  • Patrilocality and hunter-gatherer-related ancestry of populations in East-Central Europe during the Middle Bronze Age

    • The demographic history of East-Central Europe after the Neolithic period remains poorly explored, despite this region being on the confluence of various ecological zones and cultural entities. Here, the descendants of societies associated with steppe pastoralists form Early Bronze Age were followed by Middle Bronze Age populations displaying unique characteristics. Particularly, the predominance of collective burials, the scale of which, was previously seen only in the Neolithic. The extent to which this re-emergence of older traditions is a result of genetic shift or social changes in the MBA is a subject of debate. Here by analysing 91 newly generated genomes from Bronze Age individuals from present Poland and Ukraine, we discovered that Middle Bronze Age populations were formed by an additional admixture event involving a population with relatively high proportions of genetic component associated with European hunter-gatherers and that their social structure was based on, primarily patrilocal, multigenerational kin-groups. By analysing 91 Bronze Age genomes from East-Central Europe, the authors discovered that Middle Bronze Age populations were formed by an admixture event involving hunter-gatherers and that the social structure of resulting population was primarily patrilocal.

  • Matrisome Properties of Scaffolds Direct Fibroblasts in Idiopathic Pulmonary Fibrosis

    • In idiopathic pulmonary fibrosis (IPF) structural properties of the extracellular matrix (ECM) are altered and influence cellular responses through cell-matrix interactions. Scaffolds (decellularized tissue) derived from subpleural healthy and IPF lungs were examined regarding biomechanical properties and ECM composition of proteins (the matrisome). Scaffolds were repopulated with healthy fibroblasts cultured under static stretch with heavy isotope amino acids (SILAC), to examine newly synthesized proteins over time. IPF scaffolds were characterized by increased tissue density, stiffness, ultimate force, and differential expressions of matrisome proteins compared to healthy scaffolds. Collagens, proteoglycans, and ECM glycoproteins were increased in IPF scaffolds, however while specific basement membrane (BM) proteins such as laminins and collagen IV were decreased, nidogen-2 was also increased. Findings were confirmed with histology, clearly showing a disorganized BM. Fibroblasts produced scaffold-specific proteins mimicking preexisting scaffold composition, where 11 out of 20 BM proteins were differentially expressed, along with increased periostin and proteoglycans production. We demonstrate how matrisome changes affect fibroblast activity using novel approaches to study temporal differences, where IPF scaffolds support a disorganized BM and upregulation of disease-associated proteins. These matrix-directed cellular responses emphasize the IPF matrisome and specifically the BM components as important factors for disease progression.

  • Spike-Dependent Opsonization Indicates Both Dose-Dependent Inhibition of Phagocytosis and That Non-Neutralizing Antibodies Can Confer Protection to SARS-CoV-2

    • Spike-specific antibodies are central to effective COVID19 immunity. Research efforts have focused on antibodies that neutralize the ACE2-Spike interaction but not on non-neutralizing antibodies. Antibody-dependent phagocytosis is an immune mechanism enhanced by opsonization, where typically, more bound antibodies trigger a stronger phagocyte response. Here, we show that Spike-specific antibodies, dependent on concentration, can either enhance or reduce Spike-bead phagocytosis by monocytes independently of the antibody neutralization potential. Surprisingly, we find that both convalescent patient plasma and patient-derived monoclonal antibodies lead to maximum opsonization already at low levels of bound antibodies and is reduced as antibody binding to Spike protein increases. Moreover, we show that this Spike-dependent modulation of opsonization correlate with the outcome in an experimental SARS-CoV-2 infection model. These results suggest that the levels of anti-Spike antibodies could influence monocyte-mediated immune functions and propose that non-neutralizing antibodies could confer protection to SARS-CoV-2 infection by mediating phagocytosis.

    • Background: The St Gallen surrogate definition of the intrinsic subtypes of breast cancer consist of five subgroups based on estrogen receptor (ER), progesterone receptor (PgR), human epidermal growth factor receptor type 2 (HER2), and Ki-67. PgR and Ki-67 are used for discriminating between the Luminal A-like and Luminal B-like (HER2-negative) subtypes. Histological grade (G) has prognostic value in breast cancer; however, its relationship to the St Gallen subtypes is not clear. Based on a previous pilot study, we hypothesized that G could be a primary discriminator for ER-positive/HER2-negative breast cancers that were G1 or G3, whereas Ki-67 and PgR could provide additional prognostic information specifically for patients with G2 tumors. To test this hypothesis, a larger patient cohort was examined. Patients and methods: Six hundred seventy-one patients (amp;gt;= 35 years of age, pT1-2, pN0-1) with ER-positive/HER2-negative breast cancer and complete data for PgR, Ki-67, G, lymph node status, tumor size, age, and distant disease-free survival (DDFS; median follow-up 9.2 years) were included. Results: Luminal A-like tumors were mostly G1 or G2 (90%) whereas Luminal B-like tumors were mostly G2 or G3 (87%) and corresponded with good and poor DDFS, respectively. In Luminal B-like tumors that were G1 (n = 23), no metastasis occurred, whereas 14 of 40 Luminal A-like tumors that were G3 metastasized. In the G2 subgroup, low PgR and high Ki-67 were associated with an increased risk of distant metastases, hazard ratio (HR) and 95% confidence interval (CI) 1.8 (0.95-3.4) and 1.5 (0.80-2.8), respectively. Conclusions: Patients with ER-positive/HER2-negative/G1 breast cancer have a good prognosis, similar to that of Luminal A-like, while those with ER-positive/HER2-negative/G3 breast cancer have a worse prognosis, similar to that of Luminal B-like, when assessed independently of PgR and Ki-67. Therapy decisions based on Ki-67 and PgR might thus be restricted to the subgroup G2.

    • Therapeutic strategies directed at the tumor surfaceome (TS), including checkpoint inhibitor blocking antibodies, antibody drug conjugates (ADCs), and chimeric antigen receptor T (CAR-T) cells, provide a new armament to fight cancer. However, a remaining bottleneck is the lack of strategies to comprehensively interrogate patient tumors for potential TS targets. Here, we have developed a platform (tumor surfaceome mapping [TS-MAP]) integrated with a newly curated TS classifier (SURFME) that allows profiling of primary 3D cultures and intact patient glioma tumors with preserved tissue architecture. Moreover, TS-MAP specifically identifies proteins capable of endocytosis as tractable targets for ADCs and other modalities requiring toxic payload internalization. In high-grade gliomas that remain among the most aggressive forms of cancer, we show that cellular spatial organization (2D vs. 3D) fundamentally transforms the surfaceome and endocytome (e.g., integrins, proteoglycans, semaphorins, and cancer stem cell markers) with general implications for target screening approaches, as exemplified by an ADC targeting EGFR. The TS-MAP platform was further applied to profile the surfaceome and endocytome landscape in a cohort of freshly resected gliomas. We found a highly diverse TS repertoire between patient tumors, not directly associated with grade and histology, which highlights the need for individualized approaches. Our data provide additional layers of understanding fundamental to the future development of immunotherapy strategies, as well as procedures for proteomics-based target identification and selection. The TS-MAP platform should be widely applicable in efforts aiming at a better understanding of how to harness the TS for personalized immunotherapy.

