  • Monogenic Primary Immunodeficiency Disorder Associated with Common Variable Immunodeficiency and Autoimmunity
  • 2020
  • Ingår i: International archives of allergy and immunology. - : S. Karger AG. - 1423-0097 .- 1018-2438. ; 181:9, s. 706-714
  • Tidskriftsartikel (refereegranskat)abstract
    • <b><i>Background:</i></b> Common variable immunodeficiency (CVID) is the most frequent primary immunodeficiency disorder mainly characterized by recurrent bacterial infections besides other immunological defects including loss of or dysfunction of B cells and decreased immunoglobulin levels. In this study, our aim is to evaluate clinical, immunological, and molecular data of patients with a primary clinical diagnosis of CVID and autoimmune phenotype with a confirmed genetic diagnosis. <b><i>Methods:</i></b> Among 297 patients with CVID, who were registered in the Iranian Primary Immunodeficiency Registry at Children’s Medical Center Hospital in Iran, 83 patients have been genetically examined and 27 patients with autoimmunity and confirmed genetic mutations were selected for analysis. Whole-exome sequencing and confirmatory Sanger sequencing methods were used for the study population. A questionnaire was retrospectively filled for all patients to evaluate demographic, laboratory, clinical, and genetic data. <b><i>Results:</i></b> In the 27 studied patients, 11 different genetic defects were identified, and the most common mutated gene was <i>LRBA</i>, reported in 17 (63.0%) patients. Two patients (7.7%) showed autoimmune complications as the first presentation of immunodeficiency. Eleven patients (40.7%) developed one type of autoimmunity, and 16 patients (59.3%) progressed to poly-autoimmunity. Most of the patients with mono-autoimmunity (<i>n</i> = 9, 90.0%) primarily developed infectious complications, while in patients with poly-autoimmunity, the most common first presentation was enteropathy (<i>n</i> = 6, 37.6%). In 13 patients (61.9%), the diagnosis of autoimmune disorders preceded the diagnosis of primary immunodeficiency. The most frequent autoimmune manifestations were hematologic (40.7%), gastrointestinal (48.1%), rheumatologic (25.9%), and dermatologic (22.2%) disorders. Patients with poly-autoimmunity had lower regulatory T cells than patients with mono-autoimmunity. <b><i>Conclusion:</i></b> In our cohort, the diagnosis of autoimmune disorders preceded the diagnosis of primary immunodeficiency in most patients. This association highlights the fact that patients referring with autoimmune manifestations should be evaluated for humoral immunity.

  • Healthy ecosystems for human and animal health : Science diplomacy for responsible development in the Arctic The Nordic Centre of Excellence, Clinf.org (Climate-change effects on the epidemiology of infectious diseases and the impacts on Northern societies)
  • 2021
  • Ingår i: Polar Record. - : Cambridge University Press (CUP). - 0032-2474 .- 1475-3057. ; 57
  • Forskningsöversikt (refereegranskat)abstract
    • Climate warming is occurring most rapidly in the Arctic, which is both a sentinel and a driver of further global change. Ecosystems and human societies are already affected by warming. Permafrost thaws and species are on the move, bringing pathogens and vectors to virgin areas. During a five-year project, the CLINF - a Nordic Center of Excellence, funded by the Nordic Council of Ministers, has worked with the One Health concept, integrating environmental data with human and animal disease data in predictive models and creating maps of dynamic processes affecting the spread of infectious diseases. It is shown that tularemia outbreaks can be predicted even at a regional level with a manageable level of uncertainty. To decrease uncertainty, rapid development of new and harmonised technologies and databases is needed from currently highly heterogeneous data sources. A major source of uncertainty for the future of contaminants and infectious diseases in the Arctic, however, is associated with which paths the majority of the globe chooses to follow in the future. Diplomacy is one of the most powerful tools Arctic nations have to influence these choices of other nations, supported by Arctic science and One Health approaches that recognise the interconnection between people, animals, plants and their shared environment at the local, regional, national and global levels as essential for achieving a sustainable development for both the Arctic and the globe.

