  • Association Between Bone Mineral Density and Autoantibodies in Patients With Rheumatoid Arthritis

    • Objective: Autoantibodies, such as anti–citrullinated protein antibodies (ACPAs), have been described as inducing bone loss in rheumatoid arthritis (RA), which can also be reflected by bone mineral density (BMD). We therefore examined the association between osteoporosis and autoantibodies in two independent RA cohorts. Methods: Dual x-ray absorptiometry (DXA) of the lumbar spine and left hip was performed in 408 Dutch patients with early RA during 5 years of follow-up and in 198 Swedish patients with early RA during 10 years of follow-up. The longitudinal effect of ACPAs and other autoantibodies on several BMD measures was assessed using generalized estimating equations. Results: In the Dutch cohort, significantly lower BMD at baseline was observed in ACPA-positive patients compared to ACPA-negative patients, with an estimated marginal mean BMD in the left hip of 0.92 g/cm2 (95% confidence interval [95% CI] 0.91–0.93) versus 0.95 g/cm2 (95% CI 0.93–0.97) (P = 0.01). In line with this, significantly lower Z scores at baseline were noted in the ACPA-positive group compared to the ACPA-negative group (estimated marginal mean Z score in the left hip of 0.18 [95% CI 0.08–0.29] versus 0.48 [95% CI 0.33–0.63]) (P < 0.01). However, despite clear differences at baseline, ACPA positivity was not associated with greater decrease in absolute BMD or Z scores over time. Furthermore, there was no association between BMD and higher levels of ACPAs or other autoantibodies (rheumatoid factor and anti–carbamylated protein antibodies). In the Swedish cohort, ACPA-positive patients tended to have a higher prevalence of osteopenia at baseline (P = 0.04), but again, ACPA positivity was not associated with an increased prevalence of osteopenia or osteoporosis over time. Conclusion: The presence of ACPAs is associated with significantly lower BMD at baseline, but not with greater BMD loss over time in treated RA patients. These results suggest that ACPAs alone do not appear to contribute to bone loss after disease onset when disease activity is well-managed. © 2020 The Authors.     • Forskningen vid Tekniska högskolan i Luleå syftar till att vinna ny kunskap inom områden som är betydelsfulla för landet och regionen. Forskarutbildningen syftar till att utbilda licentiater och doktorer inom sådana områden.Under femårsperioden fram till och med 1995 bör den externfinansierade forskningen öka med 25 %. Vidare bör 5-10 europeiskla samarbetsprojekt etableras. För att uppmå dessa mål bör programarbete genomföras inom bland annat områdena datateknik, teknik för kallt klimat, material- konstruktions- och produktionsteknik, kvinnor och teknik samt miljöteknik.En plan bör även upprättas för ny- och återanskaffning av utrustning.En doktorsexamen och en licentiatexamen bör avläggas per tvåårsperid i varje professorsämne. För att klara detta bör varje ämne ha minst tre aktiva forskarstuderande. De forskarstuderande bör erbjudas god handledning, ett bra utbud av doktorandkurser och en god akademisk miljö.

  • Trender i kvävenedfall över Sverige 1955-2011

    • Kvävenedfallet har stor betydning för försurning och övergödning. Uppdraget har finansierats av Naturvårdsverket, med syfte att förbättra underlaget för att följa upp miljökvalitetsmålet Bara naturlig försurning . En ny, unik databas har etablerats, med de flesta mätningar som har skett i Sverige av nederbördsmängder och -kemi på öppet fält från 1955 och fram till idag. Beräkningar av nedfallet av oorganiskt kväve (NO3- + NH4+) med nederbörden (våtdepositionen) vid de sammanlagt 362 mätplatserna visar att nedfallet är högre i hela Sverige idag än när mätningarna startade 1955. Data visar en stor variation i kvävenedfallet mellan mätplatser och år, vilket gör det svårt att detektera statistiskt signifikanta förändringar över tid. Nedfallet av kväve med nederbörden till Sverige ökade signifikant under 20-årsperioden 1970-1989 i sydvästra Sverige, men inte i sydöstra eller norra Sverige. Under 20-årsperioden 1990-2009 skedde ingen förändring av nedfallet i norra eller sydvästra Sverige, men i sydöstra Sverige minskade nedfallet av olika kväveformer med nederbörden signifikant. Parallellt har kvävenedfallet uppskattats med MATCH-modellen med två olika simuleringar. Den ena simuleringen (TRENDMATCH) har använt indata från observerad meteorologi tillsammans med emissioner bestämda inom EMEP-programmet medan den andra (KLIMATMATCH) använder meteorologi från en klimatmodell och emissioner sammanställt för RCP4.5. Den med TRENDMATCH modellerade depositionen visar nedåtgående trender för såväl NO3- som NH4+ från 1990 till 2009 i alla regioner. Resultaten från KLIMATMATCH visar inte lika tydliga trender för NH4+, beroende på att de historiska emissionerna i RCP4.5 databasen både visar upp- och nedåtgående trender för perioden. Trender i kvävenedfall beräknade med MATCH-modellen är till mycket stor del styrda av trender i de emissioner som används av spridningsmodellen. Det kan finnas flera orsaker till att vi inte ser tydliga trender i kvävenedfallet under de senaste decennierna. Trendanalyser kräver data med långa tidsserier, vilket av olika orsaker ofta saknas. Om kvävenedfallet har förändrats, men förändringen är liten, så är det svårt eller omöjligt att urskilja detta ur de stora slumpmässiga mellanårsvariationerna. Det är också möjligt att nedfallet inte har förändrats, trots minskande emissioner i Europa, på grund av ändrade atmosfärskemiska förhållanden (t ex klimat och minskande svavelemissioner), eller om utsläppen av kväve från källor som är av vikt för Sverige inte har minskat i samma utsträckning som emissionerna för Europa som helhet har. Det är också svårt att uppskatta kväveemissioner, vilket i rapporten illustreras av de två emissionsuppskattningar som har använts, där den ena rapporterar minskande emissioner av ammoniak och den andra ökande. För att uppnå precision och statistiskt kunna säkerställa förhållandevis små förändringar i kvävenedfallet, i storleksordningen 20 procent under en period av 20 år, behövs mätningar vid ett stort antal platser runt om i landet. Vi föreslår en genomgång av hur de framtida mätningarna av kvävenedfallet, som utgör basen för övervakningen, bör utformas. En viktig punkt är att existerande stationer med långa tidsserier bör bevaras, något som kräver långsiktig finansiering.

