    • All human diseases involve proteins, yet our current tools to characterize and quantify them are limited. To better elucidate proteins across space, time, and molecular composition, we provide a >10 years of projection for technologies to meet the challenges that protein biology presents. With a broad perspective, we discuss grand opportunities to transition the science of proteomics into a more propulsive enterprise. Extrapolating recent trends, we describe a next generation of approaches to define, quantify, and visualize the multiple dimensions of the proteome, thereby transforming our understanding and interactions with human disease in the coming decade.

  • Integration of RNA and protein expression profiles to study human cells

    • Cellular life is highly complex. In order to expand our understanding of the workings of human cells, in particular in the context of health and disease, detailed knowledge about the underlying molecular systems is needed. The unifying theme of this thesis concerns the use of data derived from sequencing of RNA, both within the field of transcriptomics itself and as a guide for further studies at the level of protein expression. In paper I, we showed that publicly available RNA-seq datasets are consistent across different studies, requiring only light processing for the data to cluster according to biological, rather than technical characteristics. This suggests that RNA-seq has developed into a reliable and highly reproducible technology, and that the increasing amount of publicly available RNA-seq data constitutes a valuable resource for meta-analyses. In paper II, we explored the ability to extrapolate protein concentrations by the use of RNA expression levels. We showed that mRNA and corresponding steady-state protein concentrations correlate well by introducing a gene-specific RNA-to-protein conversion factor that is stable across various cell types and tissues. The results from this study indicate the utility of RNA-seq also within the field of proteomics.The second part of the thesis starts with a paper in which we used transcriptomics to guide subsequent protein studies of the molecular mechanisms underlying malignant transformation. In paper III, we applied a transcriptomics approach to a cell model for defined steps of malignant transformation, and identified several genes with interesting expression patterns whose corresponding proteins were further analyzed with subcellular spatial resolution. Several of these proteins were further studied in clinical tumor samples, confirming that this cell model provides a relevant system for studying cancer mechanisms. In paper IV, we continued to explore the transcriptional landscape in the same cell model under moderate hypoxic conditions.To conclude, this thesis demonstrates the usefulness of RNA-seq data, from a transcriptomics perspective and beyond; to guide in analyses of protein expression, with the ultimate goal to unravel the complexity of the human cell, from a holistic point of view.

  • Finding Our Way through Phenotypes.

    • Despite a large and multifaceted effort to understand the vast landscape of phenotypic data, their current form inhibits productive data analysis. The lack of a community-wide, consensus-based, human- and machine-interpretable language for describing phenotypes and their genomic and environmental contexts is perhaps the most pressing scientific bottleneck to integration across many key fields in biology, including genomics, systems biology, development, medicine, evolution, ecology, and systematics. Here we survey the current phenomics landscape, including data resources and handling, and the progress that has been made to accurately capture relevant data descriptions for phenotypes. We present an example of the kind of integration across domains that computable phenotypes would enable, and we call upon the broader biology community, publishers, and relevant funding agencies to support efforts to surmount today's data barriers and facilitate analytical reproducibility.

  • Film Tourism collaborations: a critical analysis of INTERREG destination development projects

    • This chapter discusses the different stakeholders that take part in film tourism development projects, how do they collaborate, and what is actually possible to achieve? It is a critical analysis especially when it comes to the selection of stakeholders, the project process and potential outcomes in EU-funded INTERREG projects. The analysis contributes to enhance the knowledge about the different stakeholders involved. It is evident that a new stakeholder has emerged in film tourism development projects; film commissions. Moreover, the analysis shows that it is important that a diverse range of partners collaborate. However, organizations sometimes weak knowledge of the phenomenon in play as well as of each other’s work creates problems especially in the beginning. The idea of measurability is another complicating factor since there are conflicting understandings of what is possible to measure within collaborating organizations. There is also a search for best cases that can be used as role models that totally neglects local characteristics, which raises questions of transferability. What can be concluded is that projects such as these can make a difference to single destinations with for example new policies being developed, local and international networks created and possibilities for new tourism development.

    • This chapter discusses approaches to do foresights in outdoor recreation and tourism in nature, based on research from the Nordic countries. This region has a long-standing reputation for promoting outdoor recreation, with associated policies aiming to support this tradition for the benefit of future generations and environmental engagement. To do so successfully, one key input is improving the knowledge of outdoor recreational participation, associated trends, and future development. In this chapter, we therefore review different monitoring approaches to trend analyses as a means of forecasting participation and discuss supplementary approaches to foresight in outdoor recreation. We operationalize the Nordic context through studies undertaken in Finland, Sweden, Denmark, and Norway. Our examination shows that the tradition of national surveys and on-site visitor monitoring has proven effective. However, contemporary changes in society call for additional approaches to better understand future outdoor recreation. We therefore share experiences from several different approaches to foresight in outdoor recreation. This will inform future research as well as managers, entrepreneurs, planners and policymakers in their decision-making.

