  • Variants of the interleukin-1 receptor antagonist gene are associated with fat mass in men

    • Context: Immune functions seem to have connections to variations in body fat mass. Studies of knockout mice indicate that endogenous interleukin (IL)-1 can suppress mature-onset obesity. Objective: To systematically investigate our hypotheses that single- nucleotide polymorphisms (SNPs) and/or haplotypes variants in the IL-1 gene system are associated with fat mass. Subjects: The Gothenburg osteoporosis and obesity determinants (GOOD) study is a population-based cross-sectional study of 18-20 year-old men (n = 1068), from Gothenburg, Sweden. Major findings were confirmed in elderly men (n = 3014) from the Swedish part of the osteoporotic fractures in men (MrOS) multicenter population-based study. Main Outcome Measure: The genotype distributions and their association with body fat mass in different compartments, measured with dual-energy X-ray absorptiometry (DXA). Results: Out of 15 investigated SNPs in the IL-1 receptor antagonist (IL1RN) gene, a recently identified 30 untranslated region C4T (rs4252041, minor allele frequency 4%) SNP was associated with the primary outcome total fat mass (P = 0.003) and regional fat masses, but not with lean body mass or serum IL-1 receptor 1 (IL1RN) levels. This SNP was also associated with body fat when correcting the earlier reported IL1RN_2018 T4C (rs419598) SNP (in linkage disequilibrium with a well-studied variable number tandem repeat of 86 bp). The association between rs4252041 SNP and body fat was confirmed in the older MrOS population (P = 0.03). The rs4252041 SNP was part of three haplotypes consisting of five adjacent SNPs that were identified by a sliding window approach. These haplotypes had a highly significant global association with total body fat (P < 0.001). None of the other investigated members of the IL-1 gene family displayed any SNPs that have not been described previously to be significantly associated with body fat. Conclusions: The IL1RN gene, shown to enhance obesity by suppressing IL-1 effects in experimental animals, have no previously described gene polymorphisms and haplotypes that are associated with fat, but not lean mass in two populations of men.   • Composite design for a foiling Optimist dinghy

    • In April 2017, a foiling Optimist dingy designed entirely by students, was successfully tested under standard sailing conditions in the waters outside Gothenburg. In order to achieve take of wind speeds as low as 6 m/s, a stiff and lightweight design of the dinghy and its foiling components was necessary. There have been few successful attempts to make an Optimist foil in a stable manner, as such there were no standards or recommendations available for the design. Therefore, a simulation driven structural design methodology for hydrofoils, centreboards, centreboard-to-hull connections, and necessary hull reinforcements using sandwich structures was adopted. The proposed design was then manufactured, allowing for a significantly stiffer hull and a 20% decrease in weight over a conventional Optimist. Excluding the rig and sail, the final weight came to 27 kg.

    • Digital teknik håller radikalt på att förändra vår konsumtionskultur. Appar, QR-koder och ständigt mer avancerade mobiltelefoner, samt de digitala spår användningen av dem lämnar efter sig skapar nya konsumtionsbeteenden, nya metoder för marknadsföring och försäljning av produkter, men också nya metoder för forskare att studera konsumtionsmönster. De omformar även vilka vi är som konsumenter. Att forska om den här omvandlingen innebär att undersöka tekniken i sig, de som tillverkar den, de som marknadsför den liksom de som använder den. I forskningsprojektet Digcon – Digitaliseringen av konsumtionskulturen, har forskare från Sverige och Frankrike undersökt olika aspekter av digitalisering och konsumtion. Vilka betydelser har digitala verktyg och apparater i människors vardagsliv och hur förändrar de konsumtionsmönster? Hur påverkar digitaliseringen vad det innebär att vara konsument och hur en konsument uppfattas vara. Hur omformas förståelser av kön, till exempel genom uppkomsten av modebloggar och modeappar? Vilka möjligheter till etisk konsumtion skapas genom digital teknik och vilken etik är det som skapas? I den här rapporten har vi samlat populärvetenskapliga texter från forskningsprojektet. Digcon bedrivs vid Centrum för konsumtionsvetenskap och Centre for retailing, Handelshögskolan vid Göteborgs universitet, Handelshögskolan i Stockholm och Université Tolouse II, Frankrike.

    • The PRISMA in-orbit test-bed was launched in June 2010 to demonstrate strategies and technologies for formation flying and rendezvous. OHB Sweden is the prime contractor for the project which is funded by the Swedish National Space Board (SNSB) with support from DLR, CNES, and DTU. In early September of 2011, 15 months after launch, all primary mission objectives of the PRISMA formation flying satellites had been achieved and mission success was declared. Since a significant amount of delta-V capability still remained an open call for new experiments was issued, inviting both old and new experimenters to use the capabilities of the formation. Several interested parties took the opportunity to perform their own experiments with an existing platform, each coming with new mission objectives not previously planned to be flown on the PRISMA satellites. Some of these experiments were close to what had already been achieved within the nominal mission, but some included new ways of using the formation not envisioned by the spacecraft designers. The new experiments span from data collection in specific relative orbits, with a separation from a few meters to several kilometers, to entirely new modules within the on-board software. Changing from a pre-planned technology demonstration mission to operating a commercial resource required adaptation of the original operational concept, taking into account the different levels of experience of the customers and managing the satellites between experiments. This paper describes how these new mission objectives were integrated in operations and how a sometimes very short turn-around between initial concept and experiment execution was implemented with the aid of well established validation processes, high degrees of on-board autonomy and a flexible operations team.

  • Optimus Pråm - Semi-submersible wind farm installation vessel for Blekinge Offshore

    • In Sweden the government is investing lots of resources in order to meet the energy need with clean and renewable alternatives. Since wind is an unlimited source of energy the exploitations of wind farms is of great interest. This report describes a conceptual design of an innovative offshore wind turbine installation vessel for inland sea conditions, with highest possible energy efficiency and environmental friendly performance in every detail. The customer, Blekinge Offshore, main requirement is to receive a concept with an as low installation cost as possible.The final concept Optimus Pråm includes one installation vessel, which is a semi-submersible barge, and one support vessel that supply the installation vessel with power and propulsion. The power is distributed from the support vessel to the installation vessel through a power cable. The installation vessel will be self-propelled during the installation phase using electrical motors and thrusters.The installation vessel shall be able to handle gravity foundations and fully assembled wind turbines, transported vertically to the installation site. Foundations and windmills will be fully assembled in Karlshamn port before transit to site. During transit the installation and support vessel are connected to each other in all motions except pitch. This almost total fixed connection makes the two vessels acts like one, which gives the joint vessel excellent manoeuvrability. When installing foundations, the installation vessel and the support vessel disconnect. The installation vessel places the foundation at the planned location by ballasting until the foundation has reached the seabed. After de-ballasting the installation vessel and support vessel connects again and transit back to the harbour. When installing wind turbines the two vessels are kept connected and makes highly accurate positioning for the installation possible

    • All techniques needed for proteomic analyses of plant plasma membranes are described in detail, from isolation of plasma membranes to protein identification by mass spectrometry (MS). Plasma membranes are isolated by aqueous two-phase partitioning yielding vesicles with a cytoplasmic side-in orientation and a purity of about 95%. These vesicles are turned inside-out by treatment with Brij 58, which removes soluble contaminating proteins enclosed in the vesicles as well as loosely attached proteins. The final plasma membrane preparation thus retains all integral proteins and many peripheral proteins. Proteins are separated by one-dimensional sodium dodecyl sulphate-polyacrylamide gel electrophoresis (SDS-PAGE), and protein bands are excised and digested with trypsin. Peptides in tryptic digests are separated by nanoflow liquid chromatography and either fed directly into an ESI-MS or spotted onto matrix-assisted laser desorption ionization (MALDI) plates for analysis with MALDI-MS. Finally, data processing and database searching are used for protein identification to define a plasma membrane proteome.

