    • In 2008 we published the first set of guidelines for standardizing research in autophagy. Since then, research on this topic has continued to accelerate, and many new scientists have entered the field. Our knowledge base and relevant new technologies have also been expanding. Accordingly, it is important to update these guidelines for monitoring autophagy in different organisms. Various reviews have described the range of assays that have been used for this purpose. Nevertheless, there continues to be confusion regarding acceptable methods to measure autophagy, especially in multicellular eukaryotes. A key point that needs to be emphasized is that there is a difference between measurements that monitor the numbers or volume of autophagic elements (e.g., autophagosomes or autolysosomes) at any stage of the autophagic process vs. those that measure flux through the autophagy pathway (i.e., the complete process); thus, a block in macroautophagy that results in autophagosome accumulation needs to be differentiated from stimuli that result in increased autophagic activity, defined as increased autophagy induction coupled with increased delivery to, and degradation within, lysosomes (in most higher eukaryotes and some protists such as Dictyostelium) or the vacuole (in plants and fungi). In other words, it is especially important that investigators new to the field understand that the appearance of more autophagosomes does not necessarily equate with more autophagy. In fact, in many cases, autophagosomes accumulate because of a block in trafficking to lysosomes without a concomitant change in autophagosome biogenesis, whereas an increase in autolysosomes may reflect a reduction in degradative activity. Here, we present a set of guidelines for the selection and interpretation of methods for use by investigators who aim to examine macroautophagy and related processes, as well as for reviewers who need to provide realistic and reasonable critiques of papers that are focused on these processes. These guidelines are not meant to be a formulaic set of rules, because the appropriate assays depend in part on the question being asked and the system being used. In addition, we emphasize that no individual assay is guaranteed to be the most appropriate one in every situation, and we strongly recommend the use of multiple assays to monitor autophagy. In these guidelines, we consider these various methods of assessing autophagy and what information can, or cannot, be obtained from them. Finally, by discussing the merits and limits of particular autophagy assays, we hope to encourage technical innovation in the field.

    • The International Cancer Genome Consortium (ICGC) was launched to coordinate large-scale cancer genome studies in tumours from 50 different cancer types and/or subtypes that are of clinical and societal importance across the globe. Systematic studies of more than 25,000 cancer genomes at the genomic, epigenomic and transcriptomic levels will reveal the repertoire of oncogenic mutations, uncover traces of the mutagenic influences, define clinically relevant subtypes for prognosis and therapeutic management, and enable the development of new cancer therapies.

    • The fourth generation of the Sloan Digital Sky Survey (SDSS-IV) has been in operation since 2014 July. This paper describes the second data release from this phase, and the 14th from SDSS overall (making this Data Release Fourteen or DR14). This release makes the data taken by SDSS-IV in its first two years of operation (2014-2016 July) public. Like all previous SDSS releases, DR14 is cumulative, including the most recent reductions and calibrations of all data taken by SDSS since the first phase began operations in 2000. New in DR14 is the first public release of data from the extended Baryon Oscillation Spectroscopic Survey; the first data from the second phase of the Apache Point Observatory (APO) Galactic Evolution Experiment (APOGEE-2), including stellar parameter estimates from an innovative data-driven machine-learning algorithm known as "The Cannon"; and almost twice as many data cubes from the Mapping Nearby Galaxies at APO (MaNGA) survey as were in the previous release (N = 2812 in total). This paper describes the location and format of the publicly available data from the SDSS-IV surveys. We provide references to the important technical papers describing how these data have been taken (both targeting and observation details) and processed for scientific use. The SDSS web site (www.sdss.org) has been updated for this release and provides links to data downloads, as well as tutorials and examples of data use. SDSS-IV is planning to continue to collect astronomical data until 2020 and will be followed by SDSS-V.

    • We describe the Sloan Digital Sky Survey IV (SDSS-IV), a project encompassing three major spectroscopic programs. The Apache Point Observatory Galactic Evolution Experiment 2 (APOGEE-2) is observing hundreds of thousands of Milky Way stars at high resolution and. high signal-to-noise ratios in the near-infrared. The Mapping Nearby Galaxies at Apache Point Observatory (MaNGA) survey is obtaining spatially resolved spectroscopy for thousands of nearby galaxies (median z similar to 0.03). The extended Baryon Oscillation Spectroscopic Survey (eBOSS) is mapping the galaxy, quasar, and neutral gas distributions between z similar to 0.6 and 3.5 to constrain cosmology using baryon acoustic oscillations, redshift space distortions, and the shape of the power spectrum. Within eBOSS, we are conducting two major subprograms: the SPectroscopic IDentification of eROSITA Sources (SPIDERS), investigating X-ray AGNs. and galaxies in X-ray clusters, and the Time Domain Spectroscopic Survey (TDSS), obtaining spectra of variable sources. All programs use the 2.5 m Sloan Foundation Telescope at the. Apache Point Observatory; observations there began in Summer 2014. APOGEE-2 also operates a second near-infrared spectrograph at the 2.5 m du Pont Telescope at Las Campanas Observatory, with observations beginning in early 2017. Observations at both facilities are scheduled to continue through 2020. In keeping with previous SDSS policy, SDSS-IV provides regularly scheduled public data releases; the first one, Data Release 13, was made available in 2016 July.

