  • Meta-Analyses of Genome-Wide Association Studies for Postpartum Depression

    • OBJECTIVE: Postpartum depression (PPD) is a common subtype of major depressive disorder (MDD) that is more heritable, yet is understudied in psychiatric genetics. The authors conducted meta-analyses of genome-wide association studies (GWASs) to investigate the genetic architecture of PPD.METHOD: Meta-analyses were conducted on 18 cohorts of European ancestry (17,339 PPD cases and 53,426 controls), one cohort of East Asian ancestry (975 cases and 3,780 controls), and one cohort of African ancestry (456 cases and 1,255 controls), totaling 18,770 PPD cases and 58,461 controls. Post-GWAS analyses included 1) single-nucleotide polymorphism (SNP)-based heritability ([Formula: see text]), 2) genetic correlations between PPD and other phenotypes, and 3) enrichment of the PPD GWAS findings in 27 human tissues and 265 cell types from the mouse central and peripheral nervous system.RESULTS: No SNP achieved genome-wide significance in the European or the trans-ancestry meta-analyses. The [Formula: see text] of PPD was 0.14 (SE=0.02). Significant genetic correlations were estimated for PPD with MDD, bipolar disorder, anxiety disorders, posttraumatic stress disorder, insomnia, age at menarche, and polycystic ovary syndrome. Cell-type enrichment analyses implicate inhibitory neurons in the thalamus and cholinergic neurons within septal nuclei of the hypothalamus, a pattern that differs from MDD.CONCLUSIONS: While more samples are needed to reach genome-wide levels of significance, the results presented confirm PPD as a polygenic and heritable phenotype. There is also evidence that despite a high correlation with MDD, PPD may have unique genetic components. Cell enrichment results suggest GABAergic neurons, which converge on a common mechanism with the only medication approved by the U.S. Food and Drug Administration for PPD (brexanolone).

    • We aim to investigate the aerodynamic and acoustics characteristics of a twin square jet using an implicit Large Eddy Simulation (ILES). A screeching cold jet condition, a nozzle pressure ratio (NPR) of 3.0, is considered to simulate a coupled twin-jet. A second-order central scheme with a modified version of Jameson’s artificial dissipation is adopted to damp numerical oscillations and to mimic the effect of small-scale turbulence without an explicit subgrid-scale (SGS) model. Numerical results show that the overall trends of time-averaged streamwise velocity profiles are similar to the experimental data, with the largest differences observed at locations associated with the presence of the shock-cell structures. A detailed investigation of the flow fluctuations in jet shear layers is performed. The amplitude of the velocity fluctuations is highly dependent on the location of the shear layers with respect to the twin-jet configuration (upper, lateral, or inner). The coupling mode of twin jets associated with the screech tone is determined as a symmetrical flapping mode be a two-points spacetime cross-correlation analysis. The overall trends of near-field pressure fluctuation spectra by LES agree well with the experimental results in both upstream and downstream regions. Near-field pressure fluctuation spectra by ILES agree well with the experimentally obtained spectra at different locations in the nozzle exit plane as well as at several downstream locations in the near-field acoustic region. The highest screech tone is observed at the inter-nozzle region where superposition of in-phase waves and standing waves are found. Fourier phase and amplitude fields at the fundamental frequency also confirm the symmetrical flapping mode of the twin jets by showing in-phase relations of hydrodynamic/acoustic waves and noise directivities.

    • The challenging problem of noise generation and propagation in automotive turbocharging systems is of real interest from both scientific and practical points of view. Robust and fast steady-state fluid flow calculations, complemented by acoustic analogies can represent valuable tools to be used for a quick assessment of the problem during e.g. design phase, and a starting point for more in-depth future unsteady calculations. Thus, as a part of the initial phase of a long-term project, a steady-state Reynolds Averaged Navier-Stokes (RANS) flow analysis is carried out for a specific automotive turbocharger compressor geometry. Acoustic data are extracted by means of aeroacoustics models available within the framework of the STAR-CCM+ solver (i.e. Curle and Proudman acoustic analogies, respectively). This part of the work focuses on the discussion and comparison of the aeroacoustic models, and their suitability towards predicting flow and acoustics trends corresponding to the operating conditions investigated. However, given the unsteady nature of acoustics, the project will have to develop towards an investigation of the problem using more expensive, but more accurate, Large Eddy Simulation (LES) calculations. An entire compressor map with 80 operating conditions was simulated, yielding trends in the behaviour of the performance parameters for the analysed compressor. Detailed results calculated on the same compressor speed-line for one design and one off-design operating conditions are presented in terms of time-averaged pressure coefficient, Mach number, and acoustic power distributions. A total acoustic power map has been generated based on the outcome from the Curle and Proudman acoustic models, giving an indication of the noisiest operating conditions.

    • A parallel solver of a splitting approach for Computational Aero-Acoustics (CAA) is presented. The solver is based on a hybrid approach, where the flow field and acoustic fields are computed as separate but coupled processes. The Large Eddy Simulation (LES) based flow solver is parallelized using a domain decomposition approach. The performance of the solver has been evaluated and good parallel performance has been obtained. The acoustical solver utilizes the LES data for computing the acoustical sources. The combined solver has been successfully applied for the computation of the aeroacoustic field in a model gas turbine combustion chamber.
