    • In 2008 we published the first set of guidelines for standardizing research in autophagy. Since then, research on this topic has continued to accelerate, and many new scientists have entered the field. Our knowledge base and relevant new technologies have also been expanding. Accordingly, it is important to update these guidelines for monitoring autophagy in different organisms. Various reviews have described the range of assays that have been used for this purpose. Nevertheless, there continues to be confusion regarding acceptable methods to measure autophagy, especially in multicellular eukaryotes. A key point that needs to be emphasized is that there is a difference between measurements that monitor the numbers or volume of autophagic elements (e.g., autophagosomes or autolysosomes) at any stage of the autophagic process vs. those that measure flux through the autophagy pathway (i.e., the complete process); thus, a block in macroautophagy that results in autophagosome accumulation needs to be differentiated from stimuli that result in increased autophagic activity, defined as increased autophagy induction coupled with increased delivery to, and degradation within, lysosomes (in most higher eukaryotes and some protists such as Dictyostelium) or the vacuole (in plants and fungi). In other words, it is especially important that investigators new to the field understand that the appearance of more autophagosomes does not necessarily equate with more autophagy. In fact, in many cases, autophagosomes accumulate because of a block in trafficking to lysosomes without a concomitant change in autophagosome biogenesis, whereas an increase in autolysosomes may reflect a reduction in degradative activity. Here, we present a set of guidelines for the selection and interpretation of methods for use by investigators who aim to examine macroautophagy and related processes, as well as for reviewers who need to provide realistic and reasonable critiques of papers that are focused on these processes. These guidelines are not meant to be a formulaic set of rules, because the appropriate assays depend in part on the question being asked and the system being used. In addition, we emphasize that no individual assay is guaranteed to be the most appropriate one in every situation, and we strongly recommend the use of multiple assays to monitor autophagy. In these guidelines, we consider these various methods of assessing autophagy and what information can, or cannot, be obtained from them. Finally, by discussing the merits and limits of particular autophagy assays, we hope to encourage technical innovation in the field.

    • p33(ING1b), a newly discovered candidate tumor suppressor gene and a nuclear protein, belongs to the inhibitor of growth gene family. Previous studies have shown that p33(ING1b) is involved in the restriction of cell growth and proliferation, apoptosis, tumor anchorage-independent growth, cellular senescence, maintenance of genomic stability and modulation of cell cycle checkpoints. Loss of nuclear p33(ING1b) has been observed in melanoma, seminoma, papillary thyroid carcinoma, oral squamous cell carcinoma, breast ductal cancer and acute lymphoblastic leukemia. Inactivation and/or decreased expression of p33(ING1b) have been reported in various types of cancer, including head and neck squamous cell, breast, lung, stomach, blood and brain malignancies. Since little is known about the clinicopathological significance of p33(ING1b) in esophageal squamous cell carcinoma (ESCC), this study aimed to investigate the association of p33(ING1b) expression with clinicopathological variables and particularly interesting new cysteine-histidine rich protein (PINCH) in patients with ESCC. p33(ING1b) expression was examined by immunohistochemistry in 20 normal esophageal mucosa and in 64 ESCC specimens. The results revealed that the positive expression of p33(ING1b) protein in normal squamous cells was localized in the nucleus alone and the positive rate was 95%, while in ESCCs, the positive expression was mainly in the cytoplasm, together with nuclear expression, and the positive rate was 36% (P<0.0001). Furthermore, the cases with lymph node metastasis showed a higher frequency of positive cytoplasmic expression than those without metastasis (P=0.001). The cytoplasmic expression of p33(ING1b) was positively related to PINCH expression (P<0.0001) in ESCC, and the cases positive for both proteins had a high lymph node metastasis rate (P=0.001). In conclusion, p33(ING1b) cellular compartmental shift from the nucleus to the cytoplasm may cause loss of normal cellular function and play a central role in the tumorigenesis and metastasis of ESCC.

PINCH expression is also independently correlated with poorer survival in patients with colorectal cancer. However, there is no study of PINCH in gastric cancer, therefore, the aim of this project was to investigate PINCH expression and its clinicopathological significance in gastric adenocarcinoma. less thanbrgreater than less thanbrgreater thanPatients and methods: PINCH expression was immunohistochemically examined in normal gastric mucous (n = 30) and gastric adenocarcinoma (n = 73), from gastric cancer patients. less thanbrgreater than less thanbrgreater thanResults: PINCH expression in the associated-stroma of gastric cancers was heterogeneous, and its positive rate (75%) was higher than that of normal gastric mucosa (43%, X-2 = 9.711, p = 0.002). The stronger staining was observed at the invasive edge of tumour when compared to the inner area of tumour. The rate of positive PINCH (88%) in the cases with lymph node metastasis was higher than that (52%) in the cases without metastasis (X-2 = 11.151, p = 0.001). PINCH expression was not correlated with patients gender, age, tumour size, differentiation and invasion depth (p andgt; 0.05). less thanbrgreater than less thanbrgreater thanConclusion: PINCH protein might play an important role in the tumourigenesis and metastasis of gastric adenocarcinoma.

