  • The Gaia mission
  • 2016
  • Ingår i: Astronomy and Astrophysics. - : EDP Sciences. - 0004-6361 .- 1432-0746. ; 595
  • Tidskriftsartikel (refereegranskat)abstract
    • Gaia is a cornerstone mission in the science programme of the European Space Agency (ESA). The spacecraft construction was approved in 2006, following a study in which the original interferometric concept was changed to a direct-imaging approach. Both the spacecraft and the payload were built by European industry. The involvement of the scientific community focusses on data processing for which the international Gaia Data Processing and Analysis Consortium (DPAC) was selected in 2007. Gaia was launched on 19 December 2013 and arrived at its operating point, the second Lagrange point of the Sun-Earth-Moon system, a few weeks later. The commissioning of the spacecraft and payload was completed on 19 July 2014. The nominal five-year mission started with four weeks of special, ecliptic-pole scanning and subsequently transferred into full-sky scanning mode. We recall the scientific goals of Gaia and give a description of the as-built spacecraft that is currently (mid-2016) being operated to achieve these goals. We pay special attention to the payload module, the performance of which is closely related to the scientific performance of the mission. We provide a summary of the commissioning activities and findings, followed by a description of the routine operational mode. We summarise scientific performance estimates on the basis of in-orbit operations. Several intermediate Gaia data releases are planned and the data can be retrieved from the Gaia Archive, which is available through the Gaia home page.

    • In the first production run of the WASA experiment at COSY, the eta decay into three neutral pions was measured in proton-proton interactions at a proton beam kinetic energy of 1.4 GeV. The Dalitz plot of the three pious was Studied using 1.2 x 10(5) fully reconstructed events. and the quadratic slope parameter alpha was determined to be -0.027 +/- 0.008(stat) +/- 0.005(syst). The result is consistent with previous measurements and further corroborates the importance of pion-pion final state interactions. (C) 2009 Elsevier B.V. All rights reserved.

  • Superdeformed and Triaxial States in Ca-42
  • 2016
  • Ingår i: Physical Review Letters. - : American Physical Society. - 0031-9007 .- 1079-7114. ; 117:6
  • Tidskriftsartikel (refereegranskat)abstract
    • Shape parameters of a weakly deformed ground-state band and highly deformed slightly triaxial sideband in Ca-42 were determined from E2 matrix elements measured in the first low-energy Coulomb excitation experiment performed with AGATA. The picture of two coexisting structures is well reproduced by new state-of-the-art large-scale shell model and beyond-mean-field calculations. Experimental evidence for superdeformation of the band built on 0(2)(+) has been obtained and the role of triaxiality in the A similar to 40 mass region is discussed. Furthermore, the potential of Coulomb excitation as a tool to study superdeformation has been demonstrated for the first time.

  • Ingår i: Nature. - : Springer Science and Business Media LLC. - 0028-0836 .- 1476-4687. ; 497:7448, s. 199-204
  • Tidskriftsartikel (refereegranskat)abstract
    • There is strong circumstantial evidence that certain heavy, unstable atomic nuclei are 'octupole deformed', that is, distorted into a pear shape. This contrasts with the more prevalent rugby-ball shape of nuclei with reflection-symmetric, quadrupole deformations. The elusive octupole deformed nuclei are of importance for nuclear structure theory, and also in searches for physics beyond the standard model; any measurable electric-dipole moment (a signature of the latter) is expected to be amplified in such nuclei. Here we determine electric octupole transition strengths (a direct measure of octupole correlations) for short-lived isotopes of radon and radium. Coulomb excitation experiments were performed using accelerated beams of heavy, radioactive ions. Our data on Rn-220 and Ra-224 show clear evidence for stronger octupole deformation in the latter. The results enable discrimination between differing theoretical approaches to octupole correlations, and help to constrain suitable candidates for experimental studies of atomic electric-dipole moments that might reveal extensions to the standard model.

