    • Today’s media environment provides people ample opportunities for constructing news habits fitting their preferences, but our knowledge about the dynamics of such news habits is limited. Using a four-wave panel survey from Sweden and taking a news repertoires approach, the study identifies four groups of news users labeled Public service-oriented traditionalists, Minimalists, Engaged pluralists, and Quality-oriented explorers, which are each related to news interest, trust in mainstream news media, and socio-demographic factors in distinct ways. The news repertoires are highly stable, even during profound contextual change, showing that people most often maintain their news habits and only seldom reform them.

    • PURPOSE: To elucidate the therapeutic efficacy of alpha-radioimmunotherapy of ovarian cancer in mice. This study: (i) estimated the minimum required activity (MRA), giving a reasonable high therapeutic efficacy; and (ii) calculated the specific energy to tumor cell nuclei and the metastatic cure probability (MCP) using various assumptions regarding monoclonal-antibody (mAb) distribution in measured tumors. The study was performed using the alpha-particle emitter Astatine-211 (211At) labeled to the mAb MX35 F(ab')2. METHODS AND MATERIALS: Animals were inoculated intraperitoneally with approximately 1 x 10(7) cells of the cell line NIH:OVCAR-3. Four weeks later animals were treated with 25, 50, 100, or 200 kBq 211At-MX35 F(ab')2 (n = 74). Another group of animals was treated with a nonspecific mAb: 100 kBq 211At-Rituximab F(ab')2 (n = 18). Eight weeks after treatment the animals were sacrificed and presence of macro- and microscopic tumors and ascites was determined. An MCP model was developed and compared with the experimentally determined tumor-free fraction (TFF). RESULTS: When treatment was given 4 weeks after cell inoculation, the TFFs were 25%, 22%, 50%, and 61% after treatment with 25, 50, 100, or 200 kBq (211)At-MX35 F(ab')2, respectively, the specific energy to irradiated cell nuclei varying between approximately 2 and approximately 400 Gy. CONCLUSION: As a significant increase in the therapeutic efficacy was observed between the activity levels of 50 and 100 kBq (TFF increase from 22% to 50%), the conclusion was that the MRA is approximately 100 kBq (211)At-MX35 F(ab')2. MCP was most consistent with the TFF when assuming a diffusion depth of 30 mum of the mAbs in the tumors.

    • The purpose of this work was to (a) investigate the efficacy of radioimmunotherapy using 211At-MX35 F(ab')2 or 211At-Rituximab F(ab')2 (nonspecific antibody) against differently advanced ovarian cancer in mice; (b) image the tumor growth on the peritoneum; and (c) calculate the specific energy and mean absorbed dose to tumors and critical organs. METHODS: Two experiments with 5-wk-old nude mice (n = 100 + 93), intraperitoneally inoculated with approximately 1 x 10(7) NIH:OVCAR-3 cells, were done. At either 1, 3, 4, 5, or 7 wk after inoculation animals were intraperitoneally treated with approximately 400 kBq 211At-MX35 F(ab')2 (n = 50 + 45), approximately 400 kBq 211At-Rituximab F(ab')2 (n = 25 + 24), or unlabeled Rituximab F(ab')2 (n = 25 + 24). At the time of treatment 29 animals were sacrificed and biopsies were taken for determination of tumor sizes using scanning electron microscopy (SEM). Eight weeks after each treatment the animals were sacrificed and the presence of macro- and microscopic tumors and ascites was determined. The specific energy and mean absorbed dose to tumors were calculated. The activity concentration was measured in critical organs and abdominal fluid. RESULTS: When given treatment 1, 3, 4, 5, or 7 wk after cell inoculation the tumor-free fraction (TFF) was 95%, 68%, 58%, 47%, 26%, and 100%, 80%, 20%, 20%, and 0% when treated with 211At-MX35 F(ab')2 or 211At-Rituximab F(ab')2, respectively. The SEM images revealed maximum tumor radius of approximately 30 mum 1 wk after cell inoculation, increasing to approximately 340 mum at 7 wk. Specific energy to cell nuclei varied between 0 and approximately 540 Gy, depending on assumptions regarding activity distribution and tumor size. The mean absorbed dose to thyroid, kidneys, and bone marrow was approximately 35, approximately 4, and approximately 0.3 Gy, respectively. CONCLUSION: Treatment with 211At-MX35 F(ab')2 or 211At-Rituximab F(ab')2 resulted in a TFF of 95%-100% when the tumor radius was < or =30 microm. The TFF was decreased (TFF < or = 20%) for 211At-Rituximab F(ab')2 when the tumor radius exceeded the range of the alpha-particles. The specific antibody gave for these tumor sizes a significantly better TFF, explained by a high mean absorbed dose (>22 Gy) from the activity bound to the tumor surface and probably some contribution from penetrating activity.