    • This paper documents the seventeenth data release (DR17) from the Sloan Digital Sky Surveys; the fifth and final release from the fourth phase (SDSS-IV). DR17 contains the complete release of the Mapping Nearby Galaxies at Apache Point Observatory (MaNGA) survey, which reached its goal of surveying over 10,000 nearby galaxies. The complete release of the MaNGA Stellar Library accompanies this data, providing observations of almost 30,000 stars through the MaNGA instrument during bright time. DR17 also contains the complete release of the Apache Point Observatory Galactic Evolution Experiment 2 survey that publicly releases infrared spectra of over 650,000 stars. The main sample from the Extended Baryon Oscillation Spectroscopic Survey (eBOSS), as well as the subsurvey Time Domain Spectroscopic Survey data were fully released in DR16. New single-fiber optical spectroscopy released in DR17 is from the SPectroscipic IDentification of ERosita Survey subsurvey and the eBOSS-RM program. Along with the primary data sets, DR17 includes 25 new or updated value-added catalogs. This paper concludes the release of SDSS-IV survey data. SDSS continues into its fifth phase with observations already underway for the Milky Way Mapper, Local Volume Mapper, and Black Hole Mapper surveys.

  • Anthropological contributions to historical ecology : 50 questions, infinite prospects

    • This paper presents the results of a consensus-driven process identifying 50 priority research questions for historical ecology obtained through crowdsourcing, literature reviews, and in-person workshopping. A deliberative approach was designed to maximize discussion and debate with defined outcomes. Two in-person workshops (in Sweden and Canada) over the course of two years and online discussions were peer facilitated to define specific key questions for historical ecology from anthropological and archaeological perspectives. The aim of this research is to showcase the variety of questions that reflect the broad scope for historical-ecological research trajectories across scientific disciplines. Historical ecology encompasses research concerned with decadal, centennial, and millennial human-environmental interactions, and the consequences that those relationships have in the formation of contemporary landscapes. Six interrelated themes arose from our consensus-building workshop model: (1) climate and environmental change and variability; (2) multi-scalar, multi-disciplinary; (3) biodiversity and community ecology; (4) resource and environmental management and governance; (5) methods and applications; and (6) communication and policy. The 50 questions represented by these themes highlight meaningful trends in historical ecology that distill the field down to three explicit findings. First, historical ecology is fundamentally an applied research program. Second, this program seeks to understand long-term human-environment interactions with a focus on avoiding, mitigating, and reversing adverse ecological effects. Third, historical ecology is part of convergent trends toward transdisciplinary research science, which erodes scientific boundaries between the cultural and natural.

    • Recent years have seen an increase in the amount of statistics describing different phenomena based on Big Data. This term includes data characterized not only by their large volume, but also by their variety and velocity, the organic way in which they are created, and the new types of processes needed to analyze them and make inference from them. The change in the nature of the new types of data, their availability, and the way in which they are collected and disseminated is fundamental. This change constitutes a paradigm shift for survey research. There is great potential in Big Data, but there are some fundamental challenges that have to be resolved before its full potential can be realized. This report provides examples of different types of Big Data and their potential for survey research; it also describes the Big Data process, discusses its main challenges, and considers solutions and research needs.

    • Renal cell carcinoma (RCC) has an undisputed genetic component and a stable 2:1 male to female sex ratio in its incidence across populations, suggesting possible sexual dimorphism in its genetic susceptibility. We conducted the first sex-specific genome-wide association analysis of RCC for men (3227 cases, 4916 controls) and women (1992 cases, 3095 controls) of European ancestry from two RCC genome-wide scans and replicated the top findings using an additional series of men (2261 cases, 5852 controls) and women (1399 cases, 1575 controls) from two independent cohorts of European origin. Our study confirmed sex-specific associations for two known RCC risk loci at 14q24.2 (DPF3) and 2p21(EPAS1). We also identified two additional suggestive male-specific loci at 6q24.3 (SAMD5, male odds ratio (ORmale) = 0.83 [95% CI = 0.78-0.89], Pmale = 1.71 × 10-8 compared with female odds ratio (ORfemale) = 0.98 [95% CI = 0.90-1.07], Pfemale = 0.68) and 12q23.3 (intergenic, ORmale = 0.75 [95% CI = 0.68-0.83], Pmale = 1.59 × 10-8 compared with ORfemale = 0.93 [95% CI = 0.82-1.06], Pfemale = 0.21) that attained genome-wide significance in the joint meta-analysis. Herein, we provide evidence of sex-specific associations in RCC genetic susceptibility and advocate the necessity of larger genetic and genomic studies to unravel the endogenous causes of sex bias in sexually dimorphic traits and diseases like RCC.

  • Genetic Variants Related to Longer Telomere Length are Associated with Increased Risk of Renal Cell Carcinoma.

    • BACKGROUND: Relative telomere length in peripheral blood leukocytes has been evaluated as a potential biomarker for renal cell carcinoma (RCC) risk in several studies, with conflicting findings.OBJECTIVE: We performed an analysis of genetic variants associated with leukocyte telomere length to assess the relationship between telomere length and RCC risk using Mendelian randomization, an approach unaffected by biases from temporal variability and reverse causation that might have affected earlier investigations.DESIGN, SETTING, AND PARTICIPANTS: Genotypes from nine telomere length-associated variants for 10 784 cases and 20 406 cancer-free controls from six genome-wide association studies (GWAS) of RCC were aggregated into a weighted genetic risk score (GRS) predictive of leukocyte telomere length.OUTCOME MEASUREMENTS AND STATISTICAL ANALYSIS: Odds ratios (ORs) relating the GRS and RCC risk were computed in individual GWAS datasets and combined by meta-analysis.RESULTS AND LIMITATIONS: Longer genetically inferred telomere length was associated with an increased risk of RCC (OR=2.07 per predicted kilobase increase, 95% confidence interval [CI]:=1.70-2.53, p<0.0001). As a sensitivity analysis, we excluded two telomere length variants in linkage disequilibrium (R2>0.5) with GWAS-identified RCC risk variants (rs10936599 and rs9420907) from the telomere length GRS; despite this exclusion, a statistically significant association between the GRS and RCC risk persisted (OR=1.73, 95% CI=1.36-2.21, p<0.0001). Exploratory analyses for individual histologic subtypes suggested comparable associations with the telomere length GRS for clear cell (N=5573, OR=1.93, 95% CI=1.50-2.49, p<0.0001), papillary (N=573, OR=1.96, 95% CI=1.01-3.81, p=0.046), and chromophobe RCC (N=203, OR=2.37, 95% CI=0.78-7.17, p=0.13).CONCLUSIONS: Our investigation adds to the growing body of evidence indicating some aspect of longer telomere length is important for RCC risk.PATIENT SUMMARY: Telomeres are segments of DNA at chromosome ends that maintain chromosomal stability. Our study investigated the relationship between genetic variants associated with telomere length and renal cell carcinoma risk. We found evidence suggesting individuals with inherited predisposition to longer telomere length are at increased risk of developing renal cell carcinoma.