  • 2018
  • Ingår i: Physical Review C. - : AMER PHYSICAL SOC. - 2469-9985 .- 2469-9993. ; 97:2
  • Tidskriftsartikel (refereegranskat)abstract
    • ACoulomb-excitation experiment to study electromagnetic properties of Ca-42 was performed using a 170-MeV calcium beam from the TANDEM XPU facility at INFN Laboratori Nazionali di Legnaro. gamma rays from excited states in Ca-42 were measured with the AGATA spectrometer. The magnitudes and relative signs of ten E2 matrix elements coupling six low-lying states in Ca-42, including the diagonal E2 matrix elements of 2(1)(+) and 2(2)(+) states, were determined using the least-squares code GOSIA. The obtained set of reduced E2 matrix elements was analyzed using the quadrupole sum rule method and yielded overall quadrupole deformation for 0(1),(+)(2) and 2(1,2)(+) states, as well as triaxiality for 0(1,2)(+) states, establishing the coexistence of a weakly deformed ground-state band and highly deformed slightly triaxial sideband in Ca-42. The experimental results were compared with the state-of-the-art large-scale shell-model and beyond-mean-field calculations, which reproduce well the general picture of shape coexistence in Ca-42.

  • Do nuclei go pear-shaped? Coulomb excitation of Rn-220 and Ra-224 at REX-ISOLDE (CERN)

  • Konferensbidrag (refereegranskat)abstract
    • The IS475 collaboration conducted Coulomb-excitation experiments with post-accelerated radioactive Rn-220 and Ra-224 beams at the REX-ISOLDE facility. The beam particles (E-beam: 2.83 MeV/u) were Coulomb excited using Ni-60, Cd-14, and Sn-120 scattering targets. De-excitation gamma-rays were detected employing the Miniball array and scattered particles were detected in a silicon detector. Exploiting the Coulomb-excitation code GOSIA for each nucleus several matrix elements could be obtained from the measured gamma-ray yields. The extracted < 3 parallel to E3 parallel to 0(+)> matrix element allows for the conclusion that, while Rn-220 represents an octupole vibrational system, Ra-224 has already substantial octupole correlations in its ground state. This finding has i(m)plications for the search of CP-violating Schiff moments in the atomic systems of the adjacent odd-mass nuclei.

    • Zoonomia is the largest comparative genomics resource for mammals produced to date. By aligning genomes for 240 species, we identify bases that, when mutated, are likely to affect fitness and alter disease risk. At least 332 million bases (similar to 10.7%) in the human genome are unusually conserved across species (evolutionarily constrained) relative to neutrally evolving repeats, and 4552 ultraconserved elements are nearly perfectly conserved. Of 101 million significantly constrained single bases, 80% are outside protein-coding exons and half have no functional annotations in the Encyclopedia of DNA Elements (ENCODE) resource. Changes in genes and regulatory elements are associated with exceptional mammalian traits, such as hibernation, that could inform therapeutic development. Earth's vast and imperiled biodiversity offers distinctive power for identifying genetic variants that affect genome function and organismal phenotypes.

  • Bifurcations in Nonsmooth Dynamical Systems

  • Ingår i: SIAM Review. - : Society for Industrial & Applied Mathematics (SIAM). - 0036-1445 .- 1095-7200. ; 50:4, s. 629-701
  • Forskningsöversikt (refereegranskat)abstract
    • A review is presented of the one-parameter, nonsmooth bifurcations that occur in a variety of continuous-time piecewise-smooth dynamical systems. Motivated by applications, a pragmatic approach is taken to defining a discontinuity-induced bifurcation (DIB) as a nontrivial interaction of a limit set with respect to a codimension-one discontinuity boundary in phase space. Only DIBs that are local are considered, that is, bifurcations involving equilibria or a single point of boundary interaction along a limit cycle for flows. Three classes of systems are considered, involving either state jumps, jumps in the vector field, or jumps in some derivative of the vector field. A rich array of dynamics are revealed, involving the sudden creation or disappearance of attractors, jumps to chaos, bifurcation diagrams with sharp corners, and cascades of period adding. For each kind of bifurcation identified, where possible, a kind of "normal form" or discontinuity mapping (DM) is given, together with a canonical example and an application. The goal is always to explain dynamics that may be observed in simulations of systems which include friction oscillators, impact oscillators, DC-DC converters, and problems in control theory.