    • Objective: To monitor changes in serum concentrations of cartilage oligomeric matrix protein (COMP) during a 24-h period to determine any diurnal variation, and to estimate the half life of COMP in the circulation in patients with symptomatic knee osteoarthritis and in those with rheumatoid arthritis. Methods: Serum samples were drawn every 4 h (7 samples/patient over 24 h) in 10 patients with knee osteoarthritis and 14 patients with rheumatoid arthritis. Osteoarthritis was defined radiographically and clinically (American College of Rheumatology (ACR) criteria) and rheumatoid arthritis according to the 1987 ACR criteria. Serum COMP was measured by sandwich ELISA. A statistical model for the diurnal variation in the COMP levels was developed using the computer program NONMEM. Results: No considerable changes in COMP levels were observed during the day between 08:00 and 21:00 in either group. A significant decrease in serum COMP was apparent during bed rest at night, reaching the lowest levels between 04:00 and 05:00 (p < 0.03 or better v all other time points) in patients with osteoarthritis and in those with rheumatoid arthritis. From the rate of decreasing serum COMP levels, a putative half life of COMP in the circulation was estimated to be 7.4 h. Conclusion: During normal daytime activities, serum COMP levels are constant. The decrease during the night indicates a rapid elimination of COMP once it has reached the circulation. The stable COMP levels during the day suggest that it is not necessary to further standardise the time of serum sampling in clinical practice.

    • CONTEXT: Regulation of fat mass appears to be associated with immune functions. Studies of knockout mice show that endogenous interleukin (IL)-6 can suppress mature-onset obesity. OBJECTIVE: To systematically investigate associations of single nucleotide polymorphisms (SNPs) near the IL-6 (IL6) and IL-6 receptor (IL6R) genes with body fat mass, in support for our hypothesis that variants of these genes can be associated with obesity. DESIGN AND STUDY SUBJECTS: The Gothenburg Osteoporosis and Obesity Determinants (GOOD) study is a population-based cross-sectional study of 18- to 20-year-old men (n=1049), from the Gothenburg area (Sweden). Major findings were confirmed in two additional cohorts consisting of elderly men from the Osteoporotic Fractures in Men (MrOS) Sweden (n=2851) and MrOS US (n=5611) multicenter population-based studies. MAIN OUTCOME: The genotype distributions and their association with fat mass in different compartments, measured with dual-energy X-ray absorptiometry. RESULTS: Out of 18 evaluated tag SNPs near the IL6 and IL6R genes, a recently identified SNP rs10242595 G/A (minor allele frequency=29%) 3' of the IL6 gene was negatively associated with the primary outcome total body fat mass (effect size -0.11 standard deviation (s.d.) units per A allele, P=0.02). This negative association with fat mass was also confirmed in the combined MrOS Sweden and MrOS US cohorts (effect size -0.05 s.d. units per A allele, P=0.002). When all three cohorts were combined (n=8927, Caucasian subjects), rs10242595(*)A showed a negative association with total body fat mass (effect size -0.05 s.d. units per A allele, P<0.0002). Furthermore, the rs10242595(*)A was associated with low body mass index (effect size -0.03, P<0.001) and smaller regional fat masses. None of the other SNPs investigated in the GOOD study were reproducibly associated with body fat. CONCLUSIONS: The IL6 gene polymorphism rs10242595(*)A is associated with decreased fat mass in three combined cohorts of 8927 Caucasian men.

    • This study aims to investigate focal firms' business sustainability relationships in connection with their stakeholders in supply chain networks. A questionnaire survey was sent to a sample consisting of large Swedish firms, with 107 usable questionnaires returned. The results reveal which stakeholders are of interest for firms in sustainability efforts. The results are compared with earlier findings from Norway and Spain in a triangular approach. The former is a similar country, while the latter is different to Sweden in several ways. In addition, the study uncovers which stakeholders in the supply chain network should be considered. The study also demonstrates how firms can implement business sustainability in their supply chain networks and shows the extent to which different stakeholders are considered in sustainability efforts. The study contributes to sustainability research and stakeholder theory in supply chain networks. Opposite to earlier findings, this study showed only minor influences from national culture and institutions on firms' sustainable business practices in supply chain networks. © 2022 The Authors.     • This study investigates the direct and indirect effects between economic, social and environmental dimensions of triple bottom line (TBL), based on a questionnaire survey and cross-industrial sample in Sweden. The analyses apply partial least squares structural equation models. The study tests the direct and indirect effects between economic, social, and environmental dimensions of TBL and offers additional validity and reliability to establish the measurement and structural properties between the dimensions of TBL. The study extends earlier findings by explicitly discussing how the three TBL goals relate to each other and shows how the dynamic capability view can be a fruitful lens to investigate business sustainability. Some differences in sustainability business practices caused by differences in national cultures are identified. Sustainability reporting in a strong uncertainty avoidance (UA) country happens in accordance with regulations and laws. Conversely, for weak UA cultures, reporting and compliance with regulations are ways to build trust with stakeholders. That is, reporting is more transparent and widespread in weak UA countries. The study also provides a foundation to guide companies' actions of business sustainability. The model shows companies how to establish the order of actions undertaken across economic, social, and environmental dimensions. In addition, it clarifies that the economic dimension exerts an effect on the social and environmental dimensions. The model also grasps long-term economic performance by including competitiveness and brand value, while earlier research mainly has focused on more short-term measurements as return on assets. © 2022 The Authors.     • This paper presents a custom chip for linearization of RF power amplifiers using digital predistortion. The chip has been implemented in a standard digital 0.8 μm CMOS process with standard static cells and single-phase clocking. A systolic complex multiplier based on distributed arithmetic constitutes the core of the chip. The nonlinear function is realized with a look-up table containing complex gain factors applied to the complex multiplier. The maximum clock frequency was found by means of simulation to be 105 MHz corresponding to 21 Msamples/s throughput with 3 W power consumption using 5 V supply voltage. The fabricated chip is fully functional and has been measured up to 60 MHz clock frequency with 825 mW power consumption with 3.3 V supply voltage. Operation at 1.5 V supply voltage allows 10 MHz clock frequency with 35 mW power consumption

    • A fundamentally pumped millimeter wave resistive mixer based on an HFET technology working at F-band (90-140 GHz) is described for the first time. Nonlinear simulations have been performed for this mixer based on an specially designed double δ-doped pseudomorphic HFET device developed for this application. A minimum conversion loss between 12 to 13 dB was measured with the RF fixed at different frequencies between 108 to 114 GHz at an RF power of -13 dBm. Both theoretical and experimental results are presented in this paper.

