  • Inhibition of the master regulator of Listeria monocytogenes virulence enables bacterial clearance from spacious replication vacuoles in infected macrophages

    • A hallmark of Listeria (L.) monocytogenes pathogenesis is bacterial escape from maturing entry vacuoles, which is required for rapid bacterial replication in the host cell cytoplasm and cell-to-cell spread. The bacterial transcriptional activator PrfA controls expression of key virulence factors that enable exploitation of this intracellular niche. The transcriptional activity of PrfA within infected host cells is controlled by allosteric coactivation. Inhibitory occupation of the coactivator site has been shown to impair PrfA functions, but consequences of PrfA inhibition for L. monocytogenes infection and pathogenesis are unknown. Here we report the crystal structure of PrfA with a small molecule inhibitor occupying the coactivator site at 2.0 Å resolution. Using molecular imaging and infection studies in macrophages, we demonstrate that PrfA inhibition prevents the vacuolar escape of L. monocytogenes and enables extensive bacterial replication inside spacious vacuoles. In contrast to previously described spacious Listeria-containing vacuoles, which have been implicated in supporting chronic infection, PrfA inhibition facilitated progressive clearance of intracellular L. monocytogenes from spacious vacuoles through lysosomal degradation. Thus, inhibitory occupation of the PrfA coactivator site facilitates formation of a transient intravacuolar L. monocytogenes replication niche that licenses macrophages to effectively eliminate intracellular bacteria. Our findings encourage further exploration of PrfA as a potential target for antimicrobials and highlight that intra-vacuolar residence of L. monocytogenes in macrophages is not inevitably tied to bacterial persistence.

  • Comparing psychopathological symptoms, life satisfaction, and personality traits between the WHO and APA frameworks of gaming disorder symptoms: A psychometric investigation

    • Introduction: The inclusion of Internet Gaming Disorder (IGD) in the fifth revision of the Diagnostic and Statistical Manual of Mental Disorders (DSM-5) by the American Psychiatric Association and Gaming Disorder in the 11th revision of the International Classification of Diseases (ICD-11) by the World Health Organization requires consistent psychological measures for reliable estimates. The current study aimed to investigate the psychometric properties of the Gaming Disorder Test (GDT), the Ten-Item Internet Gaming Disorder Test (IGDT-10), and the Five-Item Gaming Disorder Test (GDT-5) and to compare the WHO and the APA frameworks of gaming disorder symptoms in terms of psychopathological symptoms, life satisfaction, and personality traits.Methods: A sample of 723 Swedish gamers was recruited (29.8% women, 68.3% men, 1.9% other, Mage = 29.50 years, SD = 8.91).Results: The results indicated notable differences regarding the estimated possible risk groups between the two frameworks. However, the association between gaming disorder symptoms and personality traits, life satisfaction, and psychopathological symptoms appeared consistent across the two frameworks. The results showed excellent psychometric properties in support of the one-factor model of the GDT, IGDT-10, and GDT-5, including good reliability estimates (McDonald's omega) and evidence of construct validity. Additionally, the results demonstrated full gender and age measurement invariance of the GDT, IGDT-10, and GDT-5, indicating that gaming disorder symptoms are measured equally across the subgroups.Conclusion: These findings demonstrate that the IGDT-10, GDT-5, and GDT are appropriate measures for assessing gaming disorder symptoms and facilitating future research in Sweden.

  • Gaming motivations and gaming disorder symptoms: A systematic review and meta-analysis

