    • Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data.

    • The Visual Object Tracking challenge VOT2019 is the seventh annual tracker benchmarking activity organized by the VOT initiative. Results of 81 trackers are presented; many are state-of-the-art trackers published at major computer vision conferences or in journals in the recent years. The evaluation included the standard VOT and other popular methodologies for short-term tracking analysis as well as the standard VOT methodology for long-term tracking analysis. The VOT2019 challenge was composed of five challenges focusing on different tracking domains: (i) VOT-ST2019 challenge focused on short-term tracking in RGB, (ii) VOT-RT2019 challenge focused on "real-time" short-term tracking in RGB, (iii) VOT-LT2019 focused on long-term tracking namely coping with target disappearance and reappearance. Two new challenges have been introduced: (iv) VOT-RGBT2019 challenge focused on short-term tracking in RGB and thermal imagery and (v) VOT-RGBD2019 challenge focused on long-term tracking in RGB and depth imagery. The VOT-ST2019, VOT-RT2019 and VOT-LT2019 datasets were refreshed while new datasets were introduced for VOT-RGBT2019 and VOT-RGBD2019. The VOT toolkit has been updated to support both standard short-term, long-term tracking and tracking with multi-channel imagery. Performance of the tested trackers typically by far exceeds standard baselines. The source code for most of the trackers is publicly available from the VOT page. The dataset, the evaluation kit and the results are publicly available at the challenge website(1).

    • The Visual Object Tracking challenge VOT2016 aims at comparing short-term single-object visual trackers that do not apply pre-learned models of object appearance. Results of 70 trackers are presented, with a large number of trackers being published at major computer vision conferences and journals in the recent years. The number of tested state-of-the-art trackers makes the VOT 2016 the largest and most challenging benchmark on short-term tracking to date. For each participating tracker, a short description is provided in the Appendix. The VOT2016 goes beyond its predecessors by (i) introducing a new semi-automatic ground truth bounding box annotation methodology and (ii) extending the evaluation system with the no-reset experiment.

  • Comparative genomics reveals insights into avian genome evolution and adaptation

    • Birds are the most species-rich class of tetrapod vertebrates and have wide relevance across many research fields. We explored bird macroevolution using full genomes from 48 avian species representing all major extant clades. The avian genome is principally characterized by its constrained size, which predominantly arose because of lineage-specific erosion of repetitive elements, large segmental deletions, and gene loss. Avian genomes furthermore show a remarkably high degree of evolutionary stasis at the levels of nucleotide sequence, gene synteny, and chromosomal structure. Despite this pattern of conservation, we detected many non-neutral evolutionary changes in protein-coding genes and noncoding regions. These analyses reveal that pan-avian genomic diversity covaries with adaptations to different lifestyles and convergent evolution of traits.

  • Gene-educational attainment interactions in a multi-ancestry genome-wide meta-analysis identify novel blood pressure loci

    • Educational attainment is widely used as a surrogate for socioeconomic status (SES). Low SES is a risk factor for hypertension and high blood pressure (BP). To identify novel BP loci, we performed multi-ancestry meta-analyses accounting for gene-educational attainment interactions using two variables, “Some College” (yes/no) and “Graduated College” (yes/no). Interactions were evaluated using both a 1 degree of freedom (DF) interaction term and a 2DF joint test of genetic and interaction effects. Analyses were performed for systolic BP, diastolic BP, mean arterial pressure, and pulse pressure. We pursued genome-wide interrogation in Stage 1 studies (N = 117 438) and follow-up on promising variants in Stage 2 studies (N = 293 787) in five ancestry groups. Through combined meta-analyses of Stages 1 and 2, we identified 84 known and 18 novel BP loci at genome-wide significance level (P < 5 × 10-8). Two novel loci were identified based on the 1DF test of interaction with educational attainment, while the remaining 16 loci were identified through the 2DF joint test of genetic and interaction effects. Ten novel loci were identified in individuals of African ancestry. Several novel loci show strong biological plausibility since they involve physiologic systems implicated in BP regulation. They include genes involved in the central nervous system-adrenal signaling axis (ZDHHC17, CADPS, PIK3C2G), vascular structure and function (GNB3, CDON), and renal function (HAS2 and HAS2-AS1, SLIT3). Collectively, these findings suggest a role of educational attainment or SES in further dissection of the genetic architecture of BP.

    • Genetic variants within nearly 1000 loci are known to contribute to modulation of blood lipid levels. However, the biological pathways underlying these associations are frequently unknown, limiting understanding of these findings and hindering downstream translational efforts such as drug target discovery.To expand our understanding of the underlying biological pathways and mechanisms controlling blood lipid levels, we leverage a large multi-ancestry meta-analysis (N=1,654,960) of blood lipids to prioritize putative causal genes for 2286 lipid associations using six gene prediction approaches. Using phenome-wide association (PheWAS) scans, we identify relationships of genetically predicted lipid levels to other diseases and conditions. We confirm known pleiotropic associations with cardiovascular phenotypes and determine novel associations, notably with cholelithiasis risk. We perform sex-stratified GWAS meta-analysis of lipid levels and show that 3-5% of autosomal lipid-associated loci demonstrate sex-biased effects. Finally, we report 21 novel lipid loci identified on the X chromosome. Many of the sex-biased autosomal and X chromosome lipid loci show pleiotropic associations with sex hormones, emphasizing the role of hormone regulation in lipid metabolism.Taken together, our findings provide insights into the biological mechanisms through which associated variants lead to altered lipid levels and potentially cardiovascular disease risk.