  • ARFGAP1 Is Dynamically Associated with Lipid Droplets in Hepatocytes
  • 2014
  • Ingår i: Plos One. - : Public Library of Science (PLoS). - 1932-6203. ; 9:11
  • Tidskriftsartikel (refereegranskat)abstract
    • The ARF GTPase Activating Protein 1 (ARFGAP1) associates mainly with the cytosolic side of Golgi cisternal membranes where it participates in the formation of both COPI and clathrin-coated vesicles. In this study, we show that ARFGAP1 associates transiently with lipid droplets upon addition of oleate in cultured cells. Also, that addition of cyclic AMP shifts ARFGAP1 from lipid droplets to the Golgi apparatus and that overexpression and knockdown of ARFGAP1 affect lipid droplet formation. Examination of human liver tissue reveals that ARFGAP1 is found associated with lipid droplets at steady state in some but not all hepatocytes.

  • Hydro-climatic changes of wetlandscapes across the world
  • 2021
  • Ingår i: Scientific Reports. - : Springer Science and Business Media LLC. - 2045-2322. ; 11:1
  • Tidskriftsartikel (refereegranskat)abstract
    • Assessments of ecosystem service and function losses of wetlandscapes (i.e., wetlands and their hydrological catchments) suffer from knowledge gaps regarding impacts of ongoing hydro-climatic change. This study investigates hydro-climatic changes during 1976–2015 in 25 wetlandscapes distributed across the world’s tropical, arid, temperate and cold climate zones. Results show that the wetlandscapes were subject to precipitation (P) and temperature (T) changes consistent with mean changes over the world’s land area. However, arid and cold wetlandscapes experienced higher T increases than their respective climate zone. Also, average P decreased in arid and cold wetlandscapes, contrarily to P of arid and cold climate zones, suggesting that these wetlandscapes are located in regions of elevated climate pressures. For most wetlandscapes with available runoff (R) data, the decreases were larger in R than in P, which was attributed to aggravation of climate change impacts by enhanced evapotranspiration losses, e.g. caused by land-use changes.

  • Clinical Manifestations, Immunological Characteristics and Genetic Analysis of Patients with Hyper-Immunoglobulin M Syndrome in Iran
  • 2019
  • Ingår i: International archives of allergy and immunology. - : S. Karger AG. - 1423-0097 .- 1018-2438. ; 180:1, s. 52-63
  • Tidskriftsartikel (refereegranskat)abstract
    • <b><i>Background:</i></b> Hyper-immunoglobulin M (HIGM) syndrome is a rare heterogeneous group of primary immunodeficiency disorders characterized by low or absent serum levels of IgG and IgA along with normal or elevated serum levels of IgM. <b><i>Methods:</i></b> Clinical and immunological data were collected from the 75 patients’ medical records diagnosed in Children’s Medical Center affiliated to Tehran University Medical Sciences and other Universities of Medical Sciences in Iran. Among 75 selected patients, 48 patients (64%) were analyzed genetically using targeted and whole-exome sequencing. <b><i>Results:</i></b> The ratio of male to female was 2.9:1. The median age at the onset of the disease, time of diagnosis, and diagnostic delay were 10.5, 50, and 24 months, respectively. Pneumonia and lower respiratory tract infections (61.3%) were the most common complications. Responsible genes were identified in 35 patients (72.9%) out 48 ge<i>netically</i> analyzed patients. <i>Cluster of differentiation 40 ligand</i> gene was the most mutated gene observed in 24 patients (68.5%) followed by <i>activation-induced cytidine deaminase</i> gene in 7 patients, <i>lipopolysaccharide-responsive and beige-like anchor</i> (1 patient), <i>nuclear factor-kappa-B essential modulator</i> (1 patient), <i>phosphoinositide-3-kinase regulatory subunit 1</i> (1 patient), and <i>nuclear factor kappa B subunit 1</i> (1 patient) genes. Nineteen (25.3%) patients died during the study period, and pneumonia was the major cause of death occurred in 6 (31.6%) patients. <b><i>Conclusion:</i></b> Physicians in our country should carefully pay attention to respiratory tract infections and pneumonia, particularly in patients with a positive family history. Further investigations are required for detection of new genes and pathways resulting in HIGM phenotype.

