  • Hitomi X-ray studies of giant radio pulses from the Crab pulsar
  • 2018
  • In: Publications of the Astronomical Society of Japan. - : Oxford University Press (OUP). - 0004-6264 .- 2053-051X. ; 70:2
  • Journal article (peer-reviewed)abstract
    • To search for giant X-ray pulses correlated with the giant radio pulses (GRPs) from the Crab pulsar, we performed a simultaneous observation of the Crab pulsar with the X-ray satellite Hitomi in the 2-300 keV band and the Kashima NICT radio telescope in the 1.4-1.7 GHz band with a net exposure of about 2 ks on 2016 March 25, just before the loss of the Hitomi mission. The timing performance of the Hitomi instruments was confirmed to meet the timing requirement and about 1000 and 100 GRPs were simultaneously observed at the main pulse and inter-pulse phases, respectively, and we found no apparent correlation between the giant radio pulses and the X-ray emission in either the main pulse or inter-pulse phase. All variations are within the 2 sigma fluctuations of the X-ray fluxes at the pulse peaks, and the 3 sigma upper limits of variations of main pulse or inter-pulse GRPs are 22% or 80% of the peak flux in a 0.20 phase width, respectively, in the 2-300 keV band. The values for main pulse or inter-pulse GRPs become 25% or 110%, respectively, when the phase width is restricted to the 0.03 phase. Among the upper limits from the Hitomi satellite, those in the 4.5-10 keV and 70-300 keV bands are obtained for the first time, and those in other bands are consistent with previous reports. Numerically, the upper limits of the main pulse and inter-pulse GRPs in the 0.20 phase width are about (2.4 and 9.3) x 10(-11) erg cm(-2), respectively. No significant variability in pulse profiles implies that the GRPs originated from a local place within the magneto-sphere. Although the number of photon-emitting particles should temporarily increase to account for the brightening of the radio emission, the results do not statistically rule out variations correlated with the GRPs, because the possible X-ray enhancement may appear due to a > 0.02% brightening of the pulse-peak flux under such conditions.

  • Delta Np73 expression in thyroid neoplasms originating from follicular cells.
  • 2006
  • In: Pathology. - : Elsevier BV. - 0031-3025. ; 38:3, s. 205-9
  • Journal article (peer-reviewed)abstract
    • AIMS: p73, a homologue of p53, is known as a negative regulator of tumour progression. However, delta Np73, an isoform of p73 lacking the NH2-terminal transactivation domain plays an oncogenic role by interfering with the activity of p53 and TA (full-length transactivating isoforms) p73. In this study, we investigated the expression of delta Np73 in human thyroid neoplasms originating from follicular cells. METHODS: We immunohistochemically investigated delta Np73 expression in 223 thyroid neoplasms. Delta Np73 expression level was evaluated as the sum of positivity score and intensity score. RESULTS: Normal follicular cells did not express delta Np73, but 27.3% of follicular adenoma, 85.4% of follicular carcinoma, 99.2% of papillary carcinoma, and 95.7% of anaplastic carcinoma were positive for the transcript. Delta Np73 expression level did not differ between widely invasive and minimally invasive follicular carcinomas. In papillary carcinoma, the level was inversely linked to tumour size, extrathyroid extension, and clinically apparent metastasis. Furthermore, in anaplastic carcinoma, delta Np73 expression level was significantly lower than that in papillary carcinoma. CONCLUSIONS: Our findings indicate that delta Np73 plays a role predominantly in the early phase of papillary carcinoma progression.

  • Numerical analysis of wind-induced natural ventilation for an isolated cubic room with two openings under small mean wind pressure difference
  • 2022
  • In: Building and Environment. - : Elsevier. - 0360-1323 .- 1873-684X. ; 226
  • Journal article (peer-reviewed)abstract
    • Wind-induced natural ventilation through openings with small wind pressure differences was examined using large eddy simulation (LES) modelling. This study focused on cases in which the ventilation rate is predicted using a standard Orifice equation. The purpose of this study was to clarify how the ventilation rate is underestimated in such cases for both single-sided and double-sided openings, and to clarify the difference between the effective ventilation rate (purging flow rate, PFR) and bulk airflow rate (AFR), which have not been sufficiently and systematically understood. A simple cubic-room model with two openings was analysed using LES by varying the opening position after validation, and the ventilation rate, velocity field, and wind pressure coefficient were compared with experimental results. The PFR was determined by tracing particles, and the AFR was obtained based on the instantaneous velocity over the openings. The AFR predicted by the Orifice equation was underestimated when the difference in the mean wind pressure coefficient (Delta(C-p) over bar) was less than 0.1. The main feature of this study was to show the ventilation effectiveness defined by the PFR divided by AFR, which was approximately 70-80%, 60%, and 90% for the double-sided openings, single-sided openings on the lateral side, and windward and leeward sides, respectively. Another feature was to propose a method for estimating the AFR reflecting two key phenomena, namely pulsating flow and eddy penetration. In addition, a simple equation considering the standard deviation of wind pressure coefficient difference (sigma(Delta Cp)), and external local velocity around an opening (V-vic) was obtained.

