    • The ANTARES Neutrino Telescope was completed in May 2008 and is the first operational Neutrino Telescope in the Mediterranean Sea. The main purpose of the detector is to perform neutrino astronomy and the apparatus also offers facilities for marine and Earth sciences. This paper describes the design, the construction and the installation of the telescope in the deep sea, offshore from Toulon in France. An illustration of the detector performance is given.     • What does the future hold for the world's ecosystems and benefits that people obtain from them? While the Intergovernmental Platform on Biodiversity and Ecosystem Services (IPBES) has identified the development of scenarios as a key to helping decision makers identify potential impacts of different policy options, it currently lacks a long-term scenario strategy. IPBES will decide how it will approach scenarios at its plenary meeting on 22-28 February 2016, in Kuala Lumpur. IPBES now needs to decide whether it should create new scenarios that better explore ecosystem services and biodiversity dynamics. For IPBES to capture the social-ecological dynamics of biodiversity and ecosystem services, it is essential to engage with the great diversity of local contexts, while also including the global tele-coupling among local places. We present and compare three alternative scenario strategies that IPBES could use and then suggest a bottom-up, cross-scale scenario strategy to improve the policy relevance of future IPBES assessments. We propose five concrete steps as part of an effective, long term scenario development process for IPBES in cooperation with the scientific community.

  • Levers and leverage points for pathways to sustainability

    • 1. Humanity is on a deeply unsustainable trajectory. We are exceeding planetary boundaries and unlikely to meet many international sustainable development goals and global environmental targets. Until recently, there was no broadly accepted framework of interventions that could ignite the transformations needed to achieve these desired targets and goals.2. As a component of the IPBES Global Assessment, we conducted an iterative expert deliberation process with an extensive review of scenarios and pathways to sustainability, including the broader literature on indirect drivers, social change and sustainability transformation. We asked, what are the most important elements of pathways to sustainability?3. Applying a social-ecological systems lens, we identified eight priority points for intervention (leverage points) and five overarching strategic actions and priority interventions (levers), which appear to be key to societal transformation. The eight leverage points are: (1) Visions of a good life, (2) Total consumption and waste, (3) Latent values of responsibility, (4) Inequalities, (5) Justice and inclusion in conservation, (6) Externalities from trade and other telecouplings, (7) Responsible technology, innovation and investment, and (8) Education and knowledge generation and sharing. The five intertwined levers can be applied across the eight leverage points and more broadly. These include: (A) Incentives and capacity building, (B) Coordination across sectors and jurisdictions, (C) Pre-emptive action, (D) Adaptive decision-making and (E) Environmental law and implementation. The levers and leverage points are all non-substitutable, and each enables others, likely leading to synergistic benefits.4. Transformative change towards sustainable pathways requires more than a simple scaling-up of sustainability initiatives-it entails addressing these levers and leverage points to change the fabric of legal, political, economic and other social systems. These levers and leverage points build upon those approved within the Global Assessment's Summary for Policymakers, with the aim of enabling leaders in government, business, civil society and academia to spark transformative changes towards a more just and sustainable world.

  • Identification of conformational epitopes for human IgG on chemotaxis inhibitory protein of Staphylococcus aureus

    • Background The Chemotaxis inhibitory protein of Staphylococcus aureus (CHIPS) blocks the Complement fragment C5a receptor (C5aR) and formylated peptide receptor (FPR) and is thereby a potent inhibitor of neutrophil chemotaxis and activation of inflammatory responses. The majority of the healthy human population has antibodies against CHIPS that have been shown to interfere with its function in vitro. The aim of this study was to define potential epitopes for human antibodies on the CHIPS surface. We also initiate the process to identify a mutated CHIPS molecule that is not efficiently recognized by preformed anti-CHIPS antibodies and retains anti-inflammatory activity. Results In this paper, we panned peptide displaying phage libraries against a pool of CHIPS specific affinity-purified polyclonal human IgG. The selected peptides could be divided into two groups of sequences. The first group was the most dominant with 36 of the 48 sequenced clones represented. Binding to human affinity-purified IgG was verified by ELISA for a selection of peptide sequences in phage format. For further analysis, one peptide was chemically synthesized and antibodies affinity-purified on this peptide were found to bind the CHIPS molecule as studied by ELISA and Surface Plasmon Resonance. Furthermore, seven potential conformational epitopes responsible for antibody recognition were identified by mapping phage selected peptide sequences on the CHIPS surface as defined in the NMR structure of the recombinant CHIPS31-121 protein. Mapped epitopes were verified by in vitro mutational analysis of the CHIPS molecule. Single mutations introduced in the proposed antibody epitopes were shown to decrease antibody binding to CHIPS. The biological function in terms of C5aR signaling was studied by flow cytometry. A few mutations were shown to affect this biological function as well as the antibody binding. Conclusions Conformational epitopes recognized by human antibodies have been mapped on the CHIPS surface and amino acid residues involved in both antibody and C5aR interaction could be defined. This information has implications for the development of an effective anti-inflammatory agent based on a functional CHIPS molecule with low interaction with human IgG.

