    • We report on the final electroweak measurements performed with data taken at the Z resonance by the experiments operating at the electron-positron colliders SLC and LEP. The data consist of 17 million Z decays accumulated by the ALEPH, DELPHI, L3 and OPAL experiments at LEP, and 600 thousand Z decays by the SLID experiment using a polarised beam at SLC. The measurements include cross-sections, forward-backward asymmetries and polarised asymmetries. The mass and width of the Z boson, m(Z) and Gamma(Z), and its couplings to fermions, for example the p parameter and the effective electroweak mixing angle for leptons, are precisely measured: m(Z) = 91.1875 +/- 0.0021 GeV, Gamma(Z) = 2.4952 +/- 0.0023 GeV, rho(l) = 1.0050 +/- 0.0010, sin(2)theta(eff)(lept) = 0.23153 +/- 0.00016. The number of light neutrino species is determined to be 2.9840 +/- 0.0082, in agreement with the three observed generations of fundamental fermions. The results are compared to the predictions of the Standard Model (SM). At the Z-pole, electroweak radiative corrections beyond the running of the QED and QCD coupling constants are observed with a significance of five standard deviations, and in agreement with the Standard Model. Of the many Z-pole measurements, the forward-backward asymmetry in b-quark production shows the largest difference with respect to its SM expectation, at the level of 2.8 standard deviations. Through radiative corrections evaluated in the framework of the Standard Model, the Z-pole data are also used to predict the mass of the top quark, m(t) = 173(+10)(+13) GeV, and the mass of the W boson, m(W) = 80.363 +/- 0.032 GeV. These indirect constraints are compared to the direct measurements, providing a stringent test of the SM. Using in addition the direct measurements of m(t) and m(W), the mass of the as yet unobserved SM Higgs boson is predicted with a relative uncertainty of about 50% and found to be less than 285 GeV at 95% confidence level.   • Associations of autozygosity with a broad range of human phenotypes

    • In many species, the offspring of related parents suffer reduced reproductive success, a phenomenon known as inbreeding depression. In humans, the importance of this effect has remained unclear, partly because reproduction between close relatives is both rare and frequently associated with confounding social factors. Here, using genomic inbreeding coefficients (FROH) for >1.4 million individuals, we show that FROH is significantly associated (p < 0.0005) with apparently deleterious changes in 32 out of 100 traits analysed. These changes are associated with runs of homozygosity (ROH), but not with common variant homozygosity, suggesting that genetic variants associated with inbreeding depression are predominantly rare. The effect on fertility is striking: FROH equivalent to the offspring of first cousins is associated with a 55% decrease [95% CI 44–66%] in the odds of having children. Finally, the effects of FROH are confirmed within full-sibling pairs, where the variation in FROH is independent of all environmental confounding.

  • Effects of chemotherapy and hormonal therapy for early breast cancer on recurrence and 15-year survival: an overview of the randomised trials

    • Background Quinquennial overviews (1985-2000) of the randomised trials in early breast cancer have assessed the 5-year and 10-year effects of various systemic adjuvant therapies on breast cancer recurrence and survival. Here, we report the 10-year and 15-year effects. Methods Collaborative meta-analyses were undertaken of 194 unconfounded randomised trials of adjuvant chemotherapy or hormonal therapy that began by 1995. Many trials involved CMF (cyclophosphamide, methotrexate, fluorouracil), anthracycline-based combinations such as FAC (fluorouracil, doxombicin, cyclophosphamide) or FEC (fluorouracil, epirubicin, cyclophosphamide), tamoxifen, or ovarian suppression: none involved taxanes, trastuzumab, raloxifene, or modem aromatase inhibitors. Findings Allocation to about 6 months of anthracycline-based polychemotherapy (eg, with FAC or FEC) reduces the annual breast cancer death rate by about 38% (SE 5) for women younger than 50 years of age when diagnosed and by about 20% (SE 4) for those of age 50-69 years when diagnosed, largely irrespective of the use of tamoxifen and of oestrogen receptor (ER) status, nodal status, or other tumour characteristics. Such regimens are significantly (2p=0 . 0001 for recurrence, 2p<0 . 00001 for breast cancer mortality) more effective than CMF chemotherapy. Few women of age 70 years or older entered these chemotherapy trials. For ER-positive disease only, allocation to about 5 years of adjuvant tamoxifen reduces the annual breast cancer death rate by 31% (SE 3), largely irrespective of the use of chemotherapy and of age (<50, 50-69, &GE; 70 years), progesterone receptor status, or other tumour characteristics. 5 years is significantly (2p<0 . 00001 for recurrence, 2p=0 . 01 for breast cancer mortality) more effective than just 1-2 years of tamoxifen. For ER-positive tumours, the annual breast cancer mortality rates are similar during years 0-4 and 5-14, as are the proportional reductions in them by 5 years of tamoxifen, so the cumulative reduction in mortality is more than twice as big at 15 years as at 5 years after diagnosis. These results combine six meta-analyses: anthracycline-based versus no chemotherapy (8000 women); CMF-based versus no chemotherapy (14 000); anthracycline-based versus CMF-based chemotherapy (14 000); about 5 years of tamoxifen versus none (15 000); about 1-2 years of tamoxifen versus none (33 000); and about 5 years versus 1-2 years of tamoxifen (18 000). Finally, allocation to ovarian ablation or suppression (8000 women) also significantly reduces breast cancer mortality, but appears to do so only in the absence of other systemic treatments. For middle-aged women with ER-positive disease (the commonest type of breast cancer), the breast cancer mortality rate throughout the next 15 years would be approximately halved by 6 months of anthracycline-based chemotherapy (with a combination such as FAC or FEC) followed by 5 years of adjuvant tamoxifen. For, if mortality reductions of 38% (age <50 years) and 20% (age 50-69 years) from such chemotherapy were followed by a further reduction of 31% from tamoxifen in the risks that remain, the final mortality reductions would be 57% and 45%, respectively (and, the trial results could well have been somewhat stronger if there had been full compliance with the allocated treatments). Overall survival would be comparably improved, since these treatments have relatively small effects on mortality from the aggregate of all other causes. Interpretation Some of the widely practicable adjuvant drug treatments that were being tested in the 1980s, which substantially reduced 5-year recurrence rates (but had somewhat less effect on 5-year mortality rates), also substantially reduce 15-year mortality rates. Further improvements in long-term survival could well be available from newer drugs, or better use of older drugs.