    • In this paper we prove, for the first time, that multistability can occur in three-dimensional Fillipov type flows due to grazing-sliding bifurcations. We do this by reducing the study of the dynamics of Filippov type flows around a grazing-sliding bifurcation to the study of appropriately defined one-dimensional maps. In particular, we prove the presence of three qualitatively different types of multiple attractors born in grazing-sliding bifurcations. Namely, a period-two orbit with a sliding segment may coexist with a chaotic attractor, two stable, period-two and period-three orbits with a segment of sliding each may coexist, or a non-sliding and period-three orbit with two sliding segments may coexist.

  • Ingår i: IMA Journal of Applied Mathematics. - : Oxford University Press (OUP). - 0272-4960 .- 1464-3634. ; 81:4, s. 711-722
  • Tidskriftsartikel (refereegranskat)abstract
    • We describe two examples of three-dimensional Filippov-type flows in which multiple attractors are created by grazing-sliding bifurcations. To the best of our knowledge these are the first examples to show multistability due to a grazing-sliding bifurcation in flows. In both examples, we identify the coefficients of the normal form map describing the bifurcation, and use this to find parameters with the desired behaviour. In the first example this can be done analytically, whilst the second is a dry-friction model and the identification is numerical. This explicit correspondence between the flows and a truncated normal form map reveals an important feature of the sensitivity of the predicted dynamics: the scale of the variation of the bifurcation parameter has to be very carefully chosen. Although no detailed analysis is given, we believe that this may indicate a much greater sensitivity to parameters than experience with smooth flows might suggest. We conjecture that the grazing-sliding bifurcations leading to multistability remained unreported in the literature due to this sensitivity to parameter variations.

    • To evaluate whether antibiotic prescribing for suspected urinary tract infections in frail older adults can be reduced through a multifaceted antibiotic stewardship intervention.Pragmatic, parallel, cluster randomised controlled trial, with a five month baseline period and a seven month follow-up period.38 clusters consisting of one or more general practices (n=43) and older adult care organisations (n=43) in Poland, the Netherlands, Norway, and Sweden, from September 2019 to June 2021.1041 frail older adults aged 70 or older (Poland 325, the Netherlands 233, Norway 276, Sweden 207), contributing 411 person years to the follow-up period.Healthcare professionals received a multifaceted antibiotic stewardship intervention consisting of a decision tool for appropriate antibiotic use, supported by a toolbox with educational materials. A participatory-action-research approach was used for implementation, with sessions for education, evaluation, and local tailoring of the intervention. The control group provided care as usual. The primary outcome was the number of antibiotic prescriptions for suspected urinary tract infections per person year. Secondary outcomes included the incidence of complications, all cause hospital referrals, all cause hospital admissions, all cause mortality within 21 days after suspected urinary tract infections, and all cause mortality. The numbers of antibiotic prescriptions for suspected urinary tract infections in the follow-up period were 54 prescriptions in 202 person years (0.27 per person year) in the intervention group and 121 prescriptions in 209 person years (0.58 per person year) in the usual care group. Participants in the intervention group had a lower rate of receiving an antibiotic prescription for a suspected urinary tract infection compared with participants in the usual care group, with a rate ratio of 0.42 (95% confidence interval 0.26 to 0.68). No differences between intervention and control group were observed in the incidence of complications (<0.01 v 0.05 per person year), hospital referrals (<0.01 v 0.05), admissions to hospital (0.01 v 0.05), and mortality (0 v 0.01) within 21 days after suspected urinary tract infections, nor in all cause mortality (0.26 v 0.26).Implementation of a multifaceted antibiotic stewardship intervention safely reduced antibiotic prescribing for suspected urinary tract infections in frail older adults.ClinicalTrials.gov NCT03970356.