    • Two noncentrosymmetric (NCS) quaternary selenides, PbGa2SiSe6 (1) and PbGa2GeSe6 (2), with second-order nonlinear optical (NLO) responses, were synthesized by a conventional high-temperature solid-state reaction method. Compounds 1 and 2 are constructed by three NCS chromophores, [PbSe4], [GaSe4], and [Ga/SiSe4] or [Ga/GeSe4], with the covalent interactions between the X and Se atoms (X = Pb, Ga, Ga/Si, or Ga/Ge). They crystallize in the polar space groups Cc and Fdd2, respectively. Inspiringly, compound 2 is phase-matchable (PM) and shows high laser-induced damage threshold (LIDT) of 3.7 x AgGaS2 and wide transparent region (0.6325 mu m) in the mid-infrared (MIR) region. Most importantly, it presents extraordinary strong second harmonic generation (SHG) at 2.05 mu m radiation of about 12 x AgGaS2 at the particle size of 2545 mu m, which represents the strongest SHG among PM chalcogenides to date. The calculated major SHG tensor elements of compounds 1 and 2 are d31 = 224.7 and d12 = 222.1 pm/V, respectively, while the calculated d36 of AgGaS2 is only 21.2 pm/V.

    • Background: Expression of the meningioma-associated protein (MAC30) was increased in several types of tumors, including esophageal, gastric and colon tumors, compared to normal tissue. MAC30 expression levels gradually increased from normal colorectal mucosa to primary colorectal cancer and colorectal cancer spreading to the lymph nodes. MAC30 expression was related to survival in patients with colorectal cancer. However, there is no study on MAC30 in oral squamous cell carcinoma (OSCC). Methods: Therefore, MAC30 expression in OSCC was investigated and possible associations of MAC30 expression with clinicopathological variables in OSCC have been analyzed. MAC30 expression was immunohistochemically examined in 20 normal oral mucosa and 43 OSCC specimens. Results: Expression levels of MAC30 in the cytoplasm markedly increased from normal oral epithelial cells to primary OSCC. Strong cytoplasmic staining was significantly higher in primary OSCC compared to normal oral mucosa samples (51 vs. 20%, p = 0.019). Furthermore, MAC30 expression levels in primary tumors of patients with lymph node metastasis exceeded levels in those without metastasis (65 vs. 35%, p = 0.048), and MAC30 expression in poorly differentiated tumors was higher than in well-differentiated ones (90 vs. 39%, p = 0.005). Conclusion: Overexpression of MAC30 in the cytoplasm of OSCC may predict nodal metastasis and poor differentiation.

    • Nucleoporin 88 (Nup 88) is a component of the nuclear pore complexes (NPCs) that mediates nucleocytoplasmic trafficking of macromolecules, Nup 88 has been reported to be up-regulated in a wide variety of malignancies. Studies show that overexpression of this antigen is associated with the development, agressiveness, differentiation and prognosis in some tumours. Since no study has been carried out in the relationship between the Nup 88 expression and clinicopathological features in the patients with oral squamous cell carcinoma (OSCC), this study aimed to determine Nup 88 expression in OSCC and its clinicopathological significance. Nup 88 expression was examined by immunohistochemistry in 20 normal oral mucosa specimens and 83 OSCC tissues. The frequency of positive Nup 88 expression was gradually increased from normal oral mucosa (10%) to primary OSCC (40%, P=0.012). The Nup 88 positive rate in OSCC patient with nodal metastasis was significantly higher than those without nodal metastasis (64% vs. 21%, P=0.000085). The frequency of positive Nup 88 expression was significantly different between worse and better differentiation (80 vs. 27%, P=0.000024). Nup 88 expression was not related to the patients' gender, age, location and tumour size (P>0.05). In conclusion, Nup 88 may play an important role in tumorigenesis in oral squamous cell carcinoma. Upregulation of Nup 88 is associated with nodal metastasis and poor differentiation in oral squamous cell carcinoma.

    • Background: Particularly interesting new cysteine-histidine rich protein ( PINCH), as an adapter protein of the LIM family for signal transduction in the integrin and growth factor pathway, is upregulated in the stroma of several common types of cancers and involved in promoting tumor progression. In the present study, we examined PINCH expression in normal endometrium, atypical endometrial hyperplasia and endometrioid carcinoma, and further studied the relationships of PINCH expression with clinicopathological variables in cancer patients. Methods: PINCH expression was examined by immunohistochemistry in 23 normal endometrial samples, 18 atypical endometrial hyperplasias and 48 endometrioid endometrial carcinomas. Results: The PINCH expression in the stroma of cancer (71%) was significantly increased compared to either normal endometrium (17%, p andlt; 0.0001) or atypical hyperplasia (39%, p = 0.017), along with 9 cancers that had stronger PINCH expressions at the invasive margin of the cancers compared to the inner cancers. PINCH expression in cancer was higher in the patients with hypertension (p = 0.041) and estrogen exposure time andgt;30 years (p = 0.021). On the other hand, PINCH expression was not related to menopausal status, gravid status, blood sugar/lipid, family background of cancer, histological grade, myometrial invasion, cervical involvement, lymph nodal metastases, growth pattern, estrogen and progestogen receptors (p andgt; 0.05). Conclusion: The results suggest that PINCH seems to play a role, presently unknown, in the tumorigenesis and development of endometrial cancer that merits further study.

