    • The PREDICTS project-Projecting Responses of Ecological Diversity In Changing Terrestrial Systems (www.predicts.org.uk)-has collated from published studies a large, reasonably representative database of comparable samples of biodiversity from multiple sites that differ in the nature or intensity of human impacts relating to land use. We have used this evidence base to develop global and regional statistical models of how local biodiversity responds to these measures. We describe and make freely available this 2016 release of the database, containing more than 3.2 million records sampled at over 26,000 locations and representing over 47,000 species. We outline how the database can help in answering a range of questions in ecology and conservation biology. To our knowledge, this is the largest and most geographically and taxonomically representative database of spatial comparisons of biodiversity that has been collated to date; it will be useful to researchers and international efforts wishing to model and understand the global status of biodiversity.

  • IceCube search for neutrinos from GRB 221009A

    •  GRB 221009A is the brightest Gamma Ray Burst (GRB) ever observed. The observed extremelyhigh flux of high and very-high-energy photons provide a unique opportunity to probe the predictedneutrino counterpart to the electromagnetic emission. We have used a variety of methods to searchfor neutrinos in coincidence with the GRB over several time windows during the precursor, promptand afterglow phases of the GRB. MeV scale neutrinos are studied using photo-multiplier ratescalers which are normally used to search for galactic core-collapse supernovae neutrinos. GeVneutrinos are searched starting with DeepCore triggers. These events don’t have directionallocalization, but instead can indicate an excess in the rate of events. 10 GeV - 1 TeV and >TeVneutrinos are searched using traditional neutrino point source methods which take into accountthe direction and time of events with DeepCore and the entire IceCube detector respectively. The>TeV results include both a fast-response analysis conducted by IceCube in real-time with timewindows of T0 − 1 to T0 + 2 hours and T0 ± 1 day around the time of GRB 221009A, as well asan offline analysis with 3 new time windows up to a time window of T0 − 1 to T0 + 14 days, thelongest time period we consider. The combination of observations by IceCube covers 9 ordersof magnitude in neutrino energy, from MeV to PeV, placing upper limits across the range forpredicted neutrino emission.

  • Robust estimation of bacterial cell count from optical density

    • Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data.

    • The International Cancer Genome Consortium (ICGC) was launched to coordinate large-scale cancer genome studies in tumours from 50 different cancer types and/or subtypes that are of clinical and societal importance across the globe. Systematic studies of more than 25,000 cancer genomes at the genomic, epigenomic and transcriptomic levels will reveal the repertoire of oncogenic mutations, uncover traces of the mutagenic influences, define clinically relevant subtypes for prognosis and therapeutic management, and enable the development of new cancer therapies.

  • Determinants of morbidity and mortality following emergency abdominal surgery in children in low-income and middle-income countries

    • Background: Child health is a key priority on the global health agenda, yet the provision of essential and emergency surgery in children is patchy in resource-poor regions. This study was aimed to determine the mortality risk for emergency abdominal paediatric surgery in low-income countries globally.Methods: Multicentre, international, prospective, cohort study. Self-selected surgical units performing emergency abdominal surgery submitted prespecified data for consecutive children aged <16 years during a 2-week period between July and December 2014. The United Nation's Human Development Index (HDI) was used to stratify countries. The main outcome measure was 30-day postoperative mortality, analysed by multilevel logistic regression.Results: This study included 1409 patients from 253 centres in 43 countries; 282 children were under 2 years of age. Among them, 265 (18.8%) were from low-HDI, 450 (31.9%) from middle-HDI and 694 (49.3%) from high-HDI countries. The most common operations performed were appendectomy, small bowel resection, pyloromyotomy and correction of intussusception. After adjustment for patient and hospital risk factors, child mortality at 30 days was significantly higher in low-HDI (adjusted OR 7.14 (95% CI 2.52 to 20.23), p<0.001) and middle-HDI (4.42 (1.44 to 13.56), p=0.009) countries compared with high-HDI countries, translating to 40 excess deaths per 1000 procedures performed.Conclusions: Adjusted mortality in children following emergency abdominal surgery may be as high as 7 times greater in low-HDI and middle-HDI countries compared with high-HDI countries. Effective provision of emergency essential surgery should be a key priority for global child health agendas.

  • Pan-cancer analysis of whole genomes

    • Cancer is driven by genetic change, and the advent of massively parallel sequencing has enabled systematic documentation of this variation at the whole-genome scale1–3. Here we report the integrative analysis of 2,658 whole-cancer genomes and their matching normal tissues across 38 tumour types from the Pan-Cancer Analysis of Whole Genomes (PCAWG) Consortium of the International Cancer Genome Consortium (ICGC) and The Cancer Genome Atlas (TCGA). We describe the generation of the PCAWG resource, facilitated by international data sharing using compute clouds. On average, cancer genomes contained 4–5 driver mutations when combining coding and non-coding genomic elements; however, in around 5% of cases no drivers were identified, suggesting that cancer driver discovery is not yet complete. Chromothripsis, in which many clustered structural variants arise in a single catastrophic event, is frequently an early event in tumour evolution; in acral melanoma, for example, these events precede most somatic point mutations and affect several cancer-associated genes simultaneously. Cancers with abnormal telomere maintenance often originate from tissues with low replicative activity and show several mechanisms of preventing telomere attrition to critical levels. Common and rare germline variants affect patterns of somatic mutation, including point mutations, structural variants and somatic retrotransposition. A collection of papers from the PCAWG Consortium describes non-coding mutations that drive cancer beyond those in the TERT promoter4; identifies new signatures of mutational processes that cause base substitutions, small insertions and deletions and structural variation5,6; analyses timings and patterns of tumour evolution7; describes the diverse transcriptional consequences of somatic mutation on splicing, expression levels, fusion genes and promoter activity8,9; and evaluates a range of more-specialized features of cancer genomes8,10–18.

  • Food systems for sustainable development : proposals for a profound four-part transformation

    • Evidence shows the importance of food systems for sustainable development: they are at the nexus that links food security, nutrition, and human health, the viability of ecosystems, climate change, and social justice. However, agricultural policies tend to focus on food supply, and sometimes, on mechanisms to address negative externalities. We propose an alternative. Our starting point is that agriculture and food systems' policies should be aligned to the 2030 Agenda for Sustainable Development. This calls for deep changes in comparison with the paradigms that prevailed when steering the agricultural change in the XXth century. We identify the comprehensive food systems transformation that is needed. It has four parts: first, food systems should enable all people to benefit from nutritious and healthy food. Second, they should reflect sustainable agricultural production and food value chains. Third, they should mitigate climate change and build resilience. Fourth, they should encourage a renaissance of rural territories. The implementation of the transformation relies on (i) suitable metrics to aid decision-making, (ii) synergy of policies through convergence of local and global priorities, and (iii) enhancement of development approaches that focus on territories. We build on the work of the Milano Group, an informal group of experts convened by the UN Secretary General in Milan in 2015. Backed by a literature review, what emerges is a strategic narrative linking climate, agriculture and food, and calling for a deep transformation of food systems at scale. This is critical for achieving the Sustainable Development Goals and the Paris Agreement. The narrative highlights the needed consistency between global actions for sustainable development and numerous local-level innovations. It emphasizes the challenge of designing differentiated paths for food systems transformation responding to local and national expectations. Scientific and operational challenges are associated with the alignment and arbitration of local action within the context of global priorities.