  • Reassigning the shapes of the 0+ states in the 186Pb nucleus

    • Across the physics disciplines, the 186Pb nucleus is the only known system, where the two first excited states, together with the ground state, form a triplet of zero-spin states assigned with prolate, oblate and spherical shapes. Here we report on a precision measurement where the properties of collective transitions in 186Pb were determined in a simultaneous in-beam γ-ray and electron spectroscopy experiment employing the recoil-decay tagging technique. The feeding of the 02+ state and the interband 22+→21+ transition have been observed. We also present direct measurement of the energies of the electric monopole transitions from the excited 0+ states to the 0+ ground state. In contrast to the earlier understanding, the obtained reduced transition probability B(E2;21+→02+) value of 190(80) W.u., the transitional quadrupole moment ∣Qt(21+→02+)∣=7.7(33) eb and intensity balance arguments provide evidence to reassign the 02+ and 03+ states with predominantly prolate and oblate shape, respectively. Our work demonstrates a step-up in experimental sensitivity and paves the way for systematic studies of electric monopole transitions in this region. These electric monopole transitions probe the nuclear volume in a unique manner and provide unexploited input for development of the next-generation energy density functional models.

    • Previous genome-wide association studies (GWAS) have identified six risk loci for renal cell carcinoma (RCC). We conducted a meta-analysis of two new scans of 5,198 cases and 7,331 controls together with four existing scans, totalling 10,784 cases and 20,406 controls of European ancestry. Twenty-four loci were tested in an additional 3,182 cases and 6,301 controls. We confirm the six known RCC risk loci and identify seven new loci at 1p32.3 (rs4381241, P=3.1 × 10-10), 3p22.1 (rs67311347, P=2.5 × 10-8), 3q26.2 (rs10936602, P=8.8 × 10-9), 8p21.3 (rs2241261, P=5.8 × 10-9), 10q24.33-q25.1 (rs11813268, P=3.9 × 10-8), 11q22.3 (rs74911261, P=2.1 × 10-10) and 14q24.2 (rs4903064, P=2.2 × 10-24). Expression quantitative trait analyses suggest plausible candidate genes at these regions that may contribute to RCC susceptibility.

    • Insulin‐like growth factors (IGFs) and insulin‐like growth factor binding proteins (IGFBPs) have been implicated in the aetiology of several cancers. To better understand whether anthropometric, behavioural and sociodemographic factors may play a role in cancer risk via IGF signalling, we examined the cross‐sectional associations of these exposures with circulating concentrations of IGFs (IGF‐I and IGF‐II) and IGFBPs (IGFBP‐1, IGFBP‐2 and IGFBP‐3). The Endogenous Hormones, Nutritional Biomarkers and Prostate Cancer Collaborative Group dataset includes individual participant data from 16,024 male controls (i.e. without prostate cancer) aged 22–89 years from 22 prospective studies. Geometric means of protein concentrations were estimated using analysis of variance, adjusted for relevant covariates. Older age was associated with higher concentrations of IGFBP‐1 and IGFBP‐2 and lower concentrations of IGF‐I, IGF‐II and IGFBP‐3. Higher body mass index was associated with lower concentrations of IGFBP‐1 and IGFBP‐2. Taller height was associated with higher concentrations of IGF‐I and IGFBP‐3 and lower concentrations of IGFBP‐1. Smokers had higher concentrations of IGFBP‐1 and IGFBP‐2 and lower concentrations of IGFBP‐3 than nonsmokers. Higher alcohol consumption was associated with higher concentrations of IGF‐II and lower concentrations of IGF‐I and IGFBP‐2. African Americans had lower concentrations of IGF‐II, IGFBP‐1, IGFBP‐2 and IGFBP‐3 and Hispanics had lower IGF‐I, IGF‐II and IGFBP‐3 than non‐Hispanic whites. These findings indicate that a range of anthropometric, behavioural and sociodemographic factors are associated with circulating concentrations of IGFs and IGFBPs in men, which will lead to a greater understanding of the mechanisms through which these factors influence cancer risk.

  • Interconnections : Glass beads and trade in southern and eastern Africa and the Indian Ocean - 7th to 16th centuries AD

    • Glass beads comprise the most frequently found evidence of trade between southern Africa and the greater Indian Oceanbetween the 7th and 16th centuries AD.  In this thesis beads recovered from southern African archaeological sites are organized into series, based on morphology and chemical composition determined by LA-ICP-MS analysis.  The results are used to interpret the trade patterns and partners that linked eastern Africa to the rest of the Indian Ocean world, as well as interconnections between southern Africa andEast Africa.   Comprehensive reports on bead assemblages from several archaeological sites are presented, including: Mapungubwe, K2 and Schroda in the Shashe-Limpopo Basin; Chibuene in southern Mozambique; Hlamba Mlonga in eastern Zimbabwe; Sibudu Cave in KwaZulu-Natal, Kaole Ruins in Tanzania and Mahilaka in northwest Madagascar.  The conclusions reached show that trade relationships and socio-political development in the south were different from those on the East Coast and that changes in bead series in the south demonstrate it was fully integrated into the cycles of the Eurasian and African world-system.