  • 2006
  • Ingår i: International Journal of Bifurcation and Chaos in Applied Sciences and Engineering. - 0218-1274. ; 16:3, s. 601-629
  • Tidskriftsartikel (refereegranskat)abstract
    • This paper proposes a strategy for the classification of codimension-two discontinuity-induced bifurcations of limit cycles in piecewise smooth systems of ordinary differential equations. Such nonsmooth transitions (also known as C-bifurcations) occur when the cycle interacts with a discontinuity boundary of phase space in a nongeneric way, such as grazing contact. Several such codimension-one events have recently been identified, causing for example, period-adding or sudden onset of chaos. Here, the focus is on codimension-two grazings that are local in the sense that the dynamics can be fully described by an appropriate Poincare map from a neighborhood of the grazing point (or points) of the critical cycle to itself. It is proposed that codimension-two grazing bifurcations can be divided into three distinct types: either the grazing point is degenerate, or the grazing cycle is itself degenerate (e.g. nonhyperbolic) or we have the simultaneous occurrence of two grazing events. A careful distinction is drawn between their occurrence in systems with discontinuous states, discontinuous vector fields, or that with discontinuity in some derivative of the vector field. Examples of each kind of bifurcation are presented, mostly derived from mechanical applications. For each example, where possible, principal bifurcation curves characteristic to the codimension-two scenario are presented and general features of the dynamics discussed. Many avenues for future research are opened.

  • Ingår i: Journal of Computational and Theoretical Transport. - : Informa UK Limited. - 2332-4309 .- 2332-4325. ; 48:7, s. 263-279
  • Tidskriftsartikel (refereegranskat)abstract
    • In this paper we study stability and convergence for hp-streamline diffusion (SD) finite element method for the, relativistic, time-dependent Vlasov-Maxwell (VM) system. We consider spatial domain and velocities The objective is to show globally optimal a priori error bound of order for the SD approximation of the VM system; where is the mesh size and is the spectral order. Our estimates rely on the local version with h(K) being the diameter of the phase-space-time element K and p(K) the spectral order for K. The optimal hp estimates require an exact solution in the Sobolev space Numerical implementations, performed for examples in one space- and two velocity dimensions, are justifying the robustness of the theoretical results.

  • Ingår i: Kinetic and Related Models. - : American Institute of Mathematical Sciences (AIMS). - 1937-5093 .- 1937-5077. ; 12:1, s. 105-131
  • Tidskriftsartikel (refereegranskat)abstract
    • We study stability and convergence of hp-streamline diffusion (SD) finite element, and Nitsche's schemes for the three dimensional, relativistic (3 spatial dimension and 3 velocities), time dependent Vlasov-Maxwell system and Maxwell's equations, respectively. For the hp scheme for the Vlasov-Maxwell system, assuming that the exact solution is in the Sobolev space HS+1(Omega), we derive global a priori error bound of order O(h/p)(s+1/2), where h(= max(K) h(K)) is the mesh parameter and p(= max(K) p(K)) is the spectral order. This estimate is based on the local version with h(K) = diam K being the diameter of the phase-space-time element K and pR-is the spectral order (the degree of approximating finite element polynomial) for K. As for the Nitsche's scheme, by a simple calculus of the field equations, first we convert the Maxwell's system to an elliptic type equation. Then, combining the Nitsche's method for the spatial discretization with a second order time scheme, we obtain optimal convergence of O(h(2) +k(2)), where h is the spatial mesh size and k is the time step. Here, as in the classical literature, the second order time scheme requires higher order regularity assumptions. Numerical justification of the results, in lower dimensions, is presented and is also the subject of a forthcoming computational work [22].

