    • In 2008 we published the first set of guidelines for standardizing research in autophagy. Since then, research on this topic has continued to accelerate, and many new scientists have entered the field. Our knowledge base and relevant new technologies have also been expanding. Accordingly, it is important to update these guidelines for monitoring autophagy in different organisms. Various reviews have described the range of assays that have been used for this purpose. Nevertheless, there continues to be confusion regarding acceptable methods to measure autophagy, especially in multicellular eukaryotes. A key point that needs to be emphasized is that there is a difference between measurements that monitor the numbers or volume of autophagic elements (e.g., autophagosomes or autolysosomes) at any stage of the autophagic process vs. those that measure flux through the autophagy pathway (i.e., the complete process); thus, a block in macroautophagy that results in autophagosome accumulation needs to be differentiated from stimuli that result in increased autophagic activity, defined as increased autophagy induction coupled with increased delivery to, and degradation within, lysosomes (in most higher eukaryotes and some protists such as Dictyostelium) or the vacuole (in plants and fungi). In other words, it is especially important that investigators new to the field understand that the appearance of more autophagosomes does not necessarily equate with more autophagy. In fact, in many cases, autophagosomes accumulate because of a block in trafficking to lysosomes without a concomitant change in autophagosome biogenesis, whereas an increase in autolysosomes may reflect a reduction in degradative activity. Here, we present a set of guidelines for the selection and interpretation of methods for use by investigators who aim to examine macroautophagy and related processes, as well as for reviewers who need to provide realistic and reasonable critiques of papers that are focused on these processes. These guidelines are not meant to be a formulaic set of rules, because the appropriate assays depend in part on the question being asked and the system being used. In addition, we emphasize that no individual assay is guaranteed to be the most appropriate one in every situation, and we strongly recommend the use of multiple assays to monitor autophagy. In these guidelines, we consider these various methods of assessing autophagy and what information can, or cannot, be obtained from them. Finally, by discussing the merits and limits of particular autophagy assays, we hope to encourage technical innovation in the field.

    • Background Up-to-date evidence about levels and trends in disease and injury incidence, prevalence, and years lived with disability (YLDs) is an essential input into global, regional, and national health policies. In the Global Burden of Disease Study 2013 (GBD 2013), we estimated these quantities for acute and chronic diseases and injuries for 188 countries between 1990 and 2013. Methods Estimates were calculated for disease and injury incidence, prevalence, and YLDs using GBD 2010 methods with some important refinements. Results for incidence of acute disorders and prevalence of chronic disorders are new additions to the analysis. Key improvements include expansion to the cause and sequelae list, updated systematic reviews, use of detailed injury codes, improvements to the Bayesian meta-regression method (DisMod-MR), and use of severity splits for various causes. An index of data representativeness, showing data availability, was calculated for each cause and impairment during three periods globally and at the country level for 2013. In total, 35 620 distinct sources of data were used and documented to calculated estimates for 301 diseases and injuries and 2337 sequelae. The comorbidity simulation provides estimates for the number of sequelae, concurrently, by individuals by country, year, age, and sex. Disability weights were updated with the addition of new population-based survey data from four countries. Findings Disease and injury were highly prevalent; only a small fraction of individuals had no sequelae. Comorbidity rose substantially with age and in absolute terms from 1990 to 2013. Incidence of acute sequelae were predominantly infectious diseases and short-term injuries, with over 2 billion cases of upper respiratory infections and diarrhoeal disease episodes in 2013, with the notable exception of tooth pain due to permanent caries with more than 200 million incident cases in 2013. Conversely, leading chronic sequelae were largely attributable to non-communicable diseases, with prevalence estimates for asymptomatic permanent caries and tension-type headache of 2.4 billion and 1.6 billion, respectively. The distribution of the number of sequelae in populations varied widely across regions, with an expected relation between age and disease prevalence. YLDs for both sexes increased from 537.6 million in 1990 to 764.8 million in 2013 due to population growth and ageing, whereas the age-standardised rate decreased little from 114.87 per 1000 people to 110.31 per 1000 people between 1990 and 2013. Leading causes of YLDs included low back pain and major depressive disorder among the top ten causes of YLDs in every country. YLD rates per person, by major cause groups, indicated the main drivers of increases were due to musculoskeletal, mental, and substance use disorders, neurological disorders, and chronic respiratory diseases; however HIV/AIDS was a notable driver of increasing YLDs in sub-Saharan Africa. Also, the proportion of disability-adjusted life years due to YLDs increased globally from 21.1% in 1990 to 31.2% in 2013. Interpretation Ageing of the world's population is leading to a substantial increase in the numbers of individuals with sequelae of diseases and injuries. Rates of YLDs are declining much more slowly than mortality rates. The non-fatal dimensions of disease and injury will require more and more attention from health systems. The transition to non-fatal outcomes as the dominant source of burden of disease is occurring rapidly outside of sub-Saharan Africa. Our results can guide future health initiatives through examination of epidemiological trends and a better understanding of variation across countries.

