    • TandEM was proposed as an L-class (large) mission in response to ESA's Cosmic Vision 2015-2025 Call, and accepted for further studies, with the goal of exploring Titan and Enceladus. The mission concept is to perform in situ investigations of two worlds tied together by location and properties, whose remarkable natures have been partly revealed by the ongoing Cassini-Huygens mission. These bodies still hold mysteries requiring a complete exploration using a variety of vehicles and instruments. TandEM is an ambitious mission because its targets are two of the most exciting and challenging bodies in the Solar System. It is designed to build on but exceed the scientific and technological accomplishments of the Cassini-Huygens mission, exploring Titan and Enceladus in ways that are not currently possible (full close-up and in situ coverage over long periods of time). In the current mission architecture, TandEM proposes to deliver two medium-sized spacecraft to the Saturnian system. One spacecraft would be an orbiter with a large host of instruments which would perform several Enceladus flybys and deliver penetrators to its surface before going into a dedicated orbit around Titan alone, while the other spacecraft would carry the Titan in situ investigation components, i.e. a hot-air balloon (MontgolfiSre) and possibly several landing probes to be delivered through the atmosphere.

    • Saturn's moon Rhea had been considered massive enough to retain a thin, externally generated atmosphere capable of locally affecting Saturn's magnetosphere. The Cassini spacecraft's in situ observations reveal that energetic electrons are depleted in the moon's vicinity. The absence of a substantial exosphere implies that Rhea's magnetospheric interaction region, rather than being exclusively induced by sputtered gas and its products, likely contains solid material that can absorb magnetospheric particles. Combined observations from several instruments suggest that this material is in the form of grains and boulders up to several decimetres in size and orbits Rhea as an equatorial debris disk. Within this disk may reside denser, discrete rings or arcs of material.

  • Multi-omics for studying and understanding polar life

    • Polar ecosystems are experiencing amongst the most rapid rates of regional warming on Earth. Here, we discuss ‘omics’ approaches to investigate polar biodiversity, including the current state of the art, future perspectives and recommendations. We propose a community road map to generate and more fully exploit multi-omics data from polar organisms. These data are needed for the comprehensive evaluation of polar biodiversity and to reveal how life evolved and adapted to permanently cold environments with extreme seasonality. We argue that concerted action is required to mitigate the impact of warming on polar ecosystems via conservation efforts, to sustainably manage these unique habitats and their ecosystem services, and for the sustainable bioprospecting of novel genes and compounds for societal gain.

  • ULF Wave Transmission Across Collisionless Shocks : 2.5D Local Hybrid Simulations

    • We study the interaction of upstream ultralow frequency (ULF) waves with collisionless shocks by analyzing the outputs of 11 2D local hybrid simulation runs. Our simulated shocks have Alfvenic Mach numbers between 4.29 and 7.42 and their theta BN angles are 15 degrees, 30 degrees, 45 degrees, and 50 degrees. The ULF wave foreshocks develop upstream of all of them. The wavelength and the amplitude of the upstream waves exhibit a complex dependence on the shock's MA and theta BN. The wavelength positively correlates with both parameters, with the dependence on theta BN being much stronger. The amplitude of the ULF waves is proportional to the product of the reflected beam velocity and density, which also depend on MA and theta BN. The interaction of the ULF waves with the shock causes large-scale (several tens of upstream ion inertial lengths) shock rippling. The properties of the shock ripples are related to the ULF wave properties, namely their wavelength and amplitude. In turn, the ripples have a large impact on the ULF wave transmission across the shock because they change local shock properties (theta BN, strength), so that different sections of the same ULF wavefront encounter shock with different characteristics. Downstream fluctuations do not resemble the upstream waves in terms the wavefront extension, orientation or their wavelength. However, some features are conserved in the Fourier spectra of downstream compressive waves that present a bump or flattening at wavelengths approximately corresponding to those of the upstream ULF waves. In the transverse downstream spectra, these features are weaker.