    • Objective: We aim to identify maternal genetic affinities between the Middle to Final Neolithic (3850-2300 BC) populations from present-day Poland and possible genetic influences from the Pontic steppe.Materials and methods: We conducted ancient DNA studies from populations associated with Zlota, Globular Amphora, Funnel Beaker, and Corded Ware cultures (CWC). We sequenced genomic libraries on Illumina platform to generate 86 complete ancient mitochondrial genomes. Some of the samples were enriched for mitochondrial DNA using hybridization capture.Results: The maternal genetic composition found in Zlota-associated individuals resembled that found in people associated with the Globular Amphora culture which indicates that both groups likely originated from the same maternal genetic background. Further, these two groups were closely related to the Funnel Beaker culture-associated population. None of these groups shared a close affinity to CWC-associated people. Haplogroup U4 was present only in the CWC group and absent in Zlota group, Globular Amphora, and Funnel Beaker cultures.Discussion: The prevalence of mitochondrial haplogroups of Neolithic farmer origin identified in Early, Middle and Late Neolithic populations suggests a genetic continuity of these maternal lineages in the studied area. Although overlapping in time - and to some extent - in cultural expressions, none of the studied groups (Zlota, Globular Amphora, Funnel Beaker), shared a close genetic affinity to CWC-associated people, indicating a larger extent of cultural influence from the Pontic steppe than genetic exchange. The higher frequency of haplogroup U5b found in populations associated with Funnel Beaker, Globular Amphora, and Zlota cultures suggest a gradual maternal genetic influx from Mesolithic hunter-gatherers. Moreover, presence of haplogroup U4 in Corded Ware groups is most likely associated with the migrations from the Pontic steppe at the end of the Neolithic and supports the observed genetic distances.

    • Objective: In this work we aim to investigate the origins and genetic affinities of Bronze Age populations (2,400-1,100 BC) from the region of southern Poland and to trace maternal kinship patterns present in the burials of those populations by the use of complete mitochondrial genomes.Materials and methods: We performed ancient DNA analyses for Bronze Age individuals from present-day Poland associated with the Strzy(z) over dotow culture, the Mierzanowice culture, and the Trzciniec Cultural circle. To obtain complete mitochondrial genomes, we sequenced genomic libraries using Illumina platform. Additionally, hybridization capture was used to enrich some of the samples for mitochondrial DNA. AMS C-14-dating was conducted for 51 individuals to verify chronological and cultural attribution of the analyzed samples.Results: Complete ancient mitochondrial genomes were generated for 80 of the Bronze Age individuals from present-day Poland. The results of the population genetic analyses indicate close maternal genetic affinity between Mierzanowice, Trzciniec, and Corded Ware culture-associated populations. This is in contrast to the genetically more distant Strzy(z) over dotow people that displayed closer maternal genetic relation to steppe populations associated with the preceding Yamnaya culture and Catacomb culture, and with later Scythians. Potential maternal kinship relations were identified in burials of Mierzanowice and Trzciniec populations analyzed in this study.Discussion: Results revealed genetic continuity from the Late Neolithic Corded Ware groups to Bronze Age Mierzanowice and Trzciniec-associated populations, and possible additional genetic contribution from the steppe to the formation of the Strzy(z) over dotow-associated group at the end of 3rd millennium BC. Mitochondrial patterns indicated several pairs of potentially maternally related individuals mostly in Trzciniec-associated group.

    • Activated fibroblasts are suggested to be involved in the deposition of extracellular matrix in the formation of peribronchial fibrosis in asthma. We report the novel finding of activated elongated fibroblasts accompanied by elevated numbers of eosinophils in bronchoalveolar lavage fluid from 5 out of 12 patients with mild asthma (= 42%), whereas no fibroblasts were observed in the control subjects without asthma (n = 17). The elongated fibroblasts migrated twice as far when compared with fibroblasts from corresponding bronchial biopsies from the same patients, accompanied by an induced expression of RhoA and Rac1, indicating that the increased expression of these proteins are linked to increased migratory capabilities. Moreover, the elongated fibroblasts had an elevated production of the proteoglycans biglycan, versican, perlecan, and decorin, which correlated to an active cytoplasm in these cells. Differential expression patterns between the two fibroblast groups in motility-regulating proteins, such as cofilin, nuclear chloride ion channel protein, and heat-shock protein 20, were identified by two-dimensional electrophoresis and mass spectrometry. These findings indicate the presence of activated and mobile fibroblasts accompanied by an induced inflammatory response outside the airway epithelium in patients with mild asthma, results that may play a role in formation of airway fibrosis.

    • Background: There are potential conflicts between authorities and companies to fund new premium priced drugs especially where there are safety and/or budget concerns. Dabigatran, a new oral anticoagulant for the prevention of stroke in patients with non-valvular atrial fibrillation (AF), exemplifies this issue. Whilst new effective treatments are needed, there are issues in the elderly with dabigatran due to variable drug concentrations, no known antidote and dependence on renal elimination. Published studies have shown dabigatran to be cost-effective but there are budget concerns given the prevalence of AF. There are also issues with potentially re-designing anticoagulant services. This has resulted in activities across countries to better manage its use. Objective: To (i) review authority activities in over 30 countries and regions, (ii) use the findings to develop new models to better manage the entry of new drugs, and (iii) review the implications for all major stakeholder groups. Methodology: Descriptive review and appraisal of activities regarding dabigatran and the development of guidance for groups through an iterative process. Results: There has been a plethora of activities among authorities to manage the prescribing of dabigatran including extensive pre-launch activities, risk sharing arrangements, prescribing restrictions, and monitoring of prescribing post-launch. Reimbursement has been denied in some countries due to concerns with its budget impact and/or excessive bleeding. Development of a new model and future guidance is proposed to better manage the entry of new drugs, centering on three pillars of pre-, pen-, and post-launch activities. Conclusion: Models for introducing new drugs are essential to optimize their prescribing especially where there are concerns. Without such models, new drugs may be withdrawn prematurely and/or struggle for funding.

