    • Bio-based nanocellulose has been shown to possess impressive mechanical properties and simplicity for chemical modifications. The chemical properties are largely influenced by the surface area and functionality of the nanoscale materials. However, finding the typical cross-sections of nanocellulose, such as cellulose nanofibers (CNFs), has been a long-standing puzzle, where subtle changes in extraction methods seem to yield different shapes and dimensions. Here, we extracted CNFs from wood with two different oxidation methods and variations in degree of oxidation and high-pressure homogenization. The cross-sections of CNFs were characterized by small-angle X-ray scattering and wide-angle X-ray diffraction in dispersed and freeze-dried states, respectively, where the results were analyzed by assuming that the cross-sectional distribution was quantized with an 18-chain elementary microfibril, the building block of the cell wall. We find that the results agree well with a pseudosquare unit having a size of about 2.4 nm regardless of sample, while the aggregate level strongly depends on the extraction conditions. Furthermore, we find that aggregates have a preferred cohesion of phase boundaries parallel to the (110)-plane of the cellulose fibril, leading to a ribbon shape on average.

    • Recent molecular studies have identified substantial fungal diversity in indoor environments. Fungi and fungal particles have been linked to a range of potentially unwanted effects in the built environment, including asthma, decay of building materials, and food spoilage. The study of the built mycobiome is hampered by a number of constraints, one of which is the poor state of the metadata annotation of fungal DNA sequences from the built environment in public databases. In order to enable precise interrogation of such data – for example, “retrieve all fungal sequences recovered from bathrooms” – a workshop was organized at the University of Gothenburg (May 23-24, 2016) to annotate public fungal barcode (ITS) sequences according to the MIxS-Built Environment annotation standard (http://gensc.org/mixs/). The 36 participants assembled a total of 45,488 data points from the published literature, including the addition of 8,430 instances of countries of collection from a total of 83 countries, 5,801 instances of building types, and 3,876 instances of surface-air contaminants. The results were implemented in the UNITE database for molecular identification of fungi (http://unite.ut.ee) and were shared with other online resources. Data obtained from human/animal pathogenic fungi will furthermore be verified on culture based metadata for subsequent inclusion in the ISHAM-ITS database (http://its.mycologylab.org).

  • Tillstånd och trender för arter och deras livsmiljöer – rödlistade arter i Sverige 2015

    • 2015 års upplaga av den svenska rödlistan är den fjärde i ordningen. Den är baserad på IUCN:s rödlistningskriterier och revideras vart femte år. I rödlistan bedöms risken som enskilda arter av djur, växter och svampar löper att försvinna från Sverige. Bedömningen utförs av ArtDatabankens medarbetare i samverkan med över 100 externa experter, indelade i 14 expertkommittéer för olika organismgrupper. Under arbetet med 2015 års rödlista har tillstånd och trender bedömts för 21 600 arter och 1 318 lägre taxa (apomiktiska arter, underarter och varieteter), sammanlagt ca 22 900 taxa. Av de bedömda arterna klassificerades 2 029 som hotade (kategorierna CR, EN och VU) och 4 273 som rödlistade (inkluderar även kategorierna NT, RE och DD). Förhållandet mellan antalet rödlistade och antalet bedömda arter ar 19,8 %, vilket är ungefär samma värde som 2010 och 2005. I denna rapport jämförs antalet och andelen rödlistade arter mellan olika organismgrupper, biotoper, substrat och påverkansfaktorer. Texten ar indelad i en allmän del och åtta kapitel inriktade på olika landskapstyper. Landskapstyperna utgör en grov indelning av landets miljöer enligt följande kategorier: Skog, Jordbrukslandskap, Urbana miljöer, Fjäll, Våtmarker, Sötvatten, Havsstränder och Havsmiljöer. Skogen och jordbrukslandskapet är de artrikaste landskapstyperna med 1 800 respektive 1 400 arter som har en stark anknytning dit, och ytterligare flera hundra arter som förekommer där mer sporadiskt. De faktorer som påverkar flest rödlistade arter i Sverige är skogsavverkning och igenväxning, som båda utgör ett hot mot vardera ca 30 % av de rödlistade arterna. Avverkning minskar arealen av skog där naturliga strukturer och naturlig dynamik upprätthålls, och den orsakar därmed förlust av livsmiljöer. Igenväxning orsakas av ett antal faktorer, bland annat upphörande hävd (bete och slåtter), gödsling, trädplantering och brist på naturliga störningsregimer som t.ex. regelbundna översvämningar kring vattendrag och sjöar. Andra viktiga påverkansfaktorer är fiske, torrläggning av våtmarker, tillbakagång hos värdarter (främst alm och ask som drabbats av invasiva svampsjukdomar), klimatförändringar och konkurrens från invasiva arter. IUCN:s rödlisteindex beräknas för ett urval av de bedömda organismgrupperna. Rödlisteindex visar att skillnaderna mellan rödlistorna från 2000, 2005, 2010 och 2015 är små. Ett par undantag finns dock. Groddjur och stora däggdjur har fått en något förbättrad situation sedan 2000. Totalt förefaller det ändå som att trycket mot Sveriges artstock har förblivit relativt konstant under de senaste 15 åren.

