  • FACT : An Open-Label Randomized Phase III Study of Fulvestrant and Anastrozole in Combination Compared With Anastrozole Alone as First-Line Therapy for Patients With Receptor-Positive Postmenopausal Breast Cancer

    • Purpose: To compare the effect of therapy with anastrozole versus a combination of fulvestrant and anastrozole in women in first relapse of endocrine-responsive breast cancer. Patients and Methods: Postmenopausal women, or premenopausal women receiving a gonadotropin-releasing hormone agonist, with estrogen receptor– and/or progesterone receptor–positive disease at first relapse after primary treatment of localized disease were open-label randomly assigned to a fulvestrant loading dose (LD) regimen followed by monthly injection plus 1 mg of anastrozole daily or to 1 mg of anastrozole daily alone. The primary end point was time to progression (TTP). Results: In all, 514 women were randomly assigned to fulvestrant plus anastrozole (experimental arm; n = 258) or anastrozole (standard arm; n = 256). Approximately two thirds had received adjuvant antiestrogens, but only eight individuals had received an aromatase inhibitor. Median TTP was 10.8 and 10.2 months in the experimental versus standard arm, respectively (hazard ratio [HR] = 0.99; 95% CI, 0.81 to 1.20; P = .91); median overall survival was 37.8 and 38.2 months, respectively (HR = 1.0; 95% CI, 0.76 to 1.32; P = 1.00). Incidences of prespecified adverse events (AEs) were similar. Hot flashes were more common in the experimental arm: 63 patients (24.6%) versus 35 patients (13.8%) in the standard arm (P = .0023). Death owing to AEs was reported in 11 (4.3%) and five patients (2.0%) in the experimental versus standard arm, respectively. Conclusion: Fulvestrant (250 mg + LD regimen) in combination with anastrozole offered no clinical efficacy advantage over anastrozole monotherapy in this population of individuals with a relatively high proportion of previous adjuvant antiestrogen exposure.

    • Viruses in the family Retroviridae are found in a wide variety of vertebrate hosts. Enveloped virions are 80-100 nm in diameter with an inner core containing the viral genome and replicative enzymes. Core morphology is often characteristic for viruses within the same genus. Replication involves reverse transcription and integration into host cell DNA, resulting in a provirus. Integration into germline cells can result in a heritable provirus known as an endogenous retrovirus. This is a summary of the International Committee on Taxonomy of Viruses (ICTV) Report on the family Retroviridae, which is available at ictv.global/ report/retroviridae.

    • As computational Grids move away from the prototyping state, reliability, performance and ease of use and maintenance become focus areas of their adoption. In this paper, we describe ARC (Advanced Resource Connector) Grid middleware, where these issues have been given special consideration. We present an in-depth view of the existing components of ARC, and discuss some of the new components, functionalities and enhancements currently under development. This paper also describes architectural and technical choices that have been made to ensure scalability, stability and high performance. The core components of ARC have already been thoroughly tested in demanding production environments, where it has been in use since 2002. The main goal of this paper is to provide a first comprehensive description of ARC.

    • To reach its large computing needs, the ALICE experiment at CERN has developed its own middleware called AliEn, centralised and relying on pilot jobs. One of its strength is the automatic installation of the required packages. The Nordic countries have offered a distributed Tier-1 centre for the CERN experiments, where the job management should be done with the NorduGrid middleware ARC. We have developed an interoperation module to allow to unify several computing sites using ARC, and make them look like a single site from the point of view of AliEn. A prototype has been completed and tested out of production. This talk will present implementation details of the system and its performance in tests.

    • he paper discusses how courses in mechanics can be taught to architectural students in such a way that concepts are presented to them in a manner making mechanics an inspiration for the designing process rather than simply imposing limitations on it. In courses of this sort that have been held, emphasis has been placed on the use of software to facilitate an intuitive understanding of physical relationships in the area of mechanics, as well as on transforming such an understanding into design sketches. ForcePAD, a readily comprehensible software package of this sort developed for making schematic 2D sketches and investigating different patterns in the area of mechanics, is presented. Originally, it was developed as a tool for providing a basic introduction to mechanics for students of civil engineering. The package aims at enhancing an understanding of such factors as balance, weight, stability, rest and movement, support forces, stress fields, and deformation. The paper is based on experience with classes taught both at Lund University and at Chalmers University in Gothenburg that have made use of this package (Olsson, 2003). The types of weekly tasks students have been given, as well as how the educational conceptions utilized in these courses have influenced the design of the ForcePad software, are discussed.