  • Ingår i: Blood. - : American Society of Hematology. - 0006-4971 .- 1528-0020. ; 123:1, s. 70-77
  • Tidskriftsartikel (refereegranskat)abstract
    • Children with Down syndrome (DS) have an increased risk of B-cell precursor (BCP) acute lymphoblastic leukemia (ALL). The prognostic factors and outcome of DS-ALL patients treated in contemporary protocols are uncertain. We studied 653 DS-ALL patients enrolled in 16 international trials from 1995 to 2004. Non-DS BCP-ALL patients from the Dutch Child Oncology Group and Berlin-Frankfurt-Munster were reference cohorts. DS-ALL patients had a higher 8-year cumulative incidence of relapse (26% +/- 2% vs 15% +/- 1%, P < .001) and 2-year treatment-related mortality (TRM) (7% +/- 1% vs 2.0% +/- < 1%, P < .0001) than non-DS patients, resulting in lower 8-year event-free survival (EFS) (64% +/- 2% vs 81% +/- 2%, P < .0001) and overall survival (74% +/- 2% vs 89% +/- 1%, P < .0001). Independent favorable prognostic factors include age <6 years (hazard ratio [HR] = 0.58, P = .002), white blood cell (WBC) count <10 x 10(9)/L (HR = 0.60, P = .005), and ETV6-RUNX1 (HR = 0.14, P = .006) for EFS and age (HR = 0.48, P < .001), ETV6-RUNX1 (HR = 0.1, P = .016) and high hyperdiploidy (HeH) (HR = 0.29, P = .04) for relapse-free survival. TRM was the major cause of death in ETV6-RUNX1 and HeH DS-ALLs. Thus, while relapse is the main contributor to poorer survival in DS-ALL, infection-associated TRM was increased in all protocol elements, unrelated to treatment phase or regimen. Future strategies to improve outcome in DS-ALL should include improved supportive care throughout therapy and reduction of therapy in newly identified good-prognosis subgroups.

  • 2002
  • Ingår i: Physica D. - 0167-2789 .- 1872-8022. ; 170:04-mar, s. 175-205
  • Tidskriftsartikel (refereegranskat)abstract
    • This paper is concerned with the analysis of so-called sliding bifurcations in n-dimensional piecewise-smooth dynamical systems with discontinuous vector field. These novel bifurcations occur when the system trajectory interacts with regions on the discontinuity set where sliding is possible. The derivation of appropriate normal-form maps is detailed. It is shown that the leading-order term in the map depends on the particular bifurcation scenario considered. This is in turn related to the possible bifurcation scenarios exhibited by a periodic orbit undergoing one of the sliding bifurcations discussed in the paper. A third-order relay system serves as a numerical example.

  • 2003

  • Tidskriftsartikel (refereegranskat)abstract
    • Recent investigations of nonsmooth dynamical systems have resulted in the study of a class of novel bifurcations termed as sliding bifurcations. These bifurcations are a characteristic feature of so-called Filippov systems, that is, systems of ordinary differential equations (ODEs) with discontinuous right-hand sides. In this paper we show that sliding bifurcations also play an important role in organizing the dynamics of dry friction oscillators, which are a subclass of nonsmooth systems. After introducing the possible codimension-1 sliding bifurcations of limit cycles, we show that these bifurcations organize different types of slip to stick-slip transitions in dry friction oscillators. In particular, we show both numerically and analytically that a sliding bifurcation is an important mechanism causing the sudden jump to chaos previously unexplained in the literature on friction systems. To analyze such bifurcations we make use of a new analytical method based on the study of appropriate normal form maps describing sliding bifurcations. Also, we explain the circumstances under which the theory of so-called border-collision bifurcations can be used in order to explain the onset of complex behavior in stick-slip systems.

