  • Implementering av ekosystemtjänst-begreppet i kommunal verksamhet : slutrapport

    • Projektet ECOSIMP handlar om förutsättningen för att implementera begreppet ekosystemtjänst, i fortsättningen kallat EST-begreppet, i kommunerna, och undersöker bland annat hur kommunala tjänstemän och politiker ser på denna utmaning. En intervjustudie genomfördes med tjänstemän och politikeri de sju medverkande kommunerna. I en studie av Malmö stad undersöktes hur ekosystemtjänst-relaterade begrepp har integrerats i översiktsplaner ochutvecklats till ett verktyg i hållbar samhällsplanering. Ett annat delprojekt handlar om miljökonsekvens-bedömningar (MKB) och behovet av metodutveckling för att integrera ekosystemtjänstansatsen i MKB, där möjligheten att integrera ekosystemtjänster i den så kallade RIAM-metoden analyserades. Projektet innehåller också en analys av arbetet med att integrera ekosystemtjänsteri kommunernas klimatanpassning, så kallad ekosystembaserad klimatanpassning (EbA). Slutligen redovisas en analys av det transdisciplinära arbetssättet inom ECOSIMP-projektet. Resultaten visar att EST-begreppet idag är relativt välkänt i kommunerna och att det finns en övervägande positiv inställning till det och förhoppningar om att det ska skapa större möjlighet till miljöhänsyn. Förståelsen av begreppet behöver dock fördjupas i den kommunala verksamheten och distinktionen mellan implicit och explicit användning av EST-begreppet och den relaterade EST-ansatsen förtydligas. Ett antal hinder och möjligheter för att börja använda begreppet och för att uppnå etappmålet 2018 identifierades också. Bland annat upplevs innebörden av etappmålet 2018 som oklar, och bara en mindre del av de intervjuade i kommunerna ansåg att etappmålet skulle nås. Betydelsen av att politiker och allmänhet får kännedom om, och förståelse för, EST-begreppet betonades också. Malmö framstår som ett bra exempel på hur långsiktigt arbete för en hållbar stadsutveckling kan skapa förutsättningar att integrera ekosystemtjänster i den fysiska planeringen. Analysen av EbA i kommunerna visade att initiativ relaterade till klimatanpassning och ekosystemtjänster oftast inte är samordnade, men de skulle kunna utvecklas i den riktningen genom bättre samordning mellan kommunernas olika enheter och integrering av EbA i den långsiktiga planeringen utifrån kunskap om nutida och framtida klimatrisker. Verktyg för att värdera förändringar i EST till följd av mänsklig exploatering behövs och här föreslås en utveckling av den såkallade RIAM-metoden, som kan erbjuda ett sätt att väga in olika EST i planeringen. Den transdisciplinära analysen visar på värdet av nära samverkan mellan forskning och kommuner kring implementeringen av EST-ansatsen, men också på behovet av politiskt och ekonomiskt stöd för att frigöra tid för kommunerna att delta i sådana projekt.

  • Transcriptional Profiling of Breast Cancer Metastases Identifies Liver Metastasis-Selective Genes Associated with Adverse Outcome in Luminal A Primary Breast Cancer