  • Pan-cancer analysis of whole genomes

    • Cancer is driven by genetic change, and the advent of massively parallel sequencing has enabled systematic documentation of this variation at the whole-genome scale1–3. Here we report the integrative analysis of 2,658 whole-cancer genomes and their matching normal tissues across 38 tumour types from the Pan-Cancer Analysis of Whole Genomes (PCAWG) Consortium of the International Cancer Genome Consortium (ICGC) and The Cancer Genome Atlas (TCGA). We describe the generation of the PCAWG resource, facilitated by international data sharing using compute clouds. On average, cancer genomes contained 4–5 driver mutations when combining coding and non-coding genomic elements; however, in around 5% of cases no drivers were identified, suggesting that cancer driver discovery is not yet complete. Chromothripsis, in which many clustered structural variants arise in a single catastrophic event, is frequently an early event in tumour evolution; in acral melanoma, for example, these events precede most somatic point mutations and affect several cancer-associated genes simultaneously. Cancers with abnormal telomere maintenance often originate from tissues with low replicative activity and show several mechanisms of preventing telomere attrition to critical levels. Common and rare germline variants affect patterns of somatic mutation, including point mutations, structural variants and somatic retrotransposition. A collection of papers from the PCAWG Consortium describes non-coding mutations that drive cancer beyond those in the TERT promoter4; identifies new signatures of mutational processes that cause base substitutions, small insertions and deletions and structural variation5,6; analyses timings and patterns of tumour evolution7; describes the diverse transcriptional consequences of somatic mutation on splicing, expression levels, fusion genes and promoter activity8,9; and evaluates a range of more-specialized features of cancer genomes8,10–18.

  • A new silicon phosphide, Si12P5 : Formation conditions, structure, and properties

    • The formation conditions, structure, and properties of the recently discovered phosphide, Si12P5 (initially assigned Si7P3), have been studied using x-ray diffraction, high-resolution transmission electron microscopy, photospectrometry, and a four-point probe. The phosphide formed in amorphous Si-P alloy thin films with 30, 35 and 44 at. % P after annealing at 1000°C for 30 min. During annealing at 1100°C, the phosphide dissociated through P evaporation. The Si12P5 phase is proposed to have rhombohedral symmetry with a P31m space group and a C5W12 structure. The hexagonal lattice parameters measured were a = 6.16±0.05 Å and c = 13.17±0.01 Å. The optical band gap and the electrical resistivity of the phosphide were determined to be 1.55 eV and 5 mΩ cm, respectively. The Gibbs free energy diagram for the Si-P system was modified to include this new Si12P5 phase.

