  • A new method to assess vehicle airborne non-exhaust particles : Principle, application and emission evaluation

    • A method that could assess the whole vehicle non-exhaust particle (NEP) emission from the whole vehicle level was proposed by combining sealed housing with the chassis dynamometer. The intake and exhaust systems were connected to the out of the chamber to avoid possible effects on NEP emissions. The most distinguishing feature of the proposed method is the whole vehicle assessment of the NEP. Based on the proposed method, three vehicles were tested under various conditions. The results proved that the total NEP from the US06 cycle could be four orders of magnitude higher than that of the WLTC cycle due to the higher thermal load. The transient NEP concentration is highly associated with the friction pairs' temperature, and there are two critical temperatures respectively for volatile particles and solid particles. Mechanical friction-dominated test (WLTC) tends to produce larger particles, while thermal process-dominated tests (US06) tend to emit smaller particles. Solid particles occupy <10% of the total particles, and solid particle is dominated with the size at 10– 23 nm. The higher the brake temperature, the higher the solid particle emissions (both concentration and portion). NEP emission decreases when vehicle miles travelled increase. Total NEP number emission factors could be two orders of magnitude higher than the current PN limit for exhaust particles (6*1011#/km). The solid particle emission factor could also be ten times higher than the current PN limit. O and Fe element distribution matches well with the SEM image, which means the proposed method could successfully sample the brake particles. The carbon element weight is dominated by the organic carbon, which might mean that it's the mechanical abrasion dominate the carbon mass. The top 5 dominant element during ICP/MS analysis is Si, Na, Mg, Zn, and Al.

  • Suppression Mechanism of TiO 2 for the Partial Discharge of Oil-paper Insulation in Intensive Electric Field

    • With the rapid development of modern HVDC transmission technology, higher insulation properties are put forward on the oil-paper insulation system of the transformer, which determine the transformer service life to a certain extent. Traditional transformer oil-paper insulation is becoming increasingly difficult to meet the demands of insulation system with large capacity and miniaturization at ultra-high voltage level. In order to improve the insulation strength of oil-paper system, the insulation cellulose paper modified by TiO2 nanoparticles of different diameters (5 nm, 10 nm, 20 nm, 30 nm) were prepared, in addition, each of modified cellulose paper has different mass fraction of TiO2 nanoparticles (1%, 3%, 5%, 7% wt.). The partial discharge (PD) detection platform was established, and the partial discharge inception voltage (PDIV) values of the oil-paper insulation system with and without nanoparticles were measured. To investigate the PD characteristics, the PD waveforms and PD frequency spectrums of modified cellulose paper and the unmodified were obtained. The suppression mechanism of TiO2 nanoparticles on PD was explored through scanning electron microscope (SEM) observation. All the experiment results indicate that adding nano-TiO2 is beneficial to enhance the insulation properties of oil-paper insulation, and the optimum diameter and mass fraction of TiO2 nanoparticles to suppress oil-paper PD were obtained.

  • Ablation of ERO1A induces lethal endoplasmic reticulum stress responses and immunogenic cell death to activate anti-tumor immunity

    • Immunophenotyping of the tumor microenvironment (TME) is essential for enhancing immunotherapy effi-cacy. However, strategies for characterizing the TME exhibit significant heterogeneity. Here, we show that endoplasmic reticular oxidoreductase-1a (ERO1A) mediates an immune-suppressive TME and attenuates the response to PD-1 blockade. Ablation of ERO1A in tumor cells substantially incites anti-tumor T cell im-munity and promotes the efficacy of aPD-1 in therapeutic models. Single-cell RNA-sequencing analyses confirm that ERO1A correlates with immunosuppression and dysfunction of CD8+ T cells along anti-PD-1 treatment. In human lung cancer, high ERO1A expression is associated with a higher risk of recurrence following neoadjuvant immunotherapy. Mechanistically, ERO1A ablation impairs the balance between IRE1a and PERK signaling activities and induces lethal unfolded protein responses in tumor cells undergoing endoplasmic reticulum stress, thereby enhancing anti-tumor immunity via immunogenic cell death. These findings reveal how tumor ERO1A induces immunosuppression, highlighting its potential as a therapeutic target for cancer immunotherapy.