    • The purpose of this study is to estimate a model of male labour supply, when agents may be subject to quantity constraints. As the data contain direct information about hours constraints, this information is used in the estimation. We also quantify the importance of such rationing for the response in the hours of work to changes in wages, income and the tax system. As it turns out, quantity constraints appear only to slightly reduce the sensitivity of the average hours of work to changes in the economic environment.

  • Bilateral Prophylactic Mastectomy in Swedish Women at High Risk of Breast Cancer: A National Survey.

    • BACKGROUND/OBJECTIVE:: This study attempted a national inventory of all bilateral prophylactic mastectomies performed in Sweden between 1995 and 2005 in high-risk women without a previous breast malignancy. The primary aim was to investigate the breast cancer incidence after surgery. Secondary aims were to describe the preoperative risk assessment, operation techniques, complications, histopathological findings, and regional differences. METHODS:: Geneticists, oncologists and surgeons performing prophylactic breast surgery were asked to identify all women eligible for inclusion in their region. The medical records were reviewed in each region and the data were analyzed centrally. The BOADICEA risk assessment model was used to calculate the number of expected/prevented breast cancers during the follow-up period. RESULTS:: A total of 223 women operated on in 8 hospitals were identified. During a mean follow-up of 6.6 years, no primary breast cancer was observed compared with 12 expected cases. However, 1 woman succumbed 9 years post mastectomy to widespread adenocarcinoma of uncertain origin. Median age at operation was 40 years. A total of 58% were BRCA1/2 mutation carriers. All but 3 women underwent breast reconstruction, 208 with implants and 12 with autologous tissue. Four small, unifocal, invasive cancers and 4 ductal carcinoma in situ were found in the mastectomy specimens. The incidence of nonbreast related complications was low (3%). Implant loss due to infection/necrosis occurred in 21 women (10%) but a majority received a new implant later. In total, 64% of the women underwent at least 1unanticipated secondary operation. CONCLUSIONS:: Bilateral prophylactic mastectomy is safe and efficacious in reducing future breast cancer in asymptomatic women at high risk. Unanticipated reoperations are common. Given the small number of patients centralization seems justified.

    • Chronic myeloid leukemia (CML) is a myeloproliferative neoplasm that, if not treated, will progress into blast crisis (BC) of either myeloid or B lymphoid phenotype. The BCR-ABL1 fusion gene, encoding a constitutively active tyrosine kinase, is thought to be sufficient to cause chronic phase (CP) CML, whereas additional genetic lesions are needed for progression into CML BC. To generate a humanized CML model, we retrovirally expressed BCR-ABL1 in the cord blood CD34(+) cells and transplanted these into NOD-SCID (non-obese diabetic/severe-combined immunodeficient) interleukin-2-receptor γ-deficient mice. In primary mice, BCR-ABL1 expression induced an inflammatory-like state in the bone marrow and spleen, and mast cells were the only myeloid lineage specifically expanded by BCR-ABL1. Upon secondary transplantation, the pronounced inflammatory phenotype was lost and mainly human mast cells and macrophages were found in the bone marrow. Moreover, a striking block at the pre-B-cell stage was observed in primary mice, resulting in an accumulation of pre-B cells. A similar block in B-cell differentiation could be confirmed in primary cells from CML patients. Hence, this humanized mouse model of CML reveals previously unexplored features of CP CML and should be useful for further studies to understand the disease pathogenesis of CML.

    • Aim: Changes in community composition resulting from environmental changes modify biotic interactions and affect the distribution and density of local populations. Such changes are currently occurring in nettle-feeding butterflies in Sweden where Araschnia levana has recently expanded its range northward and is now likely to interact with resident species (Aglais urticae and Aglais io). Butterfly occurrence data collected over years and across regions enabled us to investigate how a recent range expansion of A. levana may have affected the environmental niche of resident species.Location: We focused on two regions of Sweden (Skane and Norrstrom) where A. levana has and has not established and two time periods (2001-2006 and 2009-2012) during its establishment in Skane.Methods: We performed two distinct analyses in each region using the PCA-env and the framework described in Broennimann etal. (2012). First, we described the main sources of variation in the environment. Second, in each time period and region, we characterized the realized niches of our focal species across topographic and land use gradients. Third, we quantified overlaps and differences in realized niches between and within species over time.Results: In Skane, A. levana has stabilized its distribution over time, while the distribution of the native species has shifted. These shifts depicted a consistent pattern of avoiding overlap between the native species and the environmental space occupied by A. levana, and it was stronger for A. urticae than for A. io. In both regions, we also found evidence of niche partitioning between native species.Main conclusions: Interspecific interactions are likely to affect local species distributions. It appears that the ongoing establishment of A. levana has modified local biotic interactions and induced shifts in resident species distributions. Among the mechanisms that can explain such patterns of niche partitioning, parasitoid-driven apparent competition may play an important role in this community.

    • Radiation therapy is used to treat malignant tumors in the brain and central nervous system involvement of leukemia and lymphomas in children. However, ionizing radiation causes a number of adverse long-term side effects in the brain, including cognitive impairment. Hippocampal neurogenesis is important for place learning and has been shown to be decreased by irradiation (IR) in rats and mice. In the present study, 10-day-old male mice received 6-Gy IR to the brain on postnatal day 10. We used BrdU labeling of the granule cell layer (GCL) of the hippocampus to evaluate cell proliferation and survival. An unbiased, automated platform for monitoring of behavior in a group housing environment (IntelliCage) was used to evaluate place learning 2 months after IR. We show that cranial IR impaired place learning and reduced BrdU labeling by 50% in the GCL. Cranial IR also reduced whole body weight gain 5%. We conclude that this experimental paradigm provides a novel and time-saving model to detect differences in place learning in mice subjected to IR. This method of detecting behavioral differences can be used for further studies of adverse effects of IR on hippocampal neurogenesis and possible new strategies to ameliorate the negative effects of IR on cognition.

