    • Post hoc interventions rely on having access to certain personal data - such as the gender, age, ethnicity, and sexual orientation of the persons being evaluated - in order to detect and correct for prejudice. This brings these interventions into possible tension with pertinent data protection legislation, which might restrict the processing of said data. We discuss the compatibility of post hoc interventions, more specifically the Generalized Informed Interval Scale Update (GIIU), and the General Data Protection Regulation (GDPR). In particular, we investigate the legality of applying GIIU to datasets which haven't been collected with consent from the data subjects that their data is to be processed by GIIU. We conclude that many such applications are in compliance with the GDPR, but others, specifically those where the processing includes special categories of personal data that is considered sensitive, might not be.

  • Observations of phase changes in monoolein during high viscous injection

    • Serial crystallography of membrane proteins often employs high-viscosity injectors (HVIs) to deliver micrometre-sized crystals to the X-ray beam. Typically, the carrier medium is a lipidic cubic phase (LCP) media, which can also be used to nucleate and grow the crystals. However, despite the fact that the LCP is widely used with HVIs, the potential impact of the injection process on the LCP structure has not been reported and hence is not yet well understood. The self-assembled structure of the LCP can be affected by pressure, dehydration and temperature changes, all of which occur during continuous flow injection. These changes to the LCP structure may in turn impact the results of X-ray diffraction measurements from membrane protein crystals. To investigate the influence of HVIs on the structure of the LCP we conducted a study of the phase changes in monoolein/water and monoolein/buffer mixtures during continuous flow injection, at both atmospheric pressure and under vacuum. The reservoir pressure in the HVI was tracked to determine if there is any correlation with the phase behaviour of the LCP. The results indicated that, even though the reservoir pressure underwent (at times) significant variation, this did not appear to correlate with observed phase changes in the sample stream or correspond to shifts in the LCP lattice parameter. During vacuum injection, there was a three-way coexistence of the gyroid cubic phase, diamond cubic phase and lamellar phase. During injection at atmospheric pressure, the coexistence of a cubic phase and lamellar phase in the monoolein/water mixtures was also observed. The degree to which the lamellar phase is formed was found to be strongly dependent on the co-flowing gas conditions used to stabilize the LCP stream. A combination of laboratory-based optical polarization microscopy and simulation studies was used to investigate these observations.

  • A model of an induction-stirred ladle accounting for slag and surface deformation

    • A 3 dimensional 2-phase model of an induction stirred ladle has been developed. The model is based on fundamental transport equations and includes the solution of steel- and slag-phase. Predicted velocities are in very good agreement with experimental data. Predicted velocities and turbulent kinetic energy dissipation from the model are shown to have linear relationships with input stirring force for different cylindrical ladles. Linear equations forthese relations are presented. Further calculations have shown that these linear equations also are able to predict velocities for a conical ladle. © 1999 ISIJ.

    • A fully 3-dimensional model for an inductively-stirred ladle is presented. Since the fluid flow during induction stirring is not symmetric with respect to the centre of the ladle, this model gives a more accurate description of the real stirring conditions than more traditional models that use cylindrical or polar co-ordinates. Typical output from the model are steel velocities, turbulent kinetic energy and dissipation of turbulent kinetic energy. The calculated steel velocities agree fairly well with experimental data. © Munksgaard, 1997.

    • A three-dimensional simulation model coupling heating and induction stirring in an ASEA-SKF ladle furnace was developed. Data of the heat transfer from the area to the steel bath were predicted in a separate model and included as boundary conditions in a ladle model. The are model considers the contributions of heat transferred by of each of the following mechanisms: radiation, convection, condensation and energy transported by electrons. Predictions were made to simulate the change of temperature distribution in the ladle during simultaneous heating with electrodes and stirring by induction. A first attempt was made to compare the predictions with measured temperatures from a 100 t ASEA-SKF ladle. The agreement was found to be fairly good when heat-flux data for a 25 cm are length were used as input to the ladle model. This indicates that the model can be used for more in-depth studies of the effects of heating for ladles that are inductively stirred.

    • One may begin describing the distance course “IT and Learning- rehabilitation engineering” by saying that it was a great success. Looking back, it is evident that our experience from two years of working with Internet-based teaching prior to the present course was absolutely vital to its favorable outcome. It is difficult to switch from classroom-based to Internet-based teaching. There are mechanisms in Internet-based teaching that must be learned by trial and error. Otherwise, the discrepancy between the official course description and how the course actually turns out may become too great. It is not possible to form a clear overall conception of the course by looking at its different components. The most interesting aspect of the course is what happens at the intersection of technology and educational methodology, and the choice of tools is very important to the outcome of this meeting.