  • Beyond Cockpit-ism : Four Insights to Enhance the Transformative Potential of the Sustainable Development Goals

    • The Sustainable Development Goals (SDG) have the potential to become a powerful political vision that can support the urgently needed global transition to a shared and lasting prosperity. In December 2014, the United Nations (UN) Secretary General published his report on the SDGs. However, the final goals and targets that will be adopted by the UN General Assembly in September 2015 risk falling short of expectations because of what we call "cockpit-ism": the illusion that top-down steering by governments and intergovernmental organizations alone can address global problems. In view of the limited effectiveness of intergovernmental efforts and questions about the capacity of national governments to affect change, the SDGs need to additionally mobilize new agents of change such as businesses, cities and civil society. To galvanize such a broad set of actors, multiple perspectives on sustainable development are needed that respond to the various motives and logics of change of these different actors. We propose four connected perspectives which can strengthen the universal relevance of the SDGs: "planetary boundaries" to stress the urgency of addressing environmental concerns and to target governments to take responsibility for (global) public goods; "the safe and just operating space" to highlight the interconnectedness of social and environmental concerns and its distributive consequences; "the energetic society" to benefit from the willingness of a broad group of actors worldwide to take action; and "green competition" to stimulate innovation and new business practices. To realize the transformative potential of the SDGs, these four perspectives should be reflected in the focus and content of the SDGs that will be negotiated in the run up to September 2015 and its further implementation.

  • Towards a better future for biodiversity and people : Modelling Nature Futures

    • The Nature Futures Framework (NFF) is a heuristic tool for co-creating positive futures for nature and people. It seeks to open up a diversity of futures through mainly three value perspectives on nature - Nature for Nature, Nature for Society, and Nature as Culture. This paper describes how the NFF can be applied in modelling to support decision-making. First, we describe key considerations for the NFF in developing qualitative and quantitative scenarios: i) multiple value perspectives on nature as a state space where pathways improving nature toward a frontier can be represented, ii) mutually reinforcing key feedbacks of social-ecological systems that are important for nature conservation and human wellbeing, iii) indicators of multiple knowledge systems describing the evolution of complex social-ecological dynamics. We then present three approaches to modelling Nature Futures scenarios in the review, screening, and design phases of policy processes. This paper seeks to facilitate the integration of relational values of nature in models and strengthen modelled linkages across biodiversity, nature's contributions to people, and quality of life.

    • The United Nations' discussions on defining a new set of post-2015 development goals focus on poverty eradication and sustainable development. Biodiversity and ecosystem services are essential for poverty eradication, which is also one of the foundations of the Strategic Plan for Biodiversity of the Convention on Biological Diversity (CBD). Based on an assessment of current proposals of goals and targets, and a quantitative pathway analysis to meet long term biodiversity and food security goals, this paper discusses how biodiversity and ecosystem services can be integrated into a broad set of goals and targets, and concludes with relevant target areas and means of implementation for which specific targets need to be defined. Furthermore, it responds to the call of the CBD to consider the Strategic Plan for Biodiversity and the related Aichi biodiversity targets in the post-2015 development agenda. The paper's analysis identifies three overlapping but also supplemental ways to integrate biodiversity and ecosystem services in the post-2015 agenda: integrated goals, goals addressing earth system functioning and goals addressing environmental limits. It further concludes seven target areas to be included under the goals to address biodiversity and ecosystem services in the context of food and agriculture: access to food, demand for agricultural products, sustainable intensification, ecosystem fragmentation, protected areas, essential ecosystem services and genetic diversity. The Strategic Plan for Biodiversity provides a good basis for integrating biodiversity and ecosystem services in the post-2015 development agenda. Many Aichi targets address the proposed target areas and the means of implementation discussed, while they need to be complemented with targets that specifically address human well-being, as well as institutions and governance.