  • Biomarker-Based Prediction of Recurrent Ischemic Events in Patients With Acute Coronary Syndromes

    • BACKGROUND: In patients with acute coronary syndrome (ACS), there is residual and variable risk of recurrent ischemic events.OBJECTIVES: This study aimed to develop biomarker-based prediction models for 1-year risk of cardiovascular (CV) death and myocardial infarction (MI) in patients with ACS undergoing percutaneous coronary intervention.METHODS: We included 10,713 patients from the PLATO (A Comparison of Ticagrelor [AZD6140] and Clopidogrel in Patients With Acute Coronary Syndrome) trial in the development cohort and externally validated in 3,508 patients from the TRACER (Thrombin Receptor Antagonist for Clinical Event Reduction in Acute Coronary Syndrome) trial. Variables contributing to risk of CV death/MI were assessed using Cox regression models, and a score was derived using subsets of variables approximating the full model.RESULTS: There were 632 and 190 episodes of CV death/MI in the development and validation cohorts. The most important predictors of CV death/MI were the biomarkers, growth differentiation factor 15, and N-terminal pro-B-type natriuretic peptide, which had greater prognostic value than all candidate variables. The final model included 8 items: age (A), biomarkers (B) (growth differentiation factor 15 and N-terminal pro-B-type natriuretic peptide), and clinical variables (C) (extent of coronary artery disease, previous vascular disease, Killip class, ACS type, P2Y12 inhibitor). The model, named ABC-ACS ischemia, was well calibrated and showed good discriminatory ability for 1-year risk of CV death/MI with C-indices of 0.71 and 0.72 in the development and validation cohorts, respectively. For CV death, the score performed better, with C-indices of 0.80 and 0.84 in the development and validation cohorts, respectively.CONCLUSIONS: An 8-item score for the prediction of CV death/MI was developed and validated for patients with ACS undergoing percutaneous coronary intervention. The ABC-ACS ischemia score showed good calibration and discrimination and might be useful for risk prediction and decision support in patients with ACS.     • Key to the success of developing high-performance applications for present and future heterogeneous supercomputers will be the systematic use of measurement and analysis to understand factors that affect delivered performance in the context of parallelization strategy, heterogeneous programming methodology, data partitioning, and scalable algorithm design. The evolving complexity of future exascale platforms makes it unrealistic for application teams to implement their own tools. Similarly, it is naive to expect available robust performance tools to work effectively out-of-the-box, without integration and specialization in respect to application-specific requirements and knowledge. Vlasiator is a powerful massively parallel code for accurate magnetospheric and solar wind plasma simulations. It is being ported to the LUMI HPC system for advanced modeling of the Earth’s magnetosphere and surrounding solar wind. Building on a preexisting Vlasiator performance API called Phiprof, our work significantly advances the performance measurement and analysis capabilities offered to Vlasiator using the TAU, APEX, and IPM tools. The results presented show in-depth characterization of node-level CPU/GPU and MPI communications performance. We highlight the integration of high-level Phiprof events with detailed performance data to expose opportunities for performance tuning. Our results provide important insights to optimize Vlasiator for the upcoming Exascale machines.

    • Five different low-Mach large eddy simulations are compared to the turbulent stratified flame experiments conducted at the Technical University of Darmstadt (TUD). The simulations were contributed by TUD, the Institute for Combustion Technology (ITV) at Aachen, Lund University (LUND), the EM2C laboratory at Ecole Centrale Paris, and the University of Duisburg-Essen (UDE). Combustion is modeled by a premixed flamelet tabulation with local flame thickening (TUD), a premixed flamelet progress variable approach coupled to a level set method (ITV), a 4-steps mechanism combined with implicit LES (LUND), the F-TACLES model that is based on filtered premixed flamelet tabulation (EM2C), and a flame surface density approach (UDE). An extensive comparison of simulation and experimental data is presented for the first two moments of velocity, temperature, mixture fraction, and major species mass fractions. The importance of heat-losses was assessed by comparing simulations for adiabatic and isothermal boundary conditions at the burner walls. The adiabatic computations predict a flame anchored on the burner lip, while the non-adiabatic simulations show a flame lift-off of one half pilot diameter and a better agreement with experimental evidence for temperature and species concentrations. Most simulations agree on the mean flame brush position, but it is evident that subgrid turbulence must be considered to achieve the correct turbulent flame speed. Qualitative comparisons of instantaneous snapshots of the flame show differences in the size of the resolved flame wrinkling patterns. These differences are (a) caused by the influence of the LES combustion model on the flame dynamics and (b) by the different simulation strategies in terms of grid, inlet condition and numerics.     • BACKGROUND: Growth differentiation factor 15(GDF15), a stress-responsive cytokine produced in cardiovascular cells under conditions of inflammation and oxidative stress, is emerging as an important prognostic marker in individuals with and without existing cardiovascular disease (CVD). We therefore examined the clinical and genetic correlates of circulating GDF15 concentrations, which have not been investigated collectively. METHODS: Plasma GDF15 concentrations were measured in 2991 participants in the Framingham Offspring Study who were free of clinically overt CVD (mean age, 59 years; 56% women). Clinical correlates of GDF15 were examined in multivariable analyses. We then conducted a genomewide association study of the GDF15 concentration that included participants in the Framingham Offspring Study and participants in the PIVUS (Prospective Investigation of the Vasculature in Uppsala Seniors) study. RESULTS: GDF15 was positively associated with age, smoking, antihypertensive treatment, diabetes, worse kidney function, and use of nonsteroidal antiinflammatory drugs (NSAIDs), but it was negatively associated with total cholesterol and HDL cholesterol. Clinical correlates accounted for 38% of interindividual variation in the circulating GDF15 concentration, whereas genetic factors accounted for up to 38% of the residual variability (h 2 = 0.38; P = 2.5 X 10 -11). We identified 1 locus of genomewide significance. This locus, which is on chromosome 19p13.11 and includes the GDF15 gene, is associated with GDF15 concentration (smallest P = 2.74 X 10 -32 for rs888663). Conditional analyses revealed 2 independent association signals at this locus (rs888663 and rs1054564), which were associated with altered cis gene expression in blood cell lines. CONCLUSIONS: In ambulatory individuals, both cardiometabolic risk factors and genetic factors play important roles in determining circulating GDF15 concentrations and contribute similarly to the overall variation.

