    • Very few genetic variants have been associated with depression and neuroticism, likely because of limitations on sample size in previous studies. Subjective well-being, a phenotype that is genetically correlated with both of these traits, has not yet been studied with genome-wide data. We conducted genome-wide association studies of three phenotypes: subjective well-being (n = 298,420), depressive symptoms (n = 161,460), and neuroticism (n = 170,911). We identify 3 variants associated with subjective well-being, 2 variants associated with depressive symptoms, and 11 variants associated with neuroticism, including 2 inversion polymorphisms. The two loci associated with depressive symptoms replicate in an independent depression sample. Joint analyses that exploit the high genetic correlations between the phenotypes (vertical bar(p) over cap vertical bar approximate to 0.8) strengthen the overall credibility of the findings and allow us to identify additional variants. Across our phenotypes, loci regulating expression in central nervous system and adrenal or pancreas tissues are strongly enriched for association.

  • Heterozygosity for a Loss-of-Function Mutation in GALNT2 Improves Plasma Triglyceride Clearance in Man

    • Genome-wide association studies have identified GALNT2 as a candidate gene in lipid metabolism, but it is not known how the encoded enzyme ppGalNAc-T2, which contributes to the initiation of mucin-type O-linked glycosylation, mediates this effect. In two probands with elevated plasma high-density lipoprotein cholesterol and reduced triglycerides, we identified a mutation in GALNT2. It is shown that carriers have improved postprandial triglyceride clearance, which is likely attributable to attenuated glycosylation of apolipoprotein (apo) C-III, as observed in their plasma. This protein inhibits lipoprotein lipase (LPL), which hydrolyses plasma triglycerides. We show that an apoC-III-based peptide is a substrate for ppGalNAc-T2 while its glycosylation by the mutant enzyme is impaired. In addition, neuraminidase treatment of apoC-III which removes the sialic acids from its glycan chain decreases its potential to inhibit LPL. Combined, these data suggest that ppGalNAc-T2 can affect lipid metabolism through apoC-III glycosylation, thereby establishing GALNT2 as a lipid-modifying gene.

  • Sound and vibration : effects on infants' heart rate and heart rate variability during neonatal transport

    • Aim: To measure the effect of sound and whole-body vibration on infants' heart rate and heart rate variability during ground and air ambulance transport.Methods: Sixteen infants were transported by air ambulance with ground ambulance transport to and from the airports. Whole-body vibration and sound levels were recorded and heart parameters were obtained by ECG signal.Results: Sound and whole-body vibration levels exceeded the recommended limits. Mean whole-body vibration and sound levels were 0.19m/s(2) and 73dBA, respectively. Higher whole-body vibration was associated with a lower heart rate (p<0.05), and higher sound level was linked to a higher heart rate (p=0.05). The heart rate variability was significantly higher at the end of the transport than at the beginning (p<0.01). Poorer physiologic status was associated with lower heart rate variability (p<0.001) and a lower heart rate (p<0.01). Infants wearing earmuffs had a lower heart rate (p<0.05).Conclusions: Sound and whole-body vibration during neonatal transport exceed recommended levels for adults and sound seem to have a more stressful effect on the infant than vibrations. Infants should wear earmuffs during neonatal transport because of the stress reducing effect.

  • Discovery of Species-unique Peptide Biomarkers of Bacterial Pathogens by Tandem Mass Spectrometry-based Proteotyping

    • Mass spectrometry (MS) and proteomics offer comprehensive characterization and identification of microorganisms and discovery of protein biomarkers that are applicable for diagnostics of infectious diseases. The use of biomarkers for diagnostics is widely applied in the clinic and the use of peptide biomarkers is increasingly being investigated for applications in the clinical laboratory. Respiratory-tract infections are a predominant cause for medical treatment, although, clinical assessments and standard clinical laboratory protocols are time-consuming and often inadequate for reliable diagnoses. Novel methods, preferably applied directly to clinical samples, excluding cultivation steps, are needed to improve diagnostics of infectious diseases, provide adequate treatment and reduce the use of antibiotics and associated development of antibiotic resistance. This study applied nano-liquid chromatography (LC) coupled with tandem MS, with a bioinformatics pipeline and an in-house database of curated high-quality reference genome sequences to identify species-unique peptides as potential biomarkers for four bacterial pathogens commonly found in respiratory tract infections (RTIs): Staphylococcus aureus; Moraxella catarrhalis; Haemophilus influenzae and Streptococcus pneumoniae. The species-unique peptides were initially identified in pure cultures of bacterial reference strains, reflecting the genomic variation in the four species and, furthermore, in clinical respiratory tract samples, without prior cultivation, elucidating proteins expressed in clinical conditions of infection. For each of the four bacterial pathogens, the peptide biomarker candidates most predominantly found in clinical samples, are presented. Data are available via ProteomeXchange with identifier PXD014522. As proof-of-principle, the most promising species-unique peptides were applied in targeted tandem MS-analyses of clinical samples and their relevance for identifications of the pathogens, i.e. proteotyping, was validated, thus demonstrating their potential as peptide biomarker candidates for diagnostics of infectious diseases.

