: The workplace as a lived space offers a valuable opportunity for sport-based interventions that improve health and well-being through physical activity, social relations, and learning.

  • 2018
  • Ingår i: The Science and Practice of Racket Sport for Improved Performance and Health: Special Focus on Table Tennis. - Halmstad : Halmstad University. - 9789163971297 ; , s. 22-23
  • Konferensbidrag (refereegranskat)abstract
    • Introduction: Physical activity has a positive impact on physical and psychological well-being (Delisle, Werch, Wong, Bian, & Weiler, 2010), as well as social relations and skills development (Lamu & Olsen, 2016; Perkins & Williamon, 2014). Adopting a “healthy organization” culture through health programs, with strong senior and middle management support, and using interventions can promote health at workplaces (Rajaratnam et. al., 2014). We, therefore, consider it relevant to have physical activity, social relations and skill development as the starting point in a tabletennis intervention at the workplace.Aim: The aim was to develop, implement, and evaluate an intervention for improving health and wellbeing in the workplace by introducing table tennis. Methods and results: The intervention design consisted of two workshops and five table tennis sessions. Thirteen employees from a warehouse within the retail sector participated in the intervention. The participants had various backgrounds in table tennis and sport in general, different motives to participate, and came from different groups of employees (management and stock). After an introductory workshop, two table tennis coaches held one training session a week over the fiveweek intervention period. The employees were divided in to two groups and each group had a 45-minute session. The evaluation is in progress at present. Pre- and post-measurement has been conducted using health questionnaires Short Form 36 Health Survey (SF-36) and International Physical Activity Questionnaire (IPAQ). These will be analysed using a Bayesian paired t-test analysis. Based on the results of the questionnaire focus group interviews will be conducted with participants. Three focus groups of 4-5 participants in each group. The questions will focus on 24 experiences of the intervention and reflections on future directions for table-tennis and wellbeing activities at the workplace. Finally, there will be a workshop pointing out future directions for sport-based health activities at the workplace.

  • Reaction conditions for the resolution of 2-methylalkanoic acids in esterification and hydrolysis with lipase from Candida cylindracea
  • 1991
  • Ingår i: Applied Microbiology and Biotechnology. - 0175-7598. ; 35:5, s. 572-578
  • Tidskriftsartikel (refereegranskat)abstract
    • We have demonstrated resolution of 2-methylalkanoic acids using lipase from Candida cylindracea as a catalyst. The resolution of 2-methyldecanoic acid was more successful than that of 2-methylbutyric acid both by esterification and hydrolysis. This indicates that the resolution of the acid is dependent on the chain length of the acid moiety. The chain length of the alcohol moiety of the ester affected the resolution of the long-chain acid only. Using esterification, (R)-2-methyldecanoic acid was produced in an enantiomeric excess (e.e.) of 95% (E = 40). If the enantiomeric ratio is low (E = 3.6), as in the resolution of 2-methylbutyric acid, esterification combined with a high equilibrium conversion could be used to yield the remaining acid in a high e.e. In the hydrolytic reactions, the e.e. and the equilibrium conversion were dependent on the pH and the presence of CaCl2. When octyl 2-methyldecanoate was hydrolysed at pH 8.0 in the presence of CaCl2, the (S)-acid was formed with an e.e. of 80% (E = 9), but when the hydrolysis was carried out at pH 7.5 without CaCl2, a very low e.e. and a low equilibrium conversion were observed. The latter conditions allowed the esterification of 2-methyldecanoic acid with 1-octanol even in aqueous medium.

  • Reinventing tradition : Exploring the creation of new meaning through innovations involving craft-based design
  • 2019
  • Ingår i: Creativity and Innovation Management. - : Wiley. - 0963-1690 .- 1467-8691. ; 28:1, s. 124-137
  • Tidskriftsartikel (refereegranskat)abstract
    • Many studies in the field of innovation focus on novelty and technology, but recent literature and contemporary design practices have proposed an alternative view. Innovation has instead been suggested to bring about new meanings, often driven by design, and lately the role of tradition has been highlighted as a source of innovation rather than an obstacle. However, research has thus far not explained how tradition is actually used in innovation practice. By exploring design elements from craft-based design practice, the overall aim of this study is to identify and explore how craft-based design leads to new meaning and Innovation Through Tradition (ITT). To gain further insights into craft-based design for ITT, the Research Through Design method in combination with narrative writing was used, investigating innovation from inside design practice. The empirical design work highlights the innovation potential when craft-based design and production processes deliberately use tradition, not only by embracing the value of tradition itself, but also by creating new meaning through its unexpected combination with novel components. Finally, a visualization of key elements that can be reinvented is outlined to highlight tensions between different design elements that have the potential to produce new and valuable product meanings.

