    • This study aimed to investigate the potential of converting bio-based residues from industrial production of mushrooms and tomatoes into more valuable chemicals with antifungal properties using hydrothermal liquefaction (HTL). Liquid fractions were obtained from HTL of spent substrate of Agaricus bisporus (Lange) Imbach and Pleurotus ostreatus (jacq.) P. kumm., recomposted Agaricus bisporus spent substrate, and tomato residues. The quantitative 1H NMR spectroscopy analysis revealed that the HTL liquids of all residues contained antifungal constituents like phenols and organic acids. The HTL liquids at dilutions of 10 % were able to inhibit the fungi by over 80 %. Interestingly, the fungus P. ostreatus showed tolerance to these constituents as its growth was promoted at the lowest concentration of all the HTL liquids. The HTL liquids had lower ecotoxicity than the commercial wood preservative. These results suggest that the tested residues could be a promising source of preservative chemical constituents for the wood industry.

  • The Finnish New Variant of Chlamydia trachomatis with a Single Nucleotide Polymorphism in the 23S rRNA Target Escapes Detection by the Aptima Combo 2 Test

    • In 2019, more than 200 cases of Chlamydia trachomatis negative/equivocal by the Aptima Combo 2 assay (AC2, target: 23S rRNA) with slightly elevated relative light units (RLUs), but positive by the Aptima Chlamydia trachomatis assay (ACT, target: 16S rRNA) have been detected in Finland To identify the cause of the AC2 CT false-negative specimens, we sequenced parts of the CT 23S rRNA gene in 40 specimens that were AC2 negative/equivocal but ACT positive. A single nucleotide polymorphism (SNP; C1515T in the C. trachomatis 23S rRNA gene) was revealed in 39 AC2/ACT discordant specimens. No decrease in the number of mandatorily notified C. trachomatis cases was observed nationally in Finland in 2010-2019. When RLUs obtained for AC2 negative specimens were retrospectively evaluated in 2011-2019, a continuous increase in the proportion of samples with RLUs 10-19 was observed since 2014, and a slight increase in the proportion of samples with RLUs 20-84 in 2017-2019, indicating that the Finnish new variant of C. trachomatis might have been spreading nationally for several years. This emphasizes that careful surveillance of epidemiology, positivity rate and test performance are mandatory to detect any changes affecting detection of infections.

    • Ljungan virus (LV, genus Parechovirus, family Picornaviridae) is considered currently to be a rodent-borne virus. Despite suggested human disease associations, its zoonotic potential remains unclear. To date, LV antibody prevalence in both humans and rodents has not been studied. In this study, two different LV immunofluorescence assays (LV IFAs) were developed with LV genotypes 1 (LV strain 87-012G) and 2 (LV strain 145SLG), and cross-neutralization and -reaction studies were carried out with LV strain 145SLG. Finally, a panel of 37 Finnish sera was screened for anti-LV antibodies using two different LV IFAs (LV 145SLG and LV 87-012G) and a neutralization (NT) assay (LV 145SLG), and 50 samples from Myodes glareolus by LV IFA (LV 145SLG). The LV seroprevalence study showed 38% and 18% positivity in humans and M. glareolus, respectively. LV IFAs and NT assays were compared, and the results were in good agreement. The data are the first evidence of humans and rodents coming into contact with LV in Finland. Additional studies are required in order to acquire a better understanding of the prevalence, epidemiological patterns and possible disease association of LV infections.

  • Opinion : The strength of long-term comprehensive observations to meet multiple grand challenges in different environments and in the atmosphere

    • To be able to meet global grand challenges (climate change; biodiversity loss; environmental pollution; scarcity of water, food and energy supplies; acidification; deforestation; chemicalization; pandemics), which all are closely interlinked with each other, we need comprehensive open data with proper metadata, along with open science. The large data sets from ground-based in situ observations, ground and satellite remote sensing, and multiscale modeling need to be utilized seamlessly. In this opinion paper, we demonstrate the power of the SMEAR (Station for Measuring Earth surface-Atmosphere Relations) concept via several examples, such as detection of new particle formation and the particles' subsequent growth, quantifying atmosphere-ecosystem feedback loops, and combining comprehensive observations with emergency science and services, as well as studying the effect of COVID-19 restrictions on different air quality and climate variables. The future needs and the potential of comprehensive observations of the environment are summarized.

    • The northern Eurasian regions and Arctic Ocean will very likely undergo substantial changes during the next decades. The Arctic-boreal natural environments play a crucial role in the global climate via albedo change, carbon sources and sinks as well as atmospheric aerosol production from biogenic volatile organic compounds. Furthermore, it is expected that global trade activities, demographic movement, and use of natural resources will be increasing in the Arctic regions. There is a need for a novel research approach, which not only identifies and tackles the relevant multi-disciplinary research questions, but also is able to make a holistic system analysis of the expected feedbacks. In this paper, we introduce the research agenda of the Pan-Eurasian Experiment (PEEX), a multi-scale, multi-disciplinary and international program started in 2012 (https://www.atm.helsinki.fi/peex/). PEEX sets a research approach by which large-scale research topics are investigated from a system perspective and which aims to fill the key gaps in our understanding of the feedbacks and interactions between the land-atmosphereaquatic-society continuum in the northern Eurasian region. We introduce here the state of the art for the key topics in the PEEX research agenda and present the future prospects of the research, which we see relevant in this context.

