  • In vitro capacitation of bull spermatozoa by oviductal fluid and its components

    • Sperm capacitation is crucial for fertilization. However, debate continues on exactly how, where and when capacitation is elicited in the bovine female genital tract. In this study we used merocyanine-540 and the chlortetracycline (CTC) assay to test how capacitation of bull spermatozoa is affected in vitro by exposure to oviductal fluid (ODF) collected in vivo, various glycosaminoglycans (GAGs) or bicarbonate. Following different durations of exposure, spermatozoa were stained with CTC or merocyanine-540, and evaluated with epifluorescent light microscopy or flow cytometry, respectively. Incubation time did not significantly affect capacitation. Exposure (30-120 min) to ODF capacitated (p less than 0.05) bull spermatozoa as measured by either merocyanine-540 or CTC. Hyaluronan was the only GAG that induced a significant increase in B-pattern spermatozoa (capacitated; p = 0.012) compared with controls. Dermatan sulphate also induced capacitation (merocyanine-540 high fluorescence; p = 0.035). Exposure to bicarbonate-enriched media also yielded an increase in merocyanine-540 high fluorescence (p less than 0.0001). When bicarbonate was added to the other treatments (ODF or GAGs) an equal increase in merocyanine-540 high fluorescence was noted (p less than 0.0001), compared with before addition of bicarbonate and independent of the treatment before exposure. There was no significant difference in the number of B-pattern spermatozoa when bicarbonate was added, but an significant increase in spermatozoa with an acrosome-reacted (AR)-pattern (p less than 0.0001) was observed. Exposure of spermatozoa to solubilized zonae pellucidae significantly increased the AR-pattern spermatozoa (p = 0.016). In conclusion, ODF was more potent in inducing capacitation of bull spermatozoa than the individual GAGs. Our results also indicate that bicarbonate is an effector of bull sperm capacitation.

  • Effect of freezing bull semen in two non-egg yolk extenders on post-thaw sperm quality

    • Traditionally, extenders for bull semen included egg yolk or milk, but recently there has been a move to avoid material of animal origin. The aim of this study was to evaluate the effects of two commercial extenders (based on soya lecithin and liposomes) on bull sperm quality after cryopreservation. Post-thaw sperm quality was evaluated by computer-assisted sperm analysis and flow cytometric assessment of membrane integrity, chromatin integrity, mitochondrial membrane potential, production of reactive oxygen species and tyrosine phosphorylation. Furthermore, an artificial insemination (AI) trial was conducted, and 56-day non-return rates were evaluated. Semen frozen in the liposome-based extender showed similar membrane integrity and higher mitochondrial membrane potential compared to those in the soya lecithin-based extender. Chromatin integrity and production of live H2O2+ reactive oxygen species were similar in both extenders. Less superoxide was produced in the samples extended with liposome-based extender, with or without menadione stimulation. Chromatin integrity and tyrosine phosphorylation were not affected by either type of extender. No differences in 56-day non-return rate between extenders containing soya lecithin and liposomes were observed in the AI trial (66%+/- 0.8 and 65%+/- 0.8, respectively). In conclusion, the sperm quality of bull semen frozen in the two extenders that do not contain material of animal origin was similar, although the semen frozen in the liposome-based extender had higher mitochondrial membrane potential. Either extender could be used in situations where extenders containing material of animal origin are to be avoided.

    • This study investigated the effect of continuous elevated cortisol concentrations during standing oestrus on time of ovulation and patterns Of progesterone. oestradiol and luteinising, hormone (LH) in sows. The elevation of cortisol concentrations was achieved through repeated intravenous injections of synthetic adrenocorticotropic hormone (ACTH) every 2 It for approximately 48 h, from the onset of the second standing oestrus alter weaning. Treatment was terminated when ovulation was detected (monitored by transrectal ultrasonography every 4h) or when (lie sow had received a maximum of 24 injections. The close of ACTH (2.5 mu g/kg) was chosen to mimic the cortisol concentrations seen during mixing of unfamiliar SOWS. The sows (n = 14) Were surgically fitted with jugular vein catheters and randomly divided into a control (C group) where only NaCl solution were injected) or an ACTH group. Blood samples were collected every 2 h. In parallel with the blood sampling, saliva samples for cortisol analyses were taken from eight sows before onset of treatment and from four of the sows during treatment. There was no difference in time from onset of standing, oestrus to ovulation between the two groups. The interval between the peaks of oestradiol and LH to ovulation was prolonged in the ACTH group compared to the C group (p less than 0.05). with a tendency towards all earlier decline of oestradiol in the ACTH group. Cortisol and progesterone Concentrations were significantly elevated during treatment in the ACTH group (p less than 0.001). with cortisol peak concentrations occurring between 40 and 80 min after each ACTH injection. Cortisol concentrations in saliva and Plasma were highly correlated (p less than 0.001). In conclusion, elevated cortisol concentrations from the onset of standing oestrus increase progesterone concentrations and prolong the interval between oestradiol and LH peaks to ovulation, the latter possible due to an early decline in oestradiol concentrations and a change of the LH peak outline. the effect these hormonal changes have on reproductive performance need to be further investigated.   • Genetic analysis of functional and non-functional teats in a population of Yorkshire pigs