    • Ferritin is an important iron storage protein, which is widely existed in all forms of life. Ferritin can regulate iron homeostasis when iron ions are lacking or enriched in the body, so as to avoid iron deficiency diseases and iron poisoning. Ferritin presents a hollow nanocage, which can store ions or other small molecular substances in the cavity. Therefore, ferritin shows its potential as a functional nanomaterial that can deliver nutrients or drugs in a targeted manner to improve bioavailability. Due to the special structure, the research on ferritin has attracted more and more attention in recent years. In this paper, the structural characteristics of ferritin were introduced, and the natural purification and prokaryotic expression methods of ferritin from different sources were described. At the same time, ferritin can bind to small molecules, so that it has the activity of small molecules, to construct a new type of ferritin. As a result, ferritin plays an important role as a nutrient substance, in targeted transport, and disease monitoring, etc. In conclusion, the yield of ferritin can be improved by means of molecular biology. Meanwhile, molecular modification can be used to make ferritin have unique activity and function, which lays a foundation for subsequent research.

  • Improvement of thermal stability of oyster (Crassostrea gigas) ferritin by point mutation

    • Ferritin can be widely used as functional nanomaterial. But the physiological activity of ferritin can be damaged under excessive temperatures, which affect the self-assembly property. In this study, point mutation was produced in Asp120 to Gly120 of ferritin amino acid sequence and the heat resistance was improved significantly. The thermal denaturation temperature of mutated ferritin is 89.17 degrees C and has increased by 13 degrees C more than the wildtype oyster ferritin. The effect of thermal treatment on the denaturation, aggregation state, particle size and the structure of ferritin was not changed before 90 degrees C. The computational modeling and analysis indicated that mutated ferritin promotes the overall structural stability assembly via decreasing the interaction energies of 62 percent energies in 3-fold interface. Improving the thermal stability of oyster ferritin by point mutation enhances its applications as a food ingredient.

    • Robust protein-nanomaterial surface analysis is important, but also a challenge. Thrombin plays an important role in the coagulant activity of protein corona mediated by Ca2+ ion exchanged zeolites. However, the mech-anism for this modulation remains unresolved. In this study, we proposed a combined computational and experimental approach to determine the adsorbed sites and orientations of thrombin binding to Ca2+-exchanged LTA-type (CaA) zeolite. Specifically, fourteen ensembles of simulated annealing molecular dynamics (SAMD) simulations and experimental surface residues microenvironment analysis were used to reduce the starting orientations needed for further molecular dynamics (MD) simulations. The combined MD simulations and pro -coagulant activity characterization also reveal the consequent corresponding deactivation of thrombin on CaA zeolite. It is mainly caused by two aspects: (1) the secondary structure of thrombin can change after its adsorption on the CaA zeolite. (2) The positively charged area of thrombin mediates the preferential interaction between thrombin and CaA zeolite. Some thrombin substrate sites are thus blocked by zeolite after its adsorption. This study not only provides a promising method for characterizing the protein-nanoparticle interaction, but also gives an insight into the design and application of zeolite with high procoagulant activity.

  • Binding modes of prothrombin cleavage site sequences to the factor Xa catalytic triad : Insights from atomistic simulations

    • Prothrombin is a key zymogen of the coagulation process and can be converted to thrombin by the prothrombinase complex, which consists of factor Xa (FXa), cofactor Va (FVa), and phospholipids. Prothrombin can be activated at two cleavage sites, R271 and R320, which generates two intermediates: prethrombin-2 via the initial cleavage at R271, and meizothrombin via the first cleavage at R320. Several mechanisms have been proposed to explain this activation preference, but the role of cleavage site sequences in prothrombin activation has not been thoroughly investigated. Here, we used an advanced sampling technique, parallel tempering metadynamics with a well-tempered ensemble (PTMetaDWTE), to study the binding modes of prothrombin cleavage site sequences R266AIEGRTATSEY277 (denoted as Pep271) and S315YIDGRIVEGSD326 (denoted as Pep320) to the FXa catalytic triad. Our study indicates that there exist three binding modes for Pep271 to the FXa catalytic triad but only one binding mode for Pep320 to the FXa catalytic triad. Further molecular dynamics simulations revealed that due to the strong electrostatic interactions, especially the H-bond interactions and salt bridges formed between Pep320 and FXa, the binding mode in the Pep320-FXa system is more stable than the binding modes in the Pep271-FXa system. In view of experimental observations and our results that there exists only one binding mode for Pep320 to the FXa catalytic triad and especially R320 in Pep320 can stably bind to the FXa catalytic triad, we believe that the first cleavage at R320 is favored.