    • Background and aims: The present systematic review and meta-analysis aimed to synthesize the available literature on the relationship between gaming motivations and gaming disorder symptoms. Specifically, to (1) explore what gaming motivation questionnaires and classifications are used in studies on gaming disorder symptoms and (2) investigate the relationship between motivational factors and symptoms of gaming disorder. Method: An electronic database search was conducted via EBSCO (MEDLINE and PsycINFO) and the Web of Science Core Collection. All studies using validated measurements on gaming disorder symptoms and gaming motivations and available correlation coefficients of the relationship between gaming disorder and gaming motivations were included. The meta-analyses were conducted using a random-effects model. Results: In total, 49 studies (k = 58 independent sub-samples), including 51,440 participants, out of which 46 studies (k = 55 sub-samples, n = 49,192 participants) provided data for the meta-analysis. The synthesis identified fourteen different gaming motivation instruments, seven unique motivation models, and 26 motivational factors. The meta-analysis showed statistically significant associations between gaming disorder symptoms and 23 out of 26 motivational factors, with the majority of the pooled mean effect sizes ranging from small to moderate. Moreover, large heterogeneity was observed, and the calculated prediction intervals indicated substantial variation in effects across populations and settings. Motivations related to emotional escape were robustly associated with gaming disorder symptoms. Discussion and conclusions: The present meta-analysis reinforces the importance of motivational factors in understanding problematic gaming behavior. The analysis showed significant heterogeneity in most outcomes, warranting further investigation.

  • Longitudinal effects of bilingualism on dual-tasking

    • An ongoing debate surrounds whether bilinguals outperform monolinguals in tests of executive processing. The aim of this study was to investigate if there are long-term (10 year) bilingual advantages in executive processing, as indexed by dual-task performance, in a sample that were 40–65 years at baseline. The bilingual (n = 24) and monolingual (n = 24) participants were matched on age, sex, education, fluid intelligence, and study sample. Participants performed free-recall for a 12-item list in three dual-task settings wherein they sorted cards either during encoding, retrieval, or during both encoding and retrieval of the word-list. Free recall without card sorting was used as a reference to compute dual-task costs. The results showed that bilinguals significantly outperformed monolinguals when they performed card-sorting during both encoding and retrieval of the word-list, the condition that presumably placed the highest demands on executive functioning. However, dual-task costs increased over time for bilinguals relative to monolinguals, a finding that is possibly influenced by retirement age and limited use of second language in the bilingual group.

  • Occupational cognitive complexity and episodic memory in old age

    • The aim of this study was to investigate occupational cognitive complexity of main lifetime occupation in relation to level and 15-year change in episodic memory recall in a sample of older adults (≥ 65 years, n = 780). We used latent growth curve modelling with occupational cognitive complexity (O*NET indicators) as independent variable. Subgroup analyses in a sample of middle-aged (mean: 49.9 years) men (n = 260) were additionally performed to investigate if a general cognitive ability (g) factor at age 18 was predictive of future occupational cognitive complexity and cognitive performance in midlife. For the older sample, a higher level of occupational cognitive complexity was related to a higher level of episodic recall (β = 0.15, p < .001), but the association with rate of change (β = 0.03, p = .64) was not statistically significant. In the middle-aged sample, g at age 18 was both directly (β = 0.19, p = .01) and indirectly (via years of education after age 18, ab = 0.19) predictive of midlife levels of occupational cognitive complexity. Cognitive ability at age 18 was also a direct predictor of midlife episodic recall (β = 0.60, p ≤ 0.001). Critically, entry of the early adult g factor attenuated the association between occupational complexity and cognitive level (from β = 0.21, p = .01 to β = 0.12, p = .14). Overall, our results support a pattern of preserved differentiation from early to late adulthood for individuals with different histories of occupational complexity.

    • Both the behavioural and subjective impacts of single-word spoken warnings were examined. Behaviourally, the effect of infrequently occurring warnings was studied through their disruptive impact on a visually presented serial recall task. In separate experiments, ratings of the same words were elicited. Experiment 1 showed a localized effect of the warnings (on the item immediately following the warning), with the urgently intoned warning having a greater disruptive effect than its valence (emotional content). Valence and intonation (urgency) did not interact. The performance changes were mirrored in the ratings of the words. Experiment 2 showed no systematic effect on performance of either the action-relatedness of the word or its lexicality. There was, however, a systematic effect of lexicality but not action-relatedness on ratings. The study demonstrates the feasibility of using objective performance methods to establish the likely effects of verbal warnings and the utility of using such methods for the design of alarm systems.

