  • Identification of four novel susceptibility loci for oestrogen receptor negative breast cancer

    • Common variants in 94 loci have been associated with breast cancer including 15 loci with genome-wide significant associations (P<5 x 10(-8)) with oestrogen receptor (ER)-negative breast cancer and BRCA1-associated breast cancer risk. In this study, to identify new ER-negative susceptibility loci, we performed a meta-analysis of 11 genome-wide association studies (GWAS) consisting of 4,939 ER-negative cases and 14,352 controls, combined with 7,333 ER-negative cases and 42,468 controls and 15,252 BRCA1 mutation carriers genotyped on the iCOGS array. We identify four previously unidentified loci including two loci at 13q22 near KLF5, a 2p23.2 locus near WDR43 and a 2q33 locus near PPIL3 that display genome-wide significant associations with ER-negative breast cancer. In addition, 19 known breast cancer risk loci have genome-wide significant associations and 40 had moderate associations (P<0.05) with ER-negative disease. Using functional and eQTL studies we implicate TRMT61B and WDR43 at 2p23.2 and PPIL3 at 2q33 in ER-negative breast cancer aetiology. All ER-negative loci combined account for similar to 11% of familial relative risk for ER-negative disease and may contribute to improved ER-negative and BRCA1 breast cancer risk prediction.

  • Search for Na-22 in novae supported by a novel method for measuring femtosecond nuclear lifetimes

    • Classical novae are thermonuclear explosions in stellar binary systems, and important sources of Al-26 and Na-22. While ? rays from the decay of the former radioisotope have been observed throughout the Galaxy, Na-22 remains untraceable. Its half-life (2.6 yr) would allow the observation of its 1.275 MeV ?-ray line from a cosmic source. However, the prediction of such an observation requires good knowledge of its nucleosynthesis. The Na-22(p, ?)Mg-23 reaction remains the only source of large uncertainty about the amount of Na-22 ejected. Its rate is dominated by a single resonance on the short-lived state at 7785.0(7) keV in Mg-23. Here, we propose a combined analysis of particle-particle correlations and velocity-difference profiles to measure femtosecond nuclear lifetimes. The application of this method to the study of the Mg-23 states, places strong limits on the amount of Na-22 produced in novae and constrains its detectability with future space-borne observatories. The authors report a particle-particle correlation and velocity-difference profile method to measure nuclear lifetime. The results obtained for excited states of 23Mg are used to constrain the production of 22Na in the astrophysical novae explosions.

  • No clinical utility of KRAS variant rs61764370 for ovarian or breast cancer

    • Objective Clinical genetic testing is commercially available for rs61764370, an inherited variant residing in a KRAS 3′ UTR microRNA binding site, based on suggested associations with increased ovarian and breast cancer risk as well as with survival time. However, prior studies, emphasizing particular subgroups, were relatively small. Therefore, we comprehensively evaluated ovarian and breast cancer risks as well as clinical outcome associated with rs61764370. Methods Centralized genotyping and analysis were performed for 140,012 women enrolled in the Ovarian Cancer Association Consortium (15,357 ovarian cancer patients; 30,816 controls), the Breast Cancer Association Consortium (33,530 breast cancer patients; 37,640 controls), and the Consortium of Modifiers of BRCA1 and BRCA2 (14,765 BRCA1 and 7904 BRCA2 mutation carriers). Results We found no association with risk of ovarian cancer (OR = 0.99, 95% CI 0.94-1.04, p = 0.74) or breast cancer (OR = 0.98, 95% CI 0.94-1.01, p = 0.19) and results were consistent among mutation carriers (BRCA1, ovarian cancer HR = 1.09, 95% CI 0.97-1.23, p = 0.14, breast cancer HR = 1.04, 95% CI 0.97-1.12, p = 0.27; BRCA2, ovarian cancer HR = 0.89, 95% CI 0.71-1.13, p = 0.34, breast cancer HR = 1.06, 95% CI 0.94-1.19, p = 0.35). Null results were also obtained for associations with overall survival following ovarian cancer (HR = 0.94, 95% CI 0.83-1.07, p = 0.38), breast cancer (HR = 0.96, 95% CI 0.87-1.06, p = 0.38), and all other previously-reported associations. Conclusions rs61764370 is not associated with risk of ovarian or breast cancer nor with clinical outcome for patients with these cancers. Therefore, genotyping this variant has no clinical utility related to the prediction or management of these cancers.

