  • Deep Learning-Based Classification of Cancer Cell in Leptomeningeal Metastasis on Cytomorphologic Features of Cerebrospinal Fluid

    • Background: It is a critical challenge to diagnose leptomeningeal metastasis (LM), given its technical difficulty and the lack of typical symptoms. The existing gold standard of diagnosing LM is to use positive cerebrospinal fluid (CSF) cytology, which consumes significantly more time to classify cells under a microscope.Objective: This study aims to establish a deep learning model to classify cancer cells in CSF, thus facilitating doctors to achieve an accurate and fast diagnosis of LM in an early stage.Method: The cerebrospinal fluid laboratory of Xijing Hospital provides 53,255 cells from 90 LM patients in the research. We used two deep convolutional neural networks (CNN) models to classify cells in the CSF. A five-way cell classification model (CNN1) consists of lymphocytes, monocytes, neutrophils, erythrocytes, and cancer cells. A four-way cancer cell classification model (CNN2) consists of lung cancer cells, gastric cancer cells, breast cancer cells, and pancreatic cancer cells. Here, the CNN models were constructed by Resnet-inception-V2. We evaluated the performance of the proposed models on two external datasets and compared them with the results from 42 doctors of various levels of experience in the human-machine tests. Furthermore, we develop a computer-aided diagnosis (CAD) software to generate cytology diagnosis reports in the research rapidly.Results: With respect to the validation set, the mean average precision (mAP) of CNN1 is over 95% and that of CNN2 is close to 80%. Hence, the proposed deep learning model effectively classifies cells in CSF to facilitate the screening of cancer cells. In the human-machine tests, the accuracy of CNN1 is similar to the results from experts, with higher accuracy than doctors in other levels. Moreover, the overall accuracy of CNN2 is 10% higher than that of experts, with a time consumption of only one-third of that consumed by an expert. Using the CAD software saves 90% working time of cytologists.Conclusion: A deep learning method has been developed to assist the LM diagnosis with high accuracy and low time consumption effectively. Thanks to labeled data and step-by-step training, our proposed method can successfully classify cancer cells in the CSF to assist LM diagnosis early. In addition, this unique research can predict cancer’s primary source of LM, which relies on cytomorphologic features without immunohistochemistry. Our results show that deep learning can be widely used in medical images to classify cerebrospinal fluid cells. For complex cancer classification tasks, the accuracy of the proposed method is significantly higher than that of specialist doctors, and its performance is better than that of junior doctors and interns. The application of CNNs and CAD software may ultimately aid in expediting the diagnosis and overcoming the shortage of experienced cytologists, thereby facilitating earlier treatment and improving the prognosis of LM.

    • The Visual Object Tracking Segmentation VOTS2023 challenge is the eleventh annual tracker benchmarking activity of the VOT initiative. This challenge is the first to merge short-term and long-term as well as single-target and multiple-target tracking with segmentation masks as the only target location specification. A new dataset was created; the ground truth has been withheld to prevent overfitting. New performance measures and evaluation protocols have been created along with a new toolkit and an evaluation server. Results of the presented 47 trackers indicate that modern tracking frameworks are well-suited to deal with convergence of short-term and long-term tracking and that multiple and single target tracking can be considered a single problem. A leaderboard, with participating trackers details, the source code, the datasets, and the evaluation kit are publicly available at the challenge website1

  • Heteroheptacene-based acceptors with thieno[3,2-b]pyrrole yield high-performance polymer solar cells