  • A Longitudinal Study of Memory Advantages in Bilinguals

    • Typically, studies of cognitive advantages in bilinguals have been conducted previously by using executive and inhibitory tasks (e.g. Simon task) and applying cross-sectional designs. This study longitudinally investigated bilingual advantages on episodic memory recall, verbal letter and categorical fluency during the trajectory of life. Monolingual and bilingual participants (n= 178) between 35-70 years at baseline were drawn from the Betula Prospective Cohort Study of aging, memory, and health. Results showed that bilinguals outperformed monolinguals at the first testing session and across time both in episodic memory recall and in letter fluency. No interaction with age was found indicating that the rate of change across ages was similar for bilinguals and monolinguals. As predicted and in line with studies applying cross-sectional designs, no advantages associated with bilingualism were found in the categorical fluency task. The results are discussed in the light of successful aging.

    • This study investigated if vehicle noise and whole-body vibrations produce negative after-effects on performance in an attention task and on ratings of alertness, difficulty, and annoyance. The task was applied before and after exposure, and after a rest. Participants had degraded performance immediately after exposure in all conditions, but no effect was found after the rest. No main effect was found between exposure conditions on performance, it was concluded that the degraded performance dependent on time was caused by fatigue. The results also suggested lower ratings of alertness and higher ratings of annoyance in the vibration condition compared to the control condition.

    • Unexpected task-irrelevant changes in the auditory or visual sensory channels have been shown to capture attention in an ineluctable manner and distract participants away from ongoing auditory or visual categorization tasks. We extend the study of this phenomenon by reporting the first within-participant comparison of deviance distraction in the tactile and auditory modalities. Using vibro-tactile-visual and auditory-visual cross-modal oddball tasks, we found that unexpected changes in the tactile and auditory modalities produced a number of functional similarities: A negative impact of distracter deviance on performance in the ongoing visual task, distraction on the subsequent trial (post-deviance distraction), and a similar decrease – but not the disappearance – of these effects across blocks. Despite these functional similarities, deviance distraction only correlated between the auditory and tactile modalities for the accuracy-based measure of deviance distraction and not for response latencies. Post-deviance distraction showed no correlation between modalities. Overall, the results suggest that behavioral deviance distraction may be underpinned by both modality-specific and multimodal mechanisms, while post-deviance distraction may predominantly relate to modality-specific processes.

    • This study investigates the effects of noise and whole-body vibration on saliva cortisol levels and subjectively rated difficulty and stress while performing cognitive tasks. In the first experiment, 24 men completed a logical reasoning task and a short-term memory task while, on separate occasions, being exposed to noise, vibration, combined stimuli, and control conditions. The environmental stimuli were designed to simulate the exposure from a forestry vehicle. The main finding was that participants made significantly higher ratings of stress when the noise stimulus was present, either alone or in combination with vibration. There were, however, some indications that noise sensitivity might moderate both subjective and objective measures such as higher ratings of stress and elevated cortisol levels in high noise sensitive participants. A second experiment was conducted where noise sensitivity was used as an inclusion criterion. A low sensitive and a high sensitive group were created, each containing 16 participants. The results of the second experiment, found only marginal effects of noise sensitivity and no effects at all on performance or cortisol. Increased ratings of stress and difficulty were found whenever either environmental stressor was present, whether by itself or in combination. The same result was seen even when pooling the data from both experiments. The main conclusion of the study is that relatively short exposures to noise and vibration typical of those levels that are found in industrial vehicles do not significantly affect performance in cognitive tasks nor saliva cortisol levels even if work in these environments can be experienced as more difficult or stressful.