    • Föreliggande rapport har utarbetats vid CEFOS, Centrum för forskning om offentlig sektor, Göteborgs universitet. Huvudsyftet har varit att ta fram aktuell och systematisk kunskap om den betydelse kommunstorleken har för hur den kommunala demokratin i Sverige fungerar. Rapporten bygger till största delen på reanalyser av ett befintligt omfattande empiriskt material som innefattar kommunstudier av såväl medborgare som politiker, studier av massmediers innehåll och de mer generella enkätundersökningar som sedan lång tid utförs vid SOM-institutet, Göteborgs universitet. Beträffande politiker har också nytt material samlats in. Huvudansvariga för rapporten är professor Folke Johansson och fil dr David Karlsson. Docent Bengt Johansson har medverkat med analys av massmedias roll och fil dr Ylva Norén Bretzer har medverkat med analys av förtroende för politiker.

    • Across the globe, extreme weather events have brought climate change into people’s daily lives. Extended heatwaves, droughts, and wildfires are now recurring in many regions across the globe. This study asks how the exceptional 2018 summer influenced climate change beliefs among Swedish citizens. More specifically, the study looks deeper into belief formation dynamics under intense, consonant, and extended news reporting–addressing one of the most fundamental media effects in the literature: the over-time maintenance of societal beliefs through cumulative and repetitive exposure to a dominant issue frame. Using a unique three-wave panel survey, the analysis focuses on citizens’ acceptance (and resistance) of the dominant climate change frame provided by traditional media–whether citizens believe in the existence, causes, and consequences of climate change. The findings reveal strong support for belief maintenance effects over time, but also that belief changes are possible. Even in situations of intense and consonant news reporting, acceptance (and resistance) of the dominant climate change frame depends on citizens’ personal media orientations. Both trust in traditional news media and usage of alternative online news emerge as key factors conditioning classic media effects in a fragmented and polarized media environment.

    • A fundamental precondition for the legitimacy of democracy is a high level of trust in the electoral process. It is therefore concerning that the number of appeals regarding electoral fraud has increased, that allegations of frauds circulate on social media, and that research indicates that a significant portion of the Swedish population does not fully trust that elections are conducted in a correct manner. At the same time, there is a lack of studies examining the nature of electoral distrust and the prevalence of conspiratorial beliefs regarding Swedish elections. Hence, the purpose of this study is to investigate (a) the extent of belief in irregulari-ties related to vote counting and ballot placement, (b) whether the proportion of those believing in such irregularities changes during an election campaign, and (c) the factors driving belief in conspiratorial claims of electoral fraud. The results of a panel study conducted during the 2022 election campaign indicate that the major-ity does not believe in conspiratorial claims of electoral fraud. However, there is a minority—characterized by a high degree of conspiracy thinking—who believes irregularities are common during Swedish elections. Furthermore, the results sug-gest that other explanatory factors for belief in electoral fraud include the use of right-wing alternative media and sympathy towards the Sweden Democrats.

    • Tumour infiltration by activated natural killer (A-NK) cells is a pre-requisite for tumour eradication by adoptive NK cell transfer. Extravasated A-NK cells do not always succeed in reaching the crucial target cell conjugation. Therefore, we wished to study A-NK cell locomotion and interactions with melanoma cells in a matrix environment (Matrigel) by electron, confocal and fluorescence microscopy. Two distinct patterns of A-NK cell-mediated matrix disintegration were revealed during incubation of tumour cells and A-NK cells in Matrigel: (1) A-NK cells pre-cultured for 5 days altered the homogeneous texture of the Matrigel, an initial microporous appearance became a loose filamentous meshwork by 24 h. Matrix degrading protease inhibitors could not fully prevent this, but could delay the process; and (2) A-NK cells pre-cultured for 6 days or more, instead formed large excavations in the Matrigel leaving the remaining matrix less affected compared to the effects by the younger A-NK cells. By histochemical staining with Cupromeronic Blue, the excavations were shown to contain proteoglycan material. Protease inhibitors had no discernable effect on the development of the excavations. The conspicuous capacity of A-NK cells to disintegrate extracellular matrix and the formation of large excavations seems only partially to depend on matrix-degrading proteases. Formation of extracellular proteoglycan material is suggested to facilitate A-NK cell locomotion within a matrix environment.

    • This article proposes and explores the notion of “media micro-generations”. Based on a survey of values and norms in relation to media-related behaviour in Sweden, we identify statistically significant media micro-generations. Through an analysis of the technologies that were introduced during the formative years of different media micro-generations, we propose that media micro-generations are formed with the introduction of new media technologies. Thus, the existence of media micro-generations illustrates how rapid trans- formations of media technologies can shape the moral notions of narrow age groups. It also explains why many earlier studies have detected a rather large span of years (1970-1985, in between the TV generation and the internet generation) during which no generational identity seems to have been formed.

