  • Genetic architecture of human plasma lipidome and its link to cardiovascular disease
  • 2019
  • Ingår i: Nature communications. - : Springer Science and Business Media LLC. - 2041-1723. ; 10:1, s. 4329-
  • Tidskriftsartikel (refereegranskat)abstract
    • Understanding genetic architecture of plasma lipidome could provide better insights into lipid metabolism and its link to cardiovascular diseases (CVDs). Here, we perform genome-wide association analyses of 141 lipid species (n = 2,181 individuals), followed by phenome-wide scans with 25 CVD related phenotypes (n = 511,700 individuals). We identify 35 lipid-species-associated loci (P <5 ×10−8), 10 of which associate with CVD risk including five new loci-COL5A1, GLTPD2, SPTLC3, MBOAT7 and GALNT16 (false discovery rate<0.05). We identify loci for lipid species that are shown to predict CVD e.g., SPTLC3 for CER(d18:1/24:1). We show that lipoprotein lipase (LPL) may more efficiently hydrolyze medium length triacylglycerides (TAGs) than others. Polyunsaturated lipids have highest heritability and genetic correlations, suggesting considerable genetic regulation at fatty acids levels. We find low genetic correlations between traditional lipids and lipid species. Our results show that lipidomic profiles capture information beyond traditional lipids and identify genetic variants modifying lipid levels and risk of CVD.

  • FinnGen provides genetic insights from a well-phenotyped isolated population
  • 2023
  • Ingår i: Nature. - : Springer Science and Business Media LLC. - 1476-4687 .- 0028-0836. ; 613:7944, s. 508-
  • Tidskriftsartikel (refereegranskat)abstract
    • Population isolates such as those in Finland benefit genetic research because deleterious alleles are often concentrated on a small number of low-frequency variants (0.1% ≤ minor allele frequency < 5%). These variants survived the founding bottleneck rather than being distributed over a large number of ultrarare variants. Although this effect is well established in Mendelian genetics, its value in common disease genetics is less explored1,2. FinnGen aims to study the genome and national health register data of 500,000 Finnish individuals. Given the relatively high median age of participants (63 years) and the substantial fraction of hospital-based recruitment, FinnGen is enriched for disease end points. Here we analyse data from 224,737 participants from FinnGen and study 15 diseases that have previously been investigated in large genome-wide association studies (GWASs). We also include meta-analyses of biobank data from Estonia and the United Kingdom. We identified 30 new associations, primarily low-frequency variants, enriched in the Finnish population. A GWAS of 1,932 diseases also identified 2,733 genome-wide significant associations (893 phenome-wide significant (PWS), P < 2.6 × 10–11) at 2,496 (771 PWS) independent loci with 807 (247 PWS) end points. Among these, fine-mapping implicated 148 (73 PWS) coding variants associated with 83 (42 PWS) end points. Moreover, 91 (47 PWS) had an allele frequency of <5% in non-Finnish European individuals, of which 62 (32 PWS) were enriched by more than twofold in Finland. These findings demonstrate the power of bottlenecked populations to find entry points into the biology of common diseases through low-frequency, high impact variants.

  • Investigating Mercury's Environment with the Two-Spacecraft BepiColombo Mission
  • 2020
  • Ingår i: Space Science Reviews. - : Springer Science and Business Media LLC. - 0038-6308 .- 1572-9672. ; 216:5
  • Forskningsöversikt (refereegranskat)abstract
    • The ESA-JAXA BepiColombo mission will provide simultaneous measurements from two spacecraft, offering an unprecedented opportunity to investigate magnetospheric and exospheric dynamics at Mercury as well as their interactions with the solar wind, radiation, and interplanetary dust. Many scientific instruments onboard the two spacecraft will be completely, or partially devoted to study the near-space environment of Mercury as well as the complex processes that govern it. Many issues remain unsolved even after the MESSENGER mission that ended in 2015. The specific orbits of the two spacecraft, MPO and Mio, and the comprehensive scientific payload allow a wider range of scientific questions to be addressed than those that could be achieved by the individual instruments acting alone, or by previous missions. These joint observations are of key importance because many phenomena in Mercury's environment are highly temporally and spatially variable. Examples of possible coordinated observations are described in this article, analysing the required geometrical conditions, pointing, resolutions and operation timing of different BepiColombo instruments sensors.

