  • The Miami International Evidence-based Guidelines on Minimally Invasive Pancreas Resection

    • Objective: The aim of this study was to develop and externally validate the first evidence-based guidelines on minimally invasive pancreas resection (MIPR) before and during the International Evidence-based Guidelines on Minimally Invasive Pancreas Resection (IG-MIPR) meeting in Miami (March 2019).Summary Background Data: MIPR has seen rapid development in the past decade. Promising outcomes have been reported by early adopters from high-volume centers. Subsequently, multicenter series as well as randomized controlled trials were reported; however, guidelines for clinical practice were lacking. Methods: The Scottisch Intercollegiate Guidelines Network (SIGN) methodology was used, incorporating these 4 items: systematic reviews using PubMed, Embase, and Cochrane databases to answer clinical questions, whenever possible in PICO style, the GRADE approach for assessment of the quality of evidence, the Delphi method for establishing consensus on the developed recommendations, and the AGREE-II instrument for the assessment of guideline quality and external validation. The current guidelines are cosponsored by the International Hepato-Pancreato-Biliary Association, the Americas Hepato-Pancreato-Biliary Association, the Asian-Pacific Hepato-Pancreato-Biliary Association, the European-African Hepato-Pancreato-Biliary Association, the European Association for Endoscopic Surgery, Pancreas Club, the Society of American Gastrointestinal and Endoscopic Surgery, the Society for Surgery of the Alimentary Tract, and the Society of Surgical Oncology. Results: After screening 16,069 titles, 694 studies were reviewed, and 291 were included. The final 28 recommendations covered 6 topics; laparoscopic and robotic distal pancreatectomy, central pancreatectomy, pancreatoduodenectomy, as well as patient selection, training, learning curve, and minimal annual center volume required to obtain optimal outcomes and patient safety.Conclusion: The IG-MIPR using SIGN methodology give guidance to surgeons, hospital administrators, patients, and medical societies on the use and outcome of MIPR as well as the approach to be taken regarding this challenging type of surgery. © 2019 Wolters Kluwer Health, Inc.     • The next years will see the completion of the radioactive ion beam facility SPES (Selective Production of Exotic Species) and the upgrade of the accelerators complex at Istituto Nazionale di Fisica Nucleare – Legnaro National Laboratories (LNL) opening up new possibilities in the fields of nuclear structure, nuclear dynamics, nuclear astrophysics, and applications. The nuclear physics community has organised a workshop to discuss the new physics opportunities that will be possible in the near future by employing state-of-the-art detection systems. A detailed discussion of the outcome from the workshop is presented in this report.

  • W-band high-gain low profile circularly polarized magneto-electric dipole antenna array with gap waveguide feeding technology

    • In this paper, a high gain circularly polarized (CP) aperture-coupled magneto-electric (ME) dipole antenna array excited by the air cavity in gap waveguide (GWG) feeding technology in W-band is proposed. This antenna array is composed of two layers, upper CP ME-dipole antenna subarrays and lower metallic GWG feeding networks. The planar antenna array structure is realized by the high mode air cavity feeding network, which is realized by extending the 1×2 cavity to 3×2 cavity using more shorted pins to split the electromagnetic field. The whole single-layer GWG feeding network is composed of a ridge gap waveguide (RGWG) transmission structure and short-ended vertical polarized groove gap waveguide (VP-GGWG) high mode air cavities. The transmission loss caused by the coupling and the fabrication tolerance can be reduced by combining the RGWG and GGWG in the same layer. In addition, the feeding network is fabricated all in metal with no substrate loss. Transverse slots are etched at the right position of the printed circuit board (PCB) backside to excite the CP ME-dipoles. The antenna element has stable radiation patterns and wide axial-ratio (AR) bandwidth. The proposed 3×8 planner antenna array has a maximum boresight gain of 22 dBi and an AR bandwidth of 10.8% for AR < 3 dB. In the frequency band from 87.7 to 97.7 GHz, a boresight gain above 20 dBi is achieved.