  • Safety and Protection Measures of Underground Non-Coal Mines with Mining Depth over 800 m: A Case Study in Shandong, China

    • With the increase in mining depth, the risk of ground pressure disasters in yellow gold mines is becoming more and more serious. This paper carries out a borehole test for the pressure behavior in a non-coal mining area with a mining depth of more than 800 m in the Jiaodong area. The test results show that under a depth of 1050 m, the increase in the vertical principal stress is the same as the increase in the minimum horizontal principal stress, which is about 3 MPa per 100 m. When the depth increases to 1350 m, the vertical principal stress increases by about 3% per 100 m, and the self-weight stress and the maximum horizontal principal stress maintain a steady growth rate of about 3 MPa per 100 m. In addition, based on the test results, the operation of the ground pressure monitoring system in each mine is investigated. The investigation results show that in some of the roadway and stope mines with depths of more than 800 m, varying degrees of rock mass instability have occurred, and a few mines have had sporadic slight rockbursts, accounting for about 5%. There was a stress concentration area in the lower part of the goaf formed in the early stage of mining, and slight rockburst phenomena such as rock mass ejection have occurred; meanwhile, the area stability for normal production and construction was good, and there was no obvious ground pressure. This paper compares the researched mines horizontally as well as to international high-level mines and puts forward some suggestions, including: carrying out ground pressure investigations and improving the level of intelligence, which would provide countermeasures to balance the safety risks of deep mining, reducing all kinds of safety production accidents and providing a solid basis for risk prevention and supervision.

    • Solar-powered interfacial evaporation has emerged as an innovative and sustainable technology for clean water production. However, the rapid, mass and shape-controlled fabrication of three-dimensional (3D) steam generators (SGs) for versatile hybrid applications remains challenging. Herein, composite aerogel beads with self-contained properties (i.e., hydrophilic, porous, photothermal, and durable) are developed and demonstrated for threefold hybrid applications including efficient solar-powered interfacial evaporation, water remediation, and controlled soil enrichment. The rational incorporation of selected polysaccharides enables us to fabricate bead-like aerogels with rapid gelation, continuous processing, and enhanced ion adsorption. The composite beads can attain a high water evaporation rate of 1.62 kg m-2 h-1 under 1 sun. Meanwhile, high phosphate adsorption capacity of over 120 mg g-1 is achieved in broad pH (2.5-12.4) and concentration (200-1000 mg L-1) ranges of phosphate solutions. Gratifyingly, we demonstrate the first example of recycling biomaterials from interfacial SGs for controlled nutrient release, soil enrichment, and sustainable agriculture. The phosphate-saturated beads can be gradually broken down in the soil. Macronutrients (N, P, and K) can be slowly released in 50 days, sustaining the plant germination and growth in a whole growth stage. This work shines light on the mass and controlled fabrication of aerogel beads based on double-network biopolymers, not merely scaling up solar-powered interfacial evaporation but also considering water remediation, waste material disposal, and value-added conversion.

  • Quartet DNA reference materials and datasets for comprehensively evaluating germline variant calling performance

    • BACKGROUND: Genomic DNA reference materials are widely recognized as essential for ensuring data quality in omics research. However, relying solely on reference datasets to evaluate the accuracy of variant calling results is incomplete, as they are limited to benchmark regions. Therefore, it is important to develop DNA reference materials that enable the assessment of variant detection performance across the entire genome.RESULTS: We established a DNA reference material suite from four immortalized cell lines derived from a family of parents and monozygotic twins. Comprehensive reference datasets of 4.2 million small variants and 15,000 structural variants were integrated and certified for evaluating the reliability of germline variant calls inside the benchmark regions. Importantly, the genetic built-in-truth of the Quartet family design enables estimation of the precision of variant calls outside the benchmark regions. Using the Quartet reference materials along with study samples, batch effects are objectively monitored and alleviated by training a machine learning model with the Quartet reference datasets to remove potential artifact calls. Moreover, the matched RNA and protein reference materials and datasets from the Quartet project enables cross-omics validation of variant calls from multiomics data.CONCLUSIONS: The Quartet DNA reference materials and reference datasets provide a unique resource for objectively assessing the quality of germline variant calls throughout the whole-genome regions and improving the reliability of large-scale genomic profiling.

  • Transmission of clones of carbapenem-resistant Escherichia coli between a hospital and an urban wastewater treatment plant*

    • Carbapenem-resistant Enterobacterales (CRE) constitute an urgent threat to worldwide public health. The spread of CRE is facilitated by transmission via the environment. Wastewater treatment plants (WWTPs) are considered to be important sources of antibiotic resistance and hot spots of antibiotic-resistant bacteria (ARB) which can facilitate dissemination of antibiotic resistance genes. In this study, water samples were collected over one year from a WWTP in Jinan, Shandong province, China, from different functional sites in the wastewater treatment process. Carbapenem-resistant Escherichia coli (CREC) were isolated by selective cultivation and whole-genome sequenced to investigate the occurrence and characteristics of CREC in the WWTP. A total of 77 CREC isolates were included in the study and the detection rate of CREC in the WWTP water inlet was found to be 85%. An additional 10 CREC were isolated from a nearby teaching hospital during the sampling period and included for comparison to the environmental isolates. Susceptibility testing showed that all CREC were multidrug-resistant. 6 different carbapenem resistance genes (CRGs) were detected, including blaNDM-5 (n = 75), blaNDM-1 (n = 6), blaNDM-4 (n = 3), blaNDM-6 (n = 1), blaNDM-9 (n = 1), and blaKPC-2 (n = 4). 42 CREC isolates were whole-genome sequenced with Illumina short-read sequencing. 11 of these were also sequenced with Nanopore long-read sequencing. Plasmids carrying CRGs were found to belong to IncX3 (n = 35), IncFII (n = 12), IncFIA (n = 5), IncFIB (n = 2), IncC (n = 1), and IncP6 (n = 1). Clonal dissemination of CREC belonging to ST167, ST448, and ST746 was observed between different parts of the WWTP. Furthermore, isolates from the WWTP, including an isolate belonging to the high-risk ST167 strain, were found to be clonally related to CREC isolated at the hospital. The spread of CRGs is of considerable concern and strategies to prevent environmental dissemination of this contaminant urgently needs to be implemented.