  • Quantifying extracellular matrix turnover in human lung scaffold cultures

    • Remodelling of the extracellular matrix is accomplished by altering the balance between matrix macromolecule production and degradation. However, it is not well understood how cells balance production of new matrix molecules and degradation of existing ones during tissue remodelling and regeneration. In this study, we used decellularized lung scaffolds repopulated with allogenic lung fibroblasts cultured with stable isotope labelled amino acids to quantify the balance between matrix production and degradation at a proteome-wide scale. Specific temporal dynamics of different matrisome proteins were found to correspond to the proliferative activity of the repopulating cells and the degree of extracellular deposition. The remodeling of the scaffold was characterized by an initial phase with cell proliferation and high production of cell adhesion proteins such as emilin-1 and fibronectin. Extended culture time resulted in increased levels of core matrisome proteins. In a comparison with monolayer cultures on plastic, culture in lung scaffolds lead to a pronounced accumulation of proteoglycans, such as versican and decorin, resulting in regeneration of an extracellular matrix with greater resemblance to native lung tissue compared to standard monolayer cultures. Collectively, the study presents a promising technique for increasing the understanding of cell- extracellular matrix interactions under healthy and diseased conditions.

  • Sex-based differences in pain distribution in a cohort of patients with persistent post-traumatic neck pain

    • Objectives: To analyze a cohort of 745 consecutive patients referred to a regional specialist clinic for evaluation of post-traumatic neck pain during a five-year period. Methods: A cross-sectional observational study of baseline assessments performed by multi-professional rehabilitation teams according to a standardized checklist. Results: The cohort contained nearly twice as many females as males (64% versus 36%). The type of injury did not differ between sexes. Of the entire cohort, 38% were diagnosed with widespread pain, 50% with regional pain, and 12% with local pain. The pain distribution among the females was 43% widespread, 48% regional, and 9% local, and corresponding figures among males were 29%, 53%, and 18%. Longer time between trauma and assessment did not affect pain distribution among the men, but a tendency towards more widespread pain was observed among the women. Discussion: The importance of "female sex" as risk factor for the development of persistent pain after neck trauma needs to be discussed further. The high frequency of regional and widespread pain among patients with persistent neck pain after trauma calls for both multidisciplinary assessments and treatment strategies. The relationships between different pain distribution patterns, disability, activity, and psychological factors need to be studied further.Implications for rehabilitationPatients suffering from pain and disability after neck trauma constitute a significant proportion of patients with persistent pain.The importance of the risk factor “female sex" should be further discussed in the development of persistent pain after neck trauma.The high frequency of regional and widespread pain among patients with persistent neck pain after trauma calls both for multidisciplinary assessments and treatment strategies.The relationships between different pain distribution patterns, disability, activity, and psychological factors need to be studied further.

    • Soil communities are considered to be remarkably species-rich and to have many generalist species with seemingly similar niche requirements. The composition of soil fauna communities is often highly variable even at the plot scale, and both the environment and the spatial configuration of microhabitats are regarded as important forces shaping the structure of local communities. However, to what extent these forces are important in different ecosystems is not clear. We examined the relative roles of environmental (abiotic), vegetation (biotic) and spatial variables (using Moran's eigenvector maps, MEM) for the small-scale variation in springtail (Collembola) communities in a 100 m(2) area of the forest floor of a mature Scots pine forest in central Sweden, with small variation in important environmental variables.We found that most of the small-scale variation in community composition could be explained by spatial variables, either alone or jointly with the environmental variables. Spatial variability in community composition, in turn, could be related to shifts in functional traits of the component species. Within local communities (samples), species showed a higher diversity than expected by chance in almost all examined traits, indicating that differences in resource and micro-habitat utilisation enable Collembola species to coexist. Competition between species is therefore likely to be important for structuring Collembola communities at this spatial scale. The results indicate that the spatial scale of study and heterogeneity of environmental factors influence soil fauna community assembly processes through effects on the relative importance of environmental filtering compared to filtering by limiting similarity or competitive exclusion.   • Operationalizing Industrial Ecology in the Waste Sector : Roles and tactics for circular value innovation

    • The take-make-waste approach to resource management in human production and consumption systems is contributing to a variety of environmental and social problems worldwide. Additionally, as the world’s population and affluence increase, so do the negative impacts of poor resource management. Lifting the waste management (WM) sector into a new phase of development, which takes its lead from the ideals of Industrial Ecology and circular economy, is seen by many scholars and practitioners as one potential to assist in alleviating these impacts. While there are many studies on how more efficient inter-organizational resource management is (or could be) constructed, there are relatively few business development studies which have explored novel approaches (from roles to tactics) that WM organizations might operationalize toward more efficient resource management.The aim of this thesis is to contribute to the development of knowledge and understanding of how the waste management sector can operationalize more effective and efficient resource management. In approaching this aim, two research questions guided the exploration of: 1) novel roles for WM and 2) support tactics for such roles. Grounded in the broader context of Industrial Ecology (IE) and Business Development, five studies were performed. Two studies, focused on the novel roles of inter-organizational resource management and high value secondary resource extraction, were performed through literature review and interviews, and market driver analysis respectively. In exploring support tactics, two design and proof of concept studies were carried out to investigate data analysis tools for inter-organizational resource management, and one long-term action research engagement project was coordinated to study hands-on inter-organizational collaboration tactics.The studies highlighted that the Swedish WM sector holds some key capacities for operationalizing (and in some cases, is already developing) the novel resource management roles identified: industrial symbiosis facilitator, eco-industrial park manager, holistic facility management, and high value resource extractor. However, depending on the portfolio of services to be performed in such roles, several capacities may need to be developed or strengthened. Main opportunities seen for these roles were – staying ahead of market developments, and aligning activities with organizational goals. The main general risk related to these roles was insufficient returns on investment. Looking forward, the main enablers identified were policy leadership for more balanced market mechanisms, increasing use of external knowledge, developing long term partnerships, lobbying, stockpiling resources, and carefully crafting new business models.The tools developed for strategically applying external information toward the identification of opportunities within new roles showed tactical potential. However, their implementation in broader development processes has yet to be fully validated. The hands-on exploration of change oriented collaboration, highlighted collective system framing and goal setting and face-to-face interaction as key activities for inter-organizational approaches within roles such as industrial symbiosis facilitator.Throughout the studies, several novel roles were investigated. Each of these roles will need to be individually evaluated by directing bodies of WM organizations, and evaluated from the organization’s vision and strategy. If certain roles are chosen to be explored in more detail, they will need to be developed within full business models - addressing issues such as income structure, internal processes and capacities to be developed, and key customers. Through applying IE and business development concepts and findings, WM organizations have possibilities to translate ambitious visions into novel offerings.