  • Inner southern magnetosphere observation of Mercury via SERENA ion sensors in BepiColombo mission
  • 2022
  • Ingår i: Nature Communications. - : Nature Publishing Group. - 2041-1723. ; 13:1
  • Tidskriftsartikel (refereegranskat)abstract
    • Mercury’s southern inner magnetosphere is an unexplored region as it was not observed by earlier space missions. In October 2021, BepiColombo mission has passed through this region during its first Mercury flyby. Here, we describe the observations of SERENA ion sensors nearby and inside Mercury’s magnetosphere. An intermittent high-energy signal, possibly due to an interplanetary magnetic flux rope, has been observed downstream Mercury, together with low energy solar wind. Low energy ions, possibly due to satellite outgassing, were detected outside the magnetosphere. The dayside magnetopause and bow-shock crossing were much closer to the planet than expected, signature of a highly eroded magnetosphere. Different ion populations have been observed inside the magnetosphere, like low latitude boundary layer at magnetopause inbound and partial ring current at dawn close to the planet. These observations are important for understanding the weak magnetosphere behavior so close to the Sun, revealing details never reached before.

  • TandEM : Titan and Enceladus mission
  • 2009
  • Ingår i: Experimental astronomy. - : Springer Science and Business Media LLC. - 0922-6435 .- 1572-9508. ; 23:3, s. 893-946
  • Tidskriftsartikel (refereegranskat)abstract
    • TandEM was proposed as an L-class (large) mission in response to ESA's Cosmic Vision 2015-2025 Call, and accepted for further studies, with the goal of exploring Titan and Enceladus. The mission concept is to perform in situ investigations of two worlds tied together by location and properties, whose remarkable natures have been partly revealed by the ongoing Cassini-Huygens mission. These bodies still hold mysteries requiring a complete exploration using a variety of vehicles and instruments. TandEM is an ambitious mission because its targets are two of the most exciting and challenging bodies in the Solar System. It is designed to build on but exceed the scientific and technological accomplishments of the Cassini-Huygens mission, exploring Titan and Enceladus in ways that are not currently possible (full close-up and in situ coverage over long periods of time). In the current mission architecture, TandEM proposes to deliver two medium-sized spacecraft to the Saturnian system. One spacecraft would be an orbiter with a large host of instruments which would perform several Enceladus flybys and deliver penetrators to its surface before going into a dedicated orbit around Titan alone, while the other spacecraft would carry the Titan in situ investigation components, i.e. a hot-air balloon (MontgolfiSre) and possibly several landing probes to be delivered through the atmosphere.

  • A comparison of global models for the solar wind interaction with Mars
  • 2010
  • Ingår i: Icarus. - : Elsevier BV. - 0019-1035 .- 1090-2643. ; 206:1, s. 139-151
  • Tidskriftsartikel (refereegranskat)abstract
    • We present initial results from the first community-wide effort to compare global plasma interaction model results for Mars. Seven modeling groups participated in this activity, using MHD, multi-fluid, and hybrid assumptions in their simulations. Moderate solar wind and solar EUV conditions were chosen, and the conditions were implemented in the models and run to steady state. Model output was compared in three ways to determine how pressure was partitioned and conserved in each model, the location and asymmetry of plasma boundaries and pathways for planetary ion escape, and the total escape flux of planetary oxygen ions. The two participating MHD models provided similar results, while the five sets of multi-fluid and hybrid results were different in many ways. All hybrid results, however, showed two main channels for oxygen ion escape (a pickup ion 'plume' in the hemisphere toward which the solar wind convection electric field is directed, and a channel in the opposite hemisphere of the central magnetotail), while the MHD models showed one (a roughly symmetric channel in the central magnetotail). Most models showed a transition from an upstream region dominated by plasma dynamic pressure to a magnetosheath region dominated by thermal pressure to a low altitude region dominated by magnetic pressure. However, calculated escape rates for a single ion species varied by roughly an order of magnitude for similar input conditions, suggesting that the uncertainties in both the current and integrated escape over martian history as determined by models are large. These uncertainties are in addition to those associated with the evolution of the Sun, the martian dynamo, and the early atmosphere, highlighting the challenges we face in constructing Mars' past using models.

  • Mass composition of the escaping plasma at Mars
  • 2006
  • Ingår i: Icarus. - : Elsevier BV. - 0019-1035 .- 1090-2643. ; 182:2, s. 320-328
  • Tidskriftsartikel (refereegranskat)abstract
    • Data from the Ion Mass Analyzer (IMA) sensor of the ASPERA-3 instrument suite on Mars Express have been analyzed to determine the mass composition of the escaping ion species at Mars. We have examined 77 different ion-beam events and we present the results in terms of flux ratios between the following ion species: CO2+/O+ and O-2(+)/O+. The following ratios averaged over all events and energies were identified: CO2+/O+ = 0.2 and O-2(+)/O+ = 0.9. The values measured are significantly higher, by a factor of 10 for O-2(+)/O+, than a contemporary modeled ratio for the maximum fluxes which the martian ionosphere can supply. The most abundant ion species was found to be O+, followed by O-2(+) and CO2+. We estimate the loss of CO2+ to be 4.0 x 10(24) s(-1) (0.29 kg s(-1)) by using the previous measurements of Phobos-2 in our calculations. The dependence of the ion ratios in relation to their energy ranges we studied, 0.3-3.0 keV, indicated that no clear correlation was found.

