  • To which world regions does the valence-dominance model of social perception apply?

    • Over the past 10 years, Oosterhof and Todorov's valence-dominance model has emerged as the most prominent account of how people evaluate faces on social dimensions. In this model, two dimensions (valence and dominance) underpin social judgements of faces. Because this model has primarily been developed and tested in Western regions, it is unclear whether these findings apply to other regions. We addressed this question by replicating Oosterhof and Todorov's methodology across 11 world regions, 41 countries and 11,570 participants. When we used Oosterhof and Todorov's original analysis strategy, the valence-dominance model generalized across regions. When we used an alternative methodology to allow for correlated dimensions, we observed much less generalization. Collectively, these results suggest that, while the valence-dominance model generalizes very well across regions when dimensions are forced to be orthogonal, regional differences are revealed when we use different extraction methods and correlate and rotate the dimension reduction solution.   • Social and moral psychology of COVID-19 across 69 countries

    • The COVID-19 pandemic has affected all domains of human life, including the economic and social fabric of societies. One of the central strategies for managing public health throughout the pandemic has been through persuasive messaging and collective behaviour change. To help scholars better understand the social and moral psychology behind public health behaviour, we present a dataset comprising of 51,404 individuals from 69 countries. This dataset was collected for the International Collaboration on Social & Moral Psychology of COVID-19 project (ICSMP COVID-19). This social science survey invited participants around the world to complete a series of moral and psychological measures and public health attitudes about COVID-19 during an early phase of the COVID-19 pandemic (between April and June 2020). The survey included seven broad categories of questions: COVID-19 beliefs and compliance behaviours; identity and social attitudes; ideology; health and well-being; moral beliefs and motivation; personality traits; and demographic variables. We report both raw and cleaned data, along with all survey materials, data visualisations, and psychometric evaluations of key variables.

  • Cortical thickness and resting-state cardiac function across the lifespan : A cross-sectional pooled mega-analysis

    • Understanding the association between autonomic nervous system [ANS] function and brain morphology across the lifespan provides important insights into neurovisceral mechanisms underlying health and disease. Resting-state ANS activity, indexed by measures of heart rate [HR] and its variability [HRV] has been associated with brain morphology, particularly cortical thickness [CT]. While findings have been mixed regarding the anatomical distribution and direction of the associations, these inconsistencies may be due to sex and age differences in HR/HRV and CT. Previous studies have been limited by small sample sizes, which impede the assessment of sex differences and aging effects on the association between ANS function and CT. To overcome these limitations, 20 groups worldwide contributed data collected under similar protocols of CT assessment and HR/HRV recording to be pooled in a mega-analysis (N = 1,218 (50.5% female), mean age 36.7 years (range: 12–87)). Findings suggest a decline in HRV as well as CT with increasing age. CT, particularly in the orbitofrontal cortex, explained additional variance in HRV, beyond the effects of aging. This pattern of results may suggest that the decline in HRV with increasing age is related to a decline in orbitofrontal CT. These effects were independent of sex and specific to HRV; with no significant association between CT and HR. Greater CT across the adult lifespan may be vital for the maintenance of healthy cardiac regulation via the ANS—or greater cardiac vagal activity as indirectly reflected in HRV may slow brain atrophy. Findings reveal an important association between CT and cardiac parasympathetic activity with implications for healthy aging and longevity that should be studied further in longitudinal research.

  • National identity predicts public health support during a global pandemic

    • Understanding collective behaviour is an important aspect of managing the pandemic response. Here the authors show in a large global study that participants that reported identifying more strongly with their nation reported greater engagement in public health behaviours and support for public health policies in the context of the pandemic. Changing collective behaviour and supporting non-pharmaceutical interventions is an important component in mitigating virus transmission during a pandemic. In a large international collaboration (Study 1, N = 49,968 across 67 countries), we investigated self-reported factors associated with public health behaviours (e.g., spatial distancing and stricter hygiene) and endorsed public policy interventions (e.g., closing bars and restaurants) during the early stage of the COVID-19 pandemic (April-May 2020). Respondents who reported identifying more strongly with their nation consistently reported greater engagement in public health behaviours and support for public health policies. Results were similar for representative and non-representative national samples. Study 2 (N = 42 countries) conceptually replicated the central finding using aggregate indices of national identity (obtained using the World Values Survey) and a measure of actual behaviour change during the pandemic (obtained from Google mobility reports). Higher levels of national identification prior to the pandemic predicted lower mobility during the early stage of the pandemic (r = -0.40). We discuss the potential implications of links between national identity, leadership, and public health for managing COVID-19 and future pandemics.

