    • The Global Consortium for the Classification of Fungi and fungus-like taxa is an international initiative of more than 550 mycologists to develop an electronic structure for the classification of these organisms. The members of the Consortium originate from 55 countries/regions worldwide, from a wide range of disciplines, and include senior, mid-career and early-career mycologists and plant pathologists. The Consortium will publish a biannual update of the Outline of Fungi and fungus-like taxa, to act as an international scheme for other scientists. Notes on all newly published taxa at or above the level of species will be prepared and published online on the Outline of Fungi website (https://www.outlineoffungi.org/), and these will be finally published in the biannual edition of the Outline of Fungi and fungus-like taxa. Comments on recent important taxonomic opinions on controversial topics will be included in the biannual outline. For example, 'to promote a more stable taxonomy in Fusarium given the divergences over its generic delimitation', or 'are there too many genera in the Boletales?' and even more importantly, 'what should be done with the tremendously diverse 'dark fungal taxa?' There are undeniable differences in mycologists' perceptions and opinions regarding species classification as well as the establishment of new species. Given the pluralistic nature of fungal taxonomy and its implications for species concepts and the nature of species, this consortium aims to provide a platform to better refine and stabilise fungal classification, taking into consideration views from different parties. In the future, a confidential voting system will be set up to gauge the opinions of all mycologists in the Consortium on important topics. The results of such surveys will be presented to the International Commission on the Taxonomy of Fungi (ICTF) and the Nomenclature Committee for Fungi (NCF) with opinions and percentages of votes for and against. Criticisms based on scientific evidence with regards to nomenclature, classifications, and taxonomic concepts will be welcomed, and any recommendations on specific taxonomic issues will also be encouraged; however, we will encourage professionally and ethically responsible criticisms of others' work. This biannual ongoing project will provide an outlet for advances in various topics of fungal classification, nomenclature, and taxonomic concepts and lead to a community-agreed classification scheme for the fungi and fungus-like taxa. Interested parties should contact the lead author if they would like to be involved in future outlines.

  • Effects of tree pollen on throughfall element fluxes in European forests

    • The effects of tree pollen on precipitation chemistry are not fully understood and this can lead to misinterpretations of element deposition in European forests. We investigated the relationship between forest throughfall (TF) element fluxes and the Seasonal Pollen Integral (SPIn) using linear mixed-effects modelling (LME). TF was measured in 1990-2018 during the main pollen season (MPS, arbitrary two months) in 61 managed, mostly pure, even-aged Fagus, Quercus, Pinus, and Picea stands which are part of the ICP Forests Level II network. The SPIn for the dominant tree genus was observed at 56 aerobiological monitoring stations in nearby cities. The net contribution of pollen was estimated as the TF flux in the MPS minus the fluxes in the preceding and succeeding months. In stands of Fagus and Picea, two genera that do not form large amounts of flowers every year, TF fluxes of potassium (K+), ammonium-nitrogen (NH4+-N), dissolved organic carbon (DOC), and dissolved organic nitrogen (DON) showed a positive relationship with SPIn. However- for Fagus- a negative relationship was found between TF nitrate-nitrogen (NO3--N) fluxes and SPIn. For Quercus and Pinus, two genera producing many flowers each year, SPIn displayed limited variability and no clear association with TF element fluxes. Overall, pollen contributed on average 4.1-10.6% of the annual TF fluxes of K+ > DOC > DON > NH4+--N with the highest contribution in Quercus > Fagus > Pinus > Picea stands. Tree pollen appears to affect TF inorganic nitrogen fluxes both qualitatively and quantitatively, acting as a source of NH4+--N and a sink of NO3--N. Pollen appears to play a more complex role in nutrient cycling than previously thought.

  • Outdoor airborne allergens: Characterization, behavior and monitoring in Europe

    • Aeroallergens or inhalant allergens, are proteins dispersed through the air and have the potential to induce allergic conditions such as rhinitis, conjunctivitis, and asthma. Outdoor aeroallergens are found predominantly in pollen grains and fungal spores, which are allergen carriers. Aeroallergens from pollen and fungi have seasonal emission patterns that correlate with plant pollination and fungal sporulation and are strongly associated with atmospheric weather conditions. They are released when allergen carriers come in contact with the respiratory system, e.g. the nasal mucosa. In addition, due to the rupture of allergen carriers, airborne allergen molecules may be released directly into the air in the form of micronic and submicronic particles (cytoplasmic debris, cell wall fragments, droplets etc.) or adhered onto other airborne particulate matter. Therefore, aeroallergen detection strategies must consider, in addition to the allergen carriers, the allergen molecules themselves. This review article aims to present the current knowledge on inhalant allergens in the outdoor environment, their structure, localization, and factors affecting their production, transformation, release or degradation. In addition, methods for collecting and quantifying aeroallergens are listed and thoroughly discussed. Finally, the knowledge gaps, challenges and implications associated with aeroallergen analysis are described.