  • Performance of a multiplexed serological microarray for the detection of antibodies against central nervous system pathogens
  • 2014
  • Ingår i: Journal of Microbiological Methods. - : Elsevier BV. - 0167-7012. ; 100, s. 27-31
  • Tidskriftsartikel (refereegranskat)abstract
    • Central nervous system (CNS) infections have multiple potential causative agents for which simultaneous pathogen screening can provide a useful tool. This study evaluated a multiplexed microarray for the simultaneous detection of antibodies against CNS pathogens. The performance of selected microarray antigens for the detection of IgG antibodies against herpes simplex virus 1 and 2 (HSV-1 and HSV-2), varicella-zoster virus (VZV), adenovirus, Mycoplasma pneumoniae and Borrelia burgdorferi sensu lato, was evaluated using serum sample panels tested with reference assays used in a routine diagnostic laboratory. The microarray sensitivity for HSV-1, HSV-2, VZV, adenovirus and M. pneumonia ranged from 77% to 100%, and the specificity ranged from 74% to 97%. Very variable sensitivities and specificities were found for borrelial antigens of three different VIsE protein IR(6) peptide variants (IR6p1, IR6p2, IR6p4) and three recombinant decorin binding proteins A (DbpA; DbpAla, DbpA91, DbpAG40). For single antigens, good specificity was shown for antigens of IR6p4 and DbpAla (96%), while DbpA91, IR6p1 and IR6p2 were moderately specific (88-92%). The analytical sensitivity of the microarray was dependent on the borrelial IgG concentration of the specimen. The overall performance and technical features of the platform showed that the platform supports both recombinant proteins, whole viruses and peptides as antigens. This study showed diagnostic potential for all six CNS pathogens, including Borrelia burgdorferi sensu lato, using glutaraldehyde based microarray, and further highlighted the importance of careful antigen selection and the requirement for the use of multiple borrelial antigens in order to increase specificity without a major lack of sensitivity. • Based on the current state of technology, distance measurements over a few hundred metres in air with relative uncertainties significantly better than 10_6 are still an almost impossible challenge. In the European Joint Research Project (JRP) “Metrology for long distance surveying” measurement uncertainties in GNSS-based and optical distance metrology are going to be thoroughly investigated, novel technologies and primary standards developed and guidelines to improve surveying practice in the field worked out. A better understanding and a decrease of measurement uncertainty is also targeted for the critical local tie measurement at geodetic fundamental stations.

  • Ageing properties of car fuel-lines;accelerated testing in “close-to-real” service conditions
  • 2010
  • Ingår i: Polymer testing. - : Elsevier. - 0142-9418 .- 1873-2348. ; 29:1, s. 41-48
  • Tidskriftsartikel (refereegranskat)abstract
    • The use of ethanol-based fuels and tougher restrictions on fuel emissions put a higher demand on car fuel-line (pipe) systems. In this context, it is important to be able to establish and predict properties based on measurements on pipes exposed to real or “close-to-real” environments. This paper presents a new method to age pipes in accelerated “close-to-real” conditions. In this method, the pipe is exposed to circulating fuel on the inside and to air on the outside. The method/equipment allows for non-destructive mechanical testing on “continuous” pipes. The usefulness of the ageing method/system was illustrated on polyamide-12 (PA12) pipes exposed to fuels with varying ethanol content at 50 °C and 110 °C for a maximum of, respectively, ca 3 years and 100 days. “Non-destructive” three-point bending as well as tensile testing was used to assess the ageing-induced changes in mechanical properties. The most conclusive information was that the lowest pipe extensibility (ductility) of dried, previously fuel-exposed pipes was observed at the end of the ageing periods and at the higher ethanol contents. In fact, optical microscopy showed that the tensile fractured pipes, exposed to 25/30 vol. % ethanol at 110 °C (100 h), showed no signs of macroscopic yielding. The trends were interpreted, based also on findings from previous work, as being due to the loss of plasticiser (possibly also PA12 monomers/oligomers) and material “degradation/annealing” processes, the latter involving possibly stabiliser issues.