  • Characterization of pulp with high enzymatic hydrolyzability

    • Conversion of biomass to biofuels is currently an area that attracts large interest, and lignocellulosic biomass offers the abundance and environmental attributes that can potentially support large-scale biofuel production as an alternative to petroleum-based transportation fuel.In a recent project, Innventia has developed wood based pulps optimized for conversion to biofuels. These novel pulps were produced to target a high level of enzymatic hydrolyzability. To assess the hydrolyzability of these pulps, a laboratory protocol has been established usingan enzyme mixture containing Celluclast 1.5L and Novozyme 188 with an activity of 10 FPU/g pulp (Andersen 2007). Results obtained using this protocol are assumed to be relevant for industrial conditions. In addition to assessment of the produced pulps, the results havebeen compared to commercial cellulose substrates and pulps of a variety of grades.Furthermore, supramolecular properties – specific surface area and average pore size – were determined by an in-house method utilizing solid state nuclear magnetic resonance (Larsson et al. 2013). Kappa numbers, limiting viscosities, ISO-brightness and carbohydrate compositions were determined using standard methods. Molecular mass distributions of cellulose tricarbanilates were determined by size exclusion chromatography with tetrahydrofuran mobile phase (Drechsler et al. 2000).The presentation will discuss the influence of chemical, macromolecular and supramolecular properties of commercial and novel pulp grades on the enzymatic hydrolyzability. Theprotocol used to assess of enzymatic hydrolyzability will be proposed as a benchmark test.

    • Conversion of biomass to biofuels and other products is a research area that is currently attracting a great amount of interest, particularly because such production may be envisaged as a key part of any bio-based economy. Lignocellulosic biomass is abundant and sustainable, and can therefore potentially support large-scale production of biofuel as an alternative to petroleum-based fuel.The enzymatic hydrolysability of three industrial pulps, five lab made pulps, and one microcrystalline cellulose powder was assessed using commercial cellulolytic enzymes. To gain insight into the factors that influence the hydrolysability, a thorough characterization of the samples was done, including their chemical properties (cellulose content, hemicellulose content, lignin content, and kappa number), their macromolecular properties (peak molar mass, number-average molar mass, weight-average molar mass, polydispersity, and limiting viscosity) and their supramolecular properties (fibre saturation point, specific surface area, average pore size, and crystallinity). The hydrolysability was assessed by determination of initial conversion rate and final conversion yield, with conversion yield defined as the amount of glucose in solution per unit of glucose in the substrate. Multivariate data analysis revealed that for the investigated samples the conversion of cellulose to glucose was mainly dependent on the supramolecular properties, such as specific surface area and average pore size. The molar mass distribution, the crystallinity, and the lignin content of the pulps had no significant effect on the hydrolysability of the investigated samples.In addition, experiments were carried out aiming at identifying suitable conditions for pre-treatment of wheat straw, for the purpose of making cellulose rich pulps with improved enzymatic reactivity. Two sets of conditions for pre-treatment of wheat straw were identified; a combination of low temperature alkaline washing and acid pre-hydrolysis, or high temperature acid pre-hydrolysis. Both bleached wheat straw pulps showed similar enzymatic reactivity. However, the enzymatic reactivity of both bleached wheat straw pulps was found to be significantly less than what has been achieved for wood pulps. A probable explanation for the low enzymatic reactivity of the bleached wheat straw pulp can be the small pore size, limiting the access for enzymes to the cellulose surfaces in the fibre wall interior.Text, figures and tables in an extended abstract (< 4 pages with title and references).

    • BACKGROUND: The infection caused by SARS CoV-2 has been postulated to induce a cytokine storm syndrome that results in organ failure and even death in a considerable number of patients. However, the inflammatory response in Corona virus disease-19 (Covid-19) and its potential to cause collateral organ damage has not been fully elucidated to date. This study aims to characterize the acute cytokine response in a cohort of critically ill Covid-19 patients.METHOD: 24 adults with PCR-confirmed Covid-19 were included at time of admission to intensive care a median of eleven days after initial symptoms. Eleven adult patients admitted for elective abdominal surgery with preoperative plasma samples served as controls. All patients were included after informed consent was obtained. 27 cytokines were quantified in plasma. The expression of inflammatory mediators was then related to routine inflammatory markers, SAPS3, SOFA score, organ failure and 30-day mortality.RESULTS: A general increase in cytokine expression was observed in all Covid-19 patients. A strong correlation between respiratory failure and IL-1ra, IL-4, IL-6, IL-8 and IP-10 expression was observed. Acute kidney injury development correlated well with increased levels of IL-1ra, IL-6, IL-8, IL-17a, IP-10 and MCP-1. Generally, the cohort demonstrated weaker correlations between cytokine expression and 30-day mortality out of which IL-8 showed the strongest signal in terms of mortality.CONCLUSION: The present study found that respiratory failure, acute kidney injury and 30-day mortality in critically ill Covid-19 patients are associated with moderate increases of a broad range of inflammatory mediators at time of admission.