    • Litter size is included in the goal of most pig breeding programmes. With an increased litter size, the number of functional teats, which are needed to achieve piglet weight gain and uniform litters,becomes increasingly important. The non-functional (e.g. inverted, blind,or small) teats might have a lower value for the nursing of the piglets. In Sweden, within the pig breeding company Nordic Genetics, the total number of teats, as well as the numbers of functional and non-functional teats, are recorded for purebred Yorkshire pigs both at 3 weeks of age (identity marking) and at about 5 months of age (performance testing). In 2010, 13% of the Swedish purebred Yorkshire pigs had at least one non-functional teat at performance testing (100 kg live weight). The aim of this study was to analyse both genetic and phenotypic variation among the teat traits and to analyse the associations among the teat traits,age and side fat thickness at 100 kg live weight. In the phenotypic analyses there were significant differences between males and females for all analysed teat traits. Males showed a higher number of functional teats and females three times more non-functional teats at 100kg live weight. In the genetic analyses there cords for males and females were treated as representing different traits. The estimated heritability of total number of teats and the number of functional teats, both at 3 weeks of age and at 100 kg live weight, was in the range 0.36-0.42. The heritability of the number of non-functional teats at 3 weeks of age was low, 0.02, for both males and females. Surprisingly, at 100 kg live weight, the heritability of non-functional teats was moderate,0.29, for females, but still low, 0.03, formales. Further studies are required to investigate whether there is a biological explanation for this finding, or  if there is a difference between  the genders in the recording procedure of this trait.The only  significant genetic correlation found between the teat traits and performance traits was for  male pigs; the genetic correlation for the relationship between the number of non- functional teats at 3 weeks of age and the age at 100 kg was 0.2. Thus, slowly growing pigs have a lower number of non-functional teats. Our results show that the teat number efficiently  can be improved by selection. However non-functional teats is a complex trait and  further studies need to be performed to investigate the possibilities to improve selection against the non-functional teats.

  • Identification of novel candidate genes for the inverted teat defect in sows using a genome-wide marker panel

    • The number of functional teats is an important selection criterion in pig breeding. Inherited defects of the udder, such as the inverted teat, do have a considerable negative impact on the nursing ability of the sow. To investigate the genetic background of this defect and the number of functional teats in Swedish maternal lines, samples from 230 Yorkshire pigs were selected for genotyping using the PorcineSNP60K BeadChip (Illumina Inc.), each pig with at least one inverted teat was matched with one non-affected pig (fullsib or pairs with matching herd and gender). A genome-wide association study on these 230 pigs was performed using the two-step approach implemented in GenABEL using 46,652 single nucleotide polymorphisms across all autosomes and the X chromosome. A number of significant regions were identified for the inverted teat defect on chromosomes 2, 10, and 18. Many of the regions associated with the number of functional teats were located in the same or close regions, except two associated markers on the X chromosome and one on chromosome 3. We identified some of the regions on chromosomes previously reported in one linkage and one gene expression study. We conclude, despite being able to suggest new candidate genes, that further studies are needed to better understand the biologic background of the teat development. Despite the in-depth comparison of identified regions for the inverted teat defect done here, more studies are required to allow a clear identification of genetic regions relevant for this defect across many pig populations.