    • The recent discovery of an earth-like planet around Proxima Centauri has drawn much attention to this star and its environment. We performed a series of observations of Proxima Centauri using Spectro-Polarimetric High-contrast Exoplanet REsearch (SPHERE), the planet-finder instrument installed at the European Southern Observatory (ESO) Very Large Telescope (VLT) UT3, using its near-infrared modules, InfraRed Dual-band Imager and Spectrograph (IRDIS) and IFS. No planet was detected directly, but we set upper limits on the mass up to 7 au by exploiting the AMES-COND models. Our IFS observations reveal that no planet more massive than similar to 6-7 M-Jup can be present within 1 au. The dual-band imaging camera IRDIS also enables us to probe larger separations than other techniques such as radial velocity or astrometry. We obtained mass limits of the order of 4 M-Jup at separations of 2 au or larger, representing the most stringent mass limits at separations larger than 5 au available at the moment. We also made an attempt to estimate the radius of possible planets around Proxima using the reflected light. Since the residual noise for the observations is dominated by photon noise and thermal background, longer exposures in good observing conditions could improve the achievable contrast limit further.

  • Mammal body size evolution in North America and Europe over 20 Myr: similar trends generated by different processes

    • Because body size interacts with many fundamental biological properties of a species, body size evolution can be an essential component of the generation and maintenance of biodiversity. Here we investigate how body size evolution can be linked to the clade-specific diversification dynamics in different geographical regions. We analyse an extensive body size dataset of Neogene large herbivores (covering approx. 50% of the 970 species in the orders Artiodactyla and Perissodactyla) in Europe and North America in a Bayesian framework. We reconstruct the temporal patterns of body size in each order on each continent independently, and find significant increases of minimum size in three of the continental assemblages (except European perissodactyls), suggesting an active selection for larger bodies. Assessment of trait-correlated birth-death models indicates that the common trend of body size increase is generated by different processes in different clades and regions. Larger-bodied artiodactyl species on both continents tend to have higher origination rates, and both clades in North America show strong links between large bodies and low extinction rate. Collectively, our results suggest a strong role of species selection and perhaps of higher-taxon sorting in driving body size evolution, and highlight the value of investigating evolutionary processes in a biogeographic context.

  • Challenges in Permeability Assessment for Oral Drug Product Development

    • Drug permeation across the intestinal epithelium is a prerequisite for successful oral drug delivery. The increased interest in oral administration of peptides, as well as poorly soluble and poorly permeable compounds such as drugs for targeted protein degradation, have made permeability a key parameter in oral drug product development. This review describes the various in vitro, in silico and in vivo methodologies that are applied to determine drug permeability in the human gastrointestinal tract and identifies how they are applied in the different stages of drug development. The various methods used to predict, estimate or measure permeability values, ranging from in silico and in vitro methods all the way to studies in animals and humans, are discussed with regard to their advantages, limitations and applications. A special focus is put on novel techniques such as computational approaches, gut-on-chip models and human tissue-based models, where significant progress has been made in the last few years. In addition, the impact of permeability estimations on PK predictions in PBPK modeling, the degree to which excipients can affect drug permeability in clinical studies and the requirements for colonic drug absorption are addressed.

    • Concerns about the veracity of psychological research have been growing. Many findings in psychological science are based on studies with insufficient statistical power and nonrepresentative samples, or may otherwise be limited to specific, ungeneralizable settings or populations. Crowdsourced research, a type of large-scale collaboration in which one or more research projects are conducted across multiple lab sites, offers a pragmatic solution to these and other current methodological challenges. The Psychological Science Accelerator (PSA) is a distributed network of laboratories designed to enable and support crowdsourced research projects. These projects can focus on novel research questions or replicate prior research in large, diverse samples. The PSA’s mission is to accelerate the accumulation of reliable and generalizable evidence in psychological science. Here, we describe the background, structure, principles, procedures, benefits, and challenges of the PSA. In contrast to other crowdsourced research networks, the PSA is ongoing (as opposed to time limited), efficient (in that structures and principles are reused for different projects), decentralized, diverse (in both subjects and researchers), and inclusive (of proposals, contributions, and other relevant input from anyone inside or outside the network). The PSA and other approaches to crowdsourced psychological science will advance understanding of mental processes and behaviors by enabling rigorous research and systematic examination of its generalizability.

    • BackgroundRepetitive traumatic brain injuries in American football players (AFPs) can lead to the neurodegenerative disease chronic traumatic encephalopathy (CTE). Clinical symptoms of CTE range from mood and behavioral changes to cognitive impairment, depression, and suicidality. So far, CTE cannot be diagnosed in vivo and thus specific diagnostic parameters for CTE need to be found, to observe and treat exposed athletes as early as possible. Promising blood-based biomarkers for CTE include total tau (tTau), hyperphosphorylated tau (pTau), neurofilament light protein (NF-L), glial fibrillary acidic protein (GFAP), amyloid-beta 40 (A beta 40), amyloid-beta 42 (A beta 42) and calcium-binding protein B (S100-B). Previous studies have found elevated levels of these biomarkers in subjects exposed to TBIs, whereas cerebrospinal fluid (CSF) levels of A beta 40 and A beta 42 were decreased in CTE subjects. Here, we investigated whether young AFPs already exhibit changes of these biomarker candidates during the course of a single active season.MethodsBlood samples were drawn from n = 18 American Football Players before and after a full season and n = 18 male age-matched control subjects. The plasma titers of tTau, pTau, NF-L, GFAP, A beta 40, A beta 42 and S100-B were determined. Additionally, Apathy, Depression, and Health status as well as the concussion history and medical care were assessed and analyzed for correlations.ResultsHere we show, that the selected biomarker candidates for CTE do not change significantly during the seven-month period of a single active season of American Football in blood samples of AFPs compared to healthy controls. But interestingly, they exhibit generally elevated pTau titers. Furthermore, we found correlations of depression, quality-of-life, career length, training participation and training continuation with headache after concussion with various titers.ConclusionOur data indicates, that changes of CTE marker candidates either occur slowly over several active seasons of American Football or are exclusively found in CSF. Nevertheless, our results underline the importance of a long-term assessment of these biomarker candidates, which might be possible through repeated blood biomarker monitoring in exposed athletes in the future.