    • Denna rapport handlar så mycket om "Internet inifrån" att titeln gav sig själv – det var bara det att "Internet Inifrån"-titeln redan är upptagen. Den finns sedan i fjol som beteckning på en LTH-kurs som ges i samarbete med företaget AXIS. Denna har ett renodlat inifrånperspektiv ur teknisk synvinkel. I vår kurs kommer inifrånperspektiven däremot från dem som lär sig med hjälp av Internet. Man är inne på nätet, man lär sig via nätet av sina lärare, av varandra, ja, även av sig själv, för det är så mycket i den egna personen som kommer fram, som ploppar upp, när man sitter där och arbetar just med Internet Inifrån. Nog om detta - vi valde att beteckna våra rapport med Internet inifrån två (jfr Gudfadern 2…) för att samtidigt markera att det finns flera inifrån-perspektiv på Internet. Just detta att ta användarens perspektiv, speciellt den funktionshindrade användarens, på tekniken och fördjupa sig i detta snarare än i tekniken i sig är ju f ö det som är Certecs signum totalt sett.

    • Alkyl glycoside surfactants with elongated carbohydrate chains are useful in different applications due to their improved biocompatibility. Cyclodextrin glucanotransferases can catalyse the elongation process through the coupling reaction. However, due to the presence of a hydrophobic tail, the interaction between an alkyl glycoside acceptor and the active site residues is weaker than the interaction with maltooligosaccharides at the corresponding site. Here we report the mutations of F197, G263 and E266 near the acceptor subsites in the CGTase CspCGT13 from Carboxydocella sp. The results showed that substitutions of both F197 and G263 were important for the binding of acceptor substrate dodecyl maltoside during coupling reaction. The double mutant F197Y/G263A showed enhanced coupling activity and displayed a 2-fold increase of the primary coupling product using γ-cyclodextrin as donor when compared to wildtype CspCGT13. Disproportionation activity was also reduced, which was also the case for another double mutant (F197Y/E266A) that however not showed the corresponding increase in coupling. A triple mutant F197Y/G263A/E266A maintained the increase in primary coupling product (1.8-fold increase) using dodecyl maltoside as acceptor, but disproportionation was approximately at the same level as in the double mutants. In addition, hydrolysis of starch was slightly increased by the F197Y and G263A substitutions, indicating that interactions at both positions influenced the selectivity between glycosyl and alkyl moieties.

    • This study addressed the effects of blocking spontaneous sniffing during olfactory imagery. A group of subjects (n=40) who scored high in olfactory focus and imagery ability rated the vividness in olfactory and visual imagery content under conditions of blocked sniffing, blocked vision, and a nonblocked control. The imagery stimuli consisted of 90 common words that could represent either an odor or a visual object. Blocked sniffing was expected to impair olfactory imagery vividness, but since visual imagery entails eye movements, which was not affected by the ""blocked vision"" manipulation, visual imagery ratings were effectively used as a placebo control. Confirming our hypotheses, the results showed that preventing sniffing resulted in a selectively poorer olfactory but not visual vividness, whereas blocked vision showed no effect on either the visual or olfactory vividness ratings. These observations confirm that sensorimotor activity is an important aspect for the quality of evoked olfactory images.

    • A compelling body of international research demonstrates that a positive, supportive teacher-student relationship is essential for students’ development, especially for at-risk students. In this article, the educator’s ability to build such relationships is discussed in terms of ‘relational competence’. Special educators’ relational competence is a largely unexplored topic. This article contributes by reporting on an interview study with 21 experienced special educators. The study focuses on the educators’ perceptions of their relationships and relational competence. Two research questions (RQs) are explored: (1) What is the role of social relationships in (successful) work as a special educator? and (2) How is the relational competence of special educators realised in practice? The results are presented in themes and subthemes, and are supported by significant quotes. Regarding RQ 1, the study shows that the informants perceive positive social relationships as fundamental in their work and relational competence as particularly important in their profession. The results for RQ 2 show that relational competence is realised by an accepting attitude in the ‘here and now’; by finding a personal connection to the student; and by building trusting relationships over a longer period of time. Comments on implications for special education teacher preparation are provided.

The study has three parts. In the theoretical part, a conceptualization of teachers’ relational competence using Thomas Scheff’s theory of interpersonal relationships is outlined. In the empirical (pilot) part, a methodology for prompting preservice teachers’ analyses of teacher–student relationships is described, as well as a thematic analysis of their responses. The theoretical conceptualization is then used together with the empirical data in the third part, in order to identify development needs of preservice teachers in terms of relational competence. According to the conceptualization, relational competence includes three sub-concepts: communicative, differentiation, and socio-emotional competence. From the analysis of preservice students’ texts, the article identifies development needs in relation to the three sub-concepts. The concluding discussion focuses on lessons learned from the study regarding how to promote teacher students’ relational competence.