  • Multifaceted antibiotic stewardship intervention using a participatory-action-research approach to improve antibiotic prescribing for urinary tract infections in frail elderly (ImpresU): study protocol for a European qualitative study followed by a pragmatic cluster randomised controlled trial
  • 2021
  • Ingår i: Bmj Open. - : BMJ. - 2044-6055. ; 11:10
  • Tidskriftsartikel (refereegranskat)abstract
    • Introduction Almost 60% of antibiotics in frail elderly are prescribed for alleged urinary tract infections (UTIs). A substantial part of this comprises prescriptions in case of non-specific symptoms or asymptomatic bacteriuria, for which the latest guidelines promote restrictiveness with antibiotics. We aim to reduce inappropriate antibiotic use for UTIs through an antibiotic stewardship intervention (ASI) that encourages to prescribe according to these guidelines. To develop an effective ASI, we first need a better understanding of the complex decision-making process concerning suspected UTIs in frail elderly. Moreover, the implementation approach requires tailoring to the heterogeneous elderly care setting. Methods and analysis First, we conduct a qualitative study to explore factors contributing to antibiotic prescribing for UTIs in frail elderly, using semi-structured interviews with general practitioners, nursing staff, patients and informal caregivers. Next, we perform a pragmatic cluster randomised controlled trial in elderly care organisations. A multifaceted ASI is implemented in the intervention group; the control group receives care as usual. The ASI is centred around a decision tool that promotes restrictive antibiotic use, supported by a toolbox with educational materials. For the implementation, we use a modified participatory-action-research approach, guided by the results of the qualitative study. The primary outcome is the number of antibiotic prescriptions for suspected UTIs. We aim to recruit 34 clusters with in total 680 frail elderly residents >= 70 years. Data collection takes place during a 5-month baseline period and a 7-month follow-up period. Finally, we perform a process evaluation. The study has been delayed for 6 months due to COVID-19 and is expected to end in July 2021. Ethics and dissemination Ethical approvals and/or waivers were obtained from the ethical committees in Poland, the Netherlands, Norway and Sweden. The results will be disseminated through publication in peer-reviewed journals and conference presentations.

  • Methenamine hippurate to prevent recurrent urinary tract infections in older women: protocol for a randomised, placebo-controlled trial (ImpresU)
  • 2022
  • Ingår i: BMJ Open. - : BMJ. - 2044-6055. ; 12:11
  • Tidskriftsartikel (refereegranskat)abstract
    • Introduction Methenamine hippurate is a urinary antiseptic used as preventive treatment for recurrent urinary tract infections (UTIs) in some Scandinavian countries. However, the scientific evidence for the preventive effect and safety for longer-term use is limited. The aim of this study is to assess whether methenamine hippurate can reduce the incidence of UTIs in older women with recurrent UTIs. Methods and analysis The ImpresU consortium is a collaboration between Norway, Sweden, Poland and the Netherlands. The study is a randomised, controlled, triple-blind phase IV clinical trial. Women ≥70 years with recurrent UTIs are screened for eligibility in a general practice setting. We aim to include 400 women in total, with 100 recruited from each collaborating country. The participants are randomised to treatment with methenamine hippurate 1 g or placebo tablets two times per day for a treatment period of 6 months, followed by a drug-free follow-up period of 6 months. The primary outcome is number of antibiotic treatments for UTIs during the treatment period. The secondary outcomes include number of antibiotic treatments for UTIs during the follow-up period and self-reported symptom of severity and duration of UTI episodes. Differences in complications between the treatment groups are measured as safety outcomes. We also aim to investigate whether strain characteristics or phylogenetic subgroups of Escherichia coli present in the urine culture at inclusion have a modifying effect on the outcomes. Ethics and dissemination Ethical approvals are obtained in all participating countries. The results will be communicated in peer-reviewed journals and at scientific conferences. Trial registration number ClinicalTrials.gov Registry (NCT04077580); EudraCT: 2018-002235-15.