The European Commission Cooperation in Science and Technology (COST) Action FA1203 “SMARTER” aims to make recommendations for the sustainable management of Ambrosia across Europe and for monitoring its efficiency and cost-effectiveness. The goal of the present study is to provide a baseline for spatial and temporal variations in airborne Ambrosia pollen in Europe that can be used for the management and evaluation of this noxious plant. The study covers the full range of Ambrosia artemisiifolia L. distribution over Europe (39°N–60°N; 2°W–45°E). Airborne Ambrosia pollen data for the principal flowering period of Ambrosia (August–September) recorded during a 10-year period (2004–2013) were obtained from 242 monitoring sites. The mean sum of daily average airborne Ambrosia pollen and the number of days that Ambrosia pollen was recorded in the air were analysed. The mean and standard deviation (SD) were calculated regardless of the number of years included in the study period, while trends are based on those time series with 8 or more years of data. Trends were considered significant at p<0.05. There were few significant trends in the magnitude and frequency of atmospheric Ambrosia pollen (only 8% for the mean sum of daily average Ambrosia pollen concentrations and 14% for the mean number of days Ambrosia pollen were recorded in the air). The direction of any trends varied locally and reflected changes in sources of the pollen, either in size or in distance from the monitoring station. Pollen monitoring is important for providing an early warning of the expansion of this invasive and noxious plant.

  • Variation in Artemisia pollen seasons in Central and Eastern Europe

    • This paper aims to address some gaps in current knowledge by studying temporal and spatial variations in Artemisia pollen counts (2000-2009) at 13 sites located in different biogeographical areas of Central and Eastern Europe. Analysis showed that start dates of Artemisia pollen seasons are greatly dependent on temperature during June and July, with hot summer temperatures having a tendency to delay summer flowering. However, this relationship is not linear and the rate at which seasons become later increases when mean minimum June-July temperatures reach a threshold of about 13 degrees C. No explanation for variations in pollen season intensity could be found. The geographical location or amount of urbanisation did not influence, either positively or negatively, the seasonal pollen index. Second peaks in Artemisia pollen seasons can be described as the pollen seasons of late flowering Artemisia species, and mainly occurred in the geographical area south of the Carpathian Mountains. These second peaks can significantly influence the seasonal pollen index, contributing over 50% to the season's total Artemisia pollen recorded at one site. (C) 2012 Elsevier B.V.     • The development of antigen arrays has provided researchers with great tools to identify reactivities against self or foreign antigens from body fluids. Yet, these approaches mostly do not address antibody isotypes and their effector functions even though these are key points for a more detailed understanding of disease processes. Here, we present a bead array-based assay for a multiplexed determination of antigen-specific antibody levels in parallel with their properties for complement activation. We measured the deposition of C3 fragments from serum samples to reflect the degree of complement activation via all three complement activation pathways. We utilized the assay on a bead array containing native and citrullinated peptide antigens to investigate the levels of IgG, IgM and IgA autoantibodies along with their complement activating properties in serum samples of 41 rheumatoid arthritis patients and 40 controls. Our analysis revealed significantly higher IgG reactivity against the citrullinated fibrinogen beta and filaggrin peptides as well as an IgA reactivity that was exclusive for citrullinated fibrinogen b peptide and C3 deposition in rheumatoid arthritis patients. In addition, we characterized the humoral immune response against the viral EBNA-1 antigen to demonstrate the applicability of this assay beyond autoimmune conditions. We observed that particular buffer compositions were demanded for separate measurement of antibody reactivity and complement activation, as detection of antigen-antibody complexes appeared to be masked due to C3 deposition. We also found that rheumatoid factors of IgM isotype altered C3 deposition and introduced false-positive reactivities against EBNA-1 antigen. In conclusion, the presented bead-based assay setup can be utilized to profile antibody reactivities and immune-complex induced complement activation in a high-throughput manner and could facilitate the understanding and diagnosis of several diseases where complement activation plays role in the pathomechanism.

    • Purpose: Poythress, Edens, et al. (2010) recently used cluster analysis to identify subtypes of antisocial and psychopathic offenders using a diverse collection of theoretically important clustering variables. Two predicted subtypes, primary and secondary psychopathy, were identified, in addition to non-psychopathic and (unexpectedly) "fearful" psychopathic offenders. The purpose of the present research was to determine whether these clusters could be replicated using a single self-report measure, the Psychopathic Personality Inventory (PPI; Lilienfeld & Andrews, 1996). Method: Study 1: We used discriminant function analysis (DFA) to predict cluster membership for the Poythress et al. subtypes based solely on the eight subscales of the PPI. Results: Study 1: Though overall classification accuracy with the original clusters was poor, PPI-derived subtypes differed from each other in theoretically consistent ways on several criterion measures. Method: Study 2: We used the PPI-based DFA to classify a separate sample of prison inmates from a prior PPI study (Edens et al., 2008). Results: Study 2: As predicted, inmates classified into the secondary psychopathy subgroup demonstrated the highest rates of aggressive misconduct whereas non-psychopathic were the least prone to engage in misconduct. Conclusion: The PPI may serve as a relatively simple method of identifying theoretically meaningful subtypes of psychopathic offenders.