    • Purpose: The complete molecular basis of the organ-specificity of metastasis is elusive. This study aimed to provide an independent characterization of the transcriptional landscape of breast cancer metastases with the specific objective to identify liver metastasis-selective genes of prognostic importance following primary tumor diagnosis. Experimental Design: A cohort of 304 women with advanced breast cancer was studied. Associations between the site of recurrence and clinicopathologic features were investigated. Fine-needle aspirates of metastases (n = 91) were subjected to whole-genome transcriptional profiling. Liver metastasis-selective genes were identified by significance analysis of microarray (SAM) analyses and independently validated in external datasets. Finally, the prognostic relevance of the liver metastasis-selective genes in primary breast cancer was tested. Results: Liver relapse was associated with estrogen receptor (ER) expression (P = 0.002), luminal B subtype (P = 0.01), and was prognostic for an inferior postrelapse survival (P = 0.01). The major variation in the transcriptional landscape of metastases was also associated with ER expression and molecular subtype. However, liver metastases displayed unique transcriptional fingerprints, characterized by down-regulation of extracellular matrix (i.e., stromal) genes. Importantly, we identified a 17-gene liver metastasis-selective signature, which was significantly and independently prognostic for shorter relapse-free (P < 0.001) and overall (P = 0.001) survival in ER-positive tumors. Remarkably, this signature remained independently prognostic for shorter relapse-free survival (P = 0.001) among luminal A tumors. Conclusions: Extracellular matrix (stromal) genes can be used to partition breast cancer by site of relapse and may be used to further refine prognostication in ER positive primary breast cancer.     • The paper investigates the experiences, perceptions and responses of immigrants with a Palestinian background in relation to learning about the Holocaust and the Palestinian cause in Swedish schools and visiting Holocaust sites. Data were collected from 50 immigrant students using audio-recorded and open-structured interviews. The results indicate that most informants had learned about the Holocaust in various classes, through readings, discussions and assignments, but very little about the Palestinian cause in textbooks or in school activities. The majority of informants were never asked to take part in Holocaust site visits, but if they were to be asked, 84% were willing make such visits. The majority of informants expressed sympathy with Holocaust victims, but they were not satisfied, as they felt there was too much focus on the Holocaust and too little on the Palestinian cause. This lived experience of imbalance between the two subjects resulted in reluctant attitudes towards Holocaust education among the Palestinian pupils, but this should not mainly be understood as a result of anti-Semitic sentiments among them. The results indicate that Holocaust education in Swedish schools among youngsters with a Palestinian background can hardly be treated separately from the question of the Israeli and Palestinian conflict.

    • Drugs are designed to bind their target proteins in physiologically relevant tissues and organs to modulate biological functions and elicit desirable clinical outcomes. Information about target engagement at cellular and subcellular resolution is therefore critical for guiding compound optimization in drug discovery, and for probing resistance mechanisms to targeted therapies in clinical samples. We describe a target engagement-mediated amplification (TEMA) technology, where oligonucleotide-conjugated drugs are used to visualize and measure target engagement in situ, amplified via rolling-circle replication of circularized oligonucleotide probes. We illustrate the TEMA technique using dasatinib and gefitinib, two kinase inhibitors with distinct selectivity profiles. In vitro binding by the dasatinib probe to arrays of displayed proteins accurately reproduced known selectivity profiles, while their differential binding to fixed adherent cells agreed with expectations from expression profiles of the cells. We also introduce a proximity ligation variant of TEMA to selectively investigate binding to specific target proteins of interest. This form of the assay serves to improve resolution of binding to on- and off-target proteins. In conclusion, TEMA has the potential to aid in drug development and clinical routine by conferring valuable insights in drug–target interactions at spatial resolution in protein arrays, cells and in tissues.

  • Target Engagement-Mediated Amplification for Monitoring Drug-Target Interactions in Situ

    • It is important to determine the localization of drugs or drug candidates at cellular and subcellular resolution in relevant clinical specimens. This is necessary to evaluate drug candidates from early stages of drug development to clinical evaluation of mutations potentially causing resistance to targeted therapy. We describe a technology where oligonucleotide-conjugated drug molecules are used to visualize and measure target engagement in situ via rolling-circle amplification (RCA) of circularized oligonucleotide probes (padlock probes). We established this target engagement-mediated amplification (TEMA) technique using kinase inhibitor precursor compounds, and we applied the assay to investigate target interactions by microscopy in pathology tissue sections and using flow cytometry for blood samples from patients, as well as in commercial arrays including almost half of all human proteins.  In the variant proxTEMAtechnique, in situ proximity ligation assays were performed by combining drug-DNA conjugates with antibody-DNA conjugates to specifically reveal drug binding to particular on- or off-targets in pathological tissues sections. In conclusion, the TEMA methods successfully visualize drug-target interaction by experimental and clinically approved kinase inhibitors in situ and with kinases among a large collection of arrayed proteins.