    • Versican is a proteoglycan that has many different roles in tissue homeostasis and inflammation. The biochemical structure is comprised of four different types of the core protein with attached glycosaminoglycans that can be sulphated to various extents and has the capacity to regulate differentiation of different cell types, migration, cell adhesion, proliferation, tissue stabilization and inflammation. Versican's regulatory properties are of importance during both homeostasis and changes that lead to disease progression. The glycosaminoglycans that are attached to the core protein are of the chondroitin sulfate/dermatan sulfate type and are known to be important in inflammation through interactions with cytokines and growth factors. For a more complex understanding of versican it is of importance to study the tissue niche, where the wound healing process in both healthy and diseased conditions take place. In previous studies our group has identified changes in the amount of the multifaceted versican in chronic lung disorders such as asthma, chronic obstructive pulmonary disease and bronchiolitis obliterans syndrome, which could be a result of pathologic, transforming growth factor β driven, on-going remodelling processes. Reversely, the context of versican in its niche is of great importance since versican has been reported to have a beneficial role in other contexts e.g. emphysema. Here we explore the vast mechanisms of versican in healthy lung and in lung disorders.

  • Translation and cultural adaptation of the integrated palliative care outcome scale for use in Sweden

    • Background: Patient-reported outcome measurements are important for measuring changes in patients’ health over time, evaluating the quality of care given, and improving the quality of care service. The Integrated Palliative care Outcome Scale (IPOS) is one such measure, capturing clinically important physical, psychological, social, and existential aspects of patients’ palliative care needs. When patients are unable to complete the IPOS questionnaire themselves with or without assistance, a staff version is also available. Aim: To translate and culturally adapt the IPOS Patient and Staff versions for use in the Swedish context. Method: The process comprised forward and backward translations, cultural adaptation, and expert group reviews. To validate the resulting Swedish version of IPOS, cognitive interviews were conducted with 13 patients and 15 staff from various care contexts. Preliminary results: The Swedish expert group changed some words and grammar due to minor discrepancies in the back translation process. The participants in the cognitive interviews responded positively overall to the questionnaire. Deteriorating health and not having Swedish as the native language did not cause problems in completing the questionnaire. After the first round of cognitive interviews, problematic questions and answer options were rephrased, and redundant text was deleted. Conclusion: The Swedish IPOS (Patient and Staff versions) has been validated linguistically and culturally, and is now available for clinical use. The next final step in the validation process will be to test the psychometric performance of the Swedish questionnaires.

    • BACKGROUND: To expand our clinical and scientific knowledge about holistic outcomes within palliative care, there is a need for agreed-upon patient-reported outcome measures. These patient-reported outcome measures then require translation and cultural adaptation, either from country-specific languages to English, or the other way around. The aim of this study was to translate and cross-culturally adapt the Integrated Palliative care Outcome Scale (IPOS) to the Swedish care context.METHODS: Swedish versions of IPOS Patient and IPOS Staff were developed and culturally adapted using recommended guidelines including cognitive interviews with patients (n = 13) and staff (n = 15) from different care contexts including general and specialised palliative care.RESULTS: The comprehension and judgement difficulties identified in the pre-final patient and staff versions were successfully solved during the cognitive interviewing process. IPOS was well accepted by both patients and staff, none of the questions were experienced as inappropriate, and all questions were judged important.CONCLUSIONS: In this study, we translated and culturally adapted the patient and staff versions of IPOS, and demonstrated face and content validity and acceptability of the scale through cognitive interviewing with patients and staff within residential care facility, surgical and specialised palliative home care units. Cognitive interviewing in parallel with patients and staff in rounds, with tentative analysis in between, was a suitable method for identifying and solving challenges with comprehension and evaluation in the pre-final version of IPOS. The Swedish IPOS is now available for use in a variety of clinical care settings.

    • The generation of mathematical models of biological processes, the simulation of these processes under different conditions, and the comparison and integration of multiple data sets are explicit goals of systems biology that require the knowledge of the absolute quantity of the system's components. To date, systematic estimates of cellular protein concentrations have been exceptionally scarce. Here, we provide a quantitative description of the proteome of a commonly used human cell line in two functional states, interphase and mitosis. We show that these human cultured cells express at least similar to 10 000 proteins and that the quantified proteins span a concentration range of seven orders of magnitude up to 20 000 000 copies per cell.     • Despite their significance in soil ecosystems and their use for investigations of soil ecosystem functioning and in bioindication elsewhere, springtails (Collembola) have not been well investigated in South Africa. Early recognition of their role in soil systems and sporadic systematic work has essentially characterised knowledge of the southern African fauna for some time. The situation is now changing as a consequence of systematic and ecological work on springtails. To date this research has focused mostly on the Cape Floristic Region and has revealed a much more diverse springtail fauna than previously known (136 identifiable species and an estimated 300 species for the Cape Floristic Region in total), including radiations in genera such as the isotomid Cryptopygus. Quantitative ecological work has shown that alpha diversity can be estimated readily and that the group may be useful for demonstrating land use impacts on soil biodiversity. Moreover, this ecological work has revealed that some disturbed sites, such as those dominated by Galenia africana, may be dominated by invasive springtail species. Investigation of the soil fauna involved in decomposition in Renosterveld and Fynbos has also revealed that biological decomposition has likely been underestimated in these vegetation types, and that the role of fire as the presumed predominant source of nutrient return to the soil may have to be re-examined. Ongoing research on the springtails will provide the information necessary for understanding and conserving soils: one of southern Africa's major natural assets.