  • The transcription factor TCFL5 responds to A-MYB to elaborate the male meiotic program in mice

    • In male mice, the transcription factors STRA8 and MEISON initiate meiosis I. We report that STRA8/MEISON activates the transcription factors A-MYB and TCFL5, which together reprogram gene expression after spermatogonia enter into meiosis. TCFL5 promotes the transcription of genes required for meiosis, mRNA turnover, miR-34/449 production, meiotic exit, and spermiogenesis. This transcriptional architecture is conserved in rhesus macaque, suggesting TCFL5 plays a central role in meiosis and spermiogenesis in placental mammals. Tcfl5(em1/em1) mutants are sterile, and spermatogenesis arrests at the mid- or late-pachytene stage of meiosis. Moreover, Tcfl5(+/em1) mutants produce fewer motile sperm.

    • The paper introduces a novel focal plane array (FPA) design aimed at enhancing the reliability of millimeter-wave backhaul communication links by compensating for the impact of antenna mounting structures' sways. This FPA is analyzed for an offset parabolic reflector antenna with the 40-cm aperture diameter at W-band frequencies (92-96 GHz). The FPA-fed reflector antenna generates multiple closely-overlapping beams to compensate for the expected antenna tilt range, equivalent to seven beamwidths. This study addresses major challenges associated with FPA design implementation at these high frequencies, including (i) the need for a small (≤ 0.75 λ) inter-element spacing for the required beam overlap, and (ii) manufacturing complexities of an array element ensuring optimal reflector illumination and impedance matching in the presence of mutual coupling effects. An analytical approach to the FPA pattern shaping for optimal reflector illumination is proposed and successfully validated via numerical optimization and measurements. Furthermore, our analysis of the total reflector-FPA system shows promising results, i.e. 56 - 68% antenna aperture efficiency (49 - 50 dBi directivity) for all beams with compliance to the radiation emission constraints (ETSI class 3) within ±89°.

  • Identifying influential nodes by leveraging redundant ties

    • Structure-based influential nodes identification is a long-term challenge in the study of complex networks. While global centrality-based approaches are generally considered to be more accurate and reliable, the requirements of complete network information and high computational complexity are hard to meet, limiting their applications in many practical scenarios. In addition, recent studies have highlighted the effect of cyclic structures introducing redundant paths in network connectivity and exaggerating the importance of traditional centrality measures. In this work, we develop a new centrality metric, called Multi-Spanning Tree-based Degree Centrality (MSTDC), to quantify node importance with linear complexity by leveraging redundant ties. MSTDC is calculated using the aggregation of degrees of a small number of spanning trees constructed with a few randomly selected root nodes. Experiments on synthetic and empirical networks reveal that MSTDC obtains superior performance than other benchmark network centralities in identifying influential nodes from the perspective of both maintaining network connectivity and maximizing spreading capacity. In addition, we find that MSTDC is extraordinarily effective in networks with high clustering coefficients. Our study provides novel insights into the role of redundant ties in network structural and functional analyses.

  • Quantification of mutant-allele expression at isoform level in cancer from RNA-seq data

    • Even though the role of DNA mutations in cancer is well recognized, current quantification of the RNA expression, performed either at gene or isoform level, typically ignores the mutation status. Standard methods for estimating allele-specific expression (ASE) consider gene-level expression, but the functional impact of a mutation is best assessed at isoform level. Hence our goal is to quantify the mutant–allele expression at isoform level. We have developed and implemented a method, named MAX, for quantifying mutant–allele expression given a list of mutations. For a gene of interest, a mutant reference is constructed by incorporating all possible mutant versions of the wild-type isoforms in the transcriptome annotation. The mutant reference is then used for the RNA-seq reads mapping, which in principle works similarly for any quantification tool. We apply an alternating EM algorithm to the read-count data from the mapping step. In a simulation study, MAX performs well against standard isoform-quantification methods. Also, MAX achieves higher accuracy than conventional gene-based ASE methods such as ASEP. An analysis of a real dataset of acute myeloid leukemia reveals a subgroup of NPM1-mutated patients responding well to a kinase inhibitor. Our findings indicate that quantification of mutant–allele expression at isoform level is feasible and has potential added values for assessing the functional impact of DNA mutations in cancers.

