    • The academic study of rural landscapes covers a broad range of academic disciplines and thematic, methodological and theoretical concerns and interests; including questions concerned with resource use (e.g. agriculture, forestry, water and mining), settlement, livelihoods, conflicts, conservation, culture and identity. This diversity is clearly a strength (the rich empirical and intellectual base), but also presents a challenge, as the dissemination of research findings is distributed through a plethora of publishing channels, which do not necessarily encourage exchange of results and ideas that are not already perceived as germane to already established academic networks.

    • First-principles calculations have been used to study the effect of vacancies and relaxation around the vacancy sites in substoichiometric TiC1-x. The effect of relaxation on phase stabilities, equilibrium volumes. and electronic structure of the substoichiometric phases was studied using a combined approach of pseudopotential plane wave and full-potential linear muffin-tin orbital methods. A relaxation away from the vacancies was found for the titanium atoms, the magnitude of which increased with vacancy concentration and the inclusion of nearest-neighbor carbon atom relaxation. The inclusion of local relaxations was found to correctly predict the off-stoichiometric equilibrium composition of titanium carbide. The anomalous volume behavior of TiC at small vacancy concentration is explained as an effect of the local relaxation of the atoms surrounding the vacancy sites, but we do not find that the lattice parameter of any of the studied stoichiometries is larger than that of ideal stoichiometric TiC.

    • A survey, aimed at Swedish researchers in the field of spatial data, was conducted in 2021. 113 individuals responded to the questionnaire, which gives us insight into how researchers in the field view; sharing of data, how they work with data, which metadata they think are central and what support they ask for in order to be able to handle spatial data.According to the survey a majority of the researchers share and use data shared by others in their research. Central metadata that is emphasized are coordinate systems, projections, time of the information, data capture, data content and author. The question of how to make data available is very varied among the respondents. The questionnaire responses contain a wide range of individual solutions to make the information available. A majority of researchers believe that the work with data management and accessibility has increased over the past 5 years. As an obstacle to working more with this is time, but there are also other obstacles. The researchers are seeking additional support that can be summarized as: support, technical guidance, storage, information, time and money.

    • Mutation of the melanocortin-receptor 4 (MC4R) is the most frequent cause of severe obesity in humans. Binding of agouti-related peptide (AgRP) to MC4R involves the co-receptor syndecan-3, a heparan sulfate proteoglycan. The proteoglycan can be structurally modified by the enzyme heparanase. Here we tested the hypothesis that heparanase plays a role in food intake behaviour and energy balance regulation by analysing body weight, body composition and food intake in genetically modified mice that either lack or overexpress heparanase. We also assessed food intake and body weight following acute central intracerebroventricular administration of heparanase; such treatment reduced food intake in wildtype mice, an effect that was abolished in mice lacking MC4R. By contrast, heparanase knockout mice on a high-fat diet showed increased food intake and maturity-onset obesity, with up to a 40% increase in body fat. Mice overexpressing heparanase displayed essentially the opposite phenotypes, with a reduced fat mass. These results implicate heparanase in energy balance control via the central melanocortin system. Our data indicate that heparanase acts as a negative modulator of AgRP signaling at MC4R, through cleavage of heparan sulfate chains presumably linked to syndecan-3.

    • Land use and land cover change (LUCC) has emerged as an important issue of global change with significant influences on the geochemical cycle and regional climate change. Understanding the historical changes of land use and analyzing the environmental effects of LUCC make such reconstructions important. Based on historical statistics, this article reconstructs cropland area at the municipality level (härad/kommun) in the Scandinavian Peninsula during 1875–1999. The total acreage of cropland increased 36.84 % during 1875–1930, kept stable during 1930–1950, and decreased 14.25 % during 1950–1999. The croplands of Sweden and Norway both increased before 1950 and changed in different ways during 1950–1999 when the cropland decreased by 19.79 % in Sweden but increased by 9.63 % in Norway. The counties in the south Scandinavian Peninsula owned most of the cropland, with a cropland proportion of over 20 %, and experienced relatively obvious cropland changes. Growth centers with a rate of increase of over 5 % during 1875–1910 were found in Skåne, Stockholm, the Uppsala counties in Sweden, and the area around Oslo county in Norway. The general cropland distribution showed almost no change during this period, which mainly reflects the suitability of the natural conditions for agriculture. Multi-social factors co-impacted land use activities and induced temporal and spatial variations of the cropland. These factors included food supply, world trade, wars, agriculture policies, the economic and political environment, urbanization, and national differences. Compared with the widely used global land use dataset Historical Database of the Global Environment (HYDE), this study shows different cropland change curves before 1950. This article is a case study showing that the hindcasting model of the HYDE dataset has difficulty estimating multi-determined cropland change in the Scandinavian Peninsula, and the empirical study contributes to the improvement of the accuracy of historical land use data at the regional level.