    • Bloodstream infections (BSIs), the presence of microorganisms in blood, are potentially serious conditions that can quickly develop into sepsis and life-threatening situations. When assessing proper treatment, rapid diagnosis is the key; besides clinical judgement performed by attending physicians, supporting microbiological tests typically are performed, often requiring microbial isolation and culturing steps, which increases the time required for confirming positive cases of BSI. The additional waiting time forces physicians to prescribe broad-spectrum antibiotics and empirically based treatments, before determining the precise cause of the disease. Thus, alternative and more rapid cultivation-independent methods are needed to improve clinical diagnostics, supporting prompt and accurate treatment and reducing the development of antibiotic resistance. In this study, a culture-independent workflow for pathogen detection and identification in blood samples was developed, using peptide biomarkers and applying bottom-up proteomics analyses, i.e., so-called "proteotyping". To demonstrate the feasibility of detection of blood infectious pathogens, using proteotyping, Escherichia coli and Staphylococcus aureus were included in the study, as the most prominent bacterial causes of bacteremia and sepsis, as well as Candida albicans, one of the most prominent causes of fungemia. Model systems including spiked negative blood samples, as well as positive blood cultures, without further culturing steps, were investigated. Furthermore, an experiment designed to determine the incubation time needed for correct identification of the infectious pathogens in blood cultures was performed. The results for the spiked negative blood samples showed that proteotyping was 100- to 1,000-fold more sensitive, in comparison with the MALDI-TOF MS-based approach. Furthermore, in the analyses of ten positive blood cultures each of E. coli and S. aureus, both the MALDI-TOF MS-based and proteotyping approaches were successful in the identification of E. coli, although only proteotyping could identify S. aureus correctly in all samples. Compared with the MALDI-TOF MS-based approaches, shotgun proteotyping demonstrated higher sensitivity and accuracy, and required significantly shorter incubation time before detection and identification of the correct pathogen could be accomplished.

    • The Thrombolysis in Ischemic Stroke Patients (TRISP) collaboration was a concerted effort initiated in 2010 with the purpose to address relevant research questions about the effectiveness and safety of intravenous thrombolysis (IVT). The collaboration also aims to prospectively collect data on patients undergoing endovascular treatment (EVT) and hence the name of the collaboration was changed from TRISP to EVA-TRISP. The methodology of the former TRISP registry for patients treated with IVT has already been published. This paper focuses on describing the EVT part of the registry.All centres committed to collecting predefined variables on consecutive patients prospectively. We aim for accuracy and completeness of the data and to adapt local databases to investigate novel research questions. Herein, we introduce the methodology of a recently constructed academic investigator-initiated open collaboration EVT registry built as an extension of an existing IVT registry in patients with acute ischaemic stroke (AIS).Currently, the EVA-TRISP network includes 20 stroke centres with considerable expertise in EVT and maintenance of high-quality hospital-based registries. Following several successful randomised controlled trials (RCTs), many important clinical questions remain unanswered in the (EVT) field and some of them will unlikely be investigated in future RCTs. Prospective registries with high-quality data on EVT-treated patients may help answering some of these unanswered issues, especially on safety and efficacy of EVT in specific patient subgroups.This collaborative effort aims at addressing clinically important questions on safety and efficacy of EVT in conditions not covered by RCTs. The TRISP registry generated substantial novel data supporting stroke physicians in their daily decision making considering IVT candidate patients. While providing observational data on EVT in daily clinical practice, our future findings may likewise be hypothesis generating for future research as well as for quality improvement (on EVT). The collaboration welcomes participation of further centres willing to fulfill the commitment and the outlined requirements.

  • Genome-wide association study identifies 74 loci associated with educational attainment

    • Educational attainment is strongly influenced by social and other environmental factors, but genetic factors are estimated to account for at least 20% of the variation across individuals. Here we report the results of a genome-wide association study (GWAS) for educational attainment that extends our earlier discovery sample of 101,069 individuals to 293,723 individuals, and a replication study in an independent sample of 111,349 individuals from the UK Biobank. We identify 74 genome-wide significant loci associated with the number of years of schooling completed. Single-nucleotide polymorphisms associated with educational attainment are disproportionately found in genomic regions regulating gene expression in the fetal brain. Candidate genes are preferentially expressed in neural tissue, especially during the prenatal period, and enriched for biological pathways involved in neural development. Our findings demonstrate that, even for a behavioural phenotype that is mostly environmentally determined, a well-powered GWAS identifies replicable associated genetic variants that suggest biologically relevant pathways. Because educational attainment is measured in large numbers of individuals, it will continue to be useful as a proxy phenotype in efforts to characterize the genetic influences of related phenotypes, including cognition and neuropsychiatric diseases.

    • Lightwave communications is a necessity for the information age. Optical links provide enormous bandwidth, and the optical fiber is the only medium that can meet the modern society's needs for transporting massive amounts of data over long distances. Applications range from global high-capacity networks, which constitute the backbone of the internet, to the massively parallel interconnects that provide data connectivity inside datacenters and supercomputers. Optical communications is a diverse and rapidly changing field, where experts in photonics, communications, electronics, and signal processing work side by side to meet the ever-increasing demands for higher capacity, lower cost, and lower energy consumption, while adapting the system design to novel services and technologies. Due to the interdisciplinary nature of this rich research field, Journal of Optics has invited 16 researchers, each a world-leading expert in their respective subfields, to contribute a section to this invited review article, summarizing their views on state-of-the-art and future developments in optical communications.

    • Background: Prokaryotes dominate the biosphere and regulate biogeochemical processes essential to all life. Yet, our knowledge about their biology is for the most part limited to the minority that has been successfully cultured. Molecular techniques now allow for obtaining genome sequences of uncultivated prokaryotic taxa, facilitating in-depth analyses that may ultimately improve our understanding of these key organisms. Results: We compared results from two culture-independent strategies for recovering bacterial genomes: single-amplified genomes and metagenome-assembled genomes. Single-amplified genomes were obtained from samples collected at an offshore station in the Baltic Sea Proper and compared to previously obtained metagenome-assembled genomes from a time series at the same station. Among 16 single-amplified genomes analyzed, seven were found to match metagenome-assembled genomes, affiliated with a diverse set of taxa. Notably, genome pairs between the two approaches were nearly identical (average 99.51% sequence identity; range 98.77-99.84%) across overlapping regions (30-80% of each genome). Within matching pairs, the single-amplified genomes were consistently smaller and less complete, whereas the genetic functional profiles were maintained. For the metagenome-assembled genomes, only on average 3.6% of the bases were estimated to be missing from the genomes due to wrongly binned contigs. Conclusions: The strong agreement between the single-amplified and metagenome-assembled genomes emphasizes that both methods generate accurate genome information from uncultivated bacteria. Importantly, this implies that the research questions and the available resources are allowed to determine the selection of genomics approach for microbiome studies.

