    • Over the past two decades, the development of methods for visualizing and analysing specimens digitally, in three and even four dimensions, has transformed the study of living and fossil organisms. However, the initial promise that the widespread application of such methods would facilitate access to the underlying digital data has not been fully achieved. The underlying datasets for many published studies are not readily or freely available, introducing a barrier to verification and reproducibility, and the reuse of data. There is no current agreement or policy on the amount and type of data that should be made available alongside studies that use, and in some cases are wholly reliant on, digital morphology. Here, we propose a set of recommendations for minimum standards and additional best practice for three-dimensional digital data publication, and review the issues around data storage, management and accessibility.

    • We summarise the findings of arthropods collected by fogging the canopy of 24 pine trees in two sites in Eastern and Western Norway. From the samples, taken in 1998 and in 1999, almost 30,000 specimens were determined to 512 species, with Diptera being most species rich (210 species), followed by Coleoptera (76 species) and Araneae (49 species). Of the 96 new species records, nine were new to science (5 Diptera and 4 Oribatida), two were new to the European, three to the Scandinavian and 82 to the Norwegian faunas. The paper demonstrates the need for detailed faunistical inventories of European forests.

    • Background-—Ticagrelor reduced cardiovascular death, myocardial infarction (MI), or stroke in patients with prior MI in PEGASUSTIMI 54 (Prevention of Cardiovascular Events [eg, Death From Heart or Vascular Disease, Heart Attack, or Stroke] in Patients With Prior Heart Attack Using Ticagrelor Compared to Placebo on a Background of Aspirin). MI can occur in diverse settings and with varying severity; therefore, understanding the types and sizes of MI events prevented is of clinical importance. Methods and Results-—MIs were adjudicated by a blinded clinical events committee and categorized by subtype and fold elevation of peak cardiac troponin over the upper limit of normal. A total of 1042 MIs occurred in 898 of the 21 162 randomized patients over a median follow-up of 33 months. The majority of the MIs (76%) were spontaneous (Type 1), with demand MI (Type 2) and stent thrombosis (Type 4b) accounting for 13% and 9%, respectively; sudden death (Type 3), percutaneous coronary intervention–related (Type 4a) and coronary artery bypass graft–related (Type 5) each accounted for <1%. Half of MIs (520, 50%) had a peak troponin ≥10x upper limit of normal and 21% of MIs (220) had a peak troponin ≥1009 upper limit of normal. A total of 21% (224) were ST-segment–elevation MI STEMI. Overall ticagrelor reduced MI (4.47% versus 5.25%, hazard ratio 0.83, 95% confidence interval 0.72–0.95, P=0.0055). The benefit was consistent among the subtypes, including a 31% reduction in MIs with a peak troponin ≥1009 upper limit of normal (hazard ratio 0.69, 95% confidence interval 0.53–0.92, P=0.0096) and a 40% reduction in ST-segment elevation MI (hazard ratio 0.60, 95% confidence interval 0.46–0.78, P=0.0002). Conclusions-—In stable outpatients with prior MI, the majority of recurrent MIs are spontaneous and associated with a high biomarker elevation. Ticagrelor reduces the MI consistently among subtypes and sizes including large MIs and ST-segment elevation MI. Clinical Trial Registration-URL: https://www.clinicaltrials.gov.     • The precisionFDA Truth Challenge V2 aimed to assess the state-of-the-art of variant calling in difficult-to-map regions and the Major Histocompatibility Complex (MHC). Starting with FASTQ files, 20 challenge participants applied their variant calling pipelines and submitted 64 variant callsets for one or more sequencing technologies (~35X Illumina, ~35X PacBio HiFi, and ~50X Oxford Nanopore Technologies). Submissions were evaluated following best practices for benchmarking small variants with the new GIAB benchmark sets and genome stratifications. Challenge submissions included a number of innovative methods for all three technologies, with graph-based and machine-learning methods scoring best for short-read and long-read datasets, respectively. New methods out-performed the 2016 Truth Challenge winners, and new machine-learning approaches combining multiple sequencing technologies performed particularly well. Recent developments in sequencing and variant calling have enabled benchmarking variants in challenging genomic regions, paving the way for the identification of previously unknown clinically relevant variants.

