    • Significance of the genetic component in determining the interindividual variation observed in skeletal muscle fibre composition and enzyme activities was investigated in 31 pairs of male and female monozygous (MZ) and dizygous (DZ) twins, whose ages ranged in all but one pair (11 years) from 15 to 24 years. Percent distribution of slow twitch muscle fibres and activities of Ca2+ and Ng2+ stimulated ATPases, creatine phosphokinase, myokinase, phosphorylase, lactate dehydrogenase (LDH) and distribution of its isozyme LDH-1 were all analyzed in biopsy samples taken from the vastus lateralis muscle. The data disclosed that in contrast to DZ twins the MZ twins of both sexes had an essentially identical muscle fibre composition. Calculation of the heritability estimate for this parameter gave the values of 99.5% and 92.8%, respectively for males and females. In contrast to the fibre composition presence of a significant genetic component was not observed in any of the enzyme activities studied. It was concluded that there is a predominant genetic influence on the skeletal muscle fibre composition in man, and thus also on the potential capacity of the muscles to perform work.

  • Effect of strength training on EMG of human skeletal muscle.

    • The effects of an 8 weeks period of systematic progressive strength training on the EMG activity of the leg extensor muscles (vastus lateralis and rectus femoris) were investigated in 8 healthy male subjects (22-31 yrs). After training there were indications (n.s.) of a decline in integrated EMG (IEMG) during maximal isometric knee extension as well as in the IEMG vs isometric force relationship. The averaged motor unit potential (AMUP) did not demonstrate any significant changes due to the strength training regimen. In conformity with earlier findings no or only minor alterations were observed in anthropometrics, muscle enzyme activities and fibre composition. The fibre area ratio indicated a specific effect of the training stimuli on the fast twitch muscle fibres. Thus, EMG-analyses, as employed in the present study, did not provide any conclusive additional explanation as to the mechanisms behind the well established gains in muscle strength performance induced by the applied strength training program.

  • Infections with Plasmodium falciparum during pregnancy affect VAR2CSA DBL-5 domain-specific T cell cytokine responses

    • Background: Current knowledge of human immunological responses to pregnancy-associated malaria-specific Plasmodium falciparum protein VAR2CSA concerns almost exclusively B cell-driven antibody-mediated activity. Knowledge of VAR2CSA-specific T cell-mediated activity is minimal by comparison, with only a single published report of a study investigating VAR2CSA-derived peptide-specific T cell responses. The study described here represents an attempt to redress this balance. Methods: Within the framework of a cohort study of 1037 pregnant Beninese, sub-groups were selected on the basis of the documented presence/absence of infection with P. falciparum and conducted detailed immunological assessments both at inclusion into the study and at delivery. Peripheral blood mononuclear cells were isolated, stimulated in vitro, and VAR2CSA DBL-5 domain-specific, IFN-gamma-secreting T-cell frequencies and cytokine responses were quantified using flow cytometric techniques. Multivariate analyses were used to determine primarily whether the T cell-mediated DBL5-specific activity measured was associated with infection by P. falciparum adjusted for gravidity, anaemia and other cofactors. Results: Infections with P. falciparum detected at inclusion were associated with enhanced non-specific TNF responses, whilst diminished non-specific and DBL-5-specific IL-10 responses were associated with infections detected at delivery. Infections during pregnancy led to enhanced non-specific and DBL-5-specific IFN-gamma responses detectable at delivery but to concomitantly lower DBL-5-specific CD8+ IFN-gamma responses. Prospective assessments indicated that non-specific pro-inflammatory responses detectable at inclusion in the study were associated with the occurrence of infections subsequently during pregnancy. Conclusions: The findings represent a first step in elucidating the quantity and quality of cellular immunological responses to VAR2CSA, which will help in the development of the primary vaccine candidate for prevention of pregnancy-associated malaria.