    • Avfallsprevention genom ökad materialeffektivitet ger ofta miljöfördelar, t ex i form av minskade utsläpp av växthusgaser, eftersom det bidrar till att hålla nere energiintensiv materialproduktion. Syftet med detta projekt är att undersöka om ökad materialeffektivitet också bidrar till minskade utsläpp av farliga ämnen. Minskar eller ökar utsläppen av farliga ämnen om man ökar materialeffektiviteten? Finns det någon tydlig korrelation eller ej? Med farliga ämnen menar vi framförallt ämnen som hanteras av miljömålet Giftfri miljö och i Naturvårdsverkets strategi för giftfria och resurssnåla kretslopp (GRK), men också andra ämnen som påverkar människors hälsa. Vi utgår ifrån olika strategier för ökad materialeffektivitet (se Tabell S). För varje strategi identifierar vi exempel på hur den leder till ökade eller minskade utsläpp av farliga ämnen. Vi lyfter särskilt fram exempel och slutsatser som rör byggsektorn och livsmedelskedjan, eftersom de lyfts fram som prioriterade områden i GRK-strategin. Utifrån de identifierade exemplen drar vi slutsatsen att det finns många fall där ökad materialeffektivitet också bidrar till en mer giftfri miljö (se Tabell S). I vissa fall ger materialeffektiviteten viktiga miljöfördelar utöver att själva materialproduktionen minskar. Det gäller t ex fallet med småbilar. Det finns dock fall när ökad materialeffektivitet vare sig bidrar till giftfrihet eller andra miljöförbättringar. Det finns även fall där ökad materialeffektivitet bidrar till minskat energibehov, men ändå riskerar att öka användningen av farliga ämnen och/eller spridningen av dem i miljön. Slutligen finns risk för så kallade rebound-effekter om den ökade materialeffektiviteten är kostnadseffektiv. Vår samlade bedömning är att ökad materialeffektivitet ofta ger miljöfördelar också i form av minskade utsläpp av farliga ämnen. Sambandet verkar dock vara svagare än mellan materialeffektivitet och minskade utsläpp av växthusgaser. Osäkerheten är också större, eftersom frågan om farliga utsläpp är mer komplex. Man bör t ex ta hänsyn till farligheten hos ämnet och inte bara till den använda eller utsläppta mängden. Frågan är dessutom mindre utforskad. Ämnet för vår studie omfattar i princip alla material, alla produkter och alla produktionsprocesser. Vi har långt ifrån täckt ämnet fullständigt. Viktiga delar av studien är också relativt ytliga. Våra slutsatser skulle bli säkrare och mer välgrundade om studien breddas med fler exempel och/eller fördjupas på de punkter där den är ytlig. I förlängningen kan också systemanalyser vara motiverade, t ex inför implementeringen av EUs nya ramdirektiv om avfall (EU 2008) i svensk rätt.

  • Lung complications are common in intensive care treated patients with pelvis fractures : a retrospective cohort study

    • Background: The incidence of severe respiratory complications in patients with pelvis fractures needing intensive care have not previously been studied. Therefore, the aims of this registry study were to 1) determine the number of ICU patients with pelvis fractures who had severe respiratory complications 2) whether the surgical intervention in these patients is associated with the pulmonary condition and 3) whether there is an association between lung complications and mortality. We hypothesized that acute hypoxic failure (AHF) and acute respiratory distress syndrome (ARDS) 1) are common in ICU treated patients with pelvis fractures, 2) are not related to the reconstructive surgery, or to 3) to mortality.Methods: All patients in the database cohort (n = 112), scheduled for surgical stabilization of pelvis ring and/or acetabulum fractures, admitted to the general ICU at Uppsala University Hospital between 2007 and 2014 for intensive care were included.Results: The incidence of AHF/ARDS was 67 % (75/112 patients), i.e., the percentage of patients that at any period during the ICU stay fulfilled the AHF/ARDS criteria. The incidence of AHF was 44 % and incidence of ARDS was 23 %. The patients with AHF/ARDS had more lung contusions and pneumonia than the patients without AHF/ARDS. Overall, there were no significant changes in oxygenation variables associated with surgery. However, 23 patients with pre-operative normal lung status developed AHF/ARDS in relation to the surgical procedure, whereas 12 patients with AHF/ARDS normalized their lung condition. The patients who developed AHF/ARDS had a higher incidence of lung contusion (P = 0.04) and the surgical stabilization was performed earlier (5 versus 10 days) in these patients (P = 0.03).Conclusions: We found that the incidence of respiratory failure in ICU treated patients with pelvis fractures was high, that the procedure around surgical stabilization seems to be associated with a worsening in the respiratory function in patients with lung contusion, and that mortality was low and was probably not related to the respiratory condition.

    • Thorough characterization and fundamental understanding of cellulose fibers can help us develop new, sustainable material streams and advanced functional materials. As an emerging nanomaterial, cellulose nanofibrils (CNFs) have high specific surface area and good mechanical properties; however, handling and processing challenges have limited their widespread use. This work reports an in-depth characterization of self-fibrillating cellulose fibers (SFFs) and their use in smart, responsive filters capable of regulating flow and retaining nanoscale particles. By combining direct and indirect characterization methods with polyelectrolyte swelling theories, it was shown that introduction of charges and decreased supramolecular order in the fiber wall were responsible for the exceptional swelling and nanofibrillation of SFFs. Different microscopy techniques were used to visualize the swelling of SFFs before, during, and after nanofibrillation. Through filtration and pH adjustment, smart filters prepared via in situ nanofibrillation showed an ability to regulate the flow rate through the filter and a capacity of retaining 95% of 300 nm (diameter) silica nanoparticles. This exceptionally rapid and efficient approach for making smart filters directly addresses the challenges associated with dewatering of CNFs and bridges the gap between science and technology, making the widespread use of CNFs in high-performance materials a not-so-distant reality.