    • Carbapenems are vital antibiotics, but their efficacy is increasingly compromised by metallo-β-lactamases (MBLs). Here we report the discovery and optimization of potent broad-spectrum MBL inhibitors. A high-throughput screen for NDM-1 inhibitors identified indole-2-carboxylates (InCs) as potential β-lactamase stable β-lactam mimics. Subsequent structure-activity relationship studies revealed InCs as a new class of potent MBL inhibitor, active against all MBL classes of major clinical relevance. Crystallographic studies revealed a binding mode of the InCs to MBLs that, in some regards, mimics that predicted for intact carbapenems, including with respect to maintenance of the Zn(II)-bound hydroxyl, and in other regards mimics binding observed in MBL-carbapenem product complexes. InCs restore carbapenem activity against multiple drug-resistant Gram-negative bacteria and have a low frequency of resistance. InCs also have a good in vivo safety profile, and when combined with meropenem show a strong in vivo efficacy in peritonitis and thigh mouse infection models.

    • Liquid steel samples were taken during ladle treatment and continuous casting of 37 heats of two duplex stainless steel grades. These samples were used to study inclusion characteristics and potential process control using the pulse distribution analysis with optical emission spectroscopy method (PDA/OES). In addition, a few samples were chosen for a scanning electron microscope examination. A simplified approach of presenting the inclusion composition in ternary diagrams using the PDA/OES method is discussed. More specifically, by defining an operating window, potential deviations can be identified early in the steelmaking process. The variation of the B factor for A(l) (B-Al) with process time was presented showing similar tendencies for several heats. For that reason, the B factor is considered to be a reliable parameter to study any process changes with respect to inclusion characteristics. Parameters such as B-Al and the number of inclusions per unit volume larger than 4 mu m obtained using the PDA/OES method showed a moderate correlation with a slivers index. Statistically, more data have to be collected for further studies. Nevertheless, the PDA/OES method is judged to be a promising tool to determine a slivers risk factor at an early stage of the process.

    • Calcifications in the basal ganglia are a common incidental finding and are sometimes inherited as an autosomal dominant trait ( idiopathic basal ganglia calcification (IBGC)). Recently, mutations in the PDGFRB gene coding for the platelet-derived growth factor receptor beta (PDGF-R beta) were linked to IBGC. Here we identify six families of different ancestry with nonsense and missense mutations in the gene encoding PDGF-B, the main ligand for PDGF-R beta. We also show that mice carrying hypomorphic Pdgfb alleles develop brain calcifications that show age-related expansion. The occurrence of these calcium depositions depends on the loss of endothelial PDGF-B and correlates with the degree of pericyte and blood-brain barrier deficiency. Thus, our data present a clear link between Pdgfb mutations and brain calcifications in mice, as well as between PDGFB mutations and IBGC in humans.

  • The Helicobacter pylori Genome Project: insights into H. pylori population structure from analysis of a worldwide collection of complete genomes

    • Helicobacter pylori, a dominant member of the gastric microbiota, shares co-evolutionary history with humans. This has led to the development of genetically distinct H. pylori subpopulations associated with the geographic origin of the host and with differential gastric disease risk. Here, we provide insights into H. pylori population structure as a part of the Helicobacter pylori Genome Project (HpGP), a multi-disciplinary initiative aimed at elucidating H. pylori pathogenesis and identifying new therapeutic targets. We collected 1011 well-characterized clinical strains from 50 countries and generated high-quality genome sequences. We analysed core genome diversity and population structure of the HpGP dataset and 255 worldwide reference genomes to outline the ancestral contribution to Eurasian, African, and American populations. We found evidence of substantial contribution of population hpNorthAsia and subpopulation hspUral in Northern European H. pylori. The genomes of H. pylori isolated from northern and southern Indigenous Americans differed in that bacteria isolated in northern Indigenous communities were more similar to North Asian H. pylori while the southern had higher relatedness to hpEastAsia. Notably, we also found a highly clonal yet geographically dispersed North American subpopulation, which is negative for the cag pathogenicity island, and present in 7% of sequenced US genomes. We expect the HpGP dataset and the corresponding strains to become a major asset for H. pylori genomics.