  • Rhenium Perrhenate (188ReO4) Induced Apoptosis and Reduced Cancerous Phenotype in Liver Cancer Cells
  • 2022
  • Ingår i: Cells. - : MDPI AG. - 2073-4409. ; 11:2
  • Tidskriftsartikel (refereegranskat)abstract
    • Recurrence in hepatocellular carcinoma (HCC) after conventional treatments is a crucial challenge. Despite the promising progress in advanced targeted therapies, HCC is the fourth leading cause of cancer death worldwide. Radionuclide therapy can potentially be a practical targeted approach to address this concern. Rhenium-188 (188Re) is a β-emitting radionuclide used in the clinic to induce apoptosis and inhibit cell proliferation. Although adherent cell cultures are efficient and reliable, appropriate cell-cell and cell-extracellular matrix (ECM) contact is still lacking. Thus, we herein aimed to assess 188Re as a potential therapeutic component for HCC in 2D and 3D models. The death rate in treated Huh7 and HepG2 lines was significantly higher than in untreated control groups using viability assay. After treatment with 188ReO4, Annexin/PI data indicated considerable apoptosis induction in HepG2 cells after 48 h but not Huh7 cells. Quantitative RT-PCR and western blotting data also showed increased apoptosis in response to 188ReO4 treatment. In Huh7 cells, exposure to an effective dose of 188ReO4 led to cell cycle arrest in the G2 phase. Moreover, colony formation assay confirmed post-exposure growth suppression in Huh7 and HepG2 cells. Then, the immunostaining displayed proliferation inhibition in the 188ReO4-treated cells on 3D scaffolds of liver ECM. The PI3-AKT signaling pathway was activated in 3D culture but not in 2D culture. In nude mice, Huh7 cells treated with an effective dose of 188ReO4 lost their tumor formation ability compared to the control group. These findings suggest that 188ReO4 can be a potential new therapeutic agent against HCC through induction of apoptosis and cell cycle arrest and inhibition of tumor formation. This approach can be effectively combined with antibodies and peptides for more selective and personalized therapy.

  • Sustainable futures over the next decade are rooted in soil science
  • 2022
  • Ingår i: European Journal of Soil Science. - : Wiley. - 1351-0754 .- 1365-2389. ; 73:1
  • Tidskriftsartikel (refereegranskat)abstract
    • The importance of soils to society has gained increasing recognition over the past decade, with the potential to contribute to most of the United Nations’ Sustainable Development Goals (SDGs). With unprecedented and growing demands for food, water and energy, there is an urgent need for a global effort to address the challenges of climate change and land degradation, whilst protecting soil as a natural resource. In this paper, we identify the contribution of soil science over the past decade to addressing gaps in our knowledge regarding major environmental challenges: climate change, food security, water security, urban development, and ecosystem functioning and biodiversity. Continuing to address knowledge gaps in soil science is essential for the achievement of the SDGs. However, with limited time and budget, it is also pertinent to identify effective methods of working that ensure the research carried out leads to real-world impact. Here, we suggest three strategies for the next decade of soil science, comprising a greater implementation of research into policy, interdisciplinary partnerships to evaluate function trade-offs and synergies between soils and other environmental domains, and integrating monitoring and modelling methods to ensure soil-based policies can withstand the uncertainties of the future. Highlights: We highlight the contributions of soil science to five major environmental challenges since 2010. Researchers have contributed to recommendation reports, but work is rarely translated into policy. Interdisciplinary work should assess trade-offs and synergies between soils and other domains. Integrating monitoring and modelling is key for robust and sustainable soils-based policymaking.