    • A novel high-frequency fiber Bragg grating (FBG) sensing interrogation system by using fiber Sagnac-loop-based microwave photonic filtering is proposed and experimentally demonstrated. By adopting the microwave photonic filtering, the wavelength shift of sensing FBG can be converted into amplitude variation of the modulated electronic radio-frequency (RF) signal. In the experiment, the strain applied onto the sensing FBG has been demodulated by measuring the intensity of the recovered RF signal, and by modulating the RF signal with different frequencies, different interrogation sensitivities can be achieved.

    • Microwave signal generation by using the photonic beating from a phase-shift fiber Bragg grating (PS-FBG)-based dual-wavelength laser is proposed and experimentally demonstrated. The dual-wavelength laser is formed by a linear cavity, in which a PS-FBG is used as a dual-wavelength selective component. Transversal loading on the PS-FBG enhances the birefringence of the optical fiber and consequently makes the transmission peak of the PS-FBG splitting into two sharp transmission peaks of orthogonal polarizations. The wavelength spacing between the two transmission peaks increases with the transversal loading on the PS-FBG, thus making the polarization beating frequency increase. This property is exploited in a transversal loading sensor.

    • In this work, a microchanneled chirped fiber Bragg grating (MCFBG) is proposed and fabricated through the femtosecond laser-assisted chemical etching. The microchannel (similar to 550 mu m) gives access to the external index liquid, thus inducing refractive index (RI) sensitivity to the structure. In the experiment, the transmission bands induced by the reduced effective index in the microchannel region were used to sense the surrounding RI and temperature changes. The experimental results show good agreement with the theoretical analysis. The proposed MCFBG offers enhanced RI sensitivity without degrading the robustness of the device showing good application potential as bio-chemical sensors.

    • Climatic condition is triggering human health emergencies and earth’s surface changes. Anthropogenic activities, such as built-up expansion, transportation development, industrial works, and some extreme phases, are the main reason for climate change and global warming. Air pollutants are increased gradually due to anthropogenic activities and triggering the earth’s health. Nitrogen Dioxide (NO2), Carbon Monoxide (CO), and Aerosol Optical Depth (AOD) are truthfully important for air quality measurement because those air pollutants are more harmful to the environment and human’s health. Earth observational Sentinel-5P is applied for monitoring the air pollutant and chemical conditions in the atmosphere from 2018 to 2021. The cloud computing-based Google Earth Engine (GEE) platform is applied for monitoring those air pollutants and chemical components in the atmosphere. The NO2 variation indicates high during the time because of the anthropogenic activities. Carbon Monoxide (CO) is also located high between two 1-month different maps. The 2020 and 2021 results indicate AQI change is high where 2018 and 2019 indicates low AQI throughout the year. The Kolkata have seven AQI monitoring station where high nitrogen dioxide recorded 102 (2018), 48 (2019), 26 (2020) and 98 (2021), where Delhi AQI stations recorded 99 (2018), 49 (2019), 37 (2020), and 107 (2021). Delhi, Kolkata, Mumbai, Pune, and Chennai recorded huge fluctuations of air pollutants during the study periods, where ~ 50–60% NO2 was recorded as high in the recent time. The AOD was noticed high in Uttar Pradesh in 2020. These results indicate that air pollutant investigation is much necessary for future planning and management otherwise; our planet earth is mostly affected by the anthropogenic and climatic conditions where maybe life does not exist.