  • Castration Induces Up-Regulation of Intratumoral Androgen Biosynthesis and Androgen Receptor Expression in an Orthotopic VCaP Human Prostate Cancer Xenograft Model
  • 2014
  • Ingår i: American Journal of Pathology. - : Elsevier BV. - 0002-9440. ; 184:8, s. 2163-2173
  • Tidskriftsartikel (refereegranskat)abstract
    • Androgens are key factors involved in the development and progression of prostate cancer (PCa), and PCa growth can be suppressed by androgen deprivation therapy. In a considerable proportion of men receiving androgen deprivation therapy, however, PCa progresses to castration-resistant PCa (CRPC), making the development of efficient therapies challenging. We used an orthotopic VCaP human PCa xenograft model to study cellular and molecular changes in tumors after androgen deprivation therapy (castration). Tumor growth was monitored through weekly serum prostate-specific antigen measurements, and mice with recurrent tumors after castration were randomized to treatment groups. Serum prostate-specific antigen concentrations showed significant correlation with tumor volume. Castration-resistant tumors retained concentrations of intratumoral androgen (androstenedione, testosterone, and 5 alpha-dihydrotestosterone) at Levels similar to tumors growing in intact hosts. Accordingly, castration induced up-regulation of enzymes involved in androgen synthesis (CYP17A1, AKR1C3, and HSD17B6), as well as expression of full-length androgen receptor (AR) and AR splice variants (AR-V1 and AR-V7). Furthermore, AR target gene expression was maintained in castration-resistant xenografts. The AR antagonists enzalutamide (MDV3100) and ARN-509 suppressed PSA production of castration-resistant tumors, confirming the androgen dependency of these tumors. Taken together, the findings demonstrate that our VCaP xenograft model exhibits the key characteristics of clinical CRPC and thus provides a valuable tool for identifying druggable targets and for testing therapeutic strategies targeting AR signaling in CRPC.

  • Exoplanet status report: Observation, characterization and evolution of exoplanets and their host stars
  • 2010
  • Ingår i: Solar System Research. - 1608-3423 .- 0038-0946. ; 44:4, s. 290-310
  • Tidskriftsartikel (refereegranskat)abstract
    • After the discovery of more than 400 planets beyond our Solar System, the characterization of exoplanets as well as their host stars can be considered as one of the fastest growing fields in space science during the past decade. The characterization of exoplanets can only be carried out in a well coordinated interdisciplinary way which connects planetary science, solar/stellar physics and astrophysics. We present a status report on the characterization of exoplanets and their host stars by reviewing the relevant space- and ground-based projects. One finds that the previous strategy changed from space mission concepts which were designed to search, find and characterize Earth-like rocky exoplanets to: A statistical study of planetary objects in order to get information about their abundance, an identification of potential target and finally its analysis. Spectral analysis of exoplanets is mandatory, particularly to identify bio-signatures on Earth-like planets. Direct characterization of exoplanets should be done by spectroscopy, both in the visible and in the infrared spectral range. The way leading to the direct detection and characterization of exoplanets is then paved by several questions, either concerning the pre-required science or the associated observational strategy.

  • Dietary carbohydrate modification alters serum metabolic profiles in individuals with the metabolic syndrome
  • 2010
  • Ingår i: NMCD. Nutrition Metabolism and Cardiovascular Diseases. - : Elsevier. - 0939-4753 .- 1590-3729. ; 20:4, s. 249-57
  • Tidskriftsartikel (refereegranskat)abstract
    • BACKGROUND AND AIMS: Whole-grain cereals and diets with a low glycemic index may protect against the development of type 2 diabetes and heart disease, but the mechanisms are poorly understood. We studied the effect of carbohydrate modification on serum metabolic profiles, including lipids and branched chain amino acids, and dependencies between these and specific gene expression pathways in adipose tissue.METHODS AND RESULTS: Twenty subjects with metabolic syndrome were selected from the larger FUNGENUT study population, randomized either to a diet high in oat and wheat bread and potato (OWP) or rye bread and pasta (RP). Serum metabolomics analyses were performed using ultra-performance liquid chromatography coupled to electrospray ionization mass spectrometry (UPLC/MS), gas chromatography (GC) and UPLC. In the OWP group multiple proinflammatory lysophosphatidylcholines increased, while in the RP group docosahexaenoic acid (DHA 22:6n-3) increased and isoleucine decreased. mRNA expression of stress reactions- and adipose tissue differentiation-related genes were up-regulated in adipose tissue in the OWP group. In the RP group, however, pathways related to stress reactions and insulin signaling and energy metabolism were down-regulated. The lipid profiles had the strongest association with the changes in the adipose tissue differentiation pathway when using the elastic net regression model of the lipidomic profiles on selected pathways.CONCLUSION: Our results suggest that the dietary carbohydrate modification alters the serum metabolic profile, especially in lysoPC species, and may, thus, contribute to proinflammatory processes which in turn promote adverse changes in insulin and glucose metabolism.

