  • The Global Museum: natural history collections and the future of evolutionary science and public education

    • Natural history museums are unique spaces for interdisciplinary research and educational innovation. Through extensive exhibits and public programming and by hosting rich communities of amateurs, students, and researchers at all stages of their careers, they can provide a place-based window to focus on integration of science and discovery, as well as a locus for community engagement. At the same time, like a synthesis radio telescope, when joined together through emerging digital resources, the global community of museums (the 'Global Museum') is more than the sum of its parts, allowing insights and answers to diverse biological, environmental, and societal questions at the global scale, across eons of time, and spanning vast diversity across the Tree of Life. We argue that, whereas natural history collections and museums began with a focus on describing the diversity and peculiarities of species on Earth, they are now increasingly leveraged in new ways that significantly expand their impact and relevance. These new directions include the possibility to ask new, often interdisciplinary questions in basic and applied science, such as in biomimetic design, and by contributing to solutions to climate change, global health and food security challenges. As institutions, they have long been incubators for cutting-edge research in biology while simultaneously providing core infrastructure for research on present and future societal needs. Here we explore how the intersection between pressing issues in environmental and human health and rapid technological innovation have reinforced the relevance of museum collections. We do this by providing examples as food for thought for both the broader academic community and museum scientists on the evolving role of museums. We also identify challenges to the realization of the full potential of natural history collections and the Global Museum to science and society and discuss the critical need to grow these collections. We then focus on mapping and modelling of museum data (including place-based approaches and discovery), and explore the main projects, platforms and databases enabling this growth. Finally, we aim to improve relevant protocols for the long-term storage of specimens and tissues, ensuring proper connection with tomorrow's technologies and hence further increasing the relevance of natural history museums.

    • Single specimens of the Chinese mitten crab, Eriocheir sinensis H. Milne Edwards, 1853, have been regularly reported along the western and eastern coasts of Sweden since the 1930´s. The crab has most likely been brought from overseas via the transfer of ship’s ballast water or secondarily introduced from its key European distribution areas. Since 2001 a sharp increase in the occurrence of the mitten crab has been noticed in Swedish inland waters, but the dispersal routes and distribution of the species into Sweden remain poorly known. Here we document the current and historical distribution of the Chinese mitten crab in Sweden and assess possible invasion routes. A special focus is put on the historical occurrence of crabs in Lake Vänern, which empties into the Skagerrak/North Sea and Lake Mälaren, which connects to the Baltic Sea. The existing time series available for the mitten crab from the two lakes shows a large variation in dispersal pattern between different areas within each lake. In order to detect and monitor outbreaks of the mitten crab in Sweden, an internet based reporting system was created in 2007. Museum collections and reports from the general public throughout Sweden were compared with the known occurrence of mitten crabs in Lake Vänern and Lake Mälaren. During the study, the web-based reporting system was not advertised and the general public did not get paid for reporting the information. Population densities of the crab occurred “in peaks” and unevenly over the last decade in both lakes, suggesting a pulse invasion instead of a constant supply by migration. Significant difference in male and female occurrence together with few transport possibilities between lakes suggests different invasion routes for the crabs. Reports from the general public throughout Sweden coincide with the observations from local fishermen in Lake Mälaren and Lake Vänern. Data on occurrence and abundance are discussed in relation to a potential reproduction area on the western coast of Sweden.

    • Sperm structure of the Xenacoelomorpha (Acoelomorpha plus Xenoturbellida) is updated in the light of new discoveries or new interpretations of existing data. Nemertodermatida and Acoela (Acoelomorpha) have introsperm with certain basic features in common, but all acoels lack acrosomes and usually have two flagella with unusual combinations of microtubules, whereas all nemertodermatids have small, simple acrosomes and a typical 9+2 flagellum. Xenoturbellida is currently considered as the sister taxon to Acoelomorpha. Xenoturbella bocki has an aquasperm that has almost nothing in common with the sperm of Acoelomorpha. We argue that the aquasperm ultrastructure of X. bocki has much in common with sperm of hemichordates and to some extent echinoderms, which was previously disputed. Molecular analyses have on the one hand supported a connection with deuterostomes but on the other hand have negated it, suggesting that the closest common ancestor of Xenacoelomorpha is either the Nephrozoa, Deuterostomia or Protostomia. Sperm structure is highly diverse among Xenacoelomorpha, with protostome-like traits in Acoelomorpha and deuterostome-like traits in Xenoturbella. Assuming Xenacoelomorph monophyly and ancestral introsperm in this taxon, however, suggests that the re-expression of the aquasperm form of Xenoturbella, involving some key changes in sperm morphology, is a secondarily derived state that could have occurred through progenetic spermiogenesis with the precocious development of round spermatids to maturity.