  • 80-kV(p) hepatic CT to reduce contrast medium dose in azotemic patients: a feasibility study
  • 2020
  • Ingår i: Acta Radiologica. - : SAGE Publications. - 0284-1851 .- 1600-0455. ; 61:4, s. 441-449
  • Tidskriftsartikel (refereegranskat)abstract
    • Background Low peak kilovoltage (kV(p)) computed tomography (CT) may be used to reduce contrast medium doses in patients at risk of contrast medium-induced acute kidney injury if image noise can be controlled by increasing X-ray tube loading (mAs). Purpose To evaluate objective and subjective image quality in 80-kV(p) CT with reduced contrast medium dose and compensated mAs for unchanged image noise in patients with estimated glomerular filtration rate Material and Methods 80-kV(p) CT with 300 mg I/kg in 40 patients (body mass index 18-32 kg/m(2), glomerular filtration rate <45 mL/min) and 120-kV(p) CT with 500 mg I/kg in 40 patients (body mass index = 17-30 kg/m(2), glomerular filtration rate >= 45 mL/min) was compared on mean hepatic attenuation, image noise, contrast medium enhancement, signal-to-noise ratio, contrast-to-noise ratio, effective radiation dose, and subjective image quality. Results There were no significant differences regarding median hepatic post-contrast attenuation, image noise, contrast medium enhancement, signal-to-noise ratio, contrast-to-noise ratio, or effective dose between the 80-kV(p) and 120-kV(p) cohorts: 114/110 HU; 14/14 HU; 57/53 HU; 8.0/7.4; 3.8/3.5; and 5.3/5.9 mSv, respectively. However, subjective image visual grading showed statistically significantly inferior scores for 80 kV(p) for six of eight items. After exclusion of seven inferior examinations not caused by the chosen kV(p) technique, only three items showed inferior scores for 80 kV(p). Only 5% of gradings regarding overall image quality were Conclusion Despite lower subjective image quality, objective data indicate that 80-kV(p) CT with reduced contrast medium doses and compensated mAs may have the potential to provide satisfactory diagnostic quality in patients with body mass index <30 kg/m(2), which could benefit patients at risk of contrast medium-induced acute kidney injury.

  • Can iterative reconstruction algorithms replace tube loading compensation in low kVp hepatic CT? Subjective versus objective image quality
  • 2020
  • Ingår i: Acta Radiologica Open. - : SAGE Publications. - 2058-4601 .- 2058-4601. ; 9:3
  • Tidskriftsartikel (refereegranskat)abstract
    • Background: Hepatic computed tomography (CT) with decreased peak kilovoltage (kVp) may be used to reduce contrast medium doses in patients at risk of contrast-induced acute kidney injury (CI-AKI); however, it increases image noise. To preserve image quality, noise has been controlled by X-ray tube loading (mAs) compensation (TLC), i.e. increased mAs. Another option to control image noise would be to use iterative reconstructions (IR) algorithms without TLC (No-TLC). It is unclear whether this may preserve image quality or only reduce image noise. Purpose: To evaluate image quality of 80 kVp hepatic CT with TLC and filtered back projection (FBP) compared with 80 kVp with No-TLC and IR algorithms (SAFIRE 3 and 5) in patients with eGFR <45 mL/min. Material and Methods: Forty patients (BMI 18-32 kg/m(2)) were examined with both protocols following injection of 300 mg I/kg. Hepatic attenuation, image noise, enhancement, signal-to-noise ratio (SNR), contrast-to-noise ratio (CNR), and subjective image quality were evaluated for each patient. Results: Comparing TLC/FBP with No-TLC/IR-S5, there were no significant differences regarding hepatic attenuation, image noise, enhancement, SNR and CNR: 114 vs. 115 HU, 14 vs. 14 HU, 55 vs. 57 HU, 8.0 vs. 8.4, and 3.8 vs. 4.0 in median, respectively. No-TLC/IR-S3 resulted in higher image noise and lower SNR and CNR than TLC/FBP. Subjective image quality scoring with visual grading showed statistically significantly inferior scores for IR-S5 images. Conclusion: CT of 80 kVp to reduce contrast medium dose in patients at risk of CI-AKI combined with IR algorithms with unchanged tube loading to control image noise does not provide sufficient diagnostic quality.