    • Background: There is tremendous potential for genome sequencing to improve clinical diagnosis and care once it becomes routinely accessible, but this will require formalizing research methods into clinical best practices in the areas of sequence data generation, analysis, interpretation and reporting. The CLARITY Challenge was designed to spur convergence in methods for diagnosing genetic disease starting from clinical case history and genome sequencing data. DNA samples were obtained from three families with heritable genetic disorders and genomic sequence data were donated by sequencing platform vendors. The challenge was to analyze and interpret these data with the goals of identifying disease-causing variants and reporting the findings in a clinically useful format. Participating contestant groups were solicited broadly, and an independent panel of judges evaluated their performance. Results: A total of 30 international groups were engaged. The entries reveal a general convergence of practices on most elements of the analysis and interpretation process. However, even given this commonality of approach, only two groups identified the consensus candidate variants in all disease cases, demonstrating a need for consistent fine-tuning of the generally accepted methods. There was greater diversity of the final clinical report content and in the patient consenting process, demonstrating that these areas require additional exploration and standardization. Conclusions: The CLARITY Challenge provides a comprehensive assessment of current practices for using genome sequencing to diagnose and report genetic diseases. There is remarkable convergence in bioinformatic techniques, but medical interpretation and reporting are areas that require further development by many groups.

    • BACKGROUND: The Global Burden of Disease, Injuries, and Risk Factor study 2013 (GBD 2013) is the first of a series of annual updates of the GBD. Risk factor quantification, particularly of modifiable risk factors, can help to identify emerging threats to population health and opportunities for prevention. The GBD 2013 provides a timely opportunity to update the comparative risk assessment with new data for exposure, relative risks, and evidence on the appropriate counterfactual risk distribution.METHODS: Attributable deaths, years of life lost, years lived with disability, and disability-adjusted life-years (DALYs) have been estimated for 79 risks or clusters of risks using the GBD 2010 methods. Risk-outcome pairs meeting explicit evidence criteria were assessed for 188 countries for the period 1990-2013 by age and sex using three inputs: risk exposure, relative risks, and the theoretical minimum risk exposure level (TMREL). Risks are organised into a hierarchy with blocks of behavioural, environmental and occupational, and metabolic risks at the first level of the hierarchy. The next level in the hierarchy includes nine clusters of related risks and two individual risks, with more detail provided at levels 3 and 4 of the hierarchy. Compared with GBD 2010, six new risk factors have been added: handwashing practices, occupational exposure to trichloroethylene, childhood wasting, childhood stunting, unsafe sex, and low glomerular filtration rate. For most risks, data for exposure were synthesised with a Bayesian meta-regression method, DisMod-MR 2.0, or spatial-temporal Gaussian process regression. Relative risks were based on meta-regressions of published cohort and intervention studies. Attributable burden for clusters of risks and all risks combined took into account evidence on the mediation of some risks such as high body-mass index (BMI) through other risks such as high systolic blood pressure and high cholesterol.FINDINGS: All risks combined account for 57·2% (95% uncertainty interval [UI] 55·8-58·5) of deaths and 41·6% (40·1-43·0) of DALYs. Risks quantified account for 87·9% (86·5-89·3) of cardiovascular disease DALYs, ranging to a low of 0% for neonatal disorders and neglected tropical diseases and malaria. In terms of global DALYs in 2013, six risks or clusters of risks each caused more than 5% of DALYs: dietary risks accounting for 11·3 million deaths and 241·4 million DALYs, high systolic blood pressure for 10·4 million deaths and 208·1 million DALYs, child and maternal malnutrition for 1·7 million deaths and 176·9 million DALYs, tobacco smoke for 6·1 million deaths and 143·5 million DALYs, air pollution for 5·5 million deaths and 141·5 million DALYs, and high BMI for 4·4 million deaths and 134·0 million DALYs. Risk factor patterns vary across regions and countries and with time. In sub-Saharan Africa, the leading risk factors are child and maternal malnutrition, unsafe sex, and unsafe water, sanitation, and handwashing. In women, in nearly all countries in the Americas, north Africa, and the Middle East, and in many other high-income countries, high BMI is the leading risk factor, with high systolic blood pressure as the leading risk in most of Central and Eastern Europe and south and east Asia. For men, high systolic blood pressure or tobacco use are the leading risks in nearly all high-income countries, in north Africa and the Middle East, Europe, and Asia. For men and women, unsafe sex is the leading risk in a corridor from Kenya to South Africa.INTERPRETATION: Behavioural, environmental and occupational, and metabolic risks can explain half of global mortality and more than one-third of global DALYs providing many opportunities for prevention. Of the larger risks, the attributable burden of high BMI has increased in the past 23 years. In view of the prominence of behavioural risk factors, behavioural and social science research on interventions for these risks should be strengthened.     • Background Up-to-date evidence on levels and trends for age-sex-specifi c all-cause and cause-specifi c mortality is essential for the formation of global, regional, and national health policies. In the Global Burden of Disease Study 2013 (GBD 2013) we estimated yearly deaths for 188 countries between 1990, and 2013. We used the results to assess whether there is epidemiological convergence across countries. Methods We estimated age-sex-specifi c all-cause mortality using the GBD 2010 methods with some refinements to improve accuracy applied to an updated database of vital registration, survey, and census data. We generally estimated cause of death as in the GBD 2010. Key improvements included the addition of more recent vital registration data for 72 countries, an updated verbal autopsy literature review, two new and detailed data systems for China, and more detail for Mexico, UK, Turkey, and Russia. We improved statistical models for garbage code redistribution. We used six different modelling strategies across the 240 causes; cause of death ensemble modelling (CODEm) was the dominant strategy for causes with sufficient information. Trends for Alzheimer's disease and other dementias were informed by meta-regression of prevalence studies. For pathogen-specifi c causes of diarrhoea and lower respiratory infections we used a counterfactual approach. We computed two measures of convergence (inequality) across countries: the average relative difference across all pairs of countries (Gini coefficient) and the average absolute difference across countries. To summarise broad findings, we used multiple decrement life-tables to decompose probabilities of death from birth to exact age 15 years, from exact age 15 years to exact age 50 years, and from exact age 50 years to exact age 75 years, and life expectancy at birth into major causes. For all quantities reported, we computed 95% uncertainty intervals (UIs). We constrained cause-specific fractions within each age-sex-country-year group to sum to all-cause mortality based on draws from the uncertainty distributions. Findings Global life expectancy for both sexes increased from 65.3 years (UI 65.0-65.6) in 1990, to 71.5 years (UI 71.0-71.9) in 2013, while the number of deaths increased from 47.5 million (UI 46.8-48.2) to 54.9 million (UI 53.6-56.3) over the same interval. Global progress masked variation by age and sex: for children, average absolute diff erences between countries decreased but relative diff erences increased. For women aged 25-39 years and older than 75 years and for men aged 20-49 years and 65 years and older, both absolute and relative diff erences increased. Decomposition of global and regional life expectancy showed the prominent role of reductions in age-standardised death rates for cardiovascular diseases and cancers in high-income regions, and reductions in child deaths from diarrhoea, lower respiratory infections, and neonatal causes in low-income regions. HIV/AIDS reduced life expectancy in southern sub-Saharan Africa. For most communicable causes of death both numbers of deaths and age-standardised death rates fell whereas for most non-communicable causes, demographic shifts have increased numbers of deaths but decreased age-standardised death rates. Global deaths from injury increased by 10.7%, from 4.3 million deaths in 1990 to 4.8 million in 2013; but age-standardised rates declined over the same period by 21%. For some causes of more than 100 000 deaths per year in 2013, age-standardised death rates increased between 1990 and 2013, including HIV/AIDS, pancreatic cancer, atrial fibrillation and flutter, drug use disorders, diabetes, chronic kidney disease, and sickle-cell anaemias. Diarrhoeal diseases, lower respiratory infections, neonatal causes, and malaria are still in the top five causes of death in children younger than 5 years. The most important pathogens are rotavirus for diarrhoea and pneumococcus for lower respiratory infections. Country-specific probabilities of death over three phases of life were substantially varied between and within regions. Interpretation For most countries, the general pattern of reductions in age-sex specifi c mortality has been associated with a progressive shift towards a larger share of the remaining deaths caused by non-communicable disease and injuries. Assessing epidemiological convergence across countries depends on whether an absolute or relative measure of inequality is used. Nevertheless, age-standardised death rates for seven substantial causes are increasing, suggesting the potential for reversals in some countries. Important gaps exist in the empirical data for cause of death estimates for some countries; for example, no national data for India are available for the past decade.

