    • Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data.

  • Safety and Protection Measures of Underground Non-Coal Mines with Mining Depth over 800 m: A Case Study in Shandong, China

    • With the increase in mining depth, the risk of ground pressure disasters in yellow gold mines is becoming more and more serious. This paper carries out a borehole test for the pressure behavior in a non-coal mining area with a mining depth of more than 800 m in the Jiaodong area. The test results show that under a depth of 1050 m, the increase in the vertical principal stress is the same as the increase in the minimum horizontal principal stress, which is about 3 MPa per 100 m. When the depth increases to 1350 m, the vertical principal stress increases by about 3% per 100 m, and the self-weight stress and the maximum horizontal principal stress maintain a steady growth rate of about 3 MPa per 100 m. In addition, based on the test results, the operation of the ground pressure monitoring system in each mine is investigated. The investigation results show that in some of the roadway and stope mines with depths of more than 800 m, varying degrees of rock mass instability have occurred, and a few mines have had sporadic slight rockbursts, accounting for about 5%. There was a stress concentration area in the lower part of the goaf formed in the early stage of mining, and slight rockburst phenomena such as rock mass ejection have occurred; meanwhile, the area stability for normal production and construction was good, and there was no obvious ground pressure. This paper compares the researched mines horizontally as well as to international high-level mines and puts forward some suggestions, including: carrying out ground pressure investigations and improving the level of intelligence, which would provide countermeasures to balance the safety risks of deep mining, reducing all kinds of safety production accidents and providing a solid basis for risk prevention and supervision.

  • Research on Gob-Side Entry Retaining Mining of Fully Mechanized Working Face in Steeply Inclined Coal Seam: A Case in Xinqiang Coal Mine

    • As a kind of non-coal pillar roadway support technique, gob-side entry retaining is of great significance to improve the production efficiency of a fully mechanized working face. However, the construction of the roadway is often subject to the surrounding rock conditions, the application is mainly concentrated in the nearly horizontal and gently inclined coal seam conditions, and the application in the steeply inclined coal seam conditions is relatively less. This paper is based on the gob-side entry retaining roadway construction of the 58# upper right 3# working face in the fifth district of Xinqiang Coal Mine, and describes the investigation in which we measured the advanced abutment stress, mining stress, and roof stress and analyzed the moving rule of roof. On this basis, in this work, we determined the filling parameters and process and investigated the filling effect from the perspective of the deformation of the filling body and the surrounding rock. The results show that the influence range of the advanced abutment stress in the working face is about 20~25 m, the stress in the upper part is intense, and stress in the middle and lower parts are relaxed. The setting load, the cycle-end resistance, and the time-weighted mean resistance at the upper end of working face along the direction of length are the largest, followed by the middle part, and the lower end is the minimum. When exploiting the steep inclined coal seam, the upper part of the working face is more active than the lower part, and the damaging range of overlaying strata is mainly in the upper part of the goaf. With this research, we established the filling mining process in steeply inclined coal seams and determined the relevant parameters. The gangue cement mortar filling can ensure the deformation of the filling body, the surrounding rock of the roadway is small in the process of roadway retention, and the stress of the filling body is also small, which ensure the successful retention of the roadway. This study verifies the possibility of repair-less exploitation and provides a reference for the popularization and application of the gob-side entry retaining technique in steep inclined coal seam.

    • Inflammatory bowel disease (IBD) is a recurrent chronic inflammatory condition of the intestine without any efficient therapeutic regimens. Gut microbiota, which plays an instrumental role in the development and maturation of the immune system, has been implicated in the pathogenesis of IBD. Emerging evidence has established that early-life events particularly maternal influences and antibiotic treatment are strongly correlated with the health or susceptibility to disease of an individual in later life. Thus, it is proposed that there is a critical period in infancy, during which the environmental exposures bestow a long-term pathophysiological imprint. This notion sheds new light on the development of novel approaches for the treatment, i.e., early interventions, more precisely, the prevention of many uncurable chronic inflammatory diseases like IBD. In this review, we have integrated current evidence to describe the feasibility of the "able-to-be-regulated microbiota," summarized the underlying mechanisms of the "microbiota-driven immune system education," explored the optimal intervention time window, and discussed the potential of designing early-probiotic treatment as a new prevention strategy for IBD.

  • A comparative study of satellite altimetry-based and DEM-based methods for estimating lake water volume changes

    • This study compared two different methods, the satellite altimetry-based and DEM (digital elevation model)-based, for estimating lake water volume changes. We focused on 34 lakes in China as the testing sites to compare the two methods for lake water volume changes from 2005 to 2020. The satellite altimetry-based method used water levels provided by the DAHITI (Database for Hydrological Time Series of Inland Waters) data and surface areas derived from Landsat imagery. The DEM-based method used the SRTM DEM data in combination with Landsat-derived lake extents. Our results showed a high degree of consistency in lake water volume changes estimated between the two methods (R2 . 0.90), but each method has its limitations. In terms of temporal coverage, the satellite altimetry-based method with the DAHITI data is limited by missing water level data in certain periods. The performance of the DEM-based method in extracting lake shore boundaries in regions with flat terrains (slope,1.5°) is not satisfactory. The DEM-based method has complete regional applicability (100%) in the Tibetan Plateau (TP) Lake Region, yet its effectiveness drops significantly in the Xinjiang and Eastern China Plain Lake Regions, with applicability rates of 50 and 40%, respectively.