  • Malaria Modifies Neonatal and Early-Life Toll-Like Receptor Cytokine Responses

    • Protection from infections in early life relies extensively on innate immunity, but it is unknown whether and how maternal infections modulate infants' innate immune responses, thereby altering susceptibility to infections. Plasmodium falciparum causes pregnancy-associated malaria (PAM), and epidemiological studies have shown that PAM enhances infants' susceptibility to infection with P. falciparum. We investigated how PAM-mediated exposures in utero affect innate immune responses and their relationship with infection in infancy. In a prospective study of mothers and their babies in Benin, we investigated changes in Toll-like receptor (TLR)-mediated cytokine responses related to P. falciparum infections. Whole-blood samples from 134 infants at birth and at 3, 6, and 12 months of age were stimulated with agonists specific for TLR3, TLR4, TLR7/8, and TLR9. TLR-mediated interleukin 6 (IL-6) and IL-10 production was robust at birth and then stabilized, whereas tumor necrosis factor alpha (TNF-alpha) and gamma interferon (IFN-gamma) responses were weak at birth and then increased. In multivariate analyses, maternal P. falciparum infections at delivery were associated with significantly higher TLR3-mediated IL-6 and IL-10 responses in the first 3 months of life (P<0.05) and with significantly higher TLR3-, TLR7/8-, and TLR9-mediated TNF-alpha responses between 6 and 12 months of age (P<0.05). Prospective analyses showed that higher TLR3- and TLR7/8-mediated IL-10 responses at birth were associated with a significantly higher risk of P. falciparum infection in infancy (P<0.05). Neonatal and infant intracellular TLR-mediated cytokine responses are conditioned by in utero exposure through PAM late in pregnancy. Enhanced TLR-mediated IL-10 responses at birth are associated with an increased risk of P. falciparum infection, suggesting a compromised ability to combat infection in early life.

  • Effects of ospemifene and raloxifene on hormonal status, lipids, genital tract and tolerability in postmenopausal women

    • OBJECTIVE: To compare ospemifene and raloxifene regarding their effects on hormones, lipids, genital tract, and tolerability in postmenopausal women. DESIGN: A randomized, double-blind study in which 118 healthy postmenopausal women received 30 (n = 29), 60 (n = 30), or 90 mg (n = 30) of ospemifene or 60 mg (n = 29) of raloxifene for 3 months. RESULTS: There were no significant differences in the baseline characteristics between study groups. In comparison with raloxifene, follicle-stimulating hormone levels decreased significantly more in the 90-mg ospemifene group and sex hormone-binding globulin levels increased more in all ospemifene groups. Total cholesterol and low-density lipoprotein cholesterol levels decreased more in raloxifene than in ospemifene groups, although the difference in low-density lipoprotein cholesterol between 90-mg ospemifene and raloxifene was not significant. Endometrial thickness did not change in any study group and endometrial biopsies showed atrophy in the majority of subjects at 3 months. All ospemifene groups demonstrated a clear estrogenic effect on the vaginal epithelium, as seen in Pap smears. This was in sharp contrast to the raloxifene group, which had no effect on the vaginal epithelium. Kupperman index decreased in all study groups during treatment. The adverse events were mild, mainly single cases, and no clustering of events was observed. There were no clinically significant abnormal findings in laboratory safety parameters. CONCLUSIONS: Ospemifene, at the dose of 90 mg/day, was more estrogenic than raloxifene, as shown by changes in serum follicle-stimulating hormone and sex hormone-binding globulin levels. Neither agent stimulated endometrium, but in contrast to raloxifene, ospemifene had a clear estrogenic effect in the vagina. Further studies with ospemifene are needed in subjects with vaginal atrophy.

  • Effects of exhaustive stretch-shortening cycle exercise on muscle blood flow during exercise