  • An Earth system law perspective on governing social-hydrological systems in the Anthropocene

    • The global hydrological cycle is characterized by complex interdependencies and self-regulating feedbacks that keep water in an ever-evolving state of flux at local, regional, and global levels. Increasingly, the scale of human impacts in the Anthropocene is altering the dynamics of this cycle, which presents additional challenges for water governance. Earth system law provides an important approach for addressing gaps in governance that arise from the mismatch between the global hydrological cycle and dispersed regulatory architecture across institutions and geographic regions. In this article, we articulate the potential for Earth system law to account for core hydrological problems that complicate water governance, including delay, redistribution, intertwinements, permanence, and scale. Through merging concepts from Earth system law with existing policy and legal principles, we frame an approach for addressing hydrological problems in the Anthropocene and strengthening institutional fit between established governance systems and the global hydrological cycle. We discuss how such an approach can be applied, and the challenges and implications for governing water as a cycle and complex social-hydrological system, both in research and practice.

  • Introducing Resilience Practice to Watershed Groups : What Are the Learning Effects?

    • Resilience as an organizing framework for addressing dynamics of social-ecological systems has experienced strong uptake; however, its application is nascent. This research study aimed to address the gap between resilience thinking and practice by focusing on learning, a key aspect of resilience. Two Canadian watershed groups were led in 2-day workshops focused on resilience. Learning effects were measured using a survey administered both before and after the workshop, and a qualitative survey was administered 6 months later to understand longer term effects. Short-term learning effects were similar between the two case studies, with strong cognitive and relational learning and less normative learning. Longer term effects showed enduring cognitive and normative learning in both cases; however, relational learning persisted only in the watershed where a resilience practice approach to watershed planning had been incorporated. Future research directions include refinements to the learning measurement methodology and continuing to build resilience practice literature.