  • Numerical analysis of stresses in double-lap adhesive joint under thermo-mechanical load

    • A numerical study for the double-lap adhesive joint made of similar adherends subjected to tensile and thermal loads is presented. A novel displacement coupling conditions which are able to correctly represent monoclinic materials (off-axis layers of composite laminates) are used to build a comprehensive numerical model. Two types of double-lap joints are considered in this study: metal–metal and composite-composite. In case of composite laminates, four lay-ups are evaluated: unidirectional ([08]T and [908]T) and quasi-isotropic laminates ([0/45/90/−45]S and [90/45/0/−45]S). The effect of different parameters (adherend stiffness, ply stacking sequence, adherend thickness, one-step or two-step manufacturing of the joint) on peel and shear stress distribution in the middle of the adhesive is studied. The comparison of the behaviour of single-lap and double-lap joint in relation to these parameters is made. The maximum peel and shear stress at the ends of the overlap with respect to the axial modulus of the adherends are presented in a form of the master curves. The analyses of results show that: the maximum peel and shear stress concentration at the overlap ends is reduced with the increase of the axial modulus of the adherend; the stress distribution in the adhesive layer can be improved (lower stress concentrations and level-out the curve) by changing the fibre orientation (which affect the stiffness) in plies connected to the adhesive layer.

    • A comprehensive stress analysis by means of Finite Element Method (FEM) for single-lap joint subjected to thermal and mechanical loads is presented in this paper. Simulation is used to predict the effect of residual thermal stresses (caused by difference of temperature of use and elevated temperature during the assembly of the joint) on stress distribution within adhesive layer. The residual thermal stresses are assigned to joint members as initial condition before the mechanical load is applied. The FEM model employs linear and nonlinear material model and accounts for geometrical nonlinearity. It is confirmed that the difference between the manufacturing and the ambient temperature results in high residual thermal stresses, especially in axial and lateral directions of the joint. The calculation of total stress as superposition of thermal and mechanical stresses works only for linear materials. Moreover, simultaneous application of temperature and mechanical load (applied strain in case of displacement controlled test) in FEM produces inaccurate results, since in real situation the strain is applied to already thermally loaded structure. It is also found that the residual thermal stresses may reduce the peel and shear stress concentration in the adhesive at the ends of overlap and the shear stress within the overlap.

    • The objective of this work is to evaluate the effect of residual thermal stresses, arising after assembling a single-lap joint at elevated temperature, on the inelastic thermo-mechanical stress state in the adhesive layer. The numerical analysis (FEM) employing linear and non-linear material models, with geometrical nonlinearity accounted for, is carried out. Simulating the mechanical response, the calculated thermal stresses are assigned as initial conditions to polymeric, composite and metallic joint members to reflect the loading sequence where the mechanical strain is applied to cooled-down structure. It is shown that the sequence of application matters and simulations with simultaneous application of temperature and strain give different result. Two scenarios for adhesive joints with composites are studied: joining by adhesive curing of already cured composite parts (two-step process) and curing the adhesive and the composite simultaneously in one-step (co-curing). Results show that while in-plane stresses in the adhesive are higher, the peaks of out-of-plane shear stress and peel stress (most responsible for the joint failure) at the end of the overlap are reduced due to thermal effects. In joints containing composite parts, the one-step joining scenario is more favorable than the two-step. The ply stacking sequence in the composite has significant effect on stress concentrations as well as on the plateau value of the shear stress in the adhesive.

    • Opportunities, challenges and recommended targeted actions to accelerate qualitative data sharing to address complex socio-environmental problems Socio-environmental synthesis as a research approach contributes to broader sustainability policy and practice by reusing data from disparate disciplines in innovative ways. Synthesizing diverse data sources and types of evidence can help to better conceptualize, investigate and address increasingly complex socio-environmental problems. However, sharing qualitative data for re-use remains uncommon when compared to sharing quantitative data. We argue that qualitative data present untapped opportunities for sustainability science, and discuss practical pathways to facilitate and realize the benefits from sharing and reusing qualitative data. However, these opportunities and benefits are also hindered by practical, ethical and epistemological challenges. To address these challenges and accelerate qualitative data sharing, we outline enabling conditions and suggest actions for researchers, institutions, funders, data repository managers and publishers.