  • Circulating Metabolic Biomarkers of Screen-Detected Prostate Cancer in the ProtecT Study

    • BACKGROUND: Whether associations between circulating metabolites and prostate cancer are causal is unknown. We report on the largest study of metabolites and prostate cancer (2,291 cases and 2,661 controls) and appraise causality for a subset of the prostate cancer-metabolite associations using two-sample Mendelian randomization (MR).MATERIALS AND METHODS: The case-control portion of the study was conducted in nine UK centres with men aged 50-69 years who underwent prostate-specific antigen (PSA) screening for prostate cancer within the Prostate testing for cancer and Treatment (ProtecT) trial. Two data sources were used to appraise causality: a genome-wide association study (GWAS) of metabolites in 24,925 participants and a GWAS of prostate cancer in 44,825 cases and 27,904 controls within the Association Group to Investigate Cancer Associated Alterations in the Genome (PRACTICAL) consortium.RESULTS: Thirty-five metabolites were strongly associated with prostate cancer (p <0.0014, multiple-testing threshold). These fell into four classes: i) lipids and lipoprotein subclass characteristics (total cholesterol and ratios, cholesterol esters and ratios, free cholesterol and ratios, phospholipids and ratios, and triglyceride ratios); ii) fatty acids and ratios; iii) amino acids; iv) and fluid balance. Fourteen top metabolites were proxied by genetic variables, but MR indicated these were not causal.CONCLUSIONS: We identified 35 circulating metabolites associated with prostate cancer presence, but found no evidence of causality for those 14 testable with MR. Thus, the 14 MR-tested metabolites are unlikely to be mechanistically important in prostate cancer risk.IMPACT: The metabolome provides a promising set of biomarkers that may aid prostate cancer classification.

  • Two Phase 3 Trials of Gantenerumab in Early Alzheimer's Disease

    • Background: Monoclonal antibodies that target amyloid-beta (Aβ) have the potential to slow cognitive and functional decline in persons with early Alzheimer's disease. Gantenerumab is a subcutaneously administered, fully human, anti-Aβ IgG1 monoclonal antibody with highest affinity for aggregated Aβ that has been tested for the treatment of Alzheimer's disease. Methods: We conducted two phase 3 trials (GRADUATE I and II) involving participants 50 to 90 years of age with mild cognitive impairment or mild dementia due to Alzheimer's disease and evidence of amyloid plaques on positron-emission tomography (PET) or cerebrospinal fluid (CSF) testing. Participants were randomly assigned to receive gantenerumab or placebo every 2 weeks. The primary outcome was the change from baseline in the score on the Clinical Dementia Rating scale-Sum of Boxes (CDR-SB; range, 0 to 18, with higher scores indicating greater cognitive impairment) at week 116. Results: A total of 985 and 980 participants were enrolled in the GRADUATE I and II trials, respectively. The baseline CDR-SB score was 3.7 in the GRADUATE I trial and 3.6 in the GRADUATE II trial. The change from baseline in the CDR-SB score at week 116 was 3.35 with gantenerumab and 3.65 with placebo in the GRADUATE I trial (difference, -0.31; 95% confidence interval [CI], -0.66 to 0.05; P = 0.10) and was 2.82 with gantenerumab and 3.01 with placebo in the GRADUATE II trial (difference, -0.19; 95% CI, -0.55 to 0.17; P = 0.30). At week 116, the difference in the amyloid level on PET between the gantenerumab group and the placebo group was -66.44 and -56.46 centiloids in the GRADUATE I and II trials, respectively, and amyloid-negative status was attained in 28.0% and 26.8% of the participants receiving gantenerumab in the two trials. Across both trials, participants receiving gantenerumab had lower CSF levels of phosphorylated tau 181 and higher levels of Aβ42 than those receiving placebo; the accumulation of aggregated tau on PET was similar in the two groups. Amyloid-related imaging abnormalities with edema (ARIA-E) occurred in 24.9% of the participants receiving gantenerumab, and symptomatic ARIA-E occurred in 5.0%. Conclusions: Among persons with early Alzheimer's disease, the use of gantenerumab led to a lower amyloid plaque burden than placebo at 116 weeks but was not associated with slower clinical decline.

    • The spectre of overgrazing' looms large in historical and political narratives of ecological degradation in savannah ecosystems. While pastoral exploitation is a conspicuous driver of landscape variability and modification, assumptions that such change is inevitable or necessarily negative deserve to be continuously evaluated and challenged. With reference to three case studies from Kenya - the Laikipia Plateau, the Lake Baringo basin, and the Amboseli ecosystem - we argue that the impacts of pastoralism are contingent on the diachronic interactions of locally specific environmental, political, and cultural conditions. The impacts of the compression of rangelands and restrictions on herd mobility driven by misguided conservation and economic policies are emphasised over outdated notions of pastoralist inefficiency. We review the application of overgrazing' in interpretations of the archaeological record and assess its relevance for how we interpret past socio-environmental dynamics. Any discussion of overgrazing, or any form of human-environment interaction, must acknowledge spatio-temporal context and account for historical variability in landscape ontogenies.