    • FörordI den här rapporten redovisas några första sammanställningar från en nationell enkätundersökning avseende svenskarnas fritidsaktiviteter i fjällen, deras besök i olika fjällområden, landskapsrelationer, fjällens roll i sociala medier, upplevelser av vindkraft och attityder till skyddad natur. Enkäten ingår som en del i projekten "Bortom konflikter" och "Den nya fjällupplevelsen" inom ramen för Naturvårdsverkets forskningssatsning"Storslagen fjällmiljö" (www.storslagnafjall.se). Utifrån materialet i denna rapport kommer framöver olika fördjupningar och kompletteringar att göras, vilka kommer redovisas påolika sätt–vetenskapligt och populärvetenskapligt. Det är vår förhoppning att dessa studier blir en viktig del i referensramen för alla de beslut som fattas av myndigheter, företag och politiker när det gäller det fortsatta arbetet för en hållbar utveckling i fjällen. För dagens och framtidens grupper, aktiviteter och ideal kommer att avgöra hur hållbar utvecklingen blir–och dagens besökare i fjällen är här en av nyckelgrupperna.

    • Naturen lyfts ofta fram som en viktig resurs för svenskt turism och den har stor betydelse för bilden av Sverige internationellt. Naturturism är ett begrepp som intuitivt är enkelt att förstå men som är svårare att fånga i mer exakta termer. Då det saknas en vedertagen definition saknas tillförlitliga mätningar och kunskaperna om naturturismen är bristfälliga, både som näringsverksamhet (utbud av varor och tjänster) och som företeelse (vistelser i naturen utanför den vanliga omgivningen). Syftet med föreliggande rapport är därför att beskriva naturturismen som näring och företeelse, att ge ett förslag på definition av naturturism, att granska förekommande turismstatistik och andra källor till kunskap om svenskt turism utifrån ett naturturistiskt perspektiv, samt att ge förslag hur naturturismen i Sverige bättre kan mätas och kvantifieras. Rapporten innehåller också fyra internationella fallstudier för att belysa motsvarande förhållanden i Nya Zeeland, Nordamerika, Skottland och Finland. Resultaten från arbetet kan sammanfattas i fem huvudsakliga slutsatser:Det saknas en definition: Naturturism (och liknande begrepp) används flitigt utan att det finns någon vedertagen definition. Vi föreslår en bred definition som bygger på den generella definitionen av turism.Befintlig statistik säger lite: Idag förekommande turismstatistik förmår bara i mycket begränsad utsträckning mäta naturturism.Likheter med andra länder, men också mycket vi kan lära av dem: Situationen i Sverige är inte unik, de internationella fallstudierna visar på liknande förhållanden i andra länder.Naturturismen måste mätas ur flera perspektiv: Naturturismens stora bredd gör att den måste mätas ur olika perspektiv med olika metoder. Vi föreslår fyra mätningar – två konsumentundersökningar, en producentundersökning, samt systematiska besökarstudier.Tydlig företrädare och samordning mellan aktörer behövs: Naturturismen har många inblandade aktörer och det behövs en samlande och drivande nationell företrädare där ansvar för statistik utgör en del av uppdraget.

    • Rapporten beskriver forskningsfronten inom relationsmarknadsföringsansatsen (RM) i Europa under senare år. I rapporten presenteras en genomgång av ett antal artiklar och böcker inom området RMs utveckling under senare år i Europa. ’State-of-the-Art inom området beskrivs och diskuteras genom en presentation av synen på RM generellt, ett antal modeller som används för att illustrera perspektivet samt genom en presentation av vilka ämnesinriktningar och metoder som används inom ansatsen. Slutsatser som presenteras efter genomgång av materialet är att RM har återfört fokus till de relationer som en organisation har.   I detta perspektiv är direkta, kontinuerliga och långsiktiga relationer av största betydelse för organisationers framgång. En stark betoning ligger på kundservice, värdeskapande, uppfyllande av förväntningar och inte minst kvalitet. Forskningen inom RM i Europa har, under de senaste fem åren, främst fokuserats på följande ämnesinriktningar: service/kvalitet, nätverk, "business-to-business". personal/intern marknadsföring, kund och begreppet "trust". En ständig begreppsutveckling pågår inom området.