    • Many rotating machineries are supported by bearings with clearance, which are further clamped in, compared to the bearing stiffness, weak pedestals. As have been reported in several studies, it is essential to choose a proper clearance in order to avoid unwanted vibrations. When rotating machineries supported by bearings with clearance are subjected to stationary loads (gravity, magnetic pull etc.), it may not loose contact between the shaft and the bearing (i.e. full contact). However, even in full contact unwanted dynamics can occur which is of interest in this paper. It is found that the clearance give raise to anisotropic pedestal stiffness. It is further shown that some of the resonance frequencies decrease with the clearance.

    • BACKGROUND: Although tissue microarray (TMA) studies of histopathologic material have been frequently reported in studies of malignant diseases, the question of sample size (i.e., the diameter and the number of tissue cylinders investigated) has been rarely discussed. This study addresses the methodologic question of sample size in a variety of tumor types.MATERIAL AND METHODS: Material from 29 cases of lung carcinoma (small cell, squamous cell, and adenocarcinomas) was examined immunohistochemically for Ki-67 and p53 expression in virtually constructed cylinders of different diameters. The influence of tissue sample size (i.e., different numbers of virtual cylinders) was also investigated. Results from Ki-67 evaluation were analyzed as a continuous variable, whereas p53 expression was scored. p53 evaluations based on scoring in cylinders versus scoring of whole sections were also compared. Furthermore, 10 cases of endometrial and breast carcinomas were evaluated for estrogen receptor, Ki-67, and HER2 by scoring up to five cylinders.RESULTS AND CONCLUSIONS: Tissue cylinders of 0.6 and 1.0 mm diameters were compared and found equally informative about Ki-67 expression (intraclass correlation, 0.96). A statistical approach considering intraindividual and interindividual variation data is presented, indicating that in this specific setting three cylinders per case is an adequate sample size for TMA studies. Further sampling yields only a small gain in accuracy as determined by Ki-67 quantification and p53 scoring (kappa-coefficient, 0.9). For endometrial and breast tissues, TMA scoring of three cylinders yielded excellent agreement (kappa, >0.75) compared with whole-section scoring.

    • Emissions from Heating Plant burning Biomass in the interval 0.5 - 10 MW The emissions from the heating plants in the interval 0.5 - 10 MW, burning biomass, have been investigated. The measurements have included carbon monoxide, carbon dioxide, nitrogen oxides, a number of volatile organic compounds (VOC) and polyaromatic hydrocarbons (PAH). Wood chips, wood briquettes, straw, bark, wood pellets, wood powder and peat are the fuel quantities included in the survey. The measurements were made at existing operating conditions for each plant. No adjustments to the combustion equipment were made. The measurements showed that biomass combustion can give rise to high emissions of unburnt hydrocarbons. In six out of thirteen tests, significant VOC emissions with methande, ethylene, propene and acetlyene in the interval 5 - 500 mg/MJ were recorded. In all these cases the CO emissions exceeded 1000 mg/MJ. A correlation between increasing CO content and increasing VOC content was indicated in these cases. This is most apparent for methane, which is the most abundant VOC species. In the other cases, where the CO emissions were below 500 mg/MJ, no significant VOC emissions were recorded. NOx, emissions varied for all cases between 35 and 256 mg NO2/MJ. No clear correlation between low levels of unburnt hydrocarbons and high levels of NOx could be established.

    • Climate change has been shown to shift the seasonal timing (i.e. phenology) and distribution of species. The phenological effects of climate change on living organisms have often been tested using first occurrence dates, which may be uninformative and biased. More rarely investigated is how different phases of a phenological sequence (e.g. beginning, central tendency and end) or its duration have changed over time. This type of analysis requires continuous observation throughout the phenological event over multiple years, and such data sets are rare. In this study we examined the impact of temperature on long-term change of passage timing and duration of the spring migration period in birds, and which species' traits explain species-specific variation. Data used covered 195 species from 21 European and Canadian bird observatories from which systematic daily sampling protocols were available. Migration dates were negatively associated with early spring temperature and timings had in general advanced in 57 years. Short-distance migrants advanced the beginning of their migration more than long-distance migrants when corrected for phylogenic relatedness, but such a difference was not found in other phases of migration. The advancement of migration has generally been greater for the beginning and median phases of migration relative to the end, leading to extended spring migration seasons. Duration of the migration season increased with increasing temperature. Phenological changes have also been less noticeable in Canada even when corrected for rate of change in temperature. To visualize long-term changes in phenology, we constructed the first multi-species spring migration phenology indicator to describe general changes in median migration dates in the northern hemisphere. The indicator showed an average advancement of one week during five decades across the continents (period 1959-2015). The indicator is easy to update with new data and we therefore encourage future research to investigate whether the trend towards longer periods of occurrence or emergence in spring is also evident in other migratory populations. Such phenological changes may influence detectability in monitoring schemes, and may have broader implications on population and community dynamics.