    • This paper considers the ecological residues of pastoralist occupation at the site of Maili Sita in Laikipia, central Kenya, drawing links with the archaeological record so as to contribute a fresh approach to the ephemeral settlement sites of mobile herding communities, a methodological aspect of African archaeology that remains problematic. Variations in the geochemical and micromorphological composition of soils along transects across the site are compared with vegetation distributions and satellite imagery to propose an occupation pattern not dissimilar to contemporary Cushitic-speaking groups further north. We argue that Maili Sita exemplifies the broad migratory and cultural exchange networks in place during the mid- to late second millennium AD, with pastoralist occupants who were both physically and culturally mobile.

    • Investigations of the impacts of past volcanic eruptions on climate, environment, and society require accurate chronologies. However, eruptions that are not recorded in historical documents can seldom be dated exactly. Here we use annually resolved radiocarbon (C-14) measurements to isolate the 775 CE cosmogenic C-14 peak in a subfossil birch tree that was buried by a glacial outburst flood in southern Iceland. We employ this absolute time marker to date a subglacial eruption of Katla volcano at late 822 CE to early 823 CE. We argue for correlation between the 822-823 CE eruption and a conspicuous sulfur anomaly evident in Greenland ice cores, which follows in the wake of an even larger volcanic signal (ca. 818-820 CE) as yet not attributed to a known eruption. An abrupt summer cooling in 824 CE, evident in tree-ring reconstructions for Fennoscandia and the Northern Hemisphere, suggests a climatic response to the Katla eruption. Written historical sources from Europe and China corroborate our proposed tree ring-radiocarbon-ice core linkage but also point to combined effects of eruptions occurring during this period. Our study describes the oldest precisely dated, high-latitude eruption and reveals the impact of an extended phase of volcanic forcing in the early 9th century. It also provides insight into the existence of prehistoric woodland cover and the nature of volcanism several decades before Iceland's permanent settlement began.

    • East African elephants have been hunted for their ivory for millennia but the nineteenth century witnessed strongly escalating demand from Europe and North America. It has been suggested that one consequence was that by the 1880s elephant herds along the coast had become scarce, and to meet demand, trade caravans trekked farther into interior regions of East Africa, extending the extraction frontier. The steady decimation of elephant populations coupled with the extension of trade networks have also been claimed to have triggered significant ecological and socio-economic changes that left lasting legacies across the region. To explore the feasibility of using an isotopic approach to uncover a 'moving frontier' of elephant extraction, we constructed a baseline isotope data set (delta C-13, delta N-15, delta O-18 and Sr-87/Sr-86) for historic East African elephants known to have come from three distinct regions (coastal, Rift Valley, and inland Lakes). Using the isotope results with other climate data and geographical mapping tools, it was possible to characterise elephants from different habitats across the region. This baseline data set was then used to provenance elephant ivory of unknown geographical provenance that was exported from East Africa during the late nineteenth and early twentieth centuries to determine its likely origin. This produced a better understanding of historic elephant geography in the region, and the data have the potential to be used to provenance older archaeological ivories, and to inform contemporary elephant conservation strategies.

    • In this paper, we review the potential of biocultural heritage in biodiversity protection and agricultural innovation in sub-Saharan Africa. We begin by defining the concept of biocultural heritage into four interlinked elements that are revealed through integrated landscape analysis. This concerns the transdisciplinary methods whereby biocultural heritage must be explored, and here we emphasise that reconstructing landscape histories and documenting local heritage values needs to be an integral part of the process. Ecosystem memories relate to the structuring of landscape heterogeneity through such activities as agroforestry and fire management. The positive linkages between living practices, biodiversity and soil nutrients examined here are demonstrative of the concept of ecosystem memories. Landscape memories refer to built or enhanced landscapes linked to specific land-use systems and property rights. Place memories signify practices of protection or use related to a specific place. Customary protection of burial sites and/or abandoned settlements, for example, is a common occurrence across Africa with beneficial outcomes for biodiversity and forest protection. Finally, we discuss stewardship and change. Building on local traditions, inclusivity and equity are essential to promoting the continuation and innovation of practices crucial for local sustainability and biodiversity protection, and also offer new avenues for collaboration in landscape management and conservation.

    • The structural changes of water upon deep supercooling were studied through wide-angle x-ray scattering at SwissFEL. The experimental setup had a momentum transfer range of 4.5 Å-1, which covered the principal doublet of the x-ray structure factor of water. The oxygen-oxygen structure factor was obtained for temperatures down to 228.5 ± 0.6 K. Similar to previous studies, the second diffraction peak increased strongly in amplitude as the structural change accelerated toward a local tetrahedral structure upon deep supercooling. We also observed an anomalous trend for the second peak position of the oxygen-oxygen structure factor (q2). We found that q2 exhibits an unprecedented positive partial derivative with respect to temperature for temperatures below 236 K. Based on Fourier inversion of our experimental data combined with reference data, we propose that the anomalous q2 shift originates from that a repeat spacing in the tetrahedral network, associated with all peaks in the oxygen-oxygen pair-correlation function, gives rise to a less dense local ordering that resembles that of low-density amorphous ice. The findings are consistent with that liquid water consists of a pentamer-based hydrogen-bonded network with low density upon deep supercooling.

    • A lifetime analysis using the Doppler-shift attenuation method has been performed on the Tellurium isotopes Te-110,Te-112. The experiment was performed using the Gammasphere array in conjunction with the MICROBALL charged-particle detector. Three strongly coupled bands were previously established in Te-110,Te-112 which were observed up to unusually high spins. In the current experiment, it has been possible to extract lifetime measurements using a Doppler broadened lineshape analysis on one of the Delta I = 1 band structures in Te-110. In contrast to similar Delta I = 1 structures in other nuclei in this mass region, the extracted B(M1) values did not rapidly decrease with increasing angular momentum. Instead, the strongly coupled band in Te-110 represents a deformed 1p-1h structure, rather than a weakly deformed structure showing the shears mechanism.

