  • Arsenic Directly Binds to and Activates the Yeast AP-1-Like Transcription Factor Yap8.

    • The AP-1-like transcription factor Yap8 is critical for arsenic tolerance in the yeast Saccharomyces cerevisiae. However, the mechanism by which Yap8 senses the presence of arsenic and activates transcription of detoxification genes is unknown. Here we demonstrate that Yap8 directly binds to trivalent arsenite [As(III)] in vitro and in vivo and that approximately one As(III) molecule is bound per molecule of Yap8. As(III) is coordinated by three sulfur atoms in purified Yap8, and our genetic and biochemical data identify the cysteine residues that form the binding site as Cys132, Cys137, and Cys274. As(III) binding by Yap8 does not require an additional yeast protein, and Yap8 is regulated neither at the level of localization nor at the level of DNA binding. Instead, our data are consistent with a model in which a DNA-bound form of Yap8 acts directly as an As(III) sensor. Binding of As(III) to Yap8 triggers a conformational change that in turn brings about a transcriptional response. Thus, As(III) binding to Yap8 acts as a molecular switch that converts inactive Yap8 into an active transcriptional regulator. This is the first report to demonstrate how a eukaryotic protein couples arsenic sensing to transcriptional activation.

  • Spectroscopic insight into breast cancer: profiling small extracellular vesicles lipids via infrared spectroscopy for diagnostic precision

    • Breast cancer, a leading cause of female mortality due to delayed detection owing to asymptomatic nature and limited early diagnostic tools, was investigated using a multi-modal approach. Plasma-derived small EVs from breast cancer patients (BrCa, n = 74) and healthy controls (HC, n = 30) were analyzed. Small EVs (n = 104), isolated through chemical precipitation, underwent characterization via transmission electron microscopy (TEM) and nanoparticle tracking analysis (NTA). Validation involved antibody-based tests (TSG101, CD9, CD81, CD63). Infrared spectra of small EVs were obtained, revealing significant differences in lipid acyl chains, particularly in the C–H stretching of CH3. The study focused on the lipid region (3050–2900 cm−1), identifying peaks (3015 cm−1, 2960 cm−1, 2929 cm−1) as distinctive lipid characteristics. Spectroscopic lipid-to-lipid ratios [(I3015/I2929), (I2960/I2929)] emerged as prominent breast cancer markers. Exploration of protein, nucleic acid, and carbohydrate ratios indicated variations in alpha helices, asymmetric C–H stretching vibrations, and C–O stretching at 1033 cm−1. Principal component analysis (PCA) successfully differentiated BrCa and HC small EVs, and heatmap analysis and receiver operating characteristic (ROC) curve evaluations underscored the discriminatory power of lipid ratios. Notably, (I2960/I2929) exhibited 100% sensitivity and specificity, highlighting its potential as a robust BrCa sEV marker for breast cancer detection.

  • Hard X-Ray Polarization Catalog for a Five-year Sample of Gamma-Ray Bursts Using AstroSat CZT Imager

    • The Cadmium Zinc Telluride Imager (CZTI) on board AstroSat has been regularly detecting gamma-ray bursts (GRBs) since its launch in 2015. Its sensitivity to polarization measurements at energies above 100 keV allows CZTI to attempt spectropolarimetric studies of GRBs. Here, we present the first catalog of GRB polarization measurements made by CZTI during its first five years of operation. This includes the time-integrated polarization measurements of the prompt emission of 20 GRBs in the energy range 100-600 keV. The sample includes the bright GRBs that were detected within an angle range of 0 degrees-60 degrees and 120 degrees-180 degrees where the instrument has useful polarization sensitivity and is less prone to systematics. We implement a few new modifications in the analysis to enhance the polarimetric sensitivity of the instrument. The majority of the GRBs in the sample are found to possess less/null polarization across the total bursts' duration in contrast to a small fraction of five GRBs that exhibit high polarization. The low polarization across the bursts might be due either to the burst being intrinsically weakly polarized or to a varying polarization angle within the burst even when it is highly polarized. In comparison to POLAR measurements, CZTI has detected a larger number of cases with high polarization. This may be a consequence of the higher energy window of CZTI observations, which results in the sampling of a shorter duration of burst emissions than POLAR, thereby probing emissions with less temporal variation in polarization properties.