  • Development of novel hybridized models for urban flood susceptibility mapping
  • 2020
  • Ingår i: Scientific Reports. - : Nature Research. - 2045-2322. ; 10:1
  • Tidskriftsartikel (refereegranskat)abstract
    • Floods in urban environments often result in loss of life and destruction of property, with many negative socio-economic effects. However, the application of most flood prediction models still remains challenging due to data scarcity. This creates a need to develop novel hybridized models based on historical urban flood events, using, e.g., metaheuristic optimization algorithms and wavelet analysis. The hybridized models examined in this study (Wavelet-SVR-Bat and Wavelet-SVR-GWO), designed as intelligent systems, consist of a support vector regression (SVR), integrated with a combination of wavelet transform and metaheuristic optimization algorithms, including the grey wolf optimizer (GWO), and the bat optimizer (Bat). The efficiency of the novel hybridized and standalone SVR models for spatial modeling of urban flood inundation was evaluated using different cutoff-dependent and cutoff-independent evaluation criteria, including area under the receiver operating characteristic curve (AUC), Accuracy (A), Matthews Correlation Coefficient (MCC), Misclassification Rate (MR), and F-score. The results demonstrated that both hybridized models had very high performance (Wavelet-SVR-GWO: AUC = 0.981, A = 0.92, MCC = 0.86, MR = 0.07; Wavelet-SVR-Bat: AUC = 0.972, A = 0.88, MCC = 0.76, MR = 0.11) compared with the standalone SVR (AUC = 0.917, A = 0.85, MCC = 0.7, MR = 0.15). Therefore, these hybridized models are a promising, cost-effective method for spatial modeling of urban flood susceptibility and for providing in-depth insights to guide flood preparedness and emergency response services.

  • Ingår i: International journal of STD & AIDS. - : SAGE Publications. - 0956-4624 .- 1758-1052. ; 7:5, s. 333-336
  • Tidskriftsartikel (refereegranskat)abstract
    • In a prospective cohort study on the prevalence of HPV in a group of female Swedish students, we obtained repeated cytological specimens during a period of 2 years, for cytological diagnosis and PCR detection of HPV. The group comprised 98 girls, sampled between 15 and 17 years, and 82 of them completed the study. The girls were also followed every sixth month by a structured face-to-face interview regarding sexuality. Only the sexually experienced girls harboured HPVDNA in the cervix with a cumulative prevalence of 37% and HPV16 was the most common type encountered. Almost half of the infected girls were lacking any clinical or subclinical signs of HPV infection. There was a positive correlation between the presence of HPV and the number of coital partners. The time period since coitarche also had importance for the HPV detection rate.

  • Contribution of Satellite-Based Precipitation in Hydrological Rainfall–Runoff Modeling : Case Study of the Hammam Boughrara Region in Algeria
  • 2021
  • Ingår i: Earth Systems and Environment. - : Springer Nature. - 2509-9426 .- 2509-9434. ; 5:4, s. 873-881
  • Tidskriftsartikel (refereegranskat)abstract
    • Hydrological models are viewed as powerful tools that have a major importance for managing water resources and predicting flows. It should be specified that the meteorological parameter rainfall is the main input in these models. In the current study, data from only one rainfall station are available over the analysis domain, which cannot represent the entire Hammam Boughrara watershed of Algeria. The precipitation data remotely detected by the tropical rainfall measuring mission (TRMM) provide good spatial coverage in the watershed and can be used to fill in the missing data. The use of raw TRMM data gives poor results from the simulated flow rates with a Nash–Sutcliffe efficiency NSE equal to 0.34 for the validation period that ranges from year 2000 to 2005; this is mainly due to uncertainties in the TRMM data. For this reason, it was deemed necessary to carry out a performance test of the model. The results obtained give an unsatisfactory percent bias (PBIAS) of − 46.24%, which suggests the need to perform a correction to decrease the PBIAS of satellite precipitation. For this, two methods were used: the linear regression method and the multiplicative method. These two techniques can only be applied if there is at least one rainfall measurement station available in the watershed. The obtained results are very satisfactory since the PBIAS reaches − 0.62% for the linear regression method and − 11.58% for the multiplicative method. In addition, the use of corrected TRMMs gives also very good results with a Nash–Sutcliffe efficiency that ranges from 0.74 to 0.88 for both validation and calibration periods. Overall, the current study is supportive to estimate the satellite-based rainfall, one of the very sensitive to measure the meteorological parameter, in northwestern Algeria.

