    • In jawed vertebrates, development of an adaptive immune-system is essential for protection of the born organism against otherwise life-threatening pathogens. Myeloid cells of the innate immune system are formed early in development, whereas lymphopoiesis has been suggested to initiate much later, following emergence of definitive hematopoietic stem cells (HSCs). Herein, we demonstrate that the embryonic lymphoid commitment process initiates earlier than previously appreciated, prior to emergence of definitive HSCs, through establishment of a previously unrecognized entirely immune-restricted and lymphoid-primed progenitor. Notably, this immune-restricted progenitor appears to first emerge in the yolk sac and contributes physiologically to the establishment of lymphoid and some myeloid components of the immune-system, establishing the lymphomyeloid lineage restriction process as an early and physiologically important lineage-commitment step in mammalian hematopoiesis.

    • Vascular endothelial growth factor (VEGF) stimulates angiogenesis by activating VEGF receptor-2 (VEGFR-2). The role of its homolog, placental growth factor (PlGF), remains unknown. Both VEGF and PlGF bind to VEGF receptor-1 (VEGFR-1), but it is unknown whether VEGFR-1, which exists as a soluble or a membrane-bound type, is an inert decoy or a signaling receptor for PlGF during angiogenesis. Here, we report that embryonic angiogenesis in mice was not affected by deficiency of PlGF (Pgf-/-). VEGF-B, another ligand of VEGFR-1, did not rescue development in Pgf-/- mice. However, loss of PlGF impaired angiogenesis, plasma extravasation and collateral growth during ischemia, inflammation, wound healing and cancer. Transplantation of wild-type bone marrow rescued the impaired angiogenesis and collateral growth in Pgf-/- mice, indicating that PlGF might have contributed to vessel growth in the adult by mobilizing bone-marrow-derived cells. The synergism between PlGF and VEGF was specific, as PlGF deficiency impaired the response to VEGF, but not to bFGF or histamine. VEGFR-1 was activated by PlGF, given that anti-VEGFR-1 antibodies and a Src-kinase inhibitor blocked the endothelial response to PlGF or VEGF/PlGF. By upregulating PlGF and the signaling subtype of VEGFR-1, endothelial cells amplify their responsiveness to VEGF during the 'angiogenic switch' in many pathological disorders.

    • Recent findings from studies on adults show similarities between social anxiety disorder (SAD) and posttraumatic stress in the form of recurrent memories and intrusive and distressing images of earlier aversive events. Further, treatment models for SAD in adults have been successfully developed by using transdiagnostic knowledge on posttraumatic stress symptoms (PTSS). Studies on adolescents are though missing. The present study aimed at exploring the association between PTSS and SAD in Swedish adolescents. A second aim was to study mental health services utilization in relation to these conditions. A total of 5,960 high-school students participated and reported on SAD, life time victimization, PTSS and mental health service utilization. Socially anxious adolescents reported significantly higher levels of PTSS than adolescents not reporting SAD and this difference was seen in victimized as well as non-victimized subjects. Contact with a school counselor was the most common mental health service utilization in subjects with SAD and those with elevated PTSS. In the prediction of contact with a CAP-clinic, significant odds ratios were found for a condition of SAD and elevated PTSS (OR = 4.88, 95 % CI = 3.53–6.73) but not for SAD only. Screening of PTSS in adolescents with SAD is recommended. The service of school counselors is important in detecting and helping young people with SAD and elevated PTSS. Clinical studies on SAD and PTSS in adolescents could aid in modifying treatment models for SAD.

    • COST Action FP 1303 “Performance of bio-based building materials” successfully started in October 2013 and an ambitious program was set up for the four year programme. COST Actions provide an excellent opportunity for collaborative research, e.g. in the frame of Round Robin tests.The idea of this respective test was to distribute a fairly simple test set up to as many places in Europe as possible in order to collect performance data reflecting the range of climatic exposure conditions. Furthermore we wanted to consider performance in its manifold meaning, i.e. optical, aesthetical, moisture and functional performance and durability. In contrast to traditional Round Robin tests aiming on comparative evaluation and validation of results from different test laboratories, this initiative aims on collecting performance data under climatically different exposure conditions. Therefore it was required to provide weather data from the respective test sites to allow establishing relationships between climate conditions and the following measured, which shall be evaluated regularly: decay, discolouration, development of mould and other staining fungi, corrosion, formation of cracks and moisture performance (if data logging device is included). Further details about the test and the first outcomes are presented in this paper.

    • Uncertainties in aerosol radiative forcings, especially those associated with clouds, contribute to a large extent to uncertainties in the total anthropogenic forcing. The interaction of aerosols with clouds and radiation introduces feedbacks which can affect the rate of precipitation formation. In former assessments of aerosol radiative forcings, these effects have not been quantified. Also, with global aerosol-climate models simulating interactively aerosols and cloud microphysical properties, a quantification of the aerosol forcings in the traditional way is difficult to define properly. Here we argue that fast feedbacks should be included because they act quickly compared with the time scale of global warming. We show that for different forcing agents (aerosols and greenhouse gases) the radiative forcings as traditionally defined agree rather well with estimates from a method, here referred to as radiative flux perturbations (RFP), that takes these fast feedbacks and interactions into account. Based on our results, we recommend RFP as a valid option to compare different forcing agents, and to compare the effects of particular forcing agents in different models.

    • To achieve sustainable fisheries, advice to management should be based on reliable science and unbiased data. Attaining quality data (i.e. precise and unbiased) on recreational fishing can be challenging, particularly when prior knowledge of the sector is limited and a proper sample frame of recreational fishers or vessels does not exist. In this study, a registry of access points was constructed for the Swedish south-west coast and used as a spatial sample frame in determining both effort and catches of the private boat fishery. Sampling dates, times for sampling, and access points visited were selected using probabilistic methods, ensuring unbiased results. The final multi-stage sampling design involved multiple strata, clusters, and probability selection methods and enabled first-time estimation of Swedish recreational landings of western Baltic cod by private boats to be used in stock assessment. Concurrent data collection covering aspects such as boat counts at access points, provided additional information on e.g. activity patterns. That additional information opens possibilities to refine the design of the original survey and optimize the sampling effort towards different goals, such as other fished resources. In this paper, we reflect on the challenges that limitations in initial information poses to the design and deployment of a new recreational fisheries survey. We suggest ways, whereby indirect sampling frames can be developed from initially incomplete or limited information to access the fishers and their catch. Our experience shows that, despite initial frame and knowledge limitations, full probabilistic methods are worth considering in data limited scenarios and that the design-based point estimates and variances they provide on recreational fishing effort and catches are useful in guiding initial management and the next steps of survey improvement.