    • In this article, we examine an assumption about the historic Swahili of the eastern African coast: that they were a maritime society from their beginnings in the first millennium C.E. Based on historical and archaeological data, we suggest that, despite their proximity to and use of the sea, the level of maritimity of Swahili society increased greatly over time and was only fully realized in the early second millennium C.E. Drawing on recent theorizing from other areas of the world about maritimity as well as research on the Swahili, we discuss three arenas that distinguish first- and second-millennium coastal society in terms of their maritime orientation. These are variability and discontinuity in settlement location and permanence; evidence of increased engagement with the sea through fishing and sailing technology; and specialized architectural developments involving port facilities, mosques, and houses. The implications of this study are that we must move beyond coastal location in determining maritimity; consider how the sea and its products were part of social life; and assess whether the marine environment actively influences and is influenced by broader patterns of sociocultural organization, practice, and belief within Swahili and other societies.

    • A beam of 28 MeV He-8 particles with an average intensity 2 x 10(5) ions s(-1), produced by the SPIRAL facility at GANIL, was used to initiate fusion-evaporation reactions in thick targets of Pb-201 and Bi-209. Excited states of Po-212 and At-213 were populated in (He-8,4n) reactions. The emitted gamma-rays were detected using the EXOGAM array composed of four Compton suppressed germanium Clover detectors in a close geometry configuration. Preliminary results show that Po-212 has been populated to high spin (I similar or equal to 14) with a significantly increased yield compared with our previous He-8 beam experiment. An apparently increased relative population of states up to 12(+) was observed compared with published results from a Pb-208(Be-9,alpha n)(212) Po reaction at 48 MeV. Evidence is presented for a previously unreported de-excitation pathway: 69 keV (13(-) -> 12(+)). States up to I-pi = (39/2(-)) have also been observed in At-213.

    • This study presents a multidisciplinary perspective for understanding environmental change and emerging socio-ecological interactions across the Amboseli region of southwestern Kenya. We focus on late Holocene (<5,000 cal yr. BP) changes and continuities reconstructed from sedimentary, archeological, historical records and socio-ecological models. We utilize multi-disciplinary approaches to understand environmental-ecosystem-social interactions over the longue durée and use this to simulate different land use scenarios supporting conservation and sustainable livelihoods using a socio-ecological model. Today the semi-arid Amboseli landscape supports a large livestock and wildlife population, sustained by a wide variety of plants and extensive rangelands regulated by seasonal rainfall and human activity. Our data provide insight into how large-scale and long-term interactions of climate, people, livestock, wildlife and external connections have shaped the ecosystems across the Amboseli landscape. Environmental conditions were dry between ~5,000 and 2,000 cal yr. BP, followed by two wet periods at ~2,100–1,500 and 1,400–800 cal yr. BP with short dry periods; the most recent centuries were characterized by variable climate with alternative dry and wet phases with high spatial heterogeneity. Most evident in paleo and historical records is the changing woody to grass cover ratio, driven by changes in climate and fire regimes entwined with fluctuating elephant, cattle and wild ungulate populations moderated by human activity, including elephant ivory trade intensification. Archeological perspectives on the occupation of different groups (hunter-gatherers, pastoralists, and farmers) in Amboseli region and the relationships between them are discussed. An overview of the known history of humans and elephants, expanding networks of trade, and the arrival and integration of metallurgy, livestock and domesticated crops in the wider region is provided. In recent decades, increased runoff and flooding have resulted in the expansion of wetlands and a reduction of woody vegetation, compounding problems created by increased enclosure and privatization of these landscapes. However, most of the wetlands outside of the protected area are drying up because of the intensified water extraction by the communities surrounding the National Park and on the adjacent mountains areas, who have increased in numbers, become sedentary and diversified land use around the wetlands.

    • Functional disruption of the medial temporal lobe-dependent networks is thought to underlie episodic memory deficits in aging and Alzheimer's disease. Previous studies revealed that the anterior medial temporal lobe is more vulnerable to pathological and neurodegenerative processes in Alzheimer's disease. In contrast, cognitive and structural imaging literature indicates posterior, as opposed to anterior, medial temporal lobe vulnerability in normal aging. However, the extent to which Alzheimer's and aging-related pathological processes relate to functional disruption of the medial temporal lobe-dependent brain networks is poorly understood. To address this knowledge gap, we examined functional connectivity alterations in the medial temporal lobe and its immediate functional neighbourhood-the Anterior-Temporal and Posterior-Medial brain networks-in normal agers, individuals with preclinical Alzheimer's disease and patients with Mild Cognitive Impairment or mild dementia due to Alzheimer's disease. In the Anterior-Temporal network and in the perirhinal cortex, in particular, we observed an inverted 'U-shaped' relationship between functional connectivity and Alzheimer's stage. According to our results, the preclinical phase of Alzheimer's disease is characterized by increased functional connectivity between the perirhinal cortex and other regions of the medial temporal lobe, as well as between the anterior medial temporal lobe and its one-hop neighbours in the Anterior-Temporal system. This effect is no longer present in symptomatic Alzheimer's disease. Instead, patients with symptomatic Alzheimer's disease displayed reduced hippocampal connectivity within the medial temporal lobe as well as hypoconnectivity within the Posterior-Medial system. For normal aging, our results led to three main conclusions: (i) intra-network connectivity of both the Anterior-Temporal and Posterior-Medial networks declines with age; (ii) the anterior and posterior segments of the medial temporal lobe become increasingly decoupled from each other with advancing age; and (iii) the posterior subregions of the medial temporal lobe, especially the parahippocampal cortex, are more vulnerable to age-associated loss of function than their anterior counterparts. Together, the current results highlight evolving medial temporal lobe dysfunction in Alzheimer's disease and indicate different neurobiological mechanisms of the medial temporal lobe network disruption in aging versus Alzheimer's disease.

