    • This article discusses the so-called verb-oriented method and its role in the research project Gender and Work in early modern Sweden (GaW), which is based at Uppsala University. It provides a presentation of the GaW-database, which has been designed to allow analysis according to the verb-method. Finally, the article points out that this method can be combined with a number of different theoretical approaches as long as the focus is on practices. It is therefore compatible with the approaches of e.g., Judith Butler, Michel de Certeau, and Amartya Sen. Work is defined as "time-use with the purpose of making a living" and the article discusses why data on time-use, or actual work activities, are better suited for research into early modern Swedish working life than other types of data. It shows that activities are usually described in the sources by verb-phrases, and explains how and from what sources verb-phrases are collected and analyzed within the project. In order to allow for generalizations the verb-method presupposes large amounts of data. This is the rationale for the GaW-database, which at present includes around 5000 verb-phrases and 75000 data posts.

  • EPIGENETIC ALTERATIONS IN IBD : DEFINING GEOGRAPHICAL, GENETIC, AND IMMUNEIN-FLAMMATORY INFLUENCES ON THE CIRCULATING METHYLOME

    • Introduction: DNA methylation may provide critical insights into gene-environment interactions in inflammatory bowel disease (IBD).Methods: Using the multi-centre IBD Character inception cohort (295 controls, 154 CD, 161 UC, 28 IBD-U), epigenome-wide methylation was profiled using Illumina HumanMethylation450 platform. Differentially methylated position analysis was performed using age, sex and cell proportions as covariates. Integration of paired genomic and transcriptomic layers was done with Multi-Omics Factor Analysis v2 (MOFA). Unsupervised principal component analyses were performed to examine correlates of treatment escalation and clinical predictors of disease severity.Results: We report 137 differentially methylated positions (DMP) in whole blood in IBD, including VMP1/MIR21 (p=9.11×10-15) and RPS6KA2 (6.43×10-13); with consistency seen across Scandinavia and UK. Cell of origin analysis preferentially implicated the monocyte lineage. Dysregulated loci demonstrate strong genetic influence, notably VMP1 (p=1.53×10-15). Age acceleration is seen in IBD (coefficient 0.94, p<2.2x10-16). Several immuno-active genes demonstrated highly significant correlations between methylation and gene expression in IBD, in particular OSM: IBD r -0.32, p 3.64×10-7 vs. non-IBD r -0.14, p=0.77). Multi-omic integration of methylome, genome and transcriptome also identified specific pathways that associate with immune activation, response and regulation at disease inception. At follow up, a signature of 3 DMPs (TAP1, TESPA1, RPTOR) associated with treatment escalation to biological agents or surgery (hazard ratio of 5.19 (CI:2.14-12.56, logrank p=9.70×10-4).Conclusion: This study highlights the stability of the IBD-specific circulating methylome across regions with shared ancestry. Through integrative multi-omic analyses we identify key pro-inflammatory genes that are upregulated in IBD at inception. Furthermore, differential methylation within certain genes such as TAP1 associate with disease course over time.

    • AIM: To assess the pathobiological and translational importance of whole blood transcriptomic analysis in inflammatory bowel disease (IBD).METHODS: We analyzed whole blood expression profiles from paired-end sequencing in a discovery cohort of 590 Europeans recruited across six countries in the IBD Character initiative (newly diagnosed patients with Crohn's disease [CD, n = 156], ulcerative colitis [UC, n = 167], and controls [n = 267]), exploring differential expression (DESeq2), co-expression networks (WGCNA), and transcription factor involvement (EPEE, ChEA, DoRothEA). Findings were validated by analysis of an independent replication cohort (99 CD, 100 UC, and 95 controls). In the discovery cohort, we also defined baseline expression correlates of future treatment escalation using cross-validated elastic-net and random forest modelling, along with a pragmatic ratio detection procedure.RESULTS: Disease-specific transcriptomes were defined in IBD (8697 transcripts), CD (7152), and UC (8521), with the most highly significant changes in single genes, including CD177 (log2-fold change [LFC] = 4.63, p = 4.05 × 10 -118), MCEMP1 (LFC = 2.45, p = 7.37 × 10 -109), and S100A12 (LFC = 2.31, p = 2.15 × 10 -93). Significantly over-represented pathways included IL-1 (p = 1.58 × 10 -11), IL-4, and IL-13 (p = 8.96 × 10 -9). Highly concordant results were obtained using multiple regulatory activity inference tools applied to the discovery and replication cohorts. These analyses demonstrated central roles in IBD for the transcription factors NFE2, SPI1 (PU.1), CEBPB, and IRF2, all regulators of cytokine signaling, based on a consistent signal across cohorts and transcription factor ranking methods. A number of simple transcriptome-based models were associated with the need for treatment escalation, including the binary CLEC5A/CDH2 expression ratio in UC (hazard ratio = 23.4, 95% CI 5.3-102.0).CONCLUSION: Transcriptomic analysis has allowed for a detailed characterization of IBD pathobiology, with important potential translational implications.