    • Relapsing fever (RF) is caused by different species of Borrelia transmitted by soft ticks or by the human body louse. Illness is characterized by reappearing peaks of high concentrations of spirochetes in blood, concordant with fever peaks separated by asymptomatic periods. Neuroborreliosis is one of the most severe manifestations of RF borreliosis. To understand the immune response during early RF, we analyzed immune cells in brain and kidney of mice infected with B. crocidurae during the acute infection. Our results indicate that brain defense is comprised primarily of innate immune cells. Despite the infiltration of innate immune cells, Borrelia was not completely eradicated. A failure of the host brain to clear the bacteria may give the pathogen a niche where it can persist. Using our mouse model, we revealed that Borrelia duttonii could persist in the mouse brain for up to 270 days, without being present in the circulation. The infection was silent with no change in host gene expression, and the spirochetes could re-enter the circulation after immunosuppression. We propose that the brain is used by the pathogen to evade host immunity and serves as a possible natural reservoir for B. duttonii, a spirochete that has rarely been found in any mammalian host other than man. Borrelia-induced complications during pregnancy have been reported, and are especially common in RF. In our established mouse model of gestational RF, we could show that the fetuses suffered from severe pathology and growth retardation, probably as a consequence of placental destruction. We could also show trans-placental transmission of the bacteria leading to neonatal RF. Surprisingly, pregnant dams had a lower bacterial load and less severe disease, showing that pregnancy has a protective effect during RF. We have used the gestational RF model to investigate host factors favoring disease resolution. Because the spleen is the primary organ responsible for trapping and removing blood-borne pathogens, we have compared temporal changes in spleen immune cell populations and cytokine/chemokine induction during the infection. Spleens of pregnant mice had earlier neutrophil infiltration, as well as faster and higher production of pro-inflammatory mediators. This rapid, robust response suggests a more effective host defense. Thus, an enhanced pro-inflammatory response during pregnancy imparts a distinct advantage in controlling the severity of relapsing fever infection.

  • A timescale for placental mammal diversification based on Bayesian modeling of the fossil record

    • The timing of the placental mammal radiation has been the focus of debate over the efficacy of competing methods for establishing evolutionary timescales. Molecular clock analyses estimate that placental mammals originated before the Cretaceous-Paleogene (K-Pg) mass extinction, anywhere from the Late Cretaceous to the Jurassic. However, the absence of definitive fossils of placentals before the K-Pg boundary is compatible with a post-Cretaceous origin. Nevertheless, lineage divergence must occur before it can be manifest phenotypically in descendent lineages. This, combined with the non-uniformity of the rock and fossil records, requires the fossil record to be interpreted rather than read literally. To achieve this, we introduce an extended Bayesian Brownian bridge model that estimates the age of origination and, where applicable, extinction through a probabilistic interpretation of the fossil record. The model estimates the origination of placentals in the Late Cretaceous, with ordinal crown groups originating at or after the K-Pg boundary. The results reduce the plausible interval for placental mammal origination to the younger range of molecular clock estimates. Our findings support both the Long Fuse and Soft Explosive models of placental mammal diversification, indicating that the placentals originated shortly prior to the K-Pg mass extinction. The origination of many modern mammal lineages overlapped with and followed the K-Pg mass extinction.

    • Introduction: Medulloblastoma is a malignant embryonal tumor of the cerebellum that occurs predominantly in children. To find germline genetic variants associated with medulloblastoma risk, we conducted a genome-wide association study (GWAS) including 244 medulloblastoma cases and 247 control subjects from Sweden and Denmark.Methods: Genotyping was performed using Illumina BeadChips, and untyped variants were imputed using IMPUTE2.Results: Fifty-nine variants in 11 loci were associated with increased medulloblastoma risk (p < 1 × 10–5), but none were statistically significant after adjusting for multiple testing (p < 5 × 10–8). Thirteen of these variants were genotyped, whereas 46 were imputed. Genotyped variants were further investigated in a validation study comprising 249 medulloblastoma cases and 629 control subjects. In the validation study, rs78021424 (18p11.23, PTPRM) was associated with medulloblastoma risk with OR in the same direction as in the discovery cohort (ORT = 1.59, pvalidation = 0.02). We also selected seven medulloblastoma predisposition genes for investigation using a candidate gene approach: APC, BRCA2, PALB2, PTCH1, SUFU, TP53, and GPR161. The strongest evidence for association was found for rs201458864 (PALB2, ORT = 3.76, p = 3.2 × 10–4) and rs79036813 (PTCH1, ORA = 0.42, p = 2.6 × 10–3).Conclusion: The results of this study, including a novel potential medulloblastoma risk loci at 18p11.23, are suggestive but need further validation in independent cohorts.

    • Usually, the viscoelastic (VE) response of polymers for applications in composites is obtained in uniaxial strainor stress-controlled tests. However, analyzing multimaterial structures by the Finite Element Method (FEM) or by other numerical or analytical tools, a material model in terms of a complete set of compliance functions and/or relaxation functions is required. In this paper, a methodology and exact analytical expressions for calculating the whole set of VE functions is presented based on the relaxation modulus E(t)and Poisson’s ratio v (t) determined in strain-controlled tests. The method is based on Laplace transforms, where an exact inversion is possible if a linear VE model with functions in Prony series is used. Results of the analytical model are compared with the FEM simulation, where specific boundary conditions to determine each particular VE function are used. Finally, the applicability of the so-called quasi-elastic method is investigated, where the expressions of elasticity theory are used to calculate a given viscoelastic function at an instant of time tk using the instant values of E(tk) and v(tk). For isotropic materials, the three approaches render almost coinciding results.