  • Research on Gob-Side Entry Retaining Mining of Fully Mechanized Working Face in Steeply Inclined Coal Seam: A Case in Xinqiang Coal Mine

    • As a kind of non-coal pillar roadway support technique, gob-side entry retaining is of great significance to improve the production efficiency of a fully mechanized working face. However, the construction of the roadway is often subject to the surrounding rock conditions, the application is mainly concentrated in the nearly horizontal and gently inclined coal seam conditions, and the application in the steeply inclined coal seam conditions is relatively less. This paper is based on the gob-side entry retaining roadway construction of the 58# upper right 3# working face in the fifth district of Xinqiang Coal Mine, and describes the investigation in which we measured the advanced abutment stress, mining stress, and roof stress and analyzed the moving rule of roof. On this basis, in this work, we determined the filling parameters and process and investigated the filling effect from the perspective of the deformation of the filling body and the surrounding rock. The results show that the influence range of the advanced abutment stress in the working face is about 20~25 m, the stress in the upper part is intense, and stress in the middle and lower parts are relaxed. The setting load, the cycle-end resistance, and the time-weighted mean resistance at the upper end of working face along the direction of length are the largest, followed by the middle part, and the lower end is the minimum. When exploiting the steep inclined coal seam, the upper part of the working face is more active than the lower part, and the damaging range of overlaying strata is mainly in the upper part of the goaf. With this research, we established the filling mining process in steeply inclined coal seams and determined the relevant parameters. The gangue cement mortar filling can ensure the deformation of the filling body, the surrounding rock of the roadway is small in the process of roadway retention, and the stress of the filling body is also small, which ensure the successful retention of the roadway. This study verifies the possibility of repair-less exploitation and provides a reference for the popularization and application of the gob-side entry retaining technique in steep inclined coal seam.

  • Multiancestry Genome-Wide Association Study of Lipid Levels Incorporating Gene-Alcohol Interactions

    • A person's lipid profile is influenced by genetic variants and alcohol consumption, but the contribution of interactions between these exposures has not been studied. We therefore incorporated gene-alcohol interactions into a multiancestry genome-wide association study of levels of high-density lipoprotein cholesterol, low-density lipoprotein cholesterol, and triglycerides. We included 45 studies in stage 1 (genome-wide discovery) and 66 studies in stage 2 (focused follow-up), for a total of 394,584 individuals from 5 ancestry groups. Analyses covered the period July 2014-November 2017. Genetic main effects and interaction effects were jointly assessed by means of a 2-degrees-of-freedom (df) test, and a 1-df test was used to assess the interaction effects alone. Variants at 495 loci were at least suggestively associated (P < 1 x 10(-6)) with lipid levels in stage 1 and were evaluated in stage 2, followed by combined analyses of stage 1 and stage 2. In the combined analysis of stages 1 and 2, a total of 147 independent loci were associated with lipid levels at P < 5 x 10(-8) using 2-df tests, of which 18 were novel. No genome-wide-significant associations were found testing the interaction effect alone. The novel loci included several genes (proprotein convertase subtilisin/kexin type 5 (PCSK5), vascular endothelial growth factor B (VEGFB), and apolipoprotein B mRNA editing enzyme, catalytic polypeptide 1 (APOBEC1) complementation factor (A1CF)) that have a putative role in lipid metabolism on the basis of existing evidence from cellular and experimental models.

    • In today's district heating (DH) energy market, it is common to use user functional categories in price models to determine the heat price. However, users in the same category do not necessarily have the same energy consumption patterns, which potentially leads to unfair prices and many other practical issues. Taking into account heat usage characteristics, this work proposes two data-driven methods to cluster DH users to identify similar usage patterns, using practical energy consumption data. Efforts are focused on extracting representative features of users from their daily usage profiles and duration curves, respectively. Employing clustering based on these features, the resulting typical usage patterns and user category distributions are discussed. Our results can serve as potential inputs for future energy price models, demand-side management, and load reshaping strategies.