  • High-performance glass-fibre reinforced biobased aromatic polyamide in automotive biofuel supply systems
  • 2020
  • Ingår i: Journal of Cleaner Production. - : Elsevier. - 0959-6526 .- 1879-1786. ; 263
  • Tidskriftsartikel (refereegranskat)abstract
    • The increase in service temperature and the use of renewable but also more aggressive biofuels such as biodiesel have remarkably raised demands for the durability of polymer components used in automotive fuel systems. The commonly used aliphatic polyamides (PAs) can here be problematic to use in (bio)diesel systems at high temperature in locations such as fuel pipes and fuel filter housings. In the present work, the ageing behaviour of a new class of easily melt-processable glass fibre-reinforced highly-biobased semi-aromatic polyamides (polyphthalamide, PPA) was investigated in very demanding environments (petroleum diesel (B0) and its commercial blend with 20 vol% biodiesel (B20)) at high temperature. The results showed that the PPA matrix had better barrier properties towards both petroleum and bio-based diesel compared to conventional PA12, as revealed by its lower and slower fuel uptake. The sorbed fuels plasticized the composite, decreasing the glass transition temperature (Tg), with B20 having a larger plasticizing effect on PPA than B0. Notably, the mechanical properties of the PPA composite were not negatively affected even after long exposure to both B0 and B20. This was due to the low level of fuel uptake, high Tg and intact interface between the glass fibres and PPA matrix.

  • 2018
  • Ingår i: Polymer degradation and stability. - : Elsevier Ltd. - 0141-3910 .- 1873-2321. ; 156, s. 170-179
  • Tidskriftsartikel (refereegranskat)abstract
    • The long-term performance of a polyamide-12 (PA12)-based (bio)diesel fuel line/pipe with a thin poly(ethylene-co-tetrafluoroethylene) (ETFE) inner layer was investigated in “close to real” and high-temperature isothermal conditions with fuel on the inside and air on the outside of the pipe. The inner carbon-black-containing ETFE layer resisted fuel attack, as revealed by the small fuel uptake, the very low degree of oxidation, and the unchanged electrical conductivity, glass transition and melting behaviour. The properties of the ETFE layer remained the same after exposure to all the fuel types tested (petroleum diesel, biodiesel and a blend of 80% diesel with 20% biodiesel). Because of the presence of the ETFE layer on the inside, the fuel pipe experienced noticeable changes only in the outer PA12 pipe layer through migration of plasticizer, annealing and slight oxidation. The evaporation of plasticizer was found to be diffusion-controlled and it led to an increase in the glass transition temperature of PA12 by 20 °C. This, together with a small annealing-induced increase in crystallinity, resulted in a stiffer and stronger pipe with an increase in the flexural/tensile modulus and strength. The oxidation of PA12 remained at a low level and did not lead to an embrittled pipe during the simulated lifetime of the vehicle. This study reveals that fluoropolymers have a great potential for use as fuel-contacting materials in “demanding” motor vehicle fuel line systems.

  • 2017
  • Ingår i: Polymer degradation and stability. - : Elsevier. - 0141-3910 .- 1873-2321. ; 144, s. 100-109
  • Tidskriftsartikel (refereegranskat)abstract
    • The behaviour of a polyamide (PA)-based multilayer fuel pipe was investigated in “close to real” conditions using specially-designed ageing equipment with a program designed according to known customer driving modes and conditions (a key life test). The pipe was exposed to petroleum diesel and a combination of petroleum diesel and biodiesel. The fuel exposure pattern, as well as the temperature profile, followed a specified scheme in the key life test. It allowed for the investigation and understanding of complex ageing mechanisms, often observed in multi-layer systems with a variation in the running conditions. The mechanisms involved included migration of plasticizer from the innermost PA6 layer of the pipe to the fuel, and from the PA12 outer layer to the ambient air. At the same time, fuel was absorbed in the inner layer of the pipe. The oxidation of the innermost PA6 layer was promoted by the oxidation products of biodiesel. The diffusion-limited oxidation of the PA6 layer led to the formation of a 30 μm highly oxidized zone at the inner surface of the pipe, resulting in discoloration and oxidative crosslinking of the polymer. The toughness and extensibility of the pipe decreased significantly after prolonged ageing, and the extensibility was only 7% of that of the unaged pipe after 2230 h.

  • 2022
  • Ingår i: Experimental astronomy. - : Springer Nature. - 0922-6435 .- 1572-9508.
  • Tidskriftsartikel (refereegranskat)abstract
    • In the White Paper, submitted in response to the European Space Agency (ESA) Voyage 2050 Call, we present the importance of advancing our knowledge of plasma-neutral gas interactions, and of deepening our understanding of the partially ionized environments that are ubiquitous in the upper atmospheres of planets and moons, and elsewhere in space. In future space missions, the above task requires addressing the following fundamental questions: (A) How and by how much do plasma-neutral gas interactions influence the re-distribution of externally provided energy to the composing species? (B) How and by how much do plasma-neutral gas interactions contribute toward the growth of heavy complex molecules and biomolecules? Answering these questions is an absolute prerequisite for addressing the long-standing questions of atmospheric escape, the origin of biomolecules, and their role in the evolution of planets, moons, or comets, under the influence of energy sources in the form of electromagnetic and corpuscular radiation, because low-energy ion-neutral cross-sections in space cannot be reproduced quantitatively in laboratories for conditions of satisfying, particularly, (1) low-temperatures, (2) tenuous or strong gradients or layered media, and (3) in low-gravity plasma. Measurements with a minimum core instrument package (< 15 kg) can be used to perform such investigations in many different conditions and should be included in all deep-space missions. These investigations, if specific ranges of background parameters are considered, can also be pursued for Earth, Mars, and Venus.