  • Degrees of change toward polycentric transboundary water governance : exploring the Columbia river and the lesotho highlands water project

    • Complex challenges emerging in transboundary river basins reveal a need to include a range of interests and actors in governance processes. Polycentric governance is one framework that can address this need and inform adaptive and resilient governance processes in transboundary basins as linked social and ecological systems. Here, we explore whether and how nonstate actors might be contributing to a shift in governance toward polycentric systems for the Columbia River (Canada/USA) and the Lesotho Highlands Water Project (Lesotho/South Africa). Using data gathered from 60 in-depth interviews, our empirical results illustrate four governance themes relevant to the emergence of polycentricity in the case study basins: Authority, flexibility, coordination activities, and information sharing. Although the emergence of polycentricity is limited by existing state-centric governance regimes, these regimes show evidence that polycentric traits are supplementing existing governance systems, influencing policy processes, and introducing a range of management values.

    • This paper explores the links between contemporary bordering processes, Indigenous nations traditional territories, and transboundary water governance processes, using the case of the Columbia River Treaty (CRT) modernization process. We posit the Columbia River is shared not just by two nations, but also by multiple Indigenous nations with various inter-nation borders. To-date, the implications of this in practice do not appear to mean a re-imagination of borders, changes in legal authority for CRT renegotiation and implementation, or rethinking the state-centric institutions in which governance of the Columbia River is based. Three primary themes emerged from the empirical data that illustrate: (1) a reaffirmation of state-centric discourse on borders and bordering processes in CRT modernization, while (2) at the same time we see changes in the legal landscape in Canada and the U.S. that inform the obligations of colonial governments to move towards collaboration and shared governance with Indigenous nations on a government-to-government basis on issues impacting Indigenous interests. And, (3) emerging are the seeds of governance structures that seek to engage Indigenous nations within CRT renegotiation and implementation, including potentially providing a seat at the renegotiation table and including Indigenous nations within implementation structures for a modernized CRT.

  • The Dark Side of Transformation : Latent Risks in Contemporary Sustainability Discourse

    • The notion of transformation is gaining traction in contemporary sustainability debates. New ways of theorising and supporting transformations are emerging and, so the argument goes, opening exciting spaces to (re)imagine and (re)structure radically different futures. Yet, questions remain about how the term is being translated from an academic concept into an assemblage of normative policies and practices, and how this process might shape social, political, and environmental change. Motivated by these questions, we identify five latent risks associated with discourse that frames transformation as apolitical and/or inevitable. We refer to these risks as the dark side of transformation. While we cannot predict the future of radical transformations towards sustainability, we suggest that scientists, policymakers, and practitioners need to consider such change in more inherently plural and political ways.

    • Formalised knowledge systems, including universities and research institutes, are important for contemporary societies. They are, however, also arguably failing humanity when their impact is measured against the level of progress being made in stimulating the societal changes needed to address challenges like climate change. In this research we used a novel futures-oriented and participatory approach that asked what future envisioned knowledge systems might need to look like and how we might get there. Findings suggest that envisioned future systems will need to be much more collaborative, open, diverse, egalitarian, and able to work with values and systemic issues. They will also need to go beyond producing knowledge about our world to generating wisdom about how to act within it. To get to envisioned systems we will need to rapidly scale methodological innovations, connect innovators, and creatively accelerate learning about working with intractable challenges. We will also need to create new funding schemes, a global knowledge commons, and challenge deeply held assumptions. To genuinely be a creative force in supporting longevity of human and non-human life on our planet, the shift in knowledge systems will probably need to be at the scale of the enlightenment and speed of the scientific and technological revolution accompanying the second World War. This will require bold and strategic action from governments, scientists, civic society and sustained transformational intent.

    • Policy actors address complex environmental problems by engaging in multiple and often interdependent policy issues. Policy issue interdependencies imply that efforts by actors to address separate policy issues can either reinforce (‘win–win’) or counteract (‘trade-off’) each other. Thus, if interdependent issues are managed in isolation instead of being coordinated, the most effective and well-balanced solution to the underlying problem might never be realised. This study asks if reinforcing and counteracting interdependencies have different impacts on perception and collaboration. Our empirical study of collaborative water governance in the Norrström basin, Sweden, shows that policy actors often avoid collaborating when the policy issues exhibit reinforcing interdependencies. Our evidence indicates a perceived infeasibility of acting on reinforcing interdependencies. We also find that actors do not consider counteracting interdependencies (‘trade-offs’) at all when they engage in collaboration. Further, even though actors were aware of counteracting and reinforcing interdependencies, our analyses suggest they might be less aware of the former. These findings illustrate that actors either avoid each other due to policy issue interdependencies or, at best, ignore existing interdependencies when engaging in collaboration. Our study highlights the importance of problem perception in accomplishing integrated solutions to complex environmental problems, and of how understandings of different types of interdependencies shape collaboration in environmental governance.