    • In the modern pig industry, the increasing number of piglets born per litter augments the importance of the number of functional teats in the sow. The aim of this study was to evaluate the function and importance of inverted teats during nursing and to analyze structural and functional differences between the mammary glands of inverted teats versus normal teats. Nine farrowing gilts (8 purebred Swedish Yorkshire gilts and 1 cross between Swedish Yorkshire and Norwegian Landrace) and 94 piglets (59 piglets suckling normal teats, 32 piglets suckling protruded teats [i.e., previously inverted], 2 piglets suckling inverted teats, and 3 piglets suckling considerably smaller teats) were included in the study. Teat fidelity (keeping the same teat between the nursings) was registered, excluding the first 48 h postpartum. Piglet weight was recorded daily during the first week of life and thereafter once a week until weaning at 4 wk of age. Weight and growth rate were analyzed using repeated observation mixedmodel analysis of variance. The 2 piglets that suckled the inverted teats were not able to emerge the teats and they were euthanized 4 and 8 d after birth, respectively, due to loss of BW. The average weight at weaning (28 d of age) was 8.1 kg (range 3.2-13.8 kg). In the normal teats (n = 53), the weight of the corresponding mammary gland tissue at necropsy was positively correlated to the piglet average daily weight gain during wk 2 (r = 0.33, P < 0.05), 3 (r = 0.55, P < 0.001), and 4 (r = 0.47, P < 0.001). In the protruded teats (n = 32), the weight of the corresponding mammary gland tissue was positively correlated to the piglet average daily weight gain during wk 2 (r = 0.63, P < 0.001) and 3 (r = 0.43, P < 0.05). Among the piglets nursing normal teats, 82% kept fidelity to its teat and the corresponding percent for the protruded teats was 26%. In 7 of the 9 sows, the weaning weight of the piglets suckling protruded teats was numerically lower compared to the piglets suckling normal teats, although the difference was not statistically significant. Piglets nursing small teats had lower weaning weight (4.8 kg) and the corresponding mammary tissue also had lower weight (335 g). This study indicates the importance of an adequate number of well-developed and well-functioning teats in healthy sows for the growth and development of the piglets but also emphasizes the complexity of the interaction between the nursing sow and her piglets.

    • Since January 1 2013, group housing of sows has been compulsory within the European Union (EU) in all pig holdings with more than ten sows. Sows and gilts need to be kept in groups from 4 weeks after service to 1 week before the expected time of farrowing (Article 3(4) of Directive 2008/120/EC on the protection of pigs). The legislation regarding group housing was adopted already in 2001 and a long transitional period was allowed to give member states and producers enough time for adaptation. Even so, group housing of sows still seems to be uncommon in the EU, and is also uncommon in commercial pig farming systems in the rest of the world. In this review we share our experience of the Swedish 25 years of animal welfare legislation stipulating that sows must be loose-housed which de facto means group housed. The two most important concerns related to reproductive function among group-housed sows are the occurrence of lactational oestrus when sows are group-housed during lactation, and the stress that is associated with group housing during mating and gestation. Field and clinical observations in non-lactating, group-housed sows in Sweden suggest that by making basic facts known about the pig reproductive physiology related to mating, we might achieve application of efficient batch-wise breeding without pharmacological interventions. Group housing of lactating sows has some production disadvantages and somewhat lower productivity would likely have to be expected. Recordings of behavioural indicators in different housing systems suggest a lower welfare level in stalled animals compared with group-housed ones. However, there are no consistent effects on the reproductive performance associated with different housing systems. Experimental studies suggest that the most sensitive period, regarding disturbance of reproductive functions by external stressors, is the time around oestrus. We conclude that by keeping sows according to the pig welfare-friendly Directive 2008/120/EC, it is possible to combine group-housing of sows with good reproductive performance and productivity. However, substantially increased research and development is needed to optimize these systems.