    • Objective:The objective was to study the involuntary capture of attention by spoken words varying in intonation and valence.Background:In studies of verbal alarms, the propensity of alarms to capture attention has been primarily assessed with the use of subjective ratings of their perceived urgency. Past studies suggest that such ratings vary with the alarms’ spoken urgency and content.Method:We measured attention capture by spoken words varying in valence (negative vs. neutral) and intonation (urgently vs. nonurgently spoken) through subjective ratings and behavioral measures. The key behavioral measure was the response latency to visual stimuli in the presence of spoken words breaking away from the periodical repetition of a tone.Results:The results showed that all words captured attention relative to a baseline standard tone but that this effect was partly counteracted by a relative speeding of responses for urgently compared with nonurgently spoken words. Word valence did not affect behavioral performance. Rating data showed that both intonation and valence increased significantly perceived urgency and attention grabbing without any interaction.Conclusion:The data suggest a congruency between subjective ratings and behavioral performance with respect to spoken intonation but not valence.Application:This study demonstrates the usefulness and feasibility of objective measures of attention capture to help design efficient alarm systems.

  • The Informational Constraints of Behavioral Distraction by Unexpected Sounds : The Role of Event Information

    • Sounds deviating from an otherwise repeated stream of task-irrelevant auditory stimuli (deviant sounds among standard sounds) are known to capture attention and impact negatively on ongoing behavioral performance (behavioral oddball distraction). Traditional views consider such distraction as the ineluctable consequence of the deviant sounds’ low probability of occurrence relative to that of the standard.Contrary to this contention, recent evidence demonstrates that distraction by deviant sounds is not obligatory and occurs only when sounds (standards and deviants), though to be ignored, act as useful warning cues by providing information as to whether and when a target stimulus is to be presented (Parmentier, Elsley, & Ljungberg, 2010). The present study aimed to extend this finding by disentangling the roles of event information (target’s probability of occurrence) and temporal information (target’s time of occurrence). Comparing performance in a cross-modal oddball task where standard and deviant sounds provided temporal information, event information, both, or none, we found that distraction by deviant sounds emerged when sounds conveyed event information. These results suggest that unexpected changes in a stream of sounds yield behavioral distraction to the extent that standards and deviants carry relevant goal-directed information, specifically, the likelihood of occurrence of an upcoming target.

    • Research of the distractor value of hearing the own name has shown that this self-referring stimulus captures attention in an involuntary fashion and creates distraction. The behavioral studies are few and the outcomes are not always clear cut. In this study, the distraction by 'own names' compared to control names (controlling for familiarity, gender and number of syllables) or matched neutral words was investigated in 2 experiments using a cross-modal oddball task. Participants completed a visual categorization task while exposed to either a sine wave tone as a standard stimulus (75% of thetrials) or unexpected auditory deviants (12.5% trials for eachname category in Experiment 1, and 10 % for each name category and for words in Experiment 2). Results showed deviant distraction by exposure to both the irrelevant word, own and the control name compare to the standard tone but no differences were found showing that the own name captured attention and distracted the participants more than an irrelevant word or a control name. The results elucidate the role of the own name as a potent auditory distractor and possible limitations with its theoretical significance for general theories of attention are discussed.

  • The Impact of Emotional Deviant Sounds on Emoji Faces in a Sustained Attention Task

    • The involuntary shift of attention to emotional sounds were investigated in a cross-modal oddball task in which participants categorized angry and disappointed emoji faces. Prior to each face, a standard tone was presented (80% of trials) or a deviant “disappointed” or a buzzing “angry” sound (20% of trials). The deviant trials were either congruent (e.g., disappointed sound/disappointed emoji) or incongruent trials (e.g., a disappointed sound/angry emoji). Results showed that the emotional content of the deviant sounds interacted with the processing of the faces, but that the effect was only present in the congruent trials. Participants showed deviance distraction (prolonged response times compared to standard) in the disappointed trials and facilitation (no deviance distraction) in the angry deviant trials. The facilitation (or lack of distraction) caused by the angry deviant sound in the congruent trial may have been a result of an arousal effect due to the processing of threat.