    • OBJECTIVE: Radiation therapy (RT) to the brain is often used in the treatment of children with different types of malignant diseases affecting the brain. However, RT in childhood may also have severe side effects including impaired brain maturation and intellectual development. For childhood cancer survivors these adverse effects of RT can cause lifelong disability and suffering. Therefore, there is an unmet need to limit late effects after RT. Precursor cells in the subgranular zone of the dentate gyrus (DG) in the hippocampus are particularly sensitive to irradiation (IR). This may be of significance as newly generated neurons in the DG are important for memory and learning. GH secretagogues (GHS) have previously been shown to promote neurogenesis and to have neuroprotective effects. In addition, several parts of the brain, including the hippocampus, have been shown to express the GHS receptor 1a (GHS-R1a). The aim of this study was to evaluate the potential effect of the GHS hexarelin on proliferation and survival of progenitor cells in the hippocampus after brain IR in a mouse model. DESIGN: In the present study, 10-day-old male mice received 6Gy cranial IR. Non-irradiated sham animals were used as controls. We treated one group of irradiated and one sham group with hexarelin (100mug/kg/day) for 28days and used immunohistochemical labeling of bromo-deoxy uridine (BrdU) and phospho-histone H3 of the granular cell layer of the DG to evaluate proliferation and cell survival after IR at postnatal day ten. RESULTS: Our results show that hexarelin significantly increased the number of BrdU-positive cells in the granule cell layer by approximately 50% compared to controls. CONCLUSION: The increased number of BrdU-positive cells in the granule cell layer suggests a partial restoration in the pool of proliferating cells by hexarelin after IR.

    • The vascular endothelium transduces physical stimuli within the circulation into physiological responses, which influence vascular remodelling and tissue homeostasis. Therefore, a new computerized biomechanical ex vivo perfusion system was developed, in which small intact vessels can be perfused under well-defined biomechanical forces. The system enables monitoring and regulation of vessel lumen diameter, shear stress, mean pressure, variable pulsatile pressure and flow profile, and diastolic reversal flow. Vessel lumen measuring technique is based on detection of the amount of flourescein over a vessel segment. A combination of flow resistances, on/off switches, and capacitances creates a wide range of pulsatile pressures and flow profiles. Accuracy of the diameter measurement was evaluated. The diameters of umbilical arteries were measured and compared with direct ultrasonographic measurement of the vessel diameter. As part of the validation the pulsatile pressure waveform was altered, e.g., in terms of pulse pressure, frequency, diastolic shape, and diastolic reversal flow. In a series of simulation experiments, the hemodynamic homeostasis functions of the system were successfully challenged by generating a wide range of vascular diameters in artificial and intact human vessels. We conclude that the system presented may serve as a methodological and technical platform when performing advanced hemodynamic stimulation protocols.

    • Endothelial cells are the main sensors of changes in the biomechanical flow environment and play a pivotal role in vascular homeostasis. An in vitro perfusion model was developed to study the regulatory effect on gene expression by different flow and pressure profiles. Human umbilical vein endothelial cells were grown to confluence inside capillary microslides or silicone tubes. Thereafter, they were exposed to different levels of shear stress or different levels of static or pulsatile pressure. Genes representing various hemostasis functions of the endothelial cells were analyzed. Shear stress was a more effortful stimulus than static or pulsatile tensile stress. Although shear stress affected mRNA expression of all six studied genes (tissue-type plasminogen activator [t-PA], plasminogen activator inhibitor [PAI]-1, Thrombomodulin [TM], urokinase-type plasminogen activator [u-PA], vascular cell adhesion molecule [VCAM-1], and endothelial nitric oxide synthase [eNOS]), none of the genes was found regulated by pressure. Shear stress down-regulated t-PA and VCAM-1 in a dose response-dependent way, and up-regulated TM. u-PA, eNOS, and PAI-1 were up-regulated by shear stress, but there was no obvious dose-response effect for these genes. These findings suggest that shear stress has a more powerful gene regulatory effect on endothelial gene expression than tensile stress. Low shear stress induced a more proatherogenic endothelial surface but preserved t-PA gene expression levels compared to high shear stress.

    • Streptococcus pyogenes Cas9 (SpCas9) and derived enzymes are widely used as genome editors, but their promiscuous nuclease activity often induces undesired mutations and chromosomal rearrangements. Several strategies for mapping off-target effects have emerged, but they suffer from limited sensitivity. To increase the detection sensitivity, we develop an off-target assessment workflow that uses Duplex Sequencing. The strategy increases sensitivity by one order of magnitude, identifying previously unknown SpCas9's off-target mutations in the humanized PCSK9 mouse model. To reduce off-target risks, we perform a bioinformatic search and identify a high-fidelity Cas9 variant of the II-B subfamily from Parasutterella secunda (PsCas9). PsCas9 shows improved specificity as compared to SpCas9 across multiple tested sites, both in vitro and in vivo, including the PCSK9 site. In the future, while PsCas9 will offer an alternative to SpCas9 for research and clinical use, the Duplex Sequencing workflow will enable a more sensitive assessment of Cas9 editing outcomes. SpCas9 unintended editing is a major concern. Here the authors report an off-target method using Duplex Sequencing with increased sensitivity for Cas9 mutation detection; they also identify a Cas9 variant of the II-B subfamily with intrinsic high fidelity (PsCas9) and see improved specificity.

    • Objective To investigate current odontological care routines for patients treated for head and neck cancers in the county councils/regions (C/Rs) of Sweden. Methods An invitation to fill in a web-based questionnaire was sent to dentists/dental hygienists working in dental clinics in the 12 C/Rs, treating and responsible for the odontological care of patients undergoing treatment for cancer of the head and neck. The questionnaire started with two mandatory and one non-mandatory questions, followed by questions regarding routines before (n = 28), during (n = 23), and after (n = 9) treatment, plus two additional questions, totalling 65 questions. Results Four dental hygienists and six dentists in 10 of the 12 C/Rs answered the questionnaire. Three C/Rs stated that they measure both the unstimulated and stimulated salivary secretion rate, and another C/R stated that they measure the stimulated secretion rate only. Similar recommendations were given regarding oral hygiene, salivary stimulants and substitutes, and extra fluoride. However, great variations were seen regarding recommendations for preventing and relieving oral mucositis. There were also discrepancies regarding information about the importance of avoiding smoking and alcohol. In seven C/Rs, patients visited the dental hygienist once a week during cancer treatment. Conclusion The results suggests that there are great variations in odontological care given to patients undergoing treatment for cancer of the head and neck region in different county councils/regions in Sweden. There is a need to develop and implement evidence-based guidelines to decrease the risk of oral complications and increase both the quality of life and the quality of care.