    • Background: Lanreotide Autogel® is supplied in prefilled syringes. Therefore, it is possible for patients with neuroendocrine tumors to use self-/partner-administered injections. The primary objective of this study was to assess the proportion of patients preferring self/partner injections over injections administered by health care professionals, and to describe the impact of self/partner administration on efficacy, safety, and costs. Methods: Of 62 eligible patients, 26 (42%) patients with neuroendocrine tumors treated with a stable dose of lanreotide Autogel 90 mg or 120 mg every 4 weeks agreed to participate in this Phase IV, international, open-label, crossover study, conducted at hospitals in Sweden, Norway, and Denmark. Patients were randomized to two blocks, starting with administration of lanreotide Autogel by either self/partner or a health care professional. Preference for injections administered by self/partner or health care professionals was measured, as well as efficacy, safety, and health care resource utilization (both direct and indirect costs). Results: Of 25 evaluable patients, 22 (88%) preferred self/partner injections, mainly because they experienced increased independence. Based on all patients asked to participate (n = 62), 35% preferred self/partner injections on a regular basis. There was no difference in efficacy or safety between the two administration blocks. Conclusion: Many patients with neuroendocrine tumors prefer self/partner injection of lanreotide Autogel, and are able to self/partner inject without any impact on efficacy or safety. This administration method seems to provide a good alternative for suitable patients to increase patient independence and reduce the number of clinic visits.

    • Ash formation during the bubbling fluidized bed (BFB) combustion of bark and pulp mill sludge has been studied on an industrial and bench scale. During co-firing in an industrial BFB a submicron fly ash mode was formed via condensation of volatilized K, Na, S and Cl species at 0.05-0.3 μm. The submicron mass mode below 0.3 μm made up 2.2-5.0% of the fly ash, while the share of the supermicron mass fraction was 93.6-97.2%. Elements depleted in the ultrafine ash were Ca, Si, Al, Mg, Fe, Mn, P and Ti. The bench-scale test showed that the ultrafine particle concentration was increased by a higher bed temperature and decreased due to sludge moisture. As, Cd, Pb and Rb were enriched in the ultrafine ash on a bench scale, while Ba, Co, Sr and V were depleted. Cu and Zn were enriched in the ultrafine ash during the combustion of dried sludge, but not when wet sludge was fired. Micron-size ash particles composed of non-volatile species, Ca, Si, Mg, Al, P and Mn, adhered to the bed sand, presumably by surface forces, and sintering densified the ash layer. © 1998 OPA (Overseas Publishers Association) N.V.     • In the absence of RecA-mediated cleavage of the repressor, the X prophage is exceptionally stable. We. develop a stochastic model that predicts the stability of such epigenetic states from affinities of the molecular components. We find that the stability, in particular, depends on the maximum possible cI protein production. and on the number of translated cro proteins per transcribed mRNA. We apply the model to the behavior of recently published mutants of O-R and find, in particular, that a mutant that overexpress cro behaves in a different way than what was predicted, thus suggesting that the current view of the O-R switch is incomplete. The approach described here should be generally applicable to the stability of expressed states.

    • The stratospheric ozone layer has been depleted at high and mid-latitudes as a consequence of man's pollution of the atmosphere, and this results in increasing ultraviolet-B radiation at ground level. We investigate the effects of further radiation increases on plants and ecosystems by irradiating natural sub-Arctic and Arctic vegetation with artificial W-B radiation in field experiments extending over several years. Our experimental sites are located at Abisko, in northern Sweden (68 degrees N), and Adventdalen, on the island of Spitsbergen (78 degrees N). Additional UV-B induced interspecific differences in plant response in terms of reduced (or, in one case, increased) growth, changed morphology and changed pigment content. In some cases effects seem to be accumulated from one year to another. Plant litter decomposition is retarded. We are also studying how UV-B enhancement may affect the interaction between species. In some experiments we combine UV-B enhancement with changes in other factors: carbon dioxide concentration, water availability, and temperature. In some cases the effect of radiation enhancement is modified, or even reversed, by such changes. Over a four year period we did not find any significant radiation induced change in species composition, but based on the effects on individual plant species, such changes can be expected to take place over a longer time.