    • Research shows that educators’ capability to build positive and supportive relationships is critical for educational success, particularly for at-risk students. The purpose of this article is to discuss how special educators – “speciallärare” (SL) and “specialpedagoger” (SP) – perceive relational competence working with their students’ peer relationships and with the students’ parents. The article presents an interview study of 21 experienced SL and SP. The findings indicate that SL and SP perceive relational competence as an extensive, complex, and advanced work involving short- and long-term dimensions, formal and informal social situations, and with instrumental and immanent purposes. Overall, SL and SP highlight relational competence through cooperation in which the other party is directly involved. Moreover, the thematic analysis shows that SL and SP perceive relational competence regarding peer relationships as manifested through (i) “social training”, which is carried out spontaneously and by arrangement, and (ii) “relational mediation”, which has a direct or an indirect form. In relation to students’ parents, SL and SP perceive relational competence as manifested in direct interaction, which includes two aspects: (i) “guidance” and (ii) “confirmation”. The article suggests that relational competence, as seen from the perspectives of SP and SL, can be understood in terms of method and encounters, dimensions that are important to notice in special education teacher training, as well as in further research.

    • We present experimental results and simulations on a simple method for tunable particle separation based on a combination of Deterministic Lateral Displacement (DLD) and Insulator Based Dielectrophoresis (I-DEP). Rather than deriving its tunability from its elastic properties[1], our present device uses an applied AC field to perturb the particle trajectories in the pressure-driven flow and is thereby capable of scanning the critical size over a range of factor two. Potential benefits include: extended dynamic range, facilitated fabrication and less clogging for given particle sizes, and combination of the precision afforded by DLD with the versatility of DEP.

    • We report the use of dielectrophoresis (DEP) to achieve tunability, improve dynamic range and open up for the separation of particles with regard to parameters other than hydrodynamic size in deterministic lateral displacement (DLD) devices. DLD devices have been shown capable of rapidly and continuously separating micrometer sized plastic spheres by size with a resolution of 20 nm in diameter and of being able to handle the separation of biological samples as wide ranging as bacterial artificial chromosomes and blood cells. DEP, while not exhibiting the same resolution in size separation as DLD, has the benefit of being easy to tune and can, by choosing the frequency, be used to probe a variety of particle properties. By combining DLD and DEP we open up for the advantages, while avoiding the drawbacks, of the two techniques. We present a proof of principle in which the critical size for separation of polystyrene beads is tuned in the range 2-6 microm in a single device by the application of moderate (100 V cm(-1)), low frequency (100 Hz) AC electric fields. The behaviour of the device was further investigated by performing simulations of particle trajectories, the results of which were in good qualitative agreement with experiments, indicating the potential of the method for tunable, high-resolution separations with respect to both size and polarisability.

  • Geometric control of diffusing elements on InAs semiconductor surfaces via metal contacts

    • Local geometric control of basic synthesis parameters, such as elemental composition, is important for bottom-up synthesis and top-down device definition on-chip but remains a significant challenge. Here, we propose to use lithographically defined metal stacks for regulating the surface concentrations of freely diffusing synthesis elements on compound semiconductors. This is demonstrated by geometric control of Indium droplet formation on Indium Arsenide surfaces, an important consequence of incongruent evaporation. Lithographic defined Aluminium/Palladium metal patterns induce well-defined droplet-free zones during annealing up to 600 °C, while the metal patterns retain their lateral geometry. Compositional and structural analysis is performed, as well as theoretical modelling. The Pd acts as a sink for free In atoms, lowering their surface concentration locally and inhibiting droplet formation. Al acts as a diffusion barrier altering Pd’s efficiency. The behaviour depends only on a few basic assumptions and should be applicable to lithography-epitaxial manufacturing processes of compound semiconductors in general.

    • Purpose: To compare the effect of therapy with anastrozole versus a combination of fulvestrant and anastrozole in women in first relapse of endocrine-responsive breast cancer. Patients and Methods: Postmenopausal women, or premenopausal women receiving a gonadotropin-releasing hormone agonist, with estrogen receptor– and/or progesterone receptor–positive disease at first relapse after primary treatment of localized disease were open-label randomly assigned to a fulvestrant loading dose (LD) regimen followed by monthly injection plus 1 mg of anastrozole daily or to 1 mg of anastrozole daily alone. The primary end point was time to progression (TTP). Results: In all, 514 women were randomly assigned to fulvestrant plus anastrozole (experimental arm; n = 258) or anastrozole (standard arm; n = 256). Approximately two thirds had received adjuvant antiestrogens, but only eight individuals had received an aromatase inhibitor. Median TTP was 10.8 and 10.2 months in the experimental versus standard arm, respectively (hazard ratio [HR] = 0.99; 95% CI, 0.81 to 1.20; P = .91); median overall survival was 37.8 and 38.2 months, respectively (HR = 1.0; 95% CI, 0.76 to 1.32; P = 1.00). Incidences of prespecified adverse events (AEs) were similar. Hot flashes were more common in the experimental arm: 63 patients (24.6%) versus 35 patients (13.8%) in the standard arm (P = .0023). Death owing to AEs was reported in 11 (4.3%) and five patients (2.0%) in the experimental versus standard arm, respectively. Conclusion: Fulvestrant (250 mg + LD regimen) in combination with anastrozole offered no clinical efficacy advantage over anastrozole monotherapy in this population of individuals with a relatively high proportion of previous adjuvant antiestrogen exposure.