    • Plant organ size shows remarkable uniformity within species indicating strong endogenous control. We have identified a plant growth regulatory gene, functionally and structurally homologous to human EBP1. Plant EBP1 levels are tightly regulated; gene expression is highest in developing organs and correlates with genes involved in ribosome biogenesis and function. EBP1 protein is stabilised by auxin. Elevating or decreasing EBP1 levels in transgenic plants results in a dose-dependent increase or reduction in organ growth, respectively. During early stages of organ development, EBP1 promotes cell proliferation, influences cell-size threshold for division and shortens the period of meristematic activity. In postmitotic cells, it enhances cell expansion. EBP1 is required for expression of cell cycle genes; CyclinD3;1, ribonucleotide reductase 2 and the cyclin-dependent kinase B1;1. The regulation of these genes by EBP1 is dose and auxin dependent and might rely on the effect of EBP1 to reduce RBR1 protein level. We argue that EBP1 is a conserved, dose-dependent regulator of cell growth that is connected to meristematic competence and cell proliferation via regulation of RBR1 level.

    • During the last decades (-)-deprenyl has become the golden standard of MAO-B inhibitors. It possesses dopamine potentiating and antioxidant properties; however, its effects cannot be explained solely by the enzyme inhibitory action. (-)-Deprenyl prevents the toxicity of certain selective neurotoxins and recently it was demonstrated to increase cell-cell adhesion as well. The complexity of its pharmacological effects reflects the action of both the parent compound and the active metabolites. (-)-Deprenyl and related propargylamines (DRPs) show neuroprotective features in a variety of in vitro and in vivo models that is dependent on the propargyl moiety. The main presumptive targets to date include glyceraldehyde-3-phosphate dehydrogenase, poly(ADP-ribose) polymerase, some kinase cascades, as well as pro- and antiapoptotic proteins, beside the inhibition of MAO-B.     • The paper considers the possibilities of modelling the release and dispersion of the pollen of common ragweed (Ambrosia artemisiifolia L.), a highly allergenic invasive weed, which is spreading through southern and central Europe. In order to provide timely warnings for the allergy sufferers, a model was developed for forecasting ragweed pollen concentrations in the air. The development was based on the system for integrated modelling of atmospheric composition (SILAM) and concentrated on spatio-temporal modelling of ragweed flowering season and pollen release, which constitutes the emission term. Evaluation of the new model against multi-annual ragweed pollen observations demonstrated that the model reproduces well the main ragweed pollen season in the areas with major plant presence, such as the Pannonian Plain, the Lyon area in France, the Milan region in Italy, Ukraine and southern Russia. The predicted start of the season is mostly within 3 days of the observed for the majority of stations in these areas. The temporal correlation between modelled and observed concentrations exceeds 0.6 for the bulk of the stations. Model application to the seasons of 2005–2011 indicated the regions with high ragweed pollen concentrations, in particular the areas where allergenic thresholds are exceeded. It is demonstrated that, due to long-range transport of pollen, high-concentration areas are substantially more extensive than the heavily infested territories.

    • A large number of studies have indicated recently that photosynthetic reaction center proteins (RC) bind successfully to nanostructures and their functional activity is largely retained. The major goal of current research is to find the most efficient systems and conditions for the photoelectric energy conversion and for the stability of this bio-nanocomposite. In our studies, we immobilized the RC protein on multiwalled carbon nanotubes (MWNT) through specific chemical binding to amine functional groups and through conducting polymer (poly(3-thiophene acetic acid), PTAA). Both structural (TEM, AFM) and functional (absorption change and conductivity) measurements has shown that RCs could be bound effectively to functionalized CNTs. The kinetics of the light induced absorption change indicated that RCs were still active in the composite and there was an interaction between the protein cofactors and the CNTs. The light generated photocurrent was measured in an electrochemical cell with transparent CNT electrode designed specially for this experiment.

    • An optoelectronic device, which converts light energy to electric potential, was designed and fabricated by using photosynthetic reaction centre (RC) proteins of purple bacterium Rhodobacter sphaeroides R-26, based on the structure and function of the dye sensitised organic solar cells. First, an electrochemical cell with three electrodes was created especially for this measurement. ITO covered by the MWCNT-RC containing sample served as the working electrode and the counter and the reference electrodes were platinum and Ag\AgCl, respectively. Water soluble ubiquinone-0 and ferrocene, in some experiments were used as mediators. In another experiment, the presence of the RCs in the active layer under dried conditions assured tuneable wavelength sensitivity, in general in the visible, but specially, in the near infrared (700-1000 nm) spectral range. The lifetime of the primary charge separation is in the ps time scale and that of the charge stabilisation can be modulated (at least theoretically) between ps and seconds. A successful combination of RC protein with a light energy converter device in spectroelectrochemical cell (wet conditions in buffered electrolyte) and in dried multilayer structure will be presented here.
