    • We report an angle-resolved photoemission study of the charge stripe ordered La1.6-xNd0.4SrxCuO4 (Nd-LSCO) system. A comparative and quantitative line-shape analysis is presented as the system evolves from the overdoped regime into the charge ordered phase. On the overdoped side (x = 0.20), a normal-state antinodal spectral gap opens upon cooling below 80 K. In this process, spectral weight is preserved but redistributed to larger energies. A correlation between this spectral gap and electron scattering is found. A different line shape is observed in the antinodal region of charge ordered Nd-LSCO x = 1/8. Significant low-energy spectral weight appears to be lost. These observations are discussed in terms of spectral-weight redistribution and gapping originating from charge stripe ordering.

    • Fatigue crack growth experiments were performed on surface cracked tensile specimens of Inconel 718 at 400 degreesC. The loading was carried out at constant as well as at variable amplitude. The experimental results for the mean growth rate were compared with predictions based on data obtained from testing of compact tension specimens. Both nominal data as well as data corrected from measured crack closure were used in the predictions. The corrected data provided much better predictions than the nominal ones indicating that the level of crack closure during the testing of the surface cracked specimens was much lower than in compact tension specimens.

    • Nanofibrous filter materials were prepared by electrospinning a solution of 28 wt% poly(vinylidene fluoride) in N,N-dimethylacetamide with and without the addition of 2 wt% AgNO3, Cu(NO3)(2)center dot 2.5H(2)O or ZnCl2. X-ray diffraction, scanning electron microscopy with energy dispersive X-ray spectroscopy, inductively coupled plasma mass spectroscopy, thermogravimetric analysis, contact angle measurement, nitrogen sorption, and mercury intrusion porosimetry methods were used for the characterization of physical structure as well as the chemical composition of the electrospun materials. Particle filtration efficiency and antiviral activity against the SARS-CoV-2 alpha variant were tested in order to estimate the suitability of the prepared electrospun filter mate-rials for application as indoor air filtration systems with virucidal properties. All filter materials prepared with salts demonstrated very high particle filtration efficiency (>= 98.0%). The best antiviral activity was demonstrated by a material containing Cu(NO3)(2)center dot 2.5H(2)O in the spinning solution, which displayed the decrease in the number of infectious virions by three orders of magnitude after a contact time of 12 h. Materials with the addition of AgNO3 and ZnCl2 decreased the number of infectious virions after the same contact time by only-8 and-11 times, respectively.

  • 2-cm versus 4-cm surgical excision margins for primary cutaneous melanoma thicker than 2 mm: a randomised, multicentre trial

    • Background Optimum surgical resection margins for patients with clinical stage IIA-C cutaneous melanoma thicker than 2 mm are controversial. The aim of the study was to test whether survival was different for a wide local excision margin of 2 cm compared with a 4-cm excision margin. Methods We undertook a randomised controlled trial in nine European centres. Patients with cutaneous melanoma thicker than 2 mm, at clinical stage IIA-C, were allocated to have either a 2-cm or a 4-cm surgical resection margin. Patients were randomised in a 1:1 allocation to one of the two groups and stratified by geographic region. Randomisation was done by sealed envelope or by computer generated lists with permuted blocks. Our primary endpoint was overall survival. The trial was not masked at any stage. Analyses were by intention to treat. Adverse events were not systematically recorded. The study is registered with ClinicalTrials.gov, number NCT01183936. Findings 936 patients were enrolled from Jan 22, 1992, to May 19, 2004; 465 were randomly allocated to treatment with a 2-cm resection margin, and 471 to receive treatment with a 4-cm resection margin. One patient in each group was lost to follow-up but included in the analysis. After a median follow-up of 6.7 years (IQR 4.3-9.5) 181 patients in the 2-cm margin group and 177 in the 4-cm group had died (hazard ratio 1.05, 95% CI 0.85-1.29; p=0.64). 5-year overall survival was 65% (95% CI 60-69) in the 2-cm group and 65% (60-70) in the 4-cm group (p=0.69). Interpretation Our findings suggest that a 2-cm resection margin is sufficient and safe for patients with cutaneous melanoma thicker than 2 mm.