    • Rationally utilizing and developing synthetic units is of particular significance for the design of high-performance non-fullerene small-molecule acceptors (SMAs). Here, a thieno[3,2-b]pyrrole synthetic unit was employed to develop a set of SMAs (ThPy1, ThPy2, ThPy3 and ThPy4) by changing the number or the position of the pyrrole ring in the central core based on a standard SMA of IT-4Cl, compared to which the four thieno[3,2-b]pyrrole-based acceptors exhibit bathochromic absorption and upshifted frontier orbital energy level due to the strong electron-donating ability of pyrrole. As a result, the polymer solar cells (PSCs) of the four thieno[3,2-b]pyrrole-based acceptors yield higher open-circuit voltage and lower energy loss relative to those of the IT-4Cl-based device. What is more, the ThPy3-based device achieves a power conversion efficiency (PCE) (15.3%) and an outstanding fill factor (FF) (0.771) that are superior to the IT-4Cl-based device (PCE = 12.6%, FF = 0.758). The ThPy4-based device realizes the lowest energy loss and the smallest optical band gap, and the ternary PSC device based on PM6:BTP-eC9:ThPy4 exhibits a PCE of 18.43% and a FF of 0.802. Overall, this work sheds light on the great potential of thieno[3,2-b]pyrrole-based SMAs in realizing low energy loss and high PCE. Four heteroheptacene-based acceptors using thieno[3,2-b]pyrrole building block were developed for the first time, and all the four acceptors-based devices realized high performance and low energy loss.

  • Native Conditional iSWAP Operation with Superconducting Artificial Atoms

    • Controlling the flow of quantum information is a fundamental task for quantum computers, which is unfeasible to realize on classical devices. Coherent devices, which can process quantum states are thus required to route the quantum states that encode information. In this paper we demonstrate experimentally the smallest quantum transistor with a superconducting quantum processor, which is composed of a collector qubit, an emitter qubit, and a coupler (transistor gate). The interaction strength between the collector and emitter qubits is controlled by the frequency and state of the coupler, effectively implementing a quantum switch. Through the coupler-state-dependent Heisenberg (inherent) interaction between the qubits, a single-step (native) conditional iSWAP operation can be applied. To this end, we find that it is useful to take into consideration the higher-energy level for achieving a native and high-fidelity transistor operation. By reconstructing the quantum process tomography, we obtain an operation fidelity of 92.36% when the transistor gate is open (iSWAP implementation) and 95.23% in the case of closed gate (identity gate implementation). The architecture has strong potential in quantum information processing applications with superconducting qubits.

  • Optimal charging of a superconducting quantum battery

    • Quantum batteries are miniature energy storage devices and play a very important role in quantum thermodynamics. In recent years, quantum batteries have been extensively studied, but limited in theoretical level. Here we report the experimental realization of a quantum battery based on superconducting qutrit. Our model explores dark and bright states to achieve stable and powerful charging processes, respectively. Our scheme makes use of the quantum adiabatic brachistochrone, which allows us to speed up the battery ergotropy injection. Due to the inherent interaction of the system with its surrounding, the battery exhibits a self-discharge, which is shown to be described by a supercapacitor-like self-discharging mechanism. Our results paves the way for proposals of new superconducting circuits able to store extractable work for further usage.

Moreover, P resorption strongly decreased, and nitrogen:phosphorus and carbon:phosphorus resorption ratios increased with soil P content. This demonstrates a strong link between foliar P resorption and P content in soils, and emphasizes the importance of P resorption in leaves of trees growing in soils with contrasted P content. Context: Subtropical ecosystems are generally characterized by P-deficient soils. However, P-rich soils develop in phosphate rock areas. Aims: We compared the patterns of nutrient resorption, in terms of ecological stoichiometry, for two sites naturally varying in soil P content. Methods: The resorption efficiency (percentage of a nutrient recovered from senescing leaves) and proficiency (level to which nutrient concentration is reduced in senesced leaves) of 12 elements were determined in two oak (Q. variabilis) populations growing at a P-rich or a P-deficient site in subtropical China. Results: P resorption efficiency dominated the intraspecific variation in nutrient resorption between the two sites. Q. variabilis exhibited a low P resorption at the P-rich site and a high P resorption at the P-deficient site. Both P resorption efficiency and proficiency strongly decreased with soil P content only and were positively related to the N:P and C:P ratios in green and senesced leaves. Moreover, resorption efficiency ratios of both N:P and C:P were positively associated with soil P. Conclusion: These results revealed a strong link between P resorption and P stoichiometry in response to a P deficiency in the soil, and a single- and limiting-element control pattern of P resorption. Hence, these results provide new insights into the role of P resorption in plant adaptations to geologic variations of P in the subtropics.