    • This study investigated the relationship of the health of the newborn foal and (1) number of polymorphonuclear leukocytes (PMNLs) in the amniotic fluid, (2) bacteria present in the amniotic fluid and the venous umbilical blood, and (3) bacteria present in the uterus of the newly foaled mare. A further aim was to investigate relationships between the bacteriologic findings in the amniotic fluid, umbilical blood, and uterus postpartum. Samples were taken from 50 Standardbred trotter foaling mares from a well-managed stud in Sweden. Parturition was spontaneous in all cases. Length of pregnancy, parturition and postpartum complications, health status of the foal, the time between foaling and the expulsion of the placenta, and the number of postfoaling mares becoming pregnant after insemination were recorded. Amniotic fluid was collected when the amniotic vesicle was clearly visible; it was analyzed for bacteriology and occurrence of PMNLs. Umbilical blood was analyzed for the presence of bacteria and the concentration of serum amyloid A. The uterus of themarewas swabbed for bacteriology 6 to 17 hours postpartum. A blood samplewas taken from the foal before administering plasma. The foals were divided into two groups: group 1 required up to 2 hours to rise after birth (2 hours; 31 foals) and group 2 requiredmore than two hours (>2 hours; 19 foals). The length of gestation varied between 332 and 356 days; there was no significant difference in gestation length between the two foal groups. Partus and postpartum complications occurred in a significantly higher proportion of mares giving birth to group 2 foals than group 1 foals (P ¼ 0.02), although uterine culture postpartum and the subsequent pregnancy rate per season were not different between the groups. Compromised health status was significantly higher among foals belonging to group 2 than group 1 (P ¼ 0.001). Most of the amniotic samples contained 5% or less PMNLs. Only three samples contained more than 30% PMNLs; group 2 foals had the highest percentage of PMNLs. Bacterial growth was found in both amniotic fluid (57%) and umbilical blood (35%) in mares irrespective of whether their foals were healthy or compromised. Coagulase-negative staphylococci were the most frequent bacteria. There were no differences in bacterial occurrence in amniotic fluid or in umbilical blood between the two foal groups

  • Effect of different freezing rates and thawing temperatures on cryosurvival of dromedary camel spermatozoa

    • The objective of this study was to evaluate the effect of different freezing rates and thawing temperatures on the post-thaw quality of camel spermatozoa. Ten ejaculates from five male camels were frozen at five different freezing rates, achieved by placing the straws at specific heights above the surface of liquid nitrogen for different lengths of time (4 cm for 15 min; 1 cm for 15 min; 7 cm for 15 min; 7 cm for 5 min + 4 cm for 3 min; 4 cm for 5 min + 1 cm for 3 min) followed by storage in liquid nitrogen. Two thawing temperatures (37 degrees for 30 s and 60 degrees C for 10 s) were subsequently tested. Post-thawing, the samples were evaluated for total and progressive motility, kinematics, membrane and acrosome integrity, and membrane functionality (hypoosmotic swelling test) at zero and 1 h post thawing. Total and progressive motility were significantly higher for the fastest freezing rate (at 1 cm) at 0 h (p < 0.01 for both), as were VCL (p < 0.01), VSL (p < 0.05) and STR (p < 0.05). Freezing at 4 cm produced the lowest values of STR compared to other treatments (p < 0.05). At 1 h, no differences in total motility were observed between freezing at 4 cm and 1 cm, both being significantly better than freezing rate 7 cm + 4 cm (p < 0.01). For progressive motility and VSL, only freezing at 1 cm was superior to the 7 cm + 4 cm combination (p < 0.001 and p < 0.05 respectively). Membrane integrity at 1 h was higher for freezing at 7 cm than at 1 cm (p < 0.01). For thawing temperatures, total motility and progressive motility at 0 h and 1 h (p < 0.001), and acrosome integrity at 1 h (p < 0.01) were higher for 60 degrees C thawing temperature than 37 degrees C. The kinematics VCL (p < 0.001), VSL and STR (p < 0.01), and VAP (p < 0.05) showed higher values for 60 degrees C thawing temperature than 37 degrees C at 0 h. After 1 h, higher values for VSL, VCL and VAP (p < 0.05) were observed for 60 degrees C than for 37 degrees C. In conclusion, a fast freezing rate would probably be beneficial for camel semen, and thawing should be conducted at 60 degrees C. (C) 2018 Published by Elsevier Inc.