    • Building managers and operators as at Campus Pinkafeld are interested in a cost optimal fulfilment of their energy needs. From a strategic point of view they are interested in optimal investments and upgrades. From an operative point of view they are interested in an optimal use of all available resources. This paper shows how the decision support system (DSS) of the project Energy Efficiency and Risk Management in Public Buildings (EnRiMa) will help with this challenges and the integration of the DSS with the existing energy management system (EMS) is one of the key issues for a successful project. The strategic DSS will inform the building owner about possible new technologies that might reduce the total building energy costs or environmental impact. The benefit of an operational DSS is to enable the building operator to use already adopted energy efficiency improving technologies as pre-cooling, pre-heating or any other demand response related tasks to decrease costs and emissions caused by the heating and cooling system of the building. Assuming an upper and lower limit for the room temperature, we model the effect of active equipment control (via changes to either the set point or the valve flow) on the zone temperature taking into account the external temperature, solar gains, the building shell, and internal loads. The energy required to change the zone temperature in each time period is then used to calculate the energy cost or efficiency in the objective function of an optimization problem. This paper reports on example results for Campus Pinkafeld, shows the technical approach, and that such a flexible approach can save 10% costs only on an operational level.

    • BACKGROUND The global prevalence of obesity and overweight is increasing rapidly among adults as well as among children and adolescents. Recent genome-wide association studies have provided strong support for association between variants in the FTO gene and obesity. We sequenced regions of the FTO gene to identify novel variants that are associated with obesity and related metabolic traits. RESULTS We screened exons 3 and 4 including exon-intron boundaries in FTO in 48 obese children and adolescents and identified three novel single nucleotide polymorphism in the fourth intronic region, (c.896+37A>G, c.896+117C>G and c.896+223A>G). We further genotyped c.896+223A>G in 962 subjects, 450 well-characterized obese children and adolescents and 512 adolescents with normal weight. Evidence for differences in genotype frequencies were not detected for the c.896+223A>G variant between extremely obese children and adolescents and normal weight adolescents (P=0.406, OR=1.154 (0.768-1.736)). Obese subjects with the GG genotype, however, had 30% increased fasting serum insulin levels (P=0.017) and increased degree of insulin resistance (P=0.025). There were in addition no differences in body mass index (BMI) or BMI standard deviation score (SDS) levels among the obese subjects according to genotype and the associations with insulin levels and insulin resistance remained significant when adjusting for BMI SDS. CONCLUSION These findings suggest that this novel variant in FTO is affecting metabolic phenotypes such as insulin resistance, which are not mediated through differences in BMI levels.

    • Single-layer molybdenum ditelluride (MoTe2) has attracted attention due to the smaller energy difference between the semiconducting (1H) and semimetallic (1T′) phases with respect to other two-dimensional transition metal dichalcogenides (TMDs). Understanding the phenomenon of polymorphism between these structural phases is of great fundamental and practical importance. In this paper, we report a 1H to 1T′ phase transition occurring during the chemical vapor deposition (CVD) synthesis of single-layer MoTe2 at 730 °C. The transformation originates at the heterocontact between monoclinic and hexagonal crystals and progresses to either yield a partial or complete 1H to 1T′ phase transition. Microscopic and spectroscopic analyses of the MoTe2 crystals reveal the presence of Te vacancies and mirror twin boundaries (MTB) domains in the hexagonal phase. The experimental observations and theoretical simulations indicate that the combination of heterocontact formation and Te vacancies are relevant triggering mechanisms in the observed transformation. By advancing in the understanding and controlling of the direct synthesis of lateral 1T′/1H heterostructures, this work contributes to the development of MoTe2-based electronic and optoelectronic devices with low contact resistance.

    • Synthesis of bis-4,5-diazafluoren-9-one silver(I) nitrate I (dafone = 4,5-diazafluoren-9-one) and the low temperature X-ray single crystal structure of [Ag(4,5-diazafluoren-9-one)2NO3], crystal form 1, and a re-determination of [Ag(4,5-diazafluoren-9-one)2]NO3 . H2O, crystal form 2 are presented. Crystal form 1 has a distorted trigonal planar coordination geometry around Ag(I) with an N-Ag-N bond angle of 123.45(7)o. Crystal form 2 has a perfect linear coordination around Ag, with N-Ag-N 180.0o. Compound I was characterized by 1H-NMR, biological activity and ESI-MS in DMSO at room temperature. The biological activity was determined against 6 different resistant clinical isolates; two Gram-positive (Staphylococcus aureus and Streptococcus pyogenes) and four Gram-negative (Pseudomonas aeruginosa, Klebsiella pneumoniae, Proteus mirabilis, and Salmonella sp.) in comparison with 15 known antibiotics used in the treatment of diabetic foot infections. Compound I showed broad spectrum activity against all the test organisms. P. mirabilis and S. aureus and K. pneumoniae were the most sensitive clinical isolates (MIC = 4, 6 and 4 μg ml-1, respectively). Three different hydrogels containing I or Ag2SO4 were prepared and the antimicrobial activity against Ps. aeruginosa (ATCC 15442) compared, showing more or less equal activity on a weight basis, but I seems to have a significant better performance per silver ion. The Ag(I) complex also binds more effectively to calf thymus DNA than the dafone ligand itself.

