  • Chlamydia trachomatis antigens in enteroendocrine cells and macrophages of the small bowel in patients with severe irritable bowel syndrome

    • Background: Inflammation and immune activation have repeatedly been suggested as pathogentic factors in irritable bowel syndrome (IBS). The driving force for immune activation in IBS remains unknown. The aim of our study was to find out if the obligate intracellular pathogen Chlamydia could be involved in the pathogenesis of IBS. Methods: We studied 65 patients (61 females) with IBS and 42 (29 females) healthy controls in which IBS had been excluded. Full thickness biopsies from the jejunum and mucosa biopsies from the duodenum and the jejunum were stained with a monoclonal antibody to Chlamydia lipopolysaccharide (LPS) and species-specific monoclonal antibodies to C. trachomatis and C. pneumoniae. We used polyclonal antibodies to chromogranin A, CD68, CD11c, and CD117 to identify enteroendocrine cells, macrophages, dendritic, and mast cells, respectively. Results: Chlamydia LPS was present in 89% of patients with IBS, but in only 14% of healthy controls (p < 0.001) and 79% of LPS-positive biopsies were also positive for C. trachomatis major outer membrane protein (MOMP). Staining for C. pneumoniae was negative in both patients and controls. Chlamydia LPS was detected in enteroendocrine cells of the mucosa in 90% of positive biopsies and in subepithelial macrophages in 69% of biopsies. Biopsies taken at different time points in 19 patients revealed persistence of Chlamydia LPS up to 11 years. The odds ratio for the association of Chlamydia LPS with presence of IBS (43.1; 95% CI: 13.2-140.7) is much higher than any previously described pathogenetic marker in IBS. Conclusions: We found C. trachomatis antigens in enteroendocrine cells and macrophages in the small bowel mucosa of patients with IBS. Further studies are required to clarify if the presence of such antigens has a role in the pathogenesis of IBS.

    • Failure is ubiquitous in popular and consumer culture. In this commentary, we interrogate discourses around failure and outline potential avenues of inquiry for the marketing and consumption theory disciplines. We begin by synthesizing how failure has hitherto been conceptualized in marketing theory. Then, we discuss how recent rethinking of failure in other disciplines can be meaningful for marketing thought, and propose a new agenda for marketing scholars for studying failure, that moves beyond studying failure as a primarily destructive phenomenon that arises predominantly in service encounters.

  • Graphene oxide and adsorption of chloroform: A density functional study

    • Chlorinated hydrocarbon compounds are of environmental concerns, since they are toxic to humans and other mammals, and are widespread, and exposure is hard to avoid. Understanding and improving methods to reduce the amount of the substances are important. We present an atomic-scale calculational study of the adsorption of chlorine-based substance chloroform (CHCl3) on graphene oxide, as a step in estimating the capacity of graphene oxide for filtering out such substances, e.g., from drinking water. The calculations are based on density functional theory, and the recently developed consistent-exchange functional for the van der Waals density-functional method is employed. We obtain values of the chloroform adsorption energy varying from roughly 0.2 to 0.4 eV per molecule. This is comparable to previously found results for chloroform adsorbed directly on clean graphene, using similar calculations. In a wet environment, like filters for drinking water, the graphene will not stay clean and will likely oxidize, and thus adsorption onto graphene oxide, rather than clean graphene, is a more relevant process to study.

  • Norges Våpen : Cultural Memory and Uses of History in Norwegian Black Metal

    • This dissertation examines uses of history and expressions of cultural memory in Norwegian black metal. Formed in the late 1980s and early 1990s, Norwegian black metal seemed at odds with many of the stereotypes of Norway. The genre is an extreme style of heavy metal music that has been associated with burning churches, desecrating graves, and committing murders. Yet, Norway is often perceived as wealthy with sublime natural beauty and high levels of equality. Since the late 1990s, Norwegian black metal has increasingly received positive recognition and support from Norwegian government agencies and cultural institutions who have deemed this style of music a cultural product of Norway.In exploring the relationship between Norwegian black metal and Norway, two primary questions are asked: what makes Norwegian black metal ‘Norwegian’ and what are its influences? To answer these questions, a theoretical approach based on Astrid Erll’s cultural memory complex is used. Included in this cultural memory complex are notions of individual and collective memory, both of which include concepts of nationalism as outlined by Benedict Anderson and Michael Billig. The source base for this dissertation includes the musical releases of over five hundred Norwegian black metal bands which were gathered and analyzed. Three primary categories, with corresponding subcategories, were identified to account for the ways Norwegian black metal bands have used history and expressed cultural memory over a twenty-five-year period from 1988 to 2013.This dissertation shows that Norwegian black metal has made frequent use of history and has actively negotiated parts of the identity-making process from nineteenth-century Norway. In connecting to Norwegian identity in such a way, these bands link to historically construed notions of likhet and egalitarian individualism as identified by the Norwegian anthropologists Marianne Gullestad and Thomas Hylland Eriksen. They actively reproduce many of the same essentialized notions of Norwegian identity that create and maintain ethnic boundaries on Norwegian identity. By using history and expressing cultural memory in the way that they do, Norwegian black metal bands communicate that they are firmly Norwegian while, at the same time, reinforcing ethnocentric notions of Norwegian identity.