  • Drivers of changing urban flood risk : A framework for action

    • This study focuses on drivers for changing urban flood risk. We suggest a framework for guiding climate change adaptation action concerning flood risk and manageability in cities. The identified key drivers of changing flood hazard and vulnerability are used to provide an overview of each driver's impact on flood risk and manageability at the city level. We find that identified drivers for urban flood risk can be grouped in three different priority areas with different time horizon. The first group has high impact but is manageable at city level. Typical drivers in this group are related to the physical environment such as decreasing permeability and unresponsive engineering. The second group of drivers is represented by public awareness and individual willingness to participate and urbanization and urban sprawl. These drivers may be important and are manageable for the cities and they involve both short-term and long-term measures. The third group of drivers is related to policy and long-term changes. This group is represented by economic growth and increasing values at risk, climate change, and increasing complexity of society. They have all high impact but low manageability. Managing these drivers needs to be done in a longer time perspective, e.g., by developing long-term policies and exchange of ideas.

The concept of “audience” steers attention to processes of communication between those who seek to shape legitimacy perceptions and those whose perceptions would be shaped. (De)legitimation practices may have different implications for different audiences, and the chapter suggests that audiences play an active part in the performance of (de)legitimation in global governance. Two distinctions are introduced for classifying relationships between global governance institutions and the multitude of actors that may hold legitimacy beliefs about them, namely between constituencies and observers and between targeted and self-appointed audiences. Such categories can be used for the purpose of studying patterns and variation in legitimacy beliefs across actors.

    • AIM: To study the association between inflammatory bowel disease (IBD) and genetic variations in eosinophil protein X (EPX) and eosinophil cationic protein (ECP). METHODS: DNA was extracted from ethylene diamine tetraacetic acid blood of 587 patients with Crohn's disease (CD), 592 with ulcerative colitis (UC) and 300 healthy subjects. The EPX405 (G > C, rs2013109), ECP434 (G > C, rs2073342) and ECP562 (G > C, rs2233860) gene polymorphisms were analysed, by the 5'-nuclease allelic discrimination assay. For determination of intracellular content of EPX and ECP in granulocytes, 39 blood samples was collected and extracted with a buffer containing cetyltrimethylammonium bromide. The intracellular content of EPX was analysed using an enzyme-linked immunosorbent assay. The intracellular content of ECP was analysed with the UniCAP (R) system as described by the manufacturer. Statistical tests for calculations of results were chi(2) test, Fisher's exact test, ANOVA, Student-Newman-Keuls test, and Kaplan-Meier survival curve with Log-rank test for trend, the probability values of P < 0.05 were considered statistically significant. RESULTS: The genotype frequency for males with UC and with an age of disease onset of >= 45 years (n = 57) was for ECP434 and ECP562, GG = 37%, GC = 60%, CC = 4% and GG = 51%, GC = 49%, CC = 0% respectively. This was significantly different from the healthy subject's genotype frequencies of ECP434 (GG = 57%, GC = 38%, CC = 5%; P = 0.010) and ECP562 (GG = 68%, GC = 29 /0,CC = 3%; P = 0.009). The genotype frequencies for females, with an age of disease onset of >= 45 years with CD (n = 62), was for the ECP434 and ECP562 genotypes GG = 37%, GC = 52%, CC = 11% and GG = 48%, GC = 47% and CC = 5% respectively. This was also statistically different from healthy controls for both ECP434 (P = 0.010) and ECP562 (P = 0.013). The intracellular protein concentration of EPX and ECP was calculated in mu g/10(6) eosinophils and then correlated to the EPX 405 genotypes. The protein content of EPX was highest in the patients with the CC genotype of EPX405 (GG = 4.65, GC = 5.93, and CC = 6.57) and for ECP in the patients with the GG genotype of EPX405 (GG = 2.70, GC = 2.47 and CC = 1.90). ANOVA test demonstrated a difference in intracellular protein content for EPX (P = 0.009) and ECP (P = 0.022). The age of disease onset was linked to haplotypes of the EPX405, ECP434 and ECP562 genotypes. Kaplan Maier curve showed a difference between haplotype distributions for the females with CD (P = 0.003). The highest age of disease onset was seen in females with the EPX405CC, ECP434GC, ECP562CC haplotype (34 years) and the lowest in females with the EPX405GC, ECP434GC, ECP562GG haplotype (21 years). For males with UC there was also a difference between the highest and lowest age of the disease onset (EPX405CC, ECP434CC, ECP562CC, mean 24 years vs EPX405GC, ECP434GC, ECP562GG, mean 34 years, P = 0.0009). The relative risk for UC patients with ECP434 or ECP562-GC/CC genotypes to develop dysplasia/cancer was 2.5 (95%CI: 1.2-5.4, P = 0.01) and 2.5 (95%CI: 1.1-5.4, P = 0.02) respectively, compared to patients carrying the GG-genotypes. CONCLUSION: Polymorphisms of EPX and ECP are associated to IBD in an age and gender dependent manner, suggesting an essential role of eosinophils in the pathophysiology of IBD.