  • Sino-European Differences in the Genetic Landscape and Clinical Presentation of Pheochromocytoma and Paraganglioma

    • Context: Pheochromocytomas and paragangliomas (PPGLs) are characterized by distinct genotype-phenotype relationships according to studies largely restricted to Caucasian populations.Objective: To assess for possible differences in genetic landscapes and genotype-phenotype relationships of PPGLs in Chinese versus European populations.Design: Cross-sectional study.Setting: 2 tertiary-care centers in China and 9 in Europe.Participants: Patients with pathologically confirmed diagnosis of PPGL, including 719 Chinese and 919 Europeans.Main Outcome Measures: Next-generation sequencing performed in tumor specimens with mutations confirmed by Sanger sequencing and tested in peripheral blood if available. Frequencies of mutations were examined according to tumor location and catecholamine biochemical phenotypes.Results: Among all patients, higher frequencies of HRAS, FGFR1, and EPAS1 mutations were observed in Chinese than Europeans, whereas the reverse was observed for NF1, VHL, RET, and SDHx. Among patients with apparently sporadic PPGLs, the most frequently mutated genes in Chinese were HRAS (16.5% [13.6-19.3] vs 9.8% [7.6-12.1]) and FGFR1 (9.8% [7.6-12.11 vs 2.2% [1.1-3.3]), whereas among Europeans the most frequently mutated genes were NF1 (15.9% [13.2-18.6) vs 6.6% [4.7-8.5)) and SDHx (10.7% [8.4-13.0] vs 4.2% [2.6-5.7]). Among Europeans, almost all paragangliomas lacked appreciable production of epinephrine and identified gene mutations were largely restricted to those leading to stabilization of hypoxia inducible factors. In contrast, among Chinese there was a larger proportion of epinephrine-producing paragangliomas, mostly due to HRAS and FGFR1 mutations.Conclusions: This study establishes Sino-European differences in the genetic landscape and presentation of PPGLs, including ethnic differences in genotype-phenotype relationships indicating a paradigm shift in our understanding of the biology of these tumors.

    • Visual tracking has attracted a significant attention in the last few decades. The recent surge in the number of publications on tracking-related problems have made it almost impossible to follow the developments in the field. One of the reasons is that there is a lack of commonly accepted annotated data-sets and standardized evaluation protocols that would allow objective comparison of different tracking methods. To address this issue, the Visual Object Tracking (VOT) workshop was organized in conjunction with ICCV2013. Researchers from academia as well as industry were invited to participate in the first VOT2013 challenge which aimed at single-object visual trackers that do not apply pre-learned models of object appearance (model-free). Presented here is the VOT2013 benchmark dataset for evaluation of single-object visual trackers as well as the results obtained by the trackers competing in the challenge. In contrast to related attempts in tracker benchmarking, the dataset is labeled per-frame by visual attributes that indicate occlusion, illumination change, motion change, size change and camera motion, offering a more systematic comparison of the trackers. Furthermore, we have designed an automated system for performing and evaluating the experiments. We present the evaluation protocol of the VOT2013 challenge and the results of a comparison of 27 trackers on the benchmark dataset. The dataset, the evaluation tools and the tracker rankings are publicly available from the challenge website(1).

Loose coal gob has a hidden ignition location and a high possibility of spontaneous combustion, which makes fire prevention difficult. Therefore, based on the theory of gas seepage and the characteristics of loose coal, a model of air leakage and spontaneous combustion in gob is established in this paper. Using working face #10414 in the Yangliu coal mine as an example, the relationship between the three spontaneous coal combustion (CSC) zones and the three stress zones is analyzed and verified by combining a FLAC3D simulation with field monitoring. In addition, the influence of advancing speed on the CSC is discussed, and suggestions for fire prevention are presented. The results show that the variation in the calorific value of the CSC with increasing degree of looseness of the residual coal in the gob forms an arch‐shape. There is a one‐to‐one relationship between the distribution of the three stress zones and the three CSC zones. In addition, as the advancing speed increases, the contact time between the loose coal body and the air decreases and the possibility of CSC decreases. This study provides a scientific basis for fire prevention and control in mines.

    • Conditional temperature-sensitive (ts) mutations are valuable reagents for studying essential genes in the yeast Saccharomyces cerevisiae. We constructed 787 ts strains, covering 497 (∼45%) of the 1,101 essential yeast genes, with ∼30% of the genes represented by multiple alleles. All of the alleles are integrated into their native genomic locus in the S288C common reference strain and are linked to a kanMX selectable marker, allowing further genetic manipulation by synthetic genetic array (SGA)-based, high-throughput methods. We show two such manipulations: barcoding of 440 strains, which enables chemical-genetic suppression analysis, and the construction of arrays of strains carrying different fluorescent markers of subcellular structure, which enables quantitative analysis of phenotypes using high-content screening. Quantitative analysis of a GFP-tubulin marker identified roles for cohesin and condensin genes in spindle disassembly. This mutant collection should facilitate a wide range of systematic studies aimed at understanding the functions of essential genes.

    • As one of the most common disasters in deep mine roadway, floor heave has caused serious obstacles to mine transportation and normal production activities. The third section winch roadway in the third mining area of Qitaihe Longhu coal mine has a serious floor heave due to the large buried depths of the roadway and the semicoal rock roadway, and the maximum floor heave is 750 mm. For the problem of floor stability, this paper establishes a mechanical model to analyze the stability of roadway floor heave by analogy with the basement heave of deep foundation pit. It provides a model reference for analyzing the problem of roadway floor heave. Aiming at the problem of roadway floor heave in Longhu coal mine, the roadway model is established by using FLAC3D, and the roadway model after support is established according to the on-site support measures. Through the analysis of the distribution of roadway plastic area, stress nephogram, and displacement field simulation results, the results show that the maximum displacement of roadway roof and floor after support is reduced by 15% and 23%, but the maximum floor heave is still 770 mm, which is close to the measured floor heave of roadway. In order to solve the problem of roadway floor heave and integrate economic factors, this paper puts forward three support optimization schemes, simulates the support effect of each scheme, and finally determines that scheme 3 is the best support optimization scheme. Compared with that under the original support, the amount of floor heave is reduced by 81%, and the final amount of floor heave is 150 mm, which can meet the requirements of roadway floor deformation. The results provide a scheme and guidance for roadway support optimization