    • Electron IMRT treatments have the potential to reduce the integral dose due to the limited range of the electrons. However, bremsstrahlung produced in the scattering foils could penetrate an added electron MLC (eMLC), thus producing an unmodulated dose contribution that could become unacceptable in electron IMRT treatments. To limit this bremsstrahlung contribution, the photon MLC (xMLC) was used to track the eMLC, but with a margin to avoid penumbra widening through partial screening of the effective electron source. The purpose of this work was to study the effect of the photon-electron MLC tracking on the electron beam penumbra for different treatment head designs. Both isocentric designs and designs where the eMLC is used close to the patient (proximity geometry) have been analysed using Monte Carlo simulations. At 22.5 MeV energy, a tracking margin of 1 cm was enough to avoid penumbra degradation for a helium-filled isocentric geometry, while air-filled geometries (including proximity geometries) require a 2-3 cm margin. Illustrated by an example of a chest wall treatment by electron IMRT, the use of 1 cm tracking margin will reduce the collimator leakage contribution by a factor of 36 as compared to using a static setting of the photon collimator.

  • Energy and intensity modulated radiation therapy with electrons

    • In recent years intensity modulated radiation therapy with photons (xIMRT) has gained attention due to its ability to reduce the dose in the tissues close to the tumour volume. However, this technique also results in a large low dose volume. Electron IMRT (eIMRT) has the potential to reduce the integral dose to the patient due to the dose fall off in the electron depth dose curves. This dose fall off makes it possible to modulate the dose distribution in the direction of the beam by selecting appropriate electron energies. The use of a computer based energy selection method was examined in combination with the IMRT technique to optimise the electron dose distribution. It is clearly illustrated that the energy optimisation procedure reduces the dose to lung and heart in a breast cancer treatment. To shape the multiple electron subfields (beamlets) that are used in eIMRT, an electron multi leaf collimator (eMLC) is needed. However, photons produced in a conventional electron treatment head could penetrate such an added eMLC, thus producing an undesirable dose contribution. The leakage levels normally achieved are acceptable for standard single electron field treatments but could become unacceptably high in eIMRT treatments where a lot of small subfields are combined. To limit this photon contribution, the photon MLC (xMLC) was used to shield off large parts of the photon leakage. The effect of this xMLC shielding on the reduction of photon leakage, the electron beam penumbras, and electron output (dose level), was studied using Monte Carlo methods for different electron treatment head designs. The use of helium as a mean to reduce the electron scatter in the treatment head, and thus the perturbating effect of the xMLC on electron beam penumbra and output, was also investigated. This thesis shows that the effect of the xMLC shielding on the electron beam penumbra and output can be made negligible while still obtaining a significantly reduced x-ray leakage dose contribution. The result is a large gain in radiation protection of the patient and a better dynamic range for the eIMRT dose optimisation. For this optimisation a computer based electron energy selection method was developed and tested on two clinical cases.

    • In energy modulated electron therapy a large fraction of the segments will be arranged as abutting segments where inhomogeneities in segment matching regions must be kept as small as possible. Furthermore, the output variation between different segments should be minimized and must in all cases be well predicted. For electron therapy with add-on collimators, both the electron MLC (eMLC) and the photon MLC (xMLC) contribute to these effects when an xMLC tracking technique is utilized to reduce the x-ray induced leakage. Two add-on electron collimator geometries have been analyzed using Monte Carlo simulations: One isocentric eMLC geometry with an isocentric clearance of 35 cm and air or helium in the treatment head, and one conventional proximity geometry with a clearance of 5 cm and air in the treatment head. The electron fluence output for 22.5 MeV electrons is not significantly affected by the xMLC if the shielding margins are larger than 2-3 cm. For small field sizes and 9.6 MeV electrons, the isocentric design with helium in the treatment head or shielding margins larger than 3 cm is needed to avoid a reduced electron output. Dose inhomogeneity in the matching region of electron segments is, in general, small when collimator positions are adjusted to account for divergence in the field. The effect of xMLC tracking on the electron output can be made negligible while still obtaining a substantially reduced x-ray leakage contribution. Collimator scattering effects do not interfere significantly when abutting beam techniques are properly applied.

    • Photoneutron production on the nuclei of high-Z components of medical accelerator heads can lead to a significant secondary dose during a course of bremsstrahlung radiotherapy, However, a quantitative evaluation of secondary neutron dose requires improved data on the photoreaction yields. These have been measured as a function of photon energy, neutron energy and neutron angle for W-nat, using tagged photons at the MAX-Lab photonuclear facility in Sweden. This work presents neutron yields for W-nat(gamma, n) and compares these with the predictions of the Monte Carlo code MCNP-GN, developed specifically to simulate photoneutron production at medical accelerators.