    • Men have increasingly been dedicating time and effort to childcare. Consequently, the idea of the ‘new’ emotionally involved father has been discussed in the literature. This article focuses on narratives of divorced Swedish fathers with joint physical custody of their children. This arrangement, a new model of post-divorce parenting, has become increasingly popular in several Nordic countries. The article aims to analyse the experiences attached to and emanating from this particular form of post-divorce agreement, and how it is understood in relation to the Swedish childcare system. The fathers interviewed had a strong ambition to share things equally, as well as to carry on family practices in gender-equal ways. This desire was, however, balanced with a number of obstacles, such as work requirements, living conditions and conceptions of gender differences. In this way, the fathers’ subjective aspirations and strivings were filtered through structural and cultural conditions in society, with clear connections to Swedish family and gender politics.

    • Departing from a qualitative research approach, this article focuses on Swedish amateur Ironman triathletes and their family life. The purpose of the paper is to investigate how an elite amateur lifestyle is upheld and balanced with the demands of a sustainable family and social life. The results indicate that the process of becoming and staying an Ironman creates tensions in intimate relationships, making it hard to bring the family life puzzle together. Although the participants interviewed often talk about family life in terms of sharing things fairly equally, in terms of gender equity and involved parenthood, this seemingly is not always an easy ideal to fulfil in practice. On a broader cultural level, these findings can thus be contextualized in relation to discourses associated with the gendering of families and functions, and, of course, the gender of sport and performance.

    • This article describes and analyses the historical development of gym and fitness culture in general and doping use in this context in particular. Theoretically, the paper utilises the concept of subculture and explores how a subcultural response can be used analytically in relation to processes of cultural normalisation as well as marginalisation. The focus is on historical and symbolic negotiations that have occurred over time, between perceived expressions of extreme body cultures and sociocultural transformations in society-with a perspective on fitness doping in public discourse. Several distinct phases in the history of fitness doping are identified. First, there is an introductory phase in the mid-1950s, in which there is an optimism connected to modernity and thoughts about scientifically-engineered bodies. Secondly, in the 1960s and 70s, a distinct bodybuilding subculture is developed, cultivating previously unseen muscular male bodies. Thirdly, there is a critical phase in the 1980s and 90s, where drugs gradually become morally objectionable. The fourth phase, the fitness revolution, can be seen as a transformational phase in gym culture. The massive bodybuilding body is replaced with the well-defined and moderately muscular fitness body, but at the same time there are strong commercialised values which contribute to the development of a new doping market. Finally, it is possible to speculate on the development of a fifth phase, in which fitness doping is increasingly being filtered into mainstream gym and fitness culture, influencing the fitness doping demography.

    • This article analyses fitness professionals’ perceptions and understanding of their occupational education and pedagogical pursuance, framed within the emergence of a global fitness industry. The empirical material consists of interviews with personal trainers and group fitness instructors, as well as observations in their working environment. In addition, printed material from different occupational organisations and educational companies has been included. The narratives of the fitness professionals and a case study of Les Mills are presented and analysed through the concept of the McDonaldisation of society, or more specifically of fitness culture. The results show that, even though gym and fitness franchises differ from hamburger restaurant chains, there are crucial similarities, but also differences. One can, for example, discern a tendency towards the construction of predesigned and highly monitored programmes, such as the one developed by Les Mills. Homogenisation is also apparent when looking at the body ideals produced, as fitness professionals work on their own or clients’ bodies, which makes it possible to anticipate a global body ideal. The social and cultural patterns of self-regulation and self-government found in gym and fitness culture can be understood and analysed in a global context. What we find is an intriguing and complex mixture of regulation, control and standardisation, on the one hand, and a struggle to express the body, to be ‘free’ and to transgress the boundaries set by the commercial global fitness industry, on the other.

    • This article analyzes self-portrayals and gender constructions among female personal trainers within an Internet-mediated framework of fitness culture. The empirical material comes from a close examination of three strategically selected blogs. The result shows that some of the blogs clearly build upon what Connell calls emphasized femininity, as a means of legitimizing and constructing appropriate female fitness. In addition, there are also tendencies of sexualization in text and imagery present. As such, these self-representations are framed within a cultural history of body fitness dominated by stereotypical ways of perceiving masculinity and femininity. However, this does not capture the entire presentation of the self among the analyzed fitness bloggers. The blogs also point in the direction of ongoing negotiations and subversions of traditional gender norms. Among other things, they show how irony and humor are used as a means of questioning normative gender constructions while empowering female fitness and bodyliness.