  • Lysine demethylase LSD1 delivered via small extracellular vesicles promotes gastric cancer cell stemness

    • Several studies have examined the functions of nucleic acids in small extracellular vesicles (sEVs). However, much less is known about the protein cargos of sEVs and their functions in recipient cells. This study demonstrates the presence of lysine-specific demethylase 1 (LSD1), which is the first identified histone demethylase, in the culture medium of gastric cancer cells. We show that sEVs derived from gastric cancer cells and the plasma of patients with gastric cancer harbor LSD1. The shuttling of LSD1-containing sEVs from donor cells to recipient gastric cancer cells promotes cancer cell stemness by positively regulating the expression of Nanog, OCT4, SOX2, and CD44. Additionally, sEV-delivered LSD1 suppresses oxaliplatin response of recipient cells in vitro and in vivo, whereas LSD1-depleted sEVs do not. Taken together, we demonstrate that LSD1-loaded sEVs can promote stemness and chemoresistance to oxaliplatin. These findings suggest that the LSD1 content of sEV could serve as a biomarker to predict oxaliplatin response in gastric cancer patients.

    • Objective: To investigate the association of FXYD-3 expression with clinicopathological variables and PINCH in patients with ESCC.Patients and methods: Expression of FXYD-3 protein was immunohistochemically examined in normal esophageal mucous (n = 20) and ESCC (n = 64).  Results: Expression of FXYD-3 in the cytoplasm markedly increased from normal esophageal epithelial cells to primary ESCC ( p = 0.001). The expression of FXYD-3 was correlated with TNM stages and depth of tumour invasion. Furthermore, the cases with lymph node metastasis tended to show a higher frequency of positive expression than those without metastasis ( p = 0.086), and FXYD-3 expression tended to be positively related to the expression of PINCH (p = 0.063). Moreover, the cases positive for both proteins had the highest frequency of lymph node metastasis (p = 0.001). However, FXYD-3 expression was not correlated with patient,s gender (p = 0.847), age (p = 0.876), tumour location (p = 0.279), size (p = 0.771) , grade of differentiation (p = 0.279), and survival (p = 0.113).Conclusion: overexpression of FXYD-3 in the cytoplasm may play an important role in the tumourigenesis and development in the human ESCC, particularly in combination with PINCH expression.

  • Livin expression is an independent factor in rectal cancer patients with or without preoperative radiotherapy

    • Background: This study was aimed to investigate the expression significance of Livin in relation to radiotherapy (RT), clinicopathological and biological factors of rectal cancer patients.Methods: This study included 144 primary rectal cancer patients who participated in a Swedish clinical trial of preoperative radiotherapy. Tissue microarray samples from the excised primary rectal cancers, normal mucosa and lymph node metastases were immunostained with Livin antibody. The proliferation of colon cancer cell lines SW620 and RKO was assayed after Livin knock-down.Results: The expression of Livin was significantly increased from adjacent (P = 0.051) or distant (P = 0.028) normal mucosa to primary tumors. 15.4% (2/13) and 39.7% (52/131) patients with Livin-negative and positive tumors died at 180 months after surgery, and the difference tended to be statistically significant (P = 0.091). In multivariate analyses, the difference achieved statistical significance, independent of TNM stage, local and distant recurrence, grade of differentiation, gender, and age (odds ratio = 5.09, 95% CI: 1.01-25.64, P = 0.048). The in vitro study indicated colon cancer cells with Livin knock-down exhibited decreased proliferation compared with controls after RT.Conclusions: The expression of Livin was was independently related to survival in rectal cancer patients, suggesting Livin as a useful prognostic factor for rectal cancer patients.

    • The antiapoptotic factor Livin has been considered critical for tumor progression and poor prognosis for variant types of tumors. However, there are only limited reports regarding its expression and biological functions in colon cancer. Here, we examined Livin expression in four colon cancer cell lines (HCT116, RKO, KM12C, and SW620) in the presence or absence of cisplatin that was used as a model reagent. We found the different response to cisplatin was related to endogenous Livin expression level. From among a panel of apoptosis-related factors (p53, Bcl-2, Bcl-XL, BAX, and survivin), the expression of Livin was upregulated after cisplatin treatment in a dose-dependent manner. Both immunocytochemistry and nuclear cytoplasmic fractionation indicated Livin remained in the cytoplasm after treatment with cisplatin. In an attempt to explore the mechanism, we found the elevated expression of Livin was not due to the decreased degradation by proteosome but was enhanced at the mRNA level. Besides, cisplatin treatment activated the mammalian target of rapamycin (mTOR) pathway as shown by increased phosphorylation of Akt1, mTOR, S6K, and 4E-BP1, together with the elevated Livin. The PI3K inhibitor LY294002 inhibited both the phosphorylation of mTOR and upregulation of Livin. The stable overexpression of Livin inhibited the activation of caspase-3 and led to resistance to cisplatin, while the knockdown of Livin by siRNA rendered colon cancer cells more sensitive to cisplatin. Our study, along with others, highlighted the potential of Livin for cancer therapy in colon cancer.