  • Flow Cytometric Measurement of Blood Cells with BCR-ABL1 Fusion Protein in Chronic Myeloid Leukemia

    • Chronic myeloid leukemia (CML) is characterized in the majority of cases by a t(9;22)(q34;q11) translocation, also called the Philadelphia chromosome, giving rise to the BCR-ABL1 fusion protein. Current treatment with tyrosine kinase inhibitors is directed against the constitutively active ABL1 domain of the fusion protein, and minimal residual disease (MRD) after therapy is monitored by real-time quantitative PCR (RQ-PCR) of the fusion transcript. Here, we describe a novel approach to detect and enumerate cells positive for the BCR-ABL1 fusion protein by combining the in situ proximity ligation assay with flow cytometry as readout (PLA-flow). By targeting of the BCR and ABL1 parts of the fusion protein with one antibody each, and creating strong fluorescent signals through rolling circle amplification, PLA-flow allowed sensitive detection of cells positive for the BCR-ABL1 fusion at frequencies as low as one in 10,000. Importantly, the flow cytometric results correlated strongly to those of RQ-PCR, both in diagnostic testing and for MRD measurements over time. In summary, we believe this flow cytometry-based method can serve as an attractive approach for routine measurement of cells harboring BCR-ABL1 fusions, also allowing simultaneously assessment of other cell surface markers as well as sensitive longitudinal follow-up.

  • Acute hyperinsulinemia differentially regulates interstitial and circulating adiponectin oligomeric pattern in lean and insulin-resistant, obese individuals

    • CONTEXT: Hyperinsulinemia emerges as a negative modulator of the circulating high-molecular-weight adiponectin multimers. OBJECTIVES: Here we asked whether, in vivo, acute hyperinsulinemia regulates adiponectin formation and oligomeric complex distribution at the transcriptional or posttranslational level. DESIGN: Nine lean and nine uncomplicated obese males were studied in the postabsorptive state and during a euglycemic-hyperinsulinemic clamp combined with the microdialysis technique. Subcutaneous abdominal adipose tissue biopsies and interstitial and serum samples were taken at baseline and after the hyperinsulinemia. Adiponectin complexes were characterized by nonheating/nonreducing SDS-PAGE. RESULTS: At baseline, serum and interstitial total adiponectin levels were lower (P < 0.01) in obese than in lean subjects primarily due to a reduction of the high-molecular-weight isoforms. After hyperinsulinemia, serum and interstitial total adiponectin was reduced in both groups. The degree of adiponectin reduction was more prominent in interstitial fluid than in serum. Lean individuals showed an equal suppression of the high-, low-, and middle-molecular-weight adiponectin complexes both in serum and in situ (P < 0.01 vs. basal). In obese subjects, despite the lower interstitial adiponectin subfractions, insulin challenge reduced significantly the circulating middle-molecular-weight forms only. At the mRNA level, adiponectin and its receptors 1 and 2, as well as the abundance of the endoplasmic reticulum chaperone proteins ERp44 and Epsilonro1-Lalpha were similar within the groups, before and after the clamp. CONCLUSIONS: In human obesity, the impaired adiponectin oligomeric pattern in the circulation is mimicked at the tissue level, and hyperinsulinemia may differentially affect the compartmental distribution of the adiponectin complexes.

    • Human obesity has been associated with a dysregulation of the peripheral and adipose tissue (AT) endocannabinoid system (ES). The aim of this study was to elucidate the acute in vivo effects of insulin on gene expression of the cannabinoid type 1 (CB-1) and type 2 (CB-2) receptors, as well as of the fatty acid amide hydrolase (FAAH) in the sc abdominal adipose tissue (SCAAT). Nine lean (L) and 9 obese (OB), but otherwise healthy males were studied in the fasting state and during a euglycemic hyperinsulinemic clamp (40 mU/m2 * min(-1)). SCAAT biopsies were obtained at baseline and after 270 min of i.v. maintained hyperinsulinemia. The basal SCAAT gene expression pattern revealed an upregulation of the FAAH in the OB (p=0.03 vs L), whereas similar CB-1 and CB-2 mRNA levels were seen. Following hyperinsulinemia, the FAAH mRNA levels significantly increased approximately 2-fold in the L (p=0.01 vs baseline) but not in the OB. In contrast, insulin failed to significantly change both the adipose CB-1 and CB-2 gene expression. Finally, the FAAH gene expression positively correlated with the fasting serum insulin concentration (r 0.66; p=0.01), whereas an inverse association with the whole-body glucose disposal (r -0.58; p<0.05) was seen. Taken together, these first time observations demonstrate that the ES-related genes in the SCAAT differentially respond to hyperinsulinemia in lean/insulin-sensitive and in obese/insulin-resistant individuals. We suggest that insulin may play a key role in the obesity-linked dysregulation of the adipose ES at the gene level.