  • Strength of forest edge effects on litter-dwelling macro-arthropods across Europe is influenced by forest age and edge properties

    • AimForests are highly fragmented across Western Europe, making forest edges important features in many agricultural landscapes. Forest edges are subject to strong abiotic gradients altering the forest environment and resulting in strong biotic gradients. This has the potential to change the forest's capacity to provide multiple ecosystem services such as nutrient cycling, carbon sequestration and natural pest control. Soil organisms play a key role in this perspective; however, these taxa are rarely considered in forest edge research.LocationA latitudinal gradient of 2,000 km across Western Europe.MethodsWe sampled six dominant taxa of litter-dwelling macro-arthropods (carabid beetles, spiders, harvestmen, centipedes, millipedes and woodlice) in forest edges and interiors of 192 forest fragments in 12 agricultural landscapes. We related their abundance and community composition to distance from the edge and the interaction with forest age, edge orientation and edge contrast (contrast between land use types at either side of the edge).ResultsThree out of six macro-arthropod taxa have higher activity-density in forest edges compared to forest interiors. The abundance patterns along forest edge-to-interior gradients interacted with forest age. Forest age and edge orientation also influenced within-fragment compositional variation along the forest edge-to-interior gradient. Edge contrast influenced abundance gradients of generalist predators. In general, older forest fragments, south-oriented edges and edges along structurally more continuous land use (lower contrast between forest and adjacent land use) resulted in stronger edge-to-interior gradients while recent forests, north-oriented edges and sharp land use edges induced similarity between forest edge and interior along the forest edge-to-interior gradients in terms of species activity-density and composition.Main conclusionsEdge effects on litter-dwelling macro-arthropods are anticipated to feedback on important ecosystem services such as nutrient cycling, carbon sequestration and natural pest control from small forest fragments.

    • High-resolution satellite images of ocean color and sea surface temperature reveal an abundance of ocean fronts, vortices and filaments at scales below 10 km but measurements of ocean surface dynamics at these scales are rare. There is increasing recognition of the role played by small scale ocean processes in ocean-atmosphere coupling, upper-ocean mixing and ocean vertical transports, with advanced numerical models and in situ observations highlighting fundamental changes in dynamics when scales reach 1 km. Numerous scientific publications highlight the global impact of small oceanic scales on marine ecosystems, operational forecasts and long-term climate projections through strong ageostrophic circulations, large vertical ocean velocities and mixed layer re-stratification. Small-scale processes particularly dominate in coastal, shelf and polar seas where they mediate important exchanges between land, ocean, atmosphere and the cryosphere e.g. freshwater, pollutants. As numerical models continue to evolve towards finer spatial resolution and increasingly complex coupled atmosphere-wave-ice-ocean systems, modern observing capability lags behind, unable to deliver the high-resolution synoptic measurements of total currents, wind vectors and waves needed to advance understanding, develop better parameterizations and improve model validations, forecasts and projections. SEASTAR is a satellite mission concept that proposes to directly address this critical observational gap with synoptic two-dimensional imaging of total ocean surface current vectors and wind vectors at 1 km resolution and coincident directional wave spectra. Based on major recent advances in squinted along-track Synthetic Aperture Radar interferometry, SEASTAR is an innovative, mature concept with unique demonstrated capabilities, seeking to proceed towards spaceborne implementation within Europe and beyond.

    • The Thermal Infrared Visual Object Tracking challenge 2015, VOTTIR2015, aims at comparing short-term single-object visual trackers that work on thermal infrared (TIR) sequences and do not apply prelearned models of object appearance. VOT-TIR2015 is the first benchmark on short-term tracking in TIR sequences. Results of 24 trackers are presented. For each participating tracker, a short description is provided in the appendix. The VOT-TIR2015 challenge is based on the VOT2013 challenge, but introduces the following novelties: (i) the newly collected LTIR (Linköping TIR) dataset is used, (ii) the VOT2013 attributes are adapted to TIR data, (iii) the evaluation is performed using insights gained during VOT2013 and VOT2014 and is similar to VOT2015.

    • The Visual Object Tracking challenge 2015, VOT2015, aims at comparing short-term single-object visual trackers that do not apply pre-learned models of object appearance. Results of 62 trackers are presented. The number of tested trackers makes VOT 2015 the largest benchmark on short-term tracking to date. For each participating tracker, a short description is provided in the appendix. Features of the VOT2015 challenge that go beyond its VOT2014 predecessor are: (i) a new VOT2015 dataset twice as large as in VOT2014 with full annotation of targets by rotated bounding boxes and per-frame attribute, (ii) extensions of the VOT2014 evaluation methodology by introduction of a new performance measure. The dataset, the evaluation kit as well as the results are publicly available at the challenge website(1).

    • Throughout its history, Operational Research has evolved to include methods, models and algorithms that have been applied to a wide range of contexts. This encyclopedic article consists of two main sections: methods and applications. The first summarises the up-to-date knowledge and provides an overview of the state-of-the-art methods and key developments in the various subdomains of the field. The second offers a wide-ranging list of areas where Operational Research has been applied. The article is meant to be read in a nonlinear fashion and used as a point of reference by a diverse pool of readers: academics, researchers, students, and practitioners. The entries within the methods and applications sections are presented in alphabetical order. The authors dedicate this paper to the 2023 Turkey/Syria earthquake victims. We sincerely hope that advances in OR will play a role towards minimising the pain and suffering caused by this and future catastrophes.

    • The Cancer Genome Atlas (TCGA) Research Network has profiled and analyzed large numbers of human tumors to discover molecular aberrations at the DNA, RNA, protein and epigenetic levels. The resulting rich data provide a major opportunity to develop an integrated picture of commonalities, differences and emergent themes across tumor lineages. The Pan-Cancer initiative compares the first 12 tumor types profiled by TCGA. Analysis of the molecular aberrations and their functional roles across tumor types will teach us how to extend therapies effective in one cancer type to others with a similar genomic profile. © 2013 Nature America, Inc.     • Detailed knowledge of the shape of the seafloor is crucial to humankind. Bathymetry data is critical for safety of navigation and is used for many other applications. In an era of ongoing environmental degradation worldwide, bathymetry data (and the knowledge derived from it) play a pivotal role in using and managing the world's oceans in a way that is in accordance with the United Nations Sustainable Development Goal 14 - conserve and sustainably use the oceans, seas and marine resources for sustainable development. However, the vast majority of our oceans is still virtually unmapped, unobserved, and unexplored. Only a small fraction of the seafloor has been systematically mapped by direct measurement. The remaining bathymetry is predicted from satellite altimeter data, providing only an approximate estimation of the shape of the seafloor. Several global and regional initiatives are underway to change this situation. This paper presents a selection of these initiatives as best practice examples for bathymetry data collection, compilation and open data sharing as well as the Nippon Foundation-GEBCO (The General Bathymetric Chart of the Oceans) Seabed 2030 Project that complements and leverages these initiatives and promotes international collaboration and partnership. Several non-traditional data collection opportunities are looked at that are currently gaining momentum as well as new and innovative technologies that can increase the efficiency of collecting bathymetric data. Finally, recommendations are given toward a possible way forward into the future of seafloor mapping and toward achieving the goal of a truly global ocean bathymetry.