  • Twenty-three unsolved problems in hydrology (UPH) - a community perspective
  • 2019
  • Ingår i: Hydrological Sciences Journal. - : Informa UK Limited. - 0262-6667 .- 2150-3435. ; 64:10, s. 1141-1158
  • Tidskriftsartikel (refereegranskat)abstract
    • This paper is the outcome of a community initiative to identify major unsolved scientific problems in hydrology motivated by a need for stronger harmonisation of research efforts. The procedure involved a public consultation through online media, followed by two workshops through which a large number of potential science questions were collated, prioritised, and synthesised. In spite of the diversity of the participants (230 scientists in total), the process revealed much about community priorities and the state of our science: a preference for continuity in research questions rather than radical departures or redirections from past and current work. Questions remain focused on the process-based understanding of hydrological variability and causality at all space and time scales. Increased attention to environmental change drives a new emphasis on understanding how change propagates across interfaces within the hydrological system and across disciplinary boundaries. In particular, the expansion of the human footprint raises a new set of questions related to human interactions with nature and water cycle feedbacks in the context of complex water management problems. We hope that this reflection and synthesis of the 23 unsolved problems in hydrology will help guide research efforts for some years to come.

  • Hydrological Processes in Eucalypt and Pine Forested Headwater Catchments within Mediterranean Region
  • 2021
  • Ingår i: Water. - : MDPI AG. - 2073-4441. ; 13:10
  • Tidskriftsartikel (refereegranskat)abstract
    • Mediterranean headwater catchments have experienced major land-use changes in recent centuries, namely characterized by afforestation with fast-growing tree species (e.g., pine and eucalyptus). This paper investigates differences in the hydrological response of two forested catchments with distinct Pinus pinaster Aiton (PIN) and Eucalyptus globulus Labill. (EUC) covers over 6 hydrological years, from 2010 to 2016. The study was developed in paired catchments located in North-Central Portugal, under wet Mediterranean climate and schist bedrock. Based on rainfall and streamflow measurements, the hydrological responses were analyzed through the water-balance calculations, rainfall-runoff event analyses, and correlations between rainfall characteristics, evapotranspiration, soil moisture, overland flow, and land-cover parameters. The study period recorded a wide range of annual rainfall, between 984 and 2127 mm per years. The annual streamflow displayed greater inter-annual variation, from 208 to 1430 mm for PIN and from 217 to 1291 mm for EUC. The runoff coefficient was higher in the wettest years, with a maximum of 58% for PIN and 61% for EUC, and decreased substantially in the driest year, representing only 17% of the rainfall in PIN and 22% in EUC. Rainfall was predominant during the winter season (41%), followed by autumn (30%) and spring (24%). The streamflow displayed a seasonal behavior in both catchments, with higher runoff coefficient for EUC that for PIN. Half of the annual streamflow occurs during the winter, with the highest average runoff coefficient attained for EUC and PIN respectively being 68% and 64% of the rainfall amount. Annual evapotranspiration was relatively constant through the six years of study and showed higher average values in PIN (907 mm) than EUC (739 mm), highlighting the importance of forest type and showing that mature pine plantations lead to higher water consumption than eucalypt stands. Annual rainfall amount affects evapotranspiration rate (%), the percentage of ET increasing from 37% to 78% for PIN and from 34% to 73% for EUC between the wettest and the driest years, which raises concerns regarding the impact of climate changes on water availability in the mountain areas of the Mediterranean region.

  • Development of a novel hybrid multi-boosting neural network model for spatial prediction of urban flood
  • 2021
  • Ingår i: Geocarto International. - : Taylor and Francis Ltd.. - 1010-6049 .- 1752-0762.
  • Tidskriftsartikel (refereegranskat)abstract
    • In this study, a new hybridized machine learning algorithm for urban flood susceptibility mapping, named MultiB-MLPNN, was developed using a multi-boosting technique and MLPNN. The model was tested in Amol City, Iran, a data-scarce city in an ungauged area which is prone to severe flood inundation events and currently lacks flood prevention infrastructure. Performance of the hybridized model was compared with that of a standalone MLPNN model, random forest and boosted regression trees. Area under the curve, efficiency, true skill statistic, Matthews correlation coefficient, misclassification rate, sensitivity and specificity were used to evaluate model performance. In validation, the MultiB-MLPNN model showed the best predictive performance. The hybridized MultiB-MLPNN model is thus useful for generating realistic flood susceptibility maps for data-scarce urban areas. The maps can be used to develop risk-reduction measures to protect urban areas from devastating floods, particularly where available data are insufficient to support physically based hydrological or hydraulic models.