    • Functional Products (FP) and Product Service Systems (PSS) may be seen as integrated systems comprising hardware and support services. For such offerings, availability is key. Little research has been done on integrating Data Stream Management Systems (DSMS) for monitoring (parts of) a FP to improve system availability. This paper introduces an approach for how data stream mining may be applied to monitor hardware being part of a Functional Product. The result shows that DSMS have the potential to significantly support continuous availability awareness of industrial systems, especially important when the supplier is to supply a function with certain availability.

    • In shipyard panel-production lines several rectangular plates ae butt welded to form large panels. One problem is the tendency of hot cracking in the end portion of the butt weld. In this paper transient temperature fields and stress fields in butt-welded plates are calculated using the finite-element method. The calculations are performed for different welding speeds, for different heat inputs, and for one-sided and two-sided welding.

För svenska vattenområden beskrivs miljöutvecklingen i ett ekosystemsperspektiv, dels för att tydliggöra fiskens ekologiska roll och beskriva yttre miljöfaktorer som påverkar fiskbestånden, dels för att belysa fiskets effekter på miljön.Fiskbestånd och miljö i hav och sötvatten är utarbetad av Sveriges lantbruksuniversitet (SLU), Institutionen för akvatiska resurser (SLU Aqua), på uppdrag av Havs- och vattenmyndigheten. Rapporten sammanfattar utveckling och beståndsstatus för de kommersiellt viktigaste fisk- och kräftdjursarterna i våra vatten. Bedömningar och förvaltningsråd är baserade på Internationella Havsforskningsrådets (ICES) rådgivning, SLU Aquas nationella och regionala provfiskedata, samt yrkesfiskets rapportering.

    • We investigated productivity at the basal trophic level in 15 unproductive lakes in a gradient ranging from clear-water to brown-water (humic) lakes in northern Sweden. Primary production and bacterial production in benthic and pelagic habitats were measured to estimate the variation in energy mobilization from external energy sources (primary production plus bacterial production on allochthonous organic carbon) along the gradient. Clear-water lakes were dominated by autotrophic energy mobilization in the benthic habitat, whereas humic lakes were dominated by heterotrophic energy mobilization in the pelagic habitat. Whole-lake (benthic + pelagic) energy mobilization was negatively correlated to the light-extinction coefficient, which was determined by colored terrestrial organic matter in the lake water. Thus, variation in the concentration of terrestrial organic matter and its light-absorbing characteristics exerts strong control on the magnitude, as well as on the processes and pathways, of energy mobilization in unproductive lakes. We suggest that unproductive lakes in general are sensitive to input of terrestrial organic matter because of its effects on basal energy mobilization in both benthic and pelagic habitats.

    • This study quantified new biomass production of algae and bacteria in both benthic and pelagic habitats of clear-water lakes to contrast how carbon from the atmosphere and terrestrial sources regulates whole-lake metabolism. We studied four small unproductive lakes in subarctic northern Sweden during one summer season. The production of new biomass in both benthic and pelagic habitats was calculated as the sum of autotrophic production by algae and heterotrophic production by bacteria using allochthonous organic carbon (OC). Whole-lake production of new biomass was dominated by the benthic habitat (86% +/- 4% [mean +/- SD]) and by primary production (77% +/- 9%). Still, heterotrophic bacteria fueled by allochthonous OC constituted a significant portion of the new biomass production in both benthic (19% +/- 11%) and pelagic habitats (51% +/- 24%). In addition, overall net production (primary production minus respiration) was close to zero in the benthic habitats but highly negative (-163 +/- 81 mg C.m(-2).d(-1)) in pelagic regions of all lakes. We conclude (1) that allochthonous OC supported a significant part of total production of new biomass in both pelagic and benthic habitats, (2) that benthic habitats dominated the whole-lake production of new biomass, and (3) that respiration and net CO2 production dominated the carbon flux of the pelagic habitats and biomass production dominated the benthic carbon flux. Taken together, these findings suggest that previous investigations have greatly underestimated the productivity of clear-water lakes when benthic autotrophic production and metabolism of allochthonous OC have not been measured.

  • Competition mediated coexistence of invading intermediate consumer, ninespine stickleback, and a resident omnivorous top predator, Arctic char

    • Climate change results in changes in the geographical distribution of species. Species invasion success into a new area is dependent both on the dispersal ability of species as well as the strength and identity of biotic interactions between resident and invading species. Coexistence in intraguild predation (IGP) systems depends on the relative strength of predation and competition interactions which in turn are temperature dependent. We investigated the effects of introducing an intermediate consumer, ninespine stickleback (Pungitius pungitius), into allopatric populations of the omnivorous top predator Arctic char (Salvelinus alpinus). Introductions were performed in lakes with different climate regimes, two tundra lakes and two forest lakes that differed in average summer temperatures with 1.4 ± 0.5 ºC (average ± 1SD). We found that sticklebacks were able to invade and increase in density in both tundra and forest lakes. Sticklebacks had strong negative effects on resource densities which also was reflected in a decreased growth of small char. Increasing stickleback density had a positive effect on growth of large adults and on the maximum size of char. We conclude that stickleback presence is not limited by biological interactions in these systems but rather by dispersion ability. We suggest that the size dependency in the response of char to the invasion of sticklebacks is fundamental for the successful invasion of sticklebacks, and that size dependent interactions including cannibalism play important roles for coexistence in natural IGP-systems.

