    • Patients with inherited CARMIL2 or CD28 deficiency have defective T cell CD28 signaling, but their immunological and clinical phenotypes remain largely unknown. We show that only one of three CARMIL2 isoforms is produced and functional across leukocyte subsets. Tested mutant CARMIL2 alleles from 89 patients and 52 families impair canonical NF-κB but not AP-1 and NFAT activation in T cells stimulated via CD28. Like CD28-deficient patients, CARMIL2-deficient patients display recalcitrant warts and low blood counts of CD4+ and CD8+ memory T cells and CD4+ TREGs. Unlike CD28-deficient patients, they have low counts of NK cells and memory B cells, and their antibody responses are weak. CARMIL2 deficiency is fully penetrant by the age of 10 yr and is characterized by numerous infections, EBV+ smooth muscle tumors, and mucocutaneous inflammation, including inflammatory bowel disease. Patients with somatic reversions of a mutant allele in CD4+ T cells have milder phenotypes. Our study suggests that CARMIL2 governs immunological pathways beyond CD28.

  • The International Bathymetric Chart of the Southern Ocean Version 2

    • The Southern Ocean surrounding Antarctica is a region that is key to a range of climatic and oceanographic processes with worldwide effects, and is characterised by high biological productivity and biodiversity. Since 2013, the International Bathymetric Chart of the Southern Ocean (IBCSO) has represented the most comprehensive compilation of bathymetry for the Southern Ocean south of 60 degrees S. Recently, the IBCSO Project has combined its efforts with the Nippon Foundation - GEBCO Seabed 2030 Project supporting the goal of mapping the world's oceans by 2030. New datasets initiated a second version of IBCSO (IBCSO v2). This version extends to 50 degrees S (covering approximately 2.4 times the area of seafloor of the previous version) including the gateways of the Antarctic Circumpolar Current and the Antarctic circumpolar frontal systems. Due to increased (multibeam) data coverage, IBCSO v2 significantly improves the overall representation of the Southern Ocean seafloor and resolves many submarine landforms in more detail. This makes IBCSO v2 the most authoritative seafloor map of the area south of 50 degrees S.

    • In the recent years, our team has been working to identify key aspects of anisotropy in porous media. More than just characterising their anisotropic properties, we're interested in generating cellular media with completely designed anisotropic properties. The results of these studies have partly been published and more are to come. In this presentation, we'll present an overview of this work and how it ultimately ties to acoustics. We will introduce the key findings,discussed specific results that can be achieved and provide context and details related to the strategy developed to address the tasks.

  • The International Bathymetric Chart of the Arctic Ocean Version 4.0

    • Bathymetry (seafloor depth), is a critical parameter providing the geospatial context for a multitude of marine scientific studies. Since 1997, the International Bathymetric Chart of the Arctic Ocean (IBCAO) has been the authoritative source of bathymetry for the Arctic Ocean. IBCAO has merged its efforts with the Nippon Foundation-GEBCO-Seabed 2030 Project, with the goal of mapping all of the oceans by 2030. Here we present the latest version (IBCAO Ver. 4.0), with more than twice the resolution (200 x 200m versus 500 x 500m) and with individual depth soundings constraining three times more area of the Arctic Ocean (similar to 19.8% versus 6.7%), than the previous IBCAO Ver. 3.0 released in 2012. Modern multibeam bathymetry comprises similar to 14.3% in Ver. 4.0 compared to similar to 5.4% in Ver. 3.0. Thus, the new IBCAO Ver. 4.0 has substantially more seafloor morphological information that offers new insights into a range of submarine features and processes; for example, the improved portrayal of Greenland fjords better serves predictive modelling of the fate of the Greenland Ice Sheet.   • Combined RNA/tissue profiling identifies novel Cancer/testis genes

    • Cancer/Testis (CT) genes are induced in germ cells, repressed in somatic cells, and derepressed in somatic tumors, where these genes can contribute to cancer progression. CT gene identification requires data obtained using standardized protocols and technologies. This is a challenge because data for germ cells, gonads, normal somatic tissues, and a wide range of cancer samples stem from multiple sources and were generated over substantial periods of time. We carried out a GeneChip-based RNA profiling analysis using our own data for testis and enriched germ cells, data for somatic cancers from the Expression Project for Oncology, and data for normal somatic tissues from the Gene Omnibus Repository. We identified 478 candidate loci that include known CT genes, numerous genes associated with oncogenic processes, and novel candidates that are not referenced in the Cancer/Testis Database (). We complemented RNA expression data at the protein level for SPESP1, GALNTL5, PDCL2, and C11orf42 using cancer tissue microarrays covering malignant tumors of breast, uterus, thyroid, and kidney, as well as published RNA profiling and immunohistochemical data provided by the Human Protein Atlas (). We report that combined RNA/tissue profiling identifies novel CT genes that may be of clinical interest as therapeutical targets or biomarkers. Our findings also highlight the challenges of detecting truly germ cell-specific mRNAs and the proteins they encode in highly heterogenous testicular, somatic, and tumor tissues.