  • Characterization of cellulose supramolecular structure using solid-state NMR

    • Cellulose I isolated from wood in the form of cellulose-rich fibres, i.e. as a pulp, is a widely used raw material that holds a potential for further and more versatile use. Due to its abundance cellulose can be a benign replacement for many materials used in everydaycommodities.Isolated cellulose I is associated with a complex supramolecular structure (in the nanometresdimensional range), and in the case of cellulose-richfibres it is also associated with a complex fibre wall morphology (typical wood fibres are millimetres long and tenths of micrometres wide).The main advantage of using cellulose-rich fibres is an existence of a worldwide industry which has the processing equipment and the know-how necessary for efficient handling and processing of wood-based pulps.Utilization of cellulose I is dependent on the reactivity of the cellulose substrate, here the term reactivity is used in a broad sense. Enzymatic conversion of cellulose-rich fibres to sugars or the dissolution of cellulose for textile fibre manufacture is two examples where different aspects of the cellulosereactivity are important for efficient processing.Several methods for characterizing various aspects of cellulose are available. The degree of polymerization and the degree of cellulose crystallinity are two examples. In the case of cellulose-rich fibres its carbohydrate composition can be of importance. Traditionally lessattention has been paid to the supramolecular characteristics of cellulose although they are in a dimensional range that could exert an influence on the chemistry used.The present work deals with the characterization of the supramolecular properties of cellulose and cellulose-rich fibres and illustrates some examples where the supramolecular structure of the cellulose is a controlling factor for its reactivity. Most of the presented work is based on CP/MAS 13 C-NMR measurements. Using this technique it has been shown that robust measurements of cellulose nanostructures such aslateral fibril dimensions and lateral fibril aggregate dimensions can be obtained and how subsequently the specific surface area of the cellulose in a water-swollen state can be estimated. Moreover, by combining NMR resultswith measurements of the amount of water located inside a water-swollen fibre wall, estimates of the average fibre wall pore size can be obtained. Such results have beenrelated to data from enzymatic hydrolysis of cellulose-richfibres to illustrate the influence of supramolecular structure on enzymatic reactivity.

    • BACKGROUND: Fibroblast growth factor-23 (FGF23) is a regulator of mineral metabolism and has been suggested to play a role in vascular calcification in chronic kidney disease (CKD). Data on the association between FGF23 and atherosclerosis, both in CKD and in the community, is limited. METHODS: The total body atherosclerosis score (AS) was determined by a magnetic resonance imaging-based angiography in 306 elderly men and women, representing a subsample of the community-based PIVUS cohort. Subjects were divided into three categories based on AS: AS = 0, low AS and high AS. Serum FGF23 was measured using a two-site monoclonal antibody ELISA. RESULTS: In continuous and multi-category regression models, higher FGF23 was associated with a significant increase in the odds of having a high AS (OR 1.43, CI 1.06-1.92 to OR 3.01, CI 1.52-5.99). This association was stronger in individuals with eGFR <60 mL/min/1.73 m(2) (n = 27), reaching a nearly 6-fold increase in the odds for a high AS in the upper FGF23 tertile (OR 5.64, CI 2.78-11.5). We found weaker support for a relationship between FGF23 and the presence of atherosclerosis as subjects in the highest FGF23 tertile had an increased risk for an AS > 0 in crude models (OR 1.93, CI 1.05-3.55), but this was not statistically significant in adjusted (OR 1.42, CI 0.74-1.72) models. CONCLUSIONS: We provide novel evidence supporting an association between serum FGF23 and total body atherosclerosis in the community. Additional studies are warranted to determine the prospective relationship between FGF23 and atherosclerosis, and whether FGF23 is a modifiable cardiovascular risk factor.

    • Background: Endothelial glycocalyx regulates the endothelial function and plays an active role in maintaining vascular homeostasis. During ischemia/reperfusion, the glycocalyx is rapidly shed into the blood stream. A heparin conjugate (CHC; Corline systems AB, Uppsala, Sweden) consists of 70 heparin molecules that have the capacity to adhere strongly to biological tissues expressing heparin affinity. We hypothesized that CHC could be used to restore disrupted glycocalyx in vivo in kidneys from brain-dead pigs.Materials and Methods: Brain death was induced in male landrace pigs (n=6) by inflating a balloon catheter in the epidural space until obtaining negative cerebral perfusion. The recovered kidneys (n=5+5) were perfused by hypothermic machine perfusion (HMP) using two Lifeport kidney transporters (Organ Recovery Systems, Chicago, IL, USA). 50 mg CHC (including 25 mg biotinylated CHC) or 50 mg unfractionated heparin (control) was added to the perfusion fluid in the respective machines. In one case, the kidneys were used only for dose escalation of CHC with the same procedure.Results: CHC was detected by immunofluorescence and confocal microscopy in the inner surface of vessel walls. The binding of CHC in the kidney was confirmed indirectly by consumption of CHC from the perfusion fluid.Conclusions: In this first attempt, we show that CHC may be used to coat the vessel walls of perfused kidneys during HMP, an approach that could become useful in restoring endothelial glycocalyx of kidneys recovered from deceased donors to protect vascular endothelium and possibly ameliorate ischemia/reperfusion injuries.

    • In recent years, the Bitcoin community has collectively developed an open source platform that allows for the mining of the Bitcoin currency as well as instant global peer-to-peer payments and financial transactions using Bitcoins - without any central authority. With its theoretical roots in the Austrian School of Economics, the community can be seen as a potential threat to the mega financial institutions and governments in Europe and across the globe as the Bitcoin currency and its underlying principles challenge the long-standing fiat money system. Designed and implemented in only 2009, Bitcoin has rapidly grown from being an idea in the head of a “Japanese programmer” to becoming a legitimate currency as Bitcoin-Central was awarded an International Bank ID number and became a Payment Services Provider equal to organizations such as PayPal. However, perhaps due to its rapid growth during the past four years and the fact that the currency is primarily a virtual one, the Bitcoin community has been subject to external threats such as fraud, hacker attacks, and a lawsuit. Despite this, the community has shown significant resilience and has even shown continued exponential growth in recent months. As such, our research purpose is to investigate the process through which the Bitcoin community acts as an institutional entrepreneur. As a first step in fulfilling our research purpose, we conduct an exploratory analysis in this research-in-progress paper of the formal and informal “organizations” of Bitcoin as well as of the topical network structure of the Bitcoin community using secondary sources and the complete archive of 1.15 million English posts written by 21,903 members between 2009 and 2013. Some preliminary results and findings as well as future steps are discussed.