  • Temperature mediated effects on top consumer populations in subarctic lakes

    • The effects of temperature on top consumer populations in subarctic lake communities were studied by contrasting two lake pairs in different climate regimes: one pair on the low alpine tundra and one pair in the subalpine birch forest. We measured zooplankton and macroinvertebrate biomasses over the season and estimated population density and size structure of the top consumer Arctic char (Salvelinus alpinus). Furthermore, we modelled char growth using literature data on temperature dependent search rate, handling time and metabolic demands. The forest lakes were warmer than the tundra lakes. Char in the forest lakes were larger and had a higher individual growth compared to char in the tundra lakes, while population density and biomasses of char were not different between the forest and the tundra lakes. There were no differences in macroinvertebrate and zooplankton resource levels available for char between lake pairs. Our modeling of char growth revealed that higher temperature increased growth of char at the observed resource densities, suggesting that the higher temperature in the forest lakes was primarily the cause of the higher growth of char in these lakes. We suggest that cannibalism in char may regulate char recruitment and thereby population density and biomass of char leading to effects of increasing temperature on consumer biomass and consumer individual growth different from what is expected in pure consumer-resource systems. Our results emphasize the importance of feedbacks within ecosystems when addressing effects of climate change and increasing temperature on lake communities.

    • The polycondensation of polyesters from C-12 monomers at 95 degrees C in aqueous o/w emulsions, stabilized by acidic surfactants, has been studied in detail with a range of methods during the course of the reaction, resulting in a better understanding of the underlying reaction mechanisms. Comparisons of different surfactants, and effects of added NaCl, demonstrate that the reaction site is located at the interface between the hydrophobic core of the emulsion droplets and the surrounding water and that the reaction rate is dependent on the local concentration of oxonium ions at the reaction site. The equilibrium conversion achieved at long reaction times is, however, independent of the choice of surfactant or addition of salt, and the state of thermodynamic equilibrium is discussed thoroughly. Interestingly, a fraction of numerous "nanoparticles" (droplets in the size range <= 100 nm) have been found to develop in addition to the original fraction of droplets in the 10 mu m size range. It is suggested that these nanoparticles are formed when monomers dissolved in the aqueous phase undergo an acid-catalyzed reaction to generate water-insoluble oligomers. Once the nanoparticles are formed, the reactions in them proceed with a reaction mechanism similar to emulsion polymerization.

  • Widespread episodic thiamine deficiency in Northern Hemisphere wildlife

    • Many wildlife populations are declining at rates higher than can be explained by known threats to biodiversity. Recently, thiamine (vitamin B-1) deficiency has emerged as a possible contributing cause. Here, thiamine status was systematically investigated in three animal classes: bivalves, ray-finned fishes, and birds. Thiamine diphosphate is required as a cofactor in at least five life-sustaining enzymes that are required for basic cellular metabolism. Analysis of different phosphorylated forms of thiamine, as well as of activities and amount of holoenzyme and apoenzyme forms of thiaminedependent enzymes, revealed episodically occurring thiamine deficiency in all three animal classes. These biochemical effects were also linked to secondary effects on growth, condition, liver size, blood chemistry and composition, histopathology, swimming behaviour and endurance, parasite infestation, and reproduction. It is unlikely that the thiamine deficiency is caused by impaired phosphorylation within the cells. Rather, the results point towards insufficient amounts of thiamine in the food. By investigating a large geographic area, by extending the focus from lethal to sublethal thiamine deficiency, and by linking biochemical alterations to secondary effects, we demonstrate that the problem of thiamine deficiency is considerably more widespread and severe than previously reported.

    • Projektets huvudsakliga syfte har varit att utveckla och testa en metod för syresättning av djupvatten i Östersjön genom nedpumpning av syrerikt ytvatten med hjälp av vågdrivna syrepumpar. Inom projektet har två olika pilotanläggningar tagits fram, byggts och varit i drift mellan 2010 och 2013. En vågdriven pump (WEBAP I) testades under 2 år i Hanöbukten utanför Simrishamn och en elektrisk pump (WEBAP II) placerades i Kanholmsfjärden i Stockholms skärgård. Förutom tester for att undersöka den vågdrivna pumpens effektivitet och framtida utformning så har även effekterna av syresättning på omgivande vattens syre- och fosforkoncentrationer studerats under projektet. Med hjälp av modeller studerades uppskalning av tekniken samt beräkningar av åtgärdens klimatavtryck (carbon footprint) med hjälp av förenklad livscykelanalysmetodik (LCA). Projektet har tagit fram olika tekniska alternativ av syrepumpar för olika behov. Den vågdrivna syrepumpen använder sig direkt av vågorna som energi- och syreresurs vilket har flera fördelar, bland annat en enkel och robust konstruktion samt lokalt tillförd energi genom att vågor nyttjas. Den eldrivna pumpen är en nyutveckling som möjliggör att pumpning kan drivas av vind- och solenergi eftersom effektförbrukningen i pumpen är låg. De två testade systemen visade sig kunna pumpa ner stora mängder ytvatten, men ytterligare förbättringar och utveckling krävs för långsiktigt störningsfri funktion. Rapporten ger en del förslag till sådan utveckling.