    • By applying morphological and molecular data on two genera of the nudibranch molluscs it is shown that the tension between taxonomic practice and evolutionary processes persists. A review of the related genera Catriona and Tenellia is used to demonstrate that the fine-scale taxonomic differentiation is an important tool in the integration of morphological and molecular data. This is highlighted by the hidden species problem and provides strong argument that the genus must be kept as a maximally narrowly-defined entity. Otherwise, we are forced to compare a highly disparate species under the putatively lumped name "Tenellia". We demonstrate this in the present study by applying a suite of delimitation methods and describing a new species of Tenellia from the Baltic Sea. The new species possesses fine-scale morphological distinguishing features, which were not investigated before. The true, narrowly defined genus Tenellia represents a peculiar taxon with a clearly expressed paedomorphic characters and predominantly brackish-water habitats. The phylogenetically related genus Catriona, of which three new species are described here, clearly demonstrates different features. A lumping decision to call many morphologically and evolutionary different taxa as "Tenellia" will downgrade the taxonomic and phylogenetic resolution of the entire family Trinchesiidae to just a single genus. The dissolution of the dilemma of "lumpers & splitters", which still significantly affects taxonomy, will further help to make systematics a true evolutionary discipline.

    • The nudibranch genus Doto is taxonomically problematic in particular, and some species are described on the notion of strict monophagy. Here we perform species delimitation on NE Atlantic species, as well as placing them phylogenetically, using two markers: the mitochondrial COI and the nuclear H3. We also study the morphology of the species including radular ultrastructure and review food specificity. Specimens were first divided into potential species using ABDG on both markers, these groups were used as input species for species delimitation analyses using BPP, and analyses were performed with both markers combined and on H3 only. The analyses delimit 11 and eight species, respectively. With the exception of one species for which only COI was available, the differences are found in D. fragilis, which is split into three groups when COI is included and lumped into one with only H3. Doto hystrix is nested within these groups. We also found that specimens from Sweden seemingly close to D. maculata in external morphology have identical sequences as D. coronata. Analysis of food preferences of the species involved in the study contradicts the notion of strict monophagy within Doto.

    • © 2017 The Author(s) Studies of cumulative and long-term effects of human activities in the ocean are essential for developing realistic conservation targets. Here, we report the results of a recent national marine biodiversity inventory along the Swedish West coast between 2004 and 2009. The expedition revisited many historical localities that have been sampled with the same methods in the early twentieth century. We generated comparable datasets from our own investigation and the historical data to compare species richness, abundance, and geographic distribution of diversity. Our analysis indicates that the benthic ecosystems in the region have lost a large part of its original species richness over the last seven decades. We find evidence that especially rare species have disappeared. This process has caused a more homogenized community structure in the region and diminished historical biodiversity hotspots. We argue that the contemporary lack of rare species in the benthic ecosystems of the Kattegat and Skagerrak offers less opportunity to respond to environmental perturbations in the future and suggest improving the poor representation of rare species in the region. The study shows the value of biodiversity inventories as well as natural history collections in investigations of accumulated effects of anthropogenic activities and for re-establishing species-rich, productive, and resilient ecosystems.

    • The pteropod Limacina lesueurii (d'Orbigny 1836) is a subtropical species, which in the Atlantic Ocean shows maximum abundance between 30°N and 25°S. In the North Sea, this species has so far only been occasionally reported from the coastal waters off the northern Scotland and in the English Channel. In this short note, we for the first time report presence of L. lesueurii in two sediment cores taken in the Kosterfjord (Skagerrak, North Sea). The pteropod, generally absent or occurring in low numbers throughout both cores, showed distinctly higher abundances between 100 and 130 cm core depth, which based on dating by 137Cs, 210Pb and lead pollution records corresponds to ~1920–1950s. During this period positive sea surface temperature anomalies were reported in the North Atlantic and number of oceanic water inflows have been documented for the North Sea. Some of these inflows were accompanied by ”enormous shoals” of L. lesueurii observed in the English Channel and NE of Scotland. We hypothesize that L. lesueurii was transported into the Skagerrak in connection with these and propose this pteropod species as a new valuable stratigraphic marker for oceanic water inflows in the study area.