    • Carnivores tend to exhibit a lack of (or less pronounced) genetic structure at continental scales in both a geographic and temporal sense and this can confound the identification of post-glacial colonization patterns in this group. In this study we used genome-wide data (using genotyping by sequencing [GBS]) to reconstruct the phylogeographic history of a widespread carnivore, the red fox (Vulpes vulpes), by investigating broad-scale patterns of genomic variation, differentiation and admixture amongst contemporary populations in Europe. Using 15,003 single nucleotide polymorphisms (SNPs) from 524 individuals allowed us to identify the importance of refugial regions for the red fox in terms of endemism (e.g., Iberia). In addition, we tested multiple post-glacial recolonization scenarios of previously glaciated regions during the Last Glacial Maximum using an Approximate Bayesian Computation (ABC) approach that were unresolved from previous studies. This allowed us to identify the role of admixture from multiple source population post-Younger Dryas in the case of Scandinavia and ancient land-bridges in the colonization of the British Isles. A natural colonization of Ireland was deemed more likely than an ancient human-mediated introduction as has previously been proposed and potentially points to a larger mammalian community on the island in the early post-glacial period. Using genome-wide data has allowed us to tease apart broad-scale patterns of structure and diversity in a widespread carnivore in Europe that was not evident from using more limited marker sets and provides a foundation for next-generation phylogeographic studies in other non-model species.

  • A codimension-two scenario of sliding solutions in grazing-sliding bifurcations
  • 2006
  • Ingår i: Nonlinearity. - : IOP Publishing. - 0951-7715 .- 1361-6544. ; 19:1, s. 1-26
  • Tidskriftsartikel (refereegranskat)abstract
    • This paper investigates codimension-two bifurcations that involve grazing-sliding and fold scenarios. An analytical unfolding of this novel codimension-two bifurcation is presented. Using the discontinuity mapping techniques it is shown that the fold curve is one-sided and cubically tangent to the grazing curve locally to the codimension-two point. This theory is then applied to explain the dynamics of a dry-friction oscillator where such a codimension-two point has been found. In particular, the presence and the character of essential bifurcation curves that merge at the codimension-two point are confirmed. This allows us to study the dynamics away from the codimension-two point using a piecewise affine approximation of the normal form for grazing-sliding bifurcations and explain the dynamics observed in the friction system.

  • Antiviral activity of tannic acid modified silver nanoparticles: Potential to activate immune response in herpes genitalis
  • 2018
  • Ingår i: Viruses. - : MDPI AG. - 1999-4915. ; 10:10
  • Tidskriftsartikel (refereegranskat)abstract
    • (1) Background: Tannic acid is a plant-derived polyphenol showing antiviral activity mainly because of an interference with the viral adsorption. In this work, we tested whether the modification of silver nanoparticles with tannic acid (TA-AgNPs) can provide a microbicide with additional adjuvant properties to treat genital herpes infection. (2) Methods: The mouse model of the vaginal herpes simplex virus 2 (HSV-2) infection was used to test immune responses after treatment of the primary infection with TA-AgNPs, and later, after a re-challenge with the virus. (3) Results: The mice treated intravaginally with TA-AgNPs showed better clinical scores and lower virus titers in the vaginal tissues soon after treatment. Following a re-challenge, the vaginal tissues treated with TA-AgNPs showed a significant increase in the percentages of IFN-gamma+ CD8+ T-cells, activated B cells, and plasma cells, while the spleens contained significantly higher percentages of IFN-gamma+ NK cells and effector-memory CD8+ T cells in comparison to NaCl-treated group. TA-AgNPs-treated animals also showed significantly better titers of anti-HSV-2 neutralization antibodies in sera; and (4) Conclusions: Our findings suggest that TA-AgNPs sized 33 nm can be an effective anti-viral microbicide to be applied upon the mucosal tissues with additional adjuvant properties enhancing an anti-HSV-2 immune response following secondary challenge.

  • The state of research into children with cancer across Europe : new policies for a new decade
  • 2011
  • Ingår i: ecancermedicalscience. - : Ecancer Global Foundation. - 1754-6605. ; 5, s. 210-
  • Tidskriftsartikel (refereegranskat)abstract
    • Overcoming childhood cancers is critically dependent on the state of research. Understanding how, with whom and what the research community is doing with childhood cancers is essential for ensuring the evidence-based policies at national and European level to support children, their families and researchers. As part of the European Union funded EUROCANCERCOMS project to study and integrate cancer communications across Europe, we have carried out new research into the state of research in childhood cancers. We are very grateful for all the support we have received from colleagues in the European paediatric oncology community, and in particular from Edel Fitzgerald and Samira Essiaf from the SIOP Europe office. This report and the evidence-based policies that arise from it come at a important junction for Europe and its Member States. They provide a timely reminder that research into childhood cancers is critical and needs sustainable long-term support.