    • Different methods of real-time fibre-network visualisation have been studied. Using an extrusion-based method yields very good results, but for large networks the frame rate becomes unacceptably low. To increase the number of fibres that can be visualised in real time, a textured billboard method has been implemented. With this method, an average performance gain of 60% has been achieved, using an OpenGL implementation.

    • To meet the ever increasing demand for computational speed and use of ever larger datasets, multi GPU installations look very tempting. Lunarc and the Theoretical Astrophysics group at Lund Observatory collaborate on a pilot project to evaluate and utilize multi-GPU architectures for scientific calculations. Starting with a small workshop in 2009, continued investigations eventually lead to the procurement of the GPU-resource Timaeus, which is a four-node eight-GPU cluster with two Nvidia m2050 GPU-cards per node. The resource is housed within the larger cluster Platon and share disk-, network-and system resources with that cluster. The inauguration of Timaeus coincided with the meeting "Computational Physics with GPUs" in November 2010, hosted by the Theoretical Astrophysics group at Lund Observatory. The meeting comprised of a two-day workshop on GPU-computing and a two-day science meeting on using GPUs as a tool for computational physics research, with a particular focus on astrophysics and computational biology. Today Timaeus is used by research groups from Lund, Stockholm and Lule in fields ranging from Astrophysics to Molecular Chemistry. We are investigating the use of GPUs with commercial software packages and user supplied MPI-enabled codes. Looking ahead, Lunarc will be installing a new cluster during the summer of 2011 which will have a small number of GPU-enabled nodes that will enable us to continue working with the combination of parallel codes and GPU-computing. It is clear that the combination of GPUs/CPUs is becoming an important part of high performance computing and here we will describe what has been done at Lunarc regarding GPU-computations and how we will continue to investigate the new and coming multi-GPU servers and how they can be utilized in our environment.

    • This talk describes the various stages of ATLAS Data Challenge 2 (DC2) in what concerns usage of resources deployed via NorduGrid's Advanced Resource Connector (ARC). It also describes the integration of these resources with the ATLAS production system using the Dulcinea executor. ATLAS Data Challenge 2 (DC2), run in 2004, was designed to be a step forward in the distributed data processing. In particular, much coordination of task assignment to resources was planned to be delegated to Grid in its different flavours. An automatic production management system was designed, to direct the tasks to Grids and conventional resources. The Dulcinea executor is a part of this system that provides interface to the information system and resource brokering capabilities of the ARC middleware. The executor translates the job definitions recieved from the supervisor to the extended resource specification language (XRSL) used by the ARC middleware. It also takes advantage of the ARC middleware's built-in support for the Globus Replica Location Server (RLS) for file registration and lookup. NorduGrid's ARC has been deployed on many ATLAS-dedicated resources across the world in order to enable effective participation in ATLAS DC2. This was the first attempt to harness large amounts of strongly heterogeneous resources in various countries for a single collaborative exercise using Grid tools. This talk addresses various issues that arose during different stages of DC2 in this environment: preparation, such as ATLAS software installation; deployment of the middleware; and processing. The results and lessons are summarized as well.

    • In the conceptual design phase, solutions are reached through an iterative, high-paced and often chaotic manner. Conventional advanced structural analysis software is often too advanced and insufficiently agile to follow this high-paced work pattern. Premature use of advanced structural analysis tools can negatively affect conceptual understanding and the quality of the conceptual design. The multi-touch interfaces of today's tablet computers give the user a strong feeling of direct manipulation of objects on the screen. This is interesting for structural mechanics applications, enabling direct manipulation of the structural model on the screen so as to have a better understanding and feeling of the structural behaviour. A tablet computer application for the conceptual design phase, which uses this type of direct manipulation interface, has been developed.