    • Traumatic brain injury is a leading cause of acquired epilepsy. Initially described in 1989, lateral fluid percussion injury (LFPI) has since become the most extensively used and well-characterized rodent traumatic brain injury and post-traumatic epilepsy model. Universal findings, particularly seizures that reliably develop after an initial latent period, are evident across studies from multiple laboratories. However, the LFPI procedure is a two-stage process, requiring initial surgical attachment of a skull fluid cannula and then reanesthesia for delivery of the epidural fluid pressure wave. We now describe a modification of the original technique, termed 'rapid lateral fluid percussion injury' (rLFPI), which allows for a one-stage procedure and thus shorter operating time and reduced anesthesia exposure. Anesthetized male Long-Evans rats were subjected to rLFPI through a length of plastic tubing fitted with a pipette tip cannula with a 4-mm aperture. The cannula opening was positioned over a craniectomy of slightly smaller diameter and exposed dura such that the edges of the cannula fit tightly when pressed to the skull with a micromanipulator. Fluid percussion was then delivered immediately thereafter, in the same surgery session. rLFPI resulted in nonlethal focal cortical injury in all animals. We previously demonstrated that the rLFPI procedure resulted in post-traumatic seizures and regional gliosis, but had not examined other histopathologic elements. Now, we show apoptotic cell death confined to the perilesional cortex and chronic pathologic changes such as ipsilesional ventriculomegaly that are seen in the classic model. We conclude that the rLFPI method is a viable alternative to classic LFPI, and - being a one-stage procedure - has the advantage of shorter experiment turnaround and reduced exposure to anesthetics.

  • Interaction between AIF and CHCHD4 Regulates Respiratory Chain Biogenesis

    • Apoptosis-inducing factor (AIF) is a mitochondrial flavoprotein that, beyond its apoptotic function, is required for the normal expression of major respiratory chain complexes. Here we identified an AIF-interacting protein, CHCHD4, which is the central component of a redox-sensitive mitochondrial intermembrane space import machinery. Depletion or hypomorphic mutation of AIF caused a downregulation of CHCHD4 protein by diminishing its mitochondrial import. CHCHD4 depletion sufficed to induce a respiratory defect that mimicked that observed in AIF-deficient cells. CHCHD4 levels could be restored in AIF-deficient cells by enforcing its AIF-independent mitochondrial localization. This modified CHCHD4 protein reestablished respiratory function in AIF-deficient cells and enabled AIF-deficient embryoid bodies to undergo cavitation, a process of programmed cell death required for embryonic morphogenesis. These findings explain how AIF contributes to the biogenesis of respiratory chain complexes, and they establish an unexpected link between the vital function of AIF and the propensity of cells to undergo apoptosis.

    • The cryosphere (including, snow, glaciers, permafrost, lake and river ice) is an integral element of high- mountain regions, which are home to roughly 10% of the global population. Widespread cryosphere changes affect physical, biological and human systems in the mountains and surrounding lowlands, with impacts evident even in the ocean. Building on the IPCC’s Fifth Assessment Report (AR5), this chapter assesses new evidence on observed recent and projected changes in the mountain cryosphere as well as associated impacts, risks and adaptation measures related to natural and human systems. Impacts in response to climate changes independently of changes in the cryosphere are not assessed in this chapter. Polar mountains are included in Chapter 3, except those in Alaska and adjacent Yukon, Iceland, and Scandinavia, which are included in this chapter.

    • n this study, a series of Bi1.5Sb0.5Te 1.7Se1.3 (BSTS) flakes 80-nm to 140-μm in thickness was fabricated to investigate their metallic surface states. We report the observation of surface-dominated transport in these topological insulator BSTS nanoflakes. The achievement of surface-dominated transport can be attributed to high surface mobility (∼3000 cm2/V s) and low bulk mobility (12 cm2/V s). Up to 90% of the total conductance, the surface channel was estimated based on the thickness dependence of electrical conductance and the result of the Shubnikov-de Hass oscillations in a 200-nm BSTS. The nature of nontrivial Dirac surface states was also confirmed by the weak anti-localization effect.