    • Neurogenesis is an ongoing process in the brains of adult decapod crustaceans. However, the first-generation precursors that produce adult-born neurons, which reside in a neurogenic niche, are not self-renewing in crayfish and must be replenished. The source of these neuronal precursors is unknown. Here, we report that adult-born neurons in crayfish can be derived from hemocytes. Following adoptive transfer of 5-ethynyl-2′-deoxyuridine (EdU)-labeled hemocytes, labeled cells populate the neurogenic niche containing the first-generation neuronal precursors. Seven weeks after adoptive transfer, EdU-labeled cells are located in brain clusters 9 and 10 (where adult-born neurons differentiate) and express appropriate neurotransmitters. Moreover, the number of cells composing the neurogenic niche in crayfish is tightly correlated with total hemocyte counts (THCs) and can be manipulated by raising or lowering THC. These studies identify hemocytes as a source of adult-born neurons in crayfish and demonstrate that the immune system is a key contributor to adult neurogenesis.

    • It is generally known that discontinuities have a remarkable influence on the mechanical behaviour of rock masses. To further understand the fracture mechanisms of jointed rock masses, substantial effort has been focused on the strength anisotropy and failure characteristics of rocks/rock-like specimens containing persistent joints with different geometric parameters. However, only a few laboratory tests have considered the failure mechanism of a rock mass with 3D joints, especially for non-persistent joints with different persistence levels. In the present work, experiments on cubic rock-like specimens containing non-persistent joints (in areal extent) subjected to uniaxial compression were conducted to further investigate the influence of the joint inclination (θ) and persistence (N) on the rock mechanical properties and failure characteristics. The strength of a 3D non-persistent jointed specimen is characterized by three stages as the joint inclination angle (θ) increases from 0° to 90°. The strength of jointed specimens decreases with increasing N for all θ values, with the highest strength obtained for N = 0.42 and the lowest strength recorded for N = 0.92. Based on CT scan results, four typical fracture modes were identified: splitting, splitting + sliding, sliding, and intact failure. Overall, as the joint inclination increases, the failure mode of the specimen transforms from splitting to sliding and then to the intact failure mode. However, with decreasing joint persistence, the failure modes of some specimens will change from sliding to mixed failure (splitting + sliding).

    • The failure and mechanical behavior of transversely isotropic rock are significantly affected by the original bedding planes. Until now, few studies have been performed to investigate the influence of the geometrical and mechanical parameters of the bedding planes on the fracture characteristics of transversely isotropic rocks under planar shear fracture loading conditions. For this purpose, experimental and numerical compression-shear tests on double-notched specimens are conducted to investigate the fracturing characteristics of transversely isotropic rock under planar shear fracture loading. The experimental study that focuses on the influence of bedding plane inclination on fracture load, fracture pattern and AE evolution, and six inclination angles is conducted in this study. Based on the flat joint contact model (for the rock matrix) and smooth joint contact model (for the original bedding plane) in PFC2D (particle flow code), the microscale fracturing process of transversely isotropic rock with different inclinations is simulated and analyzed. The results show that the inclination has an important influence on the fracture load and fracture pattern, and the maximum and minimum fracture loads are obtained for specimens with inclination angles of 30° and 60°, respectively. Moreover, the strength and spacing of the original bedding planes also play an important role in fracture loads. Higher bedding plane strength and wider bedding plane spacing result in higher fracture loads. In addition, with a moderate inclination angle, transversely isotropic rock with higher bedding plane strength tends to form cracks that cut through the rock matrix. However, with the decrease in the bedding plane strength, more fractures form along the bedding planes.

    • Granular materials, such as mineral products, soil and chemical fertilizer, normally contain unsaturated free moisture. This unsaturated moisture could migrate in the granular material and its processing, resulting in handling and safety concerns due to a change in the material properties. In this work, a method of coupling the Discrete Element Method (DEM) and the Smoothed Particle Hydrodynamics (SPH) was proposed to investigate moisture migration in unsaturated granular materials under oscillation. The particles and water were simulated through DEM and SPH, respectively and a python coupling framework was programmed to perform the exchange of particle-fluid interaction forces. Subsequently, experiments with plastic pellets and a coal sample were used to calibrate the models, with DEM calibrated by an angle of repose experiment and draining experiments calibrating the coupled model. Finally, the coupled model was validated by comparing the results to those of experiments involving moisture migration under oscillation. It was found that, compared to measurements, the coupled SPH-DEM model predicted the same moisture migration trend and also provided good agreement for the prediction of the mass of drained water.

    • For dense small cell deployment in centralized baseband architecture, wireless fronthaul is a cost efficient and flexible alternative to fiber-based fronthaul links. Due to the available wide bandwidth, millimeter-wave technology is the most promising choice to realize multi-gigabit wireless transmission of the common public radio interface (CPRI). In this paper, we present a novel differential encoding scheme for differential quadrature phase-shift keying (D-QPSK) modulation, which enables data-rate adaptable CPRI transmission without modification of a standard receiver. A multi-rate D-QPSK modulator/demodulator (modem) is implemented and tested for data rates up to 10 Gbps, limited by the bandwidth of the microwave components in use. This modem is verified to fulfill the CPRI specifications with respect to multi-rate, low latency and high system performance. Therefore, it is proposed for the implementation of wireless CPRI links at millimeter-wave bands.

