    • Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data.

    • The Visual Object Tracking challenge VOT2018 is the sixth annual tracker benchmarking activity organized by the VOT initiative. Results of over eighty trackers are presented; many are state-of-the-art trackers published at major computer vision conferences or in journals in the recent years. The evaluation included the standard VOT and other popular methodologies for short-term tracking analysis and a “real-time” experiment simulating a situation where a tracker processes images as if provided by a continuously running sensor. A long-term tracking subchallenge has been introduced to the set of standard VOT sub-challenges. The new subchallenge focuses on long-term tracking properties, namely coping with target disappearance and reappearance. A new dataset has been compiled and a performance evaluation methodology that focuses on long-term tracking capabilities has been adopted. The VOT toolkit has been updated to support both standard short-term and the new long-term tracking subchallenges. Performance of the tested trackers typically by far exceeds standard baselines. The source code for most of the trackers is publicly available from the VOT page. The dataset, the evaluation kit and the results are publicly available at the challenge website (http://votchallenge.net).

    • Power transformer plays a vital role in maintaining reliable and efficient electricity supply. However, in service is increasing and growing older, much and much attention has been paid on their aging condition and life assessment than ever. The aging and degradation of transformer insulation will determine the life-time of its operation. The intensities of these depend on mechanical, thermal and environmental stresses that act on the insulations. Due to the temperature rise, the lifetime of insulation material will be shortened. The accelerated thermal aging method is often adopted to do some evaluation on the oil-paper insulation materials under different temperature for the thermal lifetime model. According to existing aging test, both insulation paper and mineral oil were aged at the same temperature. In this paper, the thermal aging experiments were implemented on the transformer main insulation model made with modified and unmodified insulation paper. The accelerated thermal aging tests were finished, and the superior electric characteristics of the nano-TiO2 and cellulose composition insulation were validated.

    • Low-grade gliomas (LGGs) are the most common childhood brain tumor in the general population and in individuals with the Neurofibromatosis type 1 (NF1) cancer predisposition syndrome. Surgical biopsy is rarely performed prior to treatment in the setting of NF1, resulting in a paucity of tumor genomic information. To define the molecular landscape of NF1-associated LGGs (NF1-LGG), we integrated clinical data, histological diagnoses, and multi-level genetic/genomic analyses on 70 individuals from 25 centers worldwide. Whereas, most tumors harbored bi-allelic NF1 inactivation as the only genetic abnormality, 11% had additional mutations. Moreover, tumors classified as non-pilocytic astrocytoma based on DNA methylation analysis were significantly more likely to harbor these additional mutations. The most common secondary alteration was FGFR1 mutation, which conferred an additional growth advantage in multiple complementary experimental murine Nf1 models. Taken together, this comprehensive characterization has important implications for the management of children with NF1-LGG, distinct from their sporadic counterparts.

  • Water and heat coupling processes and its simulation in frozen soils : Current status and future research directions

    • To date, most studies on coupled-water-and-heat processes in frozen soils haves focused on the mechanism of changes in frozen soil and the contribution of climate change, hydrological processes, and ecosystems in cold regions. Several studies have demonstrated considerable improvements in the accuracy of simulating water and heat transfer processes in cold regions. However, substantial differences remain among the different models and parameterizations because of the lack of observations and in-depth understanding of the water and heat transfer processes. Hence, it is necessary to summarize recent advances in the simulation of water-and-heat-coupling processes and challenges for further research. Therefore, we present a theory-focused summary of progress in this field considering the aspects of water flow and coupled-water-and-heat transfer. The simulation progress is discussed in terms of physical process models; one type of model only considers the heat conduction transfer processes without water flow, and the other considers coupled-water-and-heat transfer processes. Aspects of model deficiencies related to non-conductive heat transfer and soil water transfer processes in the frozen soil are also summarized. Moreover, the major parameterizations are reviewed, including phase changes, freeze–thaw fronts, thermal conductivity, hydraulic conductivity, snow processes, surface parameterization schemes, ground ice, and lower boundary conditions. While models and parameterizations can suitably capture local-scale water and heat transfer processes in frozen soil, their applications are spatiotemporally constrained, requiring further improvement. We provide a theoretical basis for further studying water and heat transfer processes in frozen soil and suggest that future research should enhance the accuracy of frozen soil parameterization and improve the understanding of the coupling of water and heat transfer processes based on improved observation techniques and high-resolution data.