    • The biotechnological applications of cellulose nanocrystals (CNCs) continue to grow due to their sustainable nature, impressive mechanical, rheological, and emulsifying properties, upscaled production capacity, and compatibility with other materials, such as protein and polysaccharides. In this study, hydrogels from CNCs and pectin, a plant cell wall polysaccharide broadly used in food and pharma, were produced by calcium ion-mediated internal ionotropic gelation (IG). In the absence of pectin, a minimum of 4 wt% CNC was needed to produce self-supporting gels by internal IG, whereas the addition of pectin at 0.5 wt% enabled hydrogel formation at CNC contents as low as 0.5 wt%. Experimental data indicate that CNCs and pectin interact to give robust and self-supporting hydrogels at solid contents below 2.5 %. Potential applications of these gels could be as carriers for controlled release, scaffolds for cell growth, or wherever else distinct and porous network morphologies are required.

    • Phenolic compounds in hydrolysates are degradation products from the lignin component of wood. They are diverse in nature and they account for some of the inhibitory activities observed during lignocellulosic fermentation. S. cerevisiae possesses the ability to convert some phenolic compounds. We are currently studying the interaction between S. cerevisiae and selected phenolic compounds namely; coniferyl aldehyde, ferulic acid and p-coumaric acid to understand the ability of S. cerevisiae to convert the selected compounds. Preliminary results show that the three phenolic compounds are being converted into several other less inhibitory phenolic compounds common to the three compounds. We hypothesised a conversion route and engineered S. cerevisiae strains to test the hypothesis, the preliminary result shows faster conversion in an engineered strain.

    • Background The invasive benthic round goby (Neogobius melanostomus) is the most successful temperate invasive fish and has spread in aquatic ecosystems on both sides of the Atlantic. Invasive species constitute powerful in situ experimental systems to study fast adaptation and directional selection on short ecological timescales and present promising case studies to understand factors involved the impressive ability of some species to colonize novel environments. We seize the unique opportunity presented by the round goby invasion to study genomic substrates potentially involved in colonization success. Results We report a highly contiguous long-read-based genome and analyze gene families that we hypothesize to relate to the ability of these fish to deal with novel environments. The analyses provide novel insights from the large evolutionary scale to the small species-specific scale. We describe expansions in specific cytochrome P450 enzymes, a remarkably diverse innate immune system, an ancient duplication in red light vision accompanied by red skin fluorescence, evolutionary patterns of epigenetic regulators, and the presence of osmoregulatory genes that may have contributed to the round goby's capacity to invade cold and salty waters. A recurring theme across all analyzed gene families is gene expansions. Conclusions The expanded innate immune system of round goby may potentially contribute to its ability to colonize novel areas. Since other gene families also feature copy number expansions in the round goby, and since other Gobiidae also feature fascinating environmental adaptations and are excellent colonizers, further long-read genome approaches across the goby family may reveal whether gene copy number expansions are more generally related to the ability to conquer new habitats in Gobiidae or in fish.

    • The effect of peak power in a high power impulse magnetron sputtering (HiPIMS) reactive deposition of TiO(2) films has been studied with respect to the deposition rate and coating properties. With increasing peak power not only the ionization of the sputtered material increases but also their energy. In order to correlate the variation in the ion energy distributions with the film properties, the phase composition, density and optical properties of the films grown with different HiPIMS-parameters have been investigated and compared to a film grown using direct current magnetron sputtering (DCMS). All experiments were performed for constant average power and pulse on time (100W and 35 mu s, respectively), different peak powers were achieved by varying the frequency of pulsing. Ion energy distributions for Ti and O and its dependence on the process conditions have been studied. It was found that films with the highest density and highest refractive index were grown under moderate HiPIMS conditions (moderate peak powers) resulting in only a small loss in mass-deposition rate compared to DCMS. It was further found that TiO2 films with anatase and rutile phases can be grown at room temperature without substrate heating and without post-deposition annealing.

    • In the further development of reactive sputter deposition, strategies which allow for stabilization of the transition zone between the metallic and compound modes, elimination of the process hysteresis, and increase of the deposition rate, are of particular interest. In this study, the hysteresis behavior and the characteristics of the transition zone during reactive high power impulse magnetron sputtering (HiPIMS) of Al and Ce targets in an Ar-O(2) atmosphere as a function of the pulsing frequency and the pumping speed are investigated. Comparison with reactive direct current magnetron sputtering (DCMS) reveals that HiPIMS allows for elimination/suppression of the hysteresis and a smoother transition from the metallic to the compound sputtering mode. For the experimental conditions employed in the present study, optimum behavior with respect to the hysteresis width is obtained at frequency values between 2 and 4 kHz, while HiPIMS processes with values below or above this range resemble the DCMS behavior. Al-O films are deposited using both HiPIMS and DCMS. Analysis of the film properties shows that elimination/suppression of the hysteresis in HiPIMS facilitates the growth of stoichiometric and transparent Al(2)O(3) at relatively high deposition rates over a wider range of experimental conditions as compared to DCMS.