  • Attention capture by sudden and unexpected changes : a multisensory perspective

    • The main focus for this thesis was cross-modal attention capture by sudden and unexpected sounds and vibrations, known as deviants, presented in a stream the same to-be-ignored stimulus. More specifically, the thesis takes a multisensory perspective and examines the possible similarities and differences in how deviant vibrations and sounds affect visual task performance (Study I), and whether the deviant and standard stimuli have to be presented within the same modality to capture attention away from visual tasks (Study II). Furthermore, by presenting spatial deviants (changing the source of the stimuli from one side of the body to the other) in audiotactile (bimodal), tactile, and auditory to-be-ignored, it explores whether bimodal stimuli are more salient compared to unimodal (Study III). In addition, Study III tested the claims that short-term memory is domain-specific.In line with previous research, Study I found that both auditory and tactile deviants captured attention away from the visual task. However, the temporal dynamics between the two modalities seem to differ. That is, it seems like practice causes the effect of vibratory deviants to reduce, whereas this is not the case for auditory deviants. This suggests that there are central mechanisms (detection of the change) and sensory-specific mechanisms.Study II found that the deviant and standard stimuli must be presented within the same modality. If attention capture by deviants is produced by a mismatch within a neural model predicting upcoming stimuli, the neural model is likely built on stimuli within each modality separately.The results of Study III revealed that spatial and verbal short-term memory are negatively affected by a spatial change in to-be-ignored sequences, but only when the change is within a bimodal sequence. These results can be taken as evidence for a unitary account of short-term memory (verbal and spatial information stored in the same storage) and that bimodal stimuli may be integrated into a unitary percept that make any change in the stream more salient.

    • It has been suggested that deviance distraction is caused by unexpected sensory events in the to-be-ignored stimuli violating the cognitive system's predictions of incoming stimuli. The majority of research has used methods where the to-be-ignored expected (standards) and the unexpected (deviants) stimuli are presented within the same modality. Less is known about the behavioral impact of deviance distraction when the to-be-ignored stimuli are presented in different modalities (e.g., standard and deviants presented in different modalities). In three experiments using cross-modal oddball tasks with mixed-modality to-be-ignored stimuli, we examined the distractive role of unexpected auditory deviants presented in a continuous stream of expected standard vibrations. The results showed that deviance distraction seems to be dependent upon the to-be-ignored stimuli being presented within the same modality, and that the simplest omission of something expected; in this case, a standard vibration may be enough to capture attention and distract performance.

  • The Effects of Unexpected and Sudden Vibrations and Sounds Does Not Disrupt Performance Similarly Over Time

    • Research on the effects of sudden and unexpected vibrations (i.e., deviants) have on visual tasks is scarce. Previous research has shown that vibrating deviants disrupt performance (i.e., deviance distraction) in visual digit categorization tasks in a similar manner as auditory deviants; however, this research has not used methods feasible for examining the temporal aspects of the effects. In our experiment, auditory and tactile stimuli were presented in different parts to examine the temporal aspects of deviance distraction of sounds and vibrations. Deviance distraction was found in both modalities. The effects of auditory deviants remained throughout the auditory part of the experiment whereas the effects of tactile deviants did not. Our results indicate that although deviance distraction may share similar mechanisms, the temporal aspects of deviance distraction might be dissimilar in the two modalities.

    • In this study a cross-modal oddball task was employed to study the effect that words spokeneither non-urgently or urgently would have on a digit categorization task and if women wouldexhibit greater behavioral inhibitory control. The words were unrelated to the task itself, butrelated to the action required to complete the task. Forty participants (21 women) conducteda computerized categorization task while exposed to a sinewave tone as a standard stimulus(75% of the trials) or a to-be ignored word (press, stop) spoken either non-urgently orurgently as unexpected auditory deviant stimulus (6.25% trials for each category). Urgentwords had sharp intonation and an average fundamental frequency (F0) ranging from 191.9(stop) to 204.6 (press) Hz. Non-urgent words had low intonation with average F0 rangingfrom 103.9.9 (stop) to 120.3 (press) Hz. As expected, deviant distraction and longerresponse times were found by exposure to the word stop, but deviant distraction was notfound to be significant with the word press or due to intonation. While the results showedthat women had in general longer reaction times, there were no gender differences foundrelated to the deviant distraction caused by word or intonation. The present results do notsupport the hypothesis that women have greater behavioral inhibitory control, but there wasevidence that the meaning of the word could influence response times.