In practice, however, environmental policy is still often contained within the traditional responsibilities of the public sector and frequently judged ineffective, particularly in the European context. Whether and how interdependencies are actually associated with collaboration between policy actors has remained difficult to establish.  This cumulative thesis focuses on interdependent environmental challenges that policy actors need to manage. Specifically, this thesis describes and analyses policy issue interdependencies and how they align with the collaborations of policy actors. In addition, this thesis explores how policy issue interdependencies can be revealed, concretised, and analysed. Interdependencies are effectively represented by networks, both as conceptual models and as analytical methods. Therefore, the studies in this thesis use a multilevel network model to explore the structural alignment between interdependencies and collaboration through the perspective of institutional fit.This thesis reports findings from two research projects. The first project focuses on policy issue interdependencies relating to regional water degradation. This project describes and analyses these interdependencies in relation to collaborative networks across administrative boundaries (Papers I–III). The second project focuses on climate change impacts that propagate through food trade dependencies. This project contributes insights into the effect of climate change on food trade networks that cross national borders, illustrating a need for global climate adaptation (Paper IV).Paper I introduces a methodological procedure for assessing policy issue interdependencies and develops policy issue networks by identifying overlapping causal relationships between policy issues and their environmental targets. By applying the procedure empirically to water governance, the paper shows that policy issue interdependencies vary in degree and type. Paper II combines the policy issue networks from Paper I with collaborative networks of policy actors in a multilevel network to analyse the impact policy issue interdependencies have on who policy actors select for collaborative partners and to clarify if and how patterns of collaboration among actors are formed. Paper III differentiates reinforcing and counteracting policy issue interdependencies and studies how these impact the perceptions and collaborations of the actors. Paper IV, shifting the focus to the global level, analyses climate change impacts related to food trade dependencies across national borders. Specifically, Paper IV investigates the impact of climate change on the structure of global food trade networks and therefore contributes a baseline scenario analysis for future studies that investigate policy issue interdependencies and policy actor collaborations on the global level.

    • INTRODUCTION: Aortic graft infection remains a considerable clinical challenge, and it is unclear which variables are associated with adverse outcomes among patients undergoing partial resection.METHODS: A retrospective, multi-institutional study of patients who underwent partial resection of infected aortic grafts from 2002 to 2014 was performed using a standard database. Baseline demographics, comorbidities, operative, and postoperative variables were recorded. The primary outcome was mortality. Descriptive statistics, Kaplan-Meier (KM) survival analysis, and Cox regression analysis were performed.RESULTS: One hundred fourteen patients at 22 medical centers in 6 countries underwent partial resection of an infected aortic graft. Seventy percent were men with median age 70 years. Ninety-seven percent had a history of open aortic bypass graft: 88 (77%) patients had infected aortobifemoral bypass, 18 (16%) had infected aortobiiliac bypass, and 1 (0.8%) had an infected thoracic graft. Infection was diagnosed at a median 4.3 years post-implant. All patients underwent partial resection followed by either extra-anatomic (47%) or in situ (53%) vascular reconstruction. Median follow-up period was 17 months (IQR 1, 50 months). Thirty-day mortality was 17.5%. The KM-estimated median survival from time of partial resection was 3.6 years. There was no significant survival difference between those undergoing in situ reconstruction or extra-anatomic bypass (P = 0.6). During follow up, 72% of repairs remained patent and 11% of patients underwent major amputation. On univariate Cox regression analysis, Candida infection was associated with increased risk of mortality (HR 2.4; P = 0.01) as well as aortoenteric fistula (HR 1.9, P = 0.03). Resection of a single graft limb only to resection of abdominal (graft main body) infection was associated with decreased risk of mortality (HR 0.57, P = 0.04), as well as those with American Society of Anesthesiologists classification less than 3 (HR 0.35, P = 0.04). Multivariate analysis did not reveal any factors significantly associated with mortality. Persistent early infection was noted in 26% of patients within 30 days postoperatively, and 39% of patients were found to have any post-repair infection during the follow-up period. Two patients (1.8%) were found to have a late reinfection without early persistent postoperative infection. Patients with any post-repair infection were older (67 vs. 60 years, P = 0.01) and less likely to have patent repairs during follow up (59% vs. 32%, P = 0.01). Patients with aortoenteric fistula had a higher rate of any post-repair infection (63% vs. 29%, P < 0.01)CONCLUSION: This large multi-center study suggests that patients who have undergone partial resection of infected aortic grafts may be at high risk of death or post-repair infection, especially older patients with abdominal infection not isolated to a single graft limb, or with Candida infection or aortoenteric fistula. Late reinfection correlated strongly with early persistent postoperative infection, raising concern for occult retained infected graft material.