  • Ingår i: Advances in Space Research. - : Elsevier BV. - 0273-1177 .- 1879-1948. ; 38:4, s. 672-679
  • Tidskriftsartikel (refereegranskat)abstract
    • MEFISTO, together with the companion instrument WPT, are planning the first-ever in situ measurements of the electric field in the magnetosphere of planet Mercury. The instruments have been selected by JAXA for inclusion in the BepiColombo/MMO payload, as part of the Plasma Wave Investigation coordinated by Kyoto University. The magnetosphere of Mercury was discovered by Mariner 10 in 1974 and will be studied further by Messenger starting in 2011. However, neither spacecraft did or will measure the electric field. Electric fields are crucial in the dynamics of a magnetosphere and for the energy and plasma transport between different regions within the magnetosphere as well as between the magnetosphere and the surrounding regions. The MEFISTO instrument will be capable of measuring electric fields from DC to 3 MHz, and will thus also allow diagnostics of waves at all frequencies of relevance to the Hermean magnetosphere. MEFISTO is a double-probe electric field instrument. The double-probe technique has strong heritage and is well proven on missions such as Viking, Polar, and Cluster. For BepiColombo, a newly developed deployment mechanism is planned which reduces the mass by a factor of about 5 compared to conventional mechanisms for 15 in long booms. We describe the basic characteristics of the instrument and briefly discuss the new developments made to tailor the instrument to flight in Mercury orbit.

    • The response of the Earth's magnetosphere to changing solar wind conditions is studied with a 3-D Magnetohydrodynamic (MHD) model. One full year (155 Cluster orbits) of the Earth's magnetosphere is simulated using Grand Unified Magnetosphere Ionosphere Coupling simulation (GUMICS-4) magnetohydrodynamic code. Real solar wind measurements are given to the code as input to create the longest lasting global magnetohydrodynamics simulation to date. The applicability of the results of the simulation depends critically on the input parameters used in the model. Therefore, the validity and the variance of the OMNIWeb data are first investigated thoroughly using Cluster measurement close to the bow shock. The OMNIWeb and the Cluster data were found to correlate very well before the bow shock. The solar wind magnetic field and plasma parameters are not changed significantly from the L-1 Lagrange point to the foreshock; therefore, the OMNIWeb data are appropriate input to the GUMICS-4. The Cluster SC3 footprints are determined by magnetic field mapping from the simulation results and the Tsyganenko (T96) model in order to compare two methods. The determined footprints are in rather good agreement with the T96. However, it was found that the footprints agree better in the Northern Hemisphere than the Southern one during quiet conditions. If the B-y is not zero, the agreement of the GUMICS-4 and T96 footprint is worse in longitude in the Southern Hemisphere. Overall, the study implies that a 3-D MHD model can increase our insight of the response of the magnetosphere to solar wind conditions.

  • Adrenals Contribute to Growth of Castration-Resistant VCaP Prostate Cancer Xenografts
  • 2018
  • Ingår i: American Journal of Pathology. - : Elsevier BV. - 0002-9440. ; 188:12, s. 2890-2901
  • Tidskriftsartikel (refereegranskat)abstract
    • The role of adrenal androgens as drivers for castration-resistant prostate cancer (CRPC) growth in humans is generally accepted; however, the value of preclinical mouse models of CRPC is debatable, because mouse adrenals do not produce steroids activating the androgen receptor. In this study, we confirmed the expression of enzymes essential for de novo synthesis of androgens in mouse adrenals, with high intratissue concentration of progesterone (P4) and moderate levels of androgens, such as androstenedione, testosterone, and dihydrotestosterone, in the adrenal glands of both intact and orchectomized (ORX) mice. ORX alone had no effect on serum P4 concentration, whereas orchectomized and adrenalectomized (ORX + ADX) resulted in a significant decrease in serum P4 and in a further reduction in the low levels of serum androgens (androstenedione, testosterone, and dihydrotestosterone), measured by mass spectrometry. In line with this, the serum prostate-specific antigen and growth of VCaP xenografts in mice after ORX + ADX were markedly reduced compared with ORX alone, and the growth difference was not abolished by a glucocorticoid treatment. Moreover, ORX + ADX altered the androgen-dependent gene expression in the tumors, similar to that recently shown for the enzalutamide treatment. These data indicate that in contrast to the current view, and similar to humans, mouse adrenals synthesize significant amounts of steroids that contribute to the androgen receptor–dependent growth of CRPC. © 2018 American Society for Investigative Pathology