    • Double photoionization spectra of the CS2 molecule have been recorded using the TOF-PEPECO technique in combination with synchrotron radiation at the photon energies h nu=220, 230, 240, 243, and 362.7 eV. The spectra were recorded in the S 2p and C 1s inner-shell ionization regions and reflect dicationic states formed out of one inner-shell vacancy and one vacancy in the valence region. MCSCF calculations were performed to model the energies of the dicationic states. The spectra associated with a S 2p vacancy are well structured and have been interpreted in some detail by comparison to conventional S 2p and valence photoelectron spectra. The lowest inner-shell-valence dicationic state is observed at the vertical double ionization energy 188.45 eV and is associated with a (2p(3/2))(-1)(2 pi(g))(-1) double vacancy. The spectrum connected to the C 1s vacancy shows a distinct line at 310.8 eV, accompanied by additional broad features at higher double ionization energies.     • This study focuses on the radiative properties of five subtropical marine stratocumulus cloud regions, on monthly mean scale. Through examination of the relation between total albedo and cloud fraction, and its variability and relation to other parameters, some of the factors controlling the reflectivity, or albedo, of the clouds in these regions are investigated. It is found that the main part of the variability in albedo at a given cloud fraction can be related to temporal, rather than spatial variability, indicating spatial homogeneity in cloud radiative properties in the studied regions. This is seen most clearly in satellite observations, but also in an ensemble of climate models. Further comparison between satellite data and output from climate models shows that there is good agreement with respect to the role of liquid water path, the parameter that can be assumed to be the primary source of variability in cloud reflectivity for a given cloud fraction. On the other hand, the influence of aerosol loading on cloud albedo differs between models and observations. The cloud-albedo effect, or cloud brightening caused by aerosol through its coupling to cloud droplet number concentration and droplet size, is found not to dominate in the satellite observations on monthly mean scale, as it appears to do on this scale in the climate models. The disagreement between models and observations is particularly strong in regions with frequent occurrence of absorbing aerosols above clouds, where satellite data contrary to the climate models indicate a scene darkening with increasing aerosol loading.

    • BACKGROUND: Clinical studies with work participation (WP) as an outcome domain pose particular methodological challenges that hamper interpretation, comparison between studies and meta-analyses.OBJECTIVES: To develop Points to Consider (PtC) for design, analysis and reporting of studies of patients with inflammatory arthritis that include WP as a primary or secondary outcome domain.METHODS: The EULAR Standardised Operating Procedures were followed. A multidisciplinary taskforce with 22 experts including patients with rheumatic diseases, from 10 EULAR countries and Canada, identified methodologic areas of concern. Two systematic literature reviews (SLR) appraised the methodology across these areas. In parallel, two surveys among professional societies and experts outside the taskforce sought for additional methodological areas or existing conducting/reporting recommendations. The taskforce formulated the PtC after presentation of the SLRs and survey results, and discussion. Consensus was obtained through informal voting, with levels of agreement obtained anonymously.RESULTS: Two overarching principles and nine PtC were formulated. The taskforce recommends to align the work-related study objective to the design, duration, and outcome domains/measurement instruments of the study (PtC: 1-3); to identify contextual factors upfront and account for them in analyses (PtC: 4); to account for interdependence of different work outcome domains and for changes in work status over time (PtC: 5-7); to present results as means as well as proportions of patients reaching predefined meaningful categories (PtC: 8) and to explicitly report volumes of productivity loss when costs are an outcome (PtC:9).CONCLUSION: Adherence to these EULAR PtC will improve the methodological quality of studies evaluating WP.

    • Whether climate change will turn cold biomes from large long-term carbon sinks into sources is hotly debated because of the great potential for ecosystem-mediated feedbacks to global climate. Critical are the direction, magnitude and generality of climate responses of plant litter decomposition. Here, we present the first quantitative analysis of the major climate-change-related drivers of litter decomposition rates in cold northern biomes worldwide.Leaf litters collected from the predominant species in 33 global change manipulation experiments in circum-arctic-alpine ecosystems were incubated simultaneously in two contrasting arctic life zones. We demonstrate that longer-term, large-scale changes to leaf litter decomposition will be driven primarily by both direct warming effects and concomitant shifts in plant growth form composition, with a much smaller role for changes in litter quality within species. Specifically, the ongoing warming-induced expansion of shrubs with recalcitrant leaf litter across cold biomes would constitute a negative feedback to global warming. Depending on the strength of other (previously reported) positive feedbacks of shrub expansion on soil carbon turnover, this may partly counteract direct warming enhancement of litter decomposition.