    • Most of the dozens of essential climate variables monitored each year in this report continued to follow their long-term trends in 2014, with several setting new records. Carbon dioxide, methane, and nitrous oxide-the major greenhouse gases released into Earth's atmosphere-once again all reached record high average atmospheric concentrations for the year. Carbon dioxide increased by 1.9 ppm to reach a globally averaged value of 397.2 ppm for 2014. Altogether, 5 major and 15 minor greenhouse gases contributed 2.94 W m-2 of direct radiative forcing, which is 36% greater than their contributions just a quarter century ago. Accompanying the record-high greenhouse gas concentrations was nominally the highest annual global surface temperature in at least 135 years of modern record keeping, according to four independent observational analyses. The warmth was distributed widely around the globe's land areas, Europe observed its warmest year on record by a large margin, with close to two dozen countries breaking their previous national temperature records; many countries in Asia had annual temperatures among their 10 warmest on record; Africa reported above-average temperatures across most of the continent throughout 2014; Australia saw its third warmest year on record, following record heat there in 2013; Mexico had its warmest year on record; and Argentina and Uruguay each had their second warmest year on record. Eastern North America was the only major region to observe a below-average annual temperature. But it was the oceans that drove the record global surface temperature in 2014. Although 2014 was largely ENSO-neutral, the globally averaged sea surface temperature (SST) was the highest on record. The warmth was particularly notable in the North Pacific Ocean where SST anomalies signaled a transition from a negative to positive phase of the Pacific decadal oscillation. In the winter of 2013/14, unusually warm water in the northeast Pacific was associated with elevated ocean heat content anomalies and elevated sea level in the region. Globally, upper ocean heat content was record high for the year, reflecting the continued increase of thermal energy in the oceans, which absorb over 90% of Earth's excess heat from greenhouse gas forcing. Owing to both ocean warming and land ice melt contributions, global mean sea level in 2014 was also record high and 67 mm greater than the 1993 annual mean, when satellite altimetry measurements began. Sea surface salinity trends over the past decade indicate that salty regions grew saltier while fresh regions became fresher, suggestive of an increased hydrological cycle over the ocean expected with global warming. As in previous years, these patterns are reflected in 2014 subsurface salinity anomalies as well. With a now decade-long trans-basin instrument array along 26°N, the Atlantic meridional overturning circulation shows a decrease in transport of-4.2 ± 2.5 Sv decade-1. Precipitation was quite variable across the globe. On balance, precipitation over the world's oceans was above average, while below average across land surfaces. Drought continued in southeastern Brazil and the western United States. Heavy rain during April-June led to devastating floods in Canada's Eastern Prairies. Above-normal summer monsoon rainfall was observed over the southern coast of West Africa, while drier conditions prevailed over the eastern Sahel. Generally, summer monsoon rainfall over eastern Africa was above normal, except in parts of western South Sudan and Ethiopia. The south Asian summer monsoon in India was below normal, with June record dry. Across the major tropical cyclone basins, 91 named storms were observed during 2014, above the 1981-2010 global average of 82. The Eastern/Central Pacific and South Indian Ocean basins experienced significantly above-normal activity in 2014; all other basins were either at or below normal. The 22 named storms in the Eastern/Central Pacific was the basin's most since 1992. Similar to 2013, the North Atlantic season was quieter than most years of the last two decades with respect to the number of storms, despite the absence of El Niño conditions during both years. In higher latitudes and at higher elevations, increased warming continued to be visible in the decline of glacier mass balance, increasing permafrost temperatures, and a deeper thawing layer in seasonally frozen soil. In the Arctic, the 2014 temperature over land areas was the fourth highest in the 115-year period of record and snow melt occurred 20-30 days earlier than the 1998-2010 average. The Greenland Ice Sheet experienced extensive melting in summer 2014. The extent of melting was above the 1981-2010 average for 90% of the melt season, contributing to the second lowest average summer albedo over Greenland since observations began in 2000 and a record-low albedo across the ice sheet for August. On the North Slope of Alaska, new record high temperatures at 20-m depth were measured at four of five permafrost observatories. In September, Arctic minimum sea ice extent was the sixth lowest since satellite records began in 1979. The eight lowest sea ice extents during this period have occurred in the last eight years. Conversely, in the Antarctic, sea ice extent countered its declining trend and set several new records in 2014, including record high monthly mean sea ice extent each month from April to November. On 20 September, a record large daily Antarctic sea ice extent of 20.14 × 106 km2 occurred. The 2014 Antarctic stratospheric ozone hole was 20.9 million km2 when averaged from 7 September to 13 October, the sixth smallest on record and continuing a decrease, albeit statistically insignificant, in area since 1998.

    • The Simons Observatory (SO) is a new cosmic microwave background experiment being built on Cerro Toco in Chile, due to begin observations in the early 2020s. We describe the scientific goals of the experiment, motivate the design, and forecast its performance. SO will measure the temperature and polarization anisotropy of the cosmic microwave background in six frequency bands centered at: 27, 39, 93, 145, 225 and 280 GHz. The initial con figuration of SO will have three small-aperture 0.5-m telescopes and one large-aperture 6-m telescope, with a total of 60,000 cryogenic bolometers. Our key science goals are to characterize the primordial perturbations, measure the number of relativistic species and the mass of neutrinos, test for deviations from a cosmological constant, improve our understanding of galaxy evolution, and constrain the duration of reionization. The small aperture telescopes will target the largest angular scales observable from Chile, mapping approximate to 10% of the sky to a white noise level of 2 mu K-arcmin in combined 93 and 145 GHz bands, to measure the primordial tensor-to-scalar ratio, r, at a target level of sigma(r) = 0.003. The large aperture telescope will map approximate to 40% of the sky at arcminute angular resolution to an expected white noise level of 6 mu K-arcmin in combined 93 and 145 GHz bands, overlapping with the majority of the Large Synoptic Survey Telescope sky region and partially with the Dark Energy Spectroscopic Instrument. With up to an order of magnitude lower polarization noise than maps from the Planck satellite, the high-resolution sky maps will constrain cosmological parameters derived from the damping tail, gravitational lensing of the microwave background, the primordial bispectrum, and the thermal and kinematic Sunyaev-Zel'dovich effects, and will aid in delensing the large-angle polarization signal to measure the tensor-to-scalar ratio. The survey will also provide a legacy catalog of 16,000 galaxy clusters and more than 20,000 extragalactic sources.