  • Local Structure and Electronic State of Atomically Dispersed Pt Supported on Nanosized CeO2

    • Single atom catalysts (SACs) have shown high activity and selectivity in a growing number of chemical reactions. Many efforts aimed at unveiling the structure-property relationships underpinning these activities and developing synthesis methods for obtaining SACs with the desired structures are hindered by the paucity of experimental methods capable of probing the attributes of local structure, electronic properties, and interaction with support-features that comprise key descriptors of their activity. In this work, we describe a combination of experimental and theoretical approaches that include photon and electron spectroscopy, scattering, and imaging methods, linked by density functional theory calculations, for providing detailed and comprehensive information on the atomic structure and electronic properties of SACs. This characterization toolbox is demonstrated here using a model single atom Pt/CeO2 catalyst prepared via a sol-gel-based synthesis method. Isolated Pt atoms together with extra oxygen atoms passivate the (100) surface of nanosized ceria. A detailed picture of the local structure of Pt nearest environment emerges from this work involving the bonding of isolated Pt2+ ions at the hollow sites of perturbed (100) surface planes of the CeO2 support, as well as a substantial (and heretofore unrecognized) strain within the CeO2 lattice in the immediate vicinity of the Pt centers. The detailed information on structural attributes provided by our approach is the key for understanding and improving the properties of SACs.

    • The ruff is a Palearctic wader with a spectacular lekking behavior where highly ornamented males compete for females1,2,3,4. This bird has one of the most remarkable mating systems in the animal kingdom, comprising three different male morphs (independents, satellites and faeders) that differ in behavior, plumage color and body size. Remarkably, the satellite and faeder morphs are controlled by dominant alleles5,6. Here we have used whole-genome sequencing and resolved the enigma of how such complex phenotypic differences can have a simple genetic basis. The Satellite and Faeder alleles are both associated with a 4.5-Mb inversion that occurred about 3.8 million years ago. We propose an evolutionary scenario where the Satellite chromosome arose by a rare recombination event about 500,000 years ago. The ruff mating system is the result of an evolutionary process in which multiple genetic changes contributing to phenotypic differences between morphs have accumulated within the inverted region.

  • Dynamic structure of active sites in ceria-supported Pt catalysts for the water gas shift reaction

    • Oxide-supported noble metal catalysts have been extensively studied for decades for the water gas shift (WGS) reaction, a catalytic transformation central to a host of large volume processes that variously utilize or produce hydrogen. There remains considerable uncertainty as to how the specific features of the active metal-support interfacial bonding-perhaps most importantly the temporal dynamic changes occurring therein-serve to enable high activity and selectivity. Here we report the dynamic characteristics of a Pt/CeO2 system at the atomic level for the WGS reaction and specifically reveal the synergistic effects of metal-support bonding at the perimeter region. We find that the perimeter Pt-0-O vacancy-Ce3+ sites are formed in the active structure, transformed at working temperatures and their appearance regulates the adsorbate behaviors. We find that the dynamic nature of this site is a key mechanistic step for the WGS reaction. Revealing the structure and dynamics of active sites is essential to understand catalytic mechanisms. Here the authors demonstrate the dynamic nature of perimeter Pt-0-O vacancy-Ce3+ sites in Pt/CeO2 and the key effects of their dynamics on the mechanism of the water gas shift reaction.