    • Sex hormone-binding globulin (SHBG) is a glycoprotein responsible for the transport and biologic availability of sex steroid hormones, primarily testosterone and estradiol. SHBG has been associated with chronic diseases including type 2 diabetes (T2D) and with hormone-sensitive cancers such as breast and prostate cancer. We performed a genome-wide association study (GWAS) meta-analysis of 21,791 individuals from 10 epidemiologic studies and validated these findings in 7,046 individuals in an additional six studies. We identified twelve genomic regions (SNPs) associated with circulating SHBG concentrations. Loci near the identified SNPs included SHBG (rs12150660, 17p13.1, p=1.8×10(-106)), PRMT6 (rs17496332, 1p13.3, p=1.4×10(-11)), GCKR (rs780093, 2p23.3, p=2.2×10(-16)), ZBTB10 (rs440837, 8q21.13, p=3.4×10(-09)), JMJD1C (rs7910927, 10q21.3, p=6.1×10(-35)), SLCO1B1 (rs4149056, 12p12.1, p=1.9×10(-08)), NR2F2 (rs8023580, 15q26.2, p=8.3×10(-12)), ZNF652 (rs2411984, 17q21.32, p=3.5×10(-14)), TDGF3 (rs1573036, Xq22.3, p=4.1×10(-14)), LHCGR (rs10454142, 2p16.3, p=1.3×10(-07)), BAIAP2L1 (rs3779195, 7q21.3, p=2.7×10(-08)), and UGT2B15 (rs293428, 4q13.2, p=5.5×10(-06)). These genes encompass multiple biologic pathways, including hepatic function, lipid metabolism, carbohydrate metabolism and T2D, androgen and estrogen receptor function, epigenetic effects, and the biology of sex steroid hormone-responsive cancers including breast and prostate cancer. We found evidence of sex-differentiated genetic influences on SHBG. In a sex-specific GWAS, the loci 4q13.2-UGT2B15 was significant in men only (men p=2.5×10(-08), women p=0.66, heterogeneity p=0.003). Additionally, three loci showed strong sex-differentiated effects: 17p13.1-SHBG and Xq22.3-TDGF3 were stronger in men, whereas 8q21.12-ZBTB10 was stronger in women. Conditional analyses identified additional signals at the SHBG gene that together almost double the proportion of variance explained at the locus. Using an independent study of 1,129 individuals, all SNPs identified in the overall or sex-differentiated or conditional analyses explained ∼15.6% and ∼8.4% of the genetic variation of SHBG concentrations in men and women, respectively. The evidence for sex-differentiated effects and allelic heterogeneity highlight the importance of considering these features when estimating complex trait variance.

    • In the building sector, net-zero energy performance targets and reduction of CO2 emissions are the main drivers for building integrated photovoltaics. Recent market studies [10,18,19] show the history and perspective of the global market in the BIPV sector and calculate a global compound annual growth rate of ~ 40 % from 2009 up to 2020.In Europe, the BIPV market is in a transition. The past decades of a slowly emerging BIPV market have been characterized by the original dominant value of BIPV: a building that communicates an image of sustainability and innovation. Payback time or "return on investment" have not been the major parameters in the decision process for applying BIPV. This is changing now. In most European countries, the new regulations on energy performance in buildings (derived from the European Energy Performance of Buildings Directive and the Directive on energy efficiency) have been translated in national regulations/laws, stating that all new (and freshly refurbished) buildings of the EU member states should be nearly zero energy buildings (nZEBs) by 2020. Thus, the time for the regulations to become mandatory is very near. It is expected that the energy performance regulations are now taking over as the main driving factor for the BIPV market and that should have huge consequences in removing the difference between a conventional building component and a BIPV component both in aesthetical and construction terms. This has led to intensified research and development aiming to create BIPV products that come in a variety of colours and sizes, while at the same time being as close as possible to existing building components, to its requirements and how it is considered by the construction industry.The given market overview of state-of-the-art coloured BIPV products clearly reveals that for all parts of a BIPV module (glass, polymers, PV-active layers), there are technical solutions available for colouring. Pilot projects utilizing coloured BIPV products have been built in numerous (mainly European) cities clearly demonstrating the maturity of these solutions.The key aspect that has been slowing down the progress of implementation of these aesthetically appealing and technically sophisticated new products into a wider market are the costs. In this respect, a lot of effort has been made to improve and optimize the relationship between colour and efficiency/power generation of BIPV elements. The results of the past and on-going innovative research and intensified fundamental considerations are also summarized in this report.Besides the colour perception of the coloured BIPV elements under solar irradiation which is essential for the acceptance of the exterior appearance of a building, also transparency and inside visual comfort of BIPV – windows and façade elements are essential for the users and inhabitants.

    • Mitochondrial complex I (CI) deficiency is the most prevalent defect in the respiratory chain in paediatric mitochondrial disease. This heterogeneous group of diseases includes serious or fatal neurological presentations such as Leigh syndrome and there are very limited evidence-based treatment options available. Here we describe that cell membrane-permeable prodrugs of the complex II substrate succinate increase ATP-linked mitochondrial respiration in CI-deficient human blood cells, fibroblasts and heart fibres. Lactate accumulation in platelets due to rotenone-induced CI inhibition is reversed and rotenone-induced increase in lactate:pyruvate ratio in white blood cells is alleviated. Metabolomic analyses demonstrate delivery and metabolism of [ 13 C]succinate. In Leigh syndrome patient fibroblasts, with a recessive NDUFS2 mutation, respiration and spare respiratory capacity are increased by prodrug administration. We conclude that prodrug-delivered succinate bypasses CI and supports electron transport, membrane potential and ATP production. This strategy offers a potential future therapy for metabolic decompensation due to mitochondrial CI dysfunction.

    • Mitochondrial dysfunction is considered a hallmark of aging. Up to now, a gradual decline of mitochondrial respiration with advancing age has mainly been demonstrated in human muscle tissue. A handful of studies have examined age-related mitochondrial dysfunction in human blood cells, and only with small sample sizes and mainly in platelets. In this study, we analyzed mitochondrial respiration in peripheral blood mononuclear cells (PBMCs) and platelets from 308 individuals across the human lifespan (0–86 years). In regression analyses, with adjustment for false discovery rate (FDR), we found age-related changes in respiratory measurements to be either small or absent. The main significant changes were an age-related relative decline in complex I-linked respiration and a corresponding rise of complex II-linked respiration in PBMCs. These results add to the understanding of mitochondrial dysfunction in aging and to its possible role in immune cell and platelet senescence.