As an alternative to solution chemistry, solid-state grinding could be used to prepare compounds 1 and 3, but the preparation of 4 and 5 in this way failed. The Ag(I) ions in the monomeric compounds 1, 2, 5, and 6 are coordinated to two ligands via the nitrogen atoms of the quinoline rings, thereby forming a distorted linear coordination geometry with Ag-N bond distances of 2.142(2)-2.336(2) angstrom and N-Ag-N bond angles of 163.62(13)degrees-172.25(13)degrees. The 1D coordination polymers 3 and 4 contain Ag(I) centers coordinating one ligand and two bridging nitrate groups, thereby forming a distorted trigonal planar coordination geometry with Ag-N bond distances of 2.2700(14) and 2.224(5) angstrom, Ag-O bond distances of 2.261(4)-2.536(5) angstrom, and N-Ag-O bond angles of 115.23(5)degrees-155.56(5)degrees. Hirshfeld surface analyses of compounds 1-6 are presented as d(norm) and curvedness maps. The d(norm) maps show different interaction sites around the Ag(I) ions, i.e., Ag center dot center dot center dot Ag interactions and possible O-H center dot center dot center dot O, C-H center dot center dot center dot O, C-H center dot center dot center dot N, and C-H center dot center dot center dot C hydrogen bonds. Curvedness maps are a good way of visualizing pi-pi tacking interactions between molecules. The antimicrobial activities of compounds 1, 2, and 6 were screened against 15 different multidrug-resistant strains of bacteria isolated from diabetic foot ulcers and compared to the antimicrobial activities of the clinically used silver sulfadiazine (SS). Compound 2 showed activity similar to SS against this set of test organisms, being active against all strains and having slightly better average silver efficiency than SS (5 vs 6 mu g Ag/mL). Against the standard nonresistant bacterial strains of Staphylococcus aureus, Pseudomonas aeruginosa, Proteus mirabilis, and Streptococcus pyogenes, compound 1 performed better than silver nitrate, with an average MIC of 6 mu g Ag/mL versus 18 mu g Ag/mL for the reference AgNO3. Electrospray ionization mass spectrometry (ESI-MS) analyses of compounds 3 and 6 in DMSO/MeOH confirm the two-coordinated Ag+ complexes in solution, and the results of the H-1 NMR titrations of DMSO solutions of 5-nitroquinoline and 8-nitroquinoline with AgNO3 in DMSO suggest that 5-nitroquinoline is more strongly coordinated to the silver ion.

    • An important factor affecting the mechanical characteristics of composite materials is the adhesion of constituents which can be characterized by the interfacial shear strength (IFSS). Most of the experimental methods for the evaluation of IFSS involve model single-fiber composites. By contrast, the method proposed by Bowyer and Bader is based on an analysis of the stress-strain curve of a short-fiber-reinforced composite in tension. In the present study, the method is applied to the experimental tension curves of short-flax-fiberreinforced polypropylene to evaluate the IFSS and the fiber orientation factor of the composite. The IFSS was found to depend on the volume fraction of fibers and the treatment to improve adhesion.

    • A previously proposed statistical approach for computing factors of safety (i.e. numerical measures of mechanical reliability) for any load bearing structure, like a vertical plant stem, is here extended to cope with organic structures whose morphological or mechanical properties have Weibull frequency distributions. This approach is illustrated using the actual lengthLand critical buckling lengthLerof flower stalks (peduncles) collected from isogenic garlic (Allium sativum) populations grown under windy field and protected glasshouse conditions. Our analyses of the data indicate thatLandLerof peduncles harvested from both populations have Weibull frequency distributions, that the factor of safety for glasshouse grown peduncles is very near unity (i.e.S=1.03), and that the factor of safety of field grown peduncles is 73% higher than that of glasshouse grown plants (i.e.S=1.73). Comparisons between theS-values computed on the basis of our formulas and on the basis of the quotient of the mean values ofLerandLfor each of the two populations indicate that the statistical method gives biologically realisticS-values and that the difference in theS-values for stems grown under protected and unprotected environmental conditions likely reflects the effects of chronic mechanical perturbation (due to wind-induced drag) on normal stem growth and development.

    • The calculation of the energy release rate of a debonding interface crack between fiber and matrix in plane strain requires a knowledge of the stress state and displacements at the neighbourhood of the crack tip. Analytical solutions which involve interpenetrations of the free boundaries of the fiber and matrix have classically been used to predict the variation of the energy release rate with the debonding angle. In this paper a numerical study of this problem as a contact problem, using Boundary Elements, is carried out. The singular stress components are modelled by means of singular discontinuous elements, a particular contact algorithm, which is very appropriate for cases like that under consideration where the fiber is much stiffer than the matrix, having been proposed. The numerical procedure shows the appearance of a contact zone with physical meaning starting at a certain value of the debonding angle, which implies the presence of a pure mode II of fracture. The values of G found by BEM are compared with those predicted by the use of an analytical solution showing clear discrepancies when the contact zone starts to develop, BEM always predicting smaller values of the energy release rate, in accordance with the absence of mode I in the presence of a contact zone.

