    • This document describes the technical layout and the expected performance of the Straw Tube Tracker (STT), the main tracking detector of the PANDA target spectrometer. The STT encloses a Micro-Vertex-Detector (MVD) for the inner tracking and is followed in beam direction by a set of GEM stations. The tasks of the STT are the measurement of the particle momentum from the reconstructed trajectory and the measurement of the specific energy loss for a particle identification. Dedicated simulations with full analysis studies of certain proton-antiproton reactions, identified as being benchmark tests for the whole PANDA scientific program, have been performed to test the STT layout and performance. The results are presented, and the time lines to construct the STT are described.

    • The structure of the Λ(1405) hyperon is an important and long-standing issue related to the K̄-nucleus interaction. The J-PARC E31 experiment has been performed to investigate the Λ(1405) spectrum shape. Because it is hard to form the Λ(1405) directly by a K̄N scattering in free space, E31 uses the d(K-, n) reaction with an incident kaon momentum of 1 GeV/c. We will identify three final states - ς-π+, ς+π-, ς0π0-so that the isospin structure of hyperon resonance states produced can be decomposed. The first physics run of the E31 experiment was performed in 2016. To enhance the statistics of the data set, we have performed the second physics run in the beginning of 2018. During the second run of E31, around 3.9×1010 kaons impacted on the deuteron target.

  • Structure near the K- + p + p threshold in the in-flight 3He(K-, Λp)n reaction

    • To search for an S = -1 di-baryonic state which decays toΛp, the 3He(K-,Λp)nmissing reaction was studied at 1.0 GeV/c. Unobserved neutrons were kinematically identified from the missing mass MX of the 3He (K-,Λp) X reaction in order to have a large acceptance for the Λpn final state. The observed Λpn events, distributed widely over the kinematically allowed region of the Dalitz plot, establish that the major component comes from a three-nucleon absorption process. A concentration of events at a specific neutron kinetic energy was observed in a region of low momentum transfer to the Λp. To account for the observed peak structure, the simplest S-wave polewas assumed to exist in the reaction channel, having a Breit-Wigner formin energy and with a Gaussian form factor. A minimum X2 method was applied to deduce its mass, MX = 2355+6 -8 (stat.) ±12 (syst.)MeV/c2, and decay width, γX = 110+19 -17 (stat.) ±27 (syst.)MeV/c2, respectively. The form factor parameter QX ∼ 400MeV/c implies that the range of the interaction is about 0.5 fm.

    • We have searched for the simplest kaonic nuclear state, K̄NN, using the in-flight 3He (K-, n) reaction at the J-PARC hadron experimental facility. In the semi-inclusive neutron missing-mass spectrum at θnlab=0°, an excess of yield was observed just below the K- pp mass-threshold, which cannot be explained by any elementary reactions [PTEP 2015, 061D01]. To understand the missing-mass spectrum of 3He (K-, n) X, we investigated the elementary (K-, n) reactions using hydrogen and deuterium targets. The p (K-, n) X missing-mass spectrum was well described by the charge-exchange reaction. However, in the d (K-, n) X spectrum, we observed an excess of yield just below the K- p mass-threshold, which was similar to that in the 3He (K-, n) X spectrum.

  • Siponimod versus placebo in secondary progressive multiple sclerosis (EXPAND): a double-blind, randomised, phase 3 study