  • Tuning sound transmission loss for multi-layer panels with anisotropic foams

    • Multilayer panels consisting of a load carrying structure, a porous material for thermal and acoustic insulation and an interior trim panel is a very common type of design for vehicles. Weight as well as total build height are usually limiting constraints on the design. The idea of using an anisotropic porous material instead of an isotropic one to improve the sound transmission loss without adding a lot of weight or thickness is explored in the paper. By using a state space formulation of the transfer matrix method transmission loss it is possible to include anisotropic material properties in the calculation. The anisotropic material is modelled by a combination of a simplified analytical model for the acoustic losses and inverse estimation of the 21 independent elastic constants of the Hooke’s tensor. The porous material, which has typical dimensions possible to 3D print, is based on a Kelvin cell micro model that has a controlled degree of anisotropy.

  • Dynamic behaviour of low- to high-density anisotropic cellular materials

    • The dynamic behaviour of a novel anisotropic cellular micro-structural geometry derived from the basic symmetric Kelvin cell is discussed for varying relative density. The cells are arranged in a cubic array and the dynamic response is studied in a classical seismic mass setup using beam elements to represent the ligaments of the cell. The eigenfrequencies and the eigenmodes of the cellular array are computed together with forced response simulations where a proportional damping model of the Young's modulus for the cell ligaments is assumed. The frequency dependence of the damping is based on a fractional derivative representation. Using a recently developed inversion method, equivalent, homogenised solid material models of the cellular array are discussed with the associated equivalent elastic properties given in terms of the 21 elastic constants of the Hooke's tensor. For the equivalent solid material models, the eigenfrequencies and eigenmodes are computed, and forced response simulations are performed assuming the same type of proportionality in the damping as the cellular array, for the same seismic mass setup. The correlation, between the eigenfrequencies and the eigenmodes, shows an overall interesting agreement between the cellular and the equivalent solid model for the quite complex deformation shapes observed. The forced response results indicate that the equivalent solid modelling accurately represents the global dynamics of the anisotropic cellular array, but needs to be further refined when local shearing deformation within the individual cells starts to be dominating.

  • Twist, tilt and stretch: From isometric Kelvin cells to anisotropic cellular materials

    • Simple geometric distortions applied to the isometric Kelvin cell structures, (the tetrakaidecahedron), are shown to result in equivalent materials with anisotropic Hooke's tensors. The equivalent material models are estimated using a recently published inversion method where the 21 independent elastic constants of the Hooke's tensor are identified. In these cell geometries, some of the faces of the Kelvin cell have been twisted and/or tilted. Numerical experiments suggest that the equivalent material models of the distorted cells exhibit variations in compression, shearing, shear-compression and shear-shear coupling moduli, which are shown to be continuous functions of the degree of twist and tilt applied. When twist and tilt are combined, it is demonstrated that full anisotropy in the elastic properties may be generated. A rotational symmetry without symmetry planes, but having either a tetragonal or a monoclinic elastic symmetry is discussed. Four cell geometries, one isometric and three distorted, were manufactured using masked stereolithography 3D printing technology and measured in a laboratory compression set-up. Results from numerical simulations are compared to the experimental in terms of the compressive modulus.

    • The paper discusses the vibro-acoustic behaviour of a recently proposed novel anisotropic cellular foam geometry derived from the basic symmetric Kelvin cell geometry. The associated equivalent elastic material properties are estimated based on a recently developed inversion method, in terms of the 21 elastic constants of the Hooke's tensor. The dynamic behaviour of the cellular material configuration and its homogeneous model by applying the equivalent material properties are studied under arbitrary excitation. The correlation between the vibro-acoustic behaviour, foam density, and material anisotropy is studied based on finite element simulations. High correlations of eigenfrequency and eigenmode shapes between the target cellular foam model and the equivalent solid model are found. A set of empirical relations are proposed, linking the micro-structure in low to high relative density for the anisotropic cellular geometries investigated. Potential applications in vibro-acoustic of the proposed anisotropic foam are discussed

    • Together with the optimal basic design, buried heterostructure quantum cascade laser (BH-QCL) with semi-insulating regrowth offers a unique possibility to achieve an effective thermal dissipation and lateral single mode. We demonstrate here the realization of BH-QCLs with a single-step regrowth of highly resistive (>1 x 10(8) ohm . cm) semi-insulating InP: Fe in <45 min for the first time in a flexible hydride vapor phase epitaxy process for burying ridges etched down to 10 to 15 mu m depth, both with and without mask overhang. The fabricated BH-QCLs emitting at similar to 4.7 and similar to 5.5 mu m were characterized. 2-mm-long 5.5-mu m lasers with a ridge width of 17 to 22 mu m, regrown with mask overhang, exhibited no leakage current. Large width and high doping in the structure did not permit high current density for continuous wave (CW) operation. 5-mm-long 4.7-mu m BH-QCLs of ridge widths varying from 6 to 14 mu m regrown without mu mask overhang, besides being spatially monomode, TM00, exhibited wall plug efficiency (WPE) of similar to 8 to 9% with an output power of 1.5 to 2.5 W at room temperature and under CW operation. Thus, we demonstrate a quick, flexible, and single-step regrowth process with good planarization for realizing buried QCLs leading to monomode, high power, and high WPE.