  • Climate for Culture : assessing the impact of climate change on the future indoor climate in historic buildings using simulations

    • Background The present study reports results from the large-scale integrated EU project "Climate for Culture". The full name, or title, of the project is Climate for Culture: damage risk assessment, economic impact and mitigation strategies for sustainable preservation of cultural heritage in times of climate change. This paper focusses on implementing high resolution regional climate models together with new building simulation tools in order to predict future outdoor and indoor climate conditions. The potential impact of gradual climate change on historic buildings and on the vast collections they contain has been assessed. Two moderate IPCC emission scenarios A1B and RCP 4.5 were used to predict indoor climates in historic buildings from the recent past until the year 2100. Risks to the building and to the interiors with valuable artifacts were assessed using damage functions. A set of generic building types based on data from existing buildings were used to transfer outdoor climate conditions to indoor conditions using high resolution climate projections for Europe and the Mediterranean. Results The high resolution climate change simulations have been performed with the regional climate model REMO over the whole of Europe including the Mediterranean region. Whole building simulation tools and a simplified building model were developed for historic buildings; they were forced with high resolution climate simulations. This has allowed maps of future climate-induced risks for historic buildings and their interiors to be produced. With this procedure future energy demands for building control can also be calculated. Conclusion With the newly developed method described here not only can outdoor risks for cultural heritage assets resulting from climate change be assessed, but also risks for indoor collections. This can be done for individual buildings as well as on a larger scale in the form of European risk maps. By using different standardized and exemplary artificial buildings in modelling climate change impact, a comparison between different regions in Europe has become possible for the first time. The methodology will serve heritage owners and managers as a decision tool, helping them to plan more effectively mitigation and adaption measures at various levels.

  • ELIXIR and Toxicology : a community in development

    • Toxicology has been an active research field for many decades, with academic, industrial and government involvement. Modern omics and computational approaches are changing the field, from merely disease-specific observational models into target-specific predictive models. Traditionally, toxicology has strong links with other fields such as biology, chemistry, pharmacology and medicine. With the rise of synthetic and new engineered materials, alongside ongoing prioritisation needs in chemical risk assessment for existing chemicals, early predictive evaluations are becoming of utmost importance to both scientific and regulatory purposes. ELIXIR is an intergovernmental organisation that brings together life science resources from across Europe. To coordinate the linkage of various life science efforts around modern predictive toxicology, the establishment of a new ELIXIR Community is seen as instrumental. In the past few years, joint efforts, building on incidental overlap, have been piloted in the context of ELIXIR. For example, the EU-ToxRisk, diXa, HeCaToS, transQST, and the nanotoxicology community have worked with the ELIXIR TeSS, Bioschemas, and Compute Platforms and activities. In 2018, a core group of interested parties wrote a proposal, outlining a sketch of what this new ELIXIR Toxicology Community would look like. A recent workshop (held September 30th to October 1st, 2020) extended this into an ELIXIR Toxicology roadmap and a shortlist of limited investment-high gain collaborations to give body to this new community. This Whitepaper outlines the results of these efforts and defines our vision of the ELIXIR Toxicology Community and how it complements other ELIXIR activities.

    • We consider infinitely renormalizable Lorenz maps with real critical exponent alpha > 1 of certain monotone combinatorial types. We prove the existence of periodic points of the renormalization operator, and that each map in the limit set of renormalization has an associated two-dimensional strong unstable manifold. For monotone families of Lorenz maps we prove that each infinitely renormalizable combinatorial type has a unique representative within the family. We also prove that each infinitely renormalizable map has no wandering intervals, is ergodic, and has a uniquely ergodic minimal Cantor attractor of measure zero.

    • This paper discusses the measurement of globalization with a view to advancing the construction of globalization indices. It critically analyzes the types of indices that can contribute to knowledge and policy on globalization. Three issues are particularly highlighted: (a) the focus of measurement (i.e. on activities or policies); (b) the dimensions of measurement (i.e. cultural, ecological, economic, political, and/or social); and (c) the units of measurement (i.e. local, national, regional, and/or global). This paper argues that a workable forward strategy should not seek to identify the single best composite globalization index, but rather should work in an interdisciplinary mode towards a set of complementary globalization indices. These quantitative analyses can then be productively blended with qualitative approaches in a fuller assessment of globalization's extent and impact.