    • Bioenergicentrum i Halland (BEH) är ett projekt som ligger inom ramen för EU:s strukturfondsprogram. Projektet genomförs i Region Hallands regi som är regionens välfärds- och utvecklingsorganisation. Arbetet som bedrivs inom BEH syftar speciellt till att driva utvecklingen mot en ökad produktion och användning av bioenergi till biogas och i förlängningen fordonsgas. Genom att satsa på att skapa förutsättningar för innovation, kunskapsutveckling och samverkan främjas tillväxt och hållbar utveckling. Vid naturbruksgymnasiet i Plönninge utanför Halmstad finns idag en biogasanläggning som beskickas med bl.a. nötgödsel och matavfall. Dessutom finns en mindre pilotanläggning som är tänkt att fungera som en del av test- och verifieringsanläggning som BEH vill bygga upp i Plönninge. Som ett led i att utveckla dessa anläggningar och kunna erbjuda möjligheten till kunskapsinsamling genomfördes projektet som beskrivs i denna rapport. Uppdraget var att genomföra försöksrötningar på labb, använda resultaten för att skapa en modell som sedan kan nyttjas som ett verktyg i det inledande arbetet med att investera i en biogasanläggning som beskickas med lantbruksbaserade substrat. Högskolan i Halmstad (HH) genomförde försöksrötningarna och Grontmij (GM) använde sedan resultaten för att skapa en modell där bl.a. substrat, förbehandlingsteknik och driftkostnader finns med. Sammanfattningsvis kan sägas att majs som substrat fungerar bäst med de valda förbehandlingsmetoderna; kemisk behandling, termisk behandling och ultraljudsbehandling. Alla förbehandlingsmetoder med majs som substrat visade på ett positivt resultat, d.v.s. det ökade gasutbytet och dess värde (kr/kWh) översteg kostnaderna för de olika förbehandlingarna. Vad som måste beaktas är att produktionskostnaderna överlag är höga, med och utan förbehandling. Modellen har konstruerats på ett sådant sätt att den ska vara användarvänlig och med möjlighet att enkelt lägga till ytterligare substrat och förbehandlingsmetoder. Upprepningar av de försöksrötningar som genomförts kommer att öka tillförlitligheten hos modellen. Den fungerar som ett verktyg i att beräkna investeringsmarginalen för förbehandlingsutrustningen baserat på det valda substratet. På detta vis kan intressenter få en första indikation på om det är ekonomiskt rimligt att gå vidare med det tänkta substratet, den valda förbehandlingsmetoden, de planerade mängderna substrat etc. En investeringskalkyl har tagits fram för en gårdsanläggning som hanterar 5 000 ton substrat eller gödsel årligen. Det motsvarar 2-3 stycken medelstora mjölkgårdar. Kalkylen är översiktlig och syftar till att ge en första indikation på kostnader för de stora komponenterna såsom substratlager, rötkammare och rötrestlager. Kringarbeten såsom utredningar, markarbeten och geoundersökningar är inte med i kalkylen då dessa omkostnader till stor del avgörs av lokalisering och de förutsättningar som finns på platsen redan från start. Generellt kan dock sägas att den absolut billigaste och enklaste gårdsbaserade biogasanläggningen innebär en investering på 2,7-4 MSEK för flytgödsel från 100-300 mjölkkor. För BEH är det viktigt att skapa en plattform där intressenter kan komma för att genomföra försöksrötningar, byta erfarenheter och samla kunskap. För att uppnå detta är det nödvändigt att kunna erbjuda kunden kompletta och kompetenta lösningar på en och samma plats. Detta innebär ett erbjudande som innefattar försöksrötningar på labb-, pilot och fullskala. Ett förslag på konstruktion av pilotanläggning med övergripande principskiss ingår i denna rapport och fungerar som ett inledande arbete i projekteringen av en större pilotanläggning. Nödvändiga driftanalyser av rötrest ska kunna göras på plats i Plönninge på laboratoriet; analyser såsom enskilda organiska syror ska kunna skickas till lämpligt laboratorium. Personal ska kunna tillhandahållas för att driva och optimera rötningen enligt kundens syften och önskemål. På detta vis fungerar Region Halland som en länk mellan teori ochpraktik, mellan liten och stor skala och mellan aktörer från olika discipliner och geografiska områden.

    • We investigated the effects of ionizing radiation on microvessel structure and complexity in the hippocampus. We also assessed neurogenesis and the neurovascular niche. Postnatal day 14 male C57BL/6 mice received a single dose of 8Gy to the whole brain and were killed 6 hours, 1 week, 7 weeks, or 1 year later. Irradiation decreased the total number of microvessels and branching points from 1 week onwards and decreased the total microvessel area 1 and 7 weeks after irradiation. After an initial increase in vascular parameter densities, concomitant with reduced growth of the hippocampus, the densities normalized with time, presumably adapting to the needs of the surrounding nonvascular tissue. Irradiation decreased the number of neural stem and progenitor cells in the hippocampus. The relative loss increased with time, resulting in almost completely ablated neurogenesis (DCX(+) cells) 1 year after irradiation (77% decreased 1 week, 86% decreased 7 weeks, and 98% decreased 1 year after irradiation compared with controls). After irradiation, the distance between undifferentiated stem cells and microvessels was unaffected, and very few dying endothelial cells were detected.     • The purpose of this study was to evaluate calf muscle endurance in a seated position 3 months after an Achilles tendon rupture and to evaluate how the ability to perform standardized seated heel-rises correlated to the single-leg standing heel-rise test and to patient-reported symptoms evaluated with the Achilles tendon Total Rupture Score (ATRS) 3 and 6 months after the injury. Ninety-three patients were included from a cohort of 101 patients participating in a prospective, randomized controlled trial comparing surgical and nonsurgical treatment after Achilles tendon rupture. Forty-seven patients were treated surgically and 46 nonsurgically. Ninety-one patients out of 93 (98%) could perform the standardized seated heel-rises. At the 3-month follow-up, there was a significant difference (P<0.001) between the injured and the healthy side performing standardized seated heel-rises. There were also significant correlations (r=0.29-0.37, P=<0.05) between the standardized seated heel-rises and ATRS 3 and 6 months after injury in the group who could not perform single-leg standing heel-rises. There were no significant differences between the surgical and nonsurgical treatment groups. The evaluation of standardized seated heel-rises appears to be a useful tool to quantify progress and predict future functional performance and patient-reported symptoms.