    • Although plant secondary metabolites are important source of new drugs, obtaining these compounds is challenging due to their high structural diversity and low abundance. The roots of Astragalus membranaceus are a popular herbal medicine worldwide. It contains a series of cycloartane-type saponins (astragalosides) as hepatoprotective and antivirus components. However, astragalosides exhibit complex sugar substitution patterns which hindered their purification and bioactivity investigation. In this work, glycosyltransferases (GT) from A. membranaceus were studied to synthesize structurally diverse astragalosides. Three new GTs, AmGT1/5 and AmGT9, were characterized as 3-O-glycosyltransferase and 25-O-glycosyltransferase of cycloastragenol respectively. AmGT1(G146V/I) variants were obtained as specific 3-O-xylosyltransferases by sequence alignment, molecular modelling and site-directed mutagenesis. A combinatorial synthesis system was established using AmGT1/5/9, AmGT1(G146V/S) and the reported AmGT8 and AmGT8(A394F). The system allowed the synthesis of 13 astragalosides in Astragalus root with conversion rates from 22.6% to 98.7%, covering most of the sugar-substitution patterns for astragalosides. In addition, AmGT1 exhibited remarkable sugar donor promiscuity to use 10 different donors, and was used to synthesize three novel astragalosides and ginsenosides. Glycosylation remarkably improved the hepatoprotective and SARS-CoV-2 inhibition activities for triterpenoids. This is one of the first attempts to produce a series of herbal constituents via combinatorial synthesis. The results provided new biocatalytic tools for saponin biosynthesis.

    • Age-related hearing loss (ARHL) is associated with cognitive dysfunction; however, the detailed underlying mechanisms remain unclear. The aim of this study is to investigate the potential underlying mechanism with a system genetics approach. A transcriptome-wide association study was performed on aged (12-32 months old) BXD mice strains. The hippocampus gene expression was obtained from 56 BXD strains, and the hearing acuity was assessed from 54 BXD strains. Further correlation analysis identified a total of 1,435 hearing-related genes in the hippocampus (p < 0.05). Pathway analysis of these genes indicated that the impaired glutamatergic synapse pathway is involved in ARHL (p = 0.0038). Further gene co-expression analysis showed that the expression level of glutamine synthetase (Gls), which is significantly correlated with ARHL (n = 26, r = -0.46, p = 0.0193), is a crucial regulator in glutamatergic synapse pathway and associated with learning and memory behavior. In this study, we present the first systematic evaluation of hippocampus gene expression pattern associated with ARHL, learning, and memory behavior. Our results provide novel potential molecular mechanisms involved in ARHL and cognitive dysfunction association.

    • Similar to the damage zone in the vicinity of faults, rock joints are composed of single or multiple fracture process zones, highlighting a high crack density and a decrease of Young’s modulus. Here specimens of limestone joint and granite joint were subjected to cyclic tensile stress of increasing amplitudes, up to failure. The cyclic tension tests were to study the elasticity and crack density adjacent to rock joints. Results show that the vast majority of tensile strain accommodates in the vicinity of rock joint, and rock joints have a nonlinear mechanical behavior during loading while a nearly linear behavior during unloading in the tensile stress-strain curves. A gradual reduction in Young’s modulus is seen with the increase of tensile strain. The cumulative crack damage after each stress cycle also induces a decrease in Young’s modulus. A stiffer response is found when switching from loading to unloading. During the unloading, a recovery in tensile strain and crack damage is observed, but some unrecoverable damage remains in the rock joint sample after each loading-unloading cycle. Based on the differential scheme, a constitutive model using two crack-related parameters, i.e., initial crack density and crack density increment, is developed. The model can give a micromechanical explanation and well reproduce the nonlinearity and hysteresis of tensile stress-strain curves.

    • This paper presents an implementation solution for a digital baseband receiver, which consists mainly of an analog symbol timing recovery (STR) block and a digital carrier recovery block. The STR is realized based on "one-sample-per-symbol" sampling, resulting in relaxed requirement on the A/D converters sampling speed. In this sense, the proposed implementation solution is hardware efficient. To functionally verify the solution, a proof-of-concept E-band link system is implemented and tested in the laboratory, which supports 5-Gbit/s data traffic using 16 quadrature amplitude modulation. The test results demonstrate that the proposed solution works for high-capacity millimeter-wave radios for point-to-point links, one of the targeted applications.

    • District Heating (DH) is facing a tough competition in the market. In order to improve its competence, an effective way is to reform price models for DH. This work proposed a new dynamic price model based on the levelized cost of heat (LCOH) and the predicted hourly heat demand. A DH system in Sweden was used as a case study. Three methods were adopted to allocate the fuel cost to the variable costs of heat production, including (1) in proportion to the amount of heat and electricity generation; (2) in proportion to the exergy of generated heat and electricity; and (3) deducting the market price of electricity from the total cost. Results indicated that the LCOH-based pricie model can clearly reflect the production cost of heat. Through the comparison with other market-implemented price models, it was found that even though the market-implemented price models can, to certain extent, reflect the variations in heat demand, they cannot reflect the changes in production cost when different methods of heat production are involved. In addition, price model reforming can lead to a significant change in the expense of consumers and consequently, affect the selection of heating solution.