  • The Lives and Attitudes of Employees in Tourism : A study of seasonal workers in a Swedish ski-resort

    • In this paper the reader is introduced to the concept of seasonality and the characteristics of the labour market and work force in tourism. The concepts of employee empowerment work motivation and occupational communities are further discussed. The study is based upon field research carried out in the ski-resort of Åre and its surrounding villages. Data collection was carried out in the form of questionnaires and in-depth interviews with seasonal workers. The empirical part presented in the paper consists of defining characteristics of occupational communities, seasonal workers� views on work situation, important factors for doing a good job, and the attraction of Åre ski-resort for seasonal workers. When assessing the findings of the in-depth interviews, two different groups of workers appear. The first of these two � the resident community � is made up by individuals, who live on a permanent basis in the Åre region. This group is characterised by individuals, who devote themselves to non-work friends and family commitment during their spare time. They also show interest in career development within the tourism sector. Individuals, who only live in the Åre region during their seasonal work, make up the second group � the migrant community. This group is characterised by individuals, who have a close knit relationship with their co-workers, which may be a result of them sharing place of residence. They also share a work-based image and reference group. When developing strategies for hiring, further training and retaining these individuals, it is recommended to pay attention to the differences between these two groups. An organisation, which empowers its employees, gives them the opportunity to perform at their best knowledge. In this situation employees need to know how well they perform. The results from the questionnaire show that encouragement from the closest manager in the organisation is a very important factor for the employees� work performance. When respondents expressed their experience at work, encouragement from their co-workers received a higher, though not significant, score. Another important aspect for empowered employees is that they need to feel they are allowed to discuss ideas, problems etc as regards their work situation. This is viewed by a good half of the respondents to be an important factor for their work performance but there is only one third claiming that this is experienced at their place of work. Authority to make own decisions as regards work performance, is also a factor to be considered when empowering employees. Almost two thirds of the respondents perceived themselves as having the authority to make own decisions. Studies show that work itself may not be the most important motivational stimulus for seasonal employees, but rather the close social relationships. An overwhelming majority is of the opinion that nice co-workers are important for getting motivated in one�s work.

    • In the tourism sector, quality is closely related to employee performance. One of the most important challenges facing managers is the creation of a context within which employees feel motivated and will act in order to achieve the goals of the organisation (Lee-Ross, 1999). The aim of the study is to investigate motivation in a sample of seasonal workers in the ski-resort of Åre in northern Sweden. This study is based upon field research carried out in Åre and its surrounding villages. Data collection was carried out in the form of questionnaires and in-depth interviews. According to Herzberg�s two-factor need theory, humans have two different sets of needs and that the different elements of the work situation satisfies or dissatisfies these needs (Wright, 1989). The first set of needs concerns the basic survival needs of a person � the hygiene factors (e.g. supervision, salary and physical conditions). When these needs are satisfied they do not motivate or cause satisfaction, they only prevent dissatisfaction (Pinder, 1998). In this study evidence was found that support this theory. When reviewing the results, work-related stimuli were not important for work motivation. Other results the study question Herzberg�s argument that job content is the only way to increase work motivation (Wright, 1989). The results from the in-depth interviews indicate that social stimuli were also regarded as an important construct for the enhancement of work motivation. This conclusion is also supported by quantitative data. The second set of needs in Herzberg�s theory, the growth needs (e.g. responsibility, advancement and recognition) was measured by the construct of empowerment. The results show that the construct of empowerment to a great extent influence work motivation.

    • In the service sector, quality is closely related to employee performance. An essential feature of any successful organisation is motivated employees. Therefore, the attitude of an employee towards his/her place of work and the extent to which an employer is able to motivate employees may have a direct effect on the quality of those products offered to tourists. One of the most important challenges facing managers is the creation of a context within which employees feel motivated and will act in order to achieve the goals of the organisation. The internal marketing concept emerged from service marketing and its main concern was to get everyone who was involved in service encounters � the front line or contact staff � to perform better in the interaction with customers. The usage of the concept has extended beyond its traditional field and is now accepted in all kinds of organisations. The overall objective of the internal marketing process is to attract suitable staff members as contact personnel and to managerial positions, to have a high retention rate among the co-workers and to develop motivated and customer-conscious employees. This study is based upon field research carried out in the ski-resort of Åre situated in the northern part of Sweden. Data collection was carried out in the form of in-depth interviews. The sample for the in-depth interviews consisted of managers from four hotels in Åre, which offer both accommodation and food and beverage. The findings of this study show that attracting, developing, motivating, and retaining qualified employees were of primary concern for management and were included in a deliberate company strategy. When working with these parameters, management strongly emphasised customer-consciousness and sales-mindedness. Management of the different companies was aware of the importance of front-line personnel�s immediate effect on customer satisfaction and how important staff autonomy and know-how were for business success. There was also a great amount of understanding for the need of an information process, which were directed from and to every side of the organisation. As a result of this understanding, the companies worked with different kinds of platforms for feedback and information flows. Seasonal fluctuations in demand pose, according to management, the greatest obstacle for working with internal marketing activities. Seasonality has negative effects on the selection, recruitment, retention, training, development, and motivation of employees in the companies included in the study.