  • Hydrological challenges in urban areas
  • 2022
  • Ingår i: Advances in Chemical Pollution, Environmental Management and Protection. - : Elsevier B.V.. ; , s. 47-67
  • Bokkapitel (refereegranskat)abstract
    • Urbanization alters hydrological processes and is often associated with increasing flood risks, which threaten human wellbeing and social and economic development. The conventional paradigm of flood protection relying on structural measures based on hard engineering solutions (e.g., dams, piped systems) has proven insufficient to mitigate floods. Sustainable water management, including solutions to enhance natural processes within urban areas, is a promising approach to enhance flood resilience and address the multiple sustainability challenges faced by cities. However, implementation of solutions based on mimicking natural processes has been slow. Mainstreaming of urban sustainable flood management is inhibited by governance aspects (e.g., limited collaborative governance), and knowledge gaps on effectiveness compared with conventional engineering approaches. The increasing flood hazards driven by growing urban populations and climate change projections of increasing frequency and intensity of large precipitation events demand improvements in spatial planning. This also provides opportunities for sustainable water management mainstreaming in order to complement the relatively limited drainage capacity of conventional systems.

  • Impact of Land-Use Changes on Spatiotemporal Suspended Sediment Dynamics within a Peri-Urban Catchment
  • 2020
  • Ingår i: Water. - : MDPI AG. - 2073-4441. ; 12:3
  • Tidskriftsartikel (refereegranskat)abstract
    • Understanding sediment dynamics in peri-urban catchments constitutes a research challenge because of the spatiotemporal complexity and variability of land-uses involved. This study investigates differences in the concentration of total sediments (TSC) and suspended sediments (SSC) in the small peri-urban Mediterranean Ribeira dos Covoes catchment (40% urban area) in central Portugal. Suspended sediment responses at the catchment outlet (E) and in three upstream sub-catchments, during periods of urbanization (2011-2013) and stabilizing land-use (2017-2018) are compared for storm-event datasets encompassing similar ranges of rainstorm sizes and antecedent rainfall condition. The Quinta sub-catchment, with the lowest urban area (22%) but subject to major construction activities affecting 17% of its area, led to highest TSC and SSC during urbanization (attaining 4320 mg/L and 4184 mg/L, respectively), and a median reduction of 38% and 69%, respectively, during stabilization. Espirito Santo sub-catchment, with highest urban area (49%) and minor construction activities, displayed similar median TSC in both periods (258-240 mg/L) but highest SSC reduction (76%), highlighting the impact of the anthropogenic disturbance mainly on fine-particle sediments and a good connectivity with the stream. Porto Bordalo sub-catchment, with 39% urban area and subject to the construction of a four-line road covering 1.5% of its area, showed the lowest TSC and SSC concentrations and the lowest median reductions in both periods (31% and 64%, correspondingly), mainly because of the impact of an unplanned retention basin established with soil from the construction site. Overall, median TSC and SSC reduced 14% and 59% at E, from urbanization to stabilization. Information about sediment dynamics should guide stakeholders in establishing strategies to reduce sediment loads and mitigate the impacts on urban aquatic ecosystems.

  • 2001
  • Ingår i: Laboratory Investigation. - : Elsevier BV. - 0023-6837 .- 1530-0307. ; 81:5, s. 673-679
  • Tidskriftsartikel (refereegranskat)abstract
    • The possibility of using a new bioluminometric UNA sequencing technique, called pyrosequencing, for typing of human papillomaviruses (HPV) was investigated. A blinded pyrosequencing test was performed on an HPV test panel of 67 GP5+/GP6+ PCR-derived amplification products. The 67 clinical DNA samples were sequenced up to 25 bases and sequences were searched using BLAST. All of the samples were correctly genotyped by pyrosequencing and the results were unequivocally in accordance with the results obtained from conventional DNA sequencing. Pyrosequencing was found to be a fast and efficient tool for identifying individual HPV types. Furthermore, pyrosequencing has the capability of determining novel HPV types as well as HPV sequence variants harboring mutation(s). The method is robust and well suited for large-scale programs.