    • This article is based on written accounts posted on an online forum called Flashback.The purpose of the study was to explore how participants in this community negotiated themeanings of fitness doping and how such negotiations could be understood in terms of masculinity.The findings indicate that the Internet community studied in this article can be read as an example ofa transformational process in which ordinary rules are questioned and partly put out of play. In theworld of the bodybuilder, the marginal masculinity is, in certain senses, dominant. On the one hand,achieving a muscular and well-trained body is regarded as a core aspect of manhood within thecommunity. Marginal masculinity is thus momentarily transformed into dominant and hegemonicmasculinity. On the other hand, however, the findings also indicate that a drug-using, muscularmasculinity is constructed in negotiation with other central masculine ideals, such as the employableman and the responsible father. Found within the community is a complex and dynamic interplaybetween intersecting discourses of manhood.

    • There is currently an increasing amount of research on online fatherhood. In this article, we have used international blogs as our empirical data, in order to investigate and create an in-depth picture of fatherhood as it is expressed in various men’s stories about their everyday lives. Three distinct conceptions of fatherhood and masculinity emerge. The first of these positions could be described as a nostalgic position in relation to hegemonic masculinity. The second position is referred to here as complicity. This position indicates a changing landscape of fatherhood and family politics, moving towards gender-equal conditions. Thirdly, we have an inclusive position that reflects a gender-neutral position, and are understood less in terms of masculinity, heteronormativity and gender-divided responsibilities.

    • The focus of this study is on the development of a globalised and localised gym and fitness culture. The article takes its point of departure from three distinct levels. These are (1) organisational aspects of the culture, (2) fitness professionals’ individual trajectories, and (3) national variations and gender regimes. The findings indicate that fitness professionalism is emerging as an uncertain profession. On the one hand there are international accreditation systems that aim to ensure the status of these professionals. On the other hand, there are simultaneous tendencies towards devaluation of the status of fitness professionals, on a global level. Partly, this can be related to the standardisation of training programmes, as well as the connectedness between the service industry and feminised gender regimes. In conclusion, the globalisation of fitness is a highly gendered process, involving national body ideals and cultural variations in how fitness professionals are valued and perceived locally.

    • This article investigates how Swedish gay men pursue and understand fatherhood, using a qualitative, semi-structured methodological approach. We are interested in how practical issues and different fatherhood arrangements are understood and handled by the fathers; how the construction of “rainbow families” is understood in relation to legal issues; parental responsibilities; and thoughts regarding gender-equal and gender-neutral relationships within Swedish society and culture. The results show that the trajectory of gay men toward fatherhood is shaped by a variety of mediating factors such as interactions with agencies, clinics, attorneys, and the state. There are some significant legal, social, financial, and cultural obstacles to realizing gay parenthood. There are also cases where study participants talked about a sense of intolerance expressed toward gay parents within a gay community. Although there are still strong normative dimensions involved in the construction of modern parenthood, the grip of heteronormative views on family life and sexuality is changing; the narratives presented also represent a questioning of the hegemonic image of fatherhood in Sweden. Consequently, it is possible to discern different attempts at transgressing and changing the cultural landscape of fatherhood and the ways people build families today.

  • Men’s Achilles’ heel: prostate cancer and the reconstruction of masculinity

    • The aim of this study was to investigate how Swedish men diagnosed with prostate cancer understand the effects of their treatment in relation to sexual health and masculinity. Utilising a phenomenological and sociologically informed approach, the study involved interviews with 21 Swedish men who experienced problems following treatment. The results showed that participants’ initial response post-treatment, involved the development of new bodily understandings and socially informed strategies to handle incontinence and sexual dysfunction. Due to impotence and the loss of ejaculatory ability following treatments such as surgery, participants re-articulated the meaning of intimacy, as well as their understanding of masculinity and themselves as ageing men. Unlike in previous research, such a re-articulation of masculinity and sexual health is understood as taking place within rather than in opposition to hegemonic masculinity.