  • High intratumoral dihydrotestosterone is associated with antiandrogen resistance in VCaP prostate cancer xenografts in castrated mice
  • 2022

  • Tidskriftsartikel (refereegranskat)abstract
    • Antiandrogen treatment resistance is a major clinical concern in castration-resistant prostate cancer (CRPC) treatment. Using xenografts of VCaP cells we showed that growth of antiandrogen resistant CRPC tumors were characterized by a higher intratumor dihydrotestosterone (DHT) concentration than that of treatment responsive tumors. Furthermore, the slow tumor growth after adrenalectomy was associated with a low intratumor DHT concentration. Reactivation of androgen signaling in enzalutamide-resistant tumors was further shown by the expression of several androgen-dependent genes. The data indicate that intratumor DHT concentration and expression of several androgen-dependent genes in CRPC lesions is an indication of enzalutamide treatment resistance and an indication of the need for further androgen blockade. The presence of an androgen synthesis, independent of CYP17A1 activity, has been shown to exist in prostate cancer cells, and thus, novel androgen synthesis inhibitors are needed for the treatment of enzalutamide-resistant CRPC tumors that do not respond to abiraterone.

  • Ageing properties of polyamide-12 pipes exposed to fuels with and without ethanol
  • 2008
  • Ingår i: Polymer degradation and stability. - : Elsevier BV. - 0141-3910 .- 1873-2321. ; 93:10, s. 1846-1854
  • Tidskriftsartikel (refereegranskat)abstract
    • The chemical and physical properties of polyamide-12 (PA12) fuel pipes/lines, aged for <= 2400 h at 110 degrees C, have been investigated. The pipes, containing fuel with or without ethanol, were either of a single PA12 layer, or of two PA12 layers surrounding a poly(vinylidene fluoride) barrier layer. The molar mass of the inner surface region obtained by size-exclusion chromatography was reduced during ageing. and optical microscopy revealed a surface that was partly dissolved in the presence of ethanol. Infrared spectroscopy revealed a rapid loss of plasticiser, especially in the presence of ethanol, and the fuel contained plasticiser and other polymer related components. Immersion tests at 60 degrees C showed that the swelling of the pipe and the amount of dissolved material were greatest for the fuels with intermediate ethanol content (50 vol.%). Aged samples experienced an increase in melting point, presumably, to a large extent, due to the loss of plasticiser and/or PA12-related components. In addition, for several samples, the crystallinity seemed to increase with ageing. (C) 2008 Elsevier Ltd. All rights reserved.

  • Effects of ethanol content and temperature on the permeation of fuel through polyamide-12-based pipes
  • 2010
  • Ingår i: Polymer testing. - : elsevier. - 0142-9418 .- 1873-2348. ; 29:5, s. 603-608
  • Tidskriftsartikel (refereegranskat)abstract
    • A new test method has been developed which enables the fuel permeation properties of polyamide-12 (PA-12) - based pipes to be investigated. Fuel lines were exposed to circulating fuel with equal volume contents of toluene, isooctane and 0, 25 or 85 vol.% ethanol for up to 6 months at 60-110 degrees C. The pipes were either of a single PA12-layer or a multi-layer type, the latter having a poly(vinylidene fluoride) barrier layer between two PA12 sections. With a Thwing-Albert cup attached to the fuel line, it was possible to expose a pipe-section to surrounding air running in a separate pipe loop at a controlled flow-rate. Gas/vapour samples were collected from this loop using a syringe and, subsequently, analysed with a flame ionization detector. It was observed in the case of multi-layer pipes that the presence of ethanol increased the permeability (average values) of the "total" fuel as well as of the individual hydrocarbons. In addition, the 60 degrees C fuel permeability (85 vol.% ethanol) increased after a high temperature (110 degrees C peak) cycle, whereas the ethanol-free fuel flux seemed to decrease.

  • Sensitivity study of fluid dynamic effects on nitric oxide formation in CFB combustion of wood
  • 2002
  • Ingår i: Proc. of the 7th International Circulating Fluidized Bed. - 0920804985 ; , s. 757-764
  • Bokkapitel (övrigt vetenskapligt/konstnärligt)abstract
    • The paper presents results from simulations by a 1.5D numerical model developed to study the formation of the NO and N2O emissions in a circulating fluidized bed combustor (CFBC) under different operating conditions and burning different fuels. A comprehensive kinetic scheme for the homogeneous chemistry and a single particle model for char combustion are used. Fluiddynamic factors, including gas mixing and release of volatiles, are investigated in the case of wood combustion under normal air staging conditions. The pattern of release of volatiles, the mixing of secondary air, and the lateral mixing of gas are observed to play significant roles in the formation of the relatively high NO emissions from combustion of wood. Comparisons are made with measurement data and also with coal combustion.