  • Mucosal Gene Transcript Signatures in Treatment Naïve Inflammatory Bowel Disease : A Comparative Analysis of Disease to Symptomatic and Healthy Controls in the European IBD-Character Cohort

    • Background: Studies of the mucosal transcriptomic landscape have given new insight into the pathogenesis of inflammatory bowel disease (IBD). Recently, the predictive biomarker potential of gene expression signatures has been explored. To further investigate the mucosal gene expression in IBD, we recruited a cohort of treatment naïve patients and compared them to both symptomatic and healthy controls.Methods: Altogether, 323 subjects were included: Crohn's disease (N = 75), ulcerative colitis (N = 87) and IBD unclassified (N = 3). Additionally, there were two control groups: symptomatic controls (N = 131) and healthy controls (N = 27). Mucosal biopsies were collected during ileocolonoscopy and gene expression in inflamed and non-inflamed mucosa was explored. Gene expression profiling was performed using Agilent G3 Human Gene Expression 860K v3 One-Color microarray. We recorded information about treatment escalation to anti-TNF agents or surgery, and anti-TNF response, to explore predictive opportunities of the mucosal transcriptome.Results: Gene expression profiles in symptomatic controls in whom IBD had been excluded resembled that of IBD patients and diverged from that of healthy controls. In non-inflamed Crohn's disease and ulcerative colitis, gene set enrichment analysis revealed dysregulation of pathways involved in basic cellular biological processes. Mitochondria-associated pathways were dysregulated both in non-inflamed and inflamed Crohn's disease and ulcerative colitis (>2.6 normalized enrichment scores <-1.8). Gene expression signatures of Crohn's disease and ulcerative colitis did not predict time for treatment escalation (p = 0.175). No significant association was found between gene expression signatures and anti-TNF response.Conclusion: Non-inflamed samples are probably superior to inflamed samples when exploring gene expression signatures in IBD and might reveal underlying mechanisms central for disease initiation. The gene expression signatures of the control groups were related to if they were symptomatic or not, which may have important implications for future study designs.

    • The efficacy of serotonin reuptake inhibitors in depression and anxiety disorders suggests the gene coding for the serotonin transporter (5-HTT), SLC6A4, as a candidate of importance for these conditions. Positive findings regarding associations between polymorphisms in SLC6A4 have been reported, indicating that these polymorphisms may influence anxiety-related personality traits, as well as the risk of developing depression and suicidality. Serotonin 5-HTT availability was assessed with single photon emission computed tomography (SPECT), using I-123-beta-CIT as ligand, in a population of unmedicated male suicide attempters (n=9) and in matched controls (n=9). Two polymorphisms in SLC6A4 were assessed, including the 5-HTTLPR located in the promoter region and a variable number of tandem repeats (VNTR) polymorphism in intron 2 (STin2). In suicide attempters, but not in controls, low 5-HTT availability was associated with the S allele of 5-HTTLPR and with the 12 repeat allele of STin2. Data suggest that polymorphisms in SLC6A4 may influence the expression of the brain serotonin transporter in suicide attempters.

    • BACKGROUND: Ketamine has emerged as a fast-acting and powerful antidepressant, but no head to head trial has been performed, Here, ketamine is compared with electroconvulsive therapy (ECT), the most effective therapy for depression.METHODS: Hospitalized patients with unipolar depression were randomized (1:1) to thrice-weekly racemic ketamine (0.5 mg/kg) infusions or ECT in a parallel, open-label, non-inferiority study. The primary outcome was remission (Montgomery Åsberg Depression Rating Scale score ≤10). Secondary outcomes included adverse events (AEs), time to remission, and relapse. Treatment sessions (maximum of 12) were administered until remission or maximal effect was achieved. Remitters were followed for 12 months after the final treatment session.RESULTS: In total 186 inpatients were included and received treatment. Among patients receiving ECT, 63% remitted compared with 46% receiving ketamine infusions (P = .026; difference 95% CI 2%, 30%). Both ketamine and ECT required a median of 6 treatment sessions to induce remission. Distinct AEs were associated with each treatment. Serious and long-lasting AEs, including cases of persisting amnesia, were more common with ECT, while treatment-emergent AEs led to more dropouts in the ketamine group. Among remitters, 70% and 63%, with 57 and 61 median days in remission, relapsed within 12 months in the ketamine and ECT groups, respectively (P = .52).CONCLUSION: Remission and cumulative symptom reduction following multiple racemic ketamine infusions in severely ill patients (age 18-85 years) in an authentic clinical setting suggest that ketamine, despite being inferior to ECT, can be a safe and valuable tool in treating unipolar depression.