    • All cancers are caused by somatic mutations; however, understanding of the biological processes generating these mutations is limited. The catalogue of somatic mutations from a cancer genome bears the signatures of the mutational processes that have been operative. Here we analysed 4,938,362 mutations from 7,042 cancers and extracted more than 20 distinct mutational signatures. Some are present in many cancer types, notably a signature attributed to the APOBEC family of cytidine deaminases, whereas others are confined to a single cancer class. Certain signatures are associated with age of the patient at cancer diagnosis, known mutagenic exposures or defects in DNA maintenance, but many are of cryptic origin. In addition to these genome-wide mutational signatures, hypermutation localized to small genomic regions, 'kataegis', is found in many cancer types. The results reveal the diversity of mutational processes underlying the development of cancer, with potential implications for understanding of cancer aetiology, prevention and therapy.

  • Predicting women with depressive symptoms postpartum with machine learning methods

    • Postpartum depression (PPD) is a detrimental health condition that affects 12% of new mothers. Despite negative effects on mothers' and children's health, many women do not receive adequate care. Preventive interventions are cost-efficient among high-risk women, but our ability to identify these is poor. We leveraged the power of clinical, demographic, and psychometric data to assess if machine learning methods can make accurate predictions of postpartum depression. Data were obtained from a population-based prospective cohort study in Uppsala, Sweden, collected between 2009 and 2018 (BASIC study, n = 4313). Sub-analyses among women without previous depression were performed. The extremely randomized trees method provided robust performance with highest accuracy and well-balanced sensitivity and specificity (accuracy 73%, sensitivity 72%, specificity 75%, positive predictive value 33%, negative predictive value 94%, area under the curve 81%). Among women without earlier mental health issues, the accuracy was 64%. The variables setting women at most risk for PPD were depression and anxiety during pregnancy, as well as variables related to resilience and personality. Future clinical models that could be implemented directly after delivery might consider including these variables in order to identify women at high risk for postpartum depression to facilitate individualized follow-up and cost-effectiveness.

  • Large-Scale Gene-Centric Meta-analysis across 32 Studies Identifies Multiple Lipid Loci

    • Genome-wide association studies (GWASs) have identified many SNPs underlying variations in plasma-lipid levels. We explore whether additional loci associated with plasma-lipid phenotypes, such as high-density lipoprotein cholesterol (HDL-C), low-density lipoprotein cholesterol (LDL-C), total cholesterol (TC), and triglycerides (TGs), can be identified by a dense gene-centric approach. Our meta-analysis of 32 studies in 66,240 individuals of European ancestry was based on the custom similar to 50,000 SNP genotyping array (the ITMAT-Broad-CARe array) covering similar to 2,000 candidate genes. SNP-lipid associations were replicated either in a cohort comprising an additional 24,736 samples or within the Global Lipid Genetic Consortium. We identified four, six, ten, and four unreported SNPs in established lipid genes for HDL-C, LDL-C, TC, and TGs, respectively. We also identified several lipid-related SNPs in previously unreported genes: DGAT2, HCAR2, GPIHBP1, PPARG, and FTO for HDL-C; SOCS3, APOH, SPTY2D1, BRCA2, and VLDLR for LDL-C; SOCS3, UGT1A1, BRCA2, UBE3B, FCGR2A, CHUK, and INSIG2 for TC; and SERPINF2, C4B, GCK, GATA4, INSR, and LPAL2 for TGs. The proportion of explained phenotypic variance in the subset of studies providing individual-level data was 9.9% for HDL-C, 9.5% for LDL-C, 10.3% for TC, and 8.0% for TGs. This large meta-analysis of lipid phenotypes with the use of a dense gene-centric approach identified multiple SNPs not previously described in established lipid genes and several previously unknown loci. The explained phenotypic variance from this approach was comparable to that from a meta-analysis of GWAS data, suggesting that a focused genotyping approach can further increase the understanding of heritability of plasma lipids.

    • Canine degenerative myelopathy (DM) is a fatal neurodegenerative disease prevalent in several dog breeds. Typically, the initial progressive upper motor neuron spastic and general proprioceptive ataxia in the pelvic limbs occurs at 8 years of age or older. If euthanasia is delayed, the clinical signs will ascend, causing flaccid tetraparesis and other lower motor neuron signs. DNA samples from 38 DM-affected Pembroke Welsh corgi cases and 17 related clinically normal controls were used for genome-wide association mapping, which produced the strongest associations with markers on CFA31 in a region containing the canine SOD1 gene. SOD1 was considered a regional candidate gene because mutations in human SOD1 can cause amyotrophic lateral sclerosis (ALS), an adult-onset fatal paralytic neurodegenerative disease with both upper and lower motor neuron involvement. The resequencing of SOD1 in normal and affected dogs revealed a G to A transition, resulting in an E40K missense mutation. Homozygosity for the A allele was associated with DM in 5 dog breeds: Pembroke Welsh corgi, Boxer, Rhodesian ridgeback, German Shepherd dog, and Chesapeake Bay retriever. Microscopic examination of spinal cords from affected dogs revealed myelin and axon loss affecting the lateral white matter and neuronal cytoplasmic inclusions that bind anti-superoxide dismutase 1 antibodies. These inclusions are similar to those seen in spinal cord sections from ALS patients with SOD1 mutations. Our findings identify canine DM to be the first recognized spontaneously occurring animal model for ALS.

    • An approach to reduce CO2 emissions while simultaneously keeping the soot emissions down from compression ignition (CI) engines is to blend in short chained oxygenates into the fuel. In this work, two oxygenated fuel blends consisting of diesel, gasoline and ethanol (EtOH) in the ratio of 68:17:15 and 58:14:30 have been utilized and studied in a single cylinder light duty (LD) CI engine in terms of efficiency and emissions. The reasons of utilizing gasoline in the fuel blend is due to the emulsifying properties it has while increasing the total octane rating of the fuel to be able to run the engine with a higher fraction of premixed flame. When performing the experiments, the control parameters were set as close as possible to the original equipment manufacturer (OEM) EU5 calibration of the multi-cylinder engine to study the possibility of using such blends in close to stock LD CI engines. With the oxygenates, in particular the fuel with the higher concentration of EtOH achieved an indicated net efficiency of ∼51% inf comparison to ∼47% for diesel at 8 bar BMEP. The NOX emissions increased slightly for the double injection strategy at 13 bar BMEP from ∼13.5 g/kW h to ∼14.5 g/kW h when going from diesel fuel to the higher ethanol blend. However utilizing single injection strategy at lower loads reduces the NOX. Highest soot mass measured for diesel was ∼0.46 g/kW h in contrast to ∼0.1 g/kW h for the oxygenates. Also, soot production when running the engine on the ethanol containing fuels was not significantly affected by EGR utilization as in the case of diesel. Considering particle size distribution, the particles are reduced both in terms of mean diameter and quantity. At 1500 rpm and 2 bar BMEP an increase of over ∼300% in THC and CO was measured, however, increasing the speed and load to above 2000 rpm and 8 bar BMEP respectively, made the difference negligible due to high in-cylinder temperatures contributing to better fuel oxidation. Despite having lower cetane numbers, higher combustion stability was observed for the oxygenates fuels.