    • Maritime transports are to be regarded as a self-organized ecosystem (Kay et al., 1999) characterized by sub-optimization where historically each actor to has optimized its own operations, often giving rise to inefficiencies as a whole. In recent years however, digital transformation has challenged this by providing means for enhanced transparency in data sharing and situational awareness, enabling better coordination and improved efficiency on the whole (Lind et al. 2018a). Digital transformation drives the possibilities of creating new value by enabling higher degrees of connectivity between actors, digitally twin physical objects, drawing patterns of behaviour based on extensive sets of historical data, as well as harmonizing data sharing through standardized interfaces and communication protocols (e.g. Almirall and Casadesus-Masanell 2010; Gassman et al. 2010; Lakhani et al. 2006). To break existing patterns of behaviour and to avoid the creation of proprietary solutions that feed sub-optimization, there is a need for new inspiration and perspectives that capitalize on the opportunities that digital transformation provides. From an open innovation point of view, this means that innovators both having experience from the sector as well as from other sectors would come together, come up with, and provide new applications not previously possible or never thought about before. A core capability that the ecosystem needs to develop and ensure is data streams made accessible for those that can provide new applications aimed for the single actor and/or clusters of actors, within or outside the maritime sector (Lind et al. 2018).This has also been one of the objectives for Port Collaborative Decision Making (PortCDM), which is a concept that provides guidelines and standards for the data exchange within and between ports, between ships and ports, and between ports and hinterland operators (Lind et al 2018). Such data exchange is necessary if enhanced efficiency during port call operations is to be achieved but also facilitates open innovation within the maritime sector. In order to realise that potential, a purposive transfer of knowledge between the established actors and potential new service providers has to be established (Chesbrough 2006). We therefore set out to explore How can open innovation intermediaries accelerate acquisition in an ecosystem through the management and throughput of knowledge transfer?We address the question through a longitudinal study by applying an action research approach involving actors from the local port and students from three bachelor programs. Before we describe the specifics of the research methodology, we outline our theoretical framework in terms of how knowledge transfer can be framed within an open innovation ecosystem. After the research methodology we detail the five iterations and then discuss the effect on knowledge transfer within the ecosystem. Finally, we conclude and give directions on future research.

Endoscopically assisted free semitendinosus transfer restores ankle and preserves first metatarsophalangeal joint (MTPJ) function.Indications:The main indication for the procedure is symptomatic chronic rupture of the Achilles tendon with a palpable tendon gap.Technique Description:The procedure can be split into 4 stages: graft harvest, calcaneum and tunnel preparation, proximal graft attachment, and finally graft passage and screw insertion.Discussion/Conclusion:Following reconstruction, patients use a cast in full plantar flexion for 2 weeks, then a graduated walker for full weight-bearing.

    • Background Rupture of the Achilles tendon may result in reduced functional activity and reduced plantar flexion strength. These changes may arise from elongation of the Achilles tendon. An observational study was performed to quantify the Achilles tendon resting angle (ATRA) in patients following Achilles tendon rupture, surgical repair, and rehabilitation, respectively. Methods Between May 2012 and January 2013, 26 consecutive patients (17 men), with a mean (standard deviation, SD) age of 42 (8) years were included and evaluated following injury, repair, and at 6 weeks, 3 months, 6 months, 9 months, and 12 months, respectively (rehabilitation period). The outcome was measured using the ATRA, Achilles tendon total rupture score (ATRS), and heel-rise test. Results Following rupture, the mean (SD) absolute ATRA was 55 (8)° for the injured side compared with 43 (7)°(p < 0.001) for the noninjured side. Immediately after repair, the angle reduced to 37 (9)°(p < 0.001). The difference between the injured and noninjured sides, the relative ATRA, was -12.5 (4.3)°following injury; this was reduced to 7 (7.9)°following surgery (p < 0.001). During initial rehabilitation, at the 6-week time point, the relative ATRA was 2.6 (6.2)°(p = 0.04) and at 3 months it was -6.5 (6.5)°(p < 0.001). After the 3-month time point, there were no significant changes in the resting angle. The ATRS improved significantly (p < 0.001) during each period up to 9 months following surgery, where a score of 85 (10)°was reported. The heel-rise limb symmetry index was 66 (22)% at 9 months and 82 (14)% at 12 months. At 3 months and 6 months, the absolute ATRA correlated with the ATRS (r = 0.63, p = 0.001, N = 26 and r = 0.46, p = 0.027, N = 23, respectively). At 12 months, the absolute ATRA correlated with the heel-rise height (r = -0.63, p = 0.002, N = 22). Conclusion The ATRA increases following injury, is reduced by surgery, and then increases again during initial rehabilitation. The angle also correlates with patient-reported symptoms early in the rehabilitation phase and with heel-rise height after 1 year.     • The microstructure response of laser-powder bed fusion (L-PBF)-processed ferritic stainless steel (AISI 441) during post-heat treatments is studied in detail. Focus is on the precipitation kinetics of the Nb-rich phases: Laves (Fe2Nb) and the cubic carbo-nitride (NbC), as well as the grain structure evolution. The evolution of the precipitates is characterized using scanning and transmission electron microscopy (SEM and TEM) and the experimental results are used to calibrate precipitation kinetics simulations using the precipitation module (TC-PRISMA) within the Thermo-Calc Software package. The calculations reproduce the main trend for both the mean radii for the Laves phase and the NbC, and the amount of Laves phase, as a function of temperature. The calibrated model can be used to optimize the post-heat treatment of additively manufactured ferritic stainless steel components and offer a creator tool for process and structure linkages in an integrated computational materials engineering (ICME) framework for alloy and process development of additively manufactured ferritic steels.

    • Purpose: To report the clinical experience and results of using a microsurgical technique to decompress the arteriovenous connection in complicated branch retinal vein occlusion (BRVO) combined with haemorrhage, oedema and ischaemia.Methods: We carried out a retrospective, non-randomized, interventional case study of the surgical sheathotomy decompression procedure. We enrolled 12 patients (seven women, five men; median age 64 years) with BRVO and decreased visual acuity (VA) caused by haemorrhage, oedema and ischaemia. The mean duration of thrombosis was 7 months (2–15 months). The patients were examined for pre- and postoperative best corrected VA (BCVA), intraocular pressure (IOP) and fundus photography. Ten patients were examined with fluorescein angiography and eight with ocular coherence tomography (OCT). Postoperative progression of cataract was recorded, as were other complications. The mean follow-up time was 20 months (8–39 months).Results: Best corrected VA had improved in nine patients, was unchanged in one patient and had deteriorated in two patients at the last follow-up. Noted complications were venous haemorrhage at surgery in five patients, retinal detachment in one patient and progression of cataract in four patients.Conclusions: Microsurgical treatment with sheathotomy of BRVO is a technically feasible procedure with few complications. Postoperative increased reperfusion could explain the resolution of macular haemorrhage, oedema and ischaemia, and may improve visual function in patients with this common vascular eye disease.

    • Interleukin-4 (IL-4) contains three structurally important intramolecular disulfides that are required for the bioactivity of the cytokine. We show that the cell surface of HeLa cells and endotoxin-activated monocytes can reduce IL-4 intramolecular disulfides in the extracellular space and inhibit binding of IL-4 to the IL-4R alpha receptor. IL-4 disulfides were in vitro reduced by thioredoxin 1 (Trx1) and protein disulfide isomerase (PDI). Reduction of IL-4 disulfides by the cell surface of HeLa cells was inhibited by auranofin, an inhibitor of thioredoxin reductase that is an electron donor to both Trx1 and PDI. Both Trx1 and PDI have been shown to be located at the cell surface and our data suggests that these enzymes are involved in catalyzing reduction of IL-4 disulfides. The pro-drug N-acetylcysteine (NAC) that promotes T-helper type 1 responses was also shown to mediate the reduction of IL-4 disulfides. Our data provides evidence for a novel redox dependent pathway for regulation of cytokine activity by extracellular reduction of intramolecular disulfides at the cell surface by members of the thioredoxin enzyme family.