  • Recruitment failure of coastal predatory fish in the Baltic Sea coincident with an offshore ecosystem regime shift

    • The dominant coastal predatory fish in the southwestern Baltic Sea, perch and pike, have decreased markedly in abundance during the past decade. An investigation into their recruitment at 135 coastal sites showed that both species suffered from recruitment failures, mainly in open coastal areas. A detailed study of 15 sites showed that areas with recruitment problems were also notable for mortality of early-stage larvae at the onset of exogenous food-intake. At those sites, zooplankton abundance predicted 83 and 34% of the variation in young of the year perch and pike, respectively, suggesting that the declines were caused by recruitment failure attributable to zooplankton food limitation. Incidences of recruitment failure match in time an offshore trophic cascade that generated massive increases in planktivorous sprat and decreases in zooplankton biomass in the early 1990s. Therefore, sprat biomass explained 53% of the variation in perch recruitment from 1994 to 2007 at an open coastal site, where three-spined stickleback also increased exponentially after 2002. The results indicate that the dramatic change in the offshore ecosystem may have propagated to the coast causing declines of the dominating coastal predators perch and pike followed by an increase in the abundance of small-bodied fish.

  • Overview : Integrative and Comprehensive Understanding on Polar Environments (iCUPE) - concept and initial results

    • The role of polar regions is increasing in terms of megatrends such as globalization, new transport routes, demography, and the use of natural resources with consequent effects on regional and transported pollutant concentrations. We set up the ERA-PLANET Strand 4 project iCUPE - integrative and Comprehensive Understanding on Polar Environments to provide novel insights and observational data on global grand challenges with an Arctic focus. We utilize an integrated approach combining in situ observations, satellite remote sensing Earth observations (EOs), and multi-scale modeling to synthesize data from comprehensive long-term measurements, intensive campaigns, and satellites to deliver data products, metrics, and indicators to stakeholders concerning the environmental status, availability, and extraction of natural resources in the polar areas. The iCUPE work consists of thematic state-of-the-art research and the provision of novel data in atmospheric pollution, local sources and transboundary transport, the characterization of arctic surfaces and their changes, an assessment of the concentrations and impacts of heavy metals and persistent organic pollutants and their cycling, the quantification of emissions from natural resource extraction, and the validation and optimization of satellite Earth observation (EO) data streams. In this paper we introduce the iCUPE project and summarize initial results arising out of the integration of comprehensive in situ observations, satellite remote sensing, and multi-scale modeling in the Arctic context.

  • Transferability of predictive fish distribution models in two coastal systems

    • Species distribution modelling has emerged as a tool both for exploring niche theory and for producing distribution maps for management. To understand and predict potential effects of large scale habitat change there is a need for proper model validation and applicability also in unstudied areas. However, knowledge about factors influencing the transferability of distribution models, i.e. the accuracy of the models when applying them in a new geographical area, is limited. We have successfully modelled the larval distribution of two fish species, northern pike (Esox lucius L.) and roach (Rutilus rutilus L.), on a regional scale in the Baltic Sea using a few and easily measured environmental variables. When models were transferred from the training area to the testing area the models showed reasonable to very good discrimination (ROC 0.75 and 0.93) based on external validation using independent data separated also in time (1–2 years). The predicted larval distribution also overlapped with the distribution of young-of-the-year fish later in the season. Performance when reversing the transfer, by constructing the models in the testing area and predicting back to the original training area, was less successful. This discrepancy was species-specific and could be explained by differences in the species presence ranges along the predictor variables in the testing area compared to the training area. Our results illustrate how transferability success can be influenced by area-specific differences in the range of the predictor variables and show the necessity of validating model predictions properly.

  • Trophic overlap between expanding and contracting fish predators in a range margin undergoing change

    • Climate change is predicted to cause a freshening of the Baltic Sea, facilitating range expansions of freshwater species and contractions of marine. Resident marine flounders (Platichthys flesus) and expansive freshwater roach (Rutilus rutilus) are dominant consumers in the Baltic Sea sublittoral where they occur in partial sympatry. By comparing patterns of resource use by flounders and roach along a declining resource gradient of blue mussels (Mytilus trossulus) our aim was to explore predator functional responses and the degree of trophic overlap. Understanding the nature of density-dependent prey acquisition has important implications for predicting population dynamics of both predators and their shared prey. Results showed a highly specialized diet for both species, high reliance on blue mussels throughout the range, similar prey size preference and high trophic overlap. Highest overlap occurred where blue mussels were abundant but overlap was also high where they were scarce. Our results highlight the importance of a single food item - the blue mussel - for both species, likely promoting high population size and range expansion of roach. Findings also suggest that range expansion of roach may have a top-down structuring force on mussels that differ in severity and location from that originating from resident flounders.