  • Hydrographic observations by instrumented marine mammals in the Sea of Okhotsk
  • 2017
  • In: Polar Science. - : Elsevier BV. - 1873-9652 .- 1876-4428. ; 13, s. 56-65
  • Journal article (peer-reviewed)abstract
    • The Sea of Okhotsk is a challenging environment for obtaining in situ data and satellite observation in winter due to sea ice cover. In this study, we evaluated the validity of hydrographic observations by marine mammals (e.g., seals and sea lions) equipped with oceanographic conductivity-temperaturedepth (CTD) sensors. During 4-yr operations from 2011 to 2014, we obtained total of 997 temperature-salinity profiles in and around the Soya Strait, Iony Island, and Urup Strait. The hydrographic data were mainly obtained from May to August and the maximum profile depth in shelf regions almost reaches to the seafloor, while valuable hydrographic data under sea ice cover were also obtained. In strong thermoclines, the seal-derived data sometimes showed positive biases in salinity with spikelike signal. For these salinity biases, we applied a new thermal mass inertia correction scheme, effectively reducing spurious salinity biases in the seasonal thermocline. In the Soya Strait and the adjacent region, the detailed structure of the Soya Warm Current including the cold-water belt was well identified. Dense water up to 27.0 sigma(theta), which can be a potential source of Okhotsk Sea Intermediate Water, has flowed from the Soya Strait into the Sea of Okhotsk in mid-winter (February). In summer, around the Iony Island and Urup Strait, remarkable cold and saline waters are localized in the surface layers. These regions are also characterized by weak stratification, suggesting the occurrence of tidally induced vertical mixing. Thus, CTD-tag observations have a great potential in monitoring data-sparse regions in the Sea of Okhotsk.

  • Structure and mechanism of the mammalian fructose transporter GLUT5
  • 2015
  • In: Nature. - : Springer Science and Business Media LLC. - 0028-0836 .- 1476-4687. ; 526:7573, s. 397-
  • Journal article (peer-reviewed)abstract
    • The altered activity of the fructose transporter GLUT5, an isoform of the facilitated-diffusion glucose transporter family, has been linked to disorders such as type 2 diabetes and obesity. GLUT5 is also overexpressed in certain tumour cells, and inhibitors are potential drugs for these conditions. Here we describe the crystal structures of GLUT5 from Rattus norvegicus and Bos taurus in open outward-and open inward-facing conformations, respectively. GLUT5 has a major facilitator superfamily fold like other homologous monosaccharide transporters. On the basis of a comparison of the inward-facing structures of GLUT5 and human GLUT1, a ubiquitous glucose transporter, we show that a single point mutation is enough to switch the substrate-binding preference of GLUT5 from fructose to glucose. A comparison of the substrate-free structures of GLUT5 with occluded substrate-bound structures of Escherichia coli XylE suggests that, in addition to global rocker-switch-like re-orientation of the bundles, local asymmetric rearrangements of carboxy-terminal transmembrane bundle helices TM7 and TM10 underlie a 'gated-pore' transport mechanism in such monosaccharide transporters.

  • CYP2C9, VKORC1, and CYP4F2 polymorphisms and pediatric warfarin maintenance dose : a systematic review and meta-analysis
  • 2020
  • In: The Pharmacogenomics Journal. - : Springer Science and Business Media LLC. - 1470-269X .- 1473-1150. ; 20:2, s. 306-319
  • Research review (peer-reviewed)abstract
    • Studies on the effect of cytochrome P450 2C9 (CYP2C9), vitamin K epoxide reductase complex subunit 1 (VKORC1), and cytochrome P450 4F2 (CYP4F2) polymorphisms on warfarin maintenance dose in children are conflicting. We conducted a systematic review and meta-analysis to evaluate the effect of these polymorphisms on warfarin maintenance dose in children. We searched relevant literature using the MEDLINE, EMBASE, and Cochrane Central Register of Controlled Trial libraries without any language restrictions from their inception to 23 July 2017. Dose differences are expressed as standardized mean difference (SMD) or mean difference (MD) with 95% confidence intervals (CI). This review was registered in the PROSPERO prospective register of systematic reviews (CRD42015016172). We included a total of nine studies (745 participants) in the meta-analysis. Patients with CYP2C9 *1/*2, *1/*3, *2/*2, *2/*3, or *3/*3 required a lower warfarin maintenance dose compared with patients with CYP2C9 *1/*1 (SMD = -0.610, 95% CI: -0.802 to -0.419, I-2 = 0%). Patients with VKORC1-1639GA or AA required a lower warfarin maintenance dose compared with patients with VKORC1-1639GG (SMD = -0.666, 95% CI: -0.887 to -0.445, I-2 = 33%). However, no associations were observed between CYP4F2 polymorphisms and warfarin maintenance dose (MD = 0.005 mg/kg/day, 95% CI: -0.006 to 0.015, I-2 = 0%). These results were not affected by a sensitivity analysis. Our meta-analysis provides evidence that CYP2C9 and VKORC1 variant statuses affect warfarin maintenance dose in children, but not CYP4F2.