  • Short term outcomes after robot assisted and open cystectomy- A nation-wide population-based study

    • Introduction: We aimed to compare short term outcomes after robot assisted radical cystectomy (RARC) and open radical cystectomy (ORC) for urinary bladder cancer in a large population.Materials and methods: We included all patients without distant metastases who underwent either RARC or ORC with ileal conduit between 2011 and 2019 registered in the Bladder cancer data Base Sweden (BladderBaSe) 2.0. Primary outcome was unplanned readmissions within 90 days, and secondary out-comes within 90 days of surgery were reoperations, Clavien 3-5 complications, total days alive and out of hospital, and mortality. The analysis was carried out using multivariate regression models.Results: Out of 2905 patients, 832 were operated with RARC and 2073 with ORC. Robotic procedures were to a larger extent performed during later years, at high volume centers (47% vs 17%), more often for organ-confined disease (82% vs. 72%) and more frequently in patients with high socioeconomic status (26% vs. 21%). Patients operated with RARC were more commonly readmitted (29% vs. 25%). In multi -variable analysis RARC was associated with decreased risk of Clavien 3-5 complications (OR 0.58, 95% CI 0.47-0.72), reoperations (OR 0.53, 95% CI 0.39-0.71) and had more days alive and out of hospital (mean difference 3.7 days, 95% CI 2.4-5.0).Conclusion: This study illustrates the "real-world" effects of a gradual and nation-wide introduction of RARC. Patients operated with RARC had fewer major complications and reoperations but were more frequently readmitted compared to ORC.     • Background: Membranous expression of the anti-adhesive glycoprotein podocalyxin-like (PODXL) has previously been found to correlate with poor prognosis in several major cancer forms. Here we examined the prognostic impact of PODXL expression in urothelial bladder cancer. Methods: Immunohistochemical PODXL expression was examined in tissue microarrays with tumours from two independent cohorts of patients with urothelial bladder cancer: n = 100 (Cohort I) and n = 343 (Cohort II). The impact of PODXL expression on disease-specific survival (DSS; Cohort II), 5-year overall survival (OS; both cohorts) and 2-year progression-free survival (PFS; Cohort II) was assessed. Results: Membranous PODXL expression was significantly associated with more advanced tumour (T) stage and high-grade tumours in both cohorts, and a significantly reduced 5-year OS (unadjusted HR = 2.25 in Cohort I and 3.10 in Cohort II, adjusted HR = 2.05 in Cohort I and 2.18 in Cohort II) and DSS (unadjusted HR = 4.36, adjusted HR = 2.70). In patients with Ta and T1 tumours, membranous PODXL expression was an independent predictor of a reduced 2-year PFS (unadjusted HR = 6.19, adjusted HR = 4.60) and DSS (unadjusted HR = 8.34, adjusted HR = 7.16). Conclusion: Membranous PODXL expression is an independent risk factor for progressive disease and death in patients with urothelial bladder cancer.

    • In this work, highly porous nanopaper, i.e., sheets of papers made from non-aggregated nanofibrillated cellulose (NFC), have been surface-grafted with poly(epsilon-caprolactone) (PCL) by surface-initiated ring-opening polymerization (SI-ROP). The nanopaper has exceptionally high surface area (similar to 300 m(2)/g). The "grafting from" of the nanopapers was compared to "grafting from" of cellulose in the form of filter paper, and in both cases either titanium n-butoxide (Ti(On-Bu)(4)) or tin octoate (Sn(Oct)(2)) was utilized as a catalyst. It was found that a high surface area leads to significantly higher amount of grafted PCL in the substrates when Sn(Oct)2 was utilized as a catalyst. Up to 79 wt % PCL was successfully grafted onto the nanopapers as compared to filter paper where only 2-3 wt % PCL was grafted. However, utilizing Ti(On-Bu)4 this effect was not seen and the grafted amount was essentially similar, irrespectively of surface area. The mechanical properties of the grafted nanopaper proved to be superior to those of pure PCL films, especially at elevated temperatures. The present bottom-up preparation route of NFC-based composites allows high NFC content and provides excellent nanostructural control. This is an important advantage compared with some existing preparation routes where dispersion of the filler in the matrix is challenging.

    • In the present work, cellulose nanocrystals (CNCs) have been surface-modified either via covalent grafting or through physisorption of poly(n-butyl methacrylate) (PBMA) and employed as reinforcement in PCL. Covalent grafting was achieved by surface-initiated atom transfer radical polymerization (SI-ATRP). Two approaches were utilized for the physisorption: using either micelles of poly(dimethyl aminoethyl methacrylate)-block-poly(n-butyl methacrylate) (PDMAEMA-b-PBMA) or latex nanoparticles of poly(dimethyl aminoethyl methacrylate-co-methacrylic acid)-block-poly(n-butyl methacrylate) (P(DMAEMA-co-MAA)-b-PBMA). Block copolymers (PDMAEMA-b-PBMA)s were obtained by ATRP and subsequently micellized. Latex nanoparticles were produced via reversible addition-fragmentation chain-transfer (RAFT) mediated surfactant-free emulsion polymerization, employing polymer-induced self-assembly (PISA) for the particle formation. For a reliable comparison, the amounts of micelles/latex particles adsorbed and the amount of polymer grafted onto the CNCs were kept similar. Two different chain lengths of PBMA were targeted, below and above the critical molecular weight for chain entanglement of PBMA (M-n,M-c similar to 56 000 g mo1(-1)). Poly(epsilon-caprolactone) (PCL) nanocomposites reinforced with unmodified and modified CNCs in different weight percentages (0.5, 1, and 3 wt %) were prepared via melt extrusion. The resulting composites were evaluated by UV-vis, scanning electron microscopy (SEM), thermal gravimetric analysis (TGA), and tensile testing. All materials resulted in higher transparency, greater thermal stability, and stronger mechanical properties than unfilled PCL and nanocomposites containing unmodified CNCs. The degradation temperature of PCL reinforced with grafted CNCs was higher than that of micelle-modified CNCs, and the latter was higher than that of latex-adsorbed CNCs with a long PBMA chain length. The results clearly indicate that covalent grafting is superior to physisorption with regard to thermal and mechanical properties of the final nanocomposite. This unique study is of great value for the future design of CNC-based nanocomposites with tailored properties.

    • In this work, a series of functional nanocrystals (F-CNCs) was successfully produced by an efficient preparation method, combining acid hydrolysis and Fischer esterification with various organic acids. Functionalities such as ATRP initiators, double bonds, triple bonds, and thiols could be incorporated on CNCs. Surface modification was confirmed by FT-IR, XPS, and elemental analysis. Physical properties of FC-NCs were assessed by AFM, XRD and TGA. Moreover, ATRP initiator functionalized CNCs were utilized to graft poly(methyl methacrylate) via ATRP, thiol functionalized CNCs were reacted with Ellman's reagent to determine the thiol content and dye disperse red 13 was attached to alkyne functionalized CNCs to estimate the propiolate content. The herein presented method is a highly versatile and straightforward procedure for the preparation of F-CNCs which is believed to be a better alternative for the commonly utilized, extensive, multistep, and time consuming post functionalization methods.