    • Cross-ecosystem movements of material and energy are ubiquitous. Aquatic ecosystems typically receive material that also includes organic matter from the surrounding catchment. Terrestrial-derived (allochthonous) organic matter can enter aquatic ecosystems in dissolved or particulate form. Several studies have highlighted the importance of dissolved organic carbon to aquatic consumers, but less is known about allochthonous particulate organic carbon (POC). Similarly, most studies showing the effects of allochthonous organic carbon (OC) on aquatic consumers have investigated pelagic habitats; the effects of allochthonous OC on benthic communities are less well studied. Allochthonous inputs might further decrease primary production through light reduction, thereby potentially affecting autotrophic resource availability to consumers. Here, an enclosure experiment was carried out to test the importance of POC input and light availability on the resource use in a benthic food web of a clear-water lake. Corn starch (a C-4 plant) was used as a POC source due to its insoluble nature and its distinct carbon stable isotope value (delta C-13). The starch carbon was closely dispersed over the bottom of the enclosures to study the fate of a POC source exclusively available to sediment biota. The addition of starch carbon resulted in a clear shift in the isotopic signature of surface-dwelling herbivorous and predatory invertebrates. Although the starch carbon was added solely to the sediment surface, the carbon originating from the starch reached zooplankton. We suggest that allochthonous POC can subsidize benthic food webs directly and can be further transferred to pelagic systems, thereby highlighting the importance of benthic pathways for pelagic habitats.

    • The increasing number of new psychoactive substances made available for recreational drug use has created a challenge for clinical toxicology and drug testing laboratories. As a consequence, the routine immunoassay drug testing may become less effective due to an increased occurrence of false negative and false positive screening results. This work aimed to extend the knowledge about analytical cross-reactivity of new substances in selected CEDIA, EMIT, and KIMS immunoassays for drugs-of-abuse screening. Urine standards were prepared by spiking blank urine with 45 new substances. Authentic urine samples from intoxication cases identified by liquid chromatography-tandem mass spectrometry (LC-MS/MS) were also studied. Several new psychoactive substances were demonstrated to display cross-reactivity in the immunoassays. CEDIA Amphetamine/Ecstasy and EMIT d.a.u. Amphetamine Class tests showed the highest reactivity towards the new drugs, which was expected since many have amphetamine-like structure and activity. In the samples from authentic cases, five new substances displayed 100% detection rate in the CEDIA Amphetamine/Ecstasy test. In conclusion, cross-reactivity data in routine urine drug screening immunoassays for a number of new psychoactive substances not studied before were reported. In both spiked and authentic urine samples, some new substances showed significant cross-reactivity and are thus detectable in the routine screening methods.   • Energy structure and transition rates in the Ne-like sequence from relativistic CI calculations

    • Atomic data are important in astrophysical applications and transition rates can be used in the determination of element abundances and plasma diagnostics. To provide for the extensive data needs a number of general computer codes such as SUPERSTRUCTURE, CIV3, and ATSP2K have been developed. As an alternative to these codes, which all rely on the Breit-Pauli approximation, the fully relativistic GRASP2K code can be used. GRASP2K is based on the multiconfiguration Dirac-Hartree-Fock method and implements a bi-orthogonal transformation method that permits initial and final states in a transition array to be optimized separately, which, in many cases, leads to more accurate values of the resulting rates. The GRASP2K package also contains modules to compute diagonal and off-diagonal hyperfine interaction constants, isotope shifts, Land´e gJ factors, and splittings of magnetic sub-state in intermediate and strong magnetic fields. In this work, GRASP2K has been applied to provide highly accurate spectroscopic data for ions in the Ne-like sequence between Mg III and Kr XXVII. Valence, core-valence, and core-core correlation effects were accounted for through SD-MR expansions to increasing sets of active orbitals. In Mg III, Al IV, Si V, P VI, S VII, and Ar IX, for which experimental energies are known to high accuracy, the mean error in the calculated energies is only 0.011%. For ions with no available experimental energy levels the calculated values should be most valuable in various applications. The high accuracy of the calculated energies makes it possible, in some cases, to to point out experimental values that are in error. Babushkin (length) and Coulomb (velocity) forms of transition rates are computed and agree to within a few percent for the majority of the allowed transitions. Computed lifetimes for states belonging to the 2p33s and 2p53d configurations are in good agreement with values from beam-foil measurements as well as from accurate MCHF Breit-Pauli calculations.

    • Abstract Background The characteristics of patients who survive out-of-hospital cardiac arrest (OHCA) are incompletely known. The characteristics of survivors of OHCA during a period of 16 years in Sweden are described. Methods All the patients included in the Swedish Cardiac Arrest Registry between 1992 and 2007 in whom cardiopulmonary resuscitation was attempted and who were alive after 1 month were included in the survey. Results In all, 2432 survivors were registered. Information on initial rhythm at their first ECG recording was missing in 11%. Of the remaining 2165 survivors, 80% had a shockable rhythm and 20% had a non-shockable rhythm. Only a minority with a shockable rhythm among the bystander-witnessed cases were defibrillated within 5 min after cardiac arrest. This proportion did not change during the entry period. Among survivors found in a non-shockable rhythm, the majority were bystander-witnessed cases and a few had a delay from cardiac arrest to ambulance arrival of <5 min. Of all survivors, more women (27%) than men (18%) were found in a non-shockable rhythm (p<0.0001). During the 16 years in which the register was used for this study, the proportion of survivors found in a shockable rhythm did not change significantly. The cerebral performance categories score indicated better cerebral function among patients found in a shockable rhythm than in those found in a non-shockable rhythm. Conclusion Among survivors of OHCA, a substantial proportion was found in a non-shockable rhythm and this occurred more frequently in women than in men. The proportion of survivors found in a shockable rhythm has not changed markedly over time. Survivors found in a shockable rhythm had a better cerebral performance than survivors found in a non-shockable rhythm. The proportion of survivors who were bystander-witnessed and found in a shockable rhythm and defibrillated early is still remarkably low.