    • © 2018 Elsevier Ltd Background: No treatment has consistently shown efficacy in slowing disability progression in patients with secondary progressive multiple sclerosis (SPMS). We assessed the effect of siponimod, a selective sphingosine 1-phosphate (S1P) receptor 1,5 modulator, on disability progression in patients with SPMS. Methods: This event-driven and exposure-driven, double-blind, phase 3 trial was done at 292 hospital clinics and specialised multiple sclerosis centres in 31 countries. Using interactive response technology to assign numbers linked to treatme nt arms, patients (age 18–60 years) with SPMS and an Expanded Disability Status Scale score of 3·0–6·5 were randomly assigned (2:1) to once daily oral siponimod 2 mg or placebo for up to 3 years or until the occurrence of a prespecified number of confirmed disability progression (CDP) events. The primary endpoint was time to 3-month CDP. Efficacy was assessed for the full analysis set (ie, all randomly assigned and treated patients); safety was assessed for the safety set. This trial is registered with ClinicalTrials.gov, number NCT01665144. Findings: 1651 patients were randomly assigned between Feb 5, 2013, and June 2, 2015 (1105 to the siponimod group, and 546 to the placebo group). One patient did not sign the consent form, and five patients did not receive study drug, all of whom were in the siponimod group. 1645 patients were included in the analyses (1099 in the siponimod group and 546 in the placebo). At baseline, the mean time since first multiple sclerosis symptoms was 16·8 years (SD 8·3), and the mean time since conversion to SPMS was 3·8 years (SD 3·5); 1055 (64%) patients had not relapsed in the previous 2 years, and 918 (56%) of 1651 needed walking assistance. 903 (82%) patients receiving siponimod and 424 (78%) patients receiving placebo completed the study. 288 (26%) of 1096 patients receiving siponimod and 173 (32%) of 545 patients receiving placebo had 3-month CDP (hazard ratio 0·79, 95% CI 0·65–0·95; relative risk reduction 21%; p=0·013). Adverse events occurred in 975 (89%) of 1099 patients receiving siponimod versus 445 (82%) of 546 patients receiving placebo; serious adverse events were reported for 197 (18%) patients in the siponimod group versus 83 (15%) patients in the placebo group. Lymphopenia, increased liver transaminase concentration, bradycardia and bradyarrhythmia at treatment initiation, macular oedema, hypertension, varicella zoster reactivation, and convulsions occurred more frequently with siponimod than with placebo. Initial dose titration mitigated cardiac first-dose effects. Frequencies of infections, malignancies, and fatalities did not differ between groups. Interpretation: Siponimod reduced the risk of disability progression with a safety profile similar to that of other S1P modulators and is likely to be a useful treatment for SPMS.     • Crystallographic structures of NGF/p75NTR and proNGF/p75NTR were previously obtained in 2:1 and 2:2 stoichiometries, respectively. However, evidence shows that both stoichiometries can occur for mature neurotrophins and proneurotrophins. We used Molecular Dynamics (MD) simulations to examine the energetic and structural characteristics of these two complete systems as well as the uncomplexed forms of NGF and understand how these could translate in a new view of different biological outcomes. Here, we show that one chain at the 2:2 proNGF complex seems to be preferentially lost creating a 2:1 structure able to interact with sortilin. We also demonstrated that the structure of the neurotrophin dimers is not pre-established and suffers large structural modifications upon p75NTR binding. Moreover, our data suggests an elegant explanation for the dual role of NGF in neuronal cell death and survival, where different stoichiometries induce conformational changes that might be the basis for the different biological outcomes observed with the mature and proforms of neurotrophins.

    • The present work considers control of perturbations in the boundary layer over a flat plate by means of adaptive methods. In particular, we focus our attention on a control law based on a multi-input-multi-output (MIMO) filtered-x least-mean-square (fxLMS) adaptive algorithm. The studies are performed through direct numerical simulations. The perturbation field studied here mimics those generated by freestream turbulence with different amplitude and scales. Plasma actuators and shear-stress sensors are considered to mimic a real case scenario.

    • In resolvent analyses of turbulent channel flows it has been common practice to neglect or model the nonlinear forcing term that forms the input of the resolvent. However, the spatiotemporal structure of this term is mostly unknown. Here, this nonlinear forcing term is quantified. The Fourier transform of its two-point space-time correlation, its cross-spectral density (CSD), is computed. The CSD is evaluated for two channel flows at friction Reynolds numbers Re-tau = 179 and Re-tau = 543 via direct numerical simulations (DNS). The CSDs are computed for energetic structures typical of buffer-layer and large-scale motions, for different temporal frequencies. It is found that the forcing is structured and that its solenoidal part, which is the only one affecting the velocity field, is the combination of an oblique streamwise vortical forcing and a streamwise component that counteract each other, as in a destructive interference. It is shown that a rank-2 approximation of the forcing, with only the most energetic spectral proper orthogonal decomposition (SPOD) modes, leads to the bulk of the response. Moreover, it is found that the nonlinear forcing term has a non-negligible projection onto the linear sub-optimal forcings of resolvent analysis, which demonstrates that the linear optimal forcing is not representative of the nonlinear forcing. Finally, it is clarified that the Cess eddy-viscosity-modelled forcing improves the accuracy of resolvent analysis prediction because the modelled forcing projects onto the linear sub-optimal forcings similarly to DNS data.