  • Machine Learning Prediction of Resistance to Subinhibitory Antimicrobial Concentrations from Escherichia coli Genomes.

    • Escherichia coli is an important cause of bacterial infections worldwide, with multidrug-resistant strains incurring substantial costs on human lives. Besides therapeutic concentrations of antimicrobials in health care settings, the presence of subinhibitory antimicrobial residues in the environment and in clinics selects for antimicrobial resistance (AMR), but the underlying genetic repertoire is less well understood. Here, we used machine learning to predict the population doubling time and cell growth yield of 1,407 genetically diverse E. coli strains expanding under exposure to three subinhibitory concentrations of six classes of antimicrobials from single-nucleotide genetic variants, accessory gene variation, and the presence of known AMR genes. We predicted cell growth yields in the held-out test data with an average correlation (Spearman's ρ) of 0.63 (0.36 to 0.81 across concentrations) and cell doubling times with an average correlation of 0.59 (0.32 to 0.92 across concentrations), with moderate increases in sample size unlikely to improve predictions further. This finding points to the remaining missing heritability of growth under antimicrobial exposure being explained by effects that are too rare or weak to be captured unless sample size is dramatically increased, or by effects other than those conferred by the presence of individual single-nucleotide polymorphisms (SNPs) and genes. Predictions based on whole-genome information were generally superior to those based only on known AMR genes and were accurate for AMR resistance at therapeutic concentrations. We pinpointed genes and SNPs determining the predicted growth and thereby recapitulated many known AMR determinants. Finally, we estimated the effect sizes of resistance genes across the entire collection of strains, disclosing the growth effects for known resistance genes in each individual strain. Our results underscore the potential of predictive modeling of growth patterns from genomic data under subinhibitory concentrations of antimicrobials, although the remaining missing heritability poses a challenge for achieving the accuracy and precision required for clinical use. IMPORTANCE Predicting bacterial growth from genome sequences is important for a rapid characterization of strains in clinical diagnostics and to disclose candidate novel targets for anti-infective drugs. Previous studies have dissected the relationship between bacterial growth and genotype in mutant libraries for laboratory strains, yet no study so far has examined the predictive power of genome sequence in natural strains. In this study, we used a high-throughput phenotypic assay to measure the growth of a systematic collection of natural Escherichia coli strains and then employed machine learning models to predict bacterial growth from genomic data under nontherapeutic subinhibitory concentrations of antimicrobials that are common in nonclinical settings. We found a moderate to strong correlation between predicted and actual values for the different collected data sets. Moreover, we observed that the known resistance genes are still effective at sublethal concentrations, pointing to clinical implications of these concentrations.

    • In an era characterized by a move towards a “knowledge society”, universities are central in fostering “knowledgeability”, that is the reflexive understanding of knowledge in knowledge societies. The objective of “knowledgeability” can be met through creating a stronger link between education and research. Furthermore, overall student performance, for example in critical thinking and problem solving, can be improved if research-related activities are incorporated into the curriculum.The aim of this paper is to use international examples to discuss the research- education nexus from four different perspectives, namely context, policy, implementation and quality, with case studies from higher education institutions in Singapore and Sweden.We suggest that different integrative technologies can be used to enhance the links, but it will be essential to consider the inputs of training, service and support in using new technology. Interestingly, the act of evaluating the link between education and research will increase awareness of this linkage by stakeholders involved in both education and research. In turn the link can be strengthened, contributing to increased quality in both education and research.

    • The low predictability of earthquakes and the high uncertainty associated with their forecasts make earthquakes one of the worst natural calamities, capable of causing instant loss of life and property. Here, we discuss the studies reporting the observed anomalies in the satellite-derived Land Surface Temperature (LST) before an earthquake. We compile the conclusions of these studies and evaluate the use of remotely sensed LST anomalies as precursors of earthquakes. The arrival times and the amplitudes of the anomalies vary widely, thus making it difficult to consider them as universal markers to issue earthquake warnings. Based on the randomness in the observations of these precursors, we support employing a global-scale monitoring system to detect statistically robust anomalous geophysical signals prior to earthquakes before considering them as definite precursors.

    • Slope streaks are prevalent and intriguing dark albedo surface features on contemporary Mars. Slope streaks are readily observed in the equatorial and subequatorial dusty regolith regions with low thermal inertia. They gradually fade over decadal timescales. The proposed mechanisms for their formation vary widely based on several physicochemical and geomorphological explanations. The scientific community is divided in proposing both dry and wet mechanisms for the formation of slope streaks. Here we perform a systematic evaluation of the literature for these wet and dry mechanisms. We discuss the probable constraints on the various proposed mechanisms and provide perspectives on the plausible process driving global‐scale slope streak formation on contemporary Mars. Although per our understanding, a thorough consideration of the global distribution of slope streaks, their morphology and topography, flow characteristics, physicochemical and atmospheric coincidences, and terrestrial analogies weighs more in favor of several wet mechanisms, we acknowledge that such wet mechanisms cannot explain all the reported morphological and terrain variations of slope streaks. Thus, we suggest that explanations considering both dry and wet processes can more holistically describe all the observed morphological variations among slope streaks. We further acknowledge the constraints on the resolutions of remote sensing data and on our understanding of the Martian mineralogy, climate, and atmosphere and recommend continuous investigations in this direction using future remote sensing acquisitions and simulations. In this regard, finding more wet and dry terrestrial analogs for Martian slope streaks and studying them at high spatiotemporal resolutions can greatly improve our understanding.

    • Several interpretations of recurring slope lineae (RSL) have related RSL to the potential presence of transient liquid water on Mars. Such probable signs of liquid water have implications for Mars exploration in terms of rover safety, planetary protection during rover operations, and the current habitability of the planet. Mawrth Vallis has always been a prime target to be considered for Mars rover missions due to its rich mineralogy. Most recently, Mawrth Vallis was one of the two final candidates selected by the European Space Agency as a landing site for the ExoMars 2020 mission. Therefore, all surface features and landforms in Mawrth Vallis that may be of special interest in terms of scientific goals, rover safety, and operations must be scrutinised to better assess it for future Mars missions. Here, we report on the initial detection of RSL candidates in two craters of Mawrth Vallis. The new sightings were made outside of established RSL regions and further prompt the inclusion of a new geographical region within the RSL candidate group. Our inferences on the RSL candidates are based on several morphological and geophysical evidences and analogies: (i) the dimensions of the RSL candidates are consistent with confirmed mid-latitude RSL; (ii) albedo and thermal inertia values are comparable to those of other mid-latitude RSL sites; and (iii) features are found in a summer season image and on the steep and warmest slopes. These results denote the plausible presence of transient liquid brines close to the previously proposed landing ellipse of the ExoMars rover, rendering this site particularly relevant to the search of life. Further investigations of Mawrth Vallis carried out at higher spatial and temporal resolutions are needed to identify more of such features at local scales to maximize the scientific return from the future Mars rovers, to prevent probable biological contamination during rover operations, to evade damage to rover components as brines can be highly corrosive, and to quantify the ability of the regolith at mid-latitudes to capture atmospheric water which is relevant for in-situ-resource utilization.