    • We report the design of an artificial flower-like structure that supports a magnetic plasma in the optical domain. The structure is composed of alternating "petals" of conventional dielectrics (epsilon > 0) and plasmonic materials (Re(epsilon) < 0). The induced effective magnetic current on such a structure possesses a phase lag with respect to the incident TE-mode magnetic field, similar to the phase lag between the induced electric current and the incident TM-mode electric field on a metal wire. An analogy is thus drawn with an artificial electric plasma composed of metal wires driven by a radio frequency excitation. The effective medium of an array of flowers has a negative permeability within a certain wavelength range, thus behaving as a magnetic plasma.

    • Affibody molecules binding to the site of hormone interaction in IGF-IR (insulin-like growth factor-I receptor) were successfully selected by phage-display technology employing a competitive-elution strategy during biopanning, whereby release of receptor-bound phagemids was accomplished by competition with IGFI (insulin-like growth factor-I). In non-competitive selections, the elution of receptor-bound phagemids was performed by imidazole or low-pH incubation, which also resulted in the isolation of affibody molecules that could bind to the receptor. An ELISA-based assay showed that the affibody molecules generated by IGF-I competition during elution, in addition to affibody molecules generated in the noncompetitive selections, could compete with IGF-I for binding to the receptor. The affinities of the isolated variants to IGF-IR-overexpressing MCF-7 cells were determined and ranged from high nanomolar to 2.3 nM. The most promising variant, Z(4;40), was shown to recognize IGF- IR efficiently in several different contexts: in analyses based on flow cytometry, fluorescence microscopy and receptor pull-down from cell extracts. In addition, when Z, was added to the medium of MCF-7 cells that were dependent on IGF-I for efficient growth, it was found to have a dose-dependent growth-inhibitory effect on the cells. Applications of affibody-based reagents for quantitative and qualitative analyses of IGF- I R status, as well as applications of affibody-based reagents for therapy, are discussed.

    • This paper presents the development of a three‐dimensional discrete element model using flat‐joint and smooth‐joint contact models to investigate the effect of anisotropy on the tensile behaviour of slate, a transversely isotropic rock, under Brazilian testing from both macro and microscales. The effect of anisotropy is further realised by exploring the influence of foliation orientations (β and ψ ) on the tensile strength, fracture pattern, microcracking and stress distribution of the transversely isotropic rock. The variation of tensile strength with foliation orientation is presented. The cross‐weak‐plane fracture growth observed in laboratory is reproduced, and the criterion for which to form is also given from the aspect of foliation orientation. Furthermore, the proportional variations of microcracks well account for the effects of foliation orientation on the tensile strength and failure pattern. Finally, it is found that the existence of weak planes increases both the heterogeneity and the anisotropy of stress distributions within the transversely isotropic rock, with the degree of influence varying with the foliation orientation.

    • Combining iron and copper ores can generate an oxygen carrier that has a synergic effect of high temperature resistance and high reactivity. In this work, typical cements available in the market were studied as binders to bind magnetite and chalcopyrite to develop a suitable oxygen carrier for chemical-looping combustion (CLC). A first selection step suggested that an aluminate cement, namely CA70, could favor the generation of oxygen carrier particles having good crushing strength, good particle yield, and high reactivity. The CA70-bonded oxygen carrier was then subjected to cyclic tests with CH4, CO, and H-2 in reduction and in air oxidation at temperatures of 850, 900, and 950 degrees C with gas concentrations of 5, 10, 15, and 20% in a batch-fluidized bed reactor. The increase in temperature promoted the fuel conversion. At 950 degrees C, the conversions of CH4 and CO reached up to 80.4% and 99.2%, respectively. During more than 30 cycles, the oxygen carrier kept a similar reactivity to the fresh carrier and maintained its composition and physical properties. The oxygen transport capacity was maintained at 21-23%, and the phases were CuO, Fe2O3, Al2O3, and minor CaS. In the used sample, some grains were observed, but the morphology was not greatly changed. Agglomeration was absent during all the cycles, except for the deep reduction with H-2.

    • Hole transport materials (HTMs) have greatly advanced the progress of solution-based electronic devices in the past few years. Nevertheless, most devices employing dopant-free organic HTMs can only deliver inferior performance. In this work, we introduced a novel "polymer synergy" strategy to develop versatile dopant-free polymer HTMs for quantum dot/perovskite solar cells and photodetectors. With this synergy strategy, the optical, electrical and aggregation properties of polymer HTMs can be modulated, resulting in complementary absorption, high hole mobility, favorable energy landscape and moderate aggregation. Moreover, a clear orientational transition was observed for the developed HTMs with a 9-fold increase in the face-on/edge-on ratio, providing a highway-like carrier transport for electronic devices, as revealed by in situ characterization and ultrafast transient absorption. With these benefits, the photovoltaic and photodetection performance of quantum dot devices were boosted from 11.8% to 13.5% and from 2.95 x 10(12) to 3.41 x 10(13) Jones (over a 10-fold increase), respectively. Furthermore, the developed polymer HTMs can also significantly enhance the photovoltaic and photodetection performance of perovskite devices from 15.1% to 22.7% and from 2.7 x 10(12) to 2.17 x 10(13)Jones with the same device structure, indicating their great application potential in the emerging optoelectronics.