The aim of the study was to investigate work motivation in a sample of seasonal workers in the ski-resort of Åre in northern Sweden and data was collected by means of in-depth interviews and questionnaires. The study tested Herzberg�s Two-Factor Theory by using Structural Equation Modelling (LISREL). The results show that the seasonal workers were motivated by the so called growth factors, which consisted of responsibility, information, feedback, knowledge and training. The hygiene factors, which consisted of wage level, rewards and interpersonal relations did not have an influence on work motivation.

    • Within the Spent Fuel & Waste Science and Technology (SFWST) Program, research work continues further with the goal to better understand long-term performance of disposal systems in three main geologic rock types: clay/shale, salt, and crystalline rock. This report documents LBNL’s research activities related to investigations of crystalline host rock according to the scope of two work packages: SF-20LB01030207 “Crystalline International Collaborations – LBNL,” and SF-20LB01030203 “Crystalline Disposal R&D – LBNL.” These research activities correspond are related to key Features, Events, and Processes (FEPs), ranked in importance from medium to high, as listed in Table 7 of the Used Fuel Disposition Campaign Disposal Research and Development Roadmap (FCR&D-USED-2011-000065 REV0) (Nutt, 2011). Specifically, these research activities address FEP 2.2.01, Excavation Disturbed Zone (EZD). The results of these research activities provide important insights into understanding and predicting flow and transport processes that could occur in low-permeability crystalline rocks, in which fractures might serve as main conduits for fluid flow and radionuclide transport. The evolution of the EDZ during the excavation of the tunnel as well as the evolution of microcrack growth within the EDZ after the emplacement of backfill are critical for predicting the long- term behavior of the EDZ. A number of factors including stress, temperature, water activity, capillary pressure, chemistry, and mineralogy can affect the rock evolution, which require advanced experimental tools to study it.

This chapter explores complex relationships among popcultural phenomena, destination image creation, and tourism consumption. By taking a broader social science approach, it revisits and connects research themes, such as symbolic consumption, negotiated representations, fans and fandom, technology mediation, and media convergence.   • Generation of monospecific antibodies based on affinity capture of polyclonal antibodies

    • A method is described to generate and validate antibodies based on mapping the linear epitopes of a polyclonal antibody followed by sequential epitope-specific capture using synthetic peptides. Polyclonal antibodies directed towards four proteins RBM3, SATB2, ANLN, and CNDP1, potentially involved in human cancers, were selected and antibodies to several non-overlapping epitopes were generated and subsequently validated by Western blot, immunohistochemistry, and immunofluorescence. For all four proteins, a dramatic difference in functionality could be observed for these monospecific antibodies directed to the different epitopes. In each case, at least one antibody was obtained with full functionality across all applications, while other epitope-specific fractions showed no or little functionality. These results present a path forward to use the mapped binding sites of polyclonal antibodies to generate epitope-specific antibodies, providing an attractive approach for large-scale efforts to characterize the human proteome by antibodies.

    • Periodontal Ehlers-Danlos syndrome (pEDS) is an autosomal-dominant disorder characterized by early-onset periodontitis leading to premature loss of teeth, joint hypermobility, and mild skin findings. A locus was mapped to an approximately 5.8 Mb region at 12p13.1 but no candidate gene was identified. In an international consortium we recruited 19 independent families comprising 107 individuals with pEDS to identify the locus, characterize the clinical details in those with defined genetic causes, and try to understand the physiological basis of the condition. In 17 of these families, we identified heterozygous missense or in-frame insertion/deletion mutations in C1R (15 families) or C1S (2 families), contiguous genes in the mapped locus that encode subunits C1r and C1s of the first component of the classical complement pathway. These two proteins form a heterotetramer that then combines with six C1q subunits. Pathogenic variants involve the subunit interfaces or inter-domain hinges of C1r and C1s and are associated with intracellular retention and mild endoplasmic reticulum enlargement. Clinical features of affected individuals in these families include rapidly progressing periodontitis with onset in the teens or childhood, a previously unrecognized lack of attached gingiva, pretibial hyperpigmentation, skin and vascular fragility, easy bruising, and variable musculoskeletal symptoms. Our findings open a connection between the inflammatory classical complement pathway and connective tissue homeostasis.