    • The fetal transcription factor PLAG1 is found to be overexpressed in cancers, and has been suggested to bind the insulin like growth factor 2 (IGF2) P3 promoter, and to activate the IGF2 gene. The expression of IGF2 has partly been linked to loss of CTCF-dependent chromatin insulator function at the H19 imprinting control region (ICR). We investigated the role of PLAG1 for IGF2 regulation in Hep3B and JEG-3 cell lines. Chromatin immunoprecipitation revealed cell type-specific binding of PLAG1 to the IGF2 P3 promoter, which was substantially insensitive to recombinant PLAG1 overexpression in the endogenous context. We hypothesized that the H19 chromatin insulator may be involved in the cell type-specific PLAG1 response. By using a GFP reporter gene/insulator assay plasmid construct with and without the H19 ICR and/or an SV40 enhancer, we confirm that the effect of the insulator is specifically associated with the activity of the IGF2 P3 promoter in the GFP reporter system, and furthermore, that the reporter insulator is functional in JEG-3 but not in Hep3B cells. FACS analysis was used to assess the function of PLAG1 in low endogenously expressing, but Zn-inducible stable PLAG1 expressing JEG-3 cell clones. Considerable increase in IGF2 expression upon PLAG1 induction with a partial insulator overriding activity was found using the reporter constructs. This is in contrast to the effect of the endogenous IGF2 gene which was insensitive to PLAG1 expression in JEG-3, while modestly induced the already highly expressed IGF2 gene in Hep3B cells. We suggest that the PLAG1 binding to the IGF2 P3 promoter and IGF2 expression is cell type-specific, and that the PLAG1 transcription factor acts as a transcriptional facilitator that partially overrides the insulation by the H19 ICR.

    • Background: Traumatic brain injury (TBI) is a common cause of morbidity and mortality in children worldwide. In Scandinavia, the epidemiology of pediatric head trauma is poorly documented. This study aimed to investigate and compare the epidemiology and management of pediatric patients with isolated head trauma (IHT) and head trauma in connection with multitrauma (MHT). Methods: We conducted a retrospective review of medical records of patients < 18 years of age who attended any of the five emergency departments (ED) in Scania County in Sweden in 2016 due to head trauma. Clinical data of patients with IHT were analyzed and compared with those of patients with MHT. Results: We identified 5046 pediatric patients with head trauma, 4874 with IHT and 186 with MHT, yielding an incidence of ED visits due to head trauma of 1815/100,000 children/year. There was male predominance, and the median age was four years. Falls were the dominating trauma mechanism in IHT patients, while motor vehicle accidents dominated in MHT patients. The frequencies of CT head-scans, ward admissions and intracranial injuries (ICI) were 5.4%, 11.1% and 0.7%, respectively. Four patients (0.08%) required neurosurgical intervention. The relative risks for CT-scans and admissions to a hospital ward and ICI were 10, 4.5 and 19 times higher for MHT compared with IHT patients. Conclusion: Head trauma is a common cause of ED visits in our study. Head-CTs and ICIs were less frequent than in previous studies. MHT patients had higher rates of CT-scans, admissions, and ICIs than IHT patients, suggesting that they are separate entities that should ideally be managed using different guidelines to optimize the use of CT-scans of the head.

    • We present minimal, non-iterative solutions to the absolute pose problem for images from rolling shutter cameras. The absolute pose problem is a key problem in computer vision and rolling shutter is present in a vast majority of today's digital cameras. We discuss several camera motion models and propose two feasible rolling shutter camera models for a polynomial solver. In previous work a linearized camera model was used that required an initial estimate of the camera orientation. We show how to simplify the system of equations and make this solver faster. Furthermore, we present a first solution of the non-linearized camera orientation model using the Cayley parameterization. The new solver does not require any initial camera orientation estimate and therefore serves as a standalone solution to the rolling shutter camera pose problem from six 2D-to-3D correspondences. We show that our algorithms outperform P3P followed by a non-linear refinement using a rolling shutter model.

    • A low intake of selenium is associated with increased cardiovascular mortality. This could be reduced by supplementation with selenium and coenzyme Q10. D-dimer, a fragment of fibrin mirroring fibrinolysis, is a biomarker of thromboembolism, increased inflammation, endothelial dysfunction and is associated with cardiovascular mortality in ischemic heart disease. The objective was to examine the impact of selenium and coenzyme Q10 on the level of D-dimer, and its relationship to cardiovascular mortality. D-dimer was measured in 213 individuals at the start and after 48 months of a randomised double-blind placebo-controlled trial with selenium yeast (200 µg/day) and coenzyme Q10 (200 mg/day) (n = 106) or placebo (n = 107). The follow-up time was 4.9 years. All included individuals were low in selenium (mean 67 μg/L, SD 16.8). The differences in D-dimer concentration were evaluated by the use of T-tests, repeated measures of variance and ANCOVA analyses. At the end, a significantly lower D-dimer concentration was observed in the active treatment group in comparison with those on placebo (p = 0.006). Although D-dimer values at baseline were weakly associated with high-sensitive CRP, while being more strongly associated with soluble tumour necrosis factor receptor 1 and sP-selectin, controlling for these in the analysis there was an independent effect on D-dimer. In participants with a D-dimer level above median at baseline, the supplementation resulted in significantly lower cardiovascular mortality compared to those on placebo (p = 0.014). All results were validated with a persisting significant difference between the two groups. Therefore, supplementation with selenium and coenzyme Q10 in a group of elderly low in selenium and coenzyme Q10 prevented an increase in D-dimer and reduced the risk of cardiovascular mortality in comparison with the placebo group. The obtained results also illustrate important associations between inflammation, endothelial function and cardiovascular risk.