  • Properties of Magnetic Reconnection and FTEs on the Dayside Magnetopause With and Without Positive IMF Bx Component During Southward IMF

    • This paper describes properties and behavior of magnetic reconnection and flux transfer events (FTEs) on the dayside magnetopause using the global hybrid-Vlasov code Vlasiator. We investigate two simulation runs with and without a sunward (positive)B-x component of the interplanetary magnetic field (IMF) when the IMF is southward. The runs are two-dimensional in real space in the noon-midnight meridional (polar) plane and three-dimensional in velocity space. Solar wind input parameters are identical in the two simulations with the exception that the IMF is purely southward in one but tilted 45 degrees toward the Sun in the other. In the purely southward case (i.e., without B-x) the magnitude of the magnetos heath magnetic field component tangential to the magnetopause is larger than in the run with a sunward tilt. This is because the shock normal is perpendicular to the IMF at the equatorial plane, whereas in the other run the shock configuration is oblique and a smaller fraction of the total IMF strength is compressed at the shock crossing. Hence, the measured average and maximum reconnection rate are larger in the purely southward run. The run with tilted IMF also exhibits a north-south asymmetry in the tangential magnetic field caused by the different angle between the IMF and the bow shock normal north and south of the equator. Greater north-south asymmetries are seen in the FTE occurrence rate, size, and velocity as well; FTEs moving toward the Southern Hemisphere are larger in size and observed less frequently than FTEs in the Northern Hemisphere.

  • Ion Acceleration Efficiency at the Earth's Bow Shock : Observations and Simulation Results

    • Collisionless shocks are some of the most efficient particle accelerators in heliospheric and astrophysical plasmas. Here we study and quantify ion acceleration at Earth's bow shock with observations from NASA's Magnetospheric Multiscale (MMS) satellites and in a global hybrid-Vlasov simulation. From the MMS observations, we find that quasiparallel shocks are more efficient at accelerating ions. There, up to 15% of the available energy goes into accelerating ions above 10 times their initial energy. Above a shock-normal angle of similar to 50 degrees, essentially no energetic ions are observed downstream of the shock. We find that ion acceleration efficiency is significantly lower when the shock has a low Mach number (M ( A ) < 6) while there is little Mach number dependence for higher values. We also find that ion acceleration is lower on the flanks of the bow shock than at the subsolar point regardless of the Mach number. The observations show that a higher connection time of an upstream field line leads to somewhat higher acceleration efficiency. To complement the observations, we perform a global hybrid-Vlasov simulation with realistic solar-wind parameters with the shape and size of the bow shock. We find that the ion acceleration efficiency in the simulation shows good quantitative agreement with the MMS observations. With the combined approach of direct spacecraft observations, we quantify ion acceleration in a wide range of shock angles and Mach numbers.