    • The aim of this study was to estimate genetic parameters for longevity from Swedish crossbred sows to investigate the possibilities of selecting for this trait. Data were collected from 16 commercial piglet-producing herds, on crossbred (Landrace x Yorkshire) sows farrowing in the period 1 January 2001 to 31 December 2004. The data set with records on 10373 sows was split into two sets according to the breed of the sire, i.e. Landrace sires (LS) or Yorkshire sires (YS). Removal hazard during productive life (PL) was analysed with survival analysis, using a sire model. Stayability from first to second litter (STAY12), stayability from first to third litter (STAY13), length of productive life (LPL) and lifetime production (LTP) were analysed with linear models, using an animal model. Females after the worst sire had 1.7 times higher (progeny of LS) and 2.4 times higher (progeny of YS) risk of removal than females after the best sire. Heritability for PL was estimated at 0.06 (LS) and 0.12 (YS). The heritabilities for the linear longevity traits ranged from 0.03 to 0.08. Genetic correlations between the four linear longevity traits were all high and positive (0.6 to 1.0), as were the phenotypic correlations (0.5 to 0.8). The correlations (Spearman rank) between the sire's estimated breeding values for all the five longevity traits were all significant (P < 0.001) and moderate to strong in both data sets. Estimated breeding value (EBV) correlations between the five longevity traits and traits included in the present Swedish breeding evaluation (Quality Genetics (QG)) were significant in a few cases. Significant and favourable EBV correlations were found between age at first farrowing and both STAY12 and STAY13 (-0.20 and -0.31), as well as between litter weight at 3 weeks and LPL and UP (0.13 to 0.20). Significant and unfavourable EBV correlations were found between age at 100 kg and STAY12 (0.32), as well as between the exterior conformation score from testing station and PL (-0.20). The level of the estimated heritabilities for longevity indicates that genetic improvement of sow longevity would be possible. However, overall, there was no strong indirect selection for sow longevity with the current Swedish breeding evaluation (QG).

  • Effects of free-range and confined housing on joint health in a herd of fattening pigs

    • Background: Free-range housing, in which pigs have access to both indoor and outdoor areas, is mandatory in organic pig production in Europe, but little is known about the effects of this housing on joint health in pigs. A high level of joint condemnations at slaughter has been reported in organic free-range pigs in Sweden, compared with pigs raised in conventional confined housing. We hypothesised that biomechanical forces imposed on the joints of pigs that range freely promote the development of osteochondrosis and lead to joint condemnation. We compared the prevalence of osteochondrosis and other joint lesions (e.g. arthritis, traumatic) in the elbow and hock joints of 91 crossbred Hampshire (Yorkshire x Landrace) fattening pigs that were housed in a free-range indoor/outdoor system with that in 45 pigs housed in confined indoor pens.Results: A larger proportion of free-range than confined pigs had osteochondrosis in the elbow joints (69 vs. 50%, p < 0.05), and a higher proportion of these joints in free-range pigs showed moderate or severe lesions (33 vs. 16%, p < 0.05). The free-range pigs also showed a higher prevalence of osteochondrosis in the hock joints (83 vs. 62%, p < 0.05) and a larger proportion of these joints had moderate or severe lesions (69 vs. 33%, p < 0.001). At slaughter, 4.2% of the free-range pigs had condemned joints, all of which showed severe osteochondrosis, while no joints of confined pigs were condemned.Conclusions: In this experiment the prevalence of osteochondrosis in the elbow and the hock was higher, and lesions were more severe, in free-range than in confined pigs, suggesting that free-range housing increases the risk of acquiring osteochondrosis. Increased biomechanical stress to vulnerable joint structures may be the mechanism behind this effect, however more studies are needed to verify these results. This study suggests that modification of housing, and breeding for joints that are more adapted to free-range movement may be needed in free-range pig production. Severe osteochondrosis is a cause of joint condemnation, but the condemnation rate at slaughter underestimates the actual frequency of joint lesions and hence is a poor assessment of joint health.

A numerically positive correlation (0.8) between additive genetic breeding values for mean and for variance was found, but because of the low heritability for residual variance, the variance will increase very slowly with the mean.

    • Background Surgical castration in male piglets is painful and methods that reduce this pain are requested. This study evaluated the effect of local anaesthesia and analgesia on vocal, physiological and behavioural responses during and after castration. A second purpose was to evaluate if herdsmen can effectively administer anaesthesia. Methods Four male piglets in each of 141 litters in five herds were randomly assigned to one of four treatments: castration without local anaesthesia or analgesia (C, controls), analgesia (M, meloxicam), local anaesthesia (L, lidocaine), or both local anaesthesia and analgesia (LM). Lidocaine (L, LM) was injected at least three minutes before castration and meloxicam (M, LM) was injected after castration. During castration, vocalisation was measured and resistance movements judged. Behaviour observations were carried out on the castration day and the following day. The day after castration, castration wounds were ranked, ear and skin temperature was measured, and blood samples were collected for analysis of acute phase protein Serum Amyloid A concentration (SAA). Piglets were weighed on the castration day and at three weeks of age. Sickness treatments and mortality were recorded until three weeks of age. Results Piglets castrated with lidocaine produced calls with lower intensity (p < 0.001) and less resistance movements (p < 0.001) during castration. Piglets that were given meloxicam displayed less pain-related behaviour (huddled up, spasms, rump-scratching, stiffness and prostrated) on both the castration day (p = 0.06, n.s.) and the following day (p = 0.02). Controls had less swollen wounds compared to piglets assigned to treatments M, L and LM (p < 0.001). The proportion of piglets with high SAA concentration (over threshold values 200, 400 mg/l) was higher (p = 0.005; p = 0.05) for C + L compared to M + LM. Ear temperature was higher (p < 0.01) for controls compared to L and LM. There were no significant treatment effects for skin temperature, weight gain, sickness treatments or mortality. Conclusions The study concludes that lidocaine reduced pain during castration and that meloxicam reduced pain after castration. The study also concludes that the herdsmen were able to administer local anaesthesia effectively