    • PURPOSE: Endothelial dysfunction and inflammation are conditions which fuel atherosclerosis and ischaemic heart disease. We have previously reported reduced cardiovascular (CV) mortality following supplementation with selenium and coenzyme Q10 to 443 elderly individuals with low selenium status (mean 67 μg/L) for 4 years. Here, we wanted to evaluate a possible association between the supplementation and the plasma concentrations of the von Willebrand factor (vWf), and the plasminogen activator inhibitor-1 (PAI-1), as they, besides other functions, are also strongly associated with endothelial function.METHODS: In this sub-study, 308 individuals (active substance: 157, placebo: 151) were included. Blood samples were drawn after 6 and 36 months and vWf and PAI-1 were determined in plasma by ELISA. Changes in concentrations of the biomarkers were evaluated by the use of T tests, repeated measures of variance, and ANCOVA analyses.RESULTS: The active treatment group presented a lower level of vWf after 36 months compared with the placebo group (1.08 U/mL vs. 5.10 U/mL; p = 0.0007). The results were validated through the repeated measures of variance evaluation. The PAI-1 levels showed an equally significant decrease in the active group (26.2 ng/mL vs. 49.2 ng/mL; p = 0.0002) and were also validated through repeated measures of variance evaluation.CONCLUSION: In this sub-study on elderly receiving selenium and coenzyme Q10, or placebo we found significantly lower levels of vWf and PAI-1 in the active treatment group as compared to the placebo group. We interpret this as a better endothelial function because of the intervention, which accords with a previous finding of reduced CV mortality.

    • Engine block mounted accelerometers was used for estimating combustion phasing parameters for a heavy duty diesel engine. Several sensor locations were evaluated for its sensitivity to combustion related vibrations and mechanical noise.One sensor-cylinder combination was selected for the evaluation of a simple algorithm to detect 10% and 50% burned mass fraction. The algorithm uses the accumulated accelerometer vibration energy as a base for extracting the 10% and 50% points respectively.The results show that the angular positions for 10% and 50% burned mass fraction can be estimated with precisions of 1.5 and 3 CAD respectively. It is also concluded that the early part of the accelerometer signal has a significant influence from a mechanical noise source related to the start of injection.

    • We show that water-based porosimetry (WBP), a facile, simple, and nondestructive porosimetry technique, accurately evaluates both the pore size distribution and throat size distribution of sacrificially templated macroporous alumina. The pore size distribution and throat size distribution derived from the WBP evaluation in uptake (imbibition) and release (drainage) mode, respectively, were corroborated by mercury porosimetry and X-ray micro-computed tomography (μ-CT). In contrast with mercury porosimetry, the WBP also provided information on the presence of “dead-end pores” in the macroporous alumina.

    • This thesis presents a system analysis for co-operation in local electricity markets including distributors and customers. The purpose of co-operation is to minimise the system cost of local markets by introducing system measures, such as end-use measures and municipal co-generation plants. Co-operation will strengthen the position of local markets in the national as well as future international electricity markets. With end-use measures local markets will achieve flexibility, additional reserve capacity and ability to avoid sudden large costs for peak loads. Biomass-fired cogeneration plants can become of great importance in an international market. In Sweden there is a simultaneous demand for electricity and district heating, many local markets already include district heating systems and there are major forest areas which can contribute with renewable fuel. The system analysis is partly based on the simulation model (INDSIM) and the linear programming model (MODEST). The simulation model has been further developed (STRATO) to include calculation of system costs. Shadow price analysis has been developed in order to study incentives for system measures. Calculation procedures have been developed that describe cooperation between distributor and customer. Six case studies of a selection of real, existing local markets in Sweden are presented. The studies show the potential economical effects of co-operation measured by system costs and shadow prices. Co-operation has been considered between demand- and supply-side, electricity- and district heating systems and also between different time periods. In a typical local market with 90 000 inhabitants, if end use measures are introduced without cooperation the system cost of the distributor will increase by 14 million SEK for a time period of 25 years. If instead end-use measures are introduced in co-operation, together with a biomass-fired cogeneration plant, the system cost of the local market will be reduced by 444 million SEK. Furthermore, the use of biomass in the local market is increased from 36 to 72 % while the use of oil is decreased from 34 to 1%. Another case study of another local market (50 000 inhabitants) shows that end-use measures will reduce the system cost (excluding investment costs) of an industry by 50 % corresponding to 1.3 million SEK for one year. The end-use measures imply reduced power demand during peak load periods in the local market and increased power demand during non- peak load periods.

The goal is to enable users to self-monitor their well-being status over time and improve their life-style via tailored user guidance. Building the multisensory mirror requires addressing significant scientific and technological challenges, from touch-less data acquisition, to real-time processing and integration of multimodal data.

The potential of these fibers as reinforcement for an epoxy matrix (EP) was investigated in this work. Networks of porous wood fibers were impregnated with monomeric epoxy and cured. Microscopy images from ultramicrotomed cross sections and tensile fractured surfaces were used to study the distribution of matrix inside and around the fibers - at two different length scales. Mechanical characterization at different relative humidity showed much improved mechanical properties of biocomposites based on epoxy-impregnated fibers and they were rather insensitive to surrounding humidity. Furthermore, the mechanical properties of cellulose-fiber biocomposites were compared with those of cellulose-nanofibril (CNF) composites; strong similarities were found between the two materials. The reasons for this, some limitations and the role of specific surface area of the fiber are discussed.