    • Systemic lupus erythematosus (SLE) is a common systemic autoimmune disease with complex etiology but strong clustering in families (lambda(S) = approximately 30). We performed a genome-wide association scan using 317,501 SNPs in 720 women of European ancestry with SLE and in 2,337 controls, and we genotyped consistently associated SNPs in two additional independent sample sets totaling 1,846 affected women and 1,825 controls. Aside from the expected strong association between SLE and the HLA region on chromosome 6p21 and the previously confirmed non-HLA locus IRF5 on chromosome 7q32, we found evidence of association with replication (1.1 x 10(-7) < P(overall) < 1.6 x 10(-23); odds ratio = 0.82-1.62) in four regions: 16p11.2 (ITGAM), 11p15.5 (KIAA1542), 3p14.3 (PXK) and 1q25.1 (rs10798269). We also found evidence for association (P < 1 x 10(-5)) at FCGR2A, PTPN22 and STAT4, regions previously associated with SLE and other autoimmune diseases, as well as at > or =9 other loci (P < 2 x 10(-7)). Our results show that numerous genes, some with known immune-related functions, predispose to SLE.

    • A new four-armed molecule Th-4(DTPAB) with a triphenylamine-benzothiadiazole-triphenylamine unit as the core and 4-hexylthiophene as arms was synthesized. Solution-processed organic solar cells based on blends of Th-4(DTPAB) and PC71BM demonstrate a power conversion efficiency of 3.18% with a high open circuit voltage of 0.96 V.

    • The Visual Object Tracking challenge 2014, VOT2014, aims at comparing short-term single-object visual trackers that do not apply pre-learned models of object appearance. Results of 38 trackers are presented. The number of tested trackers makes VOT 2014 the largest benchmark on short-term tracking to date. For each participating tracker, a short description is provided in the appendix. Features of the VOT2014 challenge that go beyond its VOT2013 predecessor are introduced: (i) a new VOT2014 dataset with full annotation of targets by rotated bounding boxes and per-frame attribute, (ii) extensions of the VOT2013 evaluation methodology, (iii) a new unit for tracking speed assessment less dependent on the hardware and (iv) the VOT2014 evaluation toolkit that significantly speeds up execution of experiments. The dataset, the evaluation kit as well as the results are publicly available at the challenge website (http://​votchallenge.​net).

  • Preinvasion Assessment of Exotic Bark Beetle-Vectored Fungi to Detect Tree-Killing Pathogens

    • Exotic diseases and pests of trees have caused continental-scale disturbances in forest ecosystems and industries, and their invasions are considered largely unpredictable. We tested the concept of preinvasion assessment of not yet invasive organisms, which enables empirical risk assessment of potential invasion and impact. Our example assesses fungi associated with Old World bark and ambrosia beetles and their potential to impact North American trees. We selected 55 Asian and European scolytine beetle species using host use, economic, and regulatory criteria. We isolated 111 of their most consistent fungal associates and tested their effect on four important southeastern American pine and oak species. Our test dataset found no highly virulent pathogens that should be classified as an imminent threat. Twenty-two fungal species were minor pathogens, which may require context-dependent response for their vectors at North American borders, while most of the tested fungi displayed no significant impact. Our results are significant in three ways; they ease the concerns over multiple overseas fungus vectors suspected of heightened potential risk, they provide a basis for the focus on the prevention of introduction and establishment of species that may be of consequence, and they demonstrate that preinvasion assessment, if scaled up, can support practical risk assessment of exotic pathogens.

    • Newton's gravitational constant G may vary with time at an extremely low level. The time variability of G will affect the orbital motion of a millisecond pulsar in a binary system and cause a tiny difference between the orbital period-dependent measurement of the kinematic distance and the direct measurement of the annual parallax distance. PSR J0437-4715 is the nearest millisecond pulsar and the brightest at radio wavelengths. To explore the feasibility of achieving a parallax distance accuracy of one light-year, comparable to the recent timing result, with the technique of differential astrometry, we searched for compact radio sources quite close to PSR J0437-4715. Using existing data from the Very Large Array and the Australia Telescope Compact Array, we detected two sources with flat spectra, relatively stable flux densities of 0.9 and 1.0 mJy at 8.4 GHz and separations of 13 and 45 arcsec. With a network consisting of the Long Baseline Array and the Kunming 40-m radio telescope, we found that both sources have a point-like structure and a brightness temperature of >= 10(7) K. According to these radio inputs and the absence of counterparts in other bands, we argue that they are most likely the compact radio cores of extragalactic active galactic nuclei, rather than Galactic radio stars. The finding of these two radio active galactic nuclei will enable us to achieve a sub-pc distance accuracy with in-beam phase-referencing very-long-baseline interferometric observations and provide one of the most stringent constraints on the time variability of G in the near future.

    • Conditional temperature-sensitive (ts) mutations are valuable reagents for studying essential genes in the yeast Saccharomyces cerevisiae. We constructed 787 ts strains, covering 497 (∼45%) of the 1,101 essential yeast genes, with ∼30% of the genes represented by multiple alleles. All of the alleles are integrated into their native genomic locus in the S288C common reference strain and are linked to a kanMX selectable marker, allowing further genetic manipulation by synthetic genetic array (SGA)-based, high-throughput methods. We show two such manipulations: barcoding of 440 strains, which enables chemical-genetic suppression analysis, and the construction of arrays of strains carrying different fluorescent markers of subcellular structure, which enables quantitative analysis of phenotypes using high-content screening. Quantitative analysis of a GFP-tubulin marker identified roles for cohesin and condensin genes in spindle disassembly. This mutant collection should facilitate a wide range of systematic studies aimed at understanding the functions of essential genes.