    • Non-Technical Summary. Human societies are changing where and how water flows through the atmosphere. However, these changes in the atmospheric water cycle are not being managed, nor is there any real sense of where these changes might be headed in the future. Thus, we develop a new economic theory of atmospheric water management, and explore this theory using creative story-based scenarios. These scenarios reveal surprising possibilities for the future of atmospheric water management, ranging from a stock market for transpiration to on-demand weather. We discuss these story-based futures in the context of research and policy priorities in the present day.Technical Summary. Humanity is modifying the atmospheric water cycle, via land use, climate change, air pollution, and weather modification. Historically, atmospheric water was implicitly considered a ‘public good’ since it was neither actively consumed nor controlled. However, given anthropogenic changes, atmospheric water can become a ‘common-pool’ good (consumable) or a ‘club’ good (controllable). Moreover, advancements in weather modification presage water becoming a ‘private’ good, meaning both consumable and controllable. Given the implications, we designed a theoretical framing of atmospheric water as an economic good and used a combination of methods in order to explore possible future scenarios based on human modifications of the atmospheric water cycle. First, a systematic literature search of scholarly abstracts was used in a computational text analysis. Second, the output of the text analysis was matched to different parts of an existing economic goods framework. Then, a group of global water experts were trained and developed story-based scenarios. The resultant scenarios serve as creative investigations of the future of human modification of the atmospheric water cycle. We discuss how the scenarios can enhance anticipatory capacity in the context of both future research frontiers and potential policy pathways including transboundary governance, finance, and resource management.Social Media Summary.     • Food is essential to people and is one of the main ways in which people are connected to the world’s ecosystems. However, food systems often cause ecosystem degradation and produce ill-health, which has generated increasing calls to transform food systems to be more sustainable. The Swedish food system is currently undergoing substantial change. A varied set of local actors have created alternative sustainability initiatives that enact new ways of doing, thinking, and organizing. These actors can increase the transformative impact of their initiatives through multiple actions and a variety of amplification processes. We analyzed the actions adopted by 29 food initiatives active in the Stockholm region using information available online. We conducted 11 interviews to better understand the amplification processes of speeding up (i.e., accelerating impact), scaling up (i.e., influencing higher institutional levels), and scaling deep (i.e., changing values and mind-sets). Our results indicated that the initiatives mainly seek to stabilize and grow their impact while changing the awareness, values, and mind-sets of people concerning the food they consume (scaling deep). However, these approaches raise new questions about whether these actions subvert or reinforce current unsustainable and inequitable system dynamics. We suggest there are distinct steps that local and regional governments could take to support these local actors via collaborations with coordinated forms of initiatives, and fostering changes at the municipality level, but these steps require ongoing, adaptive approaches given the highly complex nature of transformative change and the risks of reinforcing current system dynamics.

    • BackgroundPlasmodium falciparum malaria is associated with anaemia-related morbidity, attributable to host, parasite and drug factors. We quantified the haematological response following treatment of uncomplicated P. falciparum malaria to identify the factors associated with malarial anaemia.MethodsIndividual patient data from eligible antimalarial efficacy studies of uncomplicated P. falciparum malaria, available through the WorldWide Antimalarial Resistance Network data repository prior to August 2015, were pooled using standardised methodology. The haematological response over time was quantified using a multivariable linear mixed effects model with nonlinear terms for time, and the model was then used to estimate the mean haemoglobin at day of nadir and day 7. Multivariable logistic regression quantified risk factors for moderately severe anaemia (haemoglobin < 7 g/dL) at day 0, day 3 and day 7 as well as a fractional fall >= 25% at day 3 and day 7.ResultsA total of 70,226 patients, recruited into 200 studies between 1991 and 2013, were included in the analysis: 50,859 (72.4%) enrolled in Africa, 18,451 (26.3%) in Asia and 916 (1.3%) in South America. The median haemoglobin concentration at presentation was 9.9 g/dL (range 5.0-19.7 g/dL) in Africa, 11.6 g/dL (range 5.0-20.0 g/dL) in Asia and 12.3 g/dL (range 6.9-17.9 g/dL) in South America. Moderately severe anaemia (Hb < 7g/dl) was present in 8.4% (4284/50,859) of patients from Africa, 3.3% (606/18,451) from Asia and 0.1% (1/916) from South America. The nadir haemoglobin occurred on day 2 post treatment with a mean fall from baseline of 0.57 g/dL in Africa and 1.13 g/dL in Asia. Independent risk factors for moderately severe anaemia on day 7, in both Africa and Asia, included moderately severe anaemia at baseline (adjusted odds ratio (AOR) = 16.10 and AOR = 23.00, respectively), young age (age < 1 compared to >= 12 years AOR = 12.81 and AOR = 6.79, respectively), high parasitaemia (AOR = 1.78 and AOR = 1.58, respectively) and delayed parasite clearance (AOR = 2.44 and AOR = 2.59, respectively). In Asia, patients treated with an artemisinin-based regimen were at significantly greater risk of moderately severe anaemia on day 7 compared to those treated with a non-artemisinin-based regimen (AOR = 2.06 [95%CI 1.39-3.05], p < 0.001).ConclusionsIn patients with uncomplicated P. falciparum malaria, the nadir haemoglobin occurs 2 days after starting treatment. Although artemisinin-based treatments increase the rate of parasite clearance, in Asia they are associated with a greater risk of anaemia during recovery.