    • Sleep-disordered breathing (SDB) is common in patients with heart failure (HF) and leads to disturbed sleep. The objective of this study was to determine the persistent effects of long-term nocturnal oxygen treatment in patients with severe HF regarding (1) objective outcomes, such as steep. SDB, cardiac function, and functional capacity; (2) subjective outcomes, such as self-assessed sleep difficulties, daytime sleepiness, and health-related quality of life (HRQOL); and (3) the relationship between objective and subjective outcomes. In this open nonrandomized experimental study, 22 patients, median age 71 years, with severe HF were studied before and after 3 months of receiving nocturnal oxygen. The measures used were overnight polysomnography, echocardiography, 6-minute walk test, self-assessed sleep difficulties (Uppsala Sleep Inventory-HF), daytime sleepiness (Epworth Sleepiness Scale), and HRQOL (36-ltem Short Form Health Survey and Minnesota Living with Heart Failure Questionnaire). SDB, with a 90% dominance of central sleep apnea, occurred in 41 % of the patients with severe HF before intervention. After intervention, functional capacity improved for both the whole group of patients with HF (P < .01) and HF patients with SDB (P < .05). No improvements regarding cardiac function, objective sleep, subjective sleep, or SDB were seen, except for a decrease of ‚â•4% desaturations (P < 05). HRQOL did not differ significantly between HF patients with and without SDB before or after intervention with nocturnal oxygen. Long-term nocturnal oxygen treatment improved functional capacity in patients with severe HF, with or without SDB. No improvements were seen regarding sleep, daytime sleepiness, SDB, cardiac function, or HRQOL.

    • Soluble triggering receptor expressed on myeloid cells 2 (sTREM2) in cerebrospinal fluid (CSF) has been associated with Alzheimer's disease (AD). TREM2 plays a critical role in microglial activation, survival, and phagocytosis; however, the pathophysiological role of sTREM2 in AD is not well understood. Understanding the role of sTREM2 in AD may reveal new pathological mechanisms and lead to the identification of therapeutic targets. We performed a genome-wide association study (GWAS) to identify genetic modifiers of CSF sTREM2 obtained from the Alzheimer's Disease Neuroimaging Initiative. Common variants in the membrane-spanning 4-domains subfamily A (MS4A) gene region were associated with CSF sTREM2 concentrations (rs1582763; P = 1.15 x 10(-15)); this was replicated in independent datasets. The variants associated with increased CSF sTREM2 concentrations were associated with reduced AD risk and delayed age at onset of disease. The single-nucleotide polymorphism rs1582763 modified expression of the MS4A4A and MS4A6A genes in multiple tissues, suggesting that one or both of these genes are important for modulating sTREM2 production. Using human macrophages as a proxy for microglia, we found that MS4A4A and TREM2 colocalized on lipid rafts at the plasma membrane, that sTREM2 increased with MS4A4A overexpression, and that silencing of MS4A4A reduced sTREM2 production. These genetic, molecular, and cellular findings suggest that MS4A4A modulates sTREM2. These findings also provide a mechanistic explanation for the original GWAS signal in the MS4A locus for AD risk and indicate that TREM2 may be involved in AD pathogenesis not only in TREM2 risk-variant carriers but also in those with sporadic disease.

    • This article examines whether shifts in news media attention to societal issues matter for how strong beliefs citizens have about those issues. Based on an issue signal approach, in which media salience is conceptualized as signal strength, the study analyzes whether sociotropic beliefs become more prevalent, extreme, and certain when news media salience rises, and less prevalent, extreme, and certain when media salience drops. Using a four-wave panel survey dataset collected over a two-year period, the empirical analysis links media content analyses of issue salience to panel survey data, comparing four issues with different levels of baseline salience and political controversy: violent crimes, immigration, climate change, and antibiotic resistance. The analysis shows that issue-specific news media exposure and issue-specific use of alternative media offer two different pathways to the formation of beliefs. The hypothesized relationship with news media salience was supported for the two controversial issues with high baseline salience (immigration and violent crimes), but not for climate change and antibiotic resistance. The results indicate that issue attributes matter and that audiences may respond differently to salience shifts depending on the level of controversy of the issue.