    • Auto-antibodies against the beta(1)-adrenoceptors are present in 30-40% of patients with dilated cardiomyopathy. Recently, a synthetic peptide corresponding to a sequence of the second extracellular loop of the human beta(1)-adrenoceptor (beta(1)-EC(II)) has been shown to produce endoplasmic reticulum (ER) stress, myocyte apoptosis and cardiomyopathy in immunized rabbits. To study the direct cardiac effects of anti-beta(1)-EC(II) antibody in intact animals and if they are mediated via beta(1)-adrenoceptor stimulation, we administered IgG purified from beta(1)-EC(II)-immunized rabbits to recombination activating gene 2 knock-out (Rag2(-/-)) mice every 2 weeks with and without metoprolol treatment. Serial echocardiography and cardiac catheterization showed that beta(1)-EC(II) IgG reduced cardiac systolic function after 3 months. This was associated with increase in heart weight, myocyte apoptosis, activation of caspase-3, -9 and -12, and increased ER stress as evidenced by upregulation of GRP78 and CHOP and cleavage of ATF6. The Rag2(-/-) mice also exhibited increased phosphorylation of CaMKII and p38 MAPK. Metoprolol administration, which attenuated the phosphorylation of CaMKII and p38 MAPK, reduced the ER stress, caspase activation and cell death. Finally, we employed the small-interfering RNA technology to reduce caspase-12 in cultured rat cardiomyocytes. This reduced not only the increase of cleaved caspase-12 but also of the number of myocyte apoptosis produced by beta(1)-EC(II) IgG. Thus, we conclude that ER stress plays an important role in cell death and cardiac dysfunction in beta(1)-EC(II) IgG cardiomyopathy, and the effects of beta(1)-EC(II) IgG are mediated via the beta(1)-adrenergic receptor.

    • Refrigeration systems running on transcritical CO2 cycle are considered an alternative to phase-down the use of hydrofluorocarbons (HFCs) in response to the Kigali Amendment. Responding to the need to improve the efficiency of CO2 booster systems and to identify optimum system designs, thermodynamic models of three booster systems are proposed with the use of an internal heat exchanger (IHX). Findings from this study suggest that placing the IHX with a low-temperature fluid side at the suction line of high pressure stage compressor and high-temperature fluid side at the outlet of gas cooler represents the most optimal approach in improving the coefficient of performance (COP) of the booster system. When operating in the transcritical conditions, the COP values can be improved by 6.35% at the IHX thermal effectiveness of 0.8 and by 6.48% at the ratio of medium temperature load to low temperature load of 6. Using IHX can significantly reduce the compressor discharge pressure, which can be reduced by 0.55 MPa at the ambient temperature of 40 degrees C. Furthermore, by adding IHX, the annual performance factor of CO2 system can be improved significantly by 1.68% and the annual total power consumption can be decreased by 6.48% in the tropical climate. It can be concluded that IHX can improve the COP values of a booster system when operating in the subtropical and tropical regions.

    • Evidence suggests that the autoimmune cardiomyopathy produced by a peptide corresponding to the sequence of the second extracellular loop of the beta(1)-adrenergic receptor (beta(1)-EC(II)) is mediated via a biologically active anti-beta(1)-EC(II) antibody, but the mechanism linking the antibody to myocyte apoptosis and cardiac dysfunction has not been well elucidated. Since the beta(1)-EC(II) autoantibody is a partial beta(1)-agonist, we speculate that the cardiomyopathy is produced by the beta(1)-receptor-mediated stimulation of the CaMKII-p38 MAPK-ATF6 signaling pathway and endoplasmic reticulum (ER) stress, and that excess norepinephrine (NE) exaggerates the cardiomyopathy. Rabbits were randomized to receive beta(1)-EC(II) immunization, sham immunization, NE pellet, or beta(1)-EC(II) immunization plus NE pellet for 6 mo. Heart function was measured by echocardiography and catheterization. Myocyte apoptosis was determined by terminal deoxytransferase-mediated dUTP nick-end labeling and caspase-3 activity, whereas CaMKII, MAPK family (JNK, p38, ERK), and ER stress signals (ATF6, GRP78, CHOP, caspase-12) were measured by Western blot, immunohistochemistry, and kinase activity assay. beta(1)-EC(II) immunization produced progressive LV dilation, systolic dysfunction, and myocyte apoptosis. These changes were associated with activation of GRP78 and CHOP and increased cleavage of caspase-12, as well as increased CaMKII activity, increased phosphorylation of p38 MAPK, and nucleus translocation of cleaved ATF6. NE pellet produced additive effects. In addition, KN-93 and SB 203580 abolished the induction of ER stress and cell apoptosis produced by the beta(1)-EC(II) antibody in cultured neonatal cardiomyocytes. Thus ER stress occurs in autoimmune cardiomyopathy induced by beta(1)-EC(II) peptide, and this is enhanced by increased NE and caused by activation of the beta(1)-adrenergic receptor-coupled CaMKII, p38 MAPK, and ATF6 pathway.