  • Predictors of outcome in children with disorders of mitochondrial metabolism in the pediatric intensive care unit

    • Background: The aim of this study was to identify factors predicting outcome in patients with mitochondrial disease admitted to pediatric intensive care units (PICU). Methods: Retrospective study of 2434 patients (age <21 years) admitted to a PICU from 1 January 2006 through 31 March 2016 and captured in the Virtual Pediatric Systems database with ICD9 diagnosis 277.87, disorders of mitochondrial metabolism. Factors influencing mortality and prolonged length of stay (≥14 days) were analyzed using logistic regression. Results: Predictors independently affecting mortality (adjusted odds ratios and 95% confidence intervals, p < 0.05): age 1–23 months 3.4 (1.7–6.6) and mechanical ventilation 4.7 (2.6–8.6) were risk factors; post-operative 0.2 (0.1–0.6), readmission 0.5 (0.3–0.9), and neurologic reason for admittance 0.3 (0.1–0.9) were factors reducing risk. Predictors affecting prolonged length of stay: mechanical ventilation 7.4 (5.2–10.3) and infectious reason for admittance 2.0 (1.3–3.2) were risk factors, post-operative patients 0.3 (0.2–0.5) had lower risk. The utility of PRISM and PIM2 scores in this patient group was evaluated. Conclusions: The single most predictive factor for both mortality and prolonged length of stay is the presence of mechanical ventilation. Age 1–23 months is a risk factor for mortality, and infectious reason for admittance indicates risk for prolonged length of stay. Impact: Presence of mechanical ventilation is the factor most strongly associated with negative outcome in patients with mitochondrial disease in pediatric intensive care.Age 1–23 months is a risk factor for mortality, and infectious reason for admittance indicates risk for prolonged length of stayPRISM3 and PIM2 are not as accurate in patients with mitochondrial disease as in a mixed patient population.

    • In Norway, Sweden and Finland approximately 9.000 reindeer are killed every year in traffic accidents (road and railway). In field experiments two different electronically sensor system were used. The purpose is to warn car drivers and railway engineers that reindeer are close to, or on the traffic system, and reduce the numbers of accident. The information system developed by Telespor is based on GPS/GSM radio collars. These collars use SMS to inform that the reindeer are close to the traffic system. In addition to the warning, the reindeer owners get information about the reindeer land use throughout the whole year. The collars are expensive (approx. 250 euros/unit) and need battery change every year. At Umeå University a system based on wireless sensor nodes attached to the reindeer collars is developed. This node will send out a periodic warning radio beacon. On road sticks a warning device will be attached containing a radio receiver and LED flashlights that will warn if a radio beacon is received. To further increase the warning distance it is possible to use communication between the roadside warning devices and let several devices flash their LED lights when a reindeer is detected. The radio collar (approx. 15 euros/unit) will have a theoretical battery life of more than 10 years. The radio range is around 500 meters between the radio collar and the roadside warning device, which is more than sufficient.

To access these fragments, the user uses her iPhone and the Argon web AR application (created by the Augmented Environments Lab at Georgia Institute of Technology with whom we have collaborated).

    • The radiative properties of subtropical marine stratocumulus clouds are investigated in an ensemble of current-generation global climate models from phase 5 of the Climate Model Intercomparison Project (CMIP5). Using a previously documented method for determining regional mean cloud albedo, the authors find a closer agreement with observations in the CMIP5 models as compared to the previous generation of models (phase 3 of CMIP). The multimodel average indicates regional mean, monthly mean cloud albedos ranging from 0.32 to 0.5 among 26 models and five regions, to be compared with satellite observations that indicate a range from 0.32 to 0.39 for the same five regions. The intermodel spread in cloud fraction gives rise to a spread in albedo. Within models, there is a tendency for large cloud fraction to be related to low cloud albedo and vice versa, a relationship that dampens the intermodel variability in total albedo. The intramodel variability in albedo, for a given cloud fraction, is found to be up to twice as large in magnitude in models as in satellite observations. The reason for this larger variability in models is not settled, but possible contributing factors may be imperfect representation in the models of cloud type distribution or of sensitivity to meteorological variability or aerosols. Changes in aerosol loading are found to be the likely cause of an increase in cloud albedo over time. The radiative effect of such a scene brightening in marine stratocumulus cloud regions, from preindustrial times to present day, is estimated to be up to -1W m(-2) for the global ocean, but there are no observations to verify this number.

  • The Importance of Representing Mixed-Phase Clouds for Simulating Distinctive Atmospheric States in the Arctic

    • Observations from the Surface Heat Budget of the Arctic Ocean experiment (SHEBA) suggest that the Arctic Basin is characterized by two distinctly different preferred atmospheric states during wintertime. These states appear as two peaks in the frequency distribution of surface downwelling longwave radiation (LWD), representing radiatively clear and opaque conditions. Here, the authors have investigated the occurrence and representation of these states in the widely used ECMWF Interim Re-Analysis (ERA-Interim) dataset. An interannually recurring bimodal distribution of LWD values is not a clearly observable feature in the reanalysis data. However, large differences in the simulated liquid water content of clouds in ERA-Interim compared to observations are identified and these are linked to the lack of a radiatively opaque peak in the reanalysis. Using a single-column model, dynamically controlled by data from ERA-Interim, the authors show that, by tuning the glaciation speed of supercooled liquid clouds, it is possible to reach a very good agreement between the model and observations from the SHEBA campaign in terms of LWD. The results suggest that the presence of two preferred Arctic states, as observed during SHEBA, is a recurring feature of the Arctic climate system during winter [December–March (DJFM)]. The mean increase in LWD during the Arctic winter compared to ERA-Interim is 15 W m−2. This has a substantial bearing on climate model evaluation in the Arctic as it indicates the importance of representing Arctic states in climate models and reanalysis data and that doing so could have a significant impact on winter ice thickness and surface temperatures in the Arctic.