  • The Role of Coastal Yedoma Deposits and Continental Shelf Sediments in the Arctic Ocean Silicon Cycle

    • The availability of silicon (Si) in the ocean plays an important role in regulating biogeochemical and ecological processes. The Si budget of the Arctic Ocean appears balanced, with inputs equivalent to outputs, though it is unclear how a changing climate might aggravate this balance. In this study, we focus on Si cycling in Arctic coastal areas and continental shelf sediments to better constrain the Arctic Ocean Si budget. We provide the first estimate of amorphous Si (ASi) loading from erosion of coastal Yedoma deposits (30-90 Gmol yr-1), demonstrating comparable rates to particulate Si loading from rivers (10-90 Gmol yr-1). We found a positive relationship between surface sediment ASi and organic matter content on continental shelves. Combining these values with published Arctic shelf sediment properties and burial rates we estimate 70 Gmol Si yr-1 is buried on Arctic continental shelves, equivalent to 4.5% of all Si inputs to the Arctic Ocean. Sediment dissolved Si fluxes increased with distance from river mouths along cruise transects of shelf regions influenced by major rivers in the Laptev and East Siberian seas. On an annual basis, we estimate that Arctic shelf sediments recycle approximately up to twice as much DSi (680 Gmol Si) as is loaded from rivers (340-500 Gmol Si).   • Frequency-Independent Terahertz Anomalous Hall Effect in DyCo5, Co32Fe68, and Gd27Fe73 Thin Films from DC to 40 THz

    • The anomalous Hall effect (AHE) is a fundamental spintronic charge-to-charge-current conversion phenomenon and closely related to spin-to-charge-current conversion by the spin Hall effect. Future high-speed spintronic devices will crucially rely on such conversion phenomena at terahertz (THz) frequencies. Here, it is revealed that the AHE remains operative from DC up to 40 THz with a flat frequency response in thin films of three technologically relevant magnetic materials: DyCo5, Co32Fe68, and Gd27Fe73. The frequency-dependent conductivity-tensor elements sigma(xx) and sigma(yx) are measured, and good agreement with DC measurements is found. The experimental findings are fully consistent with ab initio calculations of sigma(yx) for CoFe and highlight the role of the large Drude scattering rate (approximate to 100 THz) of metal thin films, which smears out any sharp spectral features of the THz AHE. Finally, it is found that the intrinsic contribution to the THz AHE dominates over the extrinsic mechanisms for the Co32Fe68 sample. The results imply that the AHE and related effects such as the spin Hall effect are highly promising ingredients of future THz spintronic devices reliably operating from DC to 40 THz and beyond.

    • Extracellular vesicles (EVs), through their complex cargo, can reflect the state of their cell of origin and change the functions and phenotypes of other cells. These features indicate strong biomarker and therapeutic potential and have generated broad interest, as evidenced by the steady year-on-year increase in the numbers of scientific publications about EVs. Important advances have been made in EV metrology and in understanding and applying EV biology. However, hurdles remain to realising the potential of EVs in domains ranging from basic biology to clinical applications due to challenges in EV nomenclature, separation from non-vesicular extracellular particles, characterisation and functional studies. To address the challenges and opportunities in this rapidly evolving field, the International Society for Extracellular Vesicles (ISEV) updates its ‘Minimal Information for Studies of Extracellular Vesicles’, which was first published in 2014 and then in 2018 as MISEV2014 and MISEV2018, respectively. The goal of the current document, MISEV2023, is to provide researchers with an updated snapshot of available approaches and their advantages and limitations for production, separation and characterisation of EVs from multiple sources, including cell culture, body fluids and solid tissues. In addition to presenting the latest state of the art in basic principles of EV research, this document also covers advanced techniques and approaches that are currently expanding the boundaries of the field. MISEV2023 also includes new sections on EV release and uptake and a brief discussion of in vivo approaches to study EVs. Compiling feedback from ISEV expert task forces and more than 1000 researchers, this document conveys the current state of EV research to facilitate robust scientific discoveries and move the field forward even more rapidly.

  • Renormalization of Lorenz Maps

    • This thesis is a study of the renormalization operator on Lorenz αmaps with a critical point. Lorenz maps arise naturally as first-return maps for three-dimensional geometric Lorenz flows. Renormalization is a tool for analyzing the microscopic geometry of dynamical systems undergoing a phase transition. In the first part we develop new tools to study the limit set of renormalization for Lorenz maps whose combinatorics satisfy a long return condition. This combinatorial condition leads to the construction of a relatively compact subset of Lorenz maps which is essentially invariant under renormalization. From here we can deduce topological properties of the limit set (e.g. existence of periodic points of renormalization) as well as measure theoretic properties of infinitely renormalizable maps (e.g. existence of uniquely ergodic Cantor attractors). After this, we show how Martens’ decompositions can be used to study the differentiable structure of the limit set of renormalization. We prove that each point in the limit set has a global two-dimensional unstable manifold which is a graph and that the intersection of an unstable manifold with the domain of renormalization is a Cantor set. All results in this part are stated for arbitrary real critical exponents  α> 1. In the second part we give a computer assisted proof of the existence of a hyperbolic fixed point for the renormalization operator on Lorenz maps of the simplest possible nonunimodal combinatorial type. We then show how this can be used to deduce both universality and rigidity for maps with the same combinatorial type as the fixed point. The results in this part are only stated for critical exponenta α= 2.