    • High temperature fatigue crack growth in Inconel 718 has been studied at the temperatures 450 degrees C, 500 degrees C, 550 degrees C and 650 degrees C. The tests were conducted both without hold times and with hold times of different lengths and with a mix of both. Focus has been on quantifying the effect the hold time has upon the crack growth rate and how much it damages the material. Furthermore, it has been investigated how this damage influences the actual cracking behavior, i.e. where in the loading cycle the damage contributes most to the crack growth. This damage is related to the concept of a damaged zone in front of the crack tip. The size of the damaged zone has been derived from the tests and a microscopy study to confirm the findings has also been carried out. It is found that the concept of a damaged zone can be a successful explanatory model for the observed crack growth behavior under high temperature hold time.

    • In an attempt to provide an explanation for the previously reported effect of pregnancy on the Lewis phenotype of erythrocytes, the level of Leb-active glycolipid in serum was compared with the reactions of erythrocytes, using samples obtained from 73 nonpregnant women, 74 women at the time of delivery, and 2 women at weekly intervals during their pregnancy. In this Swedish population, the frequency of the Le(a--b-) blood group increased from 11% in nonpregnant women to 36% in women at the time of delivery. Among Le(a--b+) women of all ABO groups, those who were A1 most often became (Leb-) during pregnancy. The change in phenotype occurred as early as the 24th week of gestation; the Leb antigen was again detectable within 6 weeks after delivery. The concentration of Leb glycolipid in serum, as measured by radioimmunoassay, decreased only slightly during pregnancy. The repartition of glycolipids, secondary to the increased ratio of lipoprotein to red cell mass that occurs during pregnancy, may account for the relative lack of Lewis glycolipid on erythrocytes.

  • Monitoring of kindreds with hereditary predisposition for cutaneous melanoma and dysplastic nevus syndrome: results of a Swedish preventive program

    • PURPOSE: To evaluate a program initiated in 1987 by the Swedish Melanoma Study Group aiming to provide preventive surveillance to kindreds with hereditary cutaneous melanoma and dysplastic nevus syndrome. PATIENTS AND METHODS: Overall, 2,080 individuals belonging to 280 melanoma families were followed for 14 years between 1987 and 2001 at 12 participating centers. Data were registered in a central database. RESULTS: Among 1,912 skin lesions excised during follow-up, 41 melanomas were removed in 32 individuals. Of these, 15 (37%) were in situ melanomas and 26 (63%) invasive melanomas. The median tumor thickness of invasive melanomas was 0.5 mm. Ulceration was absent in 24 of 26 invasive melanomas (92%) and 12 (46%) lacked vertical growth phase. Compared with melanomas in the general Swedish population, the melanomas identified in these kindreds during follow-up had better prognostic characteristics. All melanomas except one were diagnosed in families with two or more first-degree relatives with melanoma. Diagnosis of melanoma occurred in three of eight kindreds with germline CDKN2A mutations, supporting that families with such mutations are at increased risk for melanoma development. Of the 32 individuals who developed melanoma during follow-up, 21 (66%) had had at least one previously diagnosed melanoma. CONCLUSION: This study shows that a coordinated program aimed at detecting and offering skin surveillance in kindreds with hereditary cutaneous melanoma results in a low incidence of melanomas during the follow-up period and that the tumors that do arise have favorable prognostic characteristics.

    • Nucleosome assembly proteins (NAPs) are histone chaperones with an important role in chromatin structure and epigenetic regulation of gene expression. We find that high gene expression levels of mouse Nap1l3 are restricted to haematopoietic stem cells (HSCs) in mice. Importantly, with shRNA or CRISPR-Cas9 mediated loss of function of mouse Nap1l3 and with overexpression of the gene, the number of colony-forming cells and myeloid progenitor cells in vitro are reduced. This manifests as a striking decrease in the number of HSCs, which reduces their reconstituting activities in vivo. Downregulation of human NAP1L3 in umbilical cord blood (UCB) HSCs impairs the maintenance and proliferation of HSCs both in vitro and in vivo. NAP1L3 downregulation in UCB HSCs causes an arrest in the G0 phase of cell cycle progression and induces gene expression signatures that significantly correlate with downregulation of gene sets involved in cell cycle regulation, including E2F and MYC target genes. Moreover, we demonstrate that HOXA3 and HOXA5 genes are markedly upregulated when NAP1L3 is suppressed in UCB HSCs. Taken together, our findings establish an important role for NAP1L3 in HSC homeostasis and haematopoietic differentiation.