    • On the basis of qualitatively gathered interview material and data from various postings on a pro-doping online community called Flashback, the aim of this article is to describe and analyse how female users of performance and image enhancing drugs (PIED) understand and negotiate their use in relation to gender and the body. Positioned within post-structural feminism, the results show that there is an increasing amount of knowledge that not only targets but is also developed by and for women concerning PIED use. Traditionally scholars have connected female PIED use more or less exclusively to female bodybuilders, but as new body and femininity ideals develop the demographics of female fitness doping are widening. Although PIED use in the context of gym and fitness culture remains primarily a masculine domain, the results point towards a development in which women are increasingly becoming more integrated into a fitness community of PIED users.

    • Drawing on qualitative interviews with Mixed martial arts (MMA) athletes and stakeholders, this study aims to investigate the relationship between, on the one hand, MMA as a spectacle and imaginary world, and on the other, the fighters’ experiences of violence, pain and ‘the real’. Analytically, we are influenced by the literature on the spectacle and on hyperreality. The results show that athletes’ negotiations concerning the sport largely connect to a particular way of approaching violence – culturally and in terms of physical experience. On the one hand, there is a desire to portray MMA as a civilized and regulated sport. The athletes develop different strategies by which to handle or renegotiate the physical force and violence in the cage. On the other hand, however, the fighters’ bodily control and management of their fear sometimes breaks down. When the spectacle of the octagon becomes ‘real’, the legitimacy of the sport is questioned.

    • In Sweden, but also internationally, official regimes and public health organizations conduct fairly comprehensive antidoping measures. As a consequence, numerous ‘new’ ways to learn about and access these types of drugs have emerged. Social media and different internet forums, for example, have become part of a new self-help culture in which people can anonymously approach these substances, discuss their experiences of using them and minimize the possibility of encounters with the police. This article focuses on how the use of performance- and image-enhancing drugs (PED) is perceived and negotiated socially in the context of an internet-mediated and online community calledFlashback. The results are in accordance with similar studies on internet bodybuilding communities. That is, even though the participants to a certain degree are aware of the risks and health costs of this kind of physical regime, the gains of using PED clearly dominate the discussions. Adding to this research, this article found that in the imaginary world of online bodybuilding a number of ideas about the ‘genetic max’, as well as the ultimate possibility of exceeding one’s limits, and creating something special and extraordinary, circulate. These stories confirm in many ways the legitimate mission of searching for the ultimate bodybuilding adventure, using different means and methods to transcend the limits of the self/body.

    • This article presents analyses from an international empirical study of young fatherhood in Sweden and the UK to interrogate how welfare contexts and family policies shape young fathers’ views of parenthood. Our analyses demonstrate that despite differences in constructions of young fatherhood, whereby young parenthood is problematised in UK family policy, more so than in Sweden, young fathers in both countries express an encouraging commitment to contemporary cultural imperatives for engaged fatherhood. However, differences in welfare and parental leave systems have a clear influence on the extent to which the young men in the respective countries fulfil their parental commitments and act as local agents of change in the wider social project of gender equality. We argue that while policy processes and discourses in support of young parenthood and gender equality are currently treated as disparate concerns, their articulations with one another may instead be seen as complementary and symbiotic.

    • This article builds on data gathered through focus group interviews with Swedish mentreated for prostate cancer. First, we aim to analyse how the participants talk about andhandle sensitive issues and common side effects. Second, we investigate how theparticipants’ feelings about their disease affect or interfere with their understanding ofmasculinity. The findings illustrate how participants partly break with historical ex-pectations of men as emotionally inexpressive. Instead, in the groups they talk openlyabout incontinence, erectile dysfunction, and other sensitive issues. At the same time,these discussions can be positioned within certain discursive coping strategies. Usingjokes and intellectualizing their health narratives, combined with a problematizationand partial redefinition of gender and masculinity, most of the men adapt to their newlife situation. The men’s ways of handling their emotions can be situated in the interfacebetween discursive and gendered emotion ideologies, and lived experiences ofmasculinity.