    • A major cause of tumor treatment failure is cancer cell metastasis. Toll-like receptor 4 (TLR4)-mediated signaling has been implicated in tumor cell invasion, survival, and metastasis in a variety of cancers. In this study, we investigated the biological roles of TLR4 in prostate metastatic cell invasion and survival, and the potential of gene silencing of TLR4 using small interfering RNA (siRNA) for treatment of cancer. In cultured human prostate cancer cell lines, TLR4 were higher PC3 and DU145 as compared with the poorly metastatic LNCaP indicating that up-regulation of TLR4 was positively correlated with metastasis of tumor cell. In the highly metastatic cancer cell PC3, gene silencing of TLR4 using siRNA significantly inhibited TLR4 mRNA expression and protein level. Knockdown of TLR4 in PC3 cells resulted in a dramatic reduction of tumor cell migration and invasion as indicated by a Matrigel invasion assay. Furthermore, TLR4 siRNA suppressed cell viability and ultimately caused the induction of apoptotic cell death. The effects were associated with abrogating TLR4-mediated signaling to downstream target molecules such as myeloid differentiation factor 88 (MyD88), adaptor-inducing IFN-beta (TRIF), and interferon regulatory factor-1 (IRF-1). In a mouse prostate cancer model, administration with the plasmid construct expressing siRNA for TLR4 obviously inhibited established tumor growth and survival. These studies revealed evidence of a multifaceted signaling network operating downstream of TLR4-mediated tumor cell invasion, proliferation, and survival. Thus, RNA interference-directed targeting of TLR4 may raise the potential of its application for cancer therapy.

    • Using high-resolution extreme ultraviolet resonant inelastic x-ray scattering (EUVRIXS) spectroscopy at the Cu M-edge, we observed doping dependent spectral shifts of interorbital (dd) excitations of YBa2Cu3O7-y and La2-xSrxCuO4. With increasing the hole doping level from undoped to optimally doped superconducting compositions, the leading edge of dd excitations is found to shift towards lower energy loss in a roughly linear trend that is irrespective of the cuprate species. The magnitude of the energy shift can be explained by including a 0.15 eV Coulomb attraction between Cu3d(x2-y2) electrons and the doped holes on the surrounding oxygens in the atomic multiplet calculations. The consistent energy shift between distinct cuprate families suggests that this intersite Coulomb interaction energy scale is relatively material independent, and provides an important reference point for understanding charge density wave phenomena in the cuprate phase diagram.

    • Trans-endothelial electrical resistance (TEER) is one of the most widely used indicators to quantify the barrier integrity of endothelial layers. Over the last decade, the integration of TEER sensors into organ-on-a-chip (OOC) platforms has gained increasing interest for its efficient and effective measurement of TEER in OOCs. To date, microfabricated electrodes or direct insertion of wires has been used to integrate TEER sensors into OOCs, with each method having advantages and disadvantages. In this study, we developed a TEER-SPE chip consisting of carbon-based screen-printed electrodes (SPEs) embedded in a poly(methyl methacrylate) (PMMA)-based multi-layered microfluidic device with a porous poly(ethylene terephthalate) membrane in-between. As proof of concept, we demonstrated the successful cultures of hCMEC/D3 cells and the formation of confluent monolayers in the TEER-SPE chip and obtained TEER measurements for 4 days. Additionally, the TEER-SPE chip could detect changes in the barrier integrity due to shear stress or an inflammatory cytokine (i.e., tumor necrosis factor-& alpha;). The novel approach enables a low-cost and facile fabrication of carbon-based SPEs on PMMA substrates and the subsequent assembly of PMMA layers for rapid prototyping. Being cost-effective and cleanroom-free, our method lowers the existing logistical and technical barriers presenting itself as another step forward to the broader adoption of OOCs with TEER measurement capability.

    • We investigate the properties of the dipole frequency selective surfaces-loaded rectangular waveguide. The analysis is carried out by using the method of moments together with an algorithm that solves for a multilayer structure in the spectral domain. The eigenmodes are determined by searching for resonant solutions. Dispersion diagrams obtained from this approach are compared with those generated by commercial software simulations. Experimental validation is also performed. © 2006 IEEE.

    • The present paper will give an overview of the research on hard surfaces and hard waveguides leading to the wideband ridge gap waveguide. The overview will be supported by both computed and measured results of the performance of hard waveguides and gap waveguides, clearly showing the wide bandwidth and low losses of the latter. This is a major improvement compared to the original hard waveguides, enabled by the wideband parallel-plate cut-off fenomena used for the gap waveguides.