    • Targeting a specific chemokine/receptor axis in atherosclerosis remains challenging. Soluble receptor-based strategies are not established for chemokine receptors due to their discontinuous architecture. Macrophage migration-inhibitory factor (MIF) is an atypical chemokine that promotes atherosclerosis through CXC-motif chemokine receptor-4 (CXCR4). However, CXCR4/CXCL12 interactions also mediate atheroprotection. Here, we show that constrained 31-residue-peptides (‘msR4Ms’) designed to mimic the CXCR4-binding site to MIF, selectively bind MIF with nanomolar affinity and block MIF/CXCR4 without affecting CXCL12/CXCR4. We identify msR4M-L1, which blocks MIF- but not CXCL12-elicited CXCR4 vascular cell activities. Its potency compares well with established MIF inhibitors, whereas msR4M-L1 does not interfere with cardioprotective MIF/CD74 signaling. In vivo-administered msR4M-L1 enriches in atherosclerotic plaques, blocks arterial leukocyte adhesion, and inhibits atherosclerosis and inflammation in hyperlipidemic Apoe−/− mice in vivo. Finally, msR4M-L1 binds to MIF in plaques from human carotid-endarterectomy specimens. Together, we establish an engineered GPCR-ectodomain-based mimicry principle that differentiates between disease-exacerbating and -protective pathways and chemokine-selectively interferes with atherosclerosis.

    • Background Programmed death-1 (PD-1/CD279) is a cell-surface protein expressed in activated T cells and a subset of T lymphocytes including follicular helper T cells (TFH). The interaction between PD-1 and its ligands plays a role in immune response and evasion of malignancies. In nodal follicular lymphoma, the number of intratumoral PD-1-positive lymphocytes is associated with overall survival. Objectives To investigate 28 cases of primary cutaneous B-cell lymphoma, including the subtypes PCFCL (n = 10), PCMZL (n = 10) and DLBCL-LT (n = 8) for the number and density of PD-1-positive cells. Methods Immunohistochemical staining and a computerized morphometric analysis for evaluation were applied. The results were correlated with the clinical outcome. To distinguish between activated T cells and TFH we performed PD-1/bcl-6 double staining and compared these results with CXCL-13 staining. Double staining for PD-1 and PAX-5 was used to investigate whether tumour cells were positive for PD-1. Results The PD-1-positive cells represented tumour-infiltrating T cells (TILs). Only a minor subset was represented by TFH. Patients with DLBCL-LT had a significantly lower number of PD-1-positive TILs than those with PCMZL (P = 0·012) and PCFCL (P = 0·002) or both (P = 0·001). The difference between PCMZL and PCFCL did not reach significance (P = 0·074). The tumour cells were negative for PD-1. Conclusions A higher number of PD-1-expressing cells was found in indolent PCMZL and PCFCL than in high-malignant DLBCL-LT. The PD-1-positive cells represented not only TFH, but also other activated T cells as a part of the tumour microenvironment. The tumour cells in all investigated types of PCBCL did not show aberrant PD-1 expression. What’s already known about this topic? In nodal follicular lymphoma, the number of intratumoral programmed death (PD)-1-positive lymphocytes is associated with overall survival. No data exist for primary cutaneous B-cell lymphoma (PCBCL). What does this study add? A higher number of PD-1-expressing cells was found in indolent primary cutaneous marginal zone lymphoma and primary cutaneous follicle centre lymphoma, in contrast to the more aggressive diffuse large B-cell lymphoma, leg type. PD-1-positive cells do not represent only follicular helper T cells. The tumour cells in all investigated types of PCBCL did not show aberrant PD-1 expression. © 2013 British Association of Dermatologists.

    • We use a global hybrid-Vlasov simulation for the magnetosphere, Vlasiator, to investigate magnetosheath high-speed jets. Unlike many other hybrid-kinetic simulations, Vlasiator includes an unscaled geomagnetic dipole, indicating that the simulation spatial and temporal dimensions can be given in SI units without scaling. Thus, for the first time, this allows investigating the magnetosheath jet properties and comparing them directly with the observed jets within the Earth's magnetosheath. In the run shown in this paper, the interplanetary magnetic field (IMF) cone angle is 30°, and a foreshock develops upstream of the quasi-parallel magnetosheath. We visually detect a structure with high dynamic pressure propagating from the bow shock through the magnetosheath. The structure is confirmed as a jet using three different criteria, which have been adopted in previous observational studies. We compare these criteria against the simulation results. We find that the magnetosheath jet is an elongated structure extending earthward from the bow shock by ∼ 2.6 RE, while its size perpendicular to the direction of propagation is ∼ 0.5R/E. We also investigate the jet evolution and find that the jet originates due to the interaction of the bow shock with a high-dynamic-pressure structure that reproduces observational features associated with a short, large-amplitude magnetic structure (SLAMS). The simulation shows that magnetosheath jets can develop also under steady IMF, as inferred by observational studies. To our knowledge, this paper therefore shows the first global kinetic simulation of a magnetosheath jet, which is in accordance with three observational jet criteria and is caused by a SLAMS advecting towards the bow shock.