    • In statistics, samples are drawn from a population in a data-generating process (DGP). Standard errors measure the uncertainty in estimates of population parameters. In science, evidence is generated to test hypotheses in an evidence-generating process (EGP). We claim that EGP variation across researchers adds uncertainty-nonstandard errors (NSEs). We study NSEs by letting 164 teams test the same hypotheses on the same data. NSEs turn out to be sizable, but smaller for more reproducible or higher rated research. Adding peer-review stages reduces NSEs. We further find that this type of uncertainty is underestimated by participants.

    • COVID-19 posed threats for health and well-being directly, but it also revealed and exacerbated social–ecological inequalities, worsening hunger and poverty for millions. For those focused on transforming complex and problematic system dynamics, the question was whether such devastation could create a formative moment in which transformative change could become possible. Our study examines the experiences of change agents in six African countries engaged in efforts to create or support transformative change processes. To better understand the relationship between crisis, agency, and transformation, we explored how they navigated their changed conditions and the responses to COVID-19. We document three impacts: economic impacts, hunger, and gender-based violence and we examine how they (re)shaped the opportunity contexts for change. Finally, we identify four kinds of uncertainties that emerged as a result of policy responses, including uncertainty about the: (1) robustness of preparing a system to sustain a transformative trajectory, (2) sequencing and scaling of changes within and across systems, (3) hesitancy and exhaustion effects, and (4) long-term effects of surveillance, and we describe the associated change agent strategies. We suggest these uncertainties represent new theoretical ground for future transformations research.

    • The ability of individuals and groups to identify, assess, and pursue alternative possible futures is an essential component of their ability to deliberately and collectively respond to major sustainability challenges rather than experience unguided or forced change. Deliberately engaging in transformation processes inevitably requires imagination. We refer to imagination for transformations as interdependent cognitive and social processes that create representations of present and possible future states of the world that can inform public deliberation, policy, decision making, and behavior from the individual to the global scale. We contend that imagination is an essential capacity for securing ecological, social, economic, and cultural well-being in times of rapid and often unpredictable global change. We sketch an emerging interdisciplinary research agenda on imagination as a transformational capacity and its role in transformation processes, building on contributions to a special issue on this subject. We specifically focus on imagination in relationship to transformative agency, causation, and individual-collective dynamics. Our aim is to identify research questions and challenges that are most pressing with a view to supporting efforts of transformations toward sustainability.

    • The distinction between adaptive and transformative capacities is still not well understood, and in this study we aimed to build a transformative learning space to strengthen transformative capacities. We proposed that two capacities will be essential to transformation: the capacity to navigate emergence and cross-scale systems reflexivity. We outline our efforts to design and deliver a Global Fellowship program in social innovation, intended to strengthen these two capacities among practitioners already engaged in socially innovative work. Results indicated that the concepts, frameworks, and experiences introduced through the Fellowship led to four key insights about these capacities. Firstly, individual Fellows and their organizations were able to see some complex system dynamics that were previously invisible, which in turn, allowed Fellows to see the distribution of resources and agency across the system in new ways. Secondly, engaging with diversity is essential in social innovation and transformative change processes, and system reflexivity aided in doing this. Additionally, Fellows indicated they were able to identify different kinds of opportunities and the generative potential that can lie within social-ecological systems. Lastly, the findings demonstrate the challenging nature of crossing scales and how a transformative space, such as a Fellowship, helps to practice the experience of contestation, unpredictability, and the uncontrollable dynamics of transformation and social innovation.

    • Faced with numerous seemingly intractable social and environmental challenges, many scholars and practitioners are increasingly interested in understanding how to actively engage and transform the existing systems holding such problems in place. Although a variety of analytical models have emerged in recent years, most emphasize either the social or ecological elements of such transformations rather than their coupled nature. To address this, first we have presented a definition of the core elements of a social-ecological system (SES) that could potentially be altered in a transformation. Second, we drew on insights about transformation from three branches of literature focused on radical change, i.e., social movements, socio-technical transitions, and social innovation, and gave consideration to the similarities and differences with the current studies by resilience scholars. Drawing on these findings, we have proposed a framework that outlines the process and phases of transformative change in an SES. Future research will be able to utilize the framework as a tool for analyzing the alteration of social-ecological feedbacks, identifying critical barriers and leverage points and assessing the outcome of social-ecological transformations.

    • A growing need for innovation in water policy is increasingly recognized within water policy and governance scholarship, but the types of innovation and changes being considered or undertaken, and the conditions that enable or hinder those changes remain unclear. A systematic review of water policy reform literature was undertaken to investigate how innovation is defined in this area of scholarship and the enabling conditions or barriers shaping the innovation process. The findings of the review demonstrated that the mainstream water policy reform scholarship that examines innovation is limited. A small portion of the water policy reform literature that addresses innovation considers different types of policy changes as innovative. Therefore, the results are used to propose a typology of water policy innovations. Furthermore, the results demonstrated that preliminary knowledge about the role of policy entrepreneurs, networks, social learning, adaptive approaches, and niche experiments in the innovation process emerge in a sub-set of the water policy reform literature.