    • In this paper, we design a 60 GHz millimeter-wave gap waveguide antenna for beam steering applications. Implemented on the premise of a waveguide slot-array, radiated beams can be shone towards designated directions by sliding the pin lattice that makes up one of the walls, thereby altering the modal resonance and thus the phase pattern across the radiating aperture. The array consists of six staggered longitudinal slots. Designs by simulations indicate the ability to steer the main beam through 10 degrees in the elevation plane. The simulated realized gain is 12 dB and the bandwidth is from 54 to 62 GHz (13% fractional bandwidth). The main advantages of this antenna are that it can tailor the main beam with one simple mechanical motion, thereby circumventing the need for complicated electronic networks such as phase shifters, switches or diodes, thus lowering the costs and complexities.

    • In bioethanol production, the main by-product, 5-hydroxymethylfurfural (HMF), significantly hinders microbial fermentation. Therefore, it is crucial to explore genes related to HMF tolerance in Saccharomyces cerevisiae for enhancing the tolerance of ethanol fermentation strains. A comprehensive analysis was conducted using genome-wide deletion library scanning and SGAtools, resulting in the identification of 294 genes associated with HMF tolerance in S. cerevisiae. Further KEGG and GO enrichment analysis revealed the involvement of genes OCA1 and SIW14 in the protein phosphorylation pathway, underscoring their role in HMF tolerance. Spot test validation and subcellular structure observation demonstrated that, following a 3-h treatment with 60mM HMF, the SIW14 gene knockout strain exhibited a 12.68% increase in cells with abnormal endoplasmic reticulum (ER) and a 22.41% increase in the accumulation of reactive oxygen species compared to the BY4741 strain. These findings indicate that the SIW14 gene contributes to the protection of the ER structure within the cell and facilitates the clearance of reactive oxygen species, thereby confirming its significance as a key gene for HMF tolerance in S. cerevisiae.

  • Mechanistic Investigation of Dimethylmercury Formation Mediated by a Sulfide Mineral Surface

    • Mercury (Hg) pollution is a global environmental problem. The abiotic formation of dimethylmercury (DMeHg) from monomethylmercury (MMeHg) may account for a large portion of DMeHg in oceans. Previous experimental work has shown that abiotic formation of DMeHg from MMeHg can be facilitated by reduced sulfur groups on sulfide mineral surfaces. In that work, a mechanism was proposed in which neighboring MMeHg moieties bound to sulfide sites on a mineral surface react through an SN2-type mechanism to form DMeHg and incorporate the remaining Hg atoms into the mineral surface. Here, we perform density functional theory calculations to explore the mechanisms of DMeHg formation on the 110 surface of a CdS(s) (hawleyite) nanoparticle. We show that coordination of MMeHg substituents to adjacent reduced sulfur groups protruding from the surface indeed facilitates DMeHg formation and that the reaction proceeds through direct transmethylation from one MMeHg substituent to another. Coordination of Hg by multiple S atoms provides a transition-state stabilization and activates a C–Hg bond for methyl transfer. In addition, solvation effects play an important role in the surface reconstruction of the nanoparticle and in decreasing the energetic barrier for DMeHg formation relative to the corresponding reaction in vacuo.

    • Engineering Saccharomyces cerevisiae for biodegradation and transformation of industrial toxic substances such as catechol (CA) has received widespread attention, but the low tolerance of S. cerevisiae to CA has limited its development. The exploration and modification of genes or pathways related to CA tolerance in S. cerevisiae is an effective way to further improve the utilization efficiency of CA. This study identified 36 genes associated with CA tolerance in S. cerevisiae through genome-wide identification and bioinformatics analysis and the ERG6 knockout strain (ERG6 Delta) is the most sensitive to CA. Based on the omics analysis of ERG6 Delta under CA stress, it was found that ERG6 knockout affects pathways such as intrinsic component of membrane and pentose phosphate pathway. In addition, the study revealed that 29 genes related to the cell wall-membrane system were up-regulated by more than twice, NADPH and NADP(+) were increased by 2.48 and 4.41 times respectively, and spermidine and spermine were increased by 2.85 and 2.14 times, respectively, in ERG6 Delta. Overall, the response of cell wall-membrane system, the accumulation of spermidine and NADPH, as well as the increased levels of metabolites in pentose phosphate pathway are important findings in improving the CA resistance. This study provides a theoretical basis for improving the tolerance of strains to CA and reducing the damage caused by CA to the ecological environment and human health.