    • We have examined the properties of ultralow-frequency (ULF) waves in space (the ion foreshock, magnetosheath, and magnetosphere) and at dayside magnetometer stations (L = 1.6-6.5) during Earth's encounter with a magnetic cloud in the solar wind, which is characterized by magnetic fields with large magnitudes (similar to 14 nT) and small cone angles (similar to 30 degrees). In the foreshock, waves were excited at similar to 90 m Hz as expected from theory, but there were oscillations at other frequencies as well. Oscillations near 90 mHz were detected at the other locations in space, but they were not in general the most dominant oscillations. On the ground, pulsations in the approximate Pc2-Pc4 band (5 mHz-120 mHz) were continuously detected at all stations, with no outstanding spectral peaks near 90 mHz in the H component except at stations where the frequency of the third harmonic of standing Alfven waves had this frequency. The fundamental toroidal wave frequency was below 90 mHz at all stations. In the D component spectra, a minor spectral peak is found near 90 mHz at stations located at L < 3, and the power dropped abruptly above this frequency. Magnetospheric compressional wave power was much weaker on the nightside. A hybrid-Vlasov simulation indicates that foreshock ULF waves have short spatial scale lengths and waves transmitted into the magnetosphere are strongly attenuated away from noon.

    • The Earth's magnetosphere and its bow shock, which is formed by the interaction of the supersonic solar wind with the terrestrial magnetic field, constitute a rich natural laboratory enabling in situ investigations of universal plasma processes. Under suitable interplanetary magnetic field conditions, a foreshock with intense wave activity forms upstream of the bow shock. So-called 30 s waves, named after their typical period at Earth, are the dominant wave mode in the foreshock and play an important role in modulating the shape of the shock front and affect particle reflection at the shock. These waves are also observed inside the magnetosphere and down to the Earth's surface, but how they are transmitted through the bow shock remains unknown. By combining state-of-the-art global numerical simulations and spacecraft observations, we demonstrate that the interaction of foreshock waves with the shock generates earthward-propagating, fast-mode waves, which reach the magnetosphere. These findings give crucial insight into the interaction of waves with collisionless shocks in general and their impact on the downstream medium.

    • BackgroundActivation of vascular smooth muscle cells (VSMCs) inflammation is vital to initiate vascular disease. However, the role of human-specific long noncoding RNAs (lncRNAs) in VSMC inflammation is poorly understood.MethodsBulk RNA-seq in differentiated human VSMCs revealed a novel human-specific lncRNA calledINflammatory MKL1InteractingLongNoncoding RNA (INKILN).INKILNexpression was assessed in multiple in vitro and ex vivo models of VSMC phenotypic modulation and human atherosclerosis and abdominal aortic aneurysm (AAA) samples. The transcriptional regulation ofINKILNwas determined through luciferase reporter system and chromatin immunoprecipitation assay. Both loss- and gain-of-function approaches and multiple RNA-protein and protein-protein interaction assays were utilized to uncover the role ofINKILNin VSMC proinflammatory gene program and underlying mechanisms. Bacterial Artificial Chromosome (BAC) transgenic (Tg) mice were utilized to studyINKLINexpression and function in ligation injury-induced neointimal formation.ResultsINKILNexpression is downregulated in contractile VSMCs and induced by human atherosclerosis and abdominal aortic aneurysm.INKILNis transcriptionally activated by the p65 pathway, partially through a predicted NF-κB site within its proximal promoter.INKILNactivates the proinflammatory gene expression in cultured human VSMCs and ex vivo cultured vessels. Mechanistically,INKILNphysically interacts with and stabilizes MKL1, a key activator of VSMC inflammation through the p65/NF-κB pathway.INKILNdepletion blocks ILIβ-induced nuclear localization of both p65 and MKL1. Knockdown ofINKILNabolishes the physical interaction between p65 and MKL1, and the luciferase activity of an NF-κB reporter. Further,INKILNknockdown enhances MKL1 ubiquitination, likely through the reduced physical interaction with the deubiquitinating enzyme, USP10.INKILNis induced in injured carotid arteries and exacerbates ligation injury-induced neointimal formation in BAC Tg mice.ConclusionsThese findings elucidate an important pathway of VSMC inflammation involving anINKILN/MKL1/USP10 regulatory axis. Human BAC Tg mice offer a novel and physiologically relevant approach for investigating human-specific lncRNAs under vascular disease conditions.