    • Atherosclerosis is a chronic inflammatory disease promoted by hyperlipidemia. Several studies support FOXP3-positive regulatory T cells (Tregs) as inhibitors of atherosclerosis; however, the mechanism underlying this protection remains elusive. To define the role of FOXP3-expressing Tregs in atherosclerosis, we used the DEREG mouse, which expresses the diphtheria toxin (DT) receptor under control of the Treg-specific Foxp3 promoter, allowing for specific ablation of FOXP3(+) Tregs. Lethally irradiated, atherosclerosis-prone, low-density lipoprotein receptor-deficient (Ldlr(-/-)) mice received DEREG bone marrow and were injected with DT to eliminate FOXP3(+) Tregs. Depletion of Tregs caused a 2.1-fold increase in atherosclerosis without a concomitant increase in vascular inflammation. These mice also exhibited a 1.7-fold increase in plasma cholesterol and an atherogenic lipoprotein profile with increased levels of VLDL. Clearance of VLDL and chylomicron remnants was hampered, leading to accumulation of cholesterol-rich particles in the circulation. Functional and protein analyses complemented by gene expression array identified reduced protein expression of sortilin-1 in liver and increased plasma enzyme activity of lipoprotein lipase, hepatic lipase, and phospholipid transfer protein as mediators of the altered lipid phenotype. These results demonstrate that FOXP3(+) Tregs inhibit atherosclerosis by modulating lipoprotein metabolism.

    • Destinations associated with pop culture phenomena, such as destinations depicted in books and films, often experience increased numbers of visitors as well as strengthened and changed destination images. The pop culture phenomenon the Twilight Saga (book and film series) is in this paper used as an example to explore how a pop culture phenomenon can affect destinations, and how destinations manage this type of tourism. Case studies in Forks, WA, in the USA, Volterra, Montepulciano in Italy and British Columbia in Canada illustrate different tourism destination strategies. Forks has, for example, developed experiences based on a fictionally constructed reality connected to Twilight, which has reimagined the destination, and, thus, fabricated the authenticity of the place. Volterra and Montepulciano, on the other hand, have experienced a Twilight Saga tourism development characterised by deliberations regarding the immersion of Twilight Saga elements into their cultural heritage which has resulted in a strategy best described as guarding the authenticity of their respective destinations. Finally, British Columbia has had no strategy and exhibits little interest in Twilight tourism. The priority of the destination has been to satisfy the needs of film producers. The study elaborates on different paths of pop culture tourism development, i.e. it is not always advisable to fully exploit the potential that a pop cultural phenomenon can bring to a destination.     • Background In patients with idiopathic inflammatory myopathies (IIM), autoantibodies are associated with specific clinical phenotypes suggesting a pathogenic role of adaptive immunity. We explored if autoantibody profiles are associated with specific HLA genetic variants and clinical manifestations in IIM. Methods We included 1348 IIM patients and determined the occurrence of 14 myositis-specific or-associated autoantibodies. We used unsupervised cluster analysis to identify autoantibody-defined subgroups and logistic regression to estimate associations with clinical manifestations, HLA-DRB1, HLA-DQA1, HLA-DQB1 alleles, and amino acids imputed from genetic information of HLA class II and I molecules. Findings We identified eight subgroups with the following dominant autoantibodies: anti-Ro52, -U1RNP, -PM/Scl,-Mi2,-Jo1,-Jo1/Ro52,-TIF1 gamma or negative for all analysed autoantibodies. Associations with HLA-DRB1*11, HLA-DRB1*15, HLA-DQA1*03, and HLA-DQB1*03 were present in the anti-U1RNP-dominated subgroup. HLA-DRB1*03, HLA-DQA1*05, and HLA-DQB1*02 alleles were overrepresented in the anti-PM/Scl and anti-Jo1/ Ro52-dominated subgroups. HLA-DRB1*16, HLA-DRB1*07 alleles were most frequent in anti-Mi2 and HLA- DRB1*01 and HLA-DRB1*07 alleles in the anti-TIF1 gamma subgroup. The HLA-DRB1*13, HLA-DQA1*01 and HLA-DQB1*06 alleles were overrepresented in the negative subgroup. Significant signals from variations in class I molecules were detected in the subgroups dominated by anti-Mi2, anti-Jo1/Ro52, anti-TIF1 gamma, and the negative subgroup. Interpretation Distinct HLA class II and I associations were observed for almost all autoantibody-defined subgroups. The associations support autoantibody profiles use for classifying IIM which would likely reflect underlying pathogenic mechanisms better than classifications based on clinical symptoms and/or histopathological features.     • Popular culture tourism encompasses a range of expressive practices that attract fans traveling to destinations associated with their fandom pursuit. However, scholarship on this multifaceted phenomenon is today over-fragmented and obscured by separate disciplinary agendas and priorities. We argue that the scope and breadth of popular culture tourism calls for its interdisciplinary treatment as a distinct field. Through a scoping literature review, this paper identifies the foundational elements of its conceptual and ontological roots, extracting key insights and discursive themes that can help establish a comprehensive perspective on the study and management of popular culture tourism. Our inquiry builds common ground that can shed light on the complexity of popular culture expressions and enable their strategic role as a destination placemaking tool. Thematic areas of convergence resulting in the emergent configuration of the field are delineated, and primary research questions for the comprehensive study of popular culture tourism are outlined.