    • In the present work, poly(vinyl acetate) grafted cellulose nanocrystals (CNC-g-PVAc) were prepared via surface initiated reversible addition-fragmentation chain transfer and macromolecular design via the interchange of xanthates (SI-RAFT/MADIX) polymerization. Successful grafting of PVAc from CNC was confirmed by FT-IR and TGA analysis. PVAc nanocomposites reinforced with CNC-g-PVAc, as well as pristine CNC for comparison, of different weight percentages (0.5, 1, 3 and 5 wt%) of CNC were prepared via solvent casting. The PVAc reinforced with CNC-g-PVAc resulted in higher transparency and improved mechanical properties compared with unmodified CNC nanocomposites. The addition of 5 wt% CNC-g-PVAc increased the modulus of neat PVAc with as much as 154%. The proposed SI-RAFT/MADIX on CNC could be applied to wide range of monomers, and it is believed to be an efficient and robust method for CNC functionalization, thus expanding the potential applicability of CNC.

  • Surface Modification of Nanocellulose towards Composite Applications

    • Nanocelluloses have attracted great interest during recent decades owing to their renewability, abundancy and remarkable physical and mechanical properties. The aim of this work was to investigate new strategies for surface modification and functionalization of nanocelluloses and their subsequent incorporation in polymer-host matrices.Nanocomposites of cellulose nanofibrils (CNF) and polycaprolactone (PCL) were produced by employing CNF nanopaper (NP) as a template and surface-initiated ring-opening polymerization (SI-ROP) of ε-caprolactone (ε-CL). SI-ROP of ε-CL from filter paper (FP) was also carried out for comparison. A larger amount of PCL was grafted from NP than from FP. The grafted NP had stronger mechanical properties than neat PCL.Cellulose nanocrystal (CNC)-reinforced polyvinyl acetate (PVAc) nanocomposites were also investigated. CNC were modified via “SI-reversible addition-fragmentation chain transfer and macromolecular design via the interchange of xanthate” (SI-RAFT/MADIX) polymerization of vinyl acetate (VAc). The resulting nanocomposites exhibited improved mechanical performance than the unmodified CNC.It is generally agreed that covalent grafting is superior to physical adsorption for the modification of a reinforcing agent. However, this hypothesis has never been thoroughly investigated. CNC was modified either through covalent grafting or through physical adsorption of poly(butyl methacrylate) (PBMA). Both methods resulted in improved mechanical performance than that of pure PCL or PCL containing unmodified CNC. However, covalent grafting gave the best mechanical performance even at high relative humidity.Functionalized CNC (F-CNC) were obtained through a versatile methodology employing organic acids bearing a functional group were employed for the simultaneous acid hydrolysis and esterification of cellulose fibers. This provided a facile route for the preparation of F-CNC.

  • A comparative study of covalent grafting and physical adsorption of PCL onto cellulose

    • A growing concern for the environment has, in the past years, directed the research towards a bigger focus on new “greener” materials, such as cellulose-reinforced options. Cellulose is the most abundant organic raw material in the world and it is a versatile material. However, to be able to use it in applications where it is not inherently compatible, a modification is often necessary.1-3 One common method to achieve this modification is to graft polymers onto/from the cellulose chain. This can change the inherent properties of cellulose to attain new properties, such as dimensional stability and water repellency.3 In addition to this, it has been shown that polyectrolytes can be physiosorbed onto charged surfaces.4 Due to this, it is possible to physically modify cellulose by adsorbing a polymer through electrostatic interactions instead of attaching it with a covalent bond.5However, a more detailed investigation concerning differences of covalent and physical attachment of poly(ε-caprolactone) (PCL) onto cellulose, has to the author’s best knowledge not been performed. Therefore, this project aims to compare these two techniques. Covalently bonded PCL was grafted by surface-initiated ring opening polymerization (SI-ROP) from the cellulose. For the adsorption approach, a block copolymer consisting of PCL and a shorter segment of poly(di(methylamino)ethyl methacrylate) (PDMAEMA) was made combining ROP and atom transfer radical polymerization (ATRP). The PDMAEMA-part was then quaternized, which resulted in a cationically charged chain – a polyelectrolyte. This can then be used as an electrostatic linker allowing the PDMAEMA-PCL copolymer to be adsorbed onto the negatively charged cellulose model surface. Finally, differences between the two approaches are evaluated regarding for example surface coverage and grafting/physiosorption efficiency investigated with techniques such as atomic force microscopy (AFM).

  • Physical Tuning of Cellulose-Polymer Interactions Utilizing Cationic Block Copolymers Based on PCL and Quaternized PDMAEMA

    • In this study, the aim was to prepare and evaluate a block copolymer that can be used as a compatabilizer in cellulose fiber-reinforced biocomposites. It as an amphiphilic block copolymer consisting of poly(ε-caprolactone) (PCL), made with  ring-opening polymerization (ROP), and a shorter segment of poly(2-(dimethylamino)ethyl methacrylate) (PDMAEMA) that was synthesized with atom transfer radical polymerization (ATRP). The PDMAEMA-part was made in one single length, while the PCL-part was varied in three different lengths; in total were three block copolymers prepared. In the last step of the synthesis, the PDMAEMA-part was quaternized that turns it into a cationically charged chain – a polyelectrolyte. The block copolymers were then able to form cationic micelles in water, from where they can adsorb, under mild conditions, to anionic surfaces such as silicon oxide and cellulose-model surfaces. This provides the surface with a more hydrophobic character shown with contact angle measurements. Finally, with atomic force microscopy (AFM) force measurements, it was demonstrated that there is a clear entanglement behavior obtained between the block copolymers and a PCL surface at about 60 °C, which is of importance for the information regarding the adhesive interface in a future biocomposite.

A block copolymer consisting of PCL and a shorter segment of poly(di(methylamino)ethyl methacrylate) (PDMAEMA) was made combining ring-opening polymerization (ROP) and atom transfer radical polymerization (ATRP). The length of the PDMAEMA-part was kept constant, and the PCL-part was varied in three different lengths, yielding three separate block copolymers. As a final step, the PDMAEMA-part was quaternized, which resulted in cationically charged chains –polyelectrolytes. The charged part could then be used as an electrostatic linker allowing the PDMAEMA-PCL copolymer to be adsorbed onto negatively charged cellulose model surfaces. Finally, these cellulose model surfaces were evaluated regarding for example amount of polymer adsorbed and hydrophobic character, investigated with techniques such as quartz crystal microbalance (QCM) and contact angle measurements.

    • In this study, a comparison between covalent grafting and physical adsorption of PCL onto a nanocellulose model surface was conducted. For the covalent attachment, surface-initiated ring-opening polymerization (SI-ROP) was performed. For the physical attachment, a charged block copolymer consisting of PCL and quaternized PDMAEMA was synthesized by ROP and ATRP, and adsorbed to the cellulose. Finally, differences in between the two substrates were investigated with techniques such as AFM.