    • While gender relations in the post-1800 labour market have been subject to much scholarly scrutiny, less is known about how women and men supported themselves in the early modern period. This article discusses the reasons for this lacuna and explores various approaches that could be used to increase knowledge in the field. It describes the verb-oriented method, used by the Gender and Work project at Uppsala University and inspired by the work of Sheilagh Ogilvie. Understanding ‘work’ as ‘use of time with the goal of making a living’, the method consists in systematic collection of verb phrases such as ‘to fish herring’, ‘to sell clothes’, etc. The article also presents the database GaW, which is designed so as to make possible systematic analysis of large sets of verb phrases.

    • Migration is a fundamental but often neglected chapter in the history of European peasant societies. Knowledge of migration in Medieval and Early Modern rural Europe adds significantly to our understanding of phenomena as diverse as serfdom, the land market, social stratification, the diffusion of agricultural practices, and responses to climate change. To advance this knowledge, we need to consult both documentary evidence and archaeological data, and study diverse parts of Europe. This special issue of the Journal of Migration History collects articles devoted to various aspects of mobility among peasants and its multiple meanings. Written by historians and archaeologists, they present a plethora of perspectives, methods, and approaches to the study of migration in the past. This introductory article describes the themes and scope of the articles. We argue for the importance of migration to peasant studies but also for the integration of peasant studies into mainstream migration history.

  • Distribution and Differences : Stratification and the System of Reproduction in a Swedish Peasant Community 1620-1820

    • This dissertation examines the character, conditions and change of peasant stratification in early modern Sweden. Wherever and whenever one looks, one finds that resources were unevenly spread among peasant households. In the literature, there are different, and conflicting, views compatible with this finding. In order to explain its character, this study places peasant stratification into a broader system of resource reproduction. Resource holding, families, and individuals are studied over time.The study is based on an extended family reconstitution comprising the landholding peasants in the Mid-Swedish parish of Björskog between 1620 and 1820. Data has been compiled from cadastres, poll tax registers, parish registers, court records, and maps, and has then been related to the information on resource holding as given by tax lists and probate inventories.Six elements and three general principles are identified as fundamental to systems of resource reproduction among peasants. Starting from these, the book argues that the resource holding of a Swedish peasant household was relatively independent of family demography; that wealthy peasants were able to retain large surpluses even during the period of high rent pressure in the seventeenth century; that the reproduction of poorer peasant households was imperfect whereas the reproduction of wealthier households was extended; that wealthy peasants dominated the local community; that economic inequalities within the class of landed peasants did not increase during the seventeenth and eighteenth centuries; that land accumulation and cyclical mobility became important features of peasant society, but not until the decades around 1800: and that the peasant community was characterized by a large degree of geographical and downward social mobility.

    • This article uses results from a large historical study on working practices to argue that a marked spatial and gendered separation of home and work is not a historical constant. Based on data extracted from court records from the Swedish city of Västerås and its rural surroundings in the period 1720–1881, the authors show that both women and men carried out a considerable share of their work in homes rather than in dedicated workplaces. Work in homes was multi-faceted rather than specialized, and could be both paid and unpaid. Women were more often observed working in their own homes but did also provide domestic services against payment (washing, cleaning) in other people’s homes. Men tended to work more often in other people’s homes, for instance, as carpenters, painters and in other crafts.Work away from home usually happened in public places: streets, squares, fields and woods. Some forms of work were particularly likely to occur in such sites, for example trade, transport, agriculture and forestry. Men were conspicuous in places like harbors and customs gates, places that functioned as hubs in economic life. Work caused both women and men to travel away from home, but men travelled further afield, and this gendered difference became more important towards the end of the nineteenth century. This was also when more men started to work in dedicated workplaces such as factories.There are many similarities between eighteenth- and nineteenth-century society and the society we live in today. In both cases, women and men often worked/work in spatial proximity. In both cases, the distinction between home and work was/is far from sharp and not stably linked to gender.