  • Sub-MeV spectroscopy with AstroSat-CZT imager for gamma ray bursts

    • Cadmium-Zinc-Telluride Imager (CZTI) onboard AstroSat has been a prolific Gamma-Ray Burst (GRB) monitor. While the 2-pixel Compton scattered events (100-300 keV) are used to extract sensitive spectroscopic information, the inclusion of the low-gain pixels (similar to 20% of the detector plane) after careful calibration extends the energy range of Compton energy spectra to 600 keV. The new feature also allows single-pixel spectroscopy of the GRBs to the sub-MeV range which is otherwise limited to 150 keV. We also introduced a new noise rejection algorithm in the analysis ('Compton noise'). These new additions not only enhances the spectroscopic sensitivity of CZTI, but the sub-MeV spectroscopy will also allow proper characterization of the GRBs not detected by Fermi. This article describes the methodology of single, Compton event and veto spectroscopy in 100-900 keV combined for the GRBs detected in the first year of operation. CZTI in last five years has detected similar to 20 bright GRBs. The new methodologies, when applied on the spectral analysis for this large sample of GRBs, has the potential to improve the results significantly and help in better understanding the prompt emission mechanism.

    • Today, social media technologies are being used for an array of applications including as platforms to crowdsource funds, information and products. One of the most used social media channels for business communication is Twitter. The objective of this study is to enhance the understanding about the use of Twitter in business relationships. Through 52 interviews conducted across 5 countries in 2015, it was found that Twitter users engaged in business-to-business relationship valued characteristics such as information quality, authenticity and industry influence in the people they followed. One of the primarily uses of Twitter in the B2B context was for information exchange and innovation. The study also conducted textual analysis using the Correlated Topic Modelling Method approach on the open ended responses concerning Twitter strategy and the interviewees' perception on favouring and retweeting behaviour. It was found that Twitter had a role to play in developing and supporting innovation.

    • Scanning probe microscopy (SPM) is a useful instrument to study the surface topography of the sample at an atomic resolution. SPM system is sensitive to vibration and contamination on the surface, and therefore a vibration-free and clean environment are required to improve spatial resolution in Z-height. In this article, a detailed new design of stand-alone cost-effective, vibration-free activated carbon-based cryosorption pump integrated and its detailed integration with the SPM system is discussed. The efficiency of cryosorption pump depends on the radial temperature distribution, which was analyzed using transient thermal analysis in Ansys software. The temperature profile obtained from the thermal simulation results reveals that a spiral wound copper mesh-based partition surrounding the activated carbon in the cryosorption pump improves the temperature distribution in the radial direction. The pump can operate with a maximum of 25 L of liquid nitrogen consumption and 4.5 h of continuous operation. The activated carbon was used as an adsorbent showed a hexagonal crystal structure from the X-ray diffraction (XRD) studies and its average pore size was obtained from by Brunauer-Emmett-Teller (BET) analysis was 0.34 nm. These tests were conducted to determine the effect on the pump-down characteristics of the developed cryosorption pump. The improvement in spatial resolution was studied through detailed experimental analysis in conductive atomic force microscopy (CAFM) by piezo force microscopy (PFM) which showed a 50% decrease of the current when imaged in a vacuum environment, phase and magnitude images showed less distorted and clear visibility of the piezo domains. The magnetic interaction by magnetic force microscopy (MFM) Q-factor was studied at the resonance frequency of 61.5 kHz. MFM showed that the Q-factor improved from 20 to 500 at 1E-3 mbar operating pressure. The drift of the MFM tip with respect to holding time showed a saturated Q-factor at 500. Noncontact mode by atomic force microscopy (AFM) showed a spatial resolution enhancement of 0.89 nm.

    • Conductivity, carrier mobility, and a suitable Gibbs free energy are important criteria that determine the performance of catalysts for a hydrogen evolution reaction (HER). However, it is a challenge to combine these factors into a single compound. Herein, we discover a superior electrocatalyst for a HER in the recently identified Dirac nodal arc semimetal PtSn4. The determined turnover frequency (TOF) for each active site of PtSn4 is 1.54 H-2 s(-1) at 100 mV. This sets a benchmark for HER catalysis on Pt-based noble metals and earth-abundant metal catalysts. We make use of the robust surface states of PtSn4 as their electrons can be transferred to the adsorbed hydrogen atoms in the catalytic process more efficiently. In addition, PtSn4 displays excellent chemical and electrochemical stabilities after long-term exposure in air and long-time HER stability tests.

  • A tale of exosomes and their implication in cancer

    • Cancer is a cause of high deaths worldwide and also a huge burden for the health system. Cancer cells have unique properties such as a high rate of proliferation, self-renewal, metastasis, and treatment resistance, therefore, the development of novel diagnoses of cancers is a tedious task. Exosomes are secreted by virtually all cell types and have the ability to carry a multitude of biomolecules crucial for intercellular communication, hence, contributing a crucial part in the onset and spread of cancer. These exosomal components can be utilized in the development of markers for diagnostic and prognostic purposes for various cancers. This review emphasized primarily the following topics: exosomes structure and functions, isolation and characterization strategies of exosomes, the role of exosomal contents in cancer with a focus in particular on noncoding RNA and protein, exosomes, and the cancer microenvironment interactions, cancer stem cells, and tumor diagnosis and prognosis based on exosomes.