    • The lack of a sizeable band gap has so far prevented graphene from building effective electronic and optoelectronic devices despite its numerous exceptional properties. Intensive theoretical research reveals that a band gap larger than 1 eV can only be achieved in sub-3 nm wide graphene nanoribbons (GNRs), but real fabrication of such ultranarrow GNRs still remains a critical challenge. Herein, we demonstrate an approach for the synthesis of ultranarrow and photoluminescent semiconducting GNRs by longitudinally unzipping single-walled carbon nanotubes. Atomic force microscopy reveals the unzipping process, and the resulting 2.2 nm wide GNRs are found to emit strong and sharp photoluminescence at similar to 685 nm, demonstrating a very desirable semiconducting nature. This band gap of 1.8 eV is further confirmed by follow-up photoconductivity measurements, where a considerable photocurrent is generated, as the excitation wavelength becomes shorter than 700 nm. More importantly, our fabricated GNR field-effect transistors (FETs), by employing the hexagonal boron nitride-encapsulated heterostructure to achieve edge-bonded contacts, demonstrate a high current on/off ratio beyond 105 and carrier mobility of 840 cm(2)/V s, approaching the theoretical scattering limit in semiconducting GNRs at room temperature. Especially, highly aligned GNR bundles with lengths up to a millimeter are also achieved by prepatterning a template, and the fabricated GNR bundle FETs show a high on/off ratio reaching 10(5), well-defined saturation currents, and strong light-emitting properties. Therefore, GNRs produced by this method open a door for promising applications in graphene-based electronics and optoelectronics.

    • Community-wide blind prediction experiments such as CAPRI and CASP provide an objective measure of the current state of predictive methodology. Here we describe a community-wide assessment of methods to predict the effects of mutations on protein-protein interactions. Twenty-two groups predicted the effects of comprehensive saturation mutagenesis for two designed influenza hemagglutinin binders and the results were compared with experimental yeast display enrichment data obtained using deep sequencing. The most successful methods explicitly considered the effects of mutation on monomer stability in addition to binding affinity, carried out explicit side-chain sampling and backbone relaxation, evaluated packing, electrostatic, and solvation effects, and correctly identified around a third of the beneficial mutations. Much room for improvement remains for even the best techniques, and large-scale fitness landscapes should continue to provide an excellent test bed for continued evaluation of both existing and new prediction methodologies.   • Evaluation of the RF-MEP Method for Merging Multiple Gridded Precipitation Products in the Chongqing City, China

    • Precipitation is a major component of the water cycle. Accurate and reliable estimation of precipitation is essential for various applications. Generally, there are three main types of precipitation products: satellite based, reanalysis, and ground measurements from rain gauge stations. Each type has its advantages and disadvantages. Recent efforts have been made to develop various merging methods to improve precipitation estimates by combining multiple precipitation products. This study evaluated for the first time the performance of the random forest-based merging procedure (RF-MEP) method in enhancing the accuracy of daily precipitation estimates in Chongqing city, China with a complex terrain and sparse observational data. The RF-MEP method was used to merge three widely used gridded precipitation products (CHIRPS, ERA5-Land, and GPM IMERG) with ground measurements from a limited number of rain gauge stations to produce the merged precipitation dataset. Eight stations (approximately 70% of the available stations) were used to train the RF-MEP approach, while four stations (30%) were used for independent testing. Various statistical metrics were employed to assess the performance of the merged precipitation dataset and the three existing precipitation products against the ground measurements. Our results demonstrated that the RF-MEP approach significantly enhances the accuracy of daily precipitation estimates, surpassing the performance of the individual precipitation products and two other merging methods (the simple linear regression model and the simple averaging). Among the three existing products, ERA5-Land exhibited the best performance in capturing daily precipitation, followed by GPM IMERG, while CHIRPS performed the worst. Regarding precipitation intensity, all three existing products and the RF-MEP merged dataset performed well in capturing light precipitation events with an intensity of less than 1 mm/day, which accounts for the majority (more than 70%) of occurrences. However, all datasets showed rather poor capability in capturing precipitation events beyond 1 mm/day, with the worst performance observed for extreme heavy precipitation events exceeding 50 mm/day. The RF-MEP approach significantly improves the detection ability for all precipitation intensities, except for the most extreme intensity (>50 mm/day), where only marginal improvement is observed. Analysis of the spatial pattern of precipitation estimates and the temporal bias of daily precipitation estimates further confirms the superior performance of the RF-MEP merged precipitation dataset over the three existing products.