    • Unsteady Reynolds-averaged Navier-Stokes (URANS) simulations were performed to simulate trains with different marshalling forms and lengths entering a tunnel. Three models, including a short train, a double train and a long train, were used to analyse the influence of the train configuration on the pressure variations during a train's passage. The results of the numerical predictions were validated against existing experimental data, with which they showed good agreement. The differences in the maximum pressure peak distribution and the pressure fluctuations were analysed by means of Mach diagrams. The results show that the grouping length exerts a considerable influence on the amplitude of the pressure on the train body and that the influence of the grouping length on the pressure variation on the tunnel wall varies with the location in the tunnel. The tunnel space can be divided into three and four zones with regard to the influences on the maximum positive and negative pressure values, respectively. The different marshalling forms also influence the maximum peak values and local profiles of the pressure history curves, although this influence is much slighter than that of the train's grouping length.

    • During the actual operation of a high-speed train, the airtight performance of the carriages will change significantly. Based on a series of field tests, the external and interior pressure variations of train carriages are measured and then the dynamic pressure tightness coefficient are analysed quantitatively for investigating the airtight performance of a train passing at various speeds through tunnels of different lengths. It is found that the amplitude of the external pressure variations is proportional to the square of the train speed, while the amplitude inside the vehicle is approximately proportional to the speed to the 0.7th power due to the airtight effect. The dynamic pressure tightness coefficient increases with increasing tunnel length and decreases with increasing train speed. Regarding the effect of the carriage location, the coefficient shows an overall increasing trend from the train head to tail.

    • The Three-River Headwaters (TRH) region in the Tibetan Plateau is vulnerable to climate change; changes in summer (June–August) precipitation have a significant impact on water security and sustainability in both local and downstream areas. However, the changes in summer precipitation of different intensities over the TRH region, along with their influencing factors, remain unclear. In this study, we used observational and ERA5 reanalysis data and employed a precipitation categorization and water vapor budget analysis to quantify the categorized precipitation variations and investigate their possible linkages with the water vapor budget. Our results showed an increasing trend in summer precipitation at a rate of 0.9 mm per year (p < 0.1) during 1979–2020, with a significant dry-to-wet transition in 2002. The category ‘very heavy precipitation’ (≥10 mm d−1) contributed 65.1% of the increased summer precipitation, which occurred frequently in the northern TRH region. The dry-to-wet transition was caused by the effects of varied atmospheric circulations in each subregion. Southwesterly water vapor transport through the southern boundary was responsible for the increased net water vapor flux in the western TRH region (158.2%), while southeasterly water vapor transport through the eastern boundary was responsible for the increased net water vapor flux in the central TRH (155.2%) and eastern TRH (229.2%) regions. Therefore, we inferred that the dry-to-wet transition of summer precipitation and the increased ‘very heavy precipitation’ over the TRH was caused by increased easterly and southerly water vapor transport.

    • p73 can activate transcription of p53-responsive genes, thereby inhibiting cell growth. An alternative promoter in the TP73 gene gives rise to an N-terminally truncated isoform of p73, Delta Np73, which lacks the transactivation domain of the full length TAp73 protein. TAp73 is considered pro-apoptotic, and Delta Np73 anti-apoptotic. In this study, we overexpressed ?Np73 beta in p53 wild type and p53 mutant colon cancer cell lines and further exposed the cells to cancer therapeutic drug cisplatin. The results showed that cisplatin decreased the protein expression levels of Delta Np73 beta in a dose-dependent manner, and both TAp73 and p53 were upregulated after cisplatin treatment. Further, clonogenic potential and cell viability were decreased, and apoptotic cells increased, in p53 mutant and in p53 wild type cells. Cellular viability was significantly higher in Delta Np73 beta-cells than mock-transfected cells. However, Delta Np73 beta overexpression did not affect the cellular susceptibility to cisplatin. In conclusion, the overexpression of Delta Np73 beta increases viability in p53 wild type and p53 mutant colon cancer cells, and cisplatin induces the degradation of Delta Np73 beta in a dose-dependent manner.

    • Design concept of a novel combined antenna for cognitive radio applications is proposed and numerically verified. The antenna is composed of a wideband circular monopole and a narrowband dipole. The impedance bandwidth of the wideband monopole antenna is from 4.25-10GHz. The narrowband dipole antenna's central frequency is 5.8GHz. The isolation between the two elements is higher than 50dB, which indicates that the two elements independently co-exist within a small printed circuit board (PCB). The design concept proposed in this paper should be useful to the design and realization of compact, planar antennas and antenna modules having spectrum sensing function.

    • Our recent study showed the important role of special AT-rich sequence binding protein 1 (SATB1) in the progression of human rectal cancer. However, the value of SATB1 in response to radiotherapy (RT) for rectal cancer hasn't been reported so far. Here, SATB1 was determined using immunohistochemistry in normal mucosa, biopsy, primary cancer, and lymph node metastasis from 132 rectal cancer patients: 66 with and 66 without preoperative RT before surgery. The effect of SATB1 knockdown on radiosensitivity was assessed by proliferation-based assay and clonogenic assay. The results showed that SATB1 increased from normal mucosa to primary cancer, whereas it decreased from primary cancer to metastasis in non-RT patients. SATB1 decreased in primary cancers after RT. In RT patients, positive SATB1 was independently associated with decreased response to preoperative RT, early time to metastasis, and worse survival. SATB1 negatively correlated with ataxia telangiectasia mutated (ATM) and pRb2/p130, and positively with Ki-67 and Survivin in RT patients, and their potential interaction through different canonical pathways was identified in network ideogram. Taken together, our findings disclose for the first time that radiation decreases SATB1 expression and sensitizes cancer cells to confer clinical benefit of patients, suggesting that SATB1 is predictive of response to preoperative RT and clinical outcome in rectal cancer.