    • In this work, di- and triblock copolymers based on poly(2-(dimethylamino)ethyl methacrylate) (PDMAEMA) and poly(ε-caprolactone) (PCL) have been prepared. The PDMAEMA length was kept constant for both di- and triblock copolymers, while in the diblock copolymers the PCL length was varied in three different lengths, yielding three separate block copolymers. For the triblock blockcopolymers, on the other hand, also the PCL blocks were of the same length yielding one ABA- and one BAB-type block copolymer. In the next step, the PDMAEMA-part was quaternized to yield polyelectrolytes with either one or two charged block(s). In the final step, difference in adsorption behavior onto a negatively charged cellulose surface and subsequent alteration of surface properties was investigated. Overall, the polymers were evaluated in solid state, in solution, in water dispersion, and on cellulose surfaces with techniques including differential scanning calorimetry, size exclusion chromatography, dynamic light scattering and quartz crystal microbalance.

  • Surface Modification of Cellulose by Covalent Grafting and Physical Adsorption for Biocomposite Applications

    • There is an increasing interest to replace fossil-based materials with renewable alternatives. Cellulose fibers/nanofibrils (CNF) are sustainable options since they are biobased and biodegradable. In addition, they combine low weight with high strength; making them suitable to, for example, reinforce composites. However, to be able to use them as such, modifications are often necessary. This study therefore aimed at modifying cellulose fibers, model surfaces of cellulose and CNF. Cellulose fibers and CNF were thereafter incorporated into composite materials and evaluated.Surface-initiated ring-opening polymerization (SI-ROP) was performed to graft ε-caprolactone (ε-CL) from cellulose fibers. From these fibers, paper-sheet biocomposites were produced that could form laminate structures without the need for any addition of matrix polymer.By combining ROP and atom transfer radical polymerization (ATRP), diblock copolymers of poly(2-dimethylaminoethyl methacrylate) (PDMAEMA) and PCL were prepared. Quaternized (cationic) PDMAEMA, allowed physical adsorption of block copolymers onto anionic surfaces, and, thereby, alteration of surface energy and adhesion to a potential matrix. Furthermore, the architecture of block copolymers of PCL and PDMAEMA was varied to investigate effects on morphology/crystallinity and adsorption behavior. In addition, poly(butadiene) was also evaluated as the hydrophobic block in the form of cationic and anionic triblock copolymers.Polystyrene (PS) was covalently grafted from CNF and used as reinforcement in PS-based composites. In an attempt to determine stress transfer from matrix to CNF, a method based on Raman spectroscopy was utilized.Covalent grafting and physical adsorption of PCL from/onto CNF were compared by incorporating modified CNF in PCL matrices. Both approaches resulted in improved mechanical properties compared to unmodified CNF, but even at low amounts of modified CNF, covalent grafting gave tougher materials and indicated higher interfacial adhesion.

    • Triblock copolymers of ABA- and BAB-type consisting of poly(2-(dimethylamino)ethyl methacrylate) (PDMAEMA, A) and poly(epsilon-caprolactone) (PCL, B) have successfully been prepared. PDMAEMA-b-PCL-b-PDMAEMA (ABA) and PCL-b-PDMAEMA-b-PCL (BAB) were synthesised by a combination of ring-opening polymerisation of epsilon-CL, atom transfer radical polymerisation of DMAEMA and end-group conversion, performed through either acylation or azide-alkyne "click" chemistry. All samples were analysed by size exclusion chromatography where it was found that the evaluation of PDMAEMA-containing polymers was difficult due to the thermoresponsivity of PDMAEMA, affecting the solubility of the polymer in the temperature range at which the SEC was operated. From differential scanning calorimetry measurements it was shown that the crystallinity could be altered by changing the order of the blocks; with PDMAEMA as the outer block (ABA), the inherent crystallinity of PCL was destroyed while with PCL as the outer block (BAB), the degree of crystallinity was in the same proximity as for a PCL homopolymer.

  • Who should record surgical complications? : Results from a third-party assessment of complications after radical cystectomy

    • Objective: In Sweden complications after radical cystectomy have been reported to the nationwide population-based Swedish Cystectomy Registry since 2011. Here, validation of the reporting was assessed in two healthcare regions.Materials and methods: Complications were ascertained from patient records by a third party not involved in the care delivered to 429 randomly selected patients from 949 who had undergone radical cystectomy since 2011 in four hospitals. Without knowledge of the outcome in the primary registration, post-operative complications within 90 days post-operatively were assessed by an independent review of patient charts, and the results were compared with the primary reports in the Swedish Cystectomy Registry.Results: The third-party assessment identified post-operative complications in 310 patients (72%). Low-grade complications (Clavien-Dindo I-II) were noted in 110 (26%) of the patients in the primary registration, but increased to 182 (42%) in the validation (p < 0.00001). High-grade complications (Clavien-Dindo III-V) were reported in 113 (26%) patients in the primary registration, but in 128 (30%) of the patients in the validation (p = 0.02). According to the third-party assessment, 18 patients (4%) had Clavien-Dindo grade IV complications and 12 (3%) died within 90 days of surgery (Clavien-Dindo grade V); corresponding values in the primary registration were 15 (3%) and 9 (2%), respectively. The readmission rate within 90 days increased from 27 to 32% in the validation (p < 0.00001).Conclusions: Compared with registry data, third-party assessment revealed more complications and readmissions after radical cystectomy. Hence such evaluation may improve the validity of reported complication data.