  • The MEFISTO and WPT Electric Field Sensors of the Plasma Wave Investigation on the BepiColombo Mio Spacecraft Measurements of Low and High Frequency Electric Fields at Mercury
  • 2020
  • Ingår i: Space Science Reviews. - : Springer Nature. - 0038-6308 .- 1572-9672. ; 216:8
  • Forskningsöversikt (refereegranskat)abstract
    • This paper describes the design of MEFISTO (Mercury Electric Field In-Situ Tool) and WPT (Wire Probe Antenna) electric field sensors for Plasma Wave Investigation (PWI) on the BepiColombo Mio spacecraft (Mercury Magnetospheric Orbiter, MMO). The two sensors will enable the first observations of electric fields, plasma waves and radio waves in and around the Hermean magnetosphere and exosphere. MEFISTO and WPT are dipole antennas with 31.6 m tip-to-tip length. Each antenna element has a spherical probe at each end of the wire (15 m length). They are extended orthogonally in the spin plane of the spacecraft and enable measurements of the electric field in the frequency range of DC to 10 MHz by the connection to two sets of receivers, EWO for a lower frequency range and SORBET for higher frequencies. In the initial operations after the launch (20 Oct. 2018), we succeeded to confirm the health of both antennas and to release the launch lock of the WPT. After Mercury orbit insertion planned at the end of 2025, both sensors will be fully deployed and activate full operations of the PWI electric field measurements.

  • Ingår i: BMJ open. - : BMJ. - 2044-6055. ; 10:10, s. e039574-
  • Tidskriftsartikel (refereegranskat)abstract
    • The Finnish National Esophago-Gastric Cancer Cohort (FINEGO) was established with the aim of identifying factors that could contribute to improved outcomes in oesophago-gastric cancer. The aim of this study is to describe the patients with gastric cancer included in FINEGO.ParticipantsA total of 10 457 patients with gastric cancer or tumour diagnosis in the Finnish Cancer Registry or the Finnish Patient Registry during 1987–2016 were included in the cohort, with follow-up from Causes of Death Registry until 31 December 2016. All of the participants were at least 18 years of age, and had undergone either resectional or endoscopic mucosal surgery with curative or palliative intent.Findings to dateOf the 10 457 patients, 90.1% were identified to have cancer in both cancer and patient registries. In all, the median age was 70 at the time of surgery, 54.5% of the patients were men and 64.4% had no comorbidities. Education data were available for 31.1% of the patients, of whom the majority had had <12 years of formal education. Of the 7798 with cancer staging data available, 41.1% had a local cancer. Adenocarcinoma was the most common (94.2%) histological type. Almost all patients underwent open gastrectomy and 214% in hospitals with annual volume of more than 30 gastrectomies per year. A total of 8561 deaths occurred during the study period, of which 6474 were due to oesophago-gastric cancers. The 5-year survival was 34.6% and 5-year cancer-specific survival was 39.7%.Future plansThe data in FINEGO can be currently used for registry-based research but is being expanded by data extraction from patient records and scanning of histological samples from the Finnish biobanks. Initially, we are planning on studies on the national trends in treatment and mortality, and studies on the demographic factors and their influence on survival.

  • Towards a quantitative understanding of NOx and N2O emission formation in full-scale circulating fluidised bed combustors
  • 2001
  • Ingår i: In Proceedings of the 16th International Conference on Fluidized Bed Combustion--FBC01, held in Reno, Nevada, USA, May 13-16, 2001.
  • Konferensbidrag (refereegranskat)abstract
    • A mathematical tool is being developed for studying the nitrogen oxide emission formation in circulating fluidised bed combustors. The model is based on detailed homogeneous and heterogeneous chemical kinetics and a simplified, reasonable description of CFB hydrodynamics with presumed temperature distribution (Kilpinen et al, 1999a). With the model different fuels and fuel mixtures can be compared in regard to their nitrogen oxide emission formation tendency at typical CFBC conditions. In this paper the structure of the CFBC model and its submodels are shortly described in present form. The CFBC model is tested for nitrogen oxide prediction at normal air staging conditions in a 12 MW CFB with bituminous coal and wood chips as the fuel, respectively. Comparisons of modelling results with detailed gas concentration profiles measured inside the furnace are made. The relative importance of homogeneous and heterogeneous reactions on NO and N2O concentration profiles is illustrated based on a quantitative reaction rate analysis at different parts in the combustor. The importance of effects of radical removal on particle surfaces, and thus, a decreased CO burnout and, simultaneously, enhanced rates of catalytic bed/char reactions on nitrogen oxides’ destruction are discussed.