    • We have cloned melanocortin receptors (MCRs) from several species of fish. The MC4R and MC5R subtypes arose early in vertebrate evolution and their primary structure is remarkably conserved. Expression and pharmacological characterization of the MCRs in fish has revealed that they bind and respond to melanocortin peptides with high potency. Detailed characterization of the binding properties of the different subtypes suggests that MCRs in early vertebrates had preference for adrenocorticotropic hormone (ACTH) peptides, while the high sensitivity for the shorter proopiomelanocortin (POMC) products, such as the α-, β-, and γ-melanocyte-stimulating hormone (MSH), has appeared later, perhaps as the MCR subtypes gained more specialized functions. The MCR repertoire shows in general high similarities in their primary structures, while they are however not similar in terms of functional roles. The MCRs serve therefore as an interesting model family to understand the molecular mechanisms of how functions of the genes can diverge during evolution. In this review, we provide an overview of our recent studies on the cloning, expression, pharmacology, 3D modeling, and genomic studies of the MCRs in non-mammalian species.

    • Relatively little is known about the endocannabinoid system in human neuroblastoma cell lines. In the present study, we have investigated the expression of the genes coding for the enzymes involved in the synthesis and catabolism of endocannabinoids in the SH-SY5Y cell line. The expression of MGLL, the gene coding for the 2-arachidonoylglycerol hydrolytic enzyme monoacylglycerol lipase (MAGL), was found to be 85 and 340 fold lower than the expression levels for the genes coding for alpha/beta-hydrolase domain containing 6 and 12 (ABHD6, ABHD12), which are alternative hydrolytic enzymes for this endocannabinoid. In comparison, mRNA levels of MGLL were 1.5 fold higher than ABHD6 and 2 fold lower than the levels of ABHD12 in DU-145 human prostate cells. In functional assays, the hydrolysis of the 2-arachidonoylglycerol homologue 2-oleoylglycerol by intact SH-SY5Y cells was partially inhibited by the ABHD6 inhibitor WWL70, but not by the MAGL inhibitor JZL184, whereas the reverse was true in DU-145 cells. The combination of JZL184 + WWL70 did, however produce a significantly greater inhibition of 2-OG hydrolysis than seen with WWL70 alone in the SH-SY5Y cells. The low MGLL expression in the SH-SY5Y cells was not due to epigenetic silencing, since levels were not affected by treatment with the methylation inhibitor 5-aza-2'-deoxycytidine and/or the histone acetylase inhibitor trichostatin A. The low MGLL expression in SH-SY5Y cells should be taken into account when using these cells in experiments investigating the involvement of the endocannabinoid system in models of physiological and pathological processes.

    • Polymer composites are known to exhibit nonlinear stress-strain response due to nonlinearly elastic or plastic deformation of the matrix and damage accumulation. Mechanistic modeling of material response explicitly accounting for these interacting factors often leads to complex theories. Plasticity theory formalism provides an alternative for nonlinear deformation description of composite material. We examine the applicability of an orthotropic plasticity model, developed by Sun et al. for unidirectionally reinforced composite, to composite laminate. The response of a symmetric and balanced cross-ply glass/epoxy laminate is studied under uniaxial tensile loading at different angles to the material orthotropy axis. It is found that the associated flow rule and a quadratic approximation of the orthotropic potential function provide satisfactory description for the nonlinear strain component under monotonic loading for 15-45° off-axis angle range. The nonlinearity in on-axis loading (modulus degradation)is well described by stiffness reduction due to the cracks in transverse plies. Meanwhile the change of elastic modulus due to intralaminar cracking can be neglected in off-axis loading, but the possible intensification of nonlinear deformation in off-axis loading caused by the presence of intralaminar cracks agrees well with orthotropic potential formalism.

    • NOW (New and Old Worlds) is a global database of fossil mammal occurrences, currently containing around 68,000 locality-species entries. The database spans the last 66 million years, with its primary focus on the last 23 million years. Whereas the database contains records from all continents, the main focus and coverage of the database historically has been on Eurasia. The database includes primarily, but not exclusively, terrestrial mammals. It covers a large part of the currently known mammalian fossil record, focusing on classical and actively researched fossil localities. The database is managed in collaboration with an international advisory board of experts. Rather than a static archive, it emphasizes the continuous integration of new knowledge of the community, data curation, and consistency of scientific interpretations. The database records species occurrences at localities worldwide, as well as ecological characteristics of fossil species, geological contexts of localities and more. The NOW database is primarily used for two purposes: (1) queries about occurrences of particular taxa, their characteristics and properties of localities in the spirit of an encyclopedia; and (2) large scale research and quantitative analyses of evolutionary processes, patterns, reconstructing past environments, as well as interpreting evolutionary contexts. The data are fully open, no logging in or community membership is necessary for using the data for any purpose.