    • We investigated the sequential protein expression in gingival crevicular fluid samples during the induction (I) and resolution (R) of experimental gingivitis. Periodontally and systemically healthy volunteers (n = 20) participated in a three-week experimental gingivitis protocol, followed by debridement and two weeks of regular plaque control. Gingival crevicular fluid (GCF) samples were collected at baseline, Day 7, 14, and 21 (induction; I-phase), and at Day 21, 25, 30, and 35 (resolution; R-phase). Liquid chromatography-tandem mass spectrometry (LC-MS/MS) for label-free quantitative proteomics was applied. A total of 287 proteins were identified including 254 human, 14 bacterial, 12 fungal, and 7 yeast proteins. Ontology analysis revealed proteins primarily involved in cytoskeletal rearrangements, immune response, antimicrobial function, protein degradation, and DNA binding. There was considerable variation in the number of proteins identified, both among subjects and within subjects across time points. After pooling of samples between subjects at each time point, the levels of 59 proteins in the I-phase and 73 proteins in the R-phase were quantified longitudinally. Our data demonstrate that LC-MS/MS label-free quantitative proteomics is valuable in the assessment of the protein content of the GCF and can facilitate a better understanding of the molecular mechanisms involved in the induction and resolution of plaque-induced gingival inflammation in humans.

Opportunistically collected species observations contributed by volunteer reporters are increasingly available for species and regions for which systematically collected data are not available. However, it is unclear if they are suitable to produce reliable habitat suitability models (HSMs), and hence if the species-habitat relationships found and habitat suitability maps produced can be used with confidence to advice conservation management and address basic and applied research questions.2. We evaluated HSMs with opportunistically collected observations against HSMs with systematically collected observations. We enhanced the opportunistically collected presence-only data by adding inferred species absences. To obtain inferred absences, we asked individual reporters about their identification skills and if they reported certain species consistently and combined this information with their observations. We evaluated several HSM methods using a forest bird species, Siberian jay (Perisoreus infaustus), in Sweden: logistic regression with inferred absences, two versions of MaxEnt, a model combining presence-absence with presence-only observations and a Bayesian site-occupancy-detection model.3. All HSM methods produced nationwide habitat suitability maps of Siberian jay that agreed well with systematically collected observations (AUC: 086-0.88) and were very similar to a habitat suitability map produced from the HSM with systematically collected observations (Spearman rho: 0.94-0.98). At finer geographical scales there were differences among methods.4. At finer scale, the resulting habitat suitability maps from logistic regression with inferred absences agreed better with results from systematically collected observations than other methods. The species-habitat relationships found with logistic regression also agreed well with those found from systematically collected data and with prior expectations based on the species ecology.5. Synthesis and application. For many regions and species, systematically collected data are not available. By using inferred absences from high-quality, opportunistically collected contributions of few very active reporters in logistic regression we obtained HSMs that produced results similar to those from a systematic survey. Adding high-quality inferred absences to opportunistically collected data is likely possible for many less common species across various organism groups. Well-performing HSMs are important to facilitate applications such as spatial conservation planning and prioritization, monitoring of invasive species, understanding species habitat requirements or climate change studies.

    • Treatment of high-risk childhood neuroblastoma is a clinical challenge hampered by a lack of reliable neuroblastoma mouse models for preclinical drug testing. We have previously established invasive and metastasising patient-derived orthotopic xenografts (PDXs) from high-risk neuroblastomas that retained the genotypes and phenotypes of patient tumours. Given the important role of the tumour microenvironment in tumour progression, metastasis, and treatment responses, here we analysed the tumour microenvironment of five neuroblastoma PDXs in detail. The PDXs resembled their parent tumours and retained important stromal hallmarks of aggressive lesions including rich blood and lymphatic vascularisation, pericyte coverage, high numbers of cancer-associated fibroblasts, tumour-associated macrophages, and extracellular matrix components. Patient-derived tumour endothelial cells occasionally formed blood vessels in PDXs; however, tumour stroma was, overall, of murine origin. Lymphoid cells and lymphatic endothelial cells were found in athymic nude mice but not in NSG mice; thus, the choice of mouse strain dictates tumour microenvironmental components. The murine tumour microenvironment of orthotopic neuroblastoma PDXs reflects important hallmarks of aggressive and metastatic clinical neuroblastomas. Neuroblastoma PDXs are clinically relevant models for preclinical drug testing.