    • The history of the labouring family has gained increasing attention among European scholars in the last decades, but in Sweden, it remains an under-researched topic. Still, in the early modern period, labourers and their families made vital contributions to the country’s important mining industry. This paper examines the household economy of labouring families related to the mining industry in two Swedish areas in the late seventeenth century. On the basis on account books and court records, combined with demographic data, we explore the diversity of livelihoods and the complex web of interdependencies that made this economy feasible. We show that, while monetary remuneration was limited, wage labour in mines and metalwork gave the labouring family access to resources in the form of land, labour and credit beyond its own assets. Within the household, the man generally worked for wages, while his wife made use of the use-rights that came with his employment. The mining industry, and thereby also Swedish state finances, depended on this diverse family economy. In conclusion, interdependence, rather than the independent economic position described by classical models of early modern households, characterized the household economy of the labouring family.

    • We know very little about the unlanded population (obesuttna) in Sweden before 1750. Still, knowledge of this group is vital not only to our understanding of the proletarianization proces, but also to the understanding of rural Sweden before the changes of the 18th and 19th centuries. This article takes a step towards filling this gap. Tax registers and church records from 17th-century Västmanland shows substantial proportions of unlanded people, i.e. crofters, cottagers, soldiers, artisans, and lodgers, around 1640 and 1690. The age structure of this group, which can be studied for the latter part of the period, shows that it mostly consisted of people in working age. In conclusion, the article argues that there was a significant group of smallholders and wage labourers in 17th-century Sweden, comparable to the labouring poor of other European countries.

    • Based on the verb-oriented method and a unique collection of observations from court records, this article shows that both men and women did almost all categories of work in early modern Sweden. On the level of concrete tasks, however, there was both difference and similarity between the genders. Marital status exerted a strong influence on women’s sustenance activities, creating a clear distinction between unmarried and ever-married. These patterns were probably the effect of a labour legislation that forced young people without independent means to offer their bodies and time to masters and mistresses.

    • Questions of peasant mobility, household formation and the family-land bond all relate to the same issue: the system of peasantreproduction, or the replacement of households on a specific farm. The aim of this paper is to contribute to the description and understanding of such a system, using empirical evidence from early modern Sweden.In the paper the influence on geographic mobility and household formation by the following factors is discussed: terms of tenure, the land/labour ratio, market dependence, kinship relations, gender and wealth.The study is based on a data set including about 2,000 families living in a mid-Swedish parish (Björskog in the county of Västmanland) from year 1620 to 1820. Parish registers, tax rolls and court records are used to combine demographic information with data on land-holding, kinship relations and wealth.

    • Starting from a singular case about a late seventeenth-century maid, who disobeyed her mistress and took her to court, this article discusses the place of microhistory in the wider eld of historical research. We argue that microhistory has the potential to make invaluable contributions to our understanding of more general phenomena. This presupposes that the study of a single case is put into a larger context established by previous research. By doing this, we open up the possibility of a dialogue between previous research and the case, which in turn makes possible for the results of the microhistorian to in uence the construction of grand narratives.In our example, the actions of the maid are related to certain propositions in Swedish historical research about patriarchal society. We nd that these propositions explain the maid’s actions inadequately. Instead, we suggest an alternative explanation and reformulate the general understanding of seventeenth-century Swedish patriarchal society on this basis. Patriarchy was a culture, rather than merely an ideology and it o ered a repertoire of arguments that people in both subordinate and superior positions could use. Fathers of di erent kinds, such as household heads, priests, bishops and judges, maintained patriarchal order through a dynamic interplay in which the participation of people in subordinate positions, such as maids, were crucial. Whether this new understanding of the general context is valid or not, or if we are dealing with the exception to a rule, is not for this study but for future research to determine.The kind of microhistory that we advocate is not about “bringing history to life” or “giving it a human face”, nor is it only about falsifying general theories. This would be to underestimate the potential of microhistory. Instead, our microhistory seeks to explain the particular in such a way that it has e ect on how we understand the universal. Instead of reducing a single case to an instance of something di erent, we argue that the case should be used to understand the big picture in a new way. In this way, microhistory is able to make independent contributions to historical research and, in the end, to promote more accurate grand narratives.

    • Based on a study of a Swedish parish from the seventeenth century to the early 1800s, this article discusses the role of migration in the peasant economy. It argues that migration was a fundamental feature of the reproduction process among peasants, and that the mobility of some enabled the unchanging positions of others. Migration was a solution to the dilemma created by the dual features of peasant economies: the equivalence of the household and the production unit and the obligation to pay rent. Mobility created adaptability and resilience in peasant communities, but its selective character speaks to its link to wealth distribution, class relations and the struggle for resources.