  • Expectancy effects on serotonin and dopamine transporters during SSRI treatment of social anxiety disorder : a randomized clinical trial

    • It has been extensively debated whether selective serotonin reuptake inhibitors (SSRIs) are more efficacious than placebo in affective disorders, and it is not fully understood how SSRIs exert their beneficial effects. Along with serotonin transporter blockade, altered dopamine signaling and psychological factors may contribute. In this randomized clinical trial of participants with social anxiety disorder (SAD) we investigated how manipulation of verbally-induced expectancies, vital for placebo response, affect brain monoamine transporters and symptom improvement during SSRI treatment. Twenty-seven participants with SAD (17 men, 10 women), were randomized, to 9 weeks of overt or covert treatment with escitalopram 20 mg. The overt group received correct treatment information whereas the covert group was treated deceptively with escitalopram, described as an active placebo in a cover story. Before and after treatment, patients underwent positron emission tomography (PET) assessments with the [C-11]DASB and [C-11]PE2I radiotracers, probing brain serotonin (SERT) and dopamine (DAT) transporters. SAD symptoms were measured by the Liebowitz Social Anxiety Scale. Overt was superior to covert SSRI treatment, resulting in almost a fourfold higher rate of responders. PET results showed that SERT occupancy after treatment was unrelated to anxiety reduction and equally high in both groups. In contrast, DAT binding decreased in the right putamen, pallidum, and the left thalamus with overt SSRI treatment, and increased with covert treatment, resulting in significant group differences. DAT binding potential changes in these regions correlated negatively with symptom improvement. Findings support that the anxiolytic effects of SSRIs involve psychological factors contingent on dopaminergic neurotransmission while serotonin transporter blockade alone is insufficient for clinical response.

    • Leucine twenty homeobox gene (LEUTX) is a PAIRED (PRD)-like homeobox gene that is expressed nearly exclusively in human preimplantation embryos. We previously identified a novel transcription start site for the predicted human LEUTX gene based on the transcriptional analysis of human preimplantation embryos. The novel variant encodes a protein with a complete homeodomain. Here we provide a detailed description of the molecular cloning of the complete homeodomain-containing LEUTX. Using a human embryonic stem cell overexpression model we show that the complete homeodomain isoform is functional and sufficient to activate the transcription of a large fraction of the genes found upregulated in human embryo genome activation, whereas the previously predicted partial homeodomain isoform is largely inactive. Another PRD-like transcription factor, DPRX, appears as a powerful repressor of transcription. We propose a two-stage model of human EGA in which LEUTX acts as a transcriptional activator at 4-cell stage, and DPRX as a balancing repressor at 8-cell stage. We conclude that LEUTX is a candidate regulator of human embryo genome activation.

  • Using High Energy Angle Resolved Photoelectron Spectroscopy to Reveal the Charge Density in Solids

    • The charge density in solids is a fundamental parameter. Here we demonstrate that the charge density can be determined by the use of angle resolved photoelectron spectroscopy. The method, which involves a Fourier-like transform from momentum space to real space, is demonstrated by utilizing soft x-ray angle resolved photoelectron spectroscopy to sample the complete three-dimensional Brillouin zone of copper. It is also shown that this can be done in an energy resolved way as to extract the charge density contribution from states of a particular energy.

    • An engineering method based on fracture mechanics is suggested for life predictions of components of nodular cast iron. The method should be applicable to any structure containing defects which can cause crack initiation. Component testing was performed and the observed results were compared with model predictions. Fatigue crack growth data were obtained by testing on CT specimens even for load ratios R < 0. By accounting for crack closure the R-effect on the growth rates can also be effectively eliminated for this type of material.

    • Results are presented for fatigue crack growth at elevated temperatures during both constant amplitude and variable amplitude loading. A careful experimental investigation is presented to show that the concept of the effective stress-intensity factor range DeltaK(I)(eff) can be used to eliminate the load ratio effect on fatigue data and produce one single set of crack growth data. The fatigue crack propagation data corrected for crack closure is then used for comparison of mean crack propagation rates for variable amplitude loads with simple time histories. It is concluded that measured crack closure can not fully explain the discrepancies between measured and predicted propagation rates. A reduction factor fitted to the experimental data could be used to successfully correlate the results.

    • The influence of microstructure i.e. nodules (voids) on the fatigue threshold value in nodular cast iron is numerically investigated. It is qualitatively shown that the fatigue threshold value is unaffected by the individual nodules and that the threshold value is proportional to the bulk Young's modulus. Furthermore it is shown that nodules (voids) do not influence the crack tip conditions (stress-intensity factor and crack tip opening displacement) if the voids do not lie very close to the tip for physically long cracks. It is concluded that linear elastic fracture mechanics is valid for these types of materials and no influence of the nodules need be taken into count for fatigue calculations when dealing with long cracks.