    • An analysis of the statistics of the nonlinear terms in resolvent analysis is performed in this work for turbulent Couette flow at Reynolds number 400. Data from a direct numerical simulation of a minimal flow unit is used to compute the covariance matrix of the velocity. From the same data, we computed the nonlinear terms of the Navier-Stokes equations (treated as forcing), which allowed us to compute the covariance matrix of the forcing. The quantitative relation between the two covariances via the resolvent operator is confirmed here for the first time, accounting for relevant signal processing issues related to the windowing procedure for frequency-domain quantities. Such exact correspondence allowed the eduction of the most relevant force components for the dominant structures in this flow, which participate in the self-sustaining cycle of turbulence: (i) streamwise vortices and streaks, and (ii) spanwise-coherent fluctuations of spanwise velocity. The results show a dominance by a subset of the nonlinear terms for the prediction of the full statistics of streamwise vortices and streaks; a single term is seen to be dominant for spanwise motions. A relevant feature observed in these cases is that the forcing covariance is dominated by its first eigenfunction, showing that nonlinear terms also have a coherent structure at low frequencies in this flow. Different forcing components are also coherent between them, which leads to constructive and destructive interferences that greatly modify the flow response. These are key features of forcing 'colour' for the present flow.

  • Mammographic density : Comparison of visual assessment with fully automatic calculation on a multivendor dataset

    • To compare breast density (BD) assessment provided by an automated BD evaluator (ABDE) with that provided by a panel of experienced breast radiologists, on a multivendor dataset. Twenty-one radiologists assessed 613 screening/diagnostic digital mammograms from nine centers and six different vendors, using the BI-RADS a, b, c, and d density classification. The same mammograms were also evaluated by an ABDE providing the ratio between fibroglandular and total breast area on a continuous scale and, automatically, the BI-RADS score. A panel majority report (PMR) was used as reference standard. Agreement (kappa) and accuracy (proportion of cases correctly classified) were calculated for binary (BI-RADS a-b versus c-d) and 4-class classification. While the agreement of individual radiologists with the PMR ranged from kappa = 0.483 to kappa = 0.885, the ABDE correctly classified 563/613 mammograms (92 %). A substantial agreement for binary classification was found for individual reader pairs (kappa = 0.620, standard deviation [SD] = 0.140), individual versus PMR (kappa = 0.736, SD = 0.117), and individual versus ABDE (kappa = 0.674, SD = 0.095). Agreement between ABDE and PMR was almost perfect (kappa = 0.831). The ABDE showed an almost perfect agreement with a 21-radiologist panel in binary BD classification on a multivendor dataset, earning a chance as a reproducible alternative to visual evaluation. aEuro cent Individual BD assessment differs from PMR with kappa as low as 0.483. aEuro cent An ABDE correctly classified 92 % of mammograms with almost perfect agreement (kappa = 0.831). aEuro cent An ABDE can be a valid alternative to subjective BD assessment.

  • On the wave-cancelling nature of boundary layer transition control

    • This work deals with the feedforward active control of velocity fluctuations over incompressible 3D boundary layers. Two strategies are evaluated, the Linear Quadratic Gaussian (LQG) controller, built using the eigensystem realization algorithm (ERA), is compared to a wave-cancellation scheme, obtained via the direct inversion of the frequency-domain transfer functions of the system. For the evaluated cases, it is shown that LQG leads to a wave-cancelling signal of the incoming Tollmien-Schlichting wavepacket. Such result allows further insight into the physics behind the active control of convectively unstable flows permitting, for instance, the optimization of the transverse position for actuation via a linear stability approach.