    • Moving from a state of war or violent conflict will require a transformation, but there are no guarantees that transformations automatically lead to peace, sustainability, and justice. This review focuses on the temporary phase when a system is in limbo between the existing, dominant state and a new alternative state. We combine insights from a resilience approach to transformations with peacebuilding and transformative justice studies to focus on three roles that hybrid approaches to transformative and transitional justice may play in this phase, including 1) addressing ‘backlash’ dynamics, 2) strengthening the capacities needed to navigate cross-scale dynamics of conflict, and 3) responding to additional shocks, crises, and disturbances beyond the primary conflicts. Together, these findings advance the theoretical foundations for understanding peacebuilding as a transformative change process.

    • After tracing the antecedents of the concept and considering its intersection in social innovation research, we put forward the argument that the Anthropocene concept points to three areas of thought that are strategically imperative and must be accelerated if social innovation theory and practice is to prove transformative and respond to the challenges associated with the Anthropocene. First, we contend that the current debate on social innovation for sustainability lacks a deeper focus on human-environmental interactions and the related feedbacks, which will be necessary to understand and achieve large-scale change and transformations to global sustainability. Many innovations focus on only the social or the ecological, and we believe a more integrated approach will be needed moving forward. Second, social innovation research must confront the path-dependencies embedded within systems, and we propose that the act of "bricolage," which recombines existing elements in novel ways, will be essential, rather than single variable solutions, which currently dominate social innovation discussions. Finally, we put forward the idea that confronting the cross-scalar nature of the Anthropocene requires revisiting both the scope and temporal nature of social innovations that are most typically focused upon by scholars and funders alike. We believe the concept of the Anthropocene creates new opportunities for social innovation scholars to imagine new possibilities.

    • Creating a just and sustainable planet will require not only small changes, but also systemic transformations in how humans relate to the planet and to each other, i.e., social-ecological transformations. We suggest there is a need for collaborative environments where experimentation with new configurations of social-ecological systems can occur, and we refer to these as transformative spaces. In this paper, we seek a better understanding of how to design and enable the creation of transformative spaces in a development context. We analyse nine case studies from a previous special issue on Designing Transformative Spaces that aimed to collect examples of cutting-edge action-oriented research on transformations from the Global South. The analysis showed five design phases as being essential: Problem Definition Phase; Operationalisation Phase; Tactical Phase; Outcome Phase; and Reflection Phase. From this synthesis, we distilled five key messages that should be considered when designing research, including: (a) there are ethical dilemmas associated with creating a transformative space in a system; (b) it is important to assess the readiness of the system for change before engaging in it; (c) there is a need to balance between 'safe' and 'safe-enough' spaces for transformation; (d) convening a transformative space requires an assemblage of diverse methodological frameworks and tools; and (e) transformative spaces can act as a starting point for institutionalising transformative change. Many researchers are now engaging in transdisciplinary transformations research, and are finding themselves at the knowledge-action interface contributing to transformative space-making. We hope that by analysing experiences from across different geographies we can contribute towards better understanding of how to navigate the processes needed for the urgent global transformations that are being called for to create a more equitable and sustainable planet Earth.

    • Social-ecological systems (SES) research offers new theory and evidence to transform sustainable development to better contend with the challenges of the Anthropocene. Four insights from contemporary SES literature on (a) intertwined SES, (b) cross-scale dynamics, (c) systemic tipping points, and (d) transformational change are explored. Based on these insights, shifts in sustainable development practice are suggested to recognize and govern the complex and codeveloping social and ecological aspects of development challenges. The potential susceptibility of SES to nonlinear systemic reconfigurations is highlighted, as well as the opportunities, agency, and capacities required to foster reconfigurative transformations for sustainable development. SES research proposes the need for diverse values and beliefs that are more in tune with the deep, dynamic connections between social and ecological systems to transform development practice and to support capacities to deal with shocks and surprises. From these perspectives, SES research offers new outlooks, practices, and novel opportunity spaces from which to address the challenges of the Anthropocene.

    • We review the past decade’s widespread application of resilience science in sustainable development practice and examine whether and how resilience is reshaping this practice to better engage in complex contexts. We analyse six shifts in practice: from capitals to capacities, from objects to relations, from outcomes to processes, from closed to open systems, from generic interventions to context sensitivity, and from linear to complex causality. Innovative complexity-oriented practices have emerged, but dominant applications diverge substantially from the science, including its theoretical and methodological orientations. We highlight aspects of the six shifts that are proving challenging in practice and what is required from sustainability science.

Building on a review of existing theoretical and empirical research, we developed a theoretical framework for assessing three features identified as important to transformative potential of innovative social-ecological initiatives: (1) learning practices, (2) empowerment, and (3) networking. We applied this framework to a set of African-led and Africa-related initiatives that we selected from the SOGA database that were divided into initiatives with more or less transformative potential. We coded the presence or absence of features relating to the theoretical framework using secondary data, and then compared the initiatives using qualitative comparative analysis (QCA). This analysis revealed that of the three features tested, Networking emerged as the most important feature for transformative potential when compared amongst cases. By developing and testing a framework for the comparison of cases we provide a basis for future comparative work to further identify and test properties of cases that enable transformation.