A web survey for fans of the Twilight Saga is implemented, using the concepts of cognitive, affective, and evaluative social identity and personal, product, and situational involvement. The purpose is to examine to what degree social identity and involvement can explain pop culture fans' future intention to travel, make recommendations to others, and use social media. Findings show that pop culture fans use social media to a large extent and that these means are important for making decisions about traveling and event participation. Moreover, the chapter shows that involvement dimensions are more important than social identity dimensions to explain future intention to travel, to recommend to others, and to use social media.

    • On-screen tourism destinations provide tourists with a mixture of reality and mass-media experience. This study builds a conceptual framework of authenticity evaluating the relationship between fantasy proneness, authenticity, and destination loyalty. It is among the first to compare perceived authenticity of dual images of a destination, both the film location and the story’s setting. Using Game of Thrones and the city of Dubrovnik as a case, it applies a mixed method of interviews followed by a main survey with both closed- and open-ended questions. The primary findings show that the relationship between authenticity of the film location (Dubrovnik) and loyalty is positive, but such relationship is insignificant for the authenticity of the story’s setting (King’s Landing). This investigation enriches our understanding of the complex assessments of authenticity and expands the theory of imagination by evaluating the effects of fantasy proneness on perceived authenticity in the context of on-screen tourism.

    • The servicescape is a central concept within the service marketing literature based on rationally worked out presentations of an environment. It is vital in creating the service experience, and communicates powerful messages to the consumer about the expected quality of the experience (e.g. Arnould, Price & Tierney, 1998; Aubert-Gamet, 1997; Babin & Dardin, 1996; Baker, 1987; Baker, Grewal & Levt, 1992; Bitner, 1990; 1992; Booms & Bitner, 1982; Donovan & Rossiter, 1982; Mehrabian & Russell, 1974; Tilly, 1994). Yet, the branding and marketing of places hides the different value constructions behind such commercial activities. Furthermore, much of the servicescape research has been very conceptual, focusing on only one of the physical elements of the physical environment (Countryman and Jang, 2006). One of the objectives of this research is to attempt to fill that gap by categorising all aspects of the servicescape based on data from popular culture destinations and an elaborated model of analysis for servicescape elements. More specifically, the aim of the paper is to elaborate the notion of servicescapes towards a more holistic understanding and application of the concept. The fact that the tourism experience involves complex interactions with the physical and cultural landscapes of the tourist destination, leading to complex positive and negative impacts in the local communities, evidently looping back affecting the consumer experience, implies an extension of the concept of servicescape. In this more complex context of tourism consumption, Mossberg (2007) has suggested that the servicescape concept should be substituted with the broader experiencescape concepts to better reflect that in tourism, the service product is not limited to one company and service environment but rather a combination of service providers at a destination or experience environment. We argue that the spatial dimensions of the concept need to be even further developed to incorporate the geographies of tourism. The primary data included in the study further supports this claim. A total of 278 photos and 20 video clips captured at four popular culture tourism destinations were included in the sample. The four destinations, all related to the popular culture phenomenon the Twilight Saga, are located in USA (Forks, La Push and Port Angeles), British Columbia, Canada, and Italy (Volterra and Montepulciano). NVivo9 was employed for the analysis of data in two stages. The first stage was to analyse secondary data (literature) to develop a model for analysis of primary data (stage two). The analysis approach can be described as an a priori analysis based on a model generated from previous research within servicescape and experiencescape. However, the research design also included a post hoc analysis allowing for new nodes/(sub)categories to be developed, when applicable, from empirical data. Findings show that evidence from the popular culture tourism destinations supports previous research as regards elements of the servicescape and experiencescape concepts. In addition to this, two new empirically generated subcategories were developed clearly related to the geographies of tourism. Alas, support for a more holistic approach to the servicescape and experiencescape concepts was found.