  • Non-Blocking Testing for Network-on-Chip

    • To achieve high reliability in on-chip networks, it is necessary to test the network as frequently as possible to detect physical failures before they lead to system-level failures. A main obstacle is that the circuit under test has to be isolated, resulting in network cuts and packet blockage which limit the testing frequency. To address this issue, we propose a comprehensive network-level approach which could test multiple routers simultaneously at high speed without blocking or dropping packets. We first introduce a reconfigurable router architecture allowing the cores to keep their connections with the network while the routers are under test. A deadlock-free and highly adaptive routing algorithm is proposed to support reconfigurations for testing. In addition, a testing sequence is defined to allow testing multiple routers to avoid dropping of packets. A procedure is proposed to control the behavior of the affected packets during the transition of a router from the normal to the testing mode and vice versa. This approach neither interrupts the execution of applications nor has a significant impact on the execution time. Experiments with the PARSEC benchmarks on an 8x8 NoC-based chip multiprocessors show only 3 percent execution time increase with four routers simultaneously under test.

  • Optimization of Mass Fraction and Particle Size of TiO2 Additives in Application of HVDC Transformer Insulation

    • With the fast development of high voltage direct current (HVDC) transmission technology, higher electrical insulation properties are demanded in application of HVDC converter transformers. The nano adding in the cellulose insulation is a kind of effect method to enhance their electrical insulation properties. In this paper, nano-TiO2 particles with four different diameters (5 nm, 10 nm, 20 nm and 30 nm) and four different loading ratios (1 wt%, 3 wt%, 5 wt% and 7 wt%) are added during preparation of the insulation cellulose paper. To evaluate their dielectric properties, DC breakdown test, partial discharge inception voltage test, the accumulating properties of the surface charge dynamics and trap distribution characteristic are performed on the prepared nano-TiO2 modified paper samples. The experimental results show that the DC breakdown voltage and the partial discharge inception voltage of oil-immersed composition insulating paper are significantly improved by adding nano-TiO2 particles. According to the testing data, the optimum combination of 10 nm particle size and 5 wt% mass fractions were obtained, and the modification mechanism of nano-TiO2 of the nano-composition insulation were presented in the paper.

  • Storage, patterns, and environmental controls of soil organic carbon stocks in the permafrost regions of the Northern Hemisphere

    • Large stocks of soil organic carbon (SOC) accumulated in the Northern Hemisphere permafrost regions may be vulnerable to climatic warming, but global estimates of SOC distribution and magnitude in permafrost regions still have large uncertainties. Based on multiple high-resolution environmental variables and a compiled soil sample dataset (>3000 soil profiles), we used machine-learning methods to estimate the size and spatial distribution of SOC for the top 3 m soils in the Northern Hemisphere permafrost regions. We also identified key environmental predictors of SOC. The results showed that the SOC storage for the top 3 m soil was 1079 ± 174 Pg C across the Northern Hemisphere permafrost regions (20.8 × 106 km2), including 1057 ± 167 Pg C in the northern permafrost regions and 22 ± 7 Pg C in the Third Pole permafrost regions. The mean annual air temperature and NDVI are the main controlling factors for the spatial distribution of SOC stocks in the northern and the Third Pole permafrost regions. Our estimations were more accurate than the existing global SOC stock maps. The results improve our understanding of the regional and global permafrost carbon cycle and their feedback to the climate system.