    • Dilated human cardiomyopathy is associated with suppression of the prosurvival phosphatidylinositol-3-kinase (PI3K)/Akt and STAT3 pathways. The present study was carried out to determine if restoration of the PI3K/Akt and STAT3 activity by darbepoetin alfa improved cardiac function or reduced cardiomyocyte apoptosis in rabbit autoimmune cardiomyopathy induced by a peptide corresponding to the second extracellular loop of the ss(1)-adrenergic receptor (ss(1)-EC(II)). We found that ss(1)-EC(II) immunization produced progressive LV dilation, systolic dysfunction and myocyte apoptosis as measured by TUNEL, single-stranded DNA antibody, and active caspase-3. These changes were associated with activation of p38 mitogen-activated protein kinase (MAPK), endoplasmic reticulum stress markers (GRP78 and CHOP), and increased cleavage of procaspase-12, as well as decreased phosphorylation of Akt and STAT3, and decreased Bcl2/Bax ratio. As expected, darbepoetin alfa treatment increased phosphorylation of Akt and STAT3. It also increased the myocardial expression of erythropoietin receptor which was reduced in the failing myocardium, and improved cardiac function in the ss(1)-EC(II)-immunized animals. The latter was associated with reductions of myocyte apoptosis and cleaved caspase-3, as well as reversal of increased phosphorylation of p38-MAPK, increased ER stress, and decline in Bcl2/Bax ratio. The anti-apoptotic effects of darbepoetin alfa via Akt and STAT activation were also demonstrated in cultured cardiomyocytes treated with the anti-ss(1)-EC(II) antibody. These effects of darbepoetin alfa in vitro were prevented by LY294002 and STAT3 peptide inhibitor. Thus, we conclude that darbepoetin alfa improves cardiac function and prevents progression of dilated cardiomyopathy probably by activating the PI3K/Akt and STAT3 pathways and reducing ER stress.

    • Production scheduling has great significance for optimizing tasks distribution, reducing energy consumption and mitigating environmental degradation. Currently, the research of production scheduling considering energy consumption mainly focuses on the traditional manufacturing workshop. With the wide application of the Internet of Things (IoT) technology, the real-time data of manufacturing resources and production processes can be retrieved easily. These manufacturing data can provide opportunities for manufacturing enterprises to reduce energy consumption and enhance production efficiency. To achieve these targets, a multi-period production planning based real-time scheduling (MPPRS) approach for the loT-enabled low-carbon flexible job shop (LFJS) is presented in this study to carry out real-time scheduling based on the real-time manufacturing data. Then, the mathematical models of real-time scheduling are established to achieve production efficiency improvement and energy consumption reduction. To obtain a feasible solution, an infinitely repeated game optimization approach is used. Finally, a case study is implemented to analyse and discuss the effectiveness of the proposed method. The bubble plume produced in Ruhrstahl–Heraeus (RH) up-snorkel plays an important role during refining, since it not only acts as a bubble pump, but also provides the reaction interface. Herein, it is aimed to form a new flow pattern in the up-snorkel by using a nonradially arranged gas-injection nozzle to enhance the 260 ton RH refining process. The results show that an upward spiral steel flow is produced, when nonradial gas-injection nozzles are used in the up-snorkel. Meanwhile, some bubbles moved toward the center region of the up-snorkel, which may be caused by the centripetal effect in a rotational steel flow. This leads to a more uniform bubble distribution on the cross section of the snorkel, compared to that of the conventional case. Specifically, the circulation flow rate is increased by about 18.0%, and the mixing time are shortened by about 26.2% (criteria of ±5%), compared to that of the conventional case. In addition, the inclusion removal rate is increased by 0.5%, 4.8%, and 11.3% for the inclusion size of 20, 50, and 100 μm, respectively, compared to the conventional radial nozzle case.
