    • This is a study of the design processes employed by architect Jan Gezelius in four different projects. It consists of two intertwined investigations: the first reconstructs the history of each design process, and the second analyzes each process from a phenomenological perspective. The historical reconstructions are a series of narratives based on Gezelius's professional archives and on interviews. These reconstructions are then the object of an analytical investigation that compares the main features of Gezelius's design approach with results derived from theoretical studies analyzing the nature of design processes. An interaction between conjecture and exploration is found to be characteristic of the design processes in each of the four cases. Gezelius begins with a tentative concept, then investigates its design implications through experimentation, uncovering and exploring its inherent opportunities for the particular design task. The insights gained provide the basis for a revision of the tentative concept and further exploration as the design concept evolves in a continual process of condensation and accumulation. In the analysis, this model of conjecture and exploration is supplemented by a productive function - a generative paradigm that mediates between the pre-reflective thought patterns and the design drawings, acting simultaneously as a driving force, a probe, and a track. Further, the study describes the technique of developing tentative design concepts by comparing the present project with similar projects in a repertoire gleaned from both historical precedent and personal experience. This repertoire provides exemplary schemes and design principles. The interaction between architect and client in these cases was a negotiation between enthusiastic parties that produced satisfying results; the architect was not given free rein. The accumulation of a design concept based on interaction between tentative concepts and their exploration has no clear end - there is no speciffic solution to reach. Several different ways of terminating the design process are discussed. After the historical reconstructions and their analyses, the paper concludes with a critical commentary of some aspects of the study itself.

    • The carboxyl ester lipase (CEL) gene is highly expressed in exocrine pancreas and expression of the human CEL gene is mediated by a strong tissue-specific enhancer, which is absolutely necessary for high-level expression. The mouse promoter, on the other hand, does not contain a corresponding enhancer element, but instead is totally dependent on another pancreas-specific element. This element is identified as a pancreatic transcription factor 1 (PTF 1)-binding site. The human CEL promoter also contains a putative PTF 1 element located at a position corresponding to the essential PTF 1 site in the mouse promoter. However, nucleotide changes in the human promoter 5' flanking this PTF 1 site have created an overlapping CCAAT/enhancer-binding protein (C/EBP)-like binding motif, interfering with the binding of PTF 1. Hence, our findings provide an example of genetic divergence between species not accompanied by difference in function.

    • The human carboxyl ester lipase (CEL) is an important enzyme for the intestinal absorption of dietary lipids. The gene is highly expressed in exocrine pancreas and in the mammary gland during pregnancy and lactation. In this paper, we have focused on its transcriptional regulation in exocrine pancreas. Reporter gene analysis in cell cultures reveals that a high level of tissue-specific expression is established by the proximal 839 base pairs of the 5'-flanking region. This is due to a strong enhancer, located at -672 to -637. Transfections in mammary gland-derived cells reveal that the enhancer is pancreas-specific and does not contribute to the mammary gland expression. This indicates that the expression of the CEL gene in the mammary gland and pancreas, respectively, is due to two different regulatory systems. Further characterizations of the enhancer reveal that it is composed of two closely located cis-elements. The proximal element mediates a positive effect, whereas the distal element exerts a silencing effect on the positive proximal element. The functional enhancer complex is composed of ubiquitously expressed factors, since similar interactions are achieved with nuclear extracts from cells derived from other tissues. However, no enhancer activity is achieved in such cells. Hence, the net enhancer activity is the result of a tissue-specific balance between factors interacting with the two elements. Since none of the described cis-elements show any clear homology to known cis-elements, we propose that the interacting complex is composed of yet unidentified transcription factors.

    • Inspired by the work and attitudes of architects and engineers like Jorg Schlaich, Renzo Piano, Piero Luigi Nervi, Sverre Fehn, Ted Happold, and environments like ILEK in Stuttgart and ETH in Zurich, a vision of a new kind of architects and engineers arose at Chalmers University of Technology in the early 2000. With support from the university and the branch, a double degree Architecture and Engineering programme was developed. Since the programme started in 2006 it has been a very popular programme, and among all Swedish MSc in Engineering and Master of Architecture programmes it has almost every year been the most difficult programme to get admitted to. The concept of the programme is a 180 ects (European Credit Transfer and Accumulation System) bachelor's degree, where the fundamentals from the engineering science: mathematics, mechanics, physics and materials, is combined with history of architecture and engineering, artistic explorative courses, and the fundamentals of the architectural design process. After three years the students can choose to continue for a Master of Science in Engineering with different possible directions, from mathematics and data science to industrial ecology, acoustics, management, structural engineering, and building technology, or to continue for a Master of Architecture. For the latter they need 150 etcs minimum in pure architectural design projects. In this paper the basic concepts of the programme, the culture developed around it and the strengths we can experience in the examined students will be discussed and reflected. Today students from the programme can be found at architecture and engineering companies all over the world and are appreciated for their ability to address complex architectural and engineering design issues with attitudes, insights and skills from the both professions.