    • Abstract Aim: The influence of exhaustive stretch-shortening cycle exercise (SSC) on skeletal muscle blood flow (BF) during exercise is currently unknown. Methods: Quadriceps femoris (QF) BF was measured in eight healthy men using positron emission tomography before and 3 days after exhaustive SSC exercise. The SSC protocol consisted of maximal and submaximal drop jumps with one leg. Needle biopsies of the vastus lateralis muscles were taken immediately and 2 days after SSC for muscle endothelial nitric oxide synthase (eNOS) and interleukin-1-beta (IL-1beta) mRNA level determinations. Results: All subjects reported subjective muscle soreness after SSC (P < 0.001), which was well in line with a decrease in maximal isometric contraction force (MVC) and increase in serum creatine kinase activity (CK) (P = 0.018). After SSC muscle BF was 25% higher in entire QF (P = 0.043) and in its deep and superficial muscle regions, whereas oxygen uptake remained unchanged (P = 0.893). Muscle biopsies revealed increased IL-1beta (30 min: 152 +/- 75%, P = 0.012 and 2 days: 108 +/- 203%, P = 0.036) but decreased or unchanged eNOS (30 min; -21 +/- 57%, P = 0.050 and 2 days: +101 +/- 204%, P = 0.779) mRNA levels after SSC. Conclusion: It was concluded that fatiguing SSC exercise induces increased muscle BF during exercise, which is likely to be associated with pro-inflammatory processes in the exercised muscle.

    • Maternal parasitoses modulate fetal immune development, manifesting as altered cellular immunological activity in cord blood that may be linked to enhanced susceptibility to infections in early life. Plasmodium falciparum typifies such infections, with distinct placental infection-related changes in cord blood exemplified by expanded populations of parasite antigen-specific regulatory T cells. Here we addressed whether such early-onset cellular immunological alterations persist through infancy. Specifically, in order to assess the potential impacts of P. falciparum infections either during pregnancy or during infancy, we quantified lymphocyte subsets in cord blood and in infants' peripheral blood during the first year of life. The principal age-related changes observed, independent of infection status, concerned decreases in the frequencies of CD4(+), NKdim and NKT cells, whilst CD8(+), Treg and Teff cells' frequencies increased from birth to 12 months of age. P. falciparum infections present at delivery, but not those earlier in gestation, were associated with increased frequencies of Treg and CD8(+) T cells but fewer CD4(+) and NKT cells during infancy, thus accentuating the observed age-related patterns. Overall, P. falciparum infections arising during infancy were associated with a reversal of the trends associated with maternal infection i.e. with more CD4(+) cells, with fewer Treg and CD8(+) cells. We conclude that maternal P. falciparum infection at delivery has significant and, in some cases, year-long effects on the composition of infants' peripheral blood lymphocyte populations. Those effects are superimposed on separate and independent age-as well as infant infection-related alterations that, respectively, either match or run counter to them.

  • Influence of composition and prior deformation on phase transformation temperatures and hardness in direct quenching using physical simulation

    • For conventional reheated and quenched (RQ) steels, the level of alloying necessary to achieve a given hardenability and hardness can be estimated from calculated Jominy curves or ideal critical diameters (DI) given in ASTM A255-10(2014). However, for thermomechanically rolled direct quenched (DQ) steels, little data are available. In this study, the accuracy of the ASTM approach was estimated by designing an experiment to study the main effects of seven alloying elements (C, Mn, Cr, Ni, Mo, Nb, and V) at two levels with eight boron steels based on an eight-run resolution III partial factorial designed experiment. Continuous cooling transformation (CCT) diagrams covering cooling rates of 1.5°C/s-48°C/s were determined using Gleeble simulations with or without controlled deformation below Tnr. The effects of deformation below Tnr and the alloying elements were clearly revealed. In general, low temperature straining of the austenite led to higher levels of hardness than quenching from unstrained austenite. The start of the bainite transformation Ar3 (bainite) was modelled as a function of chemical composition and cooling rate. The data in the CCT diagrams were used to derive equivalent ideal critical diameters (DIB) for strained and unstrained prior austenite. The hardenability index DIB calculated from experimental Jominy curves generally agreed well with those of CCT data for unstrained austenite. In order to apply the ASTM A255 approach to the calculation of DIB, boron factors (BF) for steels with alloy factors (AFs) greater than 26 were estimated by extrapolating the ASTM data. However, this approach did not give satisfactory predictions for either strained or unstrained austenite. Preliminary analysis indicated that Cr, Mo, and V might be less effective at increasing hardenability than implied from their AFs. New formulae were given to allow estimations of the hardness to be expected in connection with direct quenching.