    • Archaeology, archaeometallurgy and geoarchaeology are combined in this research to examine the chronology and development of iron metallurgy and its environmental repercussions in North Pare, Tanzania. Pare was a prominent centre for iron production from at least the second half of the first millennium AD, and it has been assumed that this technologywith its demand for wood charcoalhad a significant and detrimental effect on local forest cover. This research sought to examine this claim by exploring the spatial, chronological and technological characteristics of iron production in Pare in conjunction with geoarchaeological evidence. Contrary to older assumptions, our results demonstrate that erosion processes were well established in North Pare before the documented intensification of smelting and smithing activity, and that iron production continued despite environmental changes. We suggest that although iron production may well have contributed to deforestation and erosion in Pare, it is unlikely to be the sole causal factor.

    • Iron has played an important role within East African pastoralist societies for many hundreds of years, yet the means by which iron was produced or obtained by these communities has not been thoroughly documented. The bulk of our understanding is presently based on a limited number of ethnographic and artefact studies, which have tended to focus on the functional and symbolic nature of iron objects themselves. We argue that the research presented here provides the first opportunity to add to this narrow knowledge base by reconstructing the iron production technologies of pastoralist communities in Laikipia, Kenya, using an archaeometallurgical approach. Seven furnaces and one iron-production refuse area were excavated at two discrete workshop sites in Laikipia, central Kenya, that date to the second half of the second millennium AD. The recovered archaeometallurgical materials were analysed using optical microscopy, SEM-EDS and ED-XRF. These techniques revealed that the smelting technologies in question were complex and sophisticated and that they utilised titania-rich black sands and lime-rich charcoal. Whereas the technical approach and raw materials were found to be similar at both sites studied, there was striking stylistic variation in furnace design for no apparent functional reason, which might suggest nuanced differences in the socio-cultural affiliations of the smelters who worked at these sites. This paper explores some of the possible reasons for these differences. In particular, by integrating archaeological data with existing ethnographic and ethnohistoric research from the region, we discuss the technological choices of the smelters and what this might tell us about their identities, as well as considering how future research should best be targeted in order to develop a greater understanding of the organisation of production within pastoralist central Kenya.

  • The influence of obesity-related factors in the etiology of renal cell carcinoma—A mendelian randomization study

    • Background: Several obesity-related factors have been associated with renal cell carcinoma (RCC), but it is unclear which individual factors directly influence risk. We addressed this question using genetic markers as proxies for putative risk factors and evaluated their relation to RCC risk in a mendelian randomization (MR) framework. This methodology limits bias due to confounding and is not affected by reverse causation.Methods and findings: Genetic markers associated with obesity measures, blood pressure, lipids, type 2 diabetes, insulin, and glucose were initially identified as instrumental variables, and their association with RCC risk was subsequently evaluated in a genome-wide association study (GWAS) of 10,784 RCC patients and 20,406 control participants in a 2-sample MR framework. The effect on RCC risk was estimated by calculating odds ratios (ORSD) for a standard deviation (SD) increment in each risk factor. The MR analysis indicated that higher body mass index increases the risk of RCC (ORSD: 1.56, 95% confidence interval [CI] 1.44–1.70), with comparable results for waist-to-hip ratio (ORSD: 1.63, 95% CI 1.40–1.90) and body fat percentage (ORSD: 1.66, 95% CI 1.44–1.90). This analysis further indicated that higher fasting insulin (ORSD: 1.82, 95% CI 1.30–2.55) and diastolic blood pressure (DBP; ORSD: 1.28, 95% CI 1.11–1.47), but not systolic blood pressure (ORSD: 0.98, 95% CI 0.84–1.14), increase the risk for RCC. No association with RCC risk was seen for lipids, overall type 2 diabetes, or fasting glucose.Conclusions: This study provides novel evidence for an etiological role of insulin in RCC, as well as confirmatory evidence that obesity and DBP influence RCC risk.

    • Excited states in the neutron-deficient nuclides(74)(162)W(88) and W-164(74)90 were investigated by using the gamma-ray spectrometer Jurogam. A change in structure is apparent from the first rotational alignments in W-162 and W-164, whose rotationally aligned bands are interpreted as nu(h(9/2))(2) and nu(i(13/2))(2) configurations, respectively. The level schemes have been extended using recoil (-decay) correlations with the observation of excited collective structures. Configuration assignments have been made on the basis of comparisons of the deduced aligned angular momentum, as a function of rotational frequency, with the predictions of the cranked shell model.

    • Rapid rates of land use and land cover change (LULCC) in eastern Africa and limited instances of genuinely equal partnerships involving scientists, communities and decision makers challenge the development of robust pathways toward future environmental and socioeconomic sustainability. We use a participatory modelling tool, Kesho, to assess the biophysical, socioeconomic, cultural and governance factors that influenced past (1959-1999) and present (2000-2018) LULCC in northern Tanzania and to simulate four scenarios of land cover change to the year 2030. Simulations of the scenarios used spatial modelling to integrate stakeholders' perceptions of future environmental change with social and environmental data on recent trends in LULCC. From stakeholders' perspectives, between 1959 and 2018, LULCC was influenced by climate variability, availability of natural resources, agriculture expansion, urbanization, tourism growth and legislation governing land access and natural resource management. Among other socio-environmental-political LULCC drivers, the stakeholders envisioned that from 2018 to 2030 LULCC will largely be influenced by land health, natural and economic capital, and political will in implementing land use plans and policies. The projected scenarios suggest that by 2030 agricultural land will have expanded by 8-20% under different scenarios and herbaceous vegetation and forest land cover will be reduced by 2.5-5% and 10-19% respectively. Stakeholder discussions further identified desirable futures in 2030 as those with improved infrastructure, restored degraded landscapes, effective wildlife conservation, and better farming techniques. The undesirable futures in 2030 were those characterized by land degradation, poverty, and cultural loss. Insights from our work identify the implications of future LULCC scenarios on wildlife and cultural conservation and in meeting the Sustainable Development Goals (SDGs) and targets by 2030. The Kesho approach capitalizes on knowledge exchanges among diverse stakeholders, and in the process promotes social learning, provides a sense of ownership of outputs generated, democratizes scientific understanding, and improves the quality and relevance of the outputs.

