    • Introduction: Neural correlates of sleep deprivation are not fully understood and the difference between young and older adults in this regard has received little attention. We aimed to investigate the effect of partial sleep deprivation on resting state connectivity.Methods: 30 younger (20–30 years) and 23 older (65–75 years) healthy participants underwent MR imaging after normal sleep and partial sleep deprivation (3 h sleep). We acquired two runs of eyes-open resting state functional magnetic resonance images. Participants were monitored with eye-tracking to ensure their eyes remained open during scanning.Results: Global signal variability, defined as log-transformed standard deviation of average gray matter signal, was increased following partial sleep deprivation (0.16 [0.07, 0.24], p = 0.0004). In contrast to previous studies, we did not find that partial sleep deprivation inhibited connectivity in the default mode network, nor in other major networks investigated.Conclusion: Sleep deprivation caused increased global signal variability. This novel finding should be confirmed using independent data. Our finding of no difference in default mode connectivity in the sleep deprived state, could possibly be due to stricter monitoring of participants’ wakefulness compared to some earlier studies.

    • Although the literature claims that shift work is harmful, it overlooks the fact that that many shift workers are satisfied and stay healthy. There is little knowledge of the biological mechanisms mediating the differences in susceptibility. The present study compared satisfied and dissatisfied shift workers with respect to major anabolic and catabolic hormones. Forty-two male shift workers, with an extremely rapidly rotating shift schedule, were divided into two groups according to their ratings of satisfaction with their work hours. Morning blood samples were taken during the first and last morning shift in the shift cycle. Serum was analyzed with respect to testosterone, cortisol, and prolactin. Dissatisfied shift workers had lower morning testosterone than satisfied ones, but they did not significantly differ with respect to cortisol or prolactin. Low testosterone levels were, in addition, associated with a greater sleep need, disturbed sleep/wakefulness, and an increased need for recovery after the work period, the latter being the best predictor of testosterone levels. The only change across the shift cycle concerned a significant decrease of morning cortisol at the end of the shift cycle. High morning cortisol was related to having a morning personality and fewer sleep problems before the morning shift. Dissatisfaction with the shift system seems related to lower testosterone levels, which in turn are related to disturbed sleep/wakefulness and increased need for sleep and recovery. Furthermore, morning cortisol was reduced across a shift cycle. It is suggested that reduced testosterone levels may be part of a mechanism of shift work maladjustment.

    • Subjective sleepiness at different times is often measured in studies on sleep loss, night work, or drug effects. However, the context at the time of rating may influence results. The present study examined sleepiness throughout the day at hourly intervals and during controlled activities [reading, writing, walking, social interaction (discussion), etc.] by 10-min. intervals for 3 hr. This was done on a normal working day preceded by a scheduled early rising (to invite sleepiness) for six subjects. Analysis showed a significant U-shaped pattern across the day with peaks in the early morning and late evening. A walk and social interaction were associated with low sleepiness, compared to sedentary and quiet office work. None of this was visible in the hourly ratings. There was also a pronounced afternoon increase in sleepiness, that was not observable with hourly ratings. It was concluded that there are large variations in sleepiness related to time of day and also to context and that sparse sampling of subjective sleepiness may miss much of this variation.

  • How does work impact daily sleep quality? A within-individual study using actigraphy and self-reports over the retirement transition

    • This study examined how the cessation of work at retirement affects daily measures of actigraphy-measured and self-rated sleep quality. Time in bed or asleep and stress at bedtime were examined as potential mechanisms. In total 117 employed participants who were aged 60-72 years and planned to retire soon were recruited to the Swedish Retirement Study. Sleep quality was measured in a baseline week using accelerometers, diaries, and questionnaires. Subjective sleep measures were sleep quality, restless sleep, restorative sleep, getting enough sleep, estimated wake after sleep onset, difficulties falling asleep, too early final awakening, and difficulties waking up. Actigraphy measures were sleep efficiency, wake after sleep onset, and average awakening length. After 1 and 2 years, the measurements were repeated for the now retired participants. Daily variations in sleep quality before and after retirement were analysed using multilevel modelling, with time in bed or asleep and stress at bedtime as potential mediators. We found that several self-reports of sleep improved (e.g., +0.2 standard deviations for sleep quality and +0.5 standard deviations for restorative sleep) while objective sleep quality remained unchanged or decreased slightly with retirement (e.g., -0.8% for sleep efficiency). Increased time in bed or asleep and stress at bedtime accounted partially for the improvements in self-rated sleep quality at retirement. In conclusion, actigraph-measured and self-reported sleep quality do not change in concert at retirement, highlighting the interest of studying both outcomes. The main effects of retirement from work concern subjective experiences of recovery more than sleep quality per se.