    • The hypoxia-inducible factors (HIFs) regulate the main transcriptional pathway of response to hypoxia in T cells and are negatively regulated by von Hippel-Lindau factor (VHL). But the role of HIFs in the regulation of CD4 T cell responses during infection withM. tuberculosisisn’t well understood. Here we show that mice lacking VHL in T cells (Vhl cKO) are highly susceptible to infection withM. tuberculosis, which is associated with a low accumulation of mycobacteria-specific T cells in the lungs that display reduced proliferation, altered differentiation and enhanced expression of inhibitory receptors. In contrast, HIF-1 deficiency in T cells is redundant forM. tuberculosiscontrol.Vhl cKOmice also show reduced responses to vaccination. Further, VHL promotes proper MYC-activation, cell-growth responses, DNA synthesis, proliferation and survival of CD4 T cells after TCR activation. The VHL-deficient T cell responses are rescued by the loss of HIF-1α, indicating that the increased susceptibility toM. tuberculosisinfection and the impaired responses ofVhl-deficient T cells are HIF-1-dependent.

    • The activity of ADAMTS 13, the von Willebrand factor cleaving protease, is deficient in patients with thrombotic thrombocytopenic purpura (TTP). In the present study, the phenotype of ADAMTS13 in TTP and in normal plasma was demonstrated by immunoblotting. Normal plasma (n=20) revealed a single band at 190 kD under reducing conditions using a polyclonal antibody, and a single band at 150 kD under non-reducing conditions using a monoclonal antibody. ADAMTS 13 was not detected in the plasma from patients with congenital TTP (n=5) by either antibody, whereas patients with acquired TTP (n=2) presented the normal phenotype. Following immunoadsorption of immunoglobulins, the ADAMTS 13 band was removed from the plasma of the patients with acquired TTP, but not from that of normal individuals. This indicates that ADAMTS13 is complexed with immunoglobulin in these patients. The lack of ADAMTS13 expression in the plasma from patients with hereditary TTP may indicate defective synthesis, impaired cellular secretion, or enhanced degradation in the circulation. This study differentiated between normal and TTP plasma, as well as between congenital and acquired TTP. This method may, therefore, be used as a complement in the diagnosis of TTP.

    • CRISPR/Cas9-driven cancer modeling studies are based on the disruption of tumor suppressor genes by small insertions or deletions (indels) that lead to frame-shift mutations. In addition, CRISPR/Cas9 is widely used to define the significance of cancer oncogenes and genetic dependencies in loss-of-function studies. However, how CRISPR/Cas9 influences gain-of-function oncogenic mutations is elusive. Here, we demonstrate that single guide RNA targeting exon 3 of Ctnnb1 (encoding β-catenin) results in exon skipping and generates gain-of-function isoforms in vivo. CRISPR/Cas9-mediated exon skipping of Ctnnb1 induces liver tumor formation in synergy with YAPS127A in mice. We define two distinct exon skipping-induced tumor subtypes with different histological and transcriptional features. Notably, ectopic expression of two exon-skipped β-catenin transcript isoforms together with YAPS127A phenocopies the two distinct subtypes of liver cancer. Moreover, we identify similar CTNNB1 exon-skipping events in patients with hepatocellular carcinoma. Collectively, our findings advance our understanding of β-catenin-related tumorigenesis and reveal that CRISPR/Cas9 can be repurposed, in vivo, to study gain-of-function mutations of oncogenes in cancer.