    • Semiconducting boron doped single-crystal CVD diamond has been patterned using aluminum masks and an inductively coupled plasma (ICP) etch system. For comparison insulating HPHT diamond samples were also patterned using the same process. Diamond etch rates above 200 nm/min were obtained with an O2/Ar discharge for a gas pressure of 2.5 mTorr using 600 W RF power. We have accomplished the fabrication of structures with a minimum feature size of 1 μm with vertical sidewalls in both CVD and HPHT diamond. The ICP etching produced smooth surfaces with a typical root-mean-square surface roughness of 3 nm. The dependence of etch rate on bias voltage was somewhat different for the two types of diamond. However, for all samples both the etch rate and anisotropy were found to improve with increasing bias voltage.

    • In the current contribution we will present the latest developments in the project “Modelling crash behaviour in future lightweight composite vehicles – Step 2”, involving 11 Swedish partners. On the material modelling side, a fully three-dimensional model to describe fibre kinking has recently been developed. The model is physically based and considers the fibre rotation during kink-band formation under large deformations. The FE implementation of the model is straightforward which allows for easy implementation. The validation of the model for stiffness and strength shows good correlation with the experiments. The influence of initial misalignments on the stiffness is well captured, the strength defined at the onset of unstable fibre rotation, is well predicted, and, in addition, the crushing response shows very good agreement with experimental results in terms of morphology in the crushing zone, as well as in the load response. To allow for computational efficiency, we have also developed and implemented (as a user element in LS-DYNA) an adaptive modelling strategy which allows for laminates to be initially modelled with only one element over the thickness.The user element kinematics can be adaptively enriched by introducing new degrees of freedom during the simulation to allow for more accurate stress predictions in critical regions by introducing discrete material interfaces, and for the modelling of delamination crack growth by introducing discrete crack surfaces interconnected with a cohesive zone law. In this work, special care has been taken to develop a robust method for explicit crash analysis. In the element, we also able to consider the correct intralaminar fracture toughness regularisation for various spatial discretisations. To assess and validate the models developed in the project, we have also conducted a series of bending and crushing experiments on component level. Three-point bending tests (in total 45 beams) have been conducted for three different carbon-epoxy material systems (pre-preg and vacuum infused), two different span lengths and two different lay-ups at several impact speeds. Similarly, crushing tests have been conducted for the same material systems by crushing tubes (in total 35 tubes) at various angles, with two different lay-ups and at two different loading speeds (quasi-static and dynamic). We believe that these tests serve as a very strong basis for any crash model validation.

In order to enhance effective observation and control over wild life, we have built a wireless sensor network. 25 video transmitting nodes are installed for animal behavior observation and experimental vibrotactile collars have been designed for effective control in an animal park. The goal of our research is two-folded. Firstly, to provide an interaction between digital users and animals, and monitor the animal behavior for safety purposes. Secondly, we investigate how animals can be controlled or trained based on vibrotactile stimuli instead of electric stimuli. We have designed a multimedia sensor network for human animal machine interaction. We have evaluated the effect of human animal machine state communication model in field experiments.

The value that is received from this application is both educative for the visitors and marketing for the tourist attraction. The conceptual model of the social media game builds upon the foundations of internet of things for the future version of a tourist attraction, such as the Digital Zoo.

    • The introduction of fibre reinforced polymers (FRP) in the automotive industry is strongly dependent on accurate and efficient modelling tools to predict the correct energy absorption in crash simulations. From experimental observations during axial crushing, it is obvious that in order to obtain good predictability in the simulations, delamination growth needs to be accounted for. Common practice to model delamination initiation and growth is to model each ply by separate elements, interconnected by cohesive interface elements. However, due to restrictions on the simulation time, such high-fidelity models are not feasible to use in industrial applications such as full vehicle crash simulations. We have previously presented an adaptive enrichment methodology for modelling of multiple and arbitrarily located delamination cracks using an equivalent single-layer (ESL) shell model. Due to the extreme level of complexity, crash simulations are usually performed using dynamic explicit solvers. Any adaptive methodology must therefore be adapted to fit this type of solver. This poses a problem since explicit solvers are very sensitive to modifications like refinements during the simulation. Consequently, besides yielding accurate results and being computationally efficient, the implementation must be numerically robust.

    • To introduce more fibre-reinforced polymers in cars, the automotive industry is strongly dependent on efficient modelling tools to predict the correct energy absorption in crash simulations. In this context, an adaptive modelling technique shows great potential. However, as the critical energy absorption in a crash occurs over a very short period of time, and since the deformation behaviour is very complex, car crash simulations are usually performed using explicit dynamic finite element solvers. Therefore, any practical adaptive technique must be adapted to an explicit setting in a software available to the automotive companies. In this paper, we propose an adaptive method for explicit finite element analysis and describe its implementation in the commercial finite element solver LS-DYNA. The method allows for both so-called weak discontinuities (discontinuities in strain), which are crucial for accurate stress and intralaminar damage predictions, and strong discontinuities (discontinuities in displacements), needed for a proper representation of growing delamination cracks. In particular, we detail the implementation of the proposed method into LS-DYNA and also how we propose to remedy the non-physical oscillations arising from the implementation of the adaptive scheme in a explicit dynamic setting. The paper is concluded with numerical examples where we demonstrate the potential for the adaptive approach and also perform a detailed study on its accuracy and stability.

    • In this study Moderate Resolution Imaging Spectroradiometer (MODIS) Aqua retrievals of aerosol optical thickness (AOT) at 555 nm are compared to Sun photometer measurements from Svalbard for a period of 9 years. For the 642 daily coincident measurements that were obtained, MODIS AOT generally varies within the predicted uncertainty of the retrieval over ocean (Delta AOT = +/- 0.03 +/- 0.05 . AOT). The results from the remote sensing have been used to examine the accuracy in estimates of aerosol optical properties in the Arctic, generated by global climate models and from in situ measurements at the Zeppelin station, Svalbard. AOT simulated with the Norwegian Earth System Model/Community Atmosphere Model version 4 Oslo global climate model does not reproduce the observed seasonal variability of the Arctic aerosol. The model overestimates clear-sky AOT by nearly a factor of 2 for the background summer season, while tending to underestimate the values in the spring season. Furthermore, large differences in all-sky AOT of up to 1 order of magnitude are found for the Coupled Model Intercomparison Project phase 5 model ensemble for the spring and summer seasons. Large differences between satellite/ground-based remote sensing of AOT and AOT estimated from dry and humidified scattering coefficients are found for the subarctic marine boundary layer in summer.