    • Compared with linear time invariant systems, linear periodic system can describe the periodic processes arising from nature and engineering more precisely. However, the time-varying system parameters increase the difficulty of the research on periodic system, such as stabilization and observation. This article aims to consider the observation problem of periodic systems by bridging two fundamental filtering algorithms for periodic systems with a sensor network: consensus-on-measurement-based distributed filtering (CMDF) and centralized Kalman filtering (CKF). First, one mild convergence condition based on uniformly collective observability is established for CMDF, under which the filtering performance of CMDF can be formulated as a symmetric periodic positive semidefinite solution to a discrete-time periodic Lyapunov equation. Then, the closed form of the performance gap between CMDF and CKF is presented in terms of the information fusion steps and the consensus weights of the network. Moreover, it is pointed out that the estimation error covariance of CMDF exponentially converges to the centralized one with the fusion steps tending to infinity. Altogether, these new results establish a concise and specific relationship between distributed and centralized filterings, and formulate the tradeoff between the communication cost and distributed filtering performance on periodic systems. Finally, the theoretical results are verified with numerical experiments.

    • Colloidal quantum dot (QD)-based photoelectrochemical (PEC) cells are cost-effective devices showing remarkable solar-to-fuel conversion efficiency. However, the extensive use of highly toxic elements (e.g. Pb and Cd) in QDs' synthesis and device fabrication is still a major challenge towards their practical development. Herein, we fabricate a solar-driven PEC cell based on environmentally friendly Mn-alloyed CuInS2 (MnCIS)/ZnS core/shell QDs, showing more favorable band alignment, efficient charge transfer, reduced charge recombination and lower charge transfer resistance with respect to the control device fabricated using unalloyed CuInS2 (CIS)/ZnS core/shell QDs. An unprecedented photocurrent density of ∼5.7 mA cm−2 with excellent stability was obtained for the as-fabricated MnCIS/ZnS core/shell QD-based PEC device when operated under standard one sun irradiation (AM 1.5G, 100 mW cm−2). These results indicate that the transition metal-alloyed environmentally friendly core/shell QDs are promising for next-generation solar technologies.

  • Expanding the mutation and phenotype spectrum of MYH3-associated skeletal disorders

    • Pathogenic variants in MYH3 cause distal arthrogryposis type 2A and type 2B3 as well as contractures, pterygia and spondylocarpotarsal fusion syndromes types 1A and 1B. These disorders are ultra-rare and their natural course and phenotypic variability are not well described. In this study, we summarize the clinical features and genetic findings of 17 patients from 10 unrelated families with vertebral malformations caused by dominant or recessive pathogenic variants in MYH3. Twelve novel pathogenic variants in MYH3 (NM_002470.4) were identified: three of them were de novo or inherited in autosomal dominant way and nine were inherited in autosomal recessive way. The patients had vertebral segmentation anomalies accompanied with variable joint contractures, short stature and dysmorphic facial features. There was a significant phenotypic overlap between dominant and recessive MYH3-associated conditions regarding the degree of short stature as well as the number of vertebral fusions. All monoallelic variants caused significantly decreased SMAD3 phosphorylation, which is consistent with the previously proposed pathogenic mechanism of impaired canonical TGF-β signaling. Most of the biallelic variants were predicted to be protein-truncating, while one missense variant c.4244T>G,p.(Leu1415Arg), which was inherited in an autosomal recessive way, was found to alter the phosphorylation level of p38, suggesting an inhibition of the non-canonical pathway of TGF-β signaling. In conclusion, the identification of 12 novel pathogenic variants and overlapping phenotypes in 17 affected individuals from 10 unrelated families expands the mutation and phenotype spectrum of MYH3-associated skeletal disorders. We show that disturbances of canonical or non-canonical TGF-β signaling pathways are involved in pathogenesis of MYH3-associated skeletal fusion (MASF) syndrome.