  • Spatial Tissue Mapping on Joint Biopsies from Arthritis Patients

    • Rheumatoid arthritis (RA) is a chronic inflammatory autoimmune disease that mainly affects joints, causing discomfort and pain that severely reduces the life quality of affected individuals. Its etiology is largely unknown, but some pathophysiological mechanisms have been identified. These include formation of anti-citrullinated protein antibodies (ACPAs) and rheumatic factors (RFs), local proliferation of mesenchymal cells, and recruitment of T- and B cells to the affected synovium. Lymphocyte infiltration results in elevated levels of cytokines such as Tumor Necrosis Factor alpha (TNF-α) and interleukin signaling, which in turn triggers protease activation that gradually degrades the synovium and underlying bone. In many cases RA can be effectively managed by early diagnosis followed by treatment with disease-modifying anti-rheumatic drugs (DMARDs). However, this is not true for all patients and there is currently no cure for RA. Synovial lesions in RA patients exhibit complex histopathological manifestations involving the formation of lymphoid follicles with highly organized Ectopic Lymphoid Structures (ELS). These have been extensively studied using immunostaining and other cytological methods, either by targeting a few specific molecular markers in tissue sections or by examining homogenized suspensions of complex samples, which causes a loss of local and spatial tissue information. This thesis reports the use of Spatial Transcriptomics (ST) to study gene expression in tissue samples from RA patients while preserving spatial information. The method was applied to RA biopsies from early onset and untreated RA to late-stage established disease with edema, providing comprehensive coverage of the spatio-temporal dynamics of the inflamed joints. Paper I introduces sRIN, a novel method of assessing the quality of RNA in tissue sections that is similar to RNA Integrity Number (RIN) analysis for bulk RNA but with single-cell resolution. The aim was to find ways of analyzing clinically rare samples for further processing with ST. Paper II uses ST to study tissue samples from RA joints with long-standing disease, using Spondyloarthritis (SpA) as a disease control. The resulting comprehensive transcriptomic data were used to perform in silico immune cell prediction and revealed how immune cell infiltration in RA differs from that in SpA in more detail than was previously possible using traditional pathological methods. As a follow up, Paper III investigates inflamed RA joints in even greater detail by using several adjacent tissue sections to build a three-dimensional atlas of assumed ELS areas. Finally, Paper IV uses four distinct technologies to study untreated early onset RA patients. Spatial tissue analysis with ST was combined with single cell RNA sequencing (scRNA-Seq) of fluorescence-activated cell sorted (FACS) B cells. These two methods were complemented with immunohistochemistry (IHC) for validation and sRIN to assess the quality of the clinical samples. B cells are known to play a key role in RA by producing self-reactive antibodies. This work showed that B cell maturation and ELS formation are detectable even in early onset RA, and revealed mechanisms supporting survival niches in hyperplastic joints. Overall, these studies shed new light on the complex nature of Rheumatoid arthritis, characterize the site of infection with greater granularity than was previously possible, and reveal novel disease patterns with clinical implications that warrant further study.

    • Cellulose fibers have been successfully grafted with poly(methyl acrylate) using atom transfer radical polymerization, mediated by Me6-TREN and Cu(I)Br at ambient temperature. The initially hydrophilic cellulose was first modified by reacting the hydrozyl groups with 2-bromoisobutyryl bromide whereupon methyl acrylate was grafted from the surface. The resulting polymer-grafted papers were extremely hydrophobic, θa = 133°. FT-IR analysis indicates that the amount of grafted polymer can be controlled by adding sacrificial initiator to the polymerizing system. Size exclusion chromatography of the bulk polymer revealed narrow polydispersities and a molecular weight corresponding to the ratio [M]:[I].

    • Cellulose fibers, in the form of a conventional filter paper, have been modified by reacting the hydroxyl groups on the fiber surface with 2-bromoisobutyryl bromide, followed by grafting using ATRP conditions. The papers were first grafted with methyl acrylate (MA), rendering the paper very hydrophobic as reported in an earlier work. The papers were analyzed by gravimetry, FT-IR, ESCA, and AFM. To verify that the polymerization from the surface was living, a second layer of another, hydrophilic, polymer, 2-hydroxyethyl methacrylate (HEMA), was grafted upon the PMA layer, creating a block-copolymer graft from the fibers. After the layer of PHEMA had been attached, contact angle measurements were no longer possible, because of the absorbing nature of PHEMA-grafted layer. This indicates that a copolymer had indeed been formed on the surface. FT-IR showed a large increase in carbonyl content after the PHEMA-grafting, which further proves that a layer of PHEMA was attached to the PMA layer. This goes to show that the hydrophilic/ hydrophobic behavior of a cellulose surface can be tailored by the use of living/controlled radical polymerization methods such as ATRP.

    • Atom transfer radical polymerization (ATRP) of dendritic, aliphatic macromonomers has been investigated. The macromonomers were based on acrylate functionalized 2,2-bis(methylol)propionic acid (bis-MPA) dendrons, with a flexible spacer of 10 carbons incorporated in the structure in between the polymerizable group and the dendritic wedge. Dendronized polymers of generation one, two, and three were successfully synthesized by ATRP. The polymerizations proceeded until over 80% conversion was reached, while maintaining control over polydispersity index (PDI). Plots of ln([M](0)/[M]) vs time for the polymerization of all three macromonomers showed a linear dependence, indicating that the number of propagating radicals in the reaction solution was constant throughout the reaction, when ethyl 2-bromopropionate (EBrP) was used as an initiator (i.e., radical termination was negligible). All of the resulting polymers had low PDI values and molecular weight close to the theoretical ones. The products were analyzed by H-1 and C-13 NMR spectroscopies, size exclusion chromatography (SEC), differential scanning calorimetry (DSC), and matrix-assisted laser desorption-ionization time-of-flight (MALDI-TOF).

    • Dendritic polymers are highly branched, globular architectures with multiple representations of functional groups. These nanoscale organic frameworks continue to fascinate researchers worldwide and are today under intensive investigation in application-driven research. A large number of potential application areas have been suggested for dendritic polymers, including theranostics, biosensors, optics, adhesives and coatings. The transition from potential to real applications is strongly dictated by their commercial accessibility, scaffolding ability as well as biocompatibility. A dendritic family that fulfills these requirements is based on the 2,2-bismethylolpropionic acid (bis-MPA) monomer. This critical review is the first of its kind to cover most of the research activities generated on aliphatic polyester dendritic architectures based on bis-MPA. It is apparent that these scaffolds will continue to be in the forefront of cutting-edge research as their structural variations are endless including dendrons, dendrimers, hyperbranched polymers, dendritic-linear hybrids and their hybridization with inorganic surfaces.

    • A multifunctional initiator for ATRP has been synthesized by reacting a hyperbranched polyether, based on 3-ethyl-3-(hydroxymethyl)oxetane, with 2-bromo-isobutyrylbromide. The macroinitiator contained approximately 25 initiating sites per molecule. It was used for the atom transfer radical polymerization of methyl acrylate mediated by Cu(I)Br and tris(2-(dimethylamino)ethyl)amine (Me-6-TREN) in ethyl acetate at room temperature. This yielded a co-polymer with a dendritic-linear architecture. The large number of growing chains from each macromolecule increases the probability of inter-and intramolecular reactions. In order to control these kinds of polymerizing systems and prevent them from forming a gel, the concentration of propagating radicals must be kept low. The polymerizations under these conditions were well controlled. When a ratio of initiating sites-to-catalyst of 1:0.05 was used, the polymers from all of the reactions had a low polydispersity, ranging from 1.1 to 1.4. None of the polymerizations under these conditions gave gelation. Monomer conversions as high as 65% were reached while maintaining control over the polymerization. (C) 2002 Elsevier Science Ltd.