    • Objective With the identification of mutations in the conserved telomere maintenance component 1 (CTC1) gene as the cause of Coats plus (CP) disease, it has become evident that leukoencephalopathy with calcifications and cysts (LCC) is a distinct genetic entity. Patients and Methods A total of 15 patients with LCC were identified from our database of patients with intracranial calcification. The clinical and radiological features are described. Results The median age (range) at presentation was 10 months (range, 2 days-54 years). Of the 15 patients, 9 presented with epileptic seizures, 5 with motor abnormalities, and 1 with developmental delay. Motor abnormalities developed in 14 patients and cognitive problems in 13 patients. Dense calcification occurred in the basal ganglia, thalami, dentate nucleus, brain stem, deep gyri, deep white matter, and in a pericystic distribution. Diffuse leukoencephalopathy was present in all patients, and it was usually symmetrical involving periventricular, deep, and sometimes subcortical, regions. Cysts developed in the basal ganglia, thalamus, deep white matter, cerebellum, or brain stem. In unaffected areas, normal myelination was present. No patient demonstrated cerebral atrophy. Conclusion LCC shares the neuroradiological features of CP. However, LCC is a purely neurological disorder distinguished genetically by the absence of mutations in CTC1. The molecular cause(s) of LCC has (have) not yet been determined.

    • Background and aim: Adequate velopharyngeal function and speech are main goals in the treatment of cleft palate. The objective was to investigate if there were differences in velopharyngeal competency (VPC) and hypernasality at age 5 years in children with unilateral cleft lip and palate (UCLP) operated on with different surgical methods for primary palatal repair. A secondary aim was to estimate burden of care in terms of received additional secondary surgeries and speech therapy. Design: Three parallel group, randomised clinical trials were undertaken as an international multicentre study by 10 cleft teams in five countries: Denmark, Finland, Sweden, Norway, and the UK. Methods: Three different surgical protocols for primary palatal repair were tested against a common procedure in the total cohort of 448 children born with a non-syndromic UCLP. Speech audio and video recordings of 391 children (136 girls, 255 boys) were available and perceptually analysed. The main outcome measures were VPC and hypernasality from blinded assessments. Results: There were no statistically significant differences between the prevalences in the arms in any of the trials. VPC: Trial 1, A: 58%, B: 61%; Trial 2, A: 57%, C: 54%; Trial 3, A: 35%, D: 51%. No hypernasality: Trial 1, A: 54%, B: 44%; Trial 2, A: 47%, C: 51%; Trial 3, A: 34%, D: 49%. Conclusions: No differences were found regarding VPC and hypernasality at age 5 years after different methods for primary palatal repair. The burden of care in terms of secondary pharyngeal surgeries, number of fistulae, and speech therapy visits differed.

    • Vast regions of the northern hemisphere are exposed to snowfall and seasonal frost. This has large effects on spatiotemporal distribution of infiltration and groundwater recharge processes as well as on the fate of pollutants. Therefore, snow and frost need to be central inherent elements of risk assessment and management schemes. However, snow and frost are often neglected or treated summarily or in a simplistic way by groundwater modellers. Snow deposition is uneven, and the snow is likely to sublimate, be redistributed and partly melt during the winter influencing the mass and spatial distribution of snow storage available for infiltration, the presence of ice layers within and under the snowpack and, therefore, also the spatial distribution of depths and permeability of the soil frost. In steep terrain, snowmelt may travel downhill tens of metres in hours along snow layers. The permeability of frozen soil is mainly influenced by soil type, its water and organic matter content, and the timing of the first snow in relation to the timing of sub-zero temperatures. The aim with this paper is to review the literature on snow and frost processes, modelling approaches with the purpose to visualize and emphasize the need to include these processes when modelling, managing and predicting groundwater recharge for areas exposed to seasonal snow and frost