    • Layered-metal dichalcogenides with extraordinary characteristics of vast surface area, tunable bandgap and superior adsorption capability enable the potential for application in gas sensors. However, the synthesis of effective material for enhanced response performance remains a challenge. Herein, we exploited a fascinating sensitivity and selectivity towards NO2 gas detection using SnS2 nanoflakes prepared via the hydrothermal method. SnS2 nanoflakes with a thickness of 25 nm and an average diameter of approximately 500 nm show the potential for the detection of NO2 gas at low concentrations of ppb levels. The sensing properties of the SnS2 sensors were investigated for different concentrations of NO2 at various operating temperatures. The sensor exhibits the highest gas-sensing response of 161 at 250 οC upon exposure to 5 ppm of NO2 gas with fast response and recovery times. In addition, the sensor shows excellent selectivity with a low detection limit of ppb level. The electronic structure and gas-sensing mechanism are elucidated via finding density of states, charge density, and band structure based on DFT study which is calculated by the Vienna ab-initio simulation package (VASP). The considerable small adsorption energy reveals a physisorption of the NO2 molecules on the SnS2 surface (-0.174 eV), indicating the SnS2 nanoflakes are intriguing candidates for the speedy detection of NO2 gas.

    • The increasing energy demand calls for the development of sustainable energy conversion processes. Here, the splitting of H2O to O-2 and H-2, or related fuels, constitutes an excellent example of solar-to-fuel conversion schemes. The critical component in such schemes has proven to be the catalyst responsible for mediating the four-electron oxidation of H2O to O-2. Herein, we report on the unexpected formation of a single-site Ru complex from a ligand envisioned to accommodate two metal centers. Surprising N-N bond cleavage of the designed dinuclear ligand during metal complexation resulted in a single-site Ru complex carrying a carboxylate amide motif. This ligand lowered the redox potential of the Ru complex sufficiently to permit H2O oxidation to be carried out by the mild one-electron oxidant [Ru(bpy)(3)](3+) (bpy = 2,2'-bipyridine). The work thus highlights that strongly electron-donating ligands are important elements in the design of novel, efficient H2O :oxidation catalysts.

    • Bacterial infection-related diseases are major public safety issues leads to millions of deaths annually. Herein, a porous sulfur doped graphitic carbon nitride (g-SCN) for ecofriendly, metal-free and low systemic toxicity were synthesized. Sulfur doping enables to broaden the absorption spectrum and promote the photocarriers separation for photocatalysis enhancement. Moreover, sulfur element will coordinate with nitrogen, changing the electronic state and endowing g-SCN with the property of nanozyme. More importantly, we established different models and confirmed that S-N-C coordination is the source of peroxidase (POD)-like activity through theory and experiment. The increased specific surface area of g-SCN, ascribing to the porous structure, makes it easier to trap bacteria. With the synergistic effect of photocatalysis and nanozyme, the prepared g-SCN has the ability to kill both gram-negative and gram-positive bacterium, with an antibacterial efficiency up to 100%. This work provides innovative synergistic strategy for constructing nanomaterials for highly efficient antibacterial therapy.

    • N-type organic semiconductor perylene-3,4,9,10-tetracarboxylic acid diimide (PDINH) are well-investigated photocatalyst. However, the photocatalytic potential for antibacterial therapy has been underexplored owing to the insufficient light absorption and rapid recombination of light-induced carriers. Herein, functional group-regulated is introduced by recrystallizing PDINH on the surface of GO in situ, endowing the PDINH/GO with enhanced photocatalytic properties, which harvest light energy across the full spectrum form ultraviolet to near-infrared. Simultaneously, the enhanced photogenerated carriers can activate Lewis base of GO to form an amide bond on the interface between bacteria and material, exhibiting high-efficient and steady bacteria trap. Compared with PDINH, both in vitro and in vivo all demonstrated PDINH/GO possess excellent antibacterial effect. In addition, as a non-metallic semiconductor, PDINH/GO shows capacity of enhancing epidermal cells proliferation and migration, resulting in successful infectious wound regeneration in mice and the side effects in vivo are negligible. Such the integration of wide-spectrum response, high efficiency of carrier separation, intentional bacterial capture and accelerated would healing of PDINH/GO not only enables an effective antibacterial therapy but also contributes to a successful example to activate nanomaterials by regulation of functional groups.

    • The microstructures and mechanical properties (tension strength R-m, yield strength R-t0.5 and impact energy Akv) of the low-nickel alloy steels 08Ni, 12CrNi and 20CrNi with and without heat treatment were measured and analyzed, as well as the common gathering pipeline steels (X60 and 16Mn). The results indicate that the mechanical properties of the low-nickel alloy steels with heat treatment are superior compared with other test materials, in the sequence as follows: (20CrNi) > (12CrNi) > (08Ni) > 20CrNi > 12CrNi > X60 > 08Ni > 16Mn ("(steel)" indicates the material with heat treatment, and "steel" means without). The weight-loss method was used to test the corrosion rates of the test materials exposed to a simulated oilfield environment for 168 h at 90 degrees C with the CO2 partial pressure and total pressure 2 MPa and 10 MPa, respectively. The systematical characterization with scanning electron microscope and X-ray diffraction displayed the different micro-morphology of corrosion scales formed on the surface of the test materials with FeCO3 as the only detected composition of corrosion product. That the low-nickel alloy steels with heat treatment showed a higher corrosion resistance than those without, mainly resulted from the different microstructures of the matrix, the characteristics of corrosion scale and the composition of elements.