    • ObjectivesThe aim of this study was to investigate the potential correlation between muscle mass/muscle quality and risk of complications or recurrence in patients presenting with acute uncomplicated diverticulitis. It was also to study if low muscle mass/quality correlated to prolonged hospital stay.Materials and methodsThe study population comprised 501 patients admitted to Helsingborg Hospital or Skåne University Hospital between 1 January 2015 and 31 December 2017, who had been diagnosed with acute uncomplicated diverticulitis and undergone computed tomography upon admission. The scans were used to estimate skeletal muscle mass and muscle radiation attenuation (an indicator for muscle quality). Skeletal muscle index was obtained by adjusting skeletal muscle mass to the patients’ height. Values of below the fifth percentile of a normal population were considered low.ResultsThere were no differences between the patients with normal versus those with low skeletal muscle mass, skeletal muscle index or muscle radiation attenuation regarding risk of complications or recurrence of diverticular disease. However, as only 11 patients had complications, no conclusion as to a potential correlation can be made. Low muscle quality correlated to longer hospital stay, also when adjusting for other potential confounders.ConclusionsMuscle mass/quality do not seem to serve as predictor of risk for recurrent disease in patients with acute uncomplicated diverticulitis. However, low muscle radiation attenuation was associated with prolonged hospital stay. This indicates that muscle quality, assessed by computed tomography scan, might be used in clinical practise to identify patients at risk of longer hospitalisation.

    • Achilles tendon rupture occurs relatively common (De Jonge, Br J Sports Med 45:1026-1028, 2011). Although it is the thickest and strongest tendon in the human body, it remains susceptible to injury. During the last decades, the incidence of spontaneous ruptures has been rising, which may be due to the increasing keep-fit culture. Ruptures occur most frequently in patients between the age of 30 and 50 years old, with a male predominance. Approximately 75 % of Achilles tendon ruptures occur during sports activities, especially racket games, soccer, and handball. Diagnosis is primarily clinical. However, there is still a lack of consensus on the best management of the Achilles tendon rupture. Generally, open surgical management is advocated, although over the past few years, percutaneous techniques are performed more commonly. Nonsurgical management appears to be a good alternative for those with comorbidity or patients who do not wish to have surgery. Recent systematic reviews have concluded that operative management has a lower re-rupture rate but must be balanced by the risks associated with surgery (Khan and Carey Smith, Cochrane Database Syst Rev CD003674, 2010). © Springer-Verlag Berlin Heidelberg 2012, 2015, All Rights Reserved.

    • Energy efficient cultivation is the major bottleneck for microalgal biomass production on a large scale and considered very difficult to attain at northern latitudes. In this study an unconventional method for industrial microalgae cultivation for bio-oil production using pulp and paper mill waste resources while harvesting only once a year was performed, mainly in order to investigate the energy efficiency of the process. Algae were cultivated for three months in 2014 in covered pond systems with access to flue gas and waste heat from the industry, and the biomass was recovered as thick sediment sludge after dewatering. The cultivation systems, designed to manage the waste resources, reached a promising photosynthetic efficiency of at most 1.1%, a net energy ratio (NER) of 0.25, and a projected year-round energy biomass yield per area 5.2 times higher than corresponding rapeseed production at the location. Thus, microalgae cultivation was, for the first time, proven energy efficient in a cold continental climate. Energy-rich indigenous communities quickly out-competed the oleaginous monocultures used for inoculation. The recovered biomass had higher heating values of 20–23 MJ kg− 1 and contained 14–19% oil dominated by C16 followed by C18 fatty acids. The cultivation season at 59°15′N, 14°18′E was projected to be efficient for 10 months and waste heat drying of the biomass is suggested for two winter months. The technique is proposed for carbon sequestering and energy storage in the form of microalgal sludge or dry matter for later conversion into biochemicals.

    • Background and objective: Multiple chronic conditions and recurring acute illness are frequent among elderly people. One such condition is atrial fibrillation (AF), which increases the risk of stroke up to fivefold. The aim of this study was to investigate the prevalence of AF among hospitalized frail elderly patients, their use of anticoagulation and their 12-month outcomes. Patients and methods: This was a clinical observational study of acutely hospitalized frail patients over the age of 75 years. The CHA2DS2-VASc Score was used to evaluate ischemic stroke risk in patients with AF. Clinically relevant outcomes were the composite of ischemic stroke and/or bleeding within 12 months, which was considered as primary in the analysis, ischemic stroke/transient ischemic attack (TIA), mortality, bleeding and hospital care consumption. Students (test, Fishers exact test, Mann Whitney U test and a Cox proportional hazards model were used for the analyses. Results: The prevalence of AF was 47%, and 63% of them were prescribed an anticoagulant. AF patients without anticoagulation were older, more often females, more often in residential care, and they had worse Mini Nutritional Assessment and activities of daily living scores. Of the patients without anticoagulation, 56% had a documented contraindication. In univariate analysis, there were significantly more events among AF patients without anticoagulation regarding the composite outcome of ischemic stroke and/or bleeding (hazard ratio [1112] 3.65, 95% CI = 1.70-7.86; p amp;lt; 0.001). When adjusting for potential confounders in Cox regression analysis, the difference remained significant (HR 4.54, 95% CI = 1.83-11.25; p = 0.001). Conclusion: The prevalence of AF in a hospitalized frail elderly population was 47%. Of these, 63% were prescribed anticoagulation therapy. Almost half of the patients without stroke pro-phylaxis had no documented contraindication. At 1 year, there were significantly more events in terms of ischemic stroke and/or bleeding among AF patients without anticoagulation therapy than among those with.