    • This study aims to examine how healthcare professionals (HCPs) understand the impact prostate cancer treatment can have on patients’ sense of masculinity, sexuality, and intimate life, and how they describe such issues are communicated with patients. Theoretically, HCP narratives are interpreted as part of a reflective process in which notions of hegemonic masculinity are communicated, and sometimes rethought and redefined, within the rehabilitation process. The study showed that HCPs sometimes felt unqualified to deal with issues concerning masculinity and sexuality as such topics were understood to be partially outside the medical domain of their professions. Nonetheless, HCPs engaged in such conversations with patients and described how they tried to support them in reorientating their sense of masculinity. The article concludes that, whereas HCPs tended to describe their patients’ responses to rehabilitation from an embodied and psychological perspective, their own professional and personal views on masculinity usually departed from a sociocultural level (focusing on what it means to be a man in contemporary Swedish society, suggesting that penetrative sex is overrated), where ongoing configurations of hegemonic masculinity were more evident.

    • Today, fitness gyms and private health clubs are a huge global business. Fitness has turned into a folk movement, but not one comparable to the old 20th-century movements, often connected to national sentiments, but instead a highly individualized preoccupation. In this article the historical development of modern gym and fitness culture is described and an analytically developed approach to the understanding of the emergence of this multi-billion-dollar phenomenon is developed. The analysis suggest that the techniques, tools, and physical exercises used today in gyms all over the world are the results of a physical culture developed and refined during the 20th century. The body ideals, exercises, techniques, and the pedagogy of fitness have become an increasingly international enterprise. A tentative analysis of the globalization of gym and fitness culture is developed and presented. Three important and decisive phases in the globalization of gym and fitness culture are identified and analyzed.

    • Using a minor case study of the local fitness culture scene in Tokyo, Japan, this exploratory and qualitative article aims to discuss and analyse the consequences of globalisation of fitness culture. The article focuses particularly on how fitness culture is negotiated in Japanese society – at a national, local and subjective level, and in relation to attempts to uniformly rationalise and standardise gym and fitness culture. The results indicate that the fitness geography is changing, and through the establishment of a globalised fitness culture, Japanese youth are following in the footsteps of many other countries. However, this does not mean that we are witnessing a homogenisation process, or a completely McDonaldised version of fitness culture in Japan. Instead, this cultural phenomenon is shaped and formed in particular ways, pointing towards certain strong national sentiments concerning body ideals, views on gender and exercise and relaxation. For example, the cuteness ideal has a strong influence on the way young women talk about and perceive body ideals and corporeal performances in Japan.

    • This study focuses on the physical expressions and intensity of embodiment that occur in the Ironman Triathlon. More specifically, the study investigates the transformational bodily experiences taking place during Ironman competitions. Using an ethnographic approach, a total of 29 Ironman triathletes participated in the study (15 men and 14 women). Theoretically, the article focuses on how triathletes’ bodies ‘move’ between different forms of embodiment. The results show that, in the process of disciplining the body, the athletes reconceptualized feelings of pain, nausea and even disgust, making these emotionally expressive aspects of the corpus into a part of the experience and bending them towards the pleasure of reaching potential divinity. Situated in a long tradition of philosophical and sociological explorations of the transgressing and transcending body, the study interprets and understands the performing body as a site for change and utopian possibilities. Thus the study adds to existing debates on contemporary individuals’ exploration of the existential and corporeal dimensions of modernity.

    • The widespread availability of doping and its growing prevalence among fitness groups has contributed greatly to the realization of an emergent public health issue. Emanating from an ethnographic study in Sweden, the purpose of this study was to describe and analyze the processes involved in becoming and “unbecoming” a fitness doping user. The study employs a cultural and sociological perspective as its theoretical framework and discusses how the participating users gradually develop their knowledge about the drugs and how the process of becoming a user is negotiated in relation to ideas and ideals concerning health, gender, and individual freedom and Swedish law. Regarding exit processes, (re)entering into what is perceived to be an ordinary “normal” life was seldom a straightforward process. To understand the complex and sometimes complicated transition processes involved in becoming respectively unbecoming a fitness doper, the results highlight the limitations of using stage models for understanding exit process as heuristic tools. Furthermore, the article argue for the necessity to investigate the negotiations of fitness doping, taking place in the intersection between subcultural affiliations/spaces, doping legislation, and mainstream perceptions of living a “Normal” life. It is argued that processes of (un)becoming a fitness doper are anything but linear and thus need to be understood in relation to sociocultural belonging and ongoing negotiation of the individual’s sense of self.