  • Clinical diversity and treatment results in tegumentary leishmaniasis : A European clinical report in 459 patients

    • Background Cutaneous leishmaniasis (CL) is frequent in travellers and can involve oro-nasal mucosae. Clinical presentation impacts therapeutic management. Methodology Demographic and clinical data from 459 travellers infected in 47 different countries were collected by members of the European LeishMan consortium. The infecting Leishmania species was identified in 198 patients. Principal findings Compared to Old World CL, New World CL was more frequently ulcerative (75% vs 47%), larger (3 vs 2cm), less frequently facial (17% vs 38%) and less frequently associated with mucosal involvement (2.7% vs 5.3%). Patients with mucosal lesions were older (58 vs 30 years) and more frequently immunocompromised (37% vs 3.5%) compared to patients with only skin lesions. Young adults infected in Latin America with L. braziliensis or L. guyanensis complex typically had an ulcer of the lower limbs with mucosal involvement in 5.8% of cases. Typically, infections with L. major and L. tropica acquired in Africa or the Middle East were not associated with mucosal lesions, while infections with L. infantum, acquired in Southern Europe resulted in slowly evolving facial lesions with mucosal involvement in 22% of cases. Local or systemic treatments were used in patients with different clinical presentations but resulted in similarly high cure rates (89% vs 86%). Conclusion/Significance CL acquired in L. infantum-endemic European and Mediterranean areas displays unexpected high rates of mucosal involvement comparable to those of CL acquired in Latin America, especially in immunocompromised patients. When used as per recommendations, local therapy is associated with high cure rates.

    • In this paper we will describe a positioning system based on synchronized IR light. Each node will be assigned a timeslot where they will send out an IR light. There is an IR camera that is also synchronized to the timeslots that will detect the position of each node and the ID that corresponds to the timeslot. To synchronize the clock of all nodes an IR flashlight is sent out that is detected by a photodiode on the nodes. The demo will show live video stream from a network camera where the ID and position of each node in view will be overlaid in real-time in the video.

  • A Comparison of Rule-based Analysis with Regression Methods in Understanding the Risk Factors for Study Withdrawal in a Pediatric Study

    • Regression models are extensively used in many epidemiological studies to understand the linkage between specific outcomes of interest and their risk factors. However, regression models in general examine the average effects of the risk factors and ignore subgroups with different risk profiles. As a result, interventions are often geared towards the average member of the population, without consideration of the special health needs of different subgroups within the population. This paper demonstrates the value of using rule-based analysis methods that can identify subgroups with heterogeneous risk profiles in a population without imposing assumptions on the subgroups or method. The rules define the risk pattern of subsets of individuals by not only considering the interactions between the risk factors but also their ranges. We compared the rule-based analysis results with the results from a logistic regression model in The Environmental Determinants of Diabetes in the Young (TEDDY) study. Both methods detected a similar suite of risk factors, but the rule-based analysis was superior at detecting multiple interactions between the risk factors that characterize the subgroups. A further investigation of the particular characteristics of each subgroup may detect the special health needs of the subgroup and lead to tailored interventions.

    • Collisions between vehicles and animals have increased in Norway and Europe over the last 40 years, causing economical losses as well as poor welfare for animals and people. In Norway 2 m high deer fences have been put up to prevent killings of semi-domestic reindeer at some limited road- and railway distances. However, these are very expensive and animal passages are needed. As a supplementary measure, an electronic system, aimed at warning the driver if reindeer are close to the road, was tested along the E6 main road at Saltfjellet, Norway, during March - April 2018. 235 female reindeer were equipped with radio transmitter collars (805.15.4 866 MHz). The animals were grazing in a collision exposed area at Saltfjellet. A total of 41 receivers were mounted on road sticks alongside the 4.5 km test distance. When a reindeer with transmitter was within 50 - 100 m proximity to a road stick, the receiver started blinking red. Saltfjellet reindeer herding district lost 15 reindeer in the test area from December 2017 until test start in February 2018. No reindeer were hit by cars during the test period. Nevertheless, 25% of the receivers became defective after a while, most possibly due to battery shortage. A new generation of transmitters and receivers was tested at Saltfjellet during December 2018 - April 2019. Preliminary results from this extended test are presented. We evaluate the electronic warning system as promising. However, improvements are needed in order to achieve optimal receiver and transmitter reliability in function.

    • OBJECTIVE: The aim of this study was to assess the short- and long-term prognostic significance of interleukin-18 (IL-18) levels in patients with acute coronary syndromes (ACS). METHODS AND RESULTS: In patients hospitalized with ACS (median age, 66 years; 30% females), we evaluated associations of serum IL-18 levels from day 1 (n=1261) with the short- (<3 months) and long-term (median, 7.6 years) risk of death, development of congestive heart failure (CHF), and myocardial infarction (MI). IL-18 was not significantly associated with short-term mortality. In the long term, IL-18 levels were significantly related to all-cause mortality, even after adjustment for clinical confounders (hazard ratio [HR], 1.19; 95% confidence interval, 1.07 to 1.33; P=0.002). Long-term, cardiovascular mortality was univariately related to IL-18, and the adjusted relation between noncardiovascular mortality and IL-18 was highly significant (HR, 1.36; 95% confidence interval, 1.11 to 1.67; P=0.003). IL-18 independently predicted CHF, MI, and cardiovascular death/CHF/MI in both the short and long term. Measurements from day 1 of ACS and 3 months after ACS had a similar power to predict late outcome. CONCLUSIONS: The addition of the measurement of IL-18 to clinical variables improved the prediction of risk of all-cause and noncardiovascular mortality. The association between IL-18 and noncardiovascular mortality is intriguing and warrants further study.