    • The high-resolution Sentinel-2 data potentially enable the estimation of gross primary productivity (GPP) at finer spatial resolution by better capturing the spatial variation in a heterogeneous landscapes. This study investigates the potential of 10 m resolution reflectance from the Sentinel-2 Multispectral Instrument to improve the accuracy of GPP estimation across Nordic vegetation types, compared with the 250 m and 500 m resolution reflectance from the Moderate Resolution Imaging Spectroradiometer (MODIS). We applied linear regression models with inputs of two-band enhanced vegetation index (EVI2) derived from Sentinel-2 and MODIS reflectance, respectively, together with various environmental drivers to estimate daily GPP at eight Nordic eddy covariance (EC) flux tower sites. Compared with the GPP from EC measurements, the accuracies of modelled GPP were generally high (R-2 = 0.84 for Sentinel-2; R-2 = 0.83 for MODIS), and the differences between Sentinel-2 and MODIS were minimal. This demonstrates the general consistency in GPP estimates based on the two satellite sensor systems at the Nordic regional scale. On the other hand, the model accuracy did not improve by using the higher spatial-resolution Sentinel-2 data. More analyses of different model formulations, more tests of remotely sensed indices and biophysical parameters, and analyses across a wider range of geographical locations and times will be required to achieve improved GPP estimations from Sentinel-2 satellite data.

  • Comparing efficacy and safety in catheter ablation strategies for atrial fibrillation: a network meta-analysis

    • Background: There is no consensus on the most efficient catheter ablation (CA) strategy for patients with atrial fibrillation (AF). The objective of this study was to compare the efficacy and safety of different CA strategies for AF ablation through network meta-analysis (NMA). Methods: A systematic search of PubMed, Web of Science, and CENTRAL was performed up to October 5th, 2020. Randomized controlled trials (RCT) comparing different CA approaches were included. Efficacy was defined as arrhythmia recurrence after CA and safety as any reported complication related to the procedure during a minimum follow-up time of 6 months. Results: In total, 67 RCTs (n = 9871) comparing 19 different CA strategies were included. The risk of recurrence was significantly decreased compared to pulmonary vein isolation (PVI) alone for PVI with renal denervation (RR: 0.60, CI: 0.38-0.94), PVI with ganglia-plexi ablation (RR: 0.62, CI: 0.41-0.94), PVI with additional ablation lines (RR: 0.8, CI: 0.68-0.95) and PVI in combination with bi-atrial modification (RR: 0.32, CI: 0.11-0.88). Strategies including PVI appeared superior to non-PVI strategies such as electrogram-based approaches. No significant differences in safety were observed. Conclusions: This NMA showed that PVI in combination with additional CA strategies, such as autonomic modulation and additional lines, seem to increase the efficacy of PVI alone. These strategies can be considered in treating patients with AF, since, additionally, no differences in safety were observed. This study provides decision-makers with comprehensive and comparative evidence about the efficacy and safety of different CA strategies.

    • Introduction Atrial fibrillation (AF) is the most common sustained arrhythmia. Catheter ablation (CA) of AF is an increasingly offered therapeutic approach, primary to relieve AF-related symptoms. Despite the development of new ablation approaches, there is no consensus regarding the most efficient ablation strategy. The objective of this network meta-analysis (NMA) is to compare the efficacy and safety of all different CA approaches for the treatment of patients with paroxysmal (PAF) and non-PAF (non-PAF). Methods and analysis We will perform a systematic search to identify randomised controlled trials of different CA approaches for the treatment of PAF and non-PAF, through the final search date of 1 March 2020. Information sources will include major bibliographic databases (MEDLINE, Web of Science and CENTRAL) and clinical trial registries. Our primary outcomes will be the efficacy (recurrence-free survival) and safety of different CA approaches for the treatment of AF. Secondary outcomes will be all-cause mortality and procedural time. An NMA will be performed to determine the relative effects of different catheter ablation approaches (such as pulmonary vein isolation alone or in combination with ablation lines, ablation of complex fractionated atrial electrograms, etc). In PAF, a separate analysis will be performed including different energy sources (such as radiofrequency, cryogenic and laser energy). Risk of bias assessment and sensitivity analyses will be conducted to assess the robustness of the findings to potential bias. Ethics and dissemination No ethical approval will be needed because data are collected from previous studies. The results will be presented through peer-review journals and conference presentation. Efficacy was defined as lack of arrhythmia recurrence after CA and safety as any reported complication related to the procedure during a minimum follow-up time of six months. In total, 43 RCTs comparing 11 different CA strategies involving 6701 patients were included. The risk of recurrence was significantly decreased in comparison with PVI with radiofrequency only for the following treatments: PVI with adjuvant ablation (RR: 0.79, CI: 0.65-0.97) and PVI with sympathetic modulation (RR: 0.64, CI: 0.46-0.88). However, PVI with radiofrequency was superior to non-PVI strategies (RR: 1.65, CI: 1.2-2.26). No statistically significant difference was found in safety between different CA strategies. Concerning different PVI strategies, no difference was observed either in efficacy or in safety between tested strategies. This NMA suggests that different PVI strategies are generally similar in terms of efficacy, while PVI with additional ablation or sympathetic modulation may be more effective than PVI alone. This study provides decision-makers with insights into the efficacy and safety of different CA strategies.