This study aims at studying genetic correlations between leg conformation traits scored in young Yorkshire pigs in nucleus herds and longevity traits of purebred Yorkshire sows in multiplier herds. Two datasets were available: 1) on 97,533 animals (both sexes) with information on movement and overall leg score recorded at performance testing and, 2) on 12,444 sows with information on longevity. The longevity traits were: stayability to survive up to second or third parity (STAY12/STAY13); length of productive life (LPL); number of litters (NoL); lifetime born alive (LBA) and lifetime total born (LTB). The estimated heritabilities ranged from 0.05 to 0.16. Almost all estimated genetic correlations between conformation and longevity traits were significantly favorable (better scores - better longevity). Movement showed higher correlations with longevity (0.36-0.53) compared with overall leg score (0.24- 0.33).

    • Longevity is important in pig production with respect to both economic and ethical aspects. Direct selection for longevity might be ineffective because 'true' longevity can only be recorded when a sow has been culled or died. Thus, indirect selection for longevity using information from other traits that can be recorded early in life and are genetically correlated with longevity might be an alternative. Leg conformation has been included in many breeding schemes for a number of years. However, proving that leg conformation traits are good early indicators for longevity still remains. Our aim was to study genetic associations between leg conformation traits of young (5 months; 100 kg) Swedish Yorkshire pigs in nucleus herds and longevity traits of sows in nucleus and multiplier herds. Data included 97 533 animals with information on conformation (Movement and Overall score) recorded at performance testing and 26 962 sows with information on longevity. The longevity traits were as follows: stayability from 1st to 2nd parity, lifetime number of litters and lifetime number of born alive piglets. Genetic analyses were performed with both linear models using REML and linear-threshold models using Bayesian methods. Heritabilities estimated using the Bayesian method were higher than those estimated using REML, ranging from 0.10 to 0.24 and 0.07 to 0.20, respectively. All estimated genetic correlations between conformation and longevity traits were significant and favourable. Heritabilities and genetic correlations between conformation and longevity indicate that selection on leg conformation should improve sow longevity.

    • Lameness is an issue of concern in pig production due both to animal welfare and to economical aspects. Lame sows are believed to suffer from pain and stress which is re ported to have a negative influence on reproduction. Leg conformation and locomotion traits in young animals are associated with the risk of lameness at higher age. The purpose of this study was to estimate the genetic parameters of leg conformation traits recorded at performance testing (around 5 months of age) and their genetic correlations with reproduction traits. Information on leg conformation traits from 123,307pigs scored and on reproduction traits from 22,204 litters in the first and second parity from Swedish Yorkshire nucleus herds were available for genetic analysis. Eight conformation and locomotion traits, coming from the old or the new scoring system in Sweden, included old movement, old overall leg score, new movement, new toes quality, new front leg quality, new rear leg quality, standing-under-position syndrome and new overall score. Four reproduction traits were analyzed by parity: the number of total born piglets, the number of liveborn piglets, the number of stillborn piglets and weaning to service interval. Estimates of heritabilities and genetic correlations between traits were obtained using a multi-trait linear animal mixed model. The heritability estimates were low to moderate, ranging from 0.02 to 0.20 for conformation traits and from 0.06 to 0.10 for reproduction traits.Significant genetic correlations were found between new toes quality and new overall score and the number of liveborn piglets in the 1st parity (-0.35 and -0.31, respectively), indicating that sows with even toes and better overall leg score tend to have higher number of liveborn piglets. Old movement score showed significant correlations with number of total born and number of liveborn piglets in both parities (0.20 to 0.36) and with weaning to service interval in the 2nd parity (-0.357±0.11). Similarly, standing-under-position syndrome was highly associated with number of total born and number of  liveborn piglets in both parities (-0.54 to -0.35), indicating that sows with better movement and not suffering from standing-under-position syndrome are likely to have larger litter size and shorter interval to return heat after weaning. Heritabilities and significantly favorable genetic correlation estimates suggest the possibility of simultaneous improvement of both leg quality and reproduction performance  by selecting on sound leg conformation and locomotion of young pigs.