  • Influence of plasma parameters on the growth and properties of magnetron sputtered CNx thin films

    • Carbon nitride CNx thin films were grown by unbalanced dc magnetron sputtering from a graphite target in a pure N-2 discharge, and with the substrate temperature T-s kept between 100 and 550 degrees C. A solenoid coil positioned in the vicinity of the substrate was used to support the magnetic field of the magnetron, so that the plasma could be increased near the substrate. By varying the coil current and gas pressure, the energy distribution and fluxes of N-2(+) ions and C neutrals could be varied independently of each other over a wide range. An array of Langmuir probes in the substrate position was used to monitor the radial ion flux distribution over the 75-mm-diam substrate, while the flux and energy distribution of neutrals was estimated through Monte Carlo simulations. The structure, surface roughness, and mechanical response of the films are found to be strongly dependent on the substrate temperature, and the fluxes and energies of the deposited particles. By controlling the process parameters, the film structure can thus be selected to be amorphous, graphite-like or fullerene-like. When depositing at 3 mTorr N-2 pressure, with T-s> 200 degrees C, a transition from a disordered graphite-like to a hard and elastic fullerene-like structure occurred when the ion flux was increased above similar to 0.5-1.0 mA/cm(2). The nitrogen-to-carbon concentration ratio in the films ranged from similar to 0.1 to 0.65, depending on substrate temperature and gas pressure. The nitrogen film concentration did, however, not change when varying the nitrogen ion-to-carbon atom flux ratios from similar to 1 to 20. (C) 2000 American Institute of Physics.     • Thin epitaxial TiC and VC films and superlattices have been deposited on MgO(001) by simultaneous sputtering of the metals and evaporation of C-60. It was found that epitaxial growth conditions for TiC could be maintained down to a temperature of 100 degreesC, while the epitaxial growth of VC required 200 degreesC, Epitaxial VC films were completely relaxed at all growth temperatures, while a change from a relaxed to a strained growth behavior was observed for TiC films. The structural quality of the TiC films was better than for the VC films. A general observation was that a plasma-assisted deposition process yields films with a higher quality and allows epitaxial growth at lower temperatures than for a pure coevaporation process.

    • The doubly magic N = Z nucleus Ni-56 has been investigated with two fusion-evaporation reactions; Ca-40(Si-28, 3 alpha)Ni-56 at a beam energy of 122MeV and Si-28(S-32, 2p2n)Ni-56 at 130MeV. To detect gamma-rays in coincidence with evaporated particles the Ge-detector array Gammasphere was used in conjunction with the charged-particle detector system, Microball and a 1 pi neutron detector array. Results include a significantly extended level scheme of Ni-56, which is compared to large-scale shell model calculations in the fp shell. The experimental and theoretical results agree to a large extent, with one notable exception; the theoretical model fails to predict the proper sequence of the yrast and yrare 8(+) states.

    • Box-shaped nanostructures of B-C-N compounds were synthesized by reactive sputtering of boron carbide in mixed argon and nitrogen discharges. Transmission electron microscopy showed that these nanoboxes were grown on self-patterned NaCl substrate with projected areas ranging from similar to 1x10(2) to similar to 5x10(4) nm(2), sizes 50-100 nm, and number density similar to 100 mu m(-2).   • Growth, microstructure, and mechanical properties of arc evaporated TiCxN1-x (0 <= x <= 1) films

    • TiCxN1-x films with x ranging from 0 to 1 were grown by arc evaporation by varying the flow ratio between the reactive gases. The substrates were cemented carbide inserts (WC-6 wt.% Co) which were negatively biased at 400 V, resulting in a deposition temperature of similar to 550 degrees C. The film composition, as measured by glow discharge optical emission spectroscopy, was found to vary almost linearly with the gas flow ratio. Cross-sectional transmission electron microscopy in combination with X-ray diffraction (XRD) showed that the films were of single-phase NaCl-structure with a dense columnar microstructure. The intrinsic stress analyzed using the XRD sin(2)psi method, was found to have a maximum of - 5.9 GPa in the composition range of 0.4 less than or equal to x less than or equal to 0.7 which correlated with a maximum in XRD peak broadening due to inhomogeneous strains. The hardness and Young's modulus of the as-deposited TiCxN1-x films were measured by the nanoindentation technique. A maximum in hardness of 45 GPa was found at the same composition range (0.4 Ix I 0.7) as the intrinsic stress maximum. The hardness for x = 0 (TiN) and x = 1 (TiC) were found to be 28 and 36 GPa, respectively. The Young's modulus was constant similar to 610 GPa for films with compositions up to x = 0.6, thereafter it decreased to 540 GPa at x = 1. The increase in intrinsic stress with increasing carbon content is suggested to be due to increased stability of defects created from the collision cascade or/and by a change in the defect structure itself. The fact that hardness showed a maximum at the same composition as residual stress and FWHM indicates that obstruction on dislocation movement has a major influence on the hardness of these films.     • Carbon and carbon nitride films, grown in argon or nitrogen discharges by reactive Jc magnetron sputtering of a graphite target, were characterized by in situ scanning tunneling microscopy. When the growth temperature increased from ambient to 800 degrees C, we observed a topographic evolution of the carbon films from an amorphous to a graphitelike structure, and further to a distorted-graphitic phase with curved and intersecting basal planes, and finally to a surface containing nanotubes and nanodomes. When nitrogen was incorporated into the films, distortion of the graphitic basal planes occurred at a lower temperature compared to the pure carbon case. At temperatures of similar to 200 degrees C and above, regions of a nongraphitic phase, containing a high degree of carbon sp(3) bonds were observed. Spatially resolved tunneling spectroscopic measurements indicated that the band gaps were 0, similar to 0-0.6 eV, and similar to 0.4-2.0 eV for graphitelike structures, the distorted-graphitic phase, and the nongraphitic phase, respectively. Together with ex sial x-ray photoelectron spectroscopy and reflection electron energy loss spectroscopy measurements, the results suggest that the incorporation of nitrogen promotes bending of the graphitic basal planes and thereby facilitates the formation of three-dimensional covalently bonded networks with a high degree of sp(3)-coordinated carbon atoms.