  • The SPECIES beamline at the MAX IV Laboratory : A facility for soft X-ray RIXS and APXPS

    • SPECIES is an undulator-based soft X-ray beamline that replaced the old I511 beamline at the MAX II storage ring. SPECIES is aimed at high-resolution ambient-pressure X-ray photoelectron spectroscopy (APXPS), near-edge X-ray absorption fine-structure (NEXAFS), X-ray emission spectroscopy (XES) and resonant inelastic X-ray scattering (RIXS) experiments. The beamline has two branches that use a common elliptically polarizing undulator and monochromator. The beam is switched between the two branches by changing the focusing optics after the monochromator. Both branches have separate exit slits, refocusing optics and dedicated permanent endstations. This allows very fast switching between two types of experiments and offers a unique combination of the surface-sensitive XPS and bulk-sensitive RIXS techniques both in UHV and at elevated ambient-pressure conditions on a single beamline. Another unique property of the beamline is that it reaches energies down to approximately 27 eV, which is not obtainable on other current APXPS beamlines. This allows, for instance, valence band studies under ambient-pressure conditions. In this article the main properties and performance of the beamline are presented, together with selected showcase experiments performed on the new setup.

  • Identification of proteins that specifically recognize and bind protofibrillar aggregates of amyloid-β

    • Protofibrils of the 42 amino acids long amyloid-β peptide are transient pre-fibrillar intermediates in the process of peptide aggregation into amyloid plaques and are thought to play a critical role in the pathology of Alzheimer's disease. Hence, there is a need for research reagents and potential diagnostic reagents for detection and imaging of such aggregates. Here we describe an in vitro selection of Affibody molecules that bind to protofibrils of Aβ42cc, which is a stable engineered mimic of wild type Aβ42 protofibrils. Several binders were identified that bind Aβ42cc protofibrils with low nanomolar affinities, and which also recognize wild type Aβ42 protofibrils. Dimeric head-to-tail fusion proteins with subnanomolar binding affinities, and very slow dissociation off-rates, were also constructed. A mapping of the chemical properties of the side chains onto the Affibody scaffold surface reveals three distinct adjacent surface areas of positively charged surface, nonpolar surface and a polar surface, which presumably match a corresponding surface epitope on the protofibrils. The results demonstrate that the engineered Aβ42cc is a suitable antigen for directed evolution of affinity reagents with specificity for wild type Aβ42 protofibrils.

  • Apex predator induces predator-rich ecosystem state innorthern lakes

    • Large predators are disappearing from ecosystems around the world. If predator speciesfacilitate each other’s existence through niche construction, this development could causecascading predator collapses and reduce ecosystem resilience. However, the importance offacilitation for the assembly and function of predator communities remains poorly understood.Here, we show that a large piscivorous fish, the northern pike (Esox lucius), enables theformation of a numerous and diverse predator community by inducing a dwarf ecotype ofEuropean whitefish (Coregonus lavaretus). Pike increases the standing biomass of prey-sized(<200mm) whitefish with a factor of 12.6, allowing small-gaped fish species (Percafluviatilis, Lota lota, Salvelinus alpinus and Salmo trutta) to go from small-growinggeneralists, to large-growing piscivores. Similarly, a guild of piscivorous birds (Gavia arctica,G. stellata, Sterna paradisaea, S. hirundo, Mergus serrator and M. merganser) shift from amixed diet to relying mainly on whitefish prey in presence of pike. Through this regime shift,the functional piscivore biomass in the non-pike fish community increases with a factor of14.2, and the density- and species richness of piscivorous birds increase with factors of 2.08and 2.16, respectively. Our results demonstrate how feedbacks between presence/absence ofimportant predators and the phenotype of prey may keep complex ecosystems in predator-richor predator-depleted states.