  • Phylogeny and Taxonomy of Childia (Acoela) : New characters for unraveling acoel phylogenies from molecules, ultrastructure, immunocytochemistry and confocal microscopy

    • This thesis presents a comprehensive phylogenetic and taxonomic study of an acoel subgroup, the Childiidae. Members of this taxon are characterized by well-developed male copulatory organs with conical/cylindrical stylets. The phylogenetic analyses, by means of total evidence approach, based on three molecular markers (18S and 28S rRNA genes and Histone H3) and 50 morphological characters reaffirm the non-monophyly of the Childiidae sensu Dörjes, 1968 (Actinoposthiidae and Childia+Paraphanostoma). The total evidence phylogeny strongly support the Childia+Paraphanostoma clade separate from other former members of the Childiidae, which are now placed in Actinoposthiidae. The monophyly of Childia+Paraphanostoma is well corroborated by several morphological characters. A new taxon Childia is defined, in accordance with the PhyloCode, comprising all former Paraphanostoma species and a member of the monotypic genus C. groenlandica. A new diagnosis for the current members of Childia is provided.Several structures, shown to hold promising phylogenetic signals for unraveling acoel relationships, such as musculature pattern, sperm and male copulatory organs, are investigated, using a combination of traditional and modern techniques (ultrastructure, immunocytochemistry and confocal microscopy), with main focus on Childia and its closest relatives. New characters are described and their phylogenetic significance assessed. Morphological characters relating to body-wall musculature, statocyst muscles, male copulatory organ musculature and ultrastructure, and sperm cytoplasmic granules are shown to carry important phylogenetic signals at lower taxonomic levels, while most of the characters related to sperm ultrastructure are useful at higher taxonomic levels within the Acoela. The data obtained undermine the phylogenetic use of the seminal bursa in Childia. In addition to this, it is shown that most of the classical morphological characters used in acoel taxonomy, obtained using traditional histological methods, may be misleading in identifying monophyletic entities within the Acoela.The most corroborated synapomorphies, identified in this thesis, are used in determining the taxonomic placement of a new viviparous acoel, Childia vivipara, into the taxon Childia.

  • Magnetic resonance imaging for preoperative staging of rectal cancer in clinical practice : high accuracy in predicting circumferential margin with clinical benefit

    • Objective: The aims were to determine agreement between staging of rectal cancer made by magnetic resonance imaging (MRI) and histopathological examination and the influence of MRI on choice of radiotherapy (RT) and surgical procedure. Method: In this retrospective audit, preoperative MRI was performed on 91 patients who underwent bowel resection, with 93% having total mesorectal excision. Tumour stage according to mural penetration, nodal status and circumferential resection margin (mCRM) involvement was assessed and compared with histopathology. Results: Five radiologists interpreted the images. Overall agreement between MRI and histopathology for T stage was 66%. The greatest difficulty was in distinguishing between T1, T2 and minimal T3 tumours. The accuracy for mCRM (MRI) was 86% (78/91),with an interobserver variation between 80% and 100%. In the 13 cases with no agreement between mCRM and pCRM (pathological), seven had long-term RT and nine en bloc resections, indicating that the margins initially were involved with an even higher accuracy for mCRM. Preoperative short-term RT was routine, but based on MRI findings, choice of RT was affected in 29 cases (32%); 17 patients had no RT and 12 long-term RT. The surgical procedure was affected in 17 cases (19%) with planned perirectal en bloc resections in all. CRM was involved (≤ 1 mm) in 14.7% of the 34 cases in which MRI had an effect upon choice of RT and/or surgery compared with 8.8% of the remaining 57 cases where it had no impact. Conclusion: Magnetic resonance imaging predicted CRM with high accuracy in rectal cancer. MRI could be used as a clinical guidance with high reliability as indicated by the low figures of histopathologically involved CRM.