    • <b><i>Background:</i></b> Wheat is a common food causing allergy which has implications on the quality of life. The diagnosis of IgE-mediated wheat allergy is based on the clinical history and presence of IgE antibodies (IgE-Ab) in skin or blood, and the results of an oral food challenge which is time consuming and associated with risks. An improved diagnostic workup is needed for wheat allergy. The objective was to examine the relationship between wheat challenge, CD-sens and IgE-Ab to related allergens in wheat-allergic children and investigate if a combination of different markers could enhance the prediction of challenge outcome. <b><i>Method:</i></b> Twenty-four children (aged 1-15 years) with a wheat allergy diagnosis underwent an open wheat challenge. CD-sens and IgE-Ab to wheat, hydrolyzed wheat protein (HWP), &#x03C9;-5 gliadin and timothy grass were analyzed and related to the challenge outcome. <b><i>Results:</i></b> A positive challenge was seen in 12/24 children. Children reacting to the challenge had higher IgE-Ab concentrations to wheat, &#x03C9;-5 gliadin and HWP (p < 0.01) and a tendency to higher wheat CD-sens values (p = 0.08) than nonreacting children. Combining wheat CD-sens >150 and IgE-Ab to wheat >20 kU<sub>A</sub>/l, or &#x03C9;-5 gliadin >0.1 kU<sub>A</sub>/l predicted the challenge outcome in 83% of the patients. Most children with IgE-Ab to wheat also had IgE-Ab to timothy. Seven of 9 challenge-positive children had a positive CD-sens to HWP and IgE-Ab to HWP >8 kU<sub>A</sub>/l. <b><i>Conclusion</i></b>: Combining CD-sens and IgE-Ab to wheat or wheat components could be useful in the diagnosis and follow-up of wheat-allergic children.

    • The initial stages of TiO2 growth on Si(111) under ultra-high vacuum conditions is studied using core level photoelectron spectroscopy, x-ray absorption spectroscopy, and scanning tunneling microscopy. The TiO2 film was formed by means of chemical vapor deposition of titanium(IV) isopropoxide at a sample temperature of 500 degreesC. The thickness and composition of the amorphous interface layer and its subsequent transition to crystalline anatase TiO2 are discussed. Three different stages are identified: In the initial stage (film thickness <10 Angstrom), the oxygen atoms are coordinated mainly to Si atoms giving rise to Ti atoms with oxidation states lower than 4+. At this stage, a small amount of carbon (0.15 ML) is observed. The next stage (<25 Angstrom) is best described as an amorphous TiSixOy compound in which the oxidation state of Ti is 4+ and the x and y values vary monotonically with the film thickness, from 2 to 0 and 4 to 2, respectively. Finally (>30 Angstrom) a stoichiometric TiO2 layer starts to form. The TiO2 phase is anatase and the layer consists of particles similar to10 nm wide.

  • Carbon nitride nanotubulite - densely-packed and well-aligned tubular nanostructures

    • Tubular carbon nitride (CNx, x=0.01–0.32) nanoparticles were successfully synthesized by d.c. magnetron sputtering. These tubes were grown in a highly packed form perpendicularly on a sodium chloride substrate. Their number density is estimated to be ∼1×104 per μm2 and is constant over macroscopic regions. Sub-nanometer scale chemical mapping shows that the nitrogen to carbon atomic ratio is rather constant across these tubes. This successful synthesis of a nanotubulite – made of a rather compact aggregation of tubular nanoparticles – could facilitate experimental approaches to measure mechanical or electrical transport properties of such nanotubes and to open the way to variable nanotube applications.