  • Aging of preleukemic thymocytes drives CpG island hypermethylation in T-cell acute lymphoblastic leukemia
  • 2020
  • Ingår i: Blood cancer discovery. - : American Association for Cancer Research. - 2643-3249. ; 1:3, s. 274-289
  • Tidskriftsartikel (refereegranskat)abstract
    • Cancer cells display DNA hypermethylation at specific CpG islands in comparison to their normal healthy counterparts, but the mechanism that drives this so-called CpG island methylator phenotype (CIMP) remains poorly understood. Here, we show that CpG island methylation in human T-cell acute lymphoblastic leukemia (T-ALL) mainly occurs at promoters of Polycomb Repressor Complex 2 (PRC2) target genes that are not expressed in normal or malignant T-cells and which display a reciprocal association with H3K27me3 binding. In addition, we revealed that this aberrant methylation profile reflects the epigenetic history of T-ALL and is established already in pre-leukemic, self-renewing thymocytes that precede T-ALL development. Finally, we unexpectedly uncover that this age-related CpG island hypermethylation signature in T-ALL is completely resistant to the FDA-approved hypomethylating agent Decitabine. Altogether, we here provide conceptual evidence for the involvement of a pre-leukemic phase characterized by self-renewing thymocytes in the pathogenesis of human T-ALL.

  • Teenagers and young adults with cancer in Europe: from national programmes to a European integrated coordinated project.
  • 2015
  • Ingår i: European Journal of Cancer Care. - : Hindawi Limited. - 1365-2354 .- 0961-5423.
  • Tidskriftsartikel (refereegranskat)abstract
    • Over 14 000 patients aged 15-24 are estimated to be diagnosed with cancer in the European Union (EU) each year. Teenagers and young adults (TYA) often fall down gaps between children's and adults cancer services. The specific challenges of providing optimal care to them are described, but we present a summary of recent progress. Progress to overcome these challenges is happening at different rates across Europe. We summarise the European national projects in this field but more recently we have seen the beginnings of European coordination. Within the EU 7th Funding Programme (FP7) European Network for Cancer Research in Children and Adolescents programme (ENCCA), a specific European Network for Teenagers and Young Adults with Cancer has held a series of scientific meetings, including professionals, patients and caregivers. This group has proposed unanswered research questions and agreed key features of a high-quality service that can improve outcomes for TYA with cancer, including the primacy of collaboration between adult and paediatric services to eliminate the gap in the management of TYA with cancer.

  • Antibiotic Stewardship in European Nursing Homes: Experiences From the Netherlands, Norway, Poland, and Sweden
  • 2020
  • Ingår i: Journal of the American Medical Directors Association. - : Elsevier BV. - 1525-8610. ; 21:1
  • Tidskriftsartikel (refereegranskat)abstract
    • Antibiotics are among the most widely prescribed drugs in long-term care facilities, which highlights the importance of antibiotic stewardship (ABS) in this setting. In this article, we describe the experiences with ABS in nursing homes (NHs) from the perspective of 4 European countries: the Netherlands, Norway, Poland, and Sweden. In these countries, a large variety of initiatives to develop and implement ABS in NHs have been introduced in recent years. Among these initiatives are national antibiotic prescribing surveillance systems, NH-specific prescribing guidelines, and national networks of healthcare institutions that exchange information and develop ABS policy. Several initiatives evolved as a result of political prioritization of antibiotic resistance, translated into national action plans. Experiences of the 4 countries with the presented initiatives may inspire other countries that aim to develop or improve ABS in the long-term care setting.