    • BACKGROUND: The blood-based biomarkers are approaching the clinical practice of Alzheimer's disease (AD). Chronic kidney disease (CKD) has a potential confounding effect on peripheral protein levels. It is essential to characterize the impact of renal function on AD markers.METHODS: Plasma phospho-tau181 (P-tau181), and neurofilament light (NfL) were assayed via the Simoa HD-X platform in 1189 dementia-free participants from the Shanghai Aging Study (SAS). The estimated glomerular filter rate (eGFR) was calculated. The association between renal function and blood NfL, P-tau181 was analyzed. An analysis of interactions between various demographic and comorbid factors and eGFR was conducted.RESULTS: The eGFR levels were negatively associated with plasma concentrations of NfL and P-tau181 (B = - 0.19, 95% CI - 0.224 to - 0.156, P < 0.001; B = - 0.009, 95% CI - 0.013 to -0.005, P < 0.001, respectively). After adjusting for demographic characteristics and comorbid diseases, eGFR remained significantly correlated with plasma NfL (B = - 0.010, 95% CI - 0.133 to - 0.068, P < 0.001), but not with P-tau181 (B = - 0.003, 95% CI - 0.007 to 0.001, P = 0.194). A significant interaction between age and eGFR was found for plasma NfL (Pinteraction < 0.001). In participants ≥ 70 years and with eGFR < 60 ml/min/1.73 m2, the correlation between eGFR and plasma NfL was significantly remarkable (B = - 0.790, 95% CI - 1.026 to - 0,554, P < 0.001).CONCLUSIONS: Considering renal function and age is crucial when interpreting AD biomarkers in the general aging population.

    • The effect of windbreak wall types on their windproof effects on trains has been systematically investigated by comparing the flow field around a high-speed train behind three typical windbreak walls. These three typical windbreak walls are widely used along the windy railway lines in China, which include the earth embankment–type windbreak wall (EW), the road cutting–type windbreak wall (RW), and the straight reinforced concrete–type windbreak wall (SW). We compared the time-averaged crosswind-induced flows and aerodynamic performances of high-speed trains behind these three windbreak walls with the same height using numerical simulations. The results revealed that the windbreak wall geometric shape effects on the aerodynamic load coefficients varied according to the type of aerodynamic load and carriage marshalling positions but barely varied with yaw angles. The total drag coefficients of the train in the EW and RW were approximately 50–60% of that in the SW under the two smaller yaw angles and were only 30–40% for the largest yaw angle. For the absolute value of the side force and rolling moments coefficients, the maximum values for the head and middle cars both appeared in the RW, and the corresponding minimum values were obtained in the SW. The maximum and minimum values for the tail car were obtained at the SW and EW, respectively. The maximum of the rolling moment coefficient among three carriages in the SW was approximately 45–60% of that in the EW and only 30–40% of that in the RW. The time-averaged train surface pressure coefficient and flow patterns were similar between the EW and RW, which showed apparent differences from those in the SW. The SW provided a strong blocking effect on the incoming windward airflow and avoided the direct impact on the train. Compared with the RW, the uplifting effect of the EW's windward slope on the incoming flow further reduced the crosswind effect on trains. In addition, the dominating frequency characteristics of the aerodynamic loads were significantly affected by windbreak wall types. These findings provide a systematic understanding of the time-averaged aerodynamics of trains behind these three typical windbreak walls.

    • A novel zinc-based luminescent metal-organic framework (Zn-MOF) has been successfully constructed based on a designed flexible and electron-rich N-involved linker (HL = 4-(bis(4-(pyridin-4-yl) phenyl) amino) benzoic acid). The framework of this Zn-MOF exhibits a 2-fold interpenetrated network which is composed of (3,3)-c sheets. The Zn-MOF has a strong solid state emission at 512 nm. The luminescence signal of the Zn-MOF can be quenched efficiently by trace amounts of electron-deficient nitroaromatics, especially 2,4,6-trinitrophenol (TNP). The quenching constant (K-sv) for TNP is 2.11 x 10(4) M-1, indicating that this framework can be employed as an excellent chemical sensor for identifying and quantifying TNP. This work highlights a strategy for designing a N-involved p-electron-rich enhanced ligand with nucleophilic properties for MOF-based materials as sensors. It also paves the way toward exploring other more efficient MOF materials as sensors for determining electron-deficient nitroaromatics.