    • Background: Common mental disorders (CMDs) are among the leading causes of sick leave in Sweden and other OECD countries. They result in suffering for the individual and considerable financial costs for the employer and for society at large. The occupational health service (OHS) can offer interventions in which both the individual and the work situation are taken into account. The aim of this paper is to describe the design of a study evaluating the effectiveness of an intervention given at the OHS to employees with CMDs or stress-related symptoms at work. In addition, intervention fidelity and its relation to the outcome will be assessed in a process analysis.Methods: The study is designed as a cluster randomized trial in which the participating OHS consultants are randomized into either delivering the intervention or performing care as usual. Employees with CMDs or stress-related symptoms at work are recruited consecutively by the OHS consultants. The intervention aims to improve the match between the employee and the job situation. Interviews are held individually with the employee and the nearest supervisor, after which a joint meeting with both the employee and the supervisor takes place. A participatory approach is applied by which the supervisor and the employee are guided by the OHS consultant and encouraged to actively take part in problem solving concerning the work situation. Outcomes will be assessed at baseline and at six and 12 months. A long-term follow-up at 3 years will also be performed. The primary outcome is registered sickness absence during a 1-year period after study inclusion. Secondary outcomes are mental health and work ability. The intervention's cost effectiveness, compared to treatment as usual, both for society and for the employer will be evaluated. A process evaluation by both the OHS consultants and the employee will be carried out.Discussion: The study includes analyses of the effectiveness of the intervention (clinical and economic) as well as an analysis of its implementation at the participating OHSs. Possible methodological challenges such as selection bias and risk of contamination between OHS consultants delivering the experimental condition and consultants giving usual care are discussed.

  • Intracellularly expressed rotavirus NSP4 stimulates release of serotonin (5-HT) from human enterochromaffin cells

    • Rotavirus (RV) is associated with diarrhoea and vomiting, but the mechanisms behind these symptoms remain unresolved. While RV have been shown to infect and stimulate secretion of serotonin (5-hydroxytryptamine; 5-HT) from human enterochromaffin (EC) cells and to infect EC cells in the small intestine of mice, it remains to identify which intracellularly expressed viral protein (VP) being responsible for this novel property.To address this issue, human EC cells were transfected with small interfering RNA (siRNA) targeting the structural (VP4, VP6 and VP7) and the non-structural protein 4 (NSP4) followed by infection with Rhesus rotavirus (RRV). siRNA specific to NSP4 (siRNANSP4) significantly attenuated secretion of 5-HT compared to siRNAVP4, siRNAVP6 , siRNAVP7 and non-targeting (Nt) siRNAnt. Intracellular calcium clamping with BABTA/AM showed that intracellularly expressed NSP4-stimulated secretion of 5-HT from EC cells was calcium-dependent. Furthermore RV down-regulated the 5-HT transporter (SERT) mRNA in ileum but not tryptophan hydroxylase 1 (TPH1) mRNA the rate-limiting enzyme for 5-HT synthesis. The unaffected expression of TPH1 mRNA in the intestinal segments suggests that release of 5- HT primarily originates from pre-made 5-HT rather than from newly synthesised 5-HT mRNA. Moreover, down-regulation of SERT mRNA in ileum presumably resulted in reduced re- uptake of 5-HT by SERT to EC cells and thus increased extracellular 5-HT in the small intestine. Moreover, 7/7 infant mice responded following intraperitoneal administration of 5-HT with rapid (<30 min) diarrhoea in dose-dependent manner. In the light of these results and the fact that both 5-HT and NSP4 can induce diarrhoea in mice, a disease mechanism to RV diarrhoea is proposed.

    • Rotavirus (RV) has been shown to infect and stimulate secretion of serotonin from human enterochromaffin (EC) cells and to infect EC cells in the small intestine of mice. It remains to identify which intracellularly expressed viral protein(s) is responsible for this novel property and to further establish the clinical role of serotonin in RV infection. First, we found that siRNA specifically silencing NSP4 (siRNA(NSP4)) significantly attenuated secretion of serotonin from Rhesus rotavirus (RRV) infected EC tumor cells compared to siRNA(VP4), siRNA(VP6) and siRNA(VP7). Second, intracellular calcium mobilization and diarrhoeal capacity from virulent and avirulent porcine viruses correlated with the capacity to release serotonin from EC tumor cells. Third, following administration of serotonin, all (10/10) infants, but no (0/8) adult mice, responded with diarrhoea. Finally, blocking of serotonin receptors using Ondansetron significantly attenuated murine RV (strain EDIM) diarrhoea in infant mice (2.9 vs 4.5 days). Ondansetron-treated mice (n = 11) had significantly (p amp;lt; 0.05) less diarrhoea, lower diarrhoea severity score and lower total diarrhoea output as compared to mock-treated mice (n = 9). Similarly, Ondansetron-treated mice had better weight gain than mock-treated animals (p amp;lt; 0.05). A most surprising finding was that the serotonin receptor antagonist significantly (p amp;lt; 0.05) also attenuated total viral shedding. In summary, we show that intracellularly expressed NSP4 stimulates release of serotonin from human EC tumor cells and that serotonin participates in RV diarrhoea, which can be attenuated by Ondansetron.