  • Distribution and Morphologies of Transverse Aeolian Ridges in ExoMars 2020 Rover Landing Site

    • Aeolian processes are believed to play a major role in the landscape evolution of Mars. Investigations on Martian aeolian landforms such as ripples, transverse aeolian ridges (TARs), and dunes, and aeolian sediment flux measurements are important to enhance our understanding of past and present wind regimes, the ongoing dust cycle, landscape evolution, and geochemistry. These aeolian bedforms are often comprised of loose sand and sharply undulating topography and thus pose a threat to mobility and maneuvers of Mars rovers. Here we present a first-hand account of the distribution, morphologies, and morphometrics of TARs in Oxia Planum, the recently selected ExoMars 2020 Rover landing site. The gridded mapping was performed for contiguous stretches of TARs within all the landing ellipses using 57 sub-meter high resolution imaging science experiment (HiRISE) scenes. We also provide the morphological descriptions for all types of TARs present within the landing ellipses. We use HiRISE digital terrain models (DTMs) along with the images to derive morphometric information for TARs in Oxia Planum. In general, the average areal TAR coverage was found to be 5.4% (±4.9% standard deviation), increasing from west to east within the landing ellipses. We report the average TAR morphometrics in the form of crest–ridge width (131.1 ± 106.2 m), down-wind TAR length (17.6 ± 10.1 m), wavelength (37.3 ± 11.6 m), plan view aspect ratio (7.1 ± 2.3), inter-bedform spacing (2.1 ± 1.1), slope (10.6° ± 6.1°), predominant orientations (NE-SW and E-W), and height (1.2 ± 0.8 m). While simple TARs are predominant, we report other TAR morphologies such as forked TAR, wavy TAR with associated smaller secondary ripples, barchan-like TAR, networked TAR, and mini-TARs from the region. Our results can help in planning the rover traverses in terms of both safe passage and scientific returns favoring aeolian research, particularly improving our understanding of TARs.

    • The cryosphere consists of frozen water and includes lakes/rivers/sea ice, glaciers, ice caps/sheets, snow cover, and permafrost. Because highly reflective snow and ice are the main components of the cryosphere, it plays an important role in the global energy balance. Thus, any qualitative or quantitative change in the physical properties and extents of the cryosphere affects global air circulation, ocean and air temperatures, sea level, and ocean current patterns. Due to the hardships involved in collecting ground control points and field data for high alpine glaciers or vast polar ice sheets, several researchers are currently using remote sensing. Satellites provide an effective space-borne platform for remotely sensing frozen areas at the global and regional scales. However, satellite remote sensing has several constraints, such as limited spatial and temporal resolutions and expensive data acquisition. Therefore, aerial and terrestrial remote sensing platforms and sensors are needed to cover temporal and spatial gaps for comprehensive cryospheric research. Light Detection and Ranging (LiDAR) antennas form a group of active remote sensors that can easily be deployed on all three platforms, i.e., satellite, aerial, and terrestrial. The generation of elevation data for glacial and snow-covered terrain from photogrammetry requires high contrast amongst various reflective surfaces (ice, snow, firn, and slush). Conventional passive optical remote sensors do not provide the necessary accuracy, especially due to the unavailability of reliable ground control points. However, active LiDAR sensors can fill this research gap and provide high-resolution and accurate Digital Elevation Models (DEMs). Due to the obvious advantages of LiDAR over conventional passive remote sensors, the number of LiDAR-based cryospheric studies has increased in recent years. In this review, we highlight studies that have utilised LiDAR sensors for the cryospheric research of various features, such as snow cover, polar ice sheets and their atmospheres, alpine glaciers, and permafrost. Because this technology shows immense promise for applications in future cryospheric research, we also emphasise the prospects of utilising LiDAR sensors. In this paper, a large compilation of relevant references is presented to allow readers to explore particular topics of interest.

The reason behind their formation remains unclear with proposed hypotheses for both dry and wet mechanisms. Here, we report an up-to-date distribution and morphometric investigation of Martian slope streaks. We find: (i) a remarkable coexistence of the slope streak distribution with the regions on Mars with high abundances of water-equivalent hydrogen, chlorine, and iron; (ii) favourable thermodynamic conditions for transient deliquescence and brine development in the slope streak regions; (iii) a significant concurrence of slope streak distribution with the regions of enhanced atmospheric water vapour concentration, thus suggestive of a present-day regolith-atmosphere water cycle; and (iv) terrain preferences and flow patterns supporting a wet mechanism for slope streaks. These results suggest a strong local regolith-atmosphere water coupling in the slope streak regions that leads to the formation of these fluidised features. Our conclusions can have profound astrobiological, habitability, environmental, and planetary protection implications

    • The high levels of uncertainty associated with earthquake prediction render earthquakes some of the worst natural calamities. Here, we present our observations of MODerate resolution Imaging Spectroradiometer (MODIS)-derived Land Surface Temperature (LST) anomaly for earthquakes in the largest tectonically active Himalayan and Andean mountain belts. We report the appearance of fairly detectable pre-earthquake Snow Surface Temperature (SST) anomalies. We use 16 years (2000–2015) of MODIS LST time-series data to robustly conclude our findings for three of the most destructive earthquakes that occurred in 2015 in the high mountains of Nepal, Chile, and Afghanistan. We propose the physical basis behind higher sensitivity of snow towards geothermal emissions. Although the preliminary appearance of SST anomalies and their amplitudes vary, we propose employing a global-scale monitoring system for detecting and studying such spatio-temporal geophysical signals. With the advent of improved remote sensors, we anticipate that such efforts can be another step towards improved earthquake predictions.

    • The catastrophic magnitude of life and monetary losses associated with earthquakes deserve serious attention and mitigation measures. However, in addition to the pre-earthquake and post-earthquake alleviation actions, the scientific community indeed needs to reconsider the possibilities of earthquake predictions using non-seismic precursors. A significant number of studies in the recent decades have reported several possible earthquake precursors such as anomalies in electric field, magnetic field, gas/aerosol emissions, ionospheric signals, ground water level, land surface temperature, surface deformations, animal behaviour, thermal infrared signals, atmospheric gravity waves, and lightning. Such substantial number of scientific articles and reported anomalous signals cannot be overlooked without a thoughtful appraisal. Here, we provide an opinion on the way forward for earthquake prediction in terms of challenges and possibilities while using non-seismic precursors. A general point of concern is the widely varying arrival times and the amplitudes of the anomalies, putting a question mark on their universal applicability as earthquake markers. However, a unifying concept which does not only define the physical basis of either all or most of these anomalies but which also streamlines their characterisation procedure must be the focus of future earthquake precursory research. Advancements in developing the adaptable instrumentation for in-situ observations of the claimed non-seismic precursors must be the next step and the satellite observations should not be taken as a replacement for field-based research. We support the need to standardise the precursor detection techniques and to employ a global-scale monitoring system for making any possible earthquake predictions reliable.