    • Den integrerade kustfiskövervakningen i Fjällbacka har pågått under oktobermånad sedan år 1989 och under augusti sedan 1998 (med uppehåll 1999). Övervakningen visar följande tillstånd och förändringar för fiskens status och miljögiftsbelastning:• Omgivningsfaktorer såsom siktdjup och temperatur har inte förändrats i Fjällbacka sedan undersökningarna startade. Däremot ses en ökning i de temperaturmätningar som gjorts i området under maj-oktober sedan 1989.• Fisksamhället domineras av torsk, rötsimpa, gråsej och vitling. Antalet arter i fångsten har ökat i oktoberfisket sedan provfisket startade. Bland de fångade arterna klassas gulål, lyrtorsk, sjurygg, torsk, tånglake och vitling som rödlistade av ArtDatabanken.• Signifikanta nedgångar har noterats för den kallvattengynnade arten tånglake och även för gulål och rödspotta under oktoberfisket. Oxsimpa och svart smörbult ökar på lång sikt i fångsterna under oktober. I oktoberfisket är den enda signifikanta förändringen att berggyltan ökat sedan provfisket startade.• Det har inte setts någon förändring i diversiteten i fisksamhället varken under augusti eller oktober. Den trofiska medelnivån är lägre i oktober än i augusti, men inte vid något av fiskena ses någon signifikant trend. Fångsten av stora fiskar (större än 30 cm) i oktober har minskat sedan provfisket startade.• Fångsten av tånglake var år 2014 bland de lägsta sedan provfisket startade, både under fisket i augusti och i oktober. Detta är en stor nedgång sedan året innan, då båda månadernas fisken noterade bland de högsta fångsterna av tånglake.• Tånglaken är en stationär art som föder levande yngel. Dess ekologi gör den därmed lämplig som miljöindikator. De fångade tånglakehonorna har med tiden blivit kortare, lättare och fått sämre kondition. Andelen honor med yngel som dött har ökat med tiden. Resultaten av undersökningarna indikerar att den kallvattenslevande tånglaken i Fjällbacka utsätts för stress på grund av ökande vattentemperatur.• Allt fler hälsovariabler hos tånglake i Fjällbacka uppvisar signifikanta tidstrender som tyder på att de exponeras för kemiska ämnen som påverkar olika fysiologiska funktioner. Tydliga förändringar är inducerat avgiftningssystem, stimulerat immunförsvar, en tendens till ökad oxidativ stress, samt en påverkad jonreglering hos fisken.• Förändringarna hos tånglake i Fjällbacka stämmer väl överens med den mångfacetterade symptombild som även ses hos abborrar respektive tånglakar i andra kustreferensområden (Holmön, Torhamn och Kvädöfjärden). Den komplexa symptombilden pekar på att det sannolikt är fråga om samverkanseffekter av flera olika kemiska ämnen och andra faktorer.Sveriges lantbruksuniversitet, Institutionen för akvatiska resurser. Öregrund 2015.• Halterna av bly, kvicksilver och HCH:er i tånglake har minskat signifikant under övervakningsperioden. Flera av de andra organiska miljögifterna (PCB, HCB och DDT) håller sig däremot på en stabil nivå hos tånglake i Fjällbacka medan halterna i Östersjön generellt har minskat till följd av totalförbud av ämnena. Halterna av kadmium uppvisar ingen trend över tidsperioden men ligger generellt lägre jämfört med tånglake från Östersjön. Samtliga av de undersökta ämnena ligger under deras respektive satta gränsvärden, dock ligger halten kvicksilver väldigt nära gränsvärdet.

    • Kustområdet vid Holmön i Västerbotten är sedan slutet av 1980-talet ett nationellt referensområde för Bottenviken. Här bedrivs årligen en omfattande och integrerad kustfiskövervakning i syfte att kartlägga fiskbeståndens status, fiskens hälsotillstånd och miljögiftsbelastning. Detta faktablad presenterar följande resultat och bedömningar från den integrerade kustfiskövervakningen vid Holmön under tidsperioden 1989-2014:• Omgivningsfaktorer såsom siktdjup och temperatur har inte förändrats vid Holmön sedan undersökningarna startades, även om 2000-talet inleddes med några varma somrar.• Fisksamhället, som domineras av abborre och mört, tycks gå mot ett mer karpfiskdominerat stadium. Den mest sannolika förklaringen till ökningen av karpfiskar är en ökad vattentemperatur, minskad salthalt och möjligen en påverkan av övergödning.• Den kraftiga ökningen i individtillväxt hos abborre som varit konsekvensen av gynnsamma vattentemperaturer under 2000-talets början har nu avtagit.• Tånglake och lake, som återfunnits i fångsten, är rödlistade av ArtDatabanken.• Vid Holmön uppnår de yngelbärande tånglakehonorna relativt sett högre ålder, men har en långsammare tillväxttakt än i andra referensområden i Egentliga Östersjön och i Västerhavet.• Missbildade eller döda tånglakeyngel förkommer mycket sällan vid Holmön och inga tidstrender har noterats, vare sig för frekvensen av yngel eller andelen honor med missbildade eller döda yngel. Trots att parametrarna gonadvikt och embryosomatiskt index uppvisar vikande trender, så tyder den goda yngelstatusen på goda förutsättningar för tånglaken vid Holmön.• Med tiden uppvisar allt fler hälsovariabler hos honabborrar vid Holmön signifikanta tidstrender eller starka tendenser till förändringar som tyder på att de sannolikt exponeras för kemiska ämnen som påverkar olika fysiologiska funktioner. Tydliga förändringar är inducerat avgiftningssystem, förminskade könskörtlar med färre ägg och ökad GR-aktivitet i levern, vilket tyder på ökad oxidativ stress. Dessutom noteras en påverkad kalciumreglering och förändrad rödablodcellsbild. Hos hanfiskar noteras en signifikant ökning av leverstorleken (LSI) och en minskning av hematokritvärdet, samt minskad kloridkoncentration i blodet.• Förändringarna hos abborrar vid Holmön stämmer väl överens med den mångfacetterade symptombild som även ses hos abborrar respektive tånglakar i andra kustreferensområden (Kvädöfjärden, Torhamn och Fjällbacka). Den komplexa symptombilden pekar på att det sannolikt är fråga om samverkanseffekter av flera olika kemiska ämnen.• De flesta organiska miljögifter och metaller som övervakas uppvisar dock minskande halter i abborre och tånglake över hela tidsperioden. Undantag är kadmium som ökat mellan 1995 och 2007 i tånglake, och kadmium och kvicksilver, som inte visar någon minskande trend hos abborre samt PCB och HCB i abborre, som planat ut respektive ökar under de senaste tio åren. I tånglake ses ingen tydlig minskning av organiska miljögifter som DDT och PCB, medan α-HCH minskar sedan 1995.Sammanvägd bedömning av tillståndet för kustfisk vid HolmönDen integrerade kustfiskövervakningen visar för varje år ett alltmer påverkat hälsotillstånd hos abborre trots att de flesta analyserade metaller och organiska miljögifter visar nedåtgående trender eller oförändrade halter. Den tydliga och successiva påverkan som ses på hälsotillståndet på individnivå har ännu inte resulterat i några påtagliga förändringar på bestånds- eller samhällsnivå. Den individuella tillväxttakten hos abborre har ökat, något som tyder på i övrigt gynnsamma förhållanden för abborrar i området. Hos tånglake fluktuerar halterna av miljögifter mellan åren, men artens tillstånd vid Holmön bedöms som stabilt.Det är angeläget att klarlägga om det är okända miljögifter, kända miljögifter som inte övervakas idag, eller andra bakomliggande miljöfaktorer som orsakar förändringarna i kustfiskars hälsotillstånd, och om eventuella följdeffekter kan spåras på populationsnivå.