  • Collective Learning in Innovative Networks
  • 2005
  • Konferensbidrag (övrigt vetenskapligt/konstnärligt)abstract
    • There are different forms for boundary-crossing learning in and between organizations. One growing form is network. This paper will present a study of collective learning in two innovative networks at the west coast of Sweden. One network is about regional health and growth, the other about leadership development among general directors. Representatives from different organizations meet to learn from each other’s practice. How does this learning take place? What supports and what prevents it? Does it have any effects in their own organizations? Data has been collected through questionnaires, observations, systematic reflections and focus interviews. Results show that the participants express great satisfaction with their outcomes on a personal base. It gives them inspiration, ideas, knowledge and contacts. It also gives them support, strength and self-confidence. One important factor in this context is differences. The participants are coming from different organizations, have different perspectives and experiences. The findings show interesting variances between the two networks that has effects on the type of learning. A problem is the transformation of learning from the network, by the participants into their organizations. There seems to be some important learning barriers. This will be further discussed in the paper.

  • Employee-Driven Innovation : An Intervention Using Action Research
  • 2019
  • Ingår i: Technology Innovation Management Review. - Ottawa : Carleton University. - 1927-0321. ; 9:5, s. 44-53
  • Tidskriftsartikel (refereegranskat)abstract
    • This article describes an intervention to design and test a method for employee-driven innovation and a model for learning among managers and development leaders. The empirical basis for the intervention focused on personal assistants in the home service within a municipality in Sweden. The intervention was carried out using action research in on a series of workshops with a group of employees, managers, development leaders. Using a “stage” and “stands” theatre metaphor, employees engaged in collective, innovative learning “on the stage” combined with observations and reflections from managers and development leaders “in the stands”. This article contributes a method that can generate creative ideas among the employees and a model that can stimulate experience-based learning through observations. The intervention also shows that action research can be used to develop and test methods and models.

  • Hållbara leadereffekter i teori och praktik: Delrapport 2 : Utvärderingsrapport 2020:1
  • 2020
  • Rapport (refereegranskat)abstract

Data are analyzed by theories on science in society, organizational learning, dialogue and reflection. Findings – Results show that the participants in the networks reduced their insecurity and increased their competence to handle change. The dialogue, exchange of experiences and systematic reflections gave ideas, knowledge and contacts that supported them in their work life and inspired them to take development action. Research limitations/implications – Researchers can contribute to the development process by acting as facilitators and using interactive methods for joint learning. Coming from different cultures and aiming to collaborate under equal and democratic conditions, all involved actors must have an attitude that is humble and free of an assumed prestige. Practical implications – External development networks with university participation can be a useful tool for internal development. Originality/value – Interactive research is a possible way for universities to make practical contributions at work. All academics who work with applied research should consider this approach.

  • Lärande nätverk : En social oas i utvecklingsprocessen
  • 2010

  • Managing Project Transformation in a Complex Context
  • 2007
  • Ingår i: Creativity and Innovation Management. - Oxford : Blackwell Publishing. - 0963-1690 .- 1467-8691. ; 16:1, s. 46-52
  • Tidskriftsartikel (refereegranskat)abstract
    • Complex projects are often undertaken in uncertain situations in which project participants who adopt a rational-oriented strategy tend to face difficulties in adapting and adjusting to changes that occur during the project. In this context a process-oriented strategy, although difficult to manage, can be a better way to handle project transformation. Collective learning then becomes crucial. This article describes some lessons learned when a project manager and team applied a process-oriented strategy in a complex project. Data has been collected through interactive research and evaluation with different methods including surveys, observations and interviews. The analysis shows that it is possible to organize for learning and to manage transformation in a complex context. Important factors for success are a process-oriented strategy, a creative climate, formative evaluation and systematic reflections. A major issue that is discussed in the article is the transfer of outcomes from the project to the participating organizations.

  • Konferensbidrag (övrigt vetenskapligt/konstnärligt)abstract
    • In 1997 a new university law imposed Swedish universities a third mission. In addition to research and teaching, universities now have to “collaborate with the surrounding society”.This new law was quite indistinct which raises questions about how, how much and in what way the co-operation between university and society should be performed. What obstacles and possibilities are there for real deep cooperation between such different cultures as the academic sector, the commercial private sector and the political public sector? In our paper we reflect upon the collaboration between research and practice from two different processes where we have been involved as action researchers. Both these processes were aimed to bridge the gap between the academic world and the industry and public sector. We reflect upon our role as researchers in relation to the role of the practitioners. What were our different incentives and expectations when entering a joint learning process? What did we learn and what different knowledge was there to serve our different needs? We discuss the two cases by highlighting the research methods, where researchers and practitioners in joint action take part in development processes, and how these processes both contribute theoretical knowledge and practical knowledge to as well academy as working life. But we also discuss the difficulties in handling the role as action researcher and the problems faced when people from different cultures and with different expectations act together.