  • UAV Imaging of a Martian Brine Analogue Environment in a Fluvio-Aeolian Setting

    • Understanding extraterrestrial environments and landforms through remote sensing and terrestrial analogy has gained momentum in recent years due to advances in remote sensing platforms, sensors, and computing efficiency. The seasonal brines of the largest salt plateau on Earth in Salar de Uyuni (Bolivian Altiplano) have been inadequately studied for their localized hydrodynamics and the regolith volume transport across the freshwater-brine mixing zones. These brines have recently been projected as a new analogue site for the proposed Martian brines, such as recurring slope lineae (RSL) and slope streaks. The Martian brines have been postulated to be the result of ongoing deliquescence-based salt-hydrology processes on contemporary Mars, similar to the studied Salar de Uyuni brines. As part of a field-site campaign during the cold and dry season in the latter half of August 2017, we deployed an unmanned aerial vehicle (UAV) at two sites of the Salar de Uyuni to perform detailed terrain mapping and geomorphometry. We generated high-resolution (2 cm/pixel) photogrammetric digital elevation models (DEMs) for observing and quantifying short-term terrain changes within the brines and their surroundings. The achieved co-registration for the temporal DEMs was considerably high, from which precise inferences regarding the terrain dynamics were derived. The observed average rate of bottom surface elevation change for brines was ~1.02 mm/day, with localized signs of erosion and deposition. Additionally, we observed short-term changes in the adjacent geomorphology and salt cracks. We conclude that the transferred regolith volume via such brines can be extremely low, well within the resolution limits of the remote sensors that are currently orbiting Mars, thereby making it difficult to resolve the topographic relief and terrain perturbations that are produced by such flows on Mars. Thus, the absence of observable erosion and deposition features within or around most of the proposed Martian RSL and slope streaks cannot be used to dismiss the possibility of fluidized flow within these features

    • Satellite remote sensing is an effective way to monitor vast extents of global glaciers and snowfields. However, satellite remote sensing is limited by spatial and temporal resolutions and the high costs involved in data acquisition. Unmanned aerial vehicle (UAV)-based glaciological studies are gaining pace in recent years due to their advantages over conventional remote sensing platforms. UAVs are easy to deploy, with the option of alternating the sensors working in visible, infrared, and microwave wavelengths. The high spatial resolution remote sensing data obtained from these UAV-borne sensors are a significant improvement over the data obtained by traditional remote sensing. The cost involved in data acquisition is minimal and researchers can acquire imagery according to their schedule and convenience. We discuss significant glaciological studies involving UAV as remote sensing platforms. This is the first review work, exclusively dedicated to highlight UAV as a remote sensing platform in glaciology. We examine polar and alpine applications of UAV and their future prospects in separate sections and present an extensive reference list for the readers, so that they can delve into their topic of interest. Because the technology is still widely unexplored for snow and glaciers, we put a special emphasis on discussing the future prospects of utilising UAVs for glaciological research.

    • Objective: Outcomes for intact abdominal aortic aneurysm (AAA) repair vary over time and by healthcare system, country, and surgeon. The aim of this study was to analyse peri-operative mortality for intact AAA repair in 11 countries over time and compare outcomes by gender, age, and geographical location. Methods: Prospective data on primary repair of intact AAAwere collected from11 countries through the International Consortium of Vascular Registries (ICVR) and analysed for two time periods, 2010 - 2013 and 2014 - 2016. The primary outcome was peri-operative mortality after endovascular aneurysm repair (EVAR) and open surgical repair (OSR). Multivariable logistic regression models were used to adjust for differences in patient characteristics. Results: A total of 103 715 patients were included. The percentage of patients undergoing EVAR increased from 63.6% to 71.2% (p < .001) over the study period. This proportion varied by country from 35% in Hungary to 81% in the United States. Overall peri-operative mortality decreased from 2.1% to 1.6 % (p < .001). Mortality also declined significantly over time for both OSR 4.2% to 3.6 % (p = .002) and EVAR 1.0% to 0.7% (p = .002). Mortality was significantly higher for female than male patients (3.0% vs. 1.6% p < .001). The percentage of patients > 80 years old undergoing AAA repair remained constant at 23.6% (p = .91). Peri-operative mortality was higher for patients > 80 years than for those < 80 years old (2.7% vs. 1.6% p < .001). Forty-six per cent (n = 275) of all EVAR deaths occurred in the over 80s. Conclusion: The proportion of AAA repairs performed using EVAR has increased over time. Peri-operative mortality continues to decline for both OSR and EVAR. Outcomes however were significantly worse for both women and those aged over 80, so efforts should be focused on these patient groups to further reduce elective AAA mortality rates.

  • Endochondral bone in an Early Devonian 'placoderm' from Mongolia

    • Endochondral bone is the main internal skeletal tissue of nearly all osteichthyans—the group comprising more than 60,000 living species of bony fishes and tetrapods. Chondrichthyans (sharks and their kin) are the living sister group of osteichthyans and have primarily cartilaginous endoskeletons, long considered the ancestral condition for all jawed vertebrates (gnathostomes). The absence of bone in modern jawless fishes and the absence of endochondral ossification in early fossil gnathostomes appear to lend support to this conclusion. Here we report the discovery of extensive endochondral bone in Minjinia turgenensis, a new genus and species of ‘placoderm’-like fish from the Early Devonian (Pragian) of western Mongolia described using X-ray computed microtomography. The fossil consists of a partial skull roof and braincase with anatomical details providing strong evidence of placement in the gnathostome stem group. However, its endochondral space is filled with an extensive network of fine trabeculae resembling the endochondral bone of osteichthyans. Phylogenetic analyses place this new taxon as a proximate sister group of the gnathostome crown. These results provide direct support for theories of generalized bone loss in chondrichthyans. Furthermore, they revive theories of a phylogenetically deeper origin of endochondral bone and its absence in chondrichthyans as a secondary condition.