    • Individual differences in cognitive performance increase with advancing age, reflecting marked cognitive changes in some individuals along with little or no change in others. Genetic and lifestyle factors are assumed to influence cognitive performance in aging by affecting the magnitude and extent of age-related brain changes (i.e., brain maintenance or atrophy), as well as the ability to recruit compensatory processes. The purpose of this review is to present findings from the Betula study and other longitudinal studies, with a focus on clarifying the role of key biological and environmental factors assumed to underlie individual differences in brain and cognitive aging. We discuss the vital importance of sampling, analytic methods, consideration of non-ignorable dropout, and related issues for valid conclusions on factors that influence healthy neurocognitive aging.

    • Alarm systems may take advantage of the tactile modality for allocation of attentional resources during the performance of demanding tasks in complex environments. The purpose of this study was to determine the effectiveness of a tactile warning during increasing levels of mental workload in a primary task. Three simulated flight task conditions varying in mental workload were presented while an “on-thigh” vibrotactile warning was randomly assessed. Generally, there was a decrement in overall warning response performance when task workload increased, but this tendency faded and plateaued as the level of task workload progressed. The observed pattern indicates that vibrotactile warning signals may offer a plausible mode for conveying information during increasing levels of primary task workload.

    • Introduction: The negative cognitive effects of the startle response are not yet fully understood. Ecological observations in the aviation field indicate risk for severe outcomes in complex or pressured situations, while sparse previous research suggests milder negative effects on simple cognitive tasks. Neuroticism is proposed as a factor related to the level of negative effects following startle.Methods: This study examined the effects of startle on performance in a choice reaction time task and analyzed relations between performance, neuroticism, and physiological stress.Results: Our results indicate that reaction time directly following startle was not affected, but reaction time in subsequent trials was significantly slower. Neuroticism and physiological stress were both unrelated to this performance effect.Discussion: We argue that higher complexity/demand tasks are necessary to complement the research on base cognitive functioning in relation to startle. If neuroticism is related to startle effects, this is more likely to be found in these higher demand situations.

  • Testing the Foreign Language Effect on Cognitive Reflection in Older Adults

    • An increasing number of people around the world communicate in more than one language, resulting in them having to make decisions in a foreign language on a daily basis. Interestingly, a burgeoning body of literature suggests that people’s decision-making is affected by whether they are reasoning in their native language (NL) or their foreign language (FL). According to the foreign language effect (FLe), people are less susceptible to bias in many decision-making tasks and more likely to display utilitarian cost-benefit analysis in moral decision-making when reasoning in a FL. While these differences have often been attributed to a reduced emotionality in the FL, an emerging body of literature has started to test the extent to which these could be attributable to increased deliberation in the FL. The present study tests whether increased deliberation leads to a FLe on cognitive reflection in a population of older adults (Mage = 65.1), from the successful aging project in Umeå, Sweden. We explored whether performance on a 6-item version of the cognitive reflection test (CRT) adapted to Swedish would differ between participants for whom Swedish was their NL and those for whom Swedish was their FL. The CRT is a task designed to elicit an incorrect, intuitive answer. In order to override the intuitive answer, one requires engaging in deliberative, analytical thinking to determine the correct answer. Therefore, we hypothesized that if thinking in a FL increases deliberation, then those performing the task in their FL would exhibit higher accuracy rates than those performing in their NL. Our results showed that age and level of education predicted performance on the task but performance on the CRT did not differ between the NL and the FL groups. In addition, in the FL group, proficiency in the FL was not related to performance in the CRT. Our results, therefore, do not provide evidence that thinking in a FL increases deliberation in a group of older adults performing a logical reasoning task that is not typically associated with an emotional connotation.