    • For discrete element methods (DEMs), integrating the equation of motion based on Newton's second law is an integral part of the computation. Accelerations and velocities are involved even for modeling static mechanics problems. As a consequence, the accuracy can be ruined and numerous calculation steps are required to converge. In this study, we propose a static DEM based on discontinuous deformation analysis (DDA). The force of inertia is removed to develop a set of static equilibrium equations for distinct blocks. It inherits the advantages of DDA in dealing with distinct block system such as jointed rock structures. Furthermore, the critical numerical artifact in DDA, ie, artificial springs between contact blocks, is avoided. Accurate numerical solution can be achieved in mere one calculation step. Last but not the least, since the method is formulated in the framework of mathematical programming, the implementation can be easily conducted with standard and readily available solvers. Its accuracy and efficiency are verified against a series of benchmarks found in the literature.

    • In this paper, a new formulation of three-dimensional spherical discontinuous deformation analysis (DDA) based on second-order cone programming has been proposed. Artificial springs with open-close iteration used in classic DDA have been removed, given that improper stiffness parameters might cause numerical problems. Furthermore, to account for irregular granular shapes, a rolling resistance model is incorporated in the variational formulation. The proposed formulation can be cast into a standard second-order cone programming program, which can be solved using efficient off-the-shelf optimisation solvers. The proposed approach is validated by a series of numerical examples.

    • District heating (DH) has been considered as a resource- and cost-efficient way of supplying heat and a promising method to mitigate climate change, yet it also facing growing competition from alternative technical solutions, such as heat pumps. Many DH companies are under price model restructuring process to enhance their competitiveness. This study investigated the competitiveness of DH among users which would encounter significant cost increase during the price model restructuring process through comparing the cost of different DH price models with three alternative technical solutions. The result shows that for the invested DH user, instead of DH, the most economic preferable choice is to install ground source heat pump combining with direct electrical heating.

    • District heating companies are facing multiple challenges nowadays, including increased investment and maintenance cost and critique from customers regarding opaque price model. Hence there is an urgent need to develop new price models. In this paper, a survey was carried out and identified four basic components in the price model. Three price models that represent the current situation and future trend have been extracted from the survey as well. Based on those price models, investigation was performed to study the impacts of different components in price models on customers’ cost. The result shows that customers with flatter consumption profiles can benefit from the price model that has a higher share of load demand component and use consumers’ real-time consumption data for charging. On the contrary, when a price model that has a higher share of energy component is adopted, customers with flatter consumption profiles may experience an increase in the cost.

    • District heating companies have been adapting their price models to reflect the changes in production cost caused by penetration of renewable fuels, and promote the applications of energy conservation measures that benefit the system efficiency. One of the approaches is to introduce a peak demand component in the price model, which has been proved to be effective to benefit users with lower peak demand. Whereas, this approach also significantly increase the cost for users with high peak demand. One of the measures that could help with high peak demand is installing energy storage on the demand side. In order to understand how the energy storage could change the users’ cost and help DH users to make informed decision, this study analyses the economic benefits of demand-side heat storage, namely if installing low-investment, low-tech, short-term hot-water storage on demand side could effectively repress the cost increase caused by new price models. Five types of building are considered here: multifamily house, commercial building, hospital and social services, industrial building, and office and school. One user of each type, whose costs increased the most during the price model transition process have been included. The result shows that heat storage could efficiently repress the cost increase, and all the investments will be paid back within 3 years, which means introducing heat storage is an efficient measure for cost saving under the circumstances.

    • Traditional pricing scheme of district heating is based on previous experience of system operation. This strategy does not work well under the circumstances of decreasing demand and shifting consumption pattern. Therefore new pricing strategies are needed. To have a comprehensive view on existing price models in Sweden, a price model survey was carried out among all members of the district heating quality system REKO. Four basic price components and multiple variants of them are detected in the survey. The result also shows that most of the district heating companies still use traditional methods and do not consider their customers’ consumption pattern while charging them.

    • District heating (DH) is considered to be an efficient, environmentally friendly and cost-effective method for providing heat to buildings, since electricity is usually co-generated in biomass fuelled combined heat and power (CHP) plants. This gives it an important role in the mitigation of climate change. Swedish district heating companies are currently facing multiple challenges, and are in urgent need of new price models to increase transparency and maintain their competitiveness.This thesis describes a survey carried out to understand the structure of the present price models and subsequently proposes and compares two restructured price models with the most commonly used price model. This work also investigates the impact of restructured price models on users who would encounter a significant cost increase if the restructured price models were to be introduced. The district heating costs of different price models are compared with three alternative technical solutions.The results show that price models based on the consumption pattern of users can reflect district heating companies’ cost structure. Meanwhile, adopting a pricing strategy based on users’ consumption patterns increased the incentives to reduce the peak load.Consequently, users with high load factor (flat consecutive load curve) were able to reduce costs whereas users with low load factor (steep consecutive load curve) faced possible cost increases, when the load demand cost was changed to daily or hourly peak demand based methods. Further, the most economically preferable option for the invested district heating user was to combine district heating with direct electrical heating or with a ground source heat pump.

    • How can entrepreneurs protect their wellbeing during a crisis? Does engaging agility (namely, opportunity agility and planning agility) in response to adversity help entrepreneurs safeguard their wellbeing? Activated by adversity, agility may function as a specific resilience mechanism enabling positive adaption to crisis. We studied 3162 entrepreneurs from 20 countries during the COVID-19 pandemic and found that more severe national lockdowns enhanced firm-level adversity for entrepreneurs and diminished their wellbeing. Moreover, entrepreneurs who combined opportunity agility with planning agility experienced higher wellbeing but planning agility alone lowered wellbeing. Entrepreneur agility offers a new agentic perspective to research on entrepreneur wellbeing.