    • Selection for sow longevity using information from traits, which are expressed in early life and genetically highly related to longevity, is expected to be more effective than direct selection as it can overcome the disadvantage of late recording of true longevity. Our aim was to investigate the correlation between leg conformation recorded on young pigs, litter size at first parity and longevity of Danish Landrace and Yorkshire sows. Information on conformation from 116,733 Landrace and 89,963 Yorkshire pigs and information on reproduction and longevity from 27,070 Landrace and 11,895 Yorkshire sows were analyzed. All considered traits were low to moderately heritable, ranging from 0.02 to 0.41. In general, both conformation and reproduction traits were favorably genetically correlated with longevity (0.07-0.39 and 0.00-0.58, respectively). These estimates suggest a potential of improving sow longevity by selection on conformation recorded at young age and litter size at first parity.

    • Leg weakness is one of the major reasons for culling of sows. Improvement of leg conformation is expected to improve both animal welfare and producers' profit. We summarize here the possibilities to breed for better legs/ gaits and the potential influence on reproduction and stayability, based on data on Landrace and Yorkshire sows in Sweden and Denmark. Purebred young pigs were scored for leg conformation traits at performance testing (5 months of age) in our studies. Leg conformation traits were analyzed together with litter size in parities 1 and 2 and stayability traits (culled or not culled after parity 1 or parity 2), recorded on purebred sows in nucleus and multiplier herds. Genetic (co)variances between traits were estimated. We estimated heritabilities for leg conformation traits ranging from 0.02 to 0.20 depending on breed and population. Estimated genetic correlations between leg conformation and litter size were favorable for Swedish Yorkshire (+0.15 to +0.42), but unfavorable in the Danish populations (-0.17 to -0.46). Estimated correlations between leg conformation and stayability were favorable in both Swedish Yorkshire and the Danish populations (0.12 to 0.42). Our results indicate that leg conformation can be improved by selection. Indirect selection for improved stayability based on leg conformation traits would be possible but influenced by the size of the genetic correlations between traits. Our ongoing project on genome wide association study using high density SNP chip is expected to identify chromosome regions associated with leg traits in three Danish pig populations (Landrace, Yorkshire and Duroc). Results from this study could be used as prior information in genomic selection, helping to more effectively improve leg conformation and accordingly improve sow longevity.

  • Characterization of pig production in Gulu and Soroti districts in northern and eastern Uganda

    • Owing to loss of cattle and goats during the recent civil unrest, pig farming has become popular in northern and eastern Uganda as a quick mitigation to poverty. This study was carried out to describe the characteristics of pig production in these regions. Using a cross-sectional design, data were collected from 96 households (51 from Gulu district, northern Uganda and 45 from Soroti district, eastern Uganda) raising pigs with suckling and weaned piglets. The households were selected using the snowballing method. The households were predominantly headed by adult men, of which 97% had attended at least primary education. The mean numbers of suckling, weaned, growing and adult pigs per household in Gulu and Soroti were 8 and 7.3, 4.8 and 5.3, 2.3 and 2.6 and 3.1 and 3.1, respectively, with no significant difference (p > 0.05) between the districts. Overall, the majority of households (64%, n=96) kept local breeds of pigs. The most common management method was tethering, as practiced by 67% of the study households. Home-made feeds were the most common (88%, n=96) and the wives/women provided much of the labour (60%, n=96). However, in only 23% of the households, women owned and made decisions on the pigs. Male children also owned and made decisions on pigs in 8% of the households. Natural breeding of sows with a shared boar was the most common practice (88%, n=96). The litter size at birth was 6 to 10 piglets in 78% (n= 96) of the households. Based on the clinical signs reported by the households, 38% and 23% of the herds experienced problems with diarrhoea and respiratory diseases, respectively. In total, 39% of the households were receiving professional veterinary care, when pigs fall sick. In conclusion, pig production around regional urban centers in northern and eastern Uganda is largely smallholder, practiced by farmers who have attended at least primary education, tether their pigs, depend on labour provided largely by housewives and there is inadequate veterinary care. The findings of this study point at a need for increased involvement of women in decision making in pig farming, increased pig veterinary care and investigation of the causes of diseases such as diarrhoea in order to support this major livelihood resource for the poor in northern and eastern Uganda, especially women and children.