    • Background and purpose: In locally advanced prostate cancer (PC), androgen deprivation therapy (ADT) in combination with whole prostate radiotherapy (RT) is the standard treatment. ADT affects the prostate as well as the tumour on multiparametric magnetic resonance imaging (MRI) with decreased PC conspicuity and impaired localisation of the prostate lesion. Image texture analysis has been suggested to be of aid in separating tumour from normal tissue. The aim of the study was to investigate the impact of ADT on baseline defined MRI features in prostate cancer with the goal to investigate if it might be of use in radiotherapy planning.Materials and methods: Fifty PC patients were included. Multiparametric MRI was performed before, and three months after ADT. At baseline, a tumour volume was delineated on apparent diffusion coefficient (ADC) maps with suspected tumour content and a reference volume in normal prostatic tissue. These volumes were transferred to MRIs after ADT and were analysed with first-order -and invariant Haralick -features.Results: At baseline, the median value and several of the invariant Haralick features of ADC, showed a significant difference between tumour and reference volumes. After ADT, only ADC median value could significantly differentiate the two volumes.Conclusions: Invariant Haralick -features could not distinguish between baseline MRI defined PC and normal tissue after ADT. First-order median value remained significantly different in tumour and reference volumes after ADT, but the difference was less pronounced than before ADT.

  • Sequential Comparative Study of Dielectric Response and Analyses of Oil and Paper from a Power Transformer Undergoing Repair

    • Methods, based on characterisation of dielectric response, for assessing moisture content in pressboard and paper are gaining ground. However, there is a need for calibrating obtained results with results from other techniques. This communication presents results from a study of a 50/10 kV - 40 MVA power transformer, which was sent to a workshop for repair, thus allowing for collecting samples of the solid insulation. Dielectric spectroscopy measurements in frequency domain were conducted at several occasions before and after the repair. Moisture estimation based on dielectric response measurements as well as on titration of oil samples yielded similar results to those obtained by direct titration of paper samples, as long as appropriate temperature values were used in all the analyses mentioned. The temperature value during measurements should be used for the analysis of the dielectric response, while in-service temperature of oil should be used for the evaluation based on Karl Fisher titration.

  • Changes in body composition after 2 years with rheumatoid arthritis

    • Objectives: Rheumatoid arthritis (RA) is associated with cardiovascular disease (CVD). Possible predictors for CVD are early changes in body composition. We therefore evaluated changes in lean body mass (LBM), lean mass of arms and legs (LMAL), total body fat mass (BFM), and truncal fat distribution (FD) after 2 years with RA and possible predictors. Methods: We registered 63 (45 women) RA patients with disease duration of <= 12 months at baseline and after 2 years. Disease Activity Score using 28 joint counts (DAS28), Health Assessment Questionnaire (HAQ) score, body mass index (BMI), comorbidities, smoking, and medications were recorded. Total and regional lean mass and fat mass were measured with dual energy X-ray absorptiometry (DXA). The data were compared with 63 age- and gender-matched controls. Results: LBM and LMAL at baseline in RA patients were significantly lower in men (p = 0.020 and 0.002, respectively) compared to matched controls. Truncal FD was non-significantly increased in RA patients (women p = 0.133, men p = 0.119). The age-related deterioration with decreased LBM after 2 years (p = 0.002 in women and men) and increased BFM (p < 0.001) and truncal FD (p = 0.020) in women were all significantly less pronounced in RA patients than in matched controls. Conclusions: In patients with early RA and after initiation of therapy, the age-related deterioration with decreasing LBM and increasing truncal FD was lower than in control subjects in this observational study. These potentially harmful alterations seem to be modifiable factors in patients with early RA.

    • Objectives: Osteoporosis is a known complication of rheumatoid arthritis (RA). This prospective study aimed to evaluate whether disease activity, disability, and glucocorticoid (GC) treatment in early RA were risk factors for loss of bone mineral density (BMD). Methods: We followed 97 women (mean age 58 years), for 24 months, with a history of RA of less than 12 months. At baseline, 77 women were receiving standard treatment with disease-modifying antirheumatic drugs (DMARDs) and 20 were receiving no treatment. Risk factors for osteoporosis were recorded. Disease activity score (DAS28), Health Assessment Questionnaire (HAQ) score, and medications were registered at baseline and every 6 months and calculated as areas under the curve (AUCs). Femoral neck and lumbar spine BMD were measured at baseline and after 2 years and compared to BMD in age- and gender-matched controls. Risk factors were analysed by linear regression models. Results: BMD loss was comparable to that of age-matched women in both the lumbar spine and the femoral neck, although neither was significantly different from baseline. In multivariate analyses the AUC for DAS28 was an independent predictor of changes in lumbar spine BMD (p=0.003) and that for HAQ of changes in femoral neck BMD (p=0.018). GC use was not an overall predictor of BMD loss. Conclusion: BMD loss was predicted by high disease activity and disability but not by GC treatment. With the DMARD, GC, hormone replacement therapy (HRT), and bisphosphonate treatment strategies used during the study period, the general outcome seems favourable concerning loss of BMD in patients with early RA.

    • During a 13 year period, 286 patients with cervical disc herniation and/or spondylotic spurs, were subjected to anterior decompression and vertebral interbody fusion with autologous bone. Twenty patients were re-admitted in the late postoperative period due to recurrent radicular symptoms and/or signs of myelopathy. In these patients myelography was performed again. In 14 patients spinal cord compression and/or nerve root involvement at a new level was visualized. At the operated level, however, the myelograms demonstrated a smooth anterior wall in the spinal canal. The series confirms the safety, effectiveness and reliability of the Cloward procedure in achieving long term spinal cord and nerve root decompression, and a solid vertebral interbody fusion.