    • Large parts of the northern hemisphere are covered by snow and seasonal frost. Climate warming is affecting spatiotemporal variations of snow and frost, hence influencing snowmelt infiltration, aquifer recharge and river runoff patterns. Measurement difficulties have hampered progress in properly assessing how variations in snow and frost impact snowmelt infiltration. This has led to contradicting findings. Some studies indicate that groundwater recharge response is scale dependent. It is thus important to measure snow and soil frost properties with temporal and spatial scales appropriate to improve infiltration process knowledge. The main aim with this paper is therefore to review ground based methods to measure snow properties (depth, density, water equivalent, wetness, and layering) and soil frost properties (depth, water and ice content, permeability, and distance to groundwater) and to make recommendations for process studies aiming to improve knowledge regarding infiltration in regions with seasonal frost. Ground-based radar (GBR) comes in many different combinations and can, depending on design, be used to assess both spatial and temporal variations in snow and frost so combinations of GBR and tracer techniques can be recommended and new promising methods (auocostics and self potential) are evolving, but the study design must be adapted to the scales, the aims and the resources of the study.     • Significant challenges are faced by front-line hospitality and tourism employees interacting with customers on a daily basis. These challenges and the ways they are dealt with form the focus of this thesis. As they interact, a range of incidents may arise from highly satisfactory encounters to service failures. It has been argued that in order to achieve satisfactory service encounters, employees need to feel in control, satisfied and motivated as well as being given appropriate assistance, information and training. Front-line employees in hospitality and tourism are focused on here due to the complex nature of the industry. It is commonly agreed that it is characterised by poor working conditions such as anti-social working hours, part-time and/or seasonal work, minimal on-the-job training and low levels of pay. These are all factors which may affect employee work satisfaction and motivation as well as recruitment and retention. The thesis is comprised of four studies. In Study I, hospitality and tourism employee work motivation is analysed. Study II examines front-line hotel restaurant employees’ experiences of service encounters. How the skills utilised by employees during these service encounters are acquired and developed is then explored in Study III. Study IV concludes by examining employee informal information sharing activities, which offer one way of developing these skills. The thesis’ findings paint a more positive picture of work and workers in hospitality and tourism than suggested in some of the earlier published research. This could be explained by the fact that the thesis utilises an employee perspective in its truest sense. The findings of the thesis show that employees are motivated (to large extent by factors that have been pointed out in earlier published work motivation research) and that they are active in the pursuit of developing skills necessary to create satisfactory service encounters. Another conclusion of the thesis’ findings is that the interaction between workers and guests in the service encounter provides an important opportunity for workers to feel satisfied at work.

    • This paper explores frontline hotel restaurant workers' experiences of critical service encounters by employing the Five Aspects Meal Model (FAMM) as a frame of reference. The questions raised in this study were: Which satisfactory and dissatisfactory incidents can be found in the different aspects of the restaurant experience according to employees? How can they be categorised under these aspects? A critical incident technique was employed to collect experiences of critical service encounters together with projective techniques. The findings of the study suggest that frontline workers experience critical service encounters related to four (Room, Meeting, Product, Management Control System) out of the five aspects of FAMM. Only a limited number of incidents were found in the Room aspect compared to the other three aspects. The largest proportions of incidents were found in Meeting and Product aspects of the meal experience. This insight may guide management of restaurants when making decisions in, for example, the recruitment, information and training of workers, the development of products and suitable management control systems as well as design of physical surroundings. Earlier research has explored the meal experience from a management as well as customer perspective. In this study, employees' experiences are in focus, which constitute the main research contribution of the study. © 2011 Taylor & Francis.

    • This research note suggests that the relationship between nature-based tourism entrepreneurs and money is complex and not fully understood. Based on observations from the Swedish supply data, four propositions are presented to illustrate why this is the case. We argue that (1) the identity of the entrepreneurs are not compatible with profit and growth, (2) niche markets and limited market knowledge obstruct opportunity for growth, (3) dependence of natural resources put sustainability ahead of growth and (4) nature-based tourism firms have limited control over their production process. These propositions can assist in better understanding how nature-based tourism companies can contribute to regional development and serve as a point of departure for further research.

    • The objective of this study was to understand work motivation in a sample of seasonal workers at a tourism destination strongly steered by seasonality. Furthermore, it was investigated whether seasonal workers could be divided into worker subgroups on the basis of their work motivation. A structural equations model tested Herzberg’s Two-Factor Theory of work motivation empirically. The findings of the study support the Two-Factor Theory of work motivation. Furthermore, results indicated that a migrant community of workers was significantly less concerned about wage level as well as significantly more concerned about meeting new people than resident workers. As a result of these findings, it is suggested that management of businesses in hospitality and tourism need to consider that the seasonal workforce consists of different kinds of worker subgroups, which have different needs to be satisfied.

    • There is nowadays a growing scholarship examining the intersection of popular culture and tourism. Most of this literature reveals that much of this work is grounded in an optimistic predisposition about the positive effects that tourism induced by popular culture has for locales. This is mainly featured in the dominant management discourse that feeds an overly positive tone about the benefits of popular culture for tourism development. However, there is a less loud line of scholarship originated in critical social studies that views regional popular culture tourism contexts in a more nuanced way bearing dubious outcomes. To address these shortcomings, the purpose of this scoping review is to demarcate and establish the strategic role of popular culture in regional tourism development. We put forward that a holistic approach entails the comprehensive treatment and multi-level embeddedness of popular culture tourism into regional contexts, epitomised by their territorial capital. On this basis, we present the predominant themes shaping the discourse in the field of popular culture tourism by discerning its two primary disciplinary streams, namely management and critical studies. Next, implications are drawn for the sustainable territorial touristification of places by leveraging comprehensively the development of popular culture tourism in local communities.