    • Non-typhoidal salmonellosis is of concern in humans in sub-Saharan Africa, and this is partly due to the high number of immunocompromised persons. Pork and pork products could be among the sources of these non-typhi Salmonella spp. The aim of this study was to identify Salmonella spp. in piglets and weaners in northern and eastern Uganda, characterize their antimicrobial resistance patterns and determine herd-level risk factors. Fecal samples were collected from 465 piglets and weaners from 93 herds (49 and 44 from northern and eastern Uganda, respectively). In addition, information about the herd management and potential risk factors were collected. The fecal samples were cultured for the identification of Salmonella spp. The Salmonella spp. confirmed by serotyping were further characterized by determination of minimum inhibitory concentration (MIC) to 12 antimicrobials by broth microdilution. At individual level, the total prevalence of Salmonella spp. was 12% (12.2% in northern and 11.9% in eastern Uganda). At herd level, the total prevalence was 39% (43% in northern and 34% in eastern Uganda). From 56 samples with Salmonella spp., 20 serovars were identified including two serovars identified only by their antigenic formulae. The predominant serovars were S. Zanzibar, S. Heidelberg, S. Infantis, S. Typhimurium, S. Stanleyville, S. Aberdeen and S. Kampala. In total, 57% of the 53 Salmonella spp. analyzed, originating from 27% of the herds, were resistant to at least one antimicrobial agent. The majority of drug-resistant isolates (60%) were from northern Uganda. Eight multidrug-resistant (MDR) isolates were from northern Uganda and three MDR isolates were from eastern Uganda. Increased prevalence of Salmonella spp. was associated with feeding the young and adults separately as compared to feeding the young and adults together (p = 0.043, OR = 4.3; 95% CI 1.1, 17.38). Protective factors were "intensive" method of keeping the pigs versus "tethering and roaming" (p = 0.016, OR = 0.11; 95% CI 0.02, 0.64), "intensive" method versus "semi-intensive" method (p = 0.048, OR = 0.12; 95% CI 0.01, 0.96) and cleaning feeders after every two days versus daily (p = 0.017, OR = 0.18; 95% CI 0.05, 0.72). This study has revealed a high prevalence of infection of piglets and weaners with diverse non-typhi Salmonella serovars and highlights the potential role of pork and pork products as sources of these organisms for humans. In addition, this study has identified protective factors that could be promoted to control Salmonella spp. and in antimicrobial resistance reduction programs in rural pigs from Uganda.   • A Pilot Study in Sweden on Efficacy of Benzylpenicillin, Oxytetracycline, and Florfenicol in Treatment of Acute Undifferentiated Respiratory Disease in Calves

    • Bovine respiratory disease (BRD) is a major indication for antibiotic treatment of cattle worldwide and some of the antibiotics used belong to classes of highest priority among those listed by WHO as critically important for human medicine. To preserve the efficacy of "newer" antibiotics, it has been suggested that "older" drugs should be revisited and used when possible. In this pilot study, we evaluated the efficacy of benzylpenicillin (PEN), oxytetracycline (OTC), and florfenicol (FLO) for treatment of naturally occurring BRD on two farms raising calves for slaughter. Farm personnel selected calves for enrolment, assigned calves to one of the three regimens in a systematically random manner, treated the calves, and registered the results. Overall, 117 calves were enrolled in the study. Nineteen calves relapsed in BRD before slaughter and were retreated (16.2%) and three died (2.6%). For PEN, treatment response rates after 30 days, 60 days, and until slaughter were 90.2%, 87.8%, and 80.5%, respectively; for OTC, 90.0%, 85.0%, and 85.0%, respectively; and for FLO, 86.1%, 83.3%, and 77.8%, respectively. There were no statistically significant differences in relapse, mortality, or response rates between the three treatment regimens. This indicates that PEN, OTC, and FLO were equally effective for treatment of BRD but the results need to be confirmed in a more elaborate study with a higher statistical power. The findings support the current recommendations from the Swedish Veterinary Association and the Medical Products Agency to use benzylpenicillin as a first line antibiotic for treatment of calves with undifferentiated respiratory disease in Sweden. Due to differences in the panorama of infectious agents and presence of acquired antibiotic resistance, the findings might not be applicable in other geographical areas.