    • Interaction between a solid copper jet and an electric current pulse is a complex process that has been experimentally studied by letting a jet created by a shaped charge device pass through an electrode configuration consisting of two aluminum plates with a separation distance of 150 mm. When the jet bridged the electrodes, which are connected to a charged pulsed power supply, current pulses with amplitude up to 250 kA were passed through the jet. By using flash X-ray diagnostics, the disruption of the electrified jets could be studied. In this paper, the disruption of the electrified jets is discussed and compared with disruption phenomena observed in electrically exploded metal rods in a static setup. Necks are naturally formed along a stretching jet, and in the experiments with current interaction these necks explode electrically. In the static experiments, the metal rods have small notches distributed along the rod to resemble the necks of the jet. When two neighboring necks or notches explode, the shock of the explosion compresses the intermediate jet or rod segment axially and the material is forced out radially. The disruption phenomena in the jet and rod experiments are similar with rapid expansion of the metal at explosion and at comparable velocities.

    • In this paper we present a study of the interaction between all electric current pulse and a solid copper jet. Experiments were performed using a dedicated pulsed power supply delivering a current pulse of such amplitude, rise little, and duration that the jet is efficiently affected. The copper jet was created by using a shaped charge warhead. All electrode configuration consisting of two aluminum plates with a separation distance of 150 mm was used. The discharge current pulse and the voltages at the capacitors and at the electrodes were measured to obtain data oil energy deposition in and the resistance of the jet and electrode contact region. X-ray diagnostics were used to radiograph the jet, and by analyzing the radiograph, the degree of disruption of the electrified jet could be obtained. It was found that a current pulse with an amplitude of 200-250 kA and a rise time of 16 mu s could strongly enhance the natural fragmentation of the jet. In this case, the initial electric energy was 100 kJ and about 90% of the electric energy was deposited in the jet and electrodes. At the exit of the electrode region, the jet fragments formed rings with a radial velocity of up to 200 m/s, depending oil the initial electric energy in the pulsed power supply.     • The interaction between a solid copper jet and an electric current pulse is studied. Copper jets that were created by a shaped-charge device were passed through an electrode configuration consisting of two aluminum plates with a separation distance of 150 mm. The electrodes were connected to a pulsed-power supply delivering a current pulse with amplitudes up to 250 kA. The current and voltages were measured, providing data on energy deposition in the jet and electrode contact region, and flash X-ray diagnostics were used to depict the jet during and after electrification. The shape of, and the velocity distributions along, the jet has been used to estimate the correlation between the jet mass flow through the electrodes and the electrical energy deposition. On average, 2.8 kJ/g was deposited in the jet and electrode region, which is sufficient to bring the jet up to the boiling point. A model based on the assumption of a homogenous current flow through the jet between the electrodes underestimates the energy deposition and the jet resistance by a factor 5 compared with the experiments, indicating a more complex current flow through the jet. The experimental results indicate the following mechanism for the enhancement of jet breakup. When electrified, the natural-formed necks in the jet are subjected to a higher current density compared with other parts of the jet. The higher current density results in a stronger heating and a stronger magnetic pinch force. Eventually, the jet material in the neck is evaporated and explodes electrically, resulting in a radial ejection of vaporized jet material.

  • Modeling of a small helical magnetic flux compression generator

    •  In order to gain experience in explosive pulsed power and to provide experimental data as the basis for computer modeling, a small high-explosive-driven helical magnetic flux-compression generator (FCG) was designed at the Swedish Defence Research Agency. The generator, of which three have been built, has an overall length of 300 mm and a diameter of 70 mm. It could serve as the energy source in a pulse-forming network to generate high-power pulses for various loads. This paper presents a simulation model of this helical FCG. The model, which was implemented in Matlab-Simulink, uses analytical expressions for the generator inductance. The model of resistive losses takes into account the heating of the conductors and the diffusion of the magnetic field into the conductors. The simulation results are compared with experimental data from two experiments with identical generators but with different seed currents, influencing the resistive losses. The model is used to analyze the performance of the generator.

    • In order to gain experience in explosive pulsed power and to provide experimental data for modeling, a small high-explosive-driven helical magnetic flux-compression generator (FCG) was designed at the Swedish Defence Research Agency (FOI). The generator, of which three have been built, has an overall length of 300 mm and a diameter of 70 mm. It could serve as the energy source in a pulse-forming network to generate high power pulses for various loads. This paper presents the design of, and tests with, this helical FCG. The generator had an initial inductance of 23 mu H and was operated into a load of 0.2 mu H. The generator is charged with 0.27 kg of high explosives (PBXN-5). Various types of diagnostics were used to monitor the operation of the generator, including current probes, optical fibers, and piezo gauges. With seed currents of 5.7 and 11.2 kA, final currents of 269 and 436 kA were obtained, corresponding to current amplification factors of 47 and 39. The peak of the current was reached about 30 mu s after the time of crowbar. The two generators showed only small losses in terms of 2 pi-clocking. Using signals from optical fibers, the deflection angle of the armature could be determined to be 10 degrees in good agreement with hydrodynamic simulations of the detonation process and the detonation velocity to be 8.7 km/s in agreement with tabulated value.

    • This report presents experimental setups and findings from the REDO project, which had been conducted between December 2019 and February 2023. Five main topics are covered in this report: 1) Effects of latency and field-of-view on driving performance; 2) Remote driving feedback and control; 3) Connectivity and mobile network support for remote driving; 4) Video transmission for remote driving; and 5) Laws and regulations concerning remote driving. Contents of this report dives into technical details and findings within each topic. Nevertheless, this report does not intend to repeat all detail and results published in scientific publications, and thus this report should be seen as complementary material to the published results.