    • Strong negative reactions, physical symptoms, and behavioral disruptions due to environmental odors are common in the adult population. We investigated relationships among such environmental chemosensory responsivity (CR), personality traits, affective states, and odor perception. Study 1 showed that CR and neuroticism were positively correlated in a sample of young adults (n = 101), suggesting that persons high in neuroticism respond more negatively to environmental odors. Study 2 explored the relationships among CR, noise responsivity (NR), neuroticism, and odor perception (i.e., pleasantness and intensity) in a subset of participants (n = 40). High CR was associated with high NR. Regression analyses indicated that high CR predicted higher odor intensity ratings and low olfactory threshold (high sensitivity) predicted lower pleasantness ratings. However, neuroticism was not directly associated with odor ratings or thresholds. Overall, the results suggest that CR and odor thresholds predict perceptual ratings of odors and that high CR is associated with nonchemosensory affective traits.

    • Water and ice are routinely studied with X-rays to reveal their diverse structures and anomalous properties. We employ a hybrid collisional-radiative/molecular dynamics method to explore how femtosecond X-ray pulses interact with hexagonal ice. We find that ice makes a phase transition into a crystalline plasma where its initial structure is maintained up to tens of femtoseconds. The ultrafast melting process occurs anisotropically, where different geometric configurations of the structure melt on different time scales. The transient state and anisotropic melting of crystals can be captured by X-ray diffraction, which impacts any study of crystalline structures probed by femtosecond X-ray lasers.

    • Objective: To compare the effectiveness of education (ED) plus home exercise (HE) and ED plus supervised exercise (SE) according to information provided by the Better Management of Patients With Osteoarthritis (BOA) Registry, a nationally implemented rehabilitation program for patients with hip and knee osteoarthritis (OA). In addition, we investigated whether or not the effect of the treatments differed based on the joint affected by OA (hip versus knee). Methods: We included 38,030 participants from the BOA Registry with knee or hip OA who were treated with either ED, HE, or SE. The effect of the 3 treatment options on the pain intensity reduction (range 0–10) immediately postintervention and at 12 months was estimated using a mixed-effects model adjusted for age, sex, body mass index, affected joint (hip or knee), pain at baseline, comorbidity, and level of education. Results: The participants undergoing HE or SE experienced a greater pain reduction compared to participants who received ED, both after the treatment (group mean change for ED –0.91 [95% confidence interval (95% CI) –1.15, –0.68], for HE –1.06 [95% CI –1.10, –1.01], and for SE –1.12 [95% CI –1.15, –1.08]) and at 12 months (group mean change for ED –0.58 [95% CI –0.87, –0.30], for HE –0.82 [95% CI –0.87, –0.76], and for SE –0.82 [95% CI –0.86, –0.77]). Patients with knee OA who underwent HE or SE improved more compared to patients with hip OA at both follow-ups. Conclusion: In primary care, HE and SE lead to similar reductions in pain intensity but are more effective than ED alone. In addition, people with knee OA benefit more from HE and SE than people with hip OA.

    • Cancer immunotherapy can result in durable tumor regressions in some patients. However, patients who initially respond often experience tumor progression. Here, we report mechanistic evidence of tumoral immune escape in an exemplary clinical case: a patient with metastatic melanoma who developed disease recurrence following an initial, unequivocal radiologic complete regression after T-cell–based immunotherapy. Functional cytotoxic T-cell responses, including responses to one mutant neoantigen, were amplified effectively with therapy and generated durable immunologic memory. However, these immune responses, including apparently effective surveillance of the tumor mutanome, did not prevent recurrence. Alterations of the MHC class I antigen-processing and presentation machinery (APM) in resistant cancer cells, but not antigen loss or impaired IFNγ signaling, led to impaired recognition by tumor-specific CD8þ T cells. Our results suggest that future immunotherapy combinations should take into account targeting cancer cells with intact and impaired MHC class I–related APM. Loss of target antigens or impaired IFNγ signaling does not appear to be mandatory for tumor relapse after a complete radiologic regression. Personalized studies to uncover mechanisms leading to disease recurrence within each individual patient are warranted.

    • This paper presents an empirical study on the weights of neural networks, where we interpret each model as a point in a high-dimensional space – the neural weight space. To explore the complex structure of this space, we sample from a diverse selection of training variations (dataset, optimization procedure, architecture,etc.) of neural network classifiers, and train a large number of models to represent the weight space. Then, we use a machine learning approach for analyzing and extracting information from this space. Most centrally, we train a number of novel deep meta-classifiers withthe objective of classifying different properties of the training setup by identifying their footprints in the weight space. Thus, the meta-classifiers probe for patterns induced by hyper-parameters, so that we can quantify how much, where, and when these are encoded through the optimization process. This provides a novel and complementary view for explainable AI, and we show how meta-classifiers can reveal a great deal of information about the training setup and optimization, by only considering a small subset of randomly selected consecutive weights. To promote further research on the weight space, we release the neural weight space (NWS) dataset – a collection of 320K weightsnapshots from 16K individually trained deep neural networks.