  • Temporal Characteristics of the Chinese Aviation Network and their Effects on the Spread of Infectious Diseases

    • Aviation transportation systems have developed rapidly in recent years and have become a focus for research on the modeling of epidemics. However, despite the number of studies on aggregated topological structures and their effects on the spread of disease, the temporal sequence of flights that connect different airports have not been examined. In this study, to analyze the temporal pattern of the Chinese Aviation Network (CAN), we obtain a time series of topological statistics through sliding the temporal CAN with an hourly time window. In addition, we build two types of Susceptible-Infectious (SI) spreading models to study the effects of linking sequence and temporal duration on the spread of diseases. The results reveal that the absence of links formed by flights without alternatives at dawn and night causes a significant decrease in the centralization of the network. The temporal sparsity of linking sequence slows down the spread of disease on CAN, and the duration of flights intensifies the sensitiveness of CAN to targeted infection. The results are of great significance for further understanding of the aviation network and the dynamic process, such as the propagation of delay.

  • A Comprehensive Landscape of Imaging Feature-Associated RNA Expression Profiles in Human Breast Tissue

    • The expression abundance of transcripts in nondiseased breast tissue varies among individuals. The association study of genotypes and imaging phenotypes may help us to understand this individual variation. Since existing reports mainly focus on tumors or lesion areas, the heterogeneity of pathological image features and their correlations with RNA expression profiles for nondiseased tissue are not clear. The aim of this study is to discover the association between the nucleus features and the transcriptome-wide RNAs. We analyzed both microscopic histology images and RNA-sequencing data of 456 breast tissues from the Genotype-Tissue Expression (GTEx) project and constructed an automatic computational framework. We classified all samples into four clusters based on their nucleus morphological features and discovered feature-specific gene sets. The biological pathway analysis was performed on each gene set. The proposed framework evaluates the morphological characteristics of the cell nucleus quantitatively and identifies the associated genes. We found image features that capture population variation in breast tissue associated with RNA expressions, suggesting that the variation in expression pattern affects population variation in the morphological traits of breast tissue. This study provides a comprehensive transcriptome-wide view of imaging-feature-specific RNA expression for healthy breast tissue. Such a framework could also be used for understanding the connection between RNA expression and morphology in other tissues and organs. Pathway analysis indicated that the gene sets we identified were involved in specific biological processes, such as immune processes.

    • Molecular classification of acute myeloid leukemia (AML) aids prognostic stratification and clinical management. Our aim in this study is to identify transcriptome-wide mRNAs that are specific to each of the molecular subtypes of AML. We analyzed RNA-sequencing data of 955 AML samples from three cohorts, including the BeatAML project, the Cancer Genome Atlas, and a cohort of Swedish patients to provide a comprehensive transcriptome-wide view of subtype-specific mRNA expression. We identified 729 subtype-specific mRNAs, discovered in the BeatAML project and validated in the other two cohorts. Using unique proteomics data, we also validated the presence of subtype-specific mRNAs at the protein level, yielding a rich collection of potential protein-based biomarkers for the AML community. To enable the exploration of subtype-specific mRNA expression by the broader scientific community, we provide an interactive resource to the public.

    • Safe teleoperation during critical missions, such as urban search and rescue and bomb disposal, requires careful velocity control when different types of terrain are found in the scenario. This can particularly be challenging when mission time is limited and the operator’s field of view affected. This paper presents a method for online adapting robot velocities according to the terrain classification from vision and laser readings. The classifier adapts itself to illumination variations, and can be improved online given feedback from the operator.

    • This paper emphasizes the importance of recognizing the major role played by mutual coupling effects in array antennas, especially for dense arrays with element spacing within half a wavelength. For array operation in transmit mode, backward waves emanate both from element ports that are excited by forward waves, and from non-excited ports. This overall and excitation-dependent mismatch is herein characterized in terms of a decoupling efficiency. It is shown how this decoupling efficiency reduces to the well-known impedance mismatch factor for a conventional phase-steered array (all elements excited), and to the embedded element efficiency (only one element excited) used to characterize multiple input and multiple output (MIMO) arrays. Generally, this decoupling efficiency can be used to characterize the combined effect of mismatch and coupling for any excitation of an array, for instance, the Airy pattern excitation of focal plane array (FPA) feeds for reflectors, as showcased here. The abovementioned forms of decoupling efficiencies are evaluated and discussed for an example array of idealized rectangular apertures with special emphasis on the Airy excitation.