    • The authors investigated whether PCSK9 inhibition could represent a therapeutic strategy in calcific aortic valve stenosis (CAVS). A meta-analysis of 10 studies was performed to determine the impact of the PCSK9 R46L variant on CAVS, and the authors found that CAVS was less prevalent in carriers of this variant (odds ratio: 0.80 [95% confidence interval: 0.70 to 0.91]; p = 0.0011) compared with noncarriers. PCSK9 expression was higher in the aortic valves of patients CAVS compared with control patients. In human valve interstitials cells submitted to a pro-osteogenic medium, PCSK9 levels increased and a PCSK9 neutralizing antibody significantly reduced calcium accumulation.

    • Background: Lipoprotein-associated phospholipase A2 (Lp-PLA2) activity has been shown to predict calcific aortic valve stenosis (CAVS) outcomes. Our objective was to test the association between plasma Lp-PLA2 activity and genetically elevated Lp-PLA2 mass/activity with CAVS in humans. Methods and results: Lp-PLA2 activity was measured in 890 patients undergoing cardiac surgery, including 476 patients undergoing aortic valve replacement for CAVS and 414 control patients undergoing coronary artery bypass grafting. After multivariable adjustment, Lp-PLA2 activity was positively associated with the presence of CAVS (OR=1.21 (95% CI 1.04 to 1.41) per SD increment). We selected four single nucleotide polymorphisms (SNPs) at the PLA2G7 locus associated with either Lp-PLA2 mass or activity (rs7756935, rs1421368, rs1805017 and rs4498351). Genetic association studies were performed in eight cohorts: Quebec-CAVS (1009 cases/1017 controls), UK Biobank (1350 cases/349 043 controls), European Prospective Investigation into Cancer and Nutrition-Norfolk (504 cases/20 307 controls), Genetic Epidemiology Research on Aging (3469 cases/51 723 controls), Malmö Diet and Cancer Study (682 cases/5963 controls) and three French cohorts (3123 cases/6532 controls), totalling 10 137 CAVS cases and 434 585 controls. A fixed-effect meta-analysis using the inverse-variance weighted method revealed that none of the four SNPs was associated with CAVS (OR=0.99 (95% CI 0.96 to 1.02, p=0.55) for rs7756935, 0.97 (95% CI 0.93 to 1.01, p=0.11) for rs1421368, 1.00 (95% CI 1.00 to 1.01, p=0.29) for rs1805017, and 1.00 (95% CI 0.97 to 1.04, p=0.87) for rs4498351). Conclusions: Higher Lp-PLA2 activity is significantly associated with the presence of CAVS and might represent a biomarker of CAVS in patients with heart disease. Results of our genetic association study suggest that Lp-PLA2 is however unlikely to represent a causal risk factor or therapeutic target for CAVS.

  • Diglyme based electrolytes for sodium-ion batteries

    • Sodium-ion batteries (SIBs) are currently being considered for large-scale energy storage. Optimization of SIB electrolytes is, however, still largely lacking. Here we exhaustively evaluate NaPF6 in diglyme as an electrolyte of choice, via both physicochemical properties and extensive electrochemical tests including half as well as full cells. Fundamentally, the ionic conductivity is found to be quite comparable to carbonate based electrolytes and to obey the fractional Walden rule with viscosity. We find Na metal to work well as a reference electrode and the electrochemical stability, evaluated potentiostatically for various electrodes and corroborated by DFT calculations, to be satisfactory in the entire voltage range 0-4.4 V. Galvanostatic cycling at C/10 of half and full cells using Na3V2(PO4)(3) (NVP) or Na3V2(PO4)(2)F-3 (NVPF) as cathodes and hard carbon (HC) as anodes indicates rapid capacity fading in cells with HC anodes, possibly originating in a lack of a stable SEI or by trapping of sodium. Aiming to understand this capacity fade further, we conducted a GC/MS analysis to determine electrolyte reduction products and to propose reduction pathways, concluding that oligomer and/or alkoxide formation is possible. Overall, the promising results should warrant further investigations of diglyme based electrolytes for modern SIB development, albeit avoiding HC anodes.