    • Purpose: p33ING1b, as a candidate tumour suppressor gene, has been found to be expressed a proportion of oral squamous cell carcinomas (OSCCs), however, its clinicopathological significance is not studied yet. Our aim was to investigate association of p33ING1b expression with clinicopathological variables and particularly interesting new cysteine-histidine rich protein (PINCH) in OSCCs. Methods: p33ING1b expression was immumohistochemically examined in 20 normal oral mucosa specimens and 49 OSCCs. Results: Normal squamous cells showed only p33ING1b nuclear expression (no cytoplasmic expression), with a rate of 90% positive cases. While 24% of OSCCs appeared cytoplasmic expression (11 of them with weak nuclear staining) and the rest tumours (76%) were negative for p33 ING1b. Furthermore, the cases having lymph node metastasis showed a higher frequency of positive cytoplasmic expression than those without metastasis (P = 0.03). The p33ING1b cytoplasmic expression was positively related to PINCH expression (P = 0.04), the cases positive for both proteins had a high rate of the metastasis (P = 0.03).     • Objectives: Since the launch of Global Leadership Initiative on Malnutrition (GLIM), there has been an urgent need to validate the new criteria, especially in patients with cancer. The aim of this study was to evaluate and validate the use of the GLIM criteria in patients with cancer.Method: This multicenter cohort study compared the GLIM with the scored Patient-Generated Subjective Global Assessment (sPG-SGA). The 1-y survival rate, multivariate Cox regression analysis, k-value, sensitivity, specificity, receiver operating characteristic (ROC) curve, and time-dependent ROC analysis were applied to identify the performance of the GLIM.Results: Among the 3777 patients in the study, 50.9% versus 49.1% or 36.3% versus 63.7% of the patients were defined as well-nourished and malnourished by GLIM or sPG-SGA, respectively. GLIM presented moderate consistency (k = 0.54, P < 0.001), fair sensitivity and specificity (70.5 and 88.3%) compared with sPG-SGA. There was no difference in the 1-y survival rate in malnourished patients (76.9 versus 76.4%, P = 0.711), but it was significantly different in well-nourished patients (85.8 versus 90.3%, P < 0.001) between GLIM and sPG-SGA. The above difference was eliminated after omitted nutritional risk screening (NRS)-2002 screening before GLIM (88.1 versus 90.3%, P = 0.078). Omitting NRS-2002 screening before GLIM did not change the 1-y survival rate in well-nourished or malnourished patients by GLIM with NRS-2002 screening (76.9 versus 78.9%, P = 0.099; 85.8% versus 88.1%, P = 0.092) although it significantly raised the rate of malnutrition to 72.5%. The combination of "weight loss and cancer" showed better performance than other combinations.Conclusions: GLIM could be a convenient alternative to sPG-SGA in nutrition assessment for patients with cancer. The combination of "weight loss and cancer" was better than other combinations. Considering the higher risk for malnutrition in patients with cancer, NRS-2002 screening may not be needed before GLIM.

  • Thermal decomposition behaviors of a self-intumescent flame retardant epoxy resin

    • This paper focuses on revealing the thermal decomposition behaviors of a self-intumescent flame retardant (IFR) epoxy (EP) resin (EP/15%APP-Cu2O) employing 12 wt% ammonium polyphosphate (APP) as a halogen-free flame retardant and 3 wt% copper (I) oxide (Cu2O) as char forming rate regulator. Initially, the thermal stability of EP/15%APP-Cu2O was analyzed and compared to virgin EP resin and flame retardant EP (EP/15%APP) containing 15 wt% APP as flame retardant by thermogravimetric analysis test at different heating rates under nitrogen atmosphere. It was shown that the incorporation of APP altered the decomposition pathway of EP and decreased the onset decomposition temperature. Luckily, compared to EP/15%APP, the onset decomposition temperature of EP/15%APP-Cu2O was just slightly reduced from 300.4 to 292.8 degrees C. Then, the thermal degradation kinetics of EP, EP/15%APP and EP/15%APP-Cu2O were further evaluated by Kissinger and Flynn-Wall-Ozawa methods. It was worth noted that the addition of APP or APP-Cu2O enhanced the thermal degradation activation energies of EP, which contributed to the protective effect of the char formation. Particularly, the incorporation of 3 wt% Cu2O significantly decreased the thermal degradation activation energies at the early decomposition stage of EP. This may be the main contribution for intumescent char formation, which resulted in higher fire safety of EP/15%APP-Cu2O compared to EP/15%APP. These information can potentially help to develop alternative IFR systems.

    • Objective: FXYD-3, also known as Mat-8, is a member of the FXYD protein family. It was reported that this protein can associate with and modify the transport properties of Na, K-ATPase, and may play an important role in a variety of physiological and pathological states. This protein is up-regulated in certain types of cancers (such as breast, prostate and pancreatic cancer), but down-regulated in other types of cancers (such as colon and kidney cancer). No study has been performed in gastric cancer; therefore, the aim of this project was to investigate FXYD-3 expression and its clinicopathological significance in gastric adenocarcinoma. Patients and methods: FXYD-3 protein was examined by immunohistochemistry in normal gastric mucous (n = 29) and gastric adenocarcinoma (n = 51), obtained from surgical resection of gastric cancer patients. Results: FXYD-3 protein was present in the cytoplasm of normal gastric epithelial cells or gastric cancer cells. The rate of FXYD-3 strong expression was significantly higher in cancer (51% of 51) than in normal mucosa (10% of 29, X-2=13.210, p andlt; 0.0001). FXYD-3 expressed strongly in ulcerative/infiltrating types of cancers compared to polypoid/fungating ones (X-2 = 5.765, p = 0.016). However, FXYD-3 expression was not correlated with patients gender, age, tumor size, lymph node status and histological grade (p andgt; 0.05). Conclosion: Up-regulated expression of FXYD-3 protein may be involved in tumourgenesis and invasion of gastric adenocarcinoma.