    • Rapid land use transformations and increased climatic uncertainties challenge potential sustainable development pathways for communities and wildlife in regions with strong economic reliance on natural resources. In response to the complex causes and consequences of land use change, participatory scenario development approaches have emerged as key tools for analyzing drivers of change to help chart the future of socio-ecological systems. We assess stakeholder perspectives of land use and land cover change (LULCC) and integrate co-produced scenarios of future land cover change with spatial modeling to evaluate how future LULCC in the wider Serengeti ecosystem might align or diverge with the United Nations' Sustainable Development Goals and the African Union's Agenda 2063. Across the wider Serengeti ecosystem, population growth, infrastructural development, agricultural economy, and political will in support of climate change management strategies were perceived to be the key drivers of future LULCC. Under eight scenarios, declines in forest area as a proportion of total land area ranged from 0.1% to 4% in 2030 and from 0.1% to 6% in 2063, with the preservation of forest cover linked to the level of protection provided. Futures with well-demarcated protected areas, sound land use plans, and stable governance were highly desired. In contrast, futures with severe climate change impacts and encroached and degazetted protected areas were considered undesirable. Insights gained from our study are important for guiding pathways toward achieving sustainability goals while recognizing societies' relationship with nature. The results highlight the usefulness of multi-stakeholder engagement, perspective sharing, and consensus building toward shared socio-ecological goals.

    • While management of atrial fibrillation (AF) patients is improved by guideline-conform application of anticoagulant therapy, rate control, rhythm control, and therapy of accompanying heart disease, the morbidity and mortality associated with AF remain unacceptably high. This paper describes the proceedings of the 3rd Atrial Fibrillation NETwork (AFNET)/European Heart Rhythm Association (EHRA) consensus conference that convened over 60 scientists and representatives from industry to jointly discuss emerging therapeutic and diagnostic improvements to achieve better management of AF patients. The paper covers four chapters: (i) risk factors and risk markers for AF; (ii) pathophysiological classification of AF; (iii) relevance of monitored AF duration for AF-related outcomes; and (iv) perspectives and needs for implementing better antithrombotic therapy. Relevant published literature for each section is covered, and suggestions for the improvement of management in each area are put forward. Combined, the propositions formulate a perspective to implement comprehensive management in AF.

    • High-spin states in the near proton-dripline nucleus 107Sb have been identified, and collectivity in this nucleus has been observed for the first time in the form of two rotational bands. One of the observed rotational structures is a ΔI = 1 band, and is interpreted as based on a π(g9/2)-1 ⊗ π(g7/2d5/2)2 proton configuration. A second structure has ΔI = 2 character, and is explained as being based on a πh11/2⊗[π(g9/2)-2 ⊗ π(g7/2d5/2)2] proton configuration through comparison with cranked Nilsson-Strutinsky model calculations. The calculations predict that this band terminates at a spin of 79/2 (Latin small letter h with stroke).

    • The present study examined relationships between actual and desired emotional states, meta-beliefs concerning the utility of distinct emotions, and emotion regulation strategies used by individuals in a sport situation as well as an emotion-eliciting situation from a different aspect of their lives. Participants (N = 924) reported their emotions, meta-beliefs for optimal emotional states, and their use of emotion regulation strategies across two broad categories of situations: Before sports competition, and a situation from daily life. Results indicated that prior to competition, high activation emotions such as anger, anxiety and excitement were preferred. In terms of strategy use, analyses revealed greater intention to use of strategies intended to increase pleasant and unpleasant emotions were associated with daily life. In conclusion, results indicated that meta-beliefs for optimal emotional states, and strategies used to regulate emotions vary between situations. We suggest that the ability to regulate emotions in a flexible manner to suit the specific dynamics of various situations is proposed to be helpful in the pursuit of personally meaningful goals and that training of a variety of emotion regulation skills could be beneficial.

    • Soldiers (N = 95) reported emotions and emotion regulation strategies experienced in their first parachute jump and other challenging situations. Results indicated an emotional profile characterized by feeling anxious, energetic, and happy before parachuting and playing sport. However, this pattern was not similar to the emotional responses experienced at work or in life in general. Participants reported greater use of strategies to increase unpleasant emotions an hour before parachuting than in other situations. Findings suggest that developing training protocols to increase the flexibility and versatility of emotion regulation skills might enhance the preparation of novice soldiers for military duties.

    • Excited states in [Formula Presented]Sb have been identified for the first time in a series of [Formula Presented]-spectroscopy experiments using both thin and backed targets, including neutron-fold and recoil-mass measurements to provide unambiguous channel identification. The three decoupled intruder bands observed in [Formula Presented]Sb are based upon configurations involving 2p-2h excitations across the [Formula Presented] shell gap and show the features of smooth band termination. The yrast intruder band, which has been connected to the low-spin levels, is tentatively identified up to its predicted termination at [Formula Presented]. Excellent agreement with configuration-dependent cranked Nilsson-Strutinsky calculations is obtained for the high-spin states near termination.