    • The interaction of light with multiple red blood cells was systematically investigated by the finite-difference time-domain method (FDTD). The simulations showed that the lateral multiple scattering between red blood cells is very weak and that the polarization has an almost insignificant influence on the distribution of the scattered light. The numerical results of the FDTD method were compared with the results from the Rytov approximation and the discrete dipole approximation (DDA). The agreement with the DDA was excellent.

    • Wilms tumor is the most common pediatric renal neoplasm, but few molecular prognostic markers have been identified for this tumor. Somatic deletion in the long arm of chromosome 16 (16q) is known to predict a less favorable outcome in Wilms tumor, but the underlying molecular mechanisms are not known. We show that 16q deletions are typically confined to immature anaplastic-blastic tumor elements, while deletions are absent in maturing tumor components. The smallest region of deletion overlap mapped to a 1.8-Mb segment containing the IRXB gene cluster including IRX3, IRX5, and IRX6, of which IRX3 is a recently identified regulator of tubular maturation during nephrogenesis. Tumors with 16q deletion showed a lower overall mRNA expression of IRXB genes, and 16q-deleted tumor cells failed to express IRX3 while it was expressed in differentiating tubular tumor elements with intact 16q. Consistent with a role for IRX3 in tubular differentiation, gene sets linked to Notch signaling, Rho signaling, and ion channel activity were enriched in tumors with high IRX3 expression, while WTs with low expression were enriched for gene sets linked to cell cycle progression. Low mRNA levels of IRXB genes were associated with diffuse anaplasia, high-stage disease, and death. A disturbed balance between tubular differentiation and self-renewal of anaplastic-blastic elements may thus be one mechanism linking 16q deletion to adverse outcome in Wilms tumor.

    • The influence of heat input and multiple welding cycles on the microstructure of the heat-affected zone in autogenously TIG-welded 6 mm 2507 type super duplex stainless steel plates was investigated. In order to produce multiple thermal cycles, one to four pass bead-on-plate welds were made with arc energies of 0.47 and 1.08 kJ/mm, corresponding to heat inputs of 0.37 and 0.87 kJ/mm. Several thermocouples were attached to record thermal cycles on the front and back sides of the plates. Finite element modelling was successfully done to map and correlate measured and calculated peak temperatures. Only minor changes were seen in the ferrite content at 1 and 2 mm from the fusion boundary. Nitrides formed in all passes of the low heat input samples in a region next to the fusion boundary, but only after the third and fourth passes of the high heat input samples. Sigma phase precipitated only in a zone heated to a peak temperature in the range of approximately 828 to 1028 °C. Multiple reheating was found to promote precipitation of sigma phase relatively more than slower cooling. A precipitation free zone was observed between the nitride and sigma phase bands. The precipitation behaviour could be understood from equilibrium phase diagrams, evaluation of local thermal cycles and by correlating results from the modelling and measurements of peak temperatures. It is suggested that the peak temperature, the accumulated time in the critical temperature range between approximately 828 and 1028 °C, and the number of thermal cycles are the most relevant criteria when evaluating the risk of sigma phase formation.

    • Human exposure to carbamates and organophosphates poses a serious threat to society and current pharmacological treatment is solely targeting the compounds' inhibitory effect on acetylcholinesterase. This toxicological pathway, responsible for acute symptom presentation, can be counteracted with currently available therapies such as atropine and oximes. However, there is still significant long-term morbidity and mortality. We propose mitochondrial dysfunction as an additional cellular mechanism of carbamate toxicity and suggest pharmacological targeting of mitochondria to overcome acute metabolic decompensation. Here, we investigated the effects on mitochondrial respiratory function of N-succinimidyl N-methylcarbamate (NSNM), a surrogate for carbamate insecticides, ex vivo in human platelets. Characterization of the mitochondrial toxicity of NSNM in platelets revealed a dose-dependent decrease in mitochondral oxygen consumption linked to respiratory chain complex I while the pathway through complex II was unaffected. In intact platelets, an increase in lactate production was seen, due to a compensatory shift towards anaerobic metabolism. Treatment with a cell-permeable succinate prodrug restored the NSNM-induced (100 μM) decrease in mitochondrial oxygen consumption and normalized lactate production to the level of control. We have demonstrated that carbamate-induced mitochondrial complex I dysfunction can be alleviated with a mitochondrial targeted countermeasure: a cell-permeable prodrug of the mitochondrial complex II substrate succinate.

  • Sustainable Timber Transport : Economic Aspects of Aerodynamic Reconfiguration

    • There is a need to reduce fuel consumption, and thereby reduce CO2-emissions in all parts of the transport sector. It is also well known that aerodynamic resistance affects the fuel consumption in a major way. By improving the aerodynamics of the vehicles, the fuel consumption will also decrease. A special type of transportation is that of timber, which is performed by specialized trucks with few alternative uses. This paper follows up on earlier papers concerning Swedish timber trucks where aerodynamic improvements for timber trucks were tested. By mapping the entire fleet of timber trucks in Sweden and investigating reduced fuel consumption of 2–10%, financial calculations were performed on how these improvements would affect the transport costs. Certain parameters are investigated, such as investment cost, extra changeover time and weight of installments. By combining these results with the mapping of the fleet, it can be seen under which circumstances these improvements would be sustainable. The results show that it is possible through aerodynamics to lower the transportation costs and make an investment plausible, with changeover time being the most important parameter. They also show that certain criteria for a reduced transportation cost already exist within the vehicle fleet today.