  • A new technique for reconstruction of the atrophied narrow alveolar crest in the maxilla using morselized impacted bone allograft and later placement of dental implants
  • 2008
  • Ingår i: Clinical Implant Dentistry and Related Research. - Oxon : Blackwell Publishing. - 1523-0899 .- 1708-8208. ; 10:2, s. 86-92
  • Tidskriftsartikel (refereegranskat)abstract
    • Background: In cases when the alveolar crest is too narrow to host an implant, lateral augmentation is required. The use of autogenous bone blocks harvested from the iliac crest is often demanded. One disadvantage is the associated patient morbidity.Purpose: The purpose of this study was to clinically and histologically evaluate the use of morselized impacted bone allograft, a novel technique for reconstruction of the narrow alveolar crest.Materials and methods: Two patients with completely edentulous maxillae and one partially edentulous, with a mean age of 77 years (range 76-79 years) were included in the study. The alveolar crest width was < 3 mm without possibility to place any implant. Bone grafts were taken from a bone bank in Gavle Hospital. Bone from the neck of femur heads was milled to produce bone chips. The milled bone was partially defatted by rinsing in 37 degrees C saline solution. After compression of the graft pieces with a size of 15 mm (height), 30 mm (length), and 6 mm (width), they were then fit to adapt to the buccal surface of the atrophied alveolar crest. One piece was placed to the right and one to the left side of the midline. On both sides fibrin glue was used (Tisseel (R), Baxter AG, Vienna, Austria) to stabilize the graft. After 6 months of graft healing, dental implants were placed, simultaneously biopsies were harvested and in one patient two oxidized microimplants were placed. At the time of abutment connection, microimplants were retrieved with surrounding bone for histology. Fixed screw-retained bridges were fabricated in mean of 7 months after implant surgery. Radiographs were taken before and after implant surgery and after 1 year of loading. Results: Sixteen implants with an oxidized surface were placed (TiUnite (R), Nobel Biocare AB, Goteborg, Sweden). After 1 year of functional loading, all implants were clinically stable. The marginal bone loss was 1.4 mm (SD 0.3) after 1 year of loading. The histological examination showed resorption and subsequent bone formation on the allograft particles. There were no signs of inflammatory cell infiltration in conjunction with the allograft. The two microimplants showed bone formation directly on the implant surface. Conclusions: This study shows that morselized impacted bone allograft can be used to increase the width of the atrophied narrow alveolar crest as a good alternative to autogenous bone grafts in elderly patients. The histological examination of biopsies revealed a normal incorporation process and no signs of an immunological reaction. Further studies with larger samples are of important to be able to conclude if equal results can be obtained using morselized impacted bone allograft as for autogenous bone graft.

  • LEADER and rural development policy - What's the problem represented to be?
  • 2024
  • Ingår i: Journal of Rural Studies. - Oxford : Elsevier. - 0743-0167 .- 1873-1392. ; 108
  • Tidskriftsartikel (refereegranskat)abstract
    • This article problematizes the EU Programme LEADER as a policy to improve opportunities to live and thrive in rural areas. LEADER was initiated in 1991 as a Commission initiative and has, over time, expanded and become an institutionalized public policy in the EU. LEADER is based on a particular development method - the LEADER method - which sets out fundamental principles for conducting development work. The idea is that rural development should start with Local Action Groups at the grassroots level, emphasizing networking and entrepreneurship within a geographic context. Drawing on Bacchi's methodology, What's the problem represented to be, the article aims to examine the LEADER Programme and analyze problem representations and political rationalities. The analysis shows that actors working within LEADER have developed political rationalities that partly constitute obstacles to achieving long-term impacts. It concerns issues related to the bureaucratic framework of LEADER and representations on a fixation on producing added values, an internal logic of project administration, and an emphasis highlighting the role of the individual entrepreneur. The LEADER Programme has not only been institutionalized as part of the EU and member states' administrative systems. Still, it has also created its ideological superstructure consisting of a specific narrative, methodology, language, and common forms of organizing. This gives LEADER advantages but also risks associated with internal closedness that creates subjectification between ‘them and us' and produces lived effects that hinder structural changes promoting rural areas.