    • Political accountability is fundamental in a democratic society. Societal changes such as the marketization of the public sector have, however, made accountability issues complex and negotiable. The question of who is to be held to account for policy failures is increasingly a subject of struggle within the media. The aim of this article is to examine how journalism does “accountability work” in a political setting marked by new public management. The empirical study focuses on an example of intensive news coverage of the mistreatment of elderly people in private health care, in Sweden, 2011. A corpus of 156 news items is analyzed. The analysis focuses on the use of accountability interviews, and how journalism constructs boundaries of political accountability by framing social problems. In general, the study shows that the political accountability work carried out was weak and restricted, the problems were constructed as a moral scandal instead of a policy failure.

    • This study investigates how political accountability, as a key democratic principle, is performed in the media and how the practices and representations of accountability are transformed over time by the influences of mediatization. The implications of mediatization are analyzed with a focus on how aspects of media dramaturgy and independent journalism influence news reporting of political responsibility and how politicians are held to account in the media. The effects of mediatization are understood as conditioned by other structural changes in political life, in particular depoliticization. The empirical study is designed as a comparison of news reports on two national industrial crises in 1980–1982 and 2008–2011, and is based on content analyses of daily morning newspapers, evening tabloids, and regional and local newspapers. The study provides evidence for a non-linear understanding of mediatization. Significant aspects of media dramaturgy are shown to be rather stable between the two time periods whilst the journalistic independence and interpretations increase as expected. The hypothesis of depoliticization receives some support although the results are not unambiguous.

    • Matrix metalloproteinases (MMPs) are thought to be of importance for the migratory ability of natural killer (NK) cells. Their expression and production may influence the amount of tumour-infiltrating NK cells and thereby any therapeutic capability. In this study, we sought to investigate the importance of MMPs for human NK cells' ability to degrade and migrate through the extracellular matrix (ECM). The two human NK cell lines, NK-92 and YT, migratory ability, MMP expression and production as well as their morphological appearance when cultured in the ECM equivalent Matrigel were analysed and compared. The quantitatively more migratory NK-92 cells were found to express invadopodia/podosomes at a significantly higher degree when cultured in Matrigel and gave rise to a general disintegration of the Matrigel. The NK-92 cells had a higher mRNA expression of MMP-2, -9, -13, MT1-, MT3- and MT6-MMP and a significantly higher production of MMP-9 compared to YT cells. These differences could explain the substantial functional difference observed between the two cell lines with respect to migratory capacity. In addition, the number of Matrigel invading NK-92 cells decreased significantly in the presence of the MMP inhibitor GM6001, demonstrating that MMPs have a critical function in their migration.

    • This study, based on Swedish data from three elections (2002, 2006 and 2010) and on a revised version of Clayman's and Heritage's conceptualization of aggressive questioning, examines bias in election campaign interviews with leading political figures. In the first part of the study, the prevalence of partisan bias is explored, and this analysis confirms that such bias does not exist. Informed by Conversation Analysis, a limited number of interviews from the 2006 election are investigated in the second part. This analysis also involves questions scripted by journalists, and it compares both quantitatively and qualitatively the differences between the manuscripts and live interaction. The results question the assumption that bias is solely related to journalistic values and actions. The level of aggressiveness in the interviews is also dependent on how the politicians manage the interview questions.

    • The article presents a study on the role of daily news reporting in the scrutiny of local politics. Political accountability is defined as an important aspect of scrutiny. The study shows that the kind of scrutinizing often mentioned in the literature, characterized by thorough investigations and disclosures of political wrongdoings, nearly doesn’t exist at all in the local press. However, by identifying other forms of scrutinizing more closely related to ordinary news reporting, the study shows that one third of the articles are in some aspect scrutinizing, out of 1500 analysed articles. An overall conclusion is that the local press plays an important role in communicating information and critique concerning how local authority service areas works, and how political responsibilities are fulfilled. The study also indicates that this role has been strengthened during the second half of the 20th twentieth century. The press in three different municipalities has been studied at three different points of time (1961, 1981 and 2001).

    • News interviews are contexts in which political responsibilities are articulated and negotiated. Although the accountability interview is recognized as a commonsense practice in journalism, and the research on political interviews is substantial, it partly remains to explore how responsibilities are negotiated in different forms of questioning in journalism. This study investigates three generic forms of questioning: accountability questioning, the clarifying of promises, and the principal assessments. Key features of the different forms are specified. The empirical study investigates how the questionings are performed in news on industrial crises in Sweden, in the different political regimes in the 1970s and the 2000s. The data consist of 27 news interviews with the formally responsible Ministers. The method is based on Conversation Analysis and includes detailed analyses of questions and answers. The study shows (1) how the forms of questioning differ when it comes to the action agenda and in how responsibilities are invoked and negotiated; (2) how the questionings tend to reproduce particular expectations of Governmental interventions; (3) how increased assertiveness and adversarialness in interviewing coincide with reduced expectations of political responsibilities.