    • This thesis sheds light on border crossings of popular music genres and proposes a novel approach to studying circulation of popular music across the globe. It propounds a critical discussion on the conventions and politics of knowledge production and dissemination within the scholarship that lies in the intersection of popular music studies, ethnomusicology and music sociology. The thesis points to the predominance of structure-driven and conjuncture-oriented approaches as well as issues of comprehension and representation within the scholarship, and it motivates an actor-oriented approach to account for transregional circulation of popular music genres. Through taking indie music in Istanbul as its case, the thesis develops an alternative approach called "genre transfer." It focuses on the emergence and development of indie music within the regional context of Istanbul from the 1990s to the 2010s. Apart from identifying the key agents and institutions that have been involved in the process of indie music's local legitimization, the thesis analyzes the agents' changing conditions, relations and strategies that have provided base to the transformation of their practices. In doing so, the thesis draws upon actor-oriented approaches of cultural transfer studies and remains informed by analytical and epistemological concerns laid out within the fields of relational sociology and sociology of knowledge.

    • The construction of shelters on plants by arthropods might influence other organisms via changes in colonization, community richness, species composition, and functionality. Arthropods, including beetles, caterpillars, sawflies, spiders, and wasps often interact with host plants via the construction of shelters, building a variety of structures such as leaf ties, tents, rolls, and bags; leaf and stem galls, and hollowed out stems. Such constructs might have both an adaptive value in terms of protection (i.e., serve as shelters) but may also exert a strong influence on terrestrial community diversity in the engineered and neighboring hosts via colonization by secondary occupants. Although different traits of the host plant (e.g., physical, chemical, and architectural features) may affect the potential for ecosystem engineering by insects, such effects have been, to a certain degree, overlooked. Further analyses of how plant traits affect the occurrence of shelters may therefore enrich our understanding of the organizing principles of plant-based communities. This data set includes more than 1000 unique records of ecosystem engineering by arthropods, in the form of structures built on plants. All records have been published in the literature, and span both natural structures (91% of the records) and structures artificially created by researchers (9% of the records). The data were gathered between 1932 and 2021, across more than 50 countries and several ecosystems, ranging from polar to tropical zones. In addition to data on host plants and engineers, we aggregated data on the type of constructs and the identity of inquilines using these structures. This data set highlights the importance of these subtle structures for the organization of terrestrial arthropod communities, enabling hypotheses testing in ecological studies addressing ecosystem engineering and facilitation mediated by constructs.     • We present 850 mu m dust polarization observations of the massive DR21 filament from the B-fields In STar-forming Region Observations (BISTRO) survey, using the POL-2 polarimeter and the SCUBA-2 camera on the James Clerk Maxwell Telescope. We detect ordered magnetic fields perpendicular to the parsec-scale ridge of the DR21 main filament. In the subfilaments, the magnetic fields are mainly parallel to the filamentary structures and smoothly connect to the magnetic fields of the main filament. We compare the POL-2 and Planck dust polarization observations to study the magnetic field structures of the DR21 filament on 0.1-10 pc scales. The magnetic fields revealed in the Planck data are well-aligned with those of the POL-2 data, indicating a smooth variation of magnetic fields from large to small scales. The plane-of-sky magnetic field strengths derived from angular dispersion functions of dust polarization are 0.6-1.0 mG in the DR21 filament and similar to 0.1 mG in the surrounding ambient gas. The mass-to-flux ratios are found to be magnetically supercritical in the filament and slightly subcritical to nearly critical in the ambient gas. The alignment between column density structures and magnetic fields changes from random alignment in the low-density ambient gas probed by Planck to mostly perpendicular in the high-density main filament probed by James Clerk Maxwell Telescope. The magnetic field structures of the DR21 filament are in agreement with MHD simulations of a strongly magnetized medium, suggesting that magnetic fields play an important role in shaping the DR21 main filament and subfilaments.

    • Several safe boundaries of critical Earth system processes have already been crossed due to human perturbations; not accounting for their interactions may further narrow the safe operating space for humanity. Using expert knowledge elicitation, we explored interactions among seven variables representing Earth system processes relevant to food production, identifying many interactions little explored in Earth system literature. We found that green water and land system change affect other Earth system processes strongly, while land, freshwater and ocean components of biosphere integrity are the most impacted by other Earth system processes, most notably blue water and biogeochemical flows. We also mapped a complex network of mechanisms mediating these interactions and created a future research prioritization scheme based on interaction strengths and existing knowledge gaps. Our study improves the understanding of Earth system interactions, with sustainability implications including improved Earth system modelling and more explicit biophysical limits for future food production.

    • The deep subsurface of other planetary bodies is of special interest for robotic and human exploration. The subsurface provides access to planetary interior processes, thus yielding insights into planetary formation and evolution. On Mars, the subsurface might harbour the most habitable conditions. In the context of human exploration, the subsurface can provide refugia for habitation from extreme surface conditions. We describe the fifth Mine Analogue Research (MINAR 5) programme at 1 km depth in the Boulby Mine, UK in collaboration with Spaceward Bound NASA and the Kalam Centre, India, to test instruments and methods for the robotic and human exploration of deep environments on the Moon and Mars. The geological context in Permian evaporites provides an analogue to evaporitic materials on other planetary bodies such as Mars. A wide range of sample acquisition instruments (NASA drills, Small Planetary Impulse Tool (SPLIT) robotic hammer, universal sampling bags), analytical instruments (Raman spectroscopy, Close-Up Imager, Minion DNA sequencing technology, methane stable isotope analysis, biomolecule and metabolic life detection instruments) and environmental monitoring equipment (passive air particle sampler, particle detectors and environmental monitoring equipment) was deployed in an integrated campaign. Investigations included studying the geochemical signatures of chloride and sulphate evaporitic minerals, testing methods for life detection and planetary protection around human-tended operations, and investigations on the radiation environment of the deep subsurface. The MINAR analogue activity occurs in an active mine, showing how the development of space exploration technology can be used to contribute to addressing immediate Earth-based challenges. During the campaign, in collaboration with European Space Agency (ESA), MINAR was used for astronaut familiarization with future exploration tools and techniques. The campaign was used to develop primary and secondary school and primary to secondary transition curriculum materials on-site during the campaign which was focused on a classroom extra vehicular activity simulation.

    • Adrenocortical carcinoma (ACC) is an orphan disease lacking effective systemic treatment options. The low incidence of the disease and high cost of clinical trials are major obstacles in the search for improved treatment strategies. As a novel approach, registry-based clinical trials have been introduced in clinical research, so allowing for significant cost reduction, but without compromising scientific benefit. Herein, we describe how the European Network for the Study of Adrenal Tumours (ENSAT) could transform its current registry into one fit for a clinical trial infrastructure. The rationale to perform randomized registry-based trials in ACC is outlined including an analysis of relevant limitations and challenges. We summarize a survey on this concept among ENSAT members who expressed a strong interest in the concept and rated its scientific potential as high. Legal aspects, including ethical approval of registry-based randomization were identified as potential obstacles. Finally, we describe three potential randomized registry-based clinical trials in an adjuvant setting and for advanced disease with a high potential to be executed within the framework of an advanced ENSAT registry. Thus we, therefore, provide the basis for future registry-based trials for ACC patients. This could ultimately provide proof-of-principle of how to perform more effective randomized trials for an orphan disease.