  • 2017
  • Ingår i: Frontiers in Microbiology. - : Frontiers Media SA. - 1664-302X. ; 8:APR
  • Tidskriftsartikel (refereegranskat)abstract
    • 3-Hydroxypropanoic acid (3-HP) is an important biomass-derivable platform chemical that can be converted into a number of industrially relevant compounds. There have been several attempts to produce 3-HP from renewable sources in cell factories, focusing mainly on Escherichia coli, Klebsiella pneumoniae, and Saccharomyces cerevisiae. Despite the significant progress made in this field, commercially exploitable large-scale production of 3-HP in microbial strains has still not been achieved. In this study, we investigated the potential of Bacillus subtilis as a microbial platform for bioconversion of glycerol into 3-HP. Our recombinant B. subtilis strains overexpress the two-step heterologous pathway containing glycerol dehydratase and aldehyde dehydrogenase from K. pneumoniae. Genetic engineering, driven by in silico optimization, and optimization of cultivation conditions resulted in a 3-HP titer of 10 g/L, in a standard batch cultivation. Our findings provide the first report of successful introduction of the biosynthetic pathway for conversion of glycerol into 3-HP in B. subtilis. With this relatively high titer in batch, and the robustness of B. subtilis in high density fermentation conditions, we expect that our production strains may constitute a solid basis for commercial production of 3-HP.

  • Directional Modulation Via Symbol-Level Precoding : A Way to Enhance Security

  • Tidskriftsartikel (refereegranskat)abstract
    • Wireless communication provides a wide coverage at the cost of exposing information to unintended users. As an information-theoretic paradigm, secrecy rate derives bounds for secure transmission when the channel to the eavesdropper is known. However, such bounds are shown to be restrictive in practice and may require exploitation of specialized coding schemes. In this paper, we employ the concept of directional modulation and follow a signal processing approach to enhance the security of multiuser multi-input multioutput (MIMO) communication systems when a multiantenna eavesdropper is present. Security enhancement is accomplished by increasing the symbol error rate at the eavesdropper. Unlike the information-theoretic secrecy rate paradigm, we assume that the legitimate transmitter is not aware of its channel to the eavesdropper, which is a more realistic assumption. We examine the applicability of MIMO receiving algorithms at the eavesdropper. Using the channel knowledge and the intended symbols for the users, we design security enhancing symbol-level precoders for different transmitter and eavesdropper antenna configurations. We transform each design problem to a linearly constrained quadratic program and propose two solutions, namely the iterative algorithm and one based on nonnegative least squares, at each scenario for a computationally efficient modulation. Simulation results verify the analysis and show that the designed precoders outperform the benchmark scheme in terms of both power efficiency and security enhancement.

  • Secrecy Analysis on Network Coding in Bidirectional Multibeam Satellite Communications
  • 2015
  • Ingår i: IEEE Transactions on Information Forensics and Security. - : Institute of Electrical and Electronics Engineers (IEEE). - 1556-6013 .- 1556-6021. ; 10:9, s. 1862-1874
  • Tidskriftsartikel (refereegranskat)abstract
    • Network coding is an efficient means to improve the spectrum efficiency of satellite communications. However, its resilience to eavesdropping attacks is not well understood. This paper studies the confidentiality issue in a bidirectional satellite network consisting of two mobile users who want to exchange message via a multibeam satellite using the XOR network coding protocol. We aim to maximize the sum secrecy rate by designing the optimal beamforming vector along with optimizing the return and forward link time allocation. The problem is nonconvex, and we find its optimal solution using semidefinite programming together with a 1-D search. For comparison, we also solve the sum secrecy rate maximization problem for a conventional reference scheme without using network coding. Simulation results using realistic system parameters demonstrate that the bidirectional scheme using network coding provides considerably higher secrecy rate compared with that of the conventional scheme.