    • CONTEXT: The chemokine monocyte chemoattractant protein-1 (MCP-1) is implicated in obesity-associated chronic inflammation, insulin resistance, and atherosclerosis. OBJECTIVES: The objectives of this study were to: 1) characterize the interstitial levels and the gene expression of MCP-1 in the sc abdominal adipose tissue (SCAAT), 2) elucidate the response of MCP-1 to acute hyperinsulinemia, and 3) determine the relationship between MCP-1 and arterial stiffness. DESIGN: Nine lean (L) and nine uncomplicated obese (OB) males were studied in the fasting state and during a euglycemic-hyperinsulinemic clamp combined with the microdialysis technique. Interstitial and serum MCP-1 (iMCP-1 and sMCP-1, respectively) levels, pulse wave analysis, and SCAAT biopsies were characterized at baseline and after hyperinsulinemia. RESULTS: OB showed elevated sMCP-1 (P < 0.01) but similar iMCP-1 levels as compared with L. Basal iMCP-1 concentrations were considerably higher than sMCP-1 (P < 0.0001), and a gradient between iMCP-1 and sMCP-1 levels was maintained throughout the hyperinsulinemia. At baseline, SCAAT gene expression profile revealed a "co-upregulation" of MCP-1, MCP-2, macrophage inflammatory protein-1alpha, and CD68 in OB, and whole-body glucose disposal inversely correlated with the MCP-1 gene expression. After hyperinsulinemia, MCP-1 and MCP-2 mRNA levels significantly increased in L, but not in OB. Finally, sMCP-1 excess in the OB positively correlated with the stiffer vasculature. CONCLUSIONS: These observations demonstrate similar interstitial concentrations and a differential gene response to hyperinsulinemia of MCP-1 in the SCAAT from L and OB individuals. In human obesity, we suggest the SCAAT MCP-1 gene overexpression as a biomarker of an "inflamed" adipose organ and impaired glucose metabolism.

    • Land use is a cornerstone of human civilization, but also intrinsically linked to many global sustainability challenges—from climate change to food security to the ongoing biodiversity crisis. Understanding the underlying technological, institutional and economic drivers of land-use change, and how they play out in different environmental, socio-economic and cultural contexts, is therefore important for identifying effective policies to successfully address these challenges. In this regard, much can be learned from studying long-term land-use change. We examined the evolution of European land management over the past 200 years with the aim of identifying (1) key episodes of changes in land management, and (2) their underlying technological, institutional and economic drivers. To do so, we generated narratives elaborating on the drivers of land use-change at the country level for 28 countries in Europe. We qualitatively grouped drivers into land-management regimes, and compared changes in management regimes across Europe. Our results allowed discerning seven land-management regimes, and highlighted marked heterogeneity regarding the types of management regimes occurring in a particular country, the timing and prevalence of regimes, and the conditions that result in observed bifurcations. However, we also found strong similarities across countries in the timing of certain land-management regime shifts, often in relation to institutional reforms (e.g., changes in EU agrarian policies or the emergence and collapse of the Soviet land management paradigm) or to technological innovations (e.g., drainage pipes, tillage and harvesting machinery, motorization, and synthetic fertilizers). Land reforms frequently triggered changes in land management, and the location and timing of reforms had substantial impacts on land-use outcomes. Finally, forest protection policies and voluntary cooperatives were important drivers of land-management changes. Overall, our results demonstrate that land-system changes should not be conceived as unidirectional developments following predefined trajectories, but rather as path-dependent processes that may be affected by various drivers, including sudden events.

    • We have investigated theoretically the adsorption of molecules onto graphene with divacancy defects. Using ab initio density-functional calculations, we have found that O-2, CO, N-2, B-2, and H2O molecules all interact strongly with a divacancy in a graphene layer. Along with a complex geometry of the molecule-graphene bonding, metallic behavior of the graphene layer in the presence of CO and N-2 molecules has been  found with a large density of states in the vicinity of the Fermi level suggesting an increase in the conductivity. The adsorption of N-2 is particularly interesting since the N atoms dissociate in the vicinity   of the defects and take the place where the missing C atoms of the divacancy used to sit. In this way, the defected graphene structure is healed geometrically, and at the same time doped with electron states.

    • This article presents a discussion of the emerging ethical issue of geodata privacy in geographical research. The paper highlights the importance of considering challenges to privacy when working with geographically explicit data and explores explicit ways in which researchers and practitioners can be conscious of these issues. Through summarising the key problems in this area and presenting outstanding open research areas and questions from a seminar series on geodata privacy, we highlight important considerations for future research in this field. We focus on the specific topics of appropriate anonymization, responsible data dissemination, the balance between data sharing and privacy, and the challenges posed by working across international contexts. We conclude by recommending approaches to manage various legal and ethical frameworks, raise the importance of the international context, and inspire future research to address the challenges of safeguarding sensitive geodata while promoting openness and transparency.

    • A facile and inexpensive method to produce thin films of nanostructured tungsten oxide is described. A nanocrystalline tungstite (WO3·H2O) film is spontaneously formed when a tungsten substrate is immersed in nitric acid at elevated temperatures. The resulting thin film is composed of plate-like tungstite crystals with edges preferentially directed out from the substrate surface. The tungstite can easily be transformed into WO3 by annealing. Patterned WO3·H2O/W structures can be obtained by a combination of lithographic techniques and etching. In this study, the effect of exposure time, acid concentration, and temperature on the microstructure of the films has been investigated. The potential of this inexpensive synthesis method to produce large-area coatings of nanostructured tungsten oxide as well as patterned films makes it interesting for several different applications, such as batteries, gas sensors, and photocatalysts.