  • Model-invariant Weight Distribution Descriptors for Visual Exploration of Neural Networks en Masse

    • We present a neural network representation which can be used for visually analyzing the similarities and differences in a large corpus of trained neural networks. The focus is on architecture-invariant comparisons based on network weights, estimating similarities of the statistical footprints encoded by the training setups and stochastic optimization procedures. To make this possible, we propose a novel visual descriptor of neural network weights. The visual descriptor considers local weight statistics in a model-agnostic manner by encoding the distribution of weights over different model depths. We show how such a representation can extract descriptive information, is robust to different parameterizations of a model, and is applicable to different architecture specifications. The descriptor is used to create a model atlas by projecting a model library to a 2D representation, where clusters can be found based on similar weight properties. A cluster analysis strategy makes it possible to understand the weight properties of clusters and how these connect to the different datasets and hyper-parameters used to train the models.

    • The aim of this work is to develop and test a method for generation of information on vegetation dynamics from high-spatial resolution data, such as Sentinel-2. In order to accomplish this, Sentinel-2 data were simulated from existing SPOT HRG and HRVIR scenes over Sweden. We used TIMESAT, a well-tested computer package for generating smooth seasonal profiles and generation of seasonality parameters, like start and end, length, amplitude, integrated values, seasonal maximum, derivatives, etc. The processing works on a pixel-by-pixel basis and is resistant to clouds and noise. Data gaps are handled, and quality information can be included to increase the fidelity of the fits. The pilot study demonstrated that TIMESAT was successful in fitting smooth model functions to the data, and generating seasonality parameters for the test area at 10 × 10 m resolution. We conclude that TIMESAT will be useful for generating vegetation dynamics data from high-spatial resolution data such as Sentinel-2. The smooth seasonal profiles will be extremely useful for driving high-resolution biophysical vegetation models, and the seasonality parameters will be excellent for change detection, and for studying trends in vegetation productivity and seasonality.

    • Background: Severe COVID-19 lung disease exhibits a high degree of spatial and temporal heterogeneity, with different histological features coexisting within a single individual. It is important to capture the disease complexity to support patient management and treatment strategies. We provide spatially decoded analyses on the immunopathology of diffuse alveolar damage (DAD) patterns and factors that modulate immune and structural changes in fatal COVID-19. Methods: We spatially quantified the immune and structural cells in exudative, intermediate, and advanced DAD through multiplex immunohistochemistry in autopsy lung tissue of 18 COVID-19 patients. Cytokine profiling, viral, bacteria, and fungi detection, and transcriptome analyses were performed. Findings: Spatial DAD progression was associated with expansion of immune cells, macrophages, CD8+ T cells, fibroblasts, and (lymph)angiogenesis. Viral load correlated positively with exudative DAD and negatively with disease/hospital length. In all cases, enteric bacteria were isolated, and Candida parapsilosis in eight cases. Cytokines correlated mainly with macrophages and CD8+T cells. Pro-coagulation and acute repair were enriched pathways in exudative DAD whereas intermediate/advanced DAD had a molecular profile of elevated humoral and innate immune responses and extracellular matrix production. Interpretation: Unraveling the spatial and molecular immunopathology of COVID-19 cases exposes the responses to SARS-CoV-2-induced exudative DAD and subsequent immune-modulatory and remodeling changes in proliferative/advanced DAD that occur side-by-side together with secondary infections in the lungs. These complex features have important implications for disease management and the development of novel treatments.   • Heterogeneous Ice Growth in Micron-Sized Water Droplets Due to Spontaneous Freezing

    • Understanding how ice nucleates and grows into larger crystals is of crucial importance for many research fields. The purpose of this study was to shed light on the phase and structure of ice once a nucleus is formed inside a metastable water droplet. Wide-angle X-ray scattering (WAXS) was performed on micron-sized droplets evaporatively cooled to temperatures where homogeneous nucleation occurs. We found that for our weak hits ice grows more cubic compared to the strong hits that are completely hexagonal. Due to efficient heat removal caused by evaporation, we propose that the cubicity of ice at the vicinity of the droplet’s surface is higher than for ice formed within the bulk of the droplet. Moreover, the Bragg peaks were classified based on their geometrical shapes and positions in reciprocal space, which showed that ice grows heterogeneously with a significant population of peaks indicative of truncation rods and crystal defects. Frequent occurrences of the (100) reflection with extended in-planar structure suggested that large planar ice crystals form at the droplet surface, then fracture into smaller domains to accommodate to the curvature of the droplets. Planar faulting due to misaligned domains would explain the increased cubicity close to the droplet surface.