  • Millimeter wave Wideband and Low-loss Compact Power Divider Based on Gap Waveguide: For Use in Wideband Antenna Array System
  • 2024
  • Ingår i: IEEE Access. - 2169-3536 .- 2169-3536. ; In Press
  • Tidskriftsartikel (refereegranskat)abstract
    • This paper presents a wideband and low-loss design of a compact power divider based on gap waveguide technology. The proposed power divider consists of two adjacent E-plane groove gap waveguide and a small ridge section to couple and equally divide the EM energy from the input E-plane groove gap waveguide to the two output ones in-phase. The simulation results show that the proposed waveguide power divider has about 40% impedance bandwidth while its size is 0.7λ × 0.6λ at the center frequency. An 8-way power divider is designed using the proposed 2-way power divider and the back-toback configuration of the 8-way one is fabricated to investigate the performance of the proposed design. In addition, to show the application of the presented power divider, a wideband linear horn antenna array is designed and fabricated. The measured results agree well with the simulated ones and prove the excellent low-loss and wide bandwidth of the proposed power divider over the band of interest from 50-75GHz. The measured S 11 of the entire 8-way power divider remains below -10 dB level, the insertion loss is around 1 dB over the band of interest. Also, the S 11 of the horn array integrated with the 8-way feed network remains below -10 dB and the low sidelobes of the radiation pattern of the entire horn array indicates excellent phase and amplitude balance for the power divider over the entire bandwidth of interest.

  • Fluvial bedload transport modelling: advanced ensemble tree-based models or optimized deep learning algorithms?
  • 2024
  • Ingår i: Engineering Applications of Computational Fluid Mechanics. - : Informa UK Limited. - 1994-2060 .- 1997-003X. ; 18:1
  • Tidskriftsartikel (refereegranskat)abstract
    • The potential of advanced tree-based models and optimized deep learning algorithms to predict fluvial bedload transport was explored, identifying the most flexible and accurate algorithm, and the optimum set of readily available and reliable inputs. Using 926 datasets for 20 rivers, the performance of three groups of models was tested: (1) standalone tree-based models Alternating Model Tree (AMT) and Dual Perturb and Combine Tree (DPCT); (2) ensemble tree-based models Iterative Absolute Error Regression (IAER), ensembled with AMT and DPCT; and (3) optimized deep learning models Long Short-Term Memory (LSTM) and Recurrent Neural Network (RNN) ensembled with Grey Wolf Optimizer. Comparison of the predictive performance of the models with that of commonly used empirical equations and sensitivity analysis of the driving variables revealed that: (i) the coarse grain-size percentile D90 was the most effective variable in bedload transport prediction (where Dx is the xth percentile of the bed surface grain size distribution), followed by D84, D50, flow discharge, D16, and channel slope and width; (ii) all tree-based models and optimized deep learning algorithms displayed ‘very good’ or ‘good’ performance, outperforming empirical equations; and (iii) all algorithms performed best when all input parameters were used. Thus, a range of different input variable combinations must be considered in the optimization of these models. Overall, ensemble algorithms provided more accurate predictions of bedload transport than their standalone counterpart. In particular, the ensemble tree-based model IAER-AMT performed most strongly, displaying great potential to produce robust predictions of bedload transport in coarse-grained rivers based on a few readily available flow and channel variables.

  • Power allocation for energy-constrained cognitive radios in the presence of an eavesdropper
  • 2014
  • Ingår i: 2014 IEEE International Conference on Acoustics, Speech and Signal Processing (ICASSP). - : IEEE conference proceedings. ; , s. 5695-5699
  • Konferensbidrag (refereegranskat)abstract
    • Reliable and agile spectrum sensing as well as secure communication are key requirements of a cognitive radio system. In this paper, secrecy throughput of a cognitive radio is maximized in order to determine the sensing threshold, the sensing time, and the transmission power. Constraints of the problem are defined as a lower-bound on the detection probability, an upper-bound on the average energy consumption per time-frame, and the maximum transmission power of the cognitive radio. We show that the problem can be solved by an on-off strategy where the cognitive radio only performs sensing and transmits data if the cognitive channel gain is greater than the average eavesdropper channel gain. The problem is then solved by a line-search over sensing time. Eventually, the secrecy throughput of the cognitive radio is evaluated employing the IEEE 802.15.4/Zig-Bee standard.