  • The potential of international cooperative initiatives to address key challenges of protected areas

    • Protected areas are frequently used as an important governance approach for biodiversity conservation. Even though the total area of protected areas has increased over time, the coverage, quality of management and effectiveness of these areas are still suboptimal. A large body of literature identifies four main challenges that limit the effectiveness of protected areas: lack of stakeholder participation, insufficient organisational capacity to enforce rules, poor integration across social and ecological goals, and underdeveloped accountability mechanisms for assessing management procedures. To address these challenges, scholars and policymakers increasingly debate how to foster an integrated, inclusive, and transparent “whole of society” approach to conservation. We contribute to this debate by examining the role of international cooperative initiatives (ICIs), involving non-state and subnational actors operating across national borders to steer society towards a common goal. We identify 20 ICIs that work on protected areas and analyse their potential to address the four main challenges identified in protected areas, by examining their actor constellation, governance functions, goal alignment, and monitoring and reporting mechanisms. We find that ICIs working on protected areas have the potential to directly address challenges in protected areas related to lack of capacity and accountability mechanisms, and indirectly address challenges related to lack of participation and integration across goals. We discuss these findings in relation to scholarly debates in the global environmental governance and protected areas literature respectively, as well as, to policy debates over the post-2020 Global Biodiversity Framework within the Convention on Biological Diversity (CBD).

    • Background Efficient incorporation of e-health in patients with heart failure (HF) may enhance health care efficiency and patient empowerment. We aimed to assess the effect on self-care of (i) the European Society of Cardiology/Heart Failure Association website on top of usual care, and (ii) an e-health adjusted care pathway leaving out in person routine HF nurse consultations in stable HF patients. Methods and results In a three-group parallel-randomized trial in stable HF patients from nine Dutch outpatient clinics, we compared two interventions ( website and an e-health adjusted care pathway) to usual care. The primary outcome was self-care measured with the European Heart Failure Self-care Behaviour Scale. Secondary outcomes were health status, mortality, and hospitalizations. In total, 450 patients were included. The mean age was 66.8 +/- 11.0 years, 74.2% were male, and 78.8% classified themselves as New York Heart Association I or II at baseline. After 3 months of follow-up, the mean score on the self-care scale was significantly higher in the groups using the website and the adjusted care pathway compared to usual care (73.5 vs. 70.8, 95% confidence interval 0.6-6.2; and 78.2 vs. 70.8, 95% confidence interval 3.8- 9.4, respectively). The effect attenuated, until no differences after 1 year between the groups. Quality of life showed a similar pattern. Other secondary outcomes did not clearly differ between the groups. Conclusions Both the website and an e-health adjusted care pathway improved self-care in HF patients on the short term, but not on the long term. Continuous updating of e-health facilities could be helpful to sustain effects.

    • Alzheimer’s disease (AD) is a neurodegenerative disease that eventually affects memory and behavior. The identification of biomarkers based on risk factors for AD provides insight into the disease since the exact cause of AD remains unknown. Several studies have proposed microRNAs (miRNAs) in blood as potential biomarkers for AD. Exposure to heavy metals is a potential risk factor for onset and development of AD. Blood cells of subjects that are exposed to lead detected in the circulatory system, potentially reflect molecular responses to this exposure that are similar to the response of neurons. In this study we analyzed blood cell-derived miRNAs derived from a general population as proxies of potentially AD-related mechanisms triggered by lead exposure. Subsequently, we analyzed these mechanisms in the brain tissue of AD subjects and controls. A total of four miRNAs were identified as lead exposure-associated with hsa-miR-3651, hsa-miR-150-5p and hsa-miR-664b-3p being negatively and hsa-miR-627 positively associated. In human brain derived from AD and AD control subjects all four miRNAs were detected. Moreover, two miRNAs (miR-3651, miR-664b-3p) showed significant differential expression in AD brains versus controls, in accordance with the change direction of lead exposure. The miRNAs’ gene targets were validated for expression in the human brain and were found enriched in AD-relevant pathways such as axon guidance. Moreover, we identified several AD relevant transcription factors such as CREB1 associated with the identified miRNAs. These findings suggest that the identified miRNAs are involved in the development of AD and might be useful in the development of new, less invasive biomarkers for monitoring of novel therapies or of processes involved in AD development.

    • The causes and consequences of biodiversity loss and climate change are deeply intertwined. Hundreds of existing cooperative initiatives—gathering thousands of states, regions, cities, companies, civil society organisations and communities—are potentially bending the curve on biodiversity loss and tackling climate change simultaneously. More research is needed to understand if, how and under what conditions cooperative initiatives are delivering on their promises and importantly how they can contribute to both ‘biodiversity positive outcomes’ and ‘net-zero emissions’ at the same time.
